Full textDue to the high specificity of their interactions, proteins are increasingly used in therapy and represent a vast majority of the global pharmaceutical market. Nevertheless, these molecules are fragile and therapeutic protein stability is a major concern in pharmaceutical industry. Protein degradation and aggregation can occur at every step during production, storage, transport and delivery. In this thesis, we interrogate the possible role of intermittent wetting in protein aggregation. Intermittent wetting frequently occurs in protocols involving pumping (cavitation), agitation, and liquid handling. During intermittent wetting, the air/liquid and liquid/solid interfaces meet at a triple line or triple interface, which is a local trigger for protein aggregation because it concentrates the mechanical action of the recessing fluid on the surface adsorbed proteins. We study the effect of surface intermittent wetting on insulin aggregation. Our results demonstrate that the triple interface line, where an air/water interface meets a hydrophobic surface, allows progressive protein accumulation, and finally triggers local insulin aggregation. We also show that shear stress, alone, is not detrimental for protein stability. Additionally, Additives such as polysorbates were tested, showing that the modification of the surface tension of a protein solution impacts its ability to form aggregates. Based on this work, we propose recommendations for the design of drug delivery and preparation devices in order to limit the risk of protein aggregation at the triple interface

Full textIn this doctoral thesis work, experimental and theoretical studies are carried out on the decomposition characteristics of C5F10O/N2/O2 gas mixture at the gas-solid interface of metal materials and under the discharge and thermal action, and the feasibility and safety of its application are evaluated in combination with the biosafety of C5F10O gas and arc decomposition products of C5F10O/N2/O2. Considering the long-term contact between C5F10O/N2/O2 gas mixture and the internal materials of the equipment during normal operation, the gas-solid interaction stability of C5F10O/N2/O2 gas mixture with commonly used metal copper, aluminum and silver inside the equipment is evaluated, and the mechanism of gas-solid interface interaction between C5F10O gas mixture and metal materials is clarified. Discharge and thermal fault may also occur during the long-term operation of the equipment. The failure decomposition mechanism of C5F10O/N2/O2 gas mixture is studied through experiments and simulations. The typical discharge and thermal fault decomposition characteristics of C5F10O/N2/O2 gas mixture containing different concentrations of oxygen are revealed. The composition and generation rules of decomposition products of the gas mixture under the faults are obtained, and the correlation between the types and contents of characteristic decomposition products and the fault types, as well as the regulation of oxygen to C5F10O gas mixture decomposition products and the inhibition mechanism of solid product precipitation are analyzed. In conclusion, based on the simulation and experimental results, we proposed the stability optimization scheme of silver-plated protective layer on copper surface for metal copper material with poor gas-solid stability of C5F10O/N2/O2 gas. We extracted the characteristic products characterizing the discharge and thermal fault of C5F10O/N2/O2 gas mixture, which provided a reference for the on-line fault monitoring based on the decomposition components. We tested the biosafety of C5F10O and its arc decomposition products, and evaluated its application safety based on the discharge and thermal fault decomposition characteristics of C5F10O/N2/O2 gas mixture, and proposed targeted safety protection measures and suggestions

Full textWe have developped an optical method to study the inter faces of the solid/liquid or solid/gaz type: attenuated total reflexion in surface plasmon oscillations. We present the theoretical principle of themethod, its experimental systems, and its application to the study of polymer adsorption at a solide/liquid interface. We have realized a general computer programme to analyze the experimental data and adjust the values of the thickness and refractive index of the adsorbed film. We demonstrated that to a first approximation, the variation of the reflectivity at a given incident angle, is proportionnal to the adsorbance. We realized two different experimental systems, one using a mechanical scan of the incident angle, and the other performing angular measurements with an array of photodiodes. To demonstrate experimentally the ability of the system, we chose to investigate the adsorption of polystyrene on silver surface, from solution in a good solvant (toluene), and in a solvant theta (cyclohexane). The results obtained kinetics of adsorption, the adsorption isotherm, the average thickness and concentration of the film, are corn- pared to those observed in ellipsometry and other techniques. The conclusion proposes the perspectives for the development of this powerful technique and its application to various systems

Full textThe aim of this work was to determine and quantify phenomena involved in the loss of stability of a catalyst prepared from whole dehydrated cells and used for the direct and continuous dehalogenation of gaseous VOCs. The study of the biocatatalyst instability revealed that (i) whole cells stay alive only few hours with selected operational conditions (temperature, aw, salt buffer borate concentration) but that the dehalogenases performances are not linked to this strong mortality (ii) the organic substrate and product of the reaction (1 chlorobutane et 1 butanol) do not sorb on the catalytic bed and do not inactivate the hydrolysis in the gas phase (iii) the important rehydration of the catalyst is responsible of thermal denaturation of a part of DhaA at 40°C (iv) all the HCl producted is retained by the biocatalyst and consequently reversibly inactivate it. Observation of the biocatalyst highlights that the behaviour of the biocatalyst can be explained by the existence of two different micro-environments for the enzymes (DhaA). Indeed this complex biocatalyst contains 50% of borate buffer salts that act as support for dehydrated cells. During rehydration, part of DhaA remains in the cellular matrix environment and is denaturated and another part is stabilized by a direct contact with borate buffer salts. The lyophilised cellular extract show a stability of 1 800 hours (75 days). The direct contact between salt and DhaA allow to avoid thermal denaturations and inactivation by HCl accumulation
