Pada penelitian ini, pemantauan korosi dilakukan pada sambungan las gesekan baja karbon rendah dan baja tahan karat austenitik (SS 202) yang disimulasikan dalam larutan biologis. Penelitian ini bertujuan menghitung laju korosi sambungan las gesek baja karbon rendah dan SS 202 serta mengevaluasi kekasaran permukaan yang terjadi pada zona pengelasan. Dari hasil sambungan las, dibuat sampel uji korosi dengan mewakili zona pengelasan. Sebelum perendaman, permukaan sampel dibersihkan dan dipoles sebelum perendaman menggunakan larutan elektrolit NaCl 0,9wt.%. Percobaan dijalankan pada 2, 4, 6, dan 8 minggu masing-masing sampel. Hasil penelitian menunjukkan bahwa laju korosi sambungan las gesek baja karbon rendah dan SS 202 tertinggi sebesar 0,0167 mm/tahun, dan terendah 0,0127 mm/tahun. Pada saat yang sama, area sambungan las yang memiliki kekasaran permukaan tertinggi terjadi pada zona plastis. Disimpulkan bahwa seluruh zona pengelasan menunjukkan potensi korosi yang seragam kecuali zona plastis yang menunjukkan perilaku korosi galvanik. ABSTRACT In this study, corrosion monitoring was carried out in the friction welded joint of low carbon steel and austenitic stainless steel (SS 202) simulated in a biological solution. This study aims to calculate the corrosion rate of friction welding joints of low carbon steel and SS 202 and evaluate the surface roughness that occurs in the welding zone. From the results of welded joints, corrosion test samples were made by representing the welding zone. Prior to immersion, the surface of the sample was cleaned and polished before immersion using a 0.9wt.% NaCl electrolyte solution. Experiments were run at 2, 4, 6, and 8 weeks of each sample. The results showed that the corrosion rate of friction welding joints for low carbon steel and SS 202, the highest was 0.0167 mm/year, and the lowest was 0.0127 mm/year. At the same time, the welded joint area that has the highest surface roughness occurs in the plasticized zone. It was concluded that the entire welding zone showed uniform corrosion potential except for the plasticized zone, which showed galvanic corrosion behavior.

Abstract:
Stainless steel (SS) is one of the most commonly used metallic food contact materials. It may be classified based on its microstructure whether ferritic, austenitic, martensitic, duplex or precipitation hardened. Austenitic SS, among mentioned grades, has the largest contribution to market due to its numerous industrial and domestic applications. In this study, the corrosion behavior of AISI 202 SS – a cheaper grade of stainless steel, in three different solution temperatures of citric acid was investigated using different electrochemical techniques such as open-circuit potential (OCP) measurements, potentiodynamic polarization and electrochemical impedance spectroscopy (EIS). The results were compared to that obtained from conventional AISI 304 SS. OCP, polarization and impedance measurements agreed that AISI 202 SS has comparable resistance to that of AISI 304 SS in citric acid at ambient temperature and at 50 °C. At 70 °C, results of OCP measurements suggest that AISI 304 SS exhibited greater performance as indicated by more positive OCP values in the designated solution. EIS results indicate that the two alloys have identical corrosion resistance even at 70 °C as indicated by their comparable polarization resistance (Rp). The corrosion mechanism in both alloys is charge-transfer controlled as indicated by depressed semi-circular appearance of the generated Nyquist plots. The values of corrosion current densities (icorr) extracted from polarization curves indicate that the initial corrosion rates were higher in AISI 304 than AISI 202 SS suggesting that formation of more protective film may have occurred on the former alloy.

Abstract Sjögren’s syndrome (SS) is an autoimmune disease characterized by an exocrine glands epithelium injury with dense lymphocytic infiltrates, mainly composed of activated T and B cells, involvement of the peripheral nervous, pulmonary, blood, kidney, skin, and joint. Present at the interface of genetic and environmental risk factors, histone acetylation is suspected to play a key role in SS. To clarify this point, we investigated the expression of histone deacetylases (HDACs1–10) and histone acetyltransferase (HAT-p300 CREEBP PCAF) by real time PCR in peripheral blood mononuclear cells (PBMCs) from SS patients. Elisa measured HDACs\HATs and histone H3/ H4 acetylation activity. Flow Cytometry and western blot verified histone acetylation level. The expression of HDACs1-10, p300, CREEBP and PCAF in PBMCs from SS patients was decreased in mRNA comparison with healthy controls (HCs). Total histone acetylation was reduced in PBMCs from SS patients, while no difference was observed in histone deacetylation. The extent of total histone H3, but not H4, acetylation in PBMCs from SS patients was decreased compared to that in HCs. An altered balance between HDAC and HAT activity was found in SS PBMCs. As a consequence, part of the pathogenic mechanisms in SS may be linked to epigenetic modifications, thus opening new therapeutic perspectives in SS.

Abstract Objectives Manual systematic literature reviews are becoming increasingly challenging due to the sharp rise in publications. The primary objective of this literature review was to compare manual and computer software using artificial intelligence retrieval of publications on the cutaneous manifestations of primary SS, but we also evaluated the prevalence of cutaneous manifestations in primary SS. Methods We compared manual searching and searching with the in-house computer software BIbliography BOT (BIBOT) designed for article retrieval and analysis. Both methods were used for a systematic literature review on a complex topic, i.e. the cutaneous manifestations of primary SS. Reproducibility was estimated by computing Cohen’s κ coefficients and was interpreted as follows: slight, 0–0.20; fair, 0.21–0.40; moderate, 0.41–0.60; substantial, 0.61–0.80; and almost perfect, 0.81–1. Results The manual search retrieved 855 articles and BIBOT 1042 articles. In all, 202 articles were then selected by applying exclusion criteria. Among them, 155 were retrieved by both methods, 33 by manual search only, and 14 by BIBOT only. Reliability (κ = 0.84) was almost perfect. Further selection was performed by reading the 202 articles. Cohort sizes and the nature and prevalence of cutaneous manifestations varied across publications. In all, we found 52 cutaneous manifestations reported in primary SS patients. The most described ones were cutaneous vasculitis (561 patients), xerosis (651 patients) and annular erythema (215 patients). Conclusion Among the final selection of 202 articles, 155/202 (77%) were found by the two methods but BIBOT was faster and automatically classified the articles in a chart. Combining the two methods retrieved the largest number of publications.

The pitting corrosion behavior of AISI 202 stainless steel (SS) – a low-nickel, austenitic SS grade, was investigated by means of cyclic voltammetry (CV) technique complemented by Scanning Electron Microscopy (SEM). From the starting potential, the current density decreases and changes its sign at the corrosion potential (Ecorr). The anodic response exhibits a well-defined anodic peak followed by a passive region. A noticeable increase in the anodic current density was observed after reaching the breakdown potential (Eb). The second anodic peak which may be attributed to onset of oxygen evolution was also observed. Moreover, the cyclic voltammograms revealed that hysteresis loop is absent for all the studied concentrations, indicating that AISI 202 SS in citric acid is highly resistant to pitting corrosion as also supported by the results of SEM. It was found out that the critical current density (icrit) increases with increasing citric acid concentration.

Abstract Abstract 1511 Poster Board I-534 Background What determines the degree of hemolysis and of anemia in patients with hemoglobin SS is not fully known. The rate of hemolysis and severity of anemia are ameliorated by the presence of alpha thalassemia and by higher hemoglobin F percentage. Mild G6PD deficiency in the form of G6PD-202/-376 may be associated with episodic hemolysis in individuals of African descent, but past studies indicated little influence of G6PD-202/-376 on the degree of hemolysis and anemia in sickle cell disease patients (1,2). In this study we examined the roles of single and double α-globin deletions and G6PD-202/-376 on the degree of hemolysis and the hemoglobin concentration in hemoglobin SS patients. Methods Two hundred sixty two children and adolescents with hemoglobin SS were recruited at three tertiary medical centers and studied at steady state. Principal component analysis was used to develop a hemolytic component from concentrations of lactate dehydrogenase, aspartate aminotransferase and bilirubin. PCR was used to determine the presence of α-thalassemia and G6PD-202/-376. Multivariate models were employed to determine the independent effects of these genotypes on hemoglobin concentration and degree of hemolysis. Results Single a-globin deletion was associated with an estimated 0.4 g/dL increase in steady-state hemoglobin concentration and double α-globin gene deletion with a 0.8 g/dL increase (P = 0.005 for trend) due to, progressively lower degrees of hemolysis (P = 0.004). G6PD-202/-376 was associated with an estimated 0.7 g/dL decrease in the hemoglobin concentration (P = 0.003) (Figure 1a), but this observation could not be explained by increased hemolysis. Rather, the reticulocyte count was an estimated 22% lower with G6PD-202/-376 (P = 0.032) (Figure 1b). Discussion G6PD -202/-376 may be associated with lower hemoglobin concentration in sickle cell anemia and the mechanism is probably impaired erythropoiesis rather than hemolysis. A recent study (3) indicates that G6PD is needed for definitive erythropoiesis as well as for normal survival of red blood cells in the periphery. Our present findings raise the possibility that, in the setting of the markedly increased erythropoiesis of sickle cell anemia, G6PD-202/-376 may result in impairment in erythropoiesis that is discernible in the peripheral blood hemoglobin concentration. Abstract 4-1BB is a member of the tumor-necrosis factor receptor (TNFR) superfamily. As a potent costimulatory molecule, it is critically involved in both enhancement and suppression of immune responses in a disease context-dependent manner. The role of 4-1BB in the development and onset of Sjögren’s syndrome (SS), an autoimmune disease as a result of immune system attacking self-exocrine glands with dryness of mouth and eyes as major clinical symptoms, still remains unknown. To elucidate the precise function of 4-1BB in the pathogenesis of SS, we intraperitoneally administered an agonistic anti-4-1BB monoclonal antibody to female non-obese diabetic (NOD) mice, a well-defined mouse model of this disorder, during the stage prior to SS disease onset. We observed that anti-4-1BB treatment significantly increased the salivary flow rate, indicating an effective amelioration of SS clinical manifestation. Administration of anti-4-1BB antibody markedly suppressed the leukocyte infiltration of the submandibular glands (SMGs) and reduced the percentage of CD4 T cells among total infiltrating leukocytes, without altering that of CD8 T cells and B cells with statistical significance. The decrease in the frequency of CD4 T cells was accompanied by a reduced expression of T-bet but not IL-4 in the SMGs, suggesting an inhibitory effect of 4-1BB activation on type 1 T helper cells. In addition, treatment with this agonistic anti-4-1BB also caused a significant reduction in the level of serum antinuclear antibodies. Taken together, our findings imply that 4-1BB plays a protective role against the pathogenesis of SS-like sialadenitis and its agonists could serve as promising therapeutic agents for controlling this autoimmune disorder.

Seed shape (SS) of soybean (Glycine max (L.) Merr.) is an important morphological trait that significantly affects the quality of marketable seed. Study of the genetic architecture of SS is important and basic to soybean molecular breeding. In the present study, a natural soybean population of 202 diverse accessions mainly from China was used to analyse the genetic basis of SS via genome-wide association analysis (GWAS), which was based on single-nucleotide polymorphisms (SNP) generated by specific-locus amplified fragment sequencing method. In total, 27335 SNPs were finally identified with minor allele frequencies >5%. By using GWAS, 14 quantitative trait nucleotides (QTNs) were identified to be associated with seed length, 13 with seed width and 21 with seed thickness in four tested environments. Among these QTNs, 21 QTNs overlapped or were located in the linked genomic regions of the reported quantitative trait loci related to SS or seed weight; and the other 27 QTNs were novel loci for SS. Ten QTNs showed environmental stability and were detected under at least two environments. In total, 83 genes were predicted in the 200-kbp flanking region of six stable QTNs that could be detected under >three environments. Gene-based association analysis was performed by using 38 accessions of diverse SS; 778 SNPs were found in the 83 genes based on 38 accessions, and 270 SNPs from 41 genes were found significantly associated with SS. Twenty-eight genes were environmentally stable and/or pleiotropic in controlling two or more SS-related traits at the same time. The identified loci along with the candidate genes could be of great value for studying the molecular mechanisms underlying SS and improving the potential seed yield of soybean.

Abstract Sjögren’s Syndrome (SS) is a chronic autoimmune disorder characterized by an increased type 1 interferon gene signature, autoantibody production, and salivary gland inflammation. Engagement of Stimulator of interferon genes (STING) induces type 1 interferon production. Gain of function mutations in TMEM173, encoding STING, results in a severe vasculopathy affecting the skin and lungs. We have previously reported that activation of the STING pathway in mice, using a STING agonist DMXAA, causes an SS-like disease characterized by sialoadenitis and salivary gland dysfunction. Considering that lungs are affected in 9–20% of SS patients, this study was undertaken to investigate lung involvement in DMXAA treated mice. Female C57BL/6 mice injected with DMXAA rapidly upregulated expression of IFNβ, IFNγ, and Mx1 in the lungs at 4 hours. This was followed by an increase in type 1 innate lymphoid cells on day 8. Histopathologic analysis showed the presence of peri-bronchial inflammatory infiltrates on day 35, which persisted until day 57. The lung inflammation was associated with an increased expression of multiple pro-inflammatory genes including CCl20, CXCl10, CXCl9, and IL17α. In addition, there was an increased frequency of lymphatic endothelial cells, suggestive of lymphangiogenesis. Although STING expression was seen in bronchial epithelium and alveolar cells in the lung, bone marrow chimeras between STING KO and wild type mice suggest that STING expression in myeloid cells was critical for lung inflammation. Our results show that systemic activation of the STING pathway might play an essential role in lung inflammation in SS patients.

Abstract HIV-1 envelope (Env) is densely shrouded with oligomannose- and complex-type glycans. These glycans interact with lectins, a family of carbohydrate-recognizing proteins. We evaluated lectins for the ability to block infection of HIV-1 isolates that differ in their sensitivity to neutralizing antibodies. The antibody-sensitive tier 1 viruses were blocked by lectins more easily than the resistant tier 2 viruses. When tier 2 viruses were produced with glycosidase inhibitors to express homogenously oligomannose-type glycans, they became more sensitive to mannose-binding lectins (GNA, GRFT, CVN), suggesting that glycan homogeneity increases virus sensitivity to antibodies and lectins. Further, we tested the effect of glycan content on HIV-1 capture and transmission via DC-SIGN, a C-type lectin expressed by macrophages and dendritic cells. While the capture of virus enriched with high-mannose glycans was enhanced, transmission was diminished due to increased degradation. Under a biologically relevant setting, glycan composition was influenced by the Env signal sequence (SS), and SS mutations altered DC-SIGN-mediated virus transmission. The SS mutants also displayed altered sensitivity to inhibition by lectins GNA and GRFT. Mutations in gp120 did not have these effects. Env SS sequences and glycan compositions also vary among HIV-1 strains, leading to differences in their interactions with and sensitivity to lectins. These phenotypes were associated with the amounts of terminating Manα1–3 vs Manα1–2 residues on the Env oligosaccharides and their recognition by DC-SIGN, GNA, and GRFT. These data demonstrate the importance of the Env sugar contents for HIV-1 interactions with lectins expressed by the host immune cells.

Abstract:
All materials which are intended to have in contact with food and other commodities produced or processed for human consumption are called food contact materials (FCM’s). Stainless steel (SS) – a widely known metallic FCM is used mainly in processing equipment, containers and household utensils. It is known for having numerous industrial and domestic applications worldwide due to its special characteristics of having notable corrosion resistance. However, this corrosion resistance is not all-encompassing since SS may still undergo degradation when subjected to a specific corrosion-inducing environment. SS may be classified according to its microstructure. If the atoms which make up the SS can be viewed as having a face-centered cubic structure, then the alloy is said to be austenitic. This SS grades include the conventional 300-series and the newly-developed 200-series. The former has superior corrosion resistance while the latter is far cheaper. In this study, the corrosion behavior of AISI 202 SS in two different levels of dissolved oxygen (O2) and three acid concentrations was investigated using electrochemical techniques, namely, open-circuit potential (OCP) measurements and electrochemical impedance spectroscopy (EIS). As the concentration of citric acid is increased, the measured OCP values of the alloy decreased and the polarization resistance (Rp) decreased, indicating decrease in alloy stability and decline in the corrosion resistance, respectively. With regards to effects of dissolved O2, results revealed that increasing the level of dissolved O2 has consequently increased the polarization resistance and shifted the OCP to more positive values. All the generated Nyquist plots exhibited a depressed capacitive loops indicating that corrosion in the designated solution occurred with charge transfer as the rate-determining step.

Background and Aims. Endoscopic variceal ligation (EVL) of esophageal varices alters the portal pressure. We observed the changes in 2D-shear wave elastography (2D-SWE) measurements of spleen and liver following EVL and tried to identify the predictors for rebleeding and mortality at 6 months. Methods. A prospective observational study of 202 patients who underwent EVL for bleeding esophageal varices was done. 2D-SWE measurements of liver stiffness (LS) and spleen stiffness (SS) and spleen volume (SV) were measured half an hour before, 1 hour, 2 weeks, and 6 weeks after EVL. All were followed up for 6 months for rebleeding and all-cause mortality. Results. 83 patients were in child C (41%). Difference in SV, SS, and LS at 2 and 6 weeks from baseline was noted as Delta 2 (2nd week post-EVL - pre-EVL SV, LS, and SS) and Delta 3 (6th week post EVL - pre - EVL SV, LS and SS), respectively. Mean Delta 2 VOL and Delta 3 VOL were lower in the bleeding and mortality groups. Delta 2 SS, Delta 3 SS, Delta 2 LS, and Delta 3 LS were higher in the rebleeding and mortality groups. These changes were statistically significant. AUROC in predicting rebleeding was the highest for Delta 2 VOL (0.773) and Delta 3 LS (0.764) amongst the USG parameters that performed better than MELD score (0.677). AUROC in predicting mortality was the highest for Delta 3 VOL and Delta 2 VOL-0.873 and 0.842, respectively, and higher than MELD’s (0.641). Statistically significant variables in binary logistic regression analysis for rebleeding were Delta 3 LS and Delta 3 SS and none for mortality. Conclusion. LS, SS, and SV change after EVL. Changes in liver and spleen stiffness at 6 weeks from baseline had good diagnostic accuracy for predicting rebleeding at 6 months.

O experimento foi realizado em Rolândia (PR), em esquema fatorial 6x3 (clones x porta-enxertos), em delineamento inteiramente casualizado, com cinco repetições e parcelas subdivididas em duas safras (2009/2010 e 2010/2011). As plantas foram conduzidas em sistema de pérgula, no espaçamento de 4,0 m x 2,0 m. O clone '22' sobre o 'IAC 766' destacou-se com médias superiores de produção, produtividade e número de cachos por planta, bem como menor acidez titulável (AT). O clone '211' sobre o 'IAC 572' apresentou menor AT e médias superiores para número de cachos por planta, teor de sólidos solúveis (SS) e relação SS/AT. O porta-enxerto '420-A' proporcionou aos clones de 'Concord' maiores teores de SS e maior relação SS/AT.

Plasma arc welding is an arc welding process wherein coalescence is produced by the heat obtained from a constricted arc setup between a tungsten electrode and the workpiece material. In the present work, the 6 mm thick stainless steel 202 plates are welded using plasma transferred arc welding. The welding parameters such as powder feed rate, welding voltage and welding current were carefully selected. The effect of these parameters on mechanical properties such as tensile strength, impact strength and hardness of the joints were studied. As a result, the increase in tensile strength and impact strength was obtained for the high current, low voltage and powder feed rate. But the highest hardness was attained for the joints made at low current, low feed rate and low voltage.

Pada penelitian ini menggunakan variasi jenis pendingin air garam dan oli dengan teknik pengelasan SMAW menggunakan material Baja AISI 1045 dan Stainless Steel 202 dan elektroda tipe E309 berdiameter 2 mm. Tebal material yang digunakan 5 mm. Hasil yang diperoleh dari data uji kekerasan Rockwell dan uji tarik menunjukan adanya pengaruh jenis media pendingin terhadap kekerasan dan tegangan tarik. Hasil dari kekerasan Rockwell untuk daerah las pada pendingin oli sebesar 53,30 HRC lebih tinggi pendingin air garam dengan nilai 58,40 HRC. Sedangkan untuk hasil uji tarik, pendingin oli memiliki nilai yang lebih tinggi dibandingkan pendingin air garam. pendingin air garam sebesar 0,324496 kN/mm2 dan pendingin oli sebesar 0,333392 kN/mm2.

Pengelasan logam adalah suatu proses pengelasan yang dilakukan pada dua jenis atau paduan logam yang berbeda. Pengelasan logam beda jenis banyak dipakai di berbagai industri, misalnya pembangkit listrik, industri transportasi, kontruksi sipil, dan lain-lain. Baja merupakan material yang banyak digunakan untuk kontruksi. Aplikasinya banyak disambung dengan logam lain. Penyambungannya dilakukan dengan pengelasan. Dalam penggabungan dua logam yang berbeda permasalahan yang sering timbul dalam pengelasan antara lain perbedaan titik lebur, koefisien muai, sifat fisis dan mekanis. Oleh karena itu dengan pemilihan elektroda pengelasan yang tepat akan menghasilkan sambungan yang baik. Penelitian ini bertujuan untuk mengetahui pengaruh jenis elektroda pada baja AISI 1045 dan SS202 terhadap struktur mikro dan kekuatan tarik. Elektroda yang digunakan pada penelitian ini adalah jenis E 6013 dan E 7018. Metode pengelasan menggunakan las SMAW. Struktur mikro dikarakterisasi menggunakan mikroskop optik. Kekuatan mekanik diuji menggunakan mesin uji tarik. Hasil pengamatan struktur mikro pada masing-masing spesimen menunjukkan adanya perbedaan susunan. Struktur mikro didominasi oleh ferit dan perlit. Dengan menggunakan elektroda E7018, menghasilkan perlit yang lebih halus. Berdasarkan uji tarik, terdapat perbedaaan yang signifikan. Pada spesimen E 6013 memiliki kekuatan tarik rata-rata sebesar 275,7 kN/mm2, sedangan E 7018 memiliki kekuatan rata-rata sebesar 419,5 kN/mm2. Introduction: Dabigatran etexilate is an oral direct thrombin inhibitor. Dabigatran excretion is 80% renal, so exposure increases with severity of renal failure. The US Food and Drug Administration-approved dabigatran etexilate 75 mg twice daily (BID) for patients with nonvalvular atrial fibrillation (NVAF) having severely impaired renal function (creatinine clearance: 15-30 mL/min), based on post hoc pharmacokinetic modeling. We assessed dabigatran exposure at trough and peak levels in patients with NVAF and severely impaired renal function and compared with model predictions. Methods: Patients received dabigatran etexilate (75 mg BID) for ≥7 days before blood sampling; Cpre,ss (steady-state predose concentration; trough) was taken 10 to 16 hours postdose (prior to next dose), and C2,ss (steady-state concentration; peak) was taken 2 hours (± 30 minutes) postdose. Pharmacodynamic parameters at baseline (Ebase), trough concentrations (Epre,ss), and peak concentrations (E2,ss) were assessed by established coagulation assays. Results: Of the 150 patients screened, 60 were treated, of which 40% were male and 78.3% were white; median age was 84 years. Cpre,ss values (n = 51) were close to pharmacokinetic modeling predictions with a geometric mean (gMean) of 155 ng/mL, geometric coefficient of variation (gCV) of 76.9%, and range of 15.6 to 498 ng/mL. The C2,ss values (n = 59) had a gMean of 202 ng/mL, gCV of 70.6%, and range of 42.0 to 680 ng/mL. Pharmacodynamic effects on coagulation paralleled dabigatran concentrations. Eleven (18.3%) patients had ≥1 adverse event (AE); pharmacokinetic results for these patients versus those without AEs (n = 49) were Cpre, ss: gMean = 206 versus 145 ng/mL, gCV = 64.0% versus 78.3%; C2,ss: gMean = 243 versus 193 ng/mL, gCV = 68.9% versus 70.8%. All bleeding events (8 events in 5 patients) were considered minor by the investigators. Conclusion: Dabigatran exposure levels largely confirmed earlier pharmacokinetic predictions, supporting the use of dabigatran etexilate 75 mg BID in patients with NVAF and severely impaired renal function. Purpose Bone metastasis occurs frequently in patients with endemic nasopharyngeal carcinoma (NPC). The main objective of this study is to evaluate positron emission tomography (PET) using fluorine-18–labeled fluorodeoxyglucose ([18F]FDG) and conventional skeletal scintigraphy (SS) for detecting bone metastasis at initial staging. Auxiliary objectives are to identify risk factors for bone metastasis and features associated with poor survival in patients with bone metastasis. Patients and Methods Patients with endemic NPC before initiation of treatment were enrolled. PET and SS were performed at initial staging and compared using McNemar's paired-sample test. Bone metastasis was considered to be present if there was any reliable evidence identified within 1 year after primary diagnosis. Multiple logistic regression and Cox's proportional hazards models were used for auxiliary objectives. Results Thirty (15%) of 202 eligible patients were found to have bone metastasis. [18F]FDG PET was found to be more sensitive than SS in the patient-based analysis (P = .006) and in the region-based analysis at the spine (P = .001). Advanced N stage was the only significant risk factor (P < .0001), and the coexistence of hepatic metastasis was a prognosticator of poor survival (P = .017). The survival was not significantly better for patients with bone metastasis undetected at primary staging than for those with initially detectable bone metastasis (P = .620). Conclusion [18F]FDG PET is more sensitive than SS for detecting bone metastasis in endemic NPC at initial staging, whereas SS can be considered as supplementary in this setting.

In this research, friction coefficients of engineering polymers such as nylon and polytetrafluoroethylene (PTFE) are investigated under normal load and sliding velocity. Experiments are conducted when nylon and PTFE slide against different counterface pin materials such as mild steel and stainless steel 202 (SS 202). Experiments are carried out at different normal loads 2, 4 and 6 N, and sliding velocities 0.2, 0.4 and 0.6 m/s. Results show that in general, friction coefficient of nylon decreases with the increase in normal load and sliding velocity. On the other hand, during friction process, PTFE shows different trend i.e. friction coefficient of PTFE increases with the increase in normal load and sliding velocity. Moreover, it is observed that at identical operating conditions, the values of friction coefficient of nylon and PTFE are different depending on normal load, sliding velocity and counterface material.

Abstract B cell receptors display V gene-encoded CDRs and hypervariable CDR3, where the former may harbor innate-like antigen recognition characteristics. To test this in vivo, we deployed transgenic mice containing full human CDRH3 diversity but were constrained to single human VH genes: IGHV1-69*01, a human VH reproducibly used to generate broadly neutralizing antibodies (bnAbs) targeting hemagglutinin stalk of Group 1 influenza A viruses or irrelevant IGHV1-2*02. With this system, we could experimentally evaluate the contribution of V gene encoded CDRs to forming the antibody paratope following immune challenge. Exposure to virus or hemagglutinin vaccine-nanoparticles elicited primary antibody responses targeting the bnAb epitope, but only in IGHV1-69 animals. Repeated exposure amplified off-target responses, however, prime/boost with stalk-only nanoparticle (SS-np) refocused serum antibodies to the target-epitope, expanding the human IGHV1-69 bnAb lineage to elicit prototypic IGHV1-69 Group 1 bnAbs. The bnAb pathway was expanded from a genetically endowed but low frequency population of on target germline B cell precursors (&lt;0.3% of the repertoire). IGHV1-69 therefore endows natural cognate anti-influenza targeting affinity which can be exploited through rational vaccine design to redirect antibodies against an otherwise immunologically silent ‘universal’ flu vaccine target. Remarkably, similar on-target germline B cell levels were seen in human PBMC, suggesting SS-np may elicit bnAbs in humans.

Abstract:
In present research, mathematical models have been established to predict the angular distortion in Metal Inert Gas(MIG) welding for 6mm plates of SS 202 grade for butt welded joints. The filler metal used was a continuously fed solid metal wire of stainless steel (304L).100% Argon gas was used to serve the purpose of shielding the weld pool from the atmosphere as it does not dissociate at high temperatures. This prevented any turbulence in the welding arc and deterioration in weld quality. To obtain experimental samples, the design matrix was developed using the statistical technique of central composite rotatable design (CCRD). Analysis of Variance (ANOVA) technique was used for the adequacy check of the models developed. The models developed can be used to find direct and interaction effect of the input parameters, namely welding speed (WS), voltage (V), nozzle to plate distance (NPD), torch angle (Ɵ) and wire feed rate (WFR) on the angular distortion.

Abstract Neutrophilic dermatoses are a group of inflammatory skin disorders characterized by sterile infiltrates of neutrophils. These syndromes include pyoderma gangrenosum (PG) and Sweet’s syndrome (SS), and they are associated with an increased incidence of inflammatory bowel disease, rheumatoid arthritis, and acute myeloid leukemia. IL-1β is detectable in skin lesions, and IL-1 neutralizing therapies have met with success in some patients. Splicing variants and promoter region deletions of protein tyrosine phosphatase-6 (PTPN6) are a feature of SS and PG. Mice lacking Ptpn6 develop a cutaneous inflammatory disease that is dependent on the IL-1 receptor, G-CSF, and neutrophils, but the source of IL-1 and the mechanisms of IL-1 release remain unclear. Here, we investigated the mechanisms controlling IL-1α/β release specifically from neutrophils (Ptpn6ΔPMN) by inhibiting Caspase-8-dependent apoptosis and Ripk1/Ripk3/Mlkl-regulated necroptosis. Loss of Ripk1 from neutrophils accelerated disease onset, whereas combined deletion of caspase-8 and either Ripk3 or Mlkl strongly protected Ptpn6ΔPMN mice. Ptpn6ΔPMN neutrophils displayed increased p38-dependent Ripk1-independent IL-1 and TNF production, and were prone to cell death. Together, these data emphasize dual functions for Ptpn6 in the negative regulation of p38 MAP kinase activation to control TNF and IL-1α/β transcription, and in maintaining Ripk1 function to prevent caspase-8- and Ripk3/Mlkl-dependent cell death and concomitant IL-1α/β release. These findings implicate neutrophils as the dominant producers of IL-1 in neutrophilic dermatoses, and identify novel therapeutic targets for the treatment of these skin disorders.

Discharges from pumping stations to rivers have become an important issue in central Shanghai. Based on the analysis of the contaminant concentrations and the volume of discharged water in central Shanghai over the past five years, this research has identified that the discharges from pumping stations to watercourse were composed of DWDs and WWDs. The total annual discharges were approximately 155 to 202 million cubic meters. The contaminant concentrations in the DWDs were close to those in the local domestic wastewater (DW), while the WWDs usually had higher concentrations of the contaminants than the local DW. In central Shanghai, the total contaminant discharges (2008 to 2012) from pumping stations were 72,900-95,700 t·a-1 for suspended solids (SS), 67,300-88,100 t·a-1 for chemical oxygen demand (COD), 20,200-27,100 t·a-1 for biochemical oxygen demand (BOD5), 2,000-3,000 t·a-1 for ammonia-nitrogen (NH3-N), 4,100-5,900 t·a-1 for total nitrogen (TN), and 800-1,300 t·a-1 for total phosphate (TP).

Object The aim of this study was to describe the pelvic parameters in a sample of healthy Mexican volunteers and to compare them with previously reported data for Caucasian and Asian populations. Methods This was a transversal study that included a sample population of healthy Mexican volunteers. Age, sex, and lateral radiographs of the lumbosacral region with the individual standing to obtain the pelvic parameters of pelvic tilt (PT), sacral slope (SS), pelvic incidence (PI), and lumbar lordosis (LL) were recorded in each volunteer. The data were compared with those previously published for Caucasian and Asian individuals. Results In total, 202 Mexican individuals (81 men and 121 women; mean age 46.5 years, range 18–85 years) were included. There were statistically significant differences between the Mexican and Caucasian control group with respect to PT (11.9° vs 15.78°, respectively) and PI (51.91° vs 56.68°, respectively). Comparison with the Asian population showed statistically significant differences in relation to the Mexican group in terms of the PT (11.5° vs 15.78°), PI (47.8° vs 56.68°), and SS (36.3° vs 40.89°). The mean LL was 60.17° for the Mexican group, 52.3° for the Asian group, and 61.3° for the Caucasian group. A significant difference in LL was found between the Mexican and Asian populations (p < 0.0001). Conclusions A comparison of the values for pelvic parameters and lumbar lordosis across the different population samples revealed statistically significant differences, which can be attributed to the ethnic origin of the individuals.

Background:Although mesenchymal stem cells (MSCs) transplantation have been demonstrated to be an effective therapeutic approach to treat experimental Sjögren’s syndrome (ESS)1, the specific underlying mechanisms remain to be elucidated. Myeloid-derived suppressor cells (MDSCs) were significantly increased with decreased suppressive capacity during disease development in ESS2-3. However, the therapeutic effects and mechanisms by which MSCs regulating MDSCs in SS still remain unknown.Objectives:Here we aim to explore whether regulation of MDSCs was responsible for the beneficial effects of MSC transplantation on SS.Methods:The MSCs were infused intonon-obese diabetic (NOD) mice via the tail vein. The histological features of submandibular glands, lung, saliva flow rate were evaluated. The number and immune-suppressive activity of MDSCs, the subsets of MDSCs, polymorphonuclear MDSCs (PMN-MDSCs) and monocytic-MDSCs (M-MDSCs) in NOD mice were determined. The bone marrow cells under MDSCs differentiation conditions were co-cultured with or without MSCs. The COX2 inhibitor NS-398, anti-TGF-β1, or anti-IFN-β antibodies were added to coculture medium of MSCs and MDSCs induced from bone marrow cells respectively.Results:We found that MDSCs in bone marrow and peripheral blood increased in ESS mice. MSC transplantation ameliorated SS-like syndrome and down-regulated the percentages of MDSCs, PMN-MDSCs and M-MDSCs and promoted their suppressive ability in ESS mice significantly (Figure 1). In vitro, MSCs could down-regulate the differentiation and up-regulate the suppressive ability of MDSCs. Tribological characteristics of steel combinations are observed in this study using rotating tribometer. Experiments are done under different roughnesses ofpin slides on stainless steel disc for different operating conditionsCertain trends are obtained between friction and rubbing durations. During experiment, it is foundthat there are significant relationship between friction coefficient and operating parameters. In fact, at initial stage friction coefficients of tested material are increased at a particular level with time and finally the friction becomes steady for remaining investigation period. Studies indicate that lower values of friction are obtained with higher loads for SS 202 slides against different roughnesses ofmild steel counterface. Under similar conditions,higher values of frictions are found with higher values of sliding velocity. In addition to that, wear rates show higher values with higher load and velocity conditions. Tribological characteristics are varied for different roughnesses of pin materials under experimental ranges of two influencing prameters.

La presente investigación tuvo como objetivo describir el burnout académico de los estudiantes de la carrera profesional de Educación de una universidad pública de Madre de Dios (Perú). El enfoque de investigación fue cuantitativo, el diseño no experimental, y de tipo descriptivo-transeccional. La muestra fue conformada por 202 estudiantes a quienes se les aplicó un cuestionario de datos sociodemográficos y académicos así como el Cuestionario Maslach Burnout Inventory Student Survey (MBI-SS), instrumento con adecuados niveles de validez y confiabilidad. Los hallazgos indican que los estudiantes se caracterizan por presentar niveles moderados de burnout académico y se determinó que algunas variables como el sexo, el grupo etario, la situación laboral y el año de estudio se asocian significativamente con el burnout académico. Finalmente se concluyó que es necesario que se diseñen y ejecuten programas que desarrollen los factores protectores de los estudiantes para que puedan afrontar adecuadamente situaciones adversas y estresantes y se disminuya así la prevalencia del burnout académico.

An integrated technique was investigated, aiming to provide an effective method for the treatment of biologically treated coking wastewater and to reduce pollutants to concentration values below the limits imposed by the legislation. Coking wastewater containing 270 mg/L COD and 202 times color was treated by coagulation using PAC as well as PAC +PAM, respectively. The application of coagulation using PAC+PAM resulted to a much more efficient removal of color, COD and SS in comparison with single coagulation without PAC. The optimal condition for coagulation with PAC was pH 8, dosage 75 mg/L and dynamic condition 1-minute high speed stirring followed by 15-minute low speed stirring. The optimal condition for coagulation with PAC and PAM was: PAM dosage of 6 mg/L, PAC dosage of 70 mg/L, pH of 8 and reaction time of 20 minutes. Both processes of coagulation were found to be efficient for removal of COD from coking wastewater, but ineffective in reducing color.

The influence of sulfate-reducing bacteria Desulfovibrio desulfuricans on stainless steel SS 202 corrosion in neutral media was studied in detail using weight loss and electrochemical routes. The bacterial activity resulted in material loss with an average rate of 0.015 mmpy. The scanning electron microscopy (SEM) analysis showed a significant increase in the sessile bacterial population with the immersion period. Use of 500 ppm palash (Butea monosperma) leaf extract (PLE) reduced the average corrosion rate to 0.002 mmpy. SEM analysis showed a very thin external film formation in the presence of the inhibitor. The X-ray photoelectron spectroscopy studies confirmed the presence of corrosion products such as Fe2O3 and FeS. The gas chromatography-mass spectrometry studies showed the dominant percentage of various terpenoids along with vitamin E as the main components of the PLE. Electrochemical analysis showed the existence of a diffusion barrier. The resistance offered by the diffusion barrier is high in the inhibited sample when compared to uninhibited samples.

Abstract Introduction Sickle cell disease (SCD) is not only responsible for acute vaso-occlusive events but also for chronic vasculopathy that affects many organs including kidneys. Animal studies have suggested that SCD vasculopathy mainly results from chronic hemolysis. However hemolysis markers have not been consistently associated with clinical vascular complications. Moreover, SCD vasculopathy events have been almost exclusively studied in the USA or in Europe, although more than 80% of SCD patients are born and living in sub-Saharan Africa, a very different environment. We have settled the first multinational African SCD cohort to measure the incidence of SCD vascular complications and looked for their predictive factors in sub-Saharan Africa. We present here our first results, focusing on SCD nephropathy with glomerular involvement and urinary loss of albumin. Methods CADRE is an ongoing cohort of SCD patients in five African countries: Cameroon, Gabon, Ivory Coast, Mali, and Senegal. Included subjects are enrolled at steady state and undergo clinical exam, blood sampling for hemoglobin electrophoresis, blood count, creatinine, lactate dehydrogenase (LDH) and bilirubin levels, and urine sample for albumin and creatinine levels, as well as echocardiography. Main outcome for the present study was micro or macroalbuminuria defined as urine albumin/creatinine ratio >30 mg/g, that has been shown to be predictive of chronic renal insufficiency in SCD. We differentiated two main SCD groups: 1) SS and Sβ0 and 2) SC and Sβ+. Frequency of albuminuria at inclusion was calculated in both groups, with further stratification according to age. We looked for clinical and biological correlates to albuminuria in both groups using multivariate logistic regression. Results In June 2013, 3850 SCD patients have been recruited, among them 3032 (2290 SS, 445 SC, 202 Sβ+, 95 Sβ0) had available complete inclusion data. Median age was 15 and 20 years in SS/Sβ0 and SC/Sβ+ group, respectively, with similar female/male ratio of 1.2. Frequency of albuminuria was 36.9% [34.4-38.7] in SS/Sβ0 and 16.1% [13.2-19.9] in SC/Sβ+ patients, and increased significantly with age in SS/Sβ0 patients (Fig 1). Albuminuria remained significantly higher in SS/Sβ0 group than in SC/Sβ+ group, after adjustment for age, sex, country, blood pressure and hemoglobin level (p<.001). In univariate analysis in SS/Sβ0 patients, albuminuria was associated with age (OR for 10 years= 1.2 [1.1-1.4]), systolic blood pressure (OR=1.2 [1.1-1.4]), hemoglobin (OR=0.8 [0.7-0.9]), leucocytes (OR=1.2 [1.0-1.3]), LDH (OR=1.2 [1.0-1.4]) and bilirubin (OR= 1.5 [1.3-1.9]) levels. Multivariate analysis showed that only age (p=0.01), LDH and bilirubin (p<0.001 both) remained significantly associated with albuminuria. Strength of association of hemolysis markers (hemoglobin, LDH and bilirubin) with albuminuria increased significantly with age (Fig 2). In SC/Sβ+ patients, hemoglobin level only was inversely associated with albuminuria. In the whole cohort of SCD individuals, albuminuria was associated with leg ulcers, priapism, cardiomegaly and acute chest syndrome history, after adjustment on age, blood pressure, hemoglobin and SCD genotype. Conclusion SCD glomerulopathy is frequent in sub Saharan Africa affecting more a third of patients. Children (<15 year-old) are especially prone to kidney involvement since we found prevalence of albuminuria almost twice as much as reported in developed countries. Albuminuria is strongly related to hemolysis level in SS/Sβ0 patients, and this association is increasing with age, suggesting a cumulative effect of hemolysis over time. This observation may explain the lack of association between microalbuminuria and hemolysis markers that was previously reported in pediatric SCD cohorts, unlike most adult studies. Clinical associations observed in our transversal data have to be interpreted in the light of an important survival bias since many African SCD patients die during childhood. The prognostic value of hemolysis markers and albuminuria in SCD chronic renal insufficiency has to be further established on longitudinal study. CADRE follow-up study is now ongoing and will provide valuable data on natural history of chronic vascular disease in African SCD patients of all age, in a variety of ethnic groups, in sub-Saharan Africa, where does live the large majority of SCD patients in the world. Abstract The cholinergic system it’s known as a modulator of the intestinal immune system. The aim of this work was to analyze the impact of muscarine and atropine on the production of myeloperoxidase (MPO) on lamina propia of small intestine as well as other local effects, like stimulation of goblet cells and mucus secretion. Three groups of male BALB/c mice were administred by i.p. route: a) control (SS), b) atropine (4 mg/kg) and c) muscarine (50 ng/kg). At 7 day of treatment, histological samples were obtained for Naphtol AS-D method to MPO activity analysis, immunofluorescence assay to localize MPO secreting cells and Flow Cytometry for intracellular TNF-α, IFN-γ, IL-2 and IL-6 measures. In the analysis of the histological samples it was observed that in the group treated with muscarine the number and size of goblet cells and mucus were augmented in the lumen. The number of neutrophils in the villus was also augmented in the muscarine group in comparison of treated with atropine. On the other hand, the immunofluorescence had an increased number of positive cells and activity in tissue to MPO in the muscarine group. In the Flow Cytometry analysis for lamina propia and Peyer’s patches, IFN-γ and TNF-α were augmented in the muscarine group, while IL-2 and IL-6 decreased. In this study, the MPO can be regulates immune functions such as increasing cytokines in macrophages and endothelial cells, also suppressing DC’s function of antigen presentation and switch to adaptive immunity which can lead to more innate response, and increased production of proinflamatory cytokines by lymphocytes.
