Full textThe present research aims at investigating the role of agreement on the field of the "sponsalia" in a diachronic perspective, from the archaic betrothal to the classical one. Special attention is paid to the legal protection of the betrothal "sponsio" in archaic age and to the change of mentality occurred around 90 BC, which led the legal system to deny that, because it was deemed contrary to morality. With regard to the classical age, the relevance of the variously expressed agreement of the people involved has been taken into consideration in order to delineate a premarital kind of "informed liability" system.

Full textIn 2007, Haygood examined US podcast ads to find out its pattern, and in 2010 McGowan chose to do a similar study to find out if Haygood's theories were still correct. It turned out thatHaygood's theories were still up to date and thus the theories had lasted for a long time. Therefore, this study's purpose is to investigate whether the U.S. advertising patterns can beapplied to advertisements in Swedish podcasts, even though Sweden have more laws than the US on how to advertise. In addition, the study will investigate the listener’s attitude towards advertising in podcasts, to find out what the listener considers about ad type and its placement in podcasts. This study was conducted through a combination of quantitative and qualitative methodology. To examine advertising patterns in Swedish podcasts, a quantitative method was applied, and podcasts on iTunes top ten were studied for six weeks. All podcasts were analyzed and documented and then compared with the results from McGowan's study (2010), since this one is the latest. The result showed that the bookend effect also exists in Swedish podcasts, however it appears more in US podcasts. How the advertisement is performed is also similar to the US podcasts, for example, "We’re this week sponsored by..." is similar to "Support for this podcast comes from...". Other advertising patterns that can be applied to Swedish podcasts are selfsufficient advertisements and recurring sponsors. To investigate the listener’s attitude towards podcast advertising, a qualitative method was used in the form of individual interviews. Respondents listened to a selection of advertisements from the top ten on iTunes, and was then interviewed about the advertisements. It was found that advertising is disturbing regardless of its type and placement. Sponsorship, when the program leader speaks, is preferred over traditional advertising. It is also preferred that the advertisement is placed in the beginning of a podcast, but sponsorship can make advertising placed in the middle of a podcast ok, because it can feel less interrupting with sponsorship compared to traditional advertisement.

Full textThe Betrothed was analysed from a point of view focusing on complexity starting from systemic theory. The comparison between two theoretical texts: Lettre à M.R C*** sur l’unité de temps et de lieu dans la tragédie and Del romanzo storico e, in genere, de’ componimenti misti di storia e d’invenzione, highlighted how the idea of complexity, and consequently of relationship at the core of the concept of lifelikeness, has underlied Manzoni’s criticism since its beginning and has troubled it in time. Manzoni’s progressive reflection on verisimilitude has been researched: the which has given an interpretation applied to The Novel, to a complex and articulated dimension through which Manzoni guided the readers to the contemplation of Truth embodied in historical reality, in the form of a betrothal. The ‘quarantana’, the novel given to the publisher in 1842 by Manzoni, has been analysed and a relationship between its three structural parts has been identified: l’Introduzione, thirty-eight chapters , la Storia della colonna infame. In its frame was found the architecture in which Manzoni brilliantly set his Truth.

This thesis introduces SPONGE, a project developed at the Massachusetts Institute of Technology between October 2006 and May 2007. SPONGE is a workshop model based on immersion, absorption and making connections. The project is both an artistic practice and a method for teaching and learning. My intent in the writing that follows is to give the reader a complete sense of SPONGE. I will begin with a narrative description of the first SPONGE. Once the concept of SPONGE is clear, I will put the project in an art historical context by providing a synopsis of site-specific art and social practice. The current debate about the role of aesthetics in socially based artwork underlies my discussion of the role of the visual and the sensory in SPONGE. Talking about aesthetics also invites an exploration of the autonomy of artistic praxis. I will argue that by creating a distancing framework, SPONGE avoids the dissolution of art into life, even as it celebrates content that exists outside of a traditional art context. A space is thereby preserved for criticality. With criticality comes the politics of the SPONGE, which will be considered in terms of pedagogy. I will address the role of the participant and the way in which SPONGE encourages independent thought and the assembly of ideas.
(cont.) Once SPONGE has been evaluated through the lens of aesthetics, criticality, and pedagogy, I will put forth some thoughts on the future of SPONGE. I believe the workshop has the potential to be adapted for use in almost any context and I propose that any field of inquiry may find its place in SPONGE. I have included excerpts in this text from an earlier paper entitled "The Artist as Employee." It is from that paper, and my desire to find a formal structure for my artwork, that SPONGE emerged. I will conclude this thesis with what I feel is an appropriate endpoint for a project that will continue to evolve: a section on the origins of SPONGE.

Full textMarine sponges harbor microbial communities of immense ecological and biotechnological importance. Recently, they have been focus of heightened attention due to the wide range of biologically active compounds with potential application, particularly, in chemical, cosmetic and pharmaceutical industries. However, we still lack fundamental knowledge of their microbial ecology and biotechnological potential. The development of high-throughput sequencing technologies has given rise to a new range of tools that can help us explore the biotechnological potential of sponges with incredible detail. Metagenomics, in particular, has the power to revolutionize the production of bioactive compounds produced by unculturable microorganisms. It can offer the identification of biosynthetic genes or gene clusters that can be heterologously expressed on a cultivable and suitable host. This review focus on the exploration of the biotechnological potential of sponge-associated microorganisms, and integration of molecular approaches, whose increasing efficiency can play an essential role on achieving a sustainable source of natural products.
As esponjas marinhas abrigam comunidades microbianas de grande importância ecológica e biotecnológica. Recentemente, estas têm recebido maior atenção devido ao grande número de compostos com actividade biológica, com potencial aplicação, particularmente, nas indústrias química, cosmética e farmacêutica. No entanto, a ecologia e o potencial biotecnológico dos seus microrganismos ainda permanecem largamente desconhecidos. O desenvolvimento de tecnologias de sequenciação de alta resolução deu origem a novo grupo de abordagens que nos podem ajudar a explorar o potencial biotecnológico das esponjas com um detalhe sem precedentes. As abordagens metagenómicas, em particular, tem poder para revolucionar a produção de compostos com actividade biológica produzidos por microrganismos não cultiváveis, ao permitir a identificação de genes ou clusters de genes biosintéticos com capacidade para serem expressos heterologamente num organismo hospedeiro adequado e cultivável. Esta revisão foca particularmente a exploração do potencial biotecnológico dos microrganismos associados a esponjas, e a integração de abordagens moleculares, cuja eficiência crescente pode desempenhar um papel essencial no desenvolvimento de uma fonte sustentável de produtos naturais.

Protein hot spots provide a large portion of the binding free energy during interact ions, and detecting and characterizing these hot spot regions provides insight that can be used in the development of novel drugs for t he purpose of regulating pathological pathways. In t his dissertation, I first compare t he FTMap algorithm, which detects hot spots by identifying locations where different simulated solvent-sized molecules are consistently found to have favorable interactions, to experimental methods that detect hot spots by alternative means. Specifically, I show that FTMap detects the hot spots detected by alanine scanning, and I discover two roles for residues near hot spots in protein-protein interaction (PPI) complexes. Furthermore, additional insights into the binding energetics of PPis are uniquely provided by FTMap, and these insights are important for drug-design. FTMap is then shown to detect the hot spots identified by successful fragment-screening experiments, and the additional sites detected by FTMap are shown to provide insight into the optimal regions for ligand extension for the molecules identified by t he fragment-screening experiment. Since binding sites are composed of multiple hot spots, we have recently used FTMap for binding site detection. I examine the highly accurate binding site detection algorithm, show that the success of this algorithm is a consequence of only a portion of the scoring protocol, and develop a faster protocol for binding site detection based on this insight. I also quantitate the improvement in precision obtained by using multiple probes and argue t hat the principle biophysical considerations in hot spot detection are hydrophobicity and complexity. Finally, I develop a functional-group clustering algorithm, which is informative for evaluation of the binding locations of pre-determined chemical moieties. I then provide evidence that other approaches employing FTMap results may lead to insight into selectivity. I conclude with a discussion on the nature of hot spots, and I suggest that evolutionary studies of protein divergence should provide insight into the emergence of chemical-selectivity thus providing biophysical insight into the factors that drive selectivity within hot spots.
