Abstract:
This study aims to provide a procedure for performing microscopy in fish gonads. Specimens used in this study were the ovaries and testes of fish collected in the Mekong Delta from April to July 2022. Organs at five different stages for each sex (ovary diameter from 1.16 cm in stage I to 4.04 cm in stage V; testis diameter from 0.42 in stage I to 2.39 in stage V) were studied in this procedure. The results showed that both ovaries and testes needed to be fixed at 4% formol for 24 h and under running water for eight hours. Ovaries and testes were then immersed in 500 ethanol for 24 h to initiate dehydration. Results showed that the time for dehydration and paraffin infiltration was longer in the ovary (26 to 55 h across stages) than in the testis (25 to 26 h across stages). The mean staining time of the ovary was 1.5 min (stage I) to 3.5 min (stage V) in Hematoxylin and 50 s (stage I) to 140 s (stage V) in Eosin. Whereas the staining time of testis was shorter, 1 min (stage I) to 3 min (stage V) in Hematoxylin and 30 s (stage I) to 90 s (stage V) in Eosin. This study provides a reference for further studies on the histological structure of fish gonads. These results could be applied to fish species inside and outside the Mekong Delta.

Abstract:
Purpose. The objective of the present work was to study the specific features and the effectiveness of the management of glaucoma in the children presenting with stages IV and V of retinopathy of prematurity after vitreoretinal surgical treatment of retinal detachment. Material and Methods. A total of 172 patients (299 eyes) with stages IVa, IVb, and V of retinopathy of prematurity were involved in the prospective cohort study after they had undergone the vitreoretinal surgical intervention for the treatment of retinal detachment. The duration of the follow-up period was 5 years. The treatment of glaucoma included cyclophotocoagulation and the implantation of the Ahmed glaucoma valve drainage. Results. In the early postoperative period, the anatomical success of the surgical treatment of retinal detachment at stages IVa and IVb of the retinopathy of prematurity was estimated at 95,3% and 97,1% respectively; the frequency of smoothing out of the retina associated with the formation of the retinal reattachment zones at stage V of the disease amounted to 63.6%. The frequency of the development of glaucoma was estimated to be 24,0% at stage IVa, 18,3% at stage IVb, and 9,2% at stage V. The clinical manifestations of glaucoma developing at stage IVa retinopathy of prematurity were especially well apparent in the patients with the congenital disease. The secondary neovascular glaucoma without buphthalmos was usually associated with stages IVb and V of retinopathy of prematurity. The effectiveness of cyclophotocoagulation efficacy was estimated to be 36,4% at stage IVa compared with 50,0% and 57,3% at stages IVb and V respectively. The implantation of the Ahmed glaucoma valve drainage proved efficient in 9 out of the 10 cases; still 77.8% of the patients needed a repeated revision for the evaluation of the state of the Ahmed valve drainage. The study has demonstrated that the overall success of the glaucoma treatment in the children presenting with stages IVa retinopathy of prematurity was 100% compared with 3.5% and 33.3% in those with stages IVb and V of the disease respectively. Conclusion. The glaucoma developing in the children with stages IV and V of retinopathy of prematurity is a refractory neovascular condition especially if it is associated with stages IVb and V of the primary disease.

Abstract:
The purpose - revealing of features of the structural organization of the articulate cartilage of a knee joint at patients with various stages of arthrosis. Materials and methods. Morphological researches of an articulate cartilage are visited at 52 patients with various stages of gonarthrosis. The control group was made by 5 men and 5 women in the age of from 18 till 25 years with traumatic damages of components of a knee joint. The basic group was made by 42 persons. On a stage of gonarthrosis patients have been divided into 2 subgroups: 23 patients with 1-2 stage and 19 patients with 2-3 stage of gonarthrosis. The histological material for research has been received at performance medical-diagnostic arthroscopies a knee joint. Results. It is noted, that morphological changes in an articulate cartilage at gonarthrosis are connected with change of density intercellular matrix and, depending on a stage of process, of chondrocyte dystrophy, necrosis and apoptosis are took place. In cytoplasm of chondrocyte there is a decrease in concentration of cytoplasmic organelles, lipids accumulation and electron density inclusions. Conclusion. The most expressed structural changes in the articulate cartilage, connected with local destruction of chondrocytes, lipids and electron density inclusions accumulation are noted at patients with 2-3 stage of gonarthrosis.

These cells were phenotypically classified into three developmental stages (stages II through IV) according to Nadler's criteria. All of them showed the Ig heavy chain (IgH) gene rearrangement, and 67% of the cells in stage IV had the rearranged TRG gamma, albeit seldom in other stages. We further analyzed IgH gene rearrangements in detail using upstream to DH (diversity region of IgH) region probe to distinguish DJ from V-DJ recombination. All dual genotyped cells in each stage except one case in stage II showed the V-DJ rearrangements. This suggests the cross- lineage involvement of the putative recombinase, particularly in the process of V-DJ rearrangements. We next examined the transcriptional status of Ig genes as an indirect reflection of the accessibility of these genes to the recombinase. Properly spliced IgH transcripts of normal size were observed in stage IV and surface Ig positive stage, but not in stage II nor III. However, IgH transcripts of aberrant sizes were seen in stage II, III, and also IV. Cross-lineage gene rearrangements were shown to occur frequently when normally spliced IgH gene begins to be transcribed or just before this. These findings may implicate that V-DJ or V-VDJ gene rearrangements forming functional IgH genes, induce frequently TCR or TRG gene rearrangements. We propose these dual genotypes are different in origin from those observed in stem cell leukemia.

На основании сравнительного анализа определены инвариантные элементы модели подготовки будущих педагогов к организации внеурочной деятельности: целеполагание курсов «Педагогика» и «Психология» на проектирование и формирование навыков развития личностных и метапредметных универсальных учебных действий обучающихся; включение в содержание курсов педагогики и психологии отдельных тем (модулей), направленных на формирование когнитивного компонента обучающихся; организация производственной практики на базе детских оздоровительных лагерей, ориентированной на формирование навыков организаторской (вожатской) деятельности; проектирование и реализация производственной практики на базе общеобразовательных учреждений, сосредоточенной на формирование навыков организации внеклассной деятельности. Предлагается периодизация этапов подготовки будущих учителей к организации внеурочной деятельности учащихся в России: дореволюционный (1900–1917 гг.) – этап создания самостоятельных учреждений внешкольного (дополнительного) образования для взрослых и детей; советский (1917–1991 гг.) – этап формирования государственной модели внешкольной, внеклассной работы с детьми; современный (с 1992 г. и по настоящее время) – этап формирования компетентностной модели подготовки будущих педагогов к организации внеурочной деятельности. Основанием для данной классификации служит содержание и направленность подготовки будущих учителей. Based on the comparative analysis, the invariant elements of the model of training future teachers for the organization of extracurricular activities are determined: the goal setting of the courses "Pedagogy" and "Psychology" for the design and formation of skills for the development of personal and metasubject universal educational actions of students; the inclusion in the content of the courses of pedagogy and psychology of individual topics (modules) aimed at the formation of the cognitive component of students; organization of industrial practice on the basis of children's health camps, focused on the formation of organizational skills (leadership) activities; design and implementation of industrial practice on the basis of general education institutions, focused on the formation of skills for organizing extracurricular activities.A periodization of the stages of training future teachers for the organization of extracurricular activities of students in Russia is proposed: pre-revolutionary (1900-1917) – the stage of creating independent institutions of extra-curricular (additional) education for adults and children; Soviet (1917-1991) – the stage of formation of the state model of extracurricular, extracurricular work with children; modern (from 1992 to the present) - the stage of formation of the competence model of training future teachers for the organization of extracurricular activities. The basis for this classification is the content and orientation of the training of future teachers.

AbstractA wear test of Ti2Ni/TiNi-based coating reinforced by TiC/TiB against a YG6X [94 wt.% tungsten carbide (WC) and 6 wt.% cobalt (Co)] ball in a sliding time of 712 h was carried out using a ball-on-disc reciprocating motion mode. The relationship between accumulated dissipated energy (∑E) and accumulated wear volume (∑V) was accurately established via the dissipated energy approach. Three wear stages were found: initial wear stage (0–200 min), breaking-in wear stage (200–14,520 min), and steady wear stage (14,520–42,720 min). At the initial wear stage, the relationship between ∑E and ∑V satisfied the equation ∑V=42.5288∑E+0.019. The curvilinear relationship in the other two stages was fitted into the equation ∑V=-0.0029∑E4+0.064∑E3-0.5353∑E2+2.0653∑E+0.9938. The wear mechanism in the entire wear process was revealed. ∑E at the initial wear stage was mainly used for debris formation. At the breaking-in wear stage, ∑E was dissipated by generating the friction heat, which led to the formation of a protective layer. At the steady wear stage, ∑E was used to generate friction heat as well as to damage and restore the protective layer. The detection results of the protective layer via energy dispersive spectroscopy showed that this layer was composed of oxides and WC debris.

Abstract:
Purpose. This study examined the relationship between stages of change, other psychosocial factors, and fruit and vegetable (F&V) consumption among rural African-Americans participating in a 5 a Day study. Design. The cross-sectional design assessed associations between F&V intake, stage of change, self-efficacy, beliefs, barriers, and social support. Setting. Participants were surveyed by telephone. Subjects. Subjects were 3557 adult church members (response rate, 79.1%), aged 18 and over from 10 North Carolina counties. Measures. A seven-item food frequency measured F&V intake. Stage of change was measured using four items; other psychosocial variables were measured using Likert scales. Chi-square tests and analysis of variance were used in statistical analyses. Results. The majority of participants (65 %) were in the preparation stage of change. Individuals in action/maintenance consumed an average of 6.5 daily F&V servings compared to 3.3 to 3.5 servings for those in precontemplation, contemplation, and preparation. Self-efficacy, social support, and belief about how many daily F&V servings are needed, were positively associated with stage. Barriers were most prevalent among precontemplators. Conclusions. The findings support the applicability of the stages-of-change model to dietary change among rural African-Americans. The relationship between stage, self-efficacy, social support, and barriers supports using a multicomponent intervention strategy.

AbstractHealthy buds of Nicotiana plumbaginifolia growing in the Kashmir University Botanic Garden were selected for the present study. Flower development and senescence was divided into seven stages, viz., tight bud stage (I), mature bud stage (II), pencil stage (III), partially open stage (IV), open stage (V), partially senescent stage (VI) and senescent stage (VII). Various physiological and biochemical changes were recorded at each stage of flower development and senescence. Floral diameter, fresh mass, dry mass and water content showed an increase up to flower opening (stage V) and thereafter a significant decrease was recorded as the flower development progressed towards senescence through stages VI and VII. An increase in α-amino acids, total phenols and sugars was registered towards anthesis (stage V) and a decrease in these parameters was recorded with senescence. Protease activity showed a significant increase towards senescence with a concomitant decrease in soluble proteins. Based on the quantitative analysis of various biochemical parameters, the flower opening in N. plumbaginifolia seems to be accompanied by an increase in the water content, soluble proteins, α‑amino acids and phenols. A decrease in these parameters, besides an increase in protease activity induces senescence in the beautiful flowers of N. plumbaginifolia. Understanding flower senescence may help in improving the postharvest performance of this beautiful ornamental flower to make it a potential material for the floriculture industry.

Abstract:
Background Kidney damage in chronic kidney disease (CKD) disrupts the 1?-hydroxylase enzyme, preventing the conversion of vitamin D into the active form of calcitriol. To our knowledge, no previous studies have assessed calcitriol levels in children with CKD. Decreased vitamin D levels may occur at an early stage of the disease, so it is important to evaluate calcitriol levels in children with early stage CKD. Objective To assess calcitriol levels in children with CKD according to disease stage and other characteristics. Methods This cross-sectional study was conducted on 43 pediatric CKD patients at Dr. M Djamil Hospital, Padang, Indonesia. We recorded patient characteristics and performed laboratory tests, including routine hematology, blood urea nitrogen (BUN), creatinine, uric acid, electrolytes, calcium, and calcitriol levels. Based on estimated glomerular filtration rate (GFR), patients were grouped into either early-stage (stages I and II), or advanced-stage (stages III to V) CKD. Univariate and multivariate analyses were conducted to determine the association between calcitriol levels with disease stage and other characteristics. Results The overall mean calcitriol level of our subjects was 108.77 (SD 10.79) pmol/L. Mean levels at each CKD stage from I to V were 164.28 (SD 160.90), 94.14 (SD 50.63), 72.16 (SD 13.18), 62.92 (SD 4.87), and 67.51 (SD 4.87) pmol/L, respectively. Calcitriol levels did not differ significantly by CKD stage (P=0.114) when each stage from I to V was considered separately. There was no significant difference in calcitriol levels by growth characteristics (P=0.944), etiology (P=0.311), or anemic status (P=0.104). However, low calcitriol levels were found in all subjects with advanced stage CKD, compared to 63.6% subjects with early stage CKD (P=0.004). Mean calcitriol levels were significantly lower in CKD stage IV (P=0.049) and stage V (P=0.027) compared to stage I. Conclusions The decrease in calcitriol level occurs at an early stage in CKD. Calcitriol levels are significantly lower in advanced stage than in early stage CKD.

This study aims to describe: (1) folklore structure of local legend Tujuah in Kanagarian Sikucua Timur sub-district V Koto Kampuang Dalam Padang Pariaman district, (2) social function folklore local legend Tujuah in Kanagarian Sikucua Timur sub-district V district Koto Kampuang sub-district Padang Pariaman. This type of research is qualitative research using descriptive methods. The data of this research are folklore of local legends of Tujuah in Kanagarian Sikucua Timur, Subdistrict V, Koto Kampuang Dalam, Padang Pariaman District. The data in this study were analyzed in four stages, namely: (1) the data identification stage, the data collected from the informants were identified through two stages, namely: (a) the stage of transcription, transliteration, and (b) the stage of describing the results of observation, recording, and interview, (2) Data classification or analysis stage, (3) Discussion phase and conclusion of the results of classification or data analysis, (4) Reporting phase. Based on the results of the study, it can be concluded that the folklore structure of local legend Tujuah in Kanagarian Sikucua Timur, V Subdistrict, Koto Kampuang Dalam, Padang Pariaman Regency consists of, namely: (1) language style, language style speech used by folklore in local legend Folklore Pariaman dialect, (2) point of view, (3) character and character, figures found in local folklore local legend Pujuran Tujuah is divided into two, Sutan Bagindo as the main character, and Putri Bungsu, Mandeh Rubiah, Rajo Simanta as a side character, ( 4) background, background of folklore local legend Pujuran Tujuah is a setting, time setting and social setting, (5) plot / plot, plot / plot in folklore local legend Pincuran Tujuah is a conventional plot, and has three stages: , the initial stage or introduction (beginning), middle stage or conflict (midle), and the final stage or end (end), (6) themes, and (7) mandate. Prognostic variables predictive of survival in ovarian carcinoma were studied in 91 patients with stage I–IV disease. Univariate analysis showed that clinical stage (FIGO), presence of ascites, age of patient, histologic type and grade, cellular DNA content, morphometric grade, mitotic activity index, and volume corrected mitotic index (M/V index) were prognostic. Multivariate analysis using a Cox model showed that the stage (best predictor in total material), the M/V index (best predictor in stage I tumors), the DNA content (best predictor in advanced stages), and ascites were independently prognostic. Morphometry and/or DNA cytometry should be standard in the histo-pathological evaluation of ovarian carcinomas.

Abstract Background To investigate the distribution of metastatic cancer cells in the mesentery (referred to as metastasis V) and enrich the understanding of the metastasis of colorectal cancer. Methods A total of two hundred ninety-nine patients who received colorectal operations at the Department of Gastrointestinal Surgery, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology between April 2018 and December 2018 were included. Samples were acquired from the dissected mesentery after the operation, and hematoxylin–eosin staining or immunohistochemistry was used to detect metastatic cancer cells. Pathological factors, including tumor position, tumor size, invasion depth, tumor differentiation, lymph node involvement, local vessel invasion, and perineural invasion, were recorded. Results Metastatic cancer cells in the colorectal mesentery (metastasis V) were detected in 62 of 299 patients. Metastasis V was closely correlated with tumor invasion depth, lymph node metastasis, tumor differentiation, and perineural and vessel invasion by cancer cells. Metastasis V occurred more frequently in patients with T3 stage (26.27%) and T4 stage (40.00%) than in patients with T1 and T2 stages (0% and 2%, respectively). Metastasis V was frequently detected in patients with N2a and N2b stage tumors (51.72% and 61.54%, respectively). Metastasis V was more frequently detected in patients with perineural metastasis and local vessel invasion. In addition, metastasis V incidences in colon and rectal cancer were similar. Conclusion The incidence rate of metastasis V is correlated with tumor staging factors and occurs more frequently in advanced-stage patients.

OBJECTIVES To determine the correlation between skeletal and dental maturity using the cervical vertebral maturation (CVM) index (CVMI) and Demirjian index (DI) in orthodontic patients. METHODOLOGY Panoramic and lateral cephalogram radiographs of 105 patients pretreatment records were retrieved. Tooth calcification stages for mandibular left molar were recorded using Demirjian Index on the panoramic radiograph. The skeletal maturation stages were evaluated on a lateral cephalogram through CVMI staging. Fisher’s exact test and Cramer’s V values were estimated to determine the correlation between DI and CVMI. Weighted kappa statistics were used to determine the reproducibility of inter-observer assessment of DI and CVMI. A p-value of ≤ 0.05 was taken to be statistically significant. RESULTS A statistically significant correlation is present among the various stages of DI and CVMI for the males (Fisher exact = 25.006, Cramer’s V = 0.848, p = < 0.001) and females (Fisher exact = 41.006, Cramer’s V = 0.623, p = <0.001). A comparison of DI with respect to CVMI shows a more advanced skeletal stage in males than for female subjects. CONCLUSION The correlation between CVMI and DI stages was found to be highly significant. Stage D and F of DI were significantly correlated with stage 3 and stage 4 of CVMI in females and males respectively.

ABSTRACT Free and conjugated 17α, 20β-dihydroxy-4-pregnen-3-one (17, 20β-P), 17α, 21-dihydroxy-4-pregnene-3, 20-dione (11-deoxycortisol) and 3α, 17α, 21-trihydroxy-5β-pregnan-20-one (3α, 17, 21-P-5β) were measured by radioimmunoassay in matching blood plasma and urine samples from plaice (Pleuronectes platessa) females at several ovarian maturity stages: post-vitellogenesis (IV), final oocyte maturation (V), and ovulation (VI). Free steroids were generally low in all samples. Conjugated steroids were up to 2 orders of magnitude higher in urine than in plasma samples. Conjugated 17, 20β-P was higher in stage V than in stage IV or VI females. Conjugated 11-deoxycortisol was higher in stage IV and V females. Conjugated 3α, 17, 21-P-5β was higher in stage V and VI females. These results support earlier studies which indicated that 17, 20β-P was the most likely maturation-inducing steroid (MIS) in plaice, and that the urine might be a vehicle for steroid pheromones synthesized by the gonads.

The current paper has presented the study results on the change of adaptability to water and temperature stresses and the value of productivity according to the stages of variety changing. There have been selected 13 winter bread wheat varieties developed in the FSBSI Agricultural Research Center “Donskoy”. It was found that the varieties of the first stages of the variety changing (I–IV stage, 1950–1989) practically did not differ in the degree of drought tolerance and the value of productivity of winter bread wheat. In this regard, there was carried out an analysis of changes in the adaptability of winter wheat varieties starting from the V stage of the variety changing. The degree of drought tolerance increased from the weakly-medium drought-resistant varieties (37.4–51%) belonging to the V stage of the variety changing to highly drought-resistant samples (80.4–93.3%) belonging to the VII stage of the variety changing. There was a significant productivity increase of the winter bread wheat varieties zoned from 2010 to 2019 (the VII stage of the variety changing). The largest number of stomata per unit of leaf area in the V stage of the variety changing was identified in the variety ‘Don 95’ (12.2 pcs/mm2). In the VI stage of the variety changing, the largest value of this indicator was identified in the variety ‘Don 105’ (18.8 pcs/mm2), and in the variety ‘Krasa Dona’ (26.9 pcs/mm2) in the VII stage of the variety changing. The dry weight of roots of the varieties of the V stage of the variety changing varied from 1.18 (the variety ‘Donshchina’) to 1.41 g (the variety ‘Don 95’), while in varieties of the VI stage this indicator varied from 1.97 (the variety ‘Donskoy Mayak’) to 2.29 g (the variety ‘Don 105’), and the largest dry weight of roots was identified at the VII stage of the variety changing and ranged from 2.34 (the variety ‘Don 107’) to 2.79 (the variety ‘Asket’). The same regularity was established for the change in the amount of roots according to the stages of variety changing.

The effects of protein supplementation in culture medium on development to the hatching and hatched blastocyst stages of cat in vitro-fertilized embryos were investigated. In the first experiment, presumptive zygotes derived from in vitro maturation and in vitro fertilization (IVF) were cultured in modified Earle's balanced salt solution (MK-1) supplemented with 0.4% bovine serum albumin (BSA) or 5% fetal bovine serum (FBS) for 9 days. There were no significant differences between the BSA and FBS groups with respect to the proportion of cleavage and development to the morula and blastocyst stages of zygotes. However, the presence of FBS in the medium enhanced development to the hatching blastocyst stage of zygotes compared with the BSA group (31.4% v. 7.8%). Moreover, 2.9% of zygotes cultured with FBS developed to the hatched blastocyst stage. The mean cell number of blastocysts derived from zygotes cultured with FBS was significantly higher (P&lt;0.01) than that from zygotes cultured with BSA (136.6 v.101.5). In the second experiment, embryos at the morula or blastocyst stage, which were produced by culturing in MK-1 supplemented with 0.4% BSA after IVF, were subsequently cultured in MK-1 with 0.4% BSA or 5% FBS. Significantly more morulae developed to the blastocyst (P&lt;0.05) and hatching blastocyst stages (P&lt;0.01) in the FBS group than in the BSA group (71.5% and 53.6% v. 44.9% and 6.0%, respectively). Although none of the morulae cultured with BSA developed to the hatched blastocyst stage, 11.5% of morulae cultured with FBS developed to the hatched blastocyst stage. Moreover, the proportion of development to the hatching blastocyst stage of blastocysts was significantly higher (P&lt;0.01) in the FBS group than in the BSA group (68.7% v. 9.8%). None of the blastocysts cultured with BSA developed to the hatched blastocyst stage, whereas 7.3% of blastocysts cultured with FBS developed to the hatched blastocyst stage. The results of the present study indicate that supplementation with FBS at different stages of early embryo development promotes development to the hatching and hatched blastocyst stages of cat IVF embryos.

This study aims to develop and analyze the validity of the developed e-module. This research is a type of educational design research using the Plomp model. At the validation stage, the feasibility of the e-module was assessed by three lecturers from the chemistry department, FMIPA UNP and two teachers from SMA Negeri 1 Lintau Buo. The validation results were obtained from experts consisting of three chemistry lecturers from FMIPA UNP and two chemistry teachers at SMA N 1 Lintau Buo. The research instrument used in this study was a validity questionnaire which was analyzed using the Aikens'V formula. The validation results were obtained from experts. The validation stage was carried out in two stages. The first stage obtained an average V value of 0.82 with an average content component validity with a V value of 0.75 (invalid), the linguistic component with a V value of 0. 88 (valid), the presentation component with a V value of 0.78 (invalid), the graphic component with a V value of 0.85 (valid) and the programming aspect with a V value of 0.84 (valid). The average validity obtained in the second validation stage is 0.86 (valid) with an average content component validity with a V value of 0.85 (valid), the linguistic component with a V value of 0.88 (valid), the presentation component with a V value of 0.88 (valid). V 0.83(valid), component with V value 0.85(valid) and programming aspect with V value 0.87(valid). After validation, the next step is a one to one evaluation test with a one to evaluation questionnaire instrument to three class XI students with different abilities. The conclusion from the research results is that the hydrocarbon e-module based on guided discovery learning for class XI SMA/MA which was developed is valid and can be continued for further research.

Purpose Intensive postoperative surveillance is associated with improved survival and recommended for patients with late stage (stage IIB and III) colon cancer. We hypothesized that stage I and IIA colon cancer patients would experience similar benefits. Patients and Methods Secondary analysis of data from the Clinical Outcomes of Surgical Therapy trial was performed by analyzing results according to TNM stage; early (stage I and IIA, 537 patients) and late (stage IIB and III, 254 patients) stage disease. Five-year recurrence rates were higher in patients with late (35.7%) versus early stage disease (9.5%). Early and late stage salvage rates, recurrence patterns and methods of first detection were compared by χ2 test. Results Salvage rates for early- and late-stage disease patients with recurrence were the same (35.9% v 37%; P = .9, respectively). Median survival after second surgery after recurrence was 51.2 and 35.8 months for early- and late-stage patients, respectively. Single sites of first recurrence did not significantly differ between early and late stage, but multiple sites of recurrence occurred less often in early-stage patients (3.6% v 28.6%, for early v late, respectively; P < .001). Methods of first detection of recurrence were not significantly different: carcinoembryonic antigen (29.1% v 37.4%), computed tomography scan (23.6% v 26.4%), chest x-ray (7.3% v 12.1%), and colonoscopy (12.7% v 8.8%), for early versus late stage disease, respectively. Conclusion Patients with early-stage colon cancer have similar sites of recurrence, and receive similar benefit from postrecurrence therapy as late-stage patients; implementation of surveillance guidelines for early-stage patients is appropriate.

Abstract The expressed human Ig repertoire is not an equal representation of all V(H) segments present in genomic DNA. Studies have shown that a restricted set of V(H) gene segments are over-represented in Ab repertoires of fetal/neonatal and adult B cells. Additionally, this restricted set of V(H) genes is frequently expressed by autoimmune and tumor B cells. To investigate at which developmental stage a bias in the repertoire begins, we compared the V(H)3 and V(H)4 family repertoires of pre-B and immature B cells from bone marrow and mature B cells from peripheral blood of two adults. We found that the V4-34 and V4-59 gene segments of the V(H)4 family and the V3-23 gene segment of the V(H)3 family dominate the repertoires of the surface Ig-negative early pre-B as well as immature and mature B cells. Furthermore, the pattern of utilization of other V(H)3 family members suggests that certain genes that are frequently rearranged during early stages of B cell development are subsequently disfavored during later stages of B cell maturation. We conclude that the over-representation of certain V genes could arise from sequential mechanisms operating at both early and later stages of B cell development. These V(H)-mediated mechanisms might include preferential rearrangement and/or efficiency of pairing with the surrogate light chain at the surface Ig-negative, early pre-B cell stage and ligand selection at more mature, surface Ig-positive, B cell stages.

This preliminary study aims to exploit the biomass of microalgae Chlorella vulgaris; Scenedesmus obliquus; and Chlorococcum sp. in the form of a mixed culture as raw material of alternative fuels. Microalgae were cultivated in the artificial growth medium of PHM (Provasoli Haematococcus Media) for 9 days to reach the exponential phase. Hydrolysis was carried out at a temperature variation of (oC) 25; 80; 100; 120; 140; 160 within 30 minutes by adding hydrochloric acid. Biomass fermentation by adding 50% (v/v) Saccharomyces cerevisiae for 5 day to produce alcohol compounds. The last stage is separation of the alcohol compounds from another compounds by distillation. The result showed that carbohydrate levels with color change indicator in luff schoorl solution at hydrolysis and fermentation stages severely were 12.20 mg/L. Carbohydrate levels in fermentation stage produced 17% (v/v) alcohol compounds while in the distillation stage, 98% (v/v) alcohol will be separated into intermediate compounds such as linoleic acid and methyl alcohol as a characteristic of bioethanol. At 80oC of hydrolysis temperature, 58% bioethanol was produced. Further research is needed, since the preliminary study proves the mixed culture of microalgae is potentially to be utilized in producing bioethanol.

In human, no studies are available regarding changes in kisspeptin1 receptor (KISS1R) sensitivity during pubertal transition. In this study, healthy boys were classified into 5 Tanner stages of puberty (n=5/stage). Human kisspeptin-10 was administered to boys at each Tanner stage and to adult men (n=5) as an IV bolus for comparison. Serial blood samples were collected for 30 min pre- and 120 min post-kisspeptin injection periods at 30 min interval for measuring plasma LH and testosterone levels. There was insignificant effect of kisspeptin on LH and testosterone levels in boys of Tanner stages I–III. At Tanner stage IV, the effect of kisspeptin on plasma LH was insignificant. However, a paired t-test on a log-transformed data showed a significant (P<0.05) increase in mean peak post-kisspeptin testosterone level. In Tanner stage V, a significant (P<0.05) increase was observed in mean post-kisspeptin peak LH level as compared to the mean basal LH value. Post-kisspeptin plasma testosterone levels were also significantly (P<0.05) increased as compared to the pre-kisspeptin level in Tanner stage V. Our data suggest that sensitivity of KISS1R on GnRH neurons with reference to LH stimulation in boys develops during the later part of puberty reaching to adult level at Tanner stage V. We investigated the incidence of weight gain and its related factors in patients with amyotrophic lateral sclerosis (ALS) who underwent tracheostomy and invasive ventilation (TIV). Seventy-eight patients with ALS and TIV were enrolled and followed up prospectively. We clarified the clinical profiles of patients with increased weight following TIV and examined chronological variations in their body mass index (BMI), energy intake, and serum albumin levels. Post follow-up, we determined their disease stage according to their communication impairment (stage I to V) and investigated factors associated with BMI increase following TIV. Patients with a post-TIV BMI increase ≥1.86 kg/m2 demonstrated a higher incidence of ophthalmoplegia (76.2%), total quadriplegia (61.9%), severe communication impairment (stage V; 33.3%), and hypoalbuminemia than those with a BMI increase <1.86 kg/m2. Patients with stage V communication impairment exhibited a larger and faster BMI decrease before TIV (mean −4.2 kg/m2 and −2.5 kg/m2/year, respectively); a larger BMI increase (mean +4.6 kg/m2) following TIV, despite lower energy intake; and lower albumin levels post follow-up than those with lower-stage communication impairment. Multilevel linear regression analysis demonstrated an independent association between communication impairment stages (stage V) and a post-TIV BMI increase (p = 0.030). Weight gain and hypoalbuminemia during TIV in patients with ALS were associated with the disease stage and may be attributable to the neurodegenerative processes that are peculiar to ALS.

Regular photometric observations of V1500 Gyg (Nova Cygni 1975) have been carried out at the Crimean Astrophysical Observatory of the USSR Academy of Sciences from 1975 till now. The 0.5 m telescope equipped with a TV system is used to estimate the star brightness in V.The decline of the mean brightness is presented in Figure 1. The solid lines (1) and (2) show the changes of the V brightness according to Patterson (1979) and Arkhipova and Zaitseva (1978), respectively. The dotted line is drawn according to our data (Pavlenko 1988). The arrows indicate the amplitudes of the light and colour variations.

Volvariella volvacea, also known as straw mushroom, is an edible mushroom, and a highly nutritious food source. In Malaysia, tremendous increase in the consumption of this mushroom is due to its characteristics; short cropping duration, distinct flavour, and pleasant taste. During harvesting, the maturity of V. volvacea is vital to obtain high quality produce. As the mature stage quickly succeeds the immature stage, farmers usually collect both the immature and mature stages at harvest. Thus, the objective of the present work was to evaluate the physical quality, nutrients, and health risk assessment of V. volvacea at both maturity stages. The mushrooms were cultivated on composted empty fruit bunch (EFB), and harvested after a week at immature (button) and mature (veil opening) stages. The absence of spores at the button stage could lower the metabolic activity after harvest, thus decelerating the deterioration rate. Button stage mushrooms showed significantly higher firmness and higher content of glutamic acid. In term of safety, both stages were within the dietary intake limit for heavy metals. In conclusion, the button stage could be suitable and practical for consumption and commercialisation.

This report proposes the stage volume ratio (Vo/V) as an acoustic design factor for concert halls and presents an investigation of the dependence of the acoustic parameters of an auditorium on the stage volume through computer simulation. Firstly, the ratio of the stage volume to the total volume of a concert hall was defined as Vo/V through case studies of existing concert halls. It was verified using a simple model that the stage acoustic parameter STEarly and audience acoustic parameters G and C80 decreased, whereas, the reverberation time and early decay time increased with increasing Vo/V. Secondly, a computer simulation was performed for nine existing concert halls, while changing Vo/V from −20% to 30%. The room acoustic parameters exhibited the same patterns as suggested by the simple model. Vo/V significantly affected the bass ratio and bass index. A comparison of the effects of Vo/V and the sound absorption coefficient in nine concert halls revealed that Vo/V contributed approximately 15% to the reverberation and affected the bass characteristics more substantially. Thus, Vo/V is a critical design factor when determining the warmth of the audience acoustics. The study results could be used as a basis for acoustic design in the future.

15063 Background: Pancreatic cancer (PC) shows the worst mortality rate in common malignancies, with 5-year survival rate of 4%. The only way to cure the disease is surgical resection of early stage PC. Establishment of a screening strategy to detect early stage PC is eagerly expected. REG4, a member of the regenerating islet-derived (REG) family, are secreted proteins that play a role in tissue regeneration and inflammation in digestive organs. We reported overexpression of REG4 in PC cells and serum, and preliminary data of the serum REG4 level of pancreatic disease patients including PC patients. We conducted a prospective study to evaluate the role of serum REG4 in PC. Methods: The series included 57 patients diagnosed pathologically as PC between November 2004 and December 2005. Serum REG4 was quantified by standard sandwich ELISA (Enzyme Linked Immunosorbent Assay) using original kit (MBL116: provided by Medical and Biological Laboratories Co., LTD, Japan) before treatment. The upper limit of the test was set at 3.52ng/ml and was based on studies of serum from 48 healthy control subjects. Results: With a specificity of 100%, the diagnostic sensitivity and accuracy were 63.2% and 80.0%, respectively. The ROC (receiver operating characteristic) analysis showed that area under the curve was 0.91. REG4 levels were a significant differences between PC and control (p<0.001), between each T stage and control (T1,T2, T3 or T4 v control), and between each TMN stage and control (stage 1, stage 2, stage 3 or stage 4 v control), but were not a statistical significance with T stage (T1 v T2 v T3 v T4), M stage (M0 v M1) or TNM stage (stage 1 v stage 2 v stage 3 v stage 4) in PC patients. The diagnostic sensitivity of carcinoembryonic antigen (CEA>5.0ng/ml) and carbohydrate antigen19–9 (CA19–9>50U/ml) was 56.5% and 68.4%, respectively. No significant correlation was demonstrated between REG4 and CA19–9 (coefficient of correlation [rs]=0.45). Conclusions: This study shows the potential of serum REG4 as a screening test for PC, especially for early PC. REG4 is considered to be a more useful marker in combination with CA19- 9. No significant financial relationships to disclose.

The system dynamics applied in this research on modeling a tourist destination (area) life cycle (TALC) contributes to understanding its behavior and the way that information feedback governs the use of feedback loops, delays and stocks and flows. On this basis, a system dynamic three-staged TALC model is conceptualized, with the number of visitors V as an indicator of the carrying capacities’ dynamics and the flow function V(t) to determine the TALC stages. In the first supply-dominance stage, the model indicated that arrivals are growing until the point of inflexion. After this point, arrivals continue growing (but with diminishing growth rates), indicating the beginning of the demand-dominance stage, ending up with the saturation point, i.e., the maximum number of visitors. The simulated TALC system dynamics model was then applied to five EU destinations (Living Labs) to explain their development along the observed period (2007–2019). The analysis revealed that all observed Living Labs reached the second lifecycle stage, with one entered as early as in 2015 and another in 2018. Lifecycle stage durations may significantly differ across the destinations, as do the policies used either to prevent stagnation or to restructure the offer to become more sustainable and resilient.

Five different developmental stages (I–V) were recognized including: (i) early cells and 100% of the vitellum; (ii) formation of the embryonic primordium at the animal pole of the egg; (iii) presence of dark pigmentation on the posterior part of the ocular globe; (iv) appearance of chromatophores on mouth parts and in the abdominal zone; and (v) eye pigmentation in circular–oval form, filling the entire surface. The embryonic development lasts from spawning to hatching approximately 21 days, with the second stage being the longest.

Between 1968 and 1982, 126 patients with pathologic stage (PS) IIB Hodgkin's disease were treated at Stanford University with either irradiation alone or irradiation combined with chemotherapy. Actuarial survival and freedom from relapse rates at 10 years for the overall group were 81% and 74% respectively, with no statistically significant difference between the treatment approaches. The impact of the severity and number of constitutional (B) symptoms, as defined by the Ann Arbor Conference, was analyzed. Patients who presented with all three B symptoms had significantly poorer survival and freedom from relapse compared with those patients with only one or two B symptoms (for survival differences, P = .005 and .007; for freedom from relapse differences, P = .002 and .04). Male sex was the only other prognostic factor that correlated with a poor outcome. At 10 years, the survival rate was 66% for males v 84% for females (P = .01), and the freedom from relapse rate was 75% for males v 89% for females (P = .02). The presence of extralymphatic sites of involvement, age greater than 40, or involvement of greater than three lymphoid sites had no significant adverse effect on either freedom from relapse or survival. Patients with large mediastinal masses treated with irradiation alone had a 10-year freedom from relapse rate of 54% v 81% for those treated with combined-modality therapy (P = .15), but there was no significant difference in survival rates (85% for irradiation alone v 71% for combined modality therapy). Treatment recommendations for stage IIB Hodgkin's disease are discussed.

Study of the administrative proceedings order of implementation as a whole involves the determination of the essence of individual stages of such proceedings. The stage of consideration of the case on the merits due to the specificity of the procedural actions involves a number of both procedural actions designed to ensure a full and impartial clarification of the truth in the dispute, as well as compliance with certain issues of the traditionality of the court session. In other words, consideration of the case on its merits is usually carried out within the framework of a court session, which is characterized as the procedural activity of almost all participants in the process, without exception, in one place. This allows for a more qualified (availability of experts and specialists) and objective (possibility to ask participants questions) assessment of the entire evidence base and to reach certain conclusions. The subject of the research is the norms of the current legislation and the scientific views of famous scientists regarding the meaning and content of such a stage as consideration of the case on its merits. During the research, using the methods of scientific analysis, explanation and generalization, an analysis of existing doctrinal developments and norms of current legislation, which determine the order and forms of administrative proceedings in relation to the judicial review of disputes, was carried out. The result of the study is the characterization of such a stage of administrative proceedings as consideration of the case on its merits, through the prism of understanding the tasks and goals of individual procedural actions. The definition of the essence of the case consideration is emphasized separately as a scientific term. The author's definition of the essence of the consideration of the case on the merits is formulated, that is a purposeful set of procedural actions aimed at clarifying all the necessary circumstances of the case, its resolution on the merits, i.e. (non)recognition of the actions or inactions of the parties to the case as illegal or untrue, with the aim of establishing the fairness of public legal relations in society and providing a real opportunity to the subjects of such relations to fully protect their own interests in court. The conclusions presented in the work can be used in the educational process of legal education students and during the development of new draft laws in the field of administrative legislation.

The purpose of this study was to compare measured , at specific stages with the American College of Sports Medicine-predicted , at the same stage during treadmill walking in older women. Twenty-five women (age range = 56.6 to 78.1 years; mean ± SD = 66.6 ± 5.8 years) walked on a treadmill at a speed of 2.0 mph and 0% grade for the first stage and at increased grades for each subsequent stage. The results showed considerable lack of agreement between the measured and predicted , values. Predicted , was lower than measured , at 0% grade and higher than measured , al 10.2% grade (p < .05 for both). However, predicted , was not significantly different from measured , at grades of 3.4% and 6.7%. These results are generalizable only to older women who can walk 2.0 mph on a treadmill and do not use handrail support. Future research is needed to develop more accurate equations for predicting , for older women.

Abstract Dry sliding tests for Cr–Mo (W)–V hot-work die steels were performed at 400–700 °C using a UMT-3 wear tester. The wear behavior was studied systematically to explore the general characteristics under high-temperature oxidation conditions. When a mild-to-severe oxidation wear transition occurred, the wear rate of the Cr–Mo (W)–V hot-work die steels could be divided into different stages. The wear rate in stage I remained low and increased continuously with increasing temperature in stage II. It decreased within a narrow temperature range in stage III and increased significantly at a high test temperature in stage IV. The microstructures of DM, H21, and H13 steels at 350 HV30 were observed in stage III, and the M 2C carbides were extensively analyzed. Furthermore, by examining the continuous heating transformation curves, the decrease in the wear rate in stage III was qualitatively confirmed, depending on the transformation of the M 2C carbides.
