Resumo: Estudou-se a germinação e alguns aspectos da morfologia e fisiologia de Metarhizium anisopliae (METSCH.) SOROKIN, utilizando-se meios de cultura de diversas composições e também substâncias inibidoras de diversas naturezas. Observou-se a formação e o crescimento de tubos germinativos em conidios de linhagens selvagens, mutantes e diplóides. Verificou-se nos estudos iniciais que quando os conidios de M.anisopliae produzidos em meio mínimo, em culturas de superfície, eram expostos a ambiente ventilado, perdiam cerca de 50% de seu peso, pela evaporação da água em 6 horas. Verificou-se também que quando conídios produzidos de igual modo eram inoculados em meio completo líquido aumentavam seu volume em mais de 6 horas.Os estudos subsequentes mostraram que em 12 horas de incubação, mais de 85% de conídios germinaram nos vários meios testados, tanto sólidos como liquidas. Este tempo pode ser considerado como o tempo mínimo de germinação (TGm), para a variedade anisopliae. Os estudos também sugeriram que em meio liquido e à temperatura ambiente, uma concentração de 10_ conidios/ml, parece ser em algumas situações a mais indicada. Houve variabilidade quanto ao número de tubos germinativos em conídios de M.anisopliae num determinado tempo. A germinação e o crescimento de tubos germinativos podem ser representados por um modelo exponencial. Considerando também os vários meios de cultura testados, o meio mínimo + extrato de levedura se mostrou um meio basal para a germinação de conidios e crescimento de co1ônias. Foram ensaiadas várias substâncias inibidoras da germinação de conídios como, glicerol, polietilenoglicol, benzeno, clorofórmio, m-cresol, álcool éter + alquil-sulfato de sódio e cicloheximida, cujos efeitos permitiram em muitos casos um maior sincronismo na germinação. Algumas destas substancias além de osmoreguladoras, também produziram efeitos morfogênicos, aumentando o número de tubos germinativos dentro de um determinado tempo. Com m-cresol, o efeito morfogênico foi mais duradouro. Os mutantes da 1inhagem E9 não diferiram no modo de germinação quando comparados com a linhagem selvagem de origem, o mesmo não acontecendo com os mutantes de F84. A germinação de conídios dos diplóides, revelou cada um deles germinando de maneira diferente. O trabalho permitiu concluir que a germinação de conídios é um processo extremamente complexo, que segue um programa genético, o qual se expressa no tempo e no espaço e influenciado por diversos fatores ambientais
Abstract: Were studied on the germination of the Metarhizium anisopliae (METSCH) SOROKIN, and several aspects the morphology and physiology, utilizing culture media of various compositions and also inhibiting substances of various natures. The formation and growth of germinative tubes were observed in conidia of wild strains, mutant and diploid. The initial studies showed that when conidia the M.anisopliae produced in a minimum medium, in superficial cultures, were exposed to a ventilated environment they would loose 50% of their weight, by water evaporation in 6 hours. It was noticed also that when conidia produced in an equal manner were inoculated in a complete liquid medium they would increase their volume to more than 50% in 6 hours.The subsequent studies showed that in 12 hours of incubation more than 85% of the conidia germinated in the various tested media, such as for solids, as for liquids. This length can be considered as the minimum time for germination (TGm), for the anisopliae variety. The studies also suggested that in a liquid medium at the environmental temperature, a concentration of 10_ conidia/ml, it seems to be in some situations the most indicated. There was variability as to the number of germinative tubes in M.anisopliae conidia in a determined period of time. The germination and growth of germitiative tubes can be represented by an exponential model. Considering also the various tested culture media, the minimum + yeast extract demonstrated to be a basal medium for germination of conidia and growth of colonies. Various inhibiting substances of the conidia germination were tested, as glycerol, poliethileneglycol, benzene, chloroform, m-cresol, alcohol ether + alquil-sulphate of sodium and cicloheximide, being that the effects served in many cases to increase synchronism in germination. Some of these substances, besides being osmoregulators, also produced morphogenic effects, increasing the number of germinative tubes within a certain period of time. With m-cresol the morphogenic effect lasted longer.The mutants of strain E9 did not differ in the germination manner when compared to a wild origen, the same did not occur with the mutants F84. The germination of diploids conidia showed each of them germinating in a different manner. It was concluded by the study that the germination of the conidia is an extremely complex process, following a genetic program, which express in time and space and it is influenced by several environmental factors

Resumo: o objetivo do presente trabalho foi reunir duas linhagens diferentes de Metarhizium anisopliae, e através do ciclo parassexual obter diplóides e segregantes, os quais pudessem ser estudados quanto a produção de destruxlnas, utilizando-se decromatografta de camada delgada (CCD) e gasosa, sendo esta última acoplada ao espectrômetro de massa (GC-MS). Para isto foram utilizadas duas linhagens: uma de origem norte-americana (F-84) e a outra brasileira (E-9). Estas foram submetidas à ação da luz ultra-vloleta, de maneira a obter-se mutantes morfológicos e auxotróficos. Os mutantes obtidos foram: E-9 am-pir, F-84 vio-tia e F-84 vio-nic, os quais apresentaram-se bastante estáveis com uma frequência de reversão menor que 10-6 conídios. Testes de complementação de pigmentos dos mutantes foram conduzidos e pode-se verificar que os mesmos não produzem a enzima lacase, que tem ação na formação do pigmento selvagem, e assim sendo os diplóides não apresentaram pigmentação verde. Os diplódes foram formados dos cruzamentos de E-9 am-pir x F-84 vio-tia e E-9 ampir x F-84 vto-nic. Ambos apresentaram coloração de um dos parentais (amarelo); estes foram haploidizados com Cloroneb (150µg/ml), surgindo setores amarelos e violetas de formato irregular. Para as destruxinas, foram utilizados os extratos das linhagens selvagens, mutantes, diplóides e segregantes, que foram cromatografados em CCD, apresentando um Rf próximo ao padrão 0,50. Um deles demonstrou Rr abaixo do padrão, (O,44). Na espectrometria de massa, os íons formados determinaram a presença das destruxinas A e B dada pelas frações 576, 7-577,6 e 592,7-593,b respectivamente. Todos os extratos das linhagens analisadas produziram destruxinas, com excecão de dois segregantes S/B-1a e S/B-1b, em cujos extratos não encontradas as duas destruxinas B e A e B. respectivamente
Abstract: In this work the main object was the study of destruxlns production, by diploids and segregants obtained through parasexual cycle in Metarhizium anisopliae. The two parantal strains utilized were E-9 (Brazil) and F-84 (USA), which after induction with UV-radiation produced conidial color and auxotrophic mutants. The mutants E-9 am-pir, F-84 vio-tia and F-84 vio-nic were utilized because they showed a reversion frequency of auxotrophlc markers lower than 10-6 conidia. The diploids Isolated (E-9 am-pir x F-84 vio-tia and E-9 am-pir x F-84 vio-nic) were all yellow conidiating and showed yellow and violet irregular segregants on Chloroneb (150µg/ml) . As only yellow conidia diploids were isolated it can be suggested that they are not able to produce laccase, which Is the enzyme responsible for wild type (green) pigment formation. Additional tests for laccase production were made with color mutants growing together in a same Petri dish and tt could be deduced that leccase was absent. Destruxtins were analyzed by Thin-layer chromatography (TLC), showing Rf close to the pattern 0,50 and gas chromatography combined with mass spectrography. The mass spectrography showed the presence of destruxins A and B by lons formation in fractions 576,7-577,6 and 592,7-593,6 respectively. One of them showed Rf below to the pattern (0,44). Mass spectrography showed the presence of the destruxin A and B by fractions 576,7-577,& and 592,7-593,6 respectively. AI) the strains analysed produced destruxins, but the sectors S/B-la and S/B-lb, which extracts didn't have destruxlns B and A and B, respectively

Full textThe method used in the production of entomopathogenic fungi had few changes in more than 40 years of use in Brasil. The production of large-scale biofactories is basically through solid fermentation in rice with high labor and long production time. Thus, the present study consisted in developing strategies that could be implemented in the different stages of the production system of Metarhizium anisopliae ESALQ1037, as addition to rice of inert materials (sugarcane bagasse, shavings, vegetable sponge, rice straw) and other substrates (millet, rice flakes and sago) for separation of rice grains and increased aeration, use of blastospores as inoculum in biphasic systems having as a solid substrate the rice or mixture of bran (wheat, corn, oats, rice, soybean and cotton) or growth and sporulation supports (sugarcane bagasse, vermiculite, wool, rice straw and shavings). The use of boiling sterilization to replace autoclaving and recovery of conidia by water extraction as a substitute for sieving was tested, as well as storage under high and low temperature and humidity conditions: 97% RH and 22 ºC; 97% RH and 4°C; 0% RH and 22°C; 0% RH and 4°C. The M11 liquid medium (the base of 45g L-1 glucose and 36g L-1 yeast extract) was the most suitable for the production of blastospores (3.0 x 108 blastospores / mL) and biomass. The inoculation of blastospores in rice resulted in larger and faster production of conidia (4.51 x 108 conidia / g) in 7 days of fungal growth. The wheat bran inoculated with blastospores produced in M11 medium resulted in higher conidia production (1.2 x 109 conidia / g) when compared to the bran and rice inoculated with conidial suspension (maximum of 7.8 x 108 conidia / g). The dried autoclaved sugarcane bagasse was the carrier that resulted in higher fungus production when associated with the liquid medium. The addition of moistened sugarcane bagasse and non-moistened shavings and vegetable sponges promoted increase in the production of conidia (> 3.3 x 109 conidia / g) when compared to rice only (1.0 x 109 conidia / g). Exposure of the boil rice for 10 and 15 minutes resulted in conidial production similar to the production in autoclaved rice without the presence of contaminants. Wheat bran was the most promising substrate for the fungus production especially when mixed with rice bran (1.75 x 109 conidia / g) in solid fermentation. Conidia extracted from rice by sieving or washing in water and recovered with or without the addition of clays, diatomaceous earth and kaolin showed viability higher than 92% after storage for 60 days at 22 °C and 97% RH. At 22 °C and 0% RH, only the conidia extracted from the rice by sieving and those extracted in water and recovered with the addition of black clays and kaolin had viability higher than 92% after storage for 60 days. Strategies tested in this study for the production and extraction of conidia of M. anisopliae are viable alternatives to be incorporated in the large scale production and to reduce the time of fermentation and manpower, to increasing the efficiency of conversion of substrate in conidia.
