Abstract:
In this study, interviewer-based questionnaires of 67 variables were administered to local fabricators and end-users of single-pot biomass stoves (SPBS) in Ghana. Additionally, two randomly selected traditional and improved SPBS were lab-tested using standard performance metrics. From the study, the relationship between fabricators and end-users was conceptualized based on selected indicators and assumptions. The study results indicated that the primary design resources for fabrication were patterns and templates, and that major challenges to fabrication were lack of training in design principles, standards and safety, poor emission efficiency and financial sustainability. Whereas end-users of improved SPBS were less affected by heat and smoke, end-users of traditional SPBS were mostly affected. From hypothesis test, because the calculated χ2cal = 24.05, and is greater than the tabulated χ2crit = 3.841, it is concluded that there is a relationship between heat, smoke effect and gender, and that female end-users of traditional SPBS were particularly affected during cooking. The traditional SPBS emitted more CO2 and CO than improved SPBS. Comparatively, 38% more end-users of traditional SPBS observed charcoal ash residue in the cooking area than improved SPBS users. Four basic practices of managing ashes from SPBS are developed. Finally, a fabricator and end-user framework are developed for energy sustainability and quality improvement.

Abstract:
The regulation and timing of spindle pole body (SPB) duplication and maturation in fission yeast was examined by transmission electron microscopy. When cells are arrested at G1 by nitrogen starvation, the SPB is unduplicated. On release from G1, the SPBs were duplicated after 1–2 h. In cells arrested at S by hydroxyurea, SPBs are duplicated but not mature. In G1 arrest/release experiments with cdc2.33 cells at the restrictive temperature, SPBs remained single, whereas in cells at the permissive temperature, SPBs were duplicated. In cdc10 mutant cells, the SPBs seem not only to be duplicated but also to undergo partial maturation, including invagination of the nuclear envelope underneath the SPB. There may be an S-phase–specific inhibitor of SPB maturation whose expression is under control of cdc10+. This model was examined by induction of overreplication of the genome by overexpression of rum1p or cdc18p. In cdc18p-overexpressing cells, the SPBs are duplicated but not mature, suggesting that cdc18p is one component of this feedback mechanism. In contrast, cells overexpressing rum1p have large, deformed SPBs accompanied by other features of maturation and duplication. We propose a feedback mechanism for maturation of the SPB that is coupled with exit from S to trigger morphological changes.

Abstract:
The NIMA kinase is required for mitotic nuclear pore complex disassembly and potentially controls other mitotic-specific events. To investigate this possibility, we imaged NIMA–green fluorescent protein (GFP) using four-dimensional spinning disk confocal microscopy. At mitosis NIMA-GFP locates to spindle pole bodies (SPBs), which contain Cdk1/cyclin B, followed by Aurora, TINA, and the BimC kinesin. NIMA promotes NPC disassembly in a spatially regulated manner starting near SPBs. NIMA is also required for TINA, a NIMA-interacting protein, to locate to SPBs during initiation of mitosis, and TINA is then necessary for locating NIMA back to SPBs during mitotic progression. To help expand the NIMA-TINA pathway, we affinity purified TINA and found it to uniquely copurify with An-WDR8, a WD40-domain protein conserved from humans to plants. Like TINA, An-WDR8 accumulates within nuclei during G2 but disperses from nuclei before locating to mitotic SPBs. Without An-WDR8, TINA levels are greatly reduced, whereas TINA is necessary for mitotic targeting of An-WDR8. Finally, we show that TINA is required to anchor mitotic microtubules to SPBs and, in combination with An-WDR8, for successful mitosis. The findings provide new insights into SPB targeting and indicate that the mitotic microtubule-anchoring system at SPBs involves WDR8 in complex with TINA.

Abstract:
Background: The past two decades has revealed an unprecedented increasing incidence of skin cancer within the Latinx population. Although Latino day laborers (LDLs) are at heightened risk for developing skin cancer because of the outdoor work in which they engage, there is limited research examining their intentions to engage in sun protective behaviors (SPBs). Therefore, this study sought to assess the explanatory power of the theory of planned behavior (TPB) to identify attitudinal, subjective norms, and perceived behavioral control factors associated with intentions to engage in SPB among LDLs. Methods: This cross-sectional retrospective study consists of a non-random convenience,community-based, sample of 137 LDLs residing in Mississippi and Illinois. Data were collected using a self-report survey centered on health practices and sun-protective behaviors. Results: Findings revealed that five significant factors shaped intentions to engage in SPBs, including barriers to engaging in SPBs (β =.30, P<0.001), benefits of engaging in SPBs (β =.27,P<0.001), education (β=0.20, P<0.01), and acculturation (β=0.18, P≤0.05). The independent variables tested in the model accounted for 42% of the change in intentions to engage in SPBs. Conclusion: This study demonstrates TPB’s usefulness for predicting future intentions to engage in SPBs among LDLs. Moreover, the strongest factor associated with predicting intentions to engage in SPBs among LDLs was perceived behavioral control. Thus, since SPBs are malleable, emphasis is placed on implementing interventions for this population that promote intentions and address perceived behavioral control.

Fusion of double-structured, interphase spindle pole bodies (SPBs) occurred before nuclear fusion in heterokaryotic secondary sporidia. The SPBs of two separate nuclei were juxtaposed with their long axes perpendicular to each other. Also, SPBs were observed oriented with their long axes parallel and fused to each other at both ends. Fusion apparently continued toward the midportion of the SPBs. Nuclei were observed joined together in a narrow region. These nuclei appeared to share a single SPB that was located opposite to a protuberance on both nuclei. Following fusion, the SPB apparently returned to an interphase structure.

Electrochemical biosensors, in which enzymatic biofuel cells simultaneously work as energy power and signal generators, have become a research hotspot. They display the merits of power self-support, a simplified structure, in vivo operational feasibility, online and timely monitoring, etc. Since the concept of enzymatic biofuel cell-powered biosensors (EBFC-SPBs) was first proposed, its applications in health monitoring have scored tremendous achievements. However, the creation and practical application of portable EBFC-SPBs are still impeded by the difficulty in their miniaturization. In recent years, the booming microfluidic technology has powerfully pushed forward the progress made in miniaturized and portable EBFC-SPBs. This brief review recalls and summarizes the achievements and progress made in miniaturized EBFC-SPBs. In addition, we also discuss the advantages and challenges that microfluidic and screen-printing technologies provide to wearable and disposable EBFC-SPBs.

Abstract:
This study aims to analyze the implementation of policy on character education "kasundaan" based on regulation Sumedang Regent about "Sumedang Puseur Budaya Sunda " (SPBS)." The method used is a survey, which is completed with interview and FGD. The results from the study found that: (1) SPBS was only instructed in the "SKPD" environment only in the form of Thursday using "Kasumedangan" dress and Sundanese language, (2) no education design of "kasundaan" characters in elementary school, (3) majority of students elementary schools already have the expected behavior in the context of "kasundaan" values based on Bupati's regulation on SPBS. However, a few other minorities, in fact, show behavior that is not as expected, and this become a potential learner to lead to negative behavior. Thus, it can be concluded that the implementation of character education "kasundaan" in elementary school in Sumedang regency conducted without base to regent regulation about SPBS, but refers to school curriculum only. There is no integration between the regent's regulation on SPBS and the school curriculum, so the values of "kasundaan" characters in the regent's regulation on SPBS have not yet been implemented in a structured way at the elementary school level.

Abstract:
The spindle pole body (SPB) is the equivalent of the centrosome in fission yeast. In vivo it nucleates microtubules (MTs) during mitosis, but, unlike animal centrosomes, does not act as a microtubule organizing center (MTOC) during interphase. We have studied the MT-nucleating activity of SPBs in vitro and have found that SPBs in permeabilized cells retain in vivo characteristics. SPBs in cells permeabilized during mitosis can nucleate MTs, and are recognized by two antibodies: anti-gamma-tubulin and MPM-2 which recognizes phosphoepitopes. SPBs in cells permeabilized during interphase cannot nucleate MTs and are only recognized by anti-gamma-tubulin. Interphase SPBs which cannot nucleate can be converted to a nucleation competent state by incubation in cytostatic factor (CSF)-arrested Xenopus egg extracts. After incubation, they are recognized by MPM-2, and can nucleate MTs. The conversion does not occur in Xenopus interphase extract, but occurs in Xenopus interphase extract driven into mitosis by preincubation with exogenous cyclin B. The conversion is ATP dependent and inhibited by protein kinase inhibitors and alkaline phosphatase. Purified, active, cdc2 kinase/cyclin B complex in itself is not effective for activation of MT nucleation, although some interphase SPBs are now stained with MPM-2. These results suggest that the ability of SPBs in vitro to nucleate MTs after exposure to CSF-arrested extracts is activated through a downstream pathway which is regulated by cdc2 kinase.

The rhythmic oscillating complex (ROC) is described from a series of experiments that surveyed the myoelectric activity of the avian small intestine as recorded from chronically implanted bipolar electrodes. A highly organized myoelectric event in the fasting avian small intestine, the ROC is demonstrated in detail in chickens (Gallus); it is also found in other gallinaceous birds but not in owls (Strix) or mammals. The ROC comprises rapidly propagating bursts of spike potentials (SPBs) that occur in a regular and predictable pattern: single orad SPBs alternate with groups of aborad SPBs. An average ROC in a chicken contains a mean of 78.9 +/- 2.0 (mean +/- SE) SPBs (37% orad, 63% aborad) that rapidly traverse the full length of the small intestine. The aborad SPBs move at mean velocities of 25.0 +/- 0.5 cm/s and last a mean of 0.9 +/- 0.0 s at an electrode site; the orad SPBs are faster (41.2 +/- 2.3 cm/s) and longer in duration (1.3 +/- 0.0 s). ROC activity continues for a mean of 7.6 +/- 0.2 min. ROCs occur only in a well-fasted gut as often as every 3 h and apparently for as long as the bird remains without food. Because ROCs restimulate fed-state activity in the stomach and small intestine, we hypothesize that they recycle nutritive material for further digestive activity in the distal tract.

Abstract:
The asymmetrically dividing yeast S. cerevisiae assembles a bipolar spindle well after establishing the future site of cell division (i.e. the bud neck) and the division axis (i.e. the mother-bud axis). A surveillance mechanism called spindle position checkpoint (SPOC) delays mitotic exit and cytokinesis until the spindle is properly positioned relative to the mother-bud axis, thereby ensuring the correct ploidy of the progeny. SPOC relies on the heterodimeric GTPase-activating protein Bub2/Bfa1 that inhibits the small GTPase Tem1, in turn essential for activating the mitotic exit network (MEN) kinase cascade and cytokinesis. The Bub2/Bfa1 GAP and the Tem1 GTPase form a complex at spindle poles that undergoes a remarkable asymmetry during mitosis when the spindle is properly positioned, with the complex accumulating on the bud-directed spindle pole. In contrast, the complex remains symmetrically localized on both poles of misaligned spindles. The mechanism driving asymmetry of Bub2/Bfa1/Tem1 in mitosis is unclear. Furthermore, whether asymmetry is involved in timely mitotic exit is controversial. We investigated the mechanism by which the GAP Bub2/Bfa1 controls GTP hydrolysis on Tem1 and generated a series of mutants leading to constitutive Tem1 activation. These mutants are SPOC-defective and invariably lead to symmetrical localization of Bub2/Bfa1/Tem1 at spindle poles, indicating that GTP hydrolysis is essential for asymmetry. Constitutive tethering of Bub2 or Bfa1 to both spindle poles impairs SPOC response but does not impair mitotic exit. Rather, it facilitates mitotic exit of MEN mutants, likely by increasing the residence time of Tem1 at spindle poles where it gets active. Surprisingly, all mutant or chimeric proteins leading to symmetrical localization of Bub2/Bfa1/Tem1 lead to increased symmetry at spindle poles of the Kar9 protein that mediates spindle positioning and cause spindle misalignment. Thus, asymmetry of the Bub2/Bfa1/Tem1 complex is crucial to control Kar9 distribution and spindle positioning during mitosis.

Rapporten är ett examensarbete som utförts i den avslutande delen av Högskoleingenjörsprogrammet i Maskinteknik. Examensarbetet är utfört av Anders Dedorsson Ylén, student vid fakulteten för hälsa, teknik- och naturvetenskap på Karlstads Universitet. Examensarbetet har utförts på Arvika Smide AB vars uppdrag gick ut på att införa SPS vid pressgrupp 5, samt undersöka hur väl företaget jobbar med ISO/TS 16949.  I rapporten finns en teoretisk del om SPS och ISO standarden ISO/TS 16949. Vidare beskrivs hur SPS använts som metod för att mäta maskinens duglighet och behov av underhåll, samt hur mätningarna utförts. Här beskrivs också av vad som skett vid renovering av smidespressen och vilka resultateten blev av mätningar gjorda efter arbetet. Lämplig metod för att se vart företaget befinner sig i sitt arbete med ISO/TS 16949 och förslag på förbättrat arbete med ISO/TS redovisas. Rapporten redovisar att SPS kan vara ett styrande medel vid tillståndsbaserat underhåll på en smidespress och hur företaget kan utveckla sitt arbete med ISO/TS 16949.
The report is a master thesis carried out in the final part of the Bachelor of Science education in Mechanical Engineering. The work is carried out by Anders Dedorsson Ylén, a student at the Faculty of Health, technology and natural science at the University of Karlstad. The work has been performed at Arvika Smide AB, whose mission was to introduce SPS at the pressgroup 5, and examine how well the company is working with ISO/TS 16949. The report contains a theoretical part about SPS and ISO standard ISO/TS 16949. It also describes how SPS can be used as a method for measuring machine condition and maintenance needs, and how the test's are carried out. It describes what has happened in the renovation of the forging press, and the results of measurements made after the renovation work was done. An appropriate method is presented to be able to see where the company is in its work with ISO/TS 16949 and suggestions for improved work with ISO/TS is suggested. The report shows that the SPS can be used as a guideline in condition-based maintenance of a forging press and how the company can improve its work with ISO/TS 16949.

Abstract:

The main objective of this thesis is to study how the structures of a sell dialog, via a web shop, should be designed in order to support and inspire the customer. To get the answer to this question I use a qualitative and quantitative method. The qualitative method is applied in connection with different interviews. The quantitative method is used to analyze different answers given from those who are interviewed. I also use the deductive method due to my interest of studying how reality can relate to the theories with the choice of the subject.

Moreover, this thesis is structured by three main perspectives; company, customers and techniques. The theoretical part of the thesis includes definitions and theories about electronic business, customers’ behaviour and technique, UML (Unified Modelling Language), which is a visual syntax for visualising, specifying, constructing and documenting system engineering, and UP (Unified Process), which is a method that complements UML. Furthermore, the empirical part of the thesis includes interviews with Chris Jangelöv and Patrik Cardell (Visma Spcs), who are representing the companys’ perspective on a sell dialog via a web shop. For the customers’ perspective on the subject, I collect and outline the interviews from Visma Spcs customers. Moreover, I review the interview with David Nadel, where he discusses the UML and UP techniques and also electronic business in general.

As for the results, I come to the conclusion that a web shop should be structured from a customer´s perspective and his/her inquiries concerning the products and services. UML and UP, however, are techniques that focus on the structuring of data and processes without taking the developing dialogue with the customer into serious account. These techniques do not address issues of why a web shop is used as it is, UML and UP only apply what a web shop can look like and how it could be used. Therefore, I argue that there is a lack of connection between the UML and UP techniques and the real live user and actual use. I also reached the conclusion that there should be three main building blocks when structuring a sell dialog via a web shop. These are; structure, information and navigation.

Abstract:
En central stabsprodukt i Svensk planerings- och ledningsmetod (SPL) är operationsdesign. Den traditionella operationsdesignen har kritiserats för sin linjäritet, stelbenthet och positiva progression och därmed ansetts mindre lämplig för operationer med defensiv inriktning eller stor osäkerhet. SPL, med målsättningen att vara en allomfattande planeringsmetod, har därför i 2017 års version bland annat tillförts ytterligare tre typer av operationsdesign. Denna uppsats undersöker hur SPLs typer av operationsdesign omhändertar tre defensiva operationsplaner från 1979. Undersökningen sker genom att en kvalitativ textanalys leder fram till efterhandskonstruktioner av planernas operationsdesigner. Denna följs av en kvalitativ innehållsanalys för att se hur dessa operationsdesigner omhändertar försvararens möjlighet att avgöra, undvika ett avgörande samt dennes behov av flexibilitet. Resultatet visar att alla designtyper fungerar väl när ett avgörande söks men sämre när ett avgörande försöker undvikas. Behovet av flexibilitet omhändertas bäst i den dynamiska designtypen och någorlunda väl i den traditionella. Båda har dock svårigheter i övergången till det militära slutläget om ambitionen i målsättningarna sänks. De två designtyperna tematisk och mixad, som främst är ämnade för mycket osäkra situationer, omhändertar däremot dåligt de planer som undersökts då mycket av den operativa inriktningen går förlorad. Sammantaget visar undersökningen att genom tillförsel av de nya designtyperna är SPL en bit på vägen mot en allomfattande planeringsmetod, men svårigheten att applicera stabsverktyget operationsdesign på defensiva operationer är ännu inte helt löst.

In this contribution, María-Ángeles Martínez explores the language of engagement in a short extract from Los Detectives Salvajes (Roberto Bolaño, 1998) and its English translation, The Savage Detectives (2007) within the framework of storyworld possible selves (SPSs). Her analysis focuses on the first cluster of SPS linguistics anchors, or linguistic expressions requiring a hybrid mental referent, inclusive of an intra- and an extra-diegetic perspectivizer, in the novel, and discusses its bearing on storyworld possible selves projection and narrative construal. The main narrative function of this first SPS cluster seems to be to invite the activation of readers' past selves as young, restless university students as the part of their self-concepts with a stronger engagement potential in this specific narrative experience.

Abstract:
We and others have shown that the bleeding side-effect of heparin is not related to the ex vivo anticoagulant effect of heparin, as measured by traditional clotting tests or chromo-genic assays. In recent experimental studies, using a rabbit ear bleeding model, we found that a highly sulfated low molecular weight heparin (LMWH) fraction enhanced blood loss 10-fold, whereas its unfractionated LMWH parent compound only enhanced blood loss 2-fold. We therefore, performed studies to investigate the relationship between the degree of sulfation of 7 sulfated polysaccharides (SPSs) and their bleeding side-effects usina the above model. Blood loss was measured as the increase in 51Cr-red cells lost from 5 standardized cuts in the rabbit ear. The dose of each SPS studied was 20 or 40 times the doses which were required to inhibit thrombosis by ∼75% in a jugular vein/hypercoagulation/stasis model. Ex vivo, we measured the effect of the SPSs on collagen-induced platelet aggregation. Sulfation of each SPS was measured as meq/g of SPS. The*re was a positive correlation between the degree of sulfation of the SPS and blood loss, p<0.05 (Figure a), ie. the greater the degree of sulfation, the greater the amount of blood lost. There was also a correlation between the degree of sulfation of the SPS and inhibition of platelet aggregation, p<0.001 (Figure b). Thus, the dose of each SPS that enhanced blood loss also inhibited platelet aggregation. We conclude that the degree of sulfation of SPS contributes to increased blood loss associated with heparin treatment, and that this effect is due, in part, to the inhibition of platelet aggregation.

Abstract In this study, nickel-zirconia cermet layers are produced by solution precursor plasma spraying (SPPS) and compared with suspension plasma sprayed (SPS) coatings of similar content. Although nickel is uniformly distributed in both coatings, its presence in the suspension caused problems with the SPS process. With the SSPS process, precursor solutions are fragmented into droplets in which Ni, Zr, and Y are intimately mixed, resulting in very fine microstructure without the problems encountered with the SPS process. It was also found that plasma gas enthalpy and spray distance have predominant effects on in-flight pyrolysis of the elements, and that plasma gas mixture has an impact on porosity as well as the oxidation state of the nickel.

Abstract:
Surface plasmons (SPs) are collective excitations of free electrons in a conducting medium bounded by a surface or interface with a dielectric. They can originate surface plasmon-polaritons (SPPs), which are evanescent waves involving electron density oscillations and the associated electromagnetic (EM) field whose amplitude decays exponentially with distance from the surface. SPPs are characterised by a dispersion curve (frequency vs wavevector along the interface) that lies outside of the light cone, so they cannot be excited directly by propagating light; however, it is possible to excite them using the Attenuated Total internal Reflection (ATR) method or a periodic grating. SPs in a confined geometry such as a metallic sphere are localised in space and characterised by discrete frequencies (a single one in the case of sphere, two or three in the case of an ellipsoid). They can be excited directly by propagating light if its frequency coincides with the localised SP frequency, giving rise to the Localized Surface Plasmon Resonance (LSPR). Both SPPs and LSPR are highly sensitive to the dielectric constant of the surrounding media and can be used for environment sensing (e.g. gas molecules that adsorb onto the structure’s surface).

Abstract Continuous development of oilfields fosters a growing need for the simplification of oilfield surface process systems (SPSs) to reduce operating and management costs. Wells, testing stations, transferring stations, and central processing facilities are the main facilities in an SPS; pipelines are used to connect these stations. In this system, production radius (PR) is an important index to determine which transferring station can a testing station be linked to. Different simplification plans will lead to different operating and management costs in the following production period. Therefore, the simplification plan should be carefully designed to minimize cost and facilitate management. This paper proposes an optimization method for the simplification of SPSs in oilfields. First, an evaluation model is developed based on fuzzy analytical hierarchy process (FAHP) to select the transferring stations that need to be decommissioned. Second, hydraulic and thermal calculations are performed to get the data for the calculation of PRs. Third, the PRs, including oil gathering radius, water flooding radius, and hot water washing radius are computed to determine the linkage between the transferring stations and the testing stations. Finally, a construction plan is obtained for new pipelines of the testing stations. A case study is conducted to verify the effectiveness of this method. The results show that this method is suitable for the simplification of SPSs in oilfields.

Tackling the challenges related to recruitment and retention of qualified staff – and developing innovative approaches to the delivery of social care services – is becoming increasingly urgent, particularly in rural and sparsely populated areas (SPAs). This policy brief gives an overview of examples across the Nordic Region aimed at tackling these resource challenges and exploring innovative ways of organising and delivering social care services in rural areas and SPAs. It is based on a desk study funded by the Nordic Welfare Centre (see more about this on the last page).
