Intensity Modulated Radiation Therapy (IMRT) is a modern radiotherapy treatment technique used to obtain highly conformal dose distributions. The delivery of IMRT is commonly achieved through the use of a multileaf collimator (MLC). One of the hindrances at present to the widespread use of IMRT is the increased time required for its planning, delivery and verification. In this thesis one particular method of MLC based IMRT, known as Static Multileaf Collimator based IMRT (SMLC-IMRT), has been studied along with methods for improving it???s delivery efficiency. The properties of an MLC commonly used in SMLC-IMRT have been characterised. The potential ramifications of these properties on the dosimetric accuracy of the delivered IMRT field were also investigated. An Interactive Leaf Sequencing (ILS) program was developed that allowed for the manipulation and processing of intensity maps using a variety of methods. The objective of each method was to improve the delivery efficiency whilst maintaining the dosimetric quality of the IMRT treatment. The different methods investigated were collimator angle optimisation, filtration, and intensity level optimisation. The collimator was optimised by identifying the angle at which the minimum monitor unit???s (MU???s) were required when using a sliding-window delivery method. A Savitzky-Golay filter was applied to random intensity maps and suitable filtration parameters identified for filtering clinical IMRT fields, and the intensity levels were optimised based on a deviation threshold. The deviation threshold identified the acceptable level of difference tolerable between the original and modified intensity map. Several IMRT cases were investigated and the impact of each the methods on MU???s, segments and dose distribution observed. As the complexity of IMRT fields increases the dosimetric impact of the MLC properties increases. Complex SMLC-IMRT fields require longer delivery times due to the increased number of MU???s and segments. Collimator optimisation was shown to be a fast and effective means of improving delivery efficiency with negligible dosimetric change to the optimised plan. Modifying intensity maps by applying a filter and optimising the intensity levels did reduce the complexity and improve the delivery efficiency, but also required a dosimetric compromise of the optimised plan.

Previous issue date: 2004<br>Desde a sua concepção em 1994, o modelo SMLC, Sparkle Model for Lanthanide Complexes,
tem sido utilizado apenas para o cálculo das geometrias do estado fundamental de complexos de
Eu(III). Neste trabalho, aperfeiçoamos em muito este modelo para o íon Eu(III) e iniciamos o
procedimento de extensão do mesmo para os outros íons lantanídeos. Desenvolvemos também uma
versão do modelo para actinídeos, SMAC/AM1, em particular para o íon Th(IV).
No processo de parametrização destes modelos, dois pontos são de fundamental importância: (i)
a escolha de um conjunto de treinamento capaz de representar bem o nosso universo de estruturas de
complexos e (ii) a escolha de uma função resposta capaz de possibilitar a reprodução satisfatória das
propriedades de interesse, que no nosso caso foram comprimentos e ângulos de ligação envolvendo o
íon metálico.
Na primeira etapa, a escolha certa de um pequeno, mas representativo conjunto de treinamento
foi fundamental, pois no processo de parametrização estas estruturas foram calculadas milhares de
vezes até que os parâmetros capazes de reproduzir mais exatamente as estruturas cristalográficas
fossem encontrados. Por este motivo, realizamos uma análise de agrupamentos hierárquicos para
cada íon utilizando todas as estruturas disponíveis no CSD (Cambridge Structural Database 2003).
Esta análise resultou, em geral, na classificação das estruturas em sete grupos distintos onde o
critério de separação foi o tipo de ligante presente no complexo. Assim, foi possível, para todos os
íons parametrizados, escolher um conjunto de treinamento pequeno, mas capaz de representar
qualquer tipo de complexo contendo o íon em questão.
Na outra etapa, resolvemos adotar uma função resposta em que as propriedades consideradas, no
nosso caso distâncias e ângulos de ligação, estivessem intimamente relacionadas com os parâmetros
do íon que estávamos buscando. Esta nova função resposta é composta de duas parcelas: a primeira
delas é igual ao quadrado da diferença entre as distâncias interatômicas experimentais, Ri

Durante o procedimento de parametrização para o íon Eu(III), o primeiro dos lantanídeos a ser
parametrizado, inúmeros testes foram realizados e cada termo da função resposta foi exaustivamente
analisado até que conseguíssemos encontrar um conjunto de parâmetros que tornasse o nosso método realmente robusto. Com o mesmo procedimento utilizado na concepção dos parâmetros do íon
Eu(III), parametrizamos os íons Gd(III) e Tb(III) e estendemos a metodologia para actinídeos -
Th(IV).
Os modelos foram validados de duas formas distintas. A primeira delas foi através de conjuntos
testes contendo várias estruturas para cada íon. A outra forma de validação, foi a realização de
cálculos ab initio RHF/STO-3G/ECP para sete estruturas representativas de cada um dos grupos
obtidos pela análise de agrupamentos.
A análise dos resultados obtidos mostrou que para o íon Eu(III) esta nova versão resolveu vários
problemas detectados na versão anterior, SMLC II. Dentre os mais importantes estão: (i) a
capacidade de predizer, muito bem, estruturas com qualquer tipo de ligante, ao contrário do SMLC II
que só era capaz de predizer muito bem estruturas com ligantes do tipo -dicetonas; (ii) correção dos
ligantes nitratos, que nas versões anteriores coordenavam-se de forma monodentada ao íon Eu(III) e
que nesta nova versão passaram a coordenar-se corretamente (ligante bidentado); (iii) uma melhora
razoável na descrição de ligantes pequenos, como água ou isotiocianato.
O avanço do modelo SMLC/AM1 para o íon Eu(III) foi de tal forma significativo que nos
possibilitou compará-lo com resultados obtidos a partir de metodologias ab initio. O mesmo foi feito
para os outros íons lantanídeos e actnídeos estudados. Para o íon európio (III) o erro médio absoluto
obtido com o modelo SMLC/AM1 foi de 0,115 Å e a metodologia ab initio RHF/STO-3G/ECP
apresentou um erro médio absoluto de 0,123 Å, ou seja, o modelo SMLC/AM1 foi 6% mais exato.
Para os íons Gd(III) e Tb(III), o modelo SMLC/AM1 foi, respectivamente, 16% e 31% mais exato
que a metodologia ab initio utilizada. No caso do íon actinídeo Th(IV), o erro médio absoluto obtido
com o modelo SMAC/AM1 foi de 0,126 Å e a metodologia ab initio utilizada apresentou um erro de
0,113 Å. Neste caso, o modelo SMAC/AM1 ficou a apenas 10% da metodologia RHF/STO-3G/ECP.
Com base nos resultados obtidos com este trabalho, podemos então afirmar que os modelos
SMLC/AM1 e SMAC/AM1 apresentaram um nível de exatidão comparável ou, em alguns casos,
superior ao da metodologia ab initio, RHF/STO-3G/ECP, na predição, tanto das distâncias entre o
íon Ln(III) ou Ac(IV) e os átomos que compõem o poliedro de coordenação, como também entre os
átomos do poliedro

Apoptosis signal pathways converge on the Caspases, a family of proteases that form a self-amplifying cascade, the activation of which generates the characteristic morphological and biochemical features of apoptotic cell death. The Inhibitors of Apoptosis (IAPs) are a family of proteins that bind and inactivate Caspases at both the initiation and terminal effector stages of this cascade. Over-expression of IAPs, as observed in cancer cell lines and tumour biopsy samples, results in a cellular phenotype of resistance to a variety of apoptotic stresses including chemotherapeutic drugs and irradiation. One negative regulator of IAP function, Smac (second mitochondrial activator of Caspases a.k.a.Diablo) has been identified as a mitochondrial protein that is processed and released concomitantly with cytochrome c following an apoptotic stress. Proteolytic processing of the mitochondrial signal peptide sequence generates a novel amino terminus that interacts with XIAP and disrupts XIAP-meditated Caspase-9 inhibition. By disrupting the interaction of XIAP with Caspase-9, Smac is believed to promote apoptotis. Furthermore, the structure determinations of XIAP, in conjunction with either Caspases or Smac, have led to the identification of a critical pocket and groove on the surface of each BIR domain ( Baculovirus IAP Repeat). Within XIAP BIR3, the binding pocket interacts with Caspase-9 and can be disrupted by the presence of Smac, which competes for the same site.
In this thesis work, the utility of Smac as a chemo-sensitizing agent for the treatment of cancer was enhanced by three different strategies. The first approach involved the development and characterization of an ubiquitin-Smac fusion system that circumvents the targeting of Smac to the mitochondria and results in the expression of a fully mature, biologically active Smac protein. The second approach entailed developing a strategy for increasing the affinity of Smac binding to the IAPs. A random peptide phage display screen was used to identify novel, high affinity peptide ligands that bind to the IAP-BIR3 motifs. These peptide sequences were similar in structure to the amino terminus of Smac and Caspase-9. The data generated from these two approaches showed that both over-expressed, mature Smac and Smac-like peptide sequences bind to the IAPs in vitro and in vivo and sensitize cells to chemotherapeutic drugs. The third and final approach involved developing a Smac system that was a more potent apoptosis-inducer than wild-type Smac, which would be predicted to have greater therapeutic potential. It has been shown that Smac interactions with the IAP BIR domains may accelerate IAP auto-ubiquitination and destruction via the IAP carboxy terminal RING (Really Interesting New Gene) domain. Adding an IAP-RING domain to the carboxy terminus of Smac was predicted to enhance IAP degradation while maintaining the IAPs in a non-functional protein complex. Indeed, it was observed that generating a Smac protein with RING-conferred E3 ligase activity significantly decreased IAP levels, with a concomitant increase in the sensitivity to apoptosis in the presence of chemotherapeutic agents.

In all cellular systems, the transmission of bulk genetic information during proliferation occurs in the form of chromosomes. The segregation of these entities upon cell division is of pivotal importance for all forms of life. Structural maintenance of chromosomes (SMC)-kleisin complexes are ubiquitous and essential factors that ensure proper organisation and segregation of the genetic material. Aim of this work was to elucidate evolutionary conserved features in the architecture of SMC-kleisin complexes, and to probe these features for functional relevance. We find that two major architectural themes have been constrained by evolution: (I) SMC-kleisin complexes form asymmetric assemblies with a ring-like topology, whereby a kleisin monomer bridges two different binding sites on a SMC dimer, (II) SMC-kleisin complexes form rod-like structures, whereby the SMC proteins of a given dimer are closely juxtaposed in a well-defined manner. Based on these findings, we propose that SMC-kleisin complexes from all domains of life act by a unifying mechanism.<br>Die universellen Träger genetischer Information bei der Vermehrung zellulären Lebens sind die Chromosomen. Die Segregation dieser Einheiten während der Zellteilung ist für alle Organismen unabdingbar. Structural Maintenance of Chromosomes (SMC)-Kleisin-Komplexe sind universelle und essentielle Faktoren, welche die korrekte Organisation und Segregation des Erbmaterials sicherstellen. Ziel dieser Arbeit war es, evolutionär konservierte Erscheinungsmerkmale von SMC-Kleisin-Komplexen aufzudecken, und diese Merkmale auf funktionelle Relevanz zu testen. Wir haben zwei evolutionär invariante Leitmotive der Architektur von SMC-Kleisin-Komplexen identifiziert: (I) SMC-Kleisin-Komplexe haben eine asymmetrische Konfiguration, in der ein Kleisin-Monomer zwei unterschiedliche Bindungsstellen eines SMC-Dimers miteinander verbindet, (II) SMC-Kleisin-Komplexe bilden stäbchenförmige Strukturen, in denen die SMC-Proteine eines Dimers in einer wohl definierten Art eng aneinander liegen. Basierend auf diesen Resultaten schlagen wir vor, dass sämtliche SMC-Kleisin-Komplexe aus allen phylogenetischen Domänen einen gemeinsamen Funktionsmechanismus haben.

Second mitochondrial-derived activator of caspases (Smac), also known as direct IAP-binding protein with low pI (DIABLO) is known as a proapoptotic protein that inhibits the inhibitors of apoptosis family (IAPs). The members of the IAP family were characterized by their ability to inhibit proapoptotic caspases. More recently, the IAPs have also been shown to play major roles in regulating diverse signalling events within the cell. A physiological role for Smac remains elusive, but studies have focused primarily on its roles in regulating the IAPs in the context of apoptosis. In order to critically evaluate possible physiological roles for Smac, we sought to examine non-apoptotic signalling events regulated by the IAPs, including NF-κB induction, mitochondrial antiviral (MAVS) signalling, and necroptosis. While Smac knockdown revealed no obvious roles for Smac in context of MAVS signalling or NF-κB induction, we found that Smac contributes to necroptosis induction in L929 cells. While we hypothesized that Smac null mice would be protected from necroptosis induced by experimental colitis, we unexpectedly found that colitis was more severe in Smac null mice than in wild-type mice. Further investigation of how Smac is involved in the reaction to induced colitis might reveal a physiological role for Smac.<br>"Second mitochondrial-derived activator of caspases" (Smac), aussi connu sous le nom de DIABLO pour "direct IAP-binding protein with low pI", est une protéine proapoptotique qui effectue son rôle en interagissant avec et inhibant l'action des membres de la famille des inhibiteurs d'apoptose (IAP). Les membres de la famille des IAP ont été caractérisés par leur capacité d'inhiber l'action des caspases proapoptotiques prévenant ainsi l'apoptose cellulaire. Les IAP sont aussi engagés dans diverses autres voies de signalisation, hormis leur rôle dans la régulation de l'apoptose. L'effet de Smac sur les IAP a été étudié dans le contexte de l'apoptose cellulaire et aucun autre rôle physiologique ne lui a été attribué jusqu'à nos jours. Afin d'évaluer diverses possibilités de rôles physiologiques pour Smac, nous désirions examiner les voies de signalisation non-apoptotiques régulées par les IAP, incluant l'induction de NF-κB, la signalisation antivirale mitochondriale (MAVS) ainsi que la nécroptose.Nos résultats révèlent que la sous expression de Smac n'affecte pas l'induction de NF-κB et n'interfère pas avec la voie de signalisation engagée par les MAVS. Par contre Smac induit la mort des cellules L929 par nécroptose.Pour examiner l'effet de Smac in vivo nous avons étudié son rôle dans un modèle de colite expérimentale. Nos résultats révèlent que les souris mutantes rendues nulles pour le gène Smac présentent une colite plus sévère que les souris à phénotype sauvage. Il serait ainsi nécessaire d'examiner de plus près la régulation de Smac dans le modèle de colite expérimentale pour essayer de mieux comprendre son rôle physiologique.

True anophthalmia is the most severe congenital eye malformation. With absence of the eye, optic nerve, chiasm and optic tracts. Identifying the genes that cause genetic true anophthalmia should improve our understanding of the critical processes required for development of the eye. Recessive loss-of-function mutations in SMOC1 have been identified as the cause of Ophthalmo-acromelic syndrome (OAS), a multisystem disorder which has true anophthalmia as a prominent feature with characteristic limb and facial malformations. In order to establish the function of SMOC1 in development I used the zebrafish as a model organism to support a link between SMOC-1 and BMP signalling. As a first step I characterised the genomic structure of zebrafish smoc1 gene. I was able to correct an error in the zebrafish genome (Zv8) that annotated zsmoc1 as two fragmented and rearranged orthologous loci. However, using RTPCR I could show that there is in fact a single intact zsmoc1 transcript. In addition, I was able to identify an un-annotated 5’ coding exon using 5' RACE which showed that the full open reading frame includes a signalling peptide. RT-PCR was also used to identify several novel zsmoc1 splice isoforms. To explore the link between zsmoc1 and bmp signalling I used injection of antisense morpholino oligonucleotide and capped mRNA to examine the effects of loss-of-function and overexpression respectively of smoc1 and genes functioning in the bmp signalling pathway. The resulting embryos were analysed using morphometric analysis (Kishimoto scale), a quantitative assay of dorsalisation/ventralisation and live imaging of reporter transgenic fish. I developed a quantitative RT-PCR assay for expression of dorsal (otx2 and runx3) and ventral (eve1 and gata2) marker genes. I established a reliable system for live imaging of zebrafish development between 8 hpf and 24 hpf. By combining this system with fluorescent transgenic reporters marking the eye field (rx3:gfp reporter) and BMP-signaling (BRE:gfp reporter) I was able to accurately quantitate the effect of smoc1 depletion on eye size and SMAD1/5/8 signalling in the eye. These results support the predictions from the Drosophila homologue Pent that zsmoc1 functions as an antagonist of bmp signalling. Finally, I describe my attempt to produce a zebrafish model for OAS using genome editing technology. I designed, produced and validated transcription activator like effectors nucleases (TALENs) targeted to the zsmoc1 open reading frame using the Voytas Goldengate method. I designed and optimised a novel strategy to demonstrate targeted cutting activity for in vitro validation. Following injections of the in vitro validated TALEN into zebrafish embryos I used Ion Torrent sequencing to assess the in vivo activity of the engineered TALEN pairs. Unfortunately these TALENs were not able to cut the targeted locus in vivo.

The thesis deal with a model of electronically controlled assembly minicell MAP-205 based on pneumatic drives and its debugging. Assembly minicell simulates assembling and disassembling of simple four item assembly. Minicell is controlled by Phoenix Contact ILC 150ETH programmable logic controller. The minicell control program is composed of several subroutines/subprograms providing various functionalities to assembly minicell. There is integrated HMI created in Control Web application too. It allows to display actual status of each component and to control each drive in manual mode. Communication between HMI based on PC and PLC controller is supported by Bluetooth module and OPC server.

Mestrado em Bioquímica - Bioquímica Clínica<br>Pulmonary hypertension (PH) is a multifactorial, progressive disease, characterized by vasoconstriction, pulmonary vascular remodelling and thrombosis in situ, which lead to augmentation of pulmonary arterial pressure and pulmonary vascular resistance, culminating in right heart failure and ultimately in dead. Pulmonary vascular remodelling, which is one of the major contributors to the development and progression of PH, is characterized by excessive cellular proliferation and reduced apoptosis. However, little is known
about the molecular mechanisms underlying this imbalance. So, the present work aimed to study the contribution of apoptosis to the pathogenesis of PAH, through the analysis of cardiac and pulmonary expression of survivin and Smac/DIABLO along hemodynamic and morphometric alterations in an animal model of monocrotaline (MCT)-induced PAH. Results showed that cardiomyocytes were hypertrophied 7 days after MCT injection, preceeding hemodynamic alterations which were only present at day 21. Seven days after MCT administration, survivin overexpression was notorious in the ventricles and progressively increased throughout the development of MCT-induced PAH.
On the other hand, although increased 7 days after MCT injection, lung survivin expression progressively decreased between the two days. The upregulation of Smac/DIABLO observed in lung, right and left ventricles 7 days after MCT administration, progressively decreased with the disease progression. In conclusion, data suggest that a deregulation in the balance between survivin and smac/DIABLO might be related with pulmonary vascular remodelling and cardiomyocytes hypertrophy in response to apoptotic stimuli.<br>A hipertensão pulmonar (HP), é uma doença multifactorial e progressiva, caracterizada pela vasoconstrição, remodelação vascular pulmonar e trombose in situ, com consequente aumento da pressão arterial pulmonar e da resistência vascular pulmonar, frequentemente culminando na insuficiência cardíaca direita e morte. A remodelação vascular pulmonar, um dos principais contribuintes para o desenvolvimento e progressão da HP, é caracterizada por uma proliferação celular excessiva e uma reduzida apoptose. No entanto, pouco se sabe sobre os mecanismos moleculares subjacentes a este desequilíbrio. Assim, o presente trabalho teve como objetivo principal a avaliação da contribuição da apoptose na patogénese da HAP, através da análise da expressão cardíaca e pulmonar da survivina e do Smac/DIABLO e a
sua associação às alterações hemodinâmicas e morfométricas. Para o efeito utilizou-se um modelo animal de HAP induzida por administração de monocrotalina (MCT). Os resultados demonstraram que os cardiomiócitos se encontravam hipertrofiados 7 dias após a injeção de MCT o que precedeu as alterações hemodinâmicas que se verificaram apenas ao dia 21. Sete dias após a administração de MCT observou-se sobreexpressão de survivina nos ventrículos, tendo esta aumentado progressivamente com a doença. A nível pulmonar, embora sobreexpressa ao dia 7, a expressão de survivin diminuiu com o desenvolvimento da doença. Apesar de sobreexpressa nos ventrículos e no pulmão 7 dias após injeção de MCT, a expressão do Smac/DIABLO diminuiu progressivamente com a HAP. Em conclusão, os resultados sugerem que a desregulação das vias de sinalização nas quais intervêm a survivina e o Smac/DIABLO está relacionada com a remodelação vascular pulmonar e a hipertrofia dos cardiomiócitos em resposta a estímulos apoptóticos.

It is a highly structured process that removes the cell from the body without damage to surrounding cells. The mitochondria has been shown to play a key role in apoptosis induced by a wide range of stimuli and can trigger the apoptotic pathway by release of apoptogenic factors. One of these factors is cytochrome c, a protein that normally functions in the electron transport chain but upon release it binds to the first mammalian homologue of CED-4, Apoptotic Protease Activating Factor 1 (APAF1) and induces the formation of a large caspase-activating complex. Another protein released from the mitochondria is Smac, a pro-apoptotic protein that functions by antagonising Inhibitor of Apoptosis Protein function. The data presented in this thesis demonstrates the existence of multiple splice forms of both APAF1 and Smac and investigated their roles in the apoptotic pathway. APAF1 forms the large molecular weight complex known as the apoptosome in 293 cells although epitope tagged APAF1 formed an incorrectly formed this complex in the absence of additional stimuli. APAF1 was also found to be cleaved by Caspase-3 during apoptosis and that the resulting 30 kDa fragment is associated with the inactive complex. Data also indicates that caspase-9 recruitment to the apoptosome occurs via interactions between both the CARD and the CED-4 homologous regions of APAF1. The pro-apoptotic protein Smac exists as several forms due to alternative slicing and one of these forms, Smac , is pro-apoptotic even though it lacks the N-terminal Inhibitor of Apoptosis protein binding domain. Smac is localised to the cell cortex and does not redistribute upon induction of apoptosis. These data demonstrate that Smac is able to induce apoptosis independently of Inhibitor of Apoptosis Protein binding. A polyclonal antibody to Smac / Smac was also developed and characterised.

Functional programming languages, such as SML (Standard Meta Language), Haskell and Lisp, focus on rules and matching of patterns, in contrast to procedural languages in which programs are written as a sequence of instructions. Programs in functional languages may have errors due to the misunderstanding of their properties. Testing is one of the essential activities to identify these errors and to guarantee the quality of the product under development. However, there are few initiatives and tools to support the testing of functional programs. Moreover, an important issue that is often not taken into consideration in this context is to provide a means to quantify the test activity. In this work, we establish mechanisms to investigate the aplicability of Mutation Testing for testing functional programs, written in SML. Mutation Testing is a test criterion that allows to evaluate the quality of a test set and to guide the generation of test sets. The existence of a tool to support this criterion is essential due to the large amount of information related to its application. The web tool PROTEUM/SML, developed with the aim of applying the Mutation Testing to SML, implements the mutation operators defined in this work. An example is provided to illustrate the concepts and PROTEUM/SML tool.<br>Ao contrário das linguagens procedimentais, em que os programas são escritos como uma sequência de instruções, as linguagens de programação funcional, tais como SML (Standard Meta Language), Haskell e Lisp, enfatizam regras e casamento de padrões. Os programas em linguagens funcionais podem conter erros pela falta de entendimento de suas propriedades. Assim, a atividade de teste é de grande importância na identificação desses erros, podendo fornecer evidências da qualidade do produto em teste. No entanto, existem poucas iniciativas para o teste de programas funcionais bem como no desenvolvimento de ferramentas de apoio a essa atividade. Além disso, os trabalhos existentes não possuem uma medida de cobertura da atividade de teste. Assim, motiva-se a investigação de novas formas de se realizarem testes em programas funcionais. Um critério de teste que fornece uma maneira de auxiliar na geração e na avaliação de um conjunto de casos de teste é o Teste de Mutação. Neste trabalho, estabelecem-se subsídios para a investigação da aplicabilidade do Teste de Mutação para o teste programas funcionais, escritos em SML. Devido ao grande volume de informações que estão envolvidas na aplicação do Teste de Mutação, é essencial a existência de ferramentas de apoio para o uso desse critério. A fim de viabilizar a aplicação do Teste de Mutação para SML, foi desenvolvida a ferramenta web PROTEUM/SML, que implementa os operadores de mutação definidos neste trabalho. With less than 2 hours of data collected whenpersonally driving the car, I show that yes, it is indeed possible. This is done by applying thetechniques of supervised machine learning combined in conjunction with a deep convolutional neuralnetwork. This were applied through software developed to interact between the network and the agentinside the virtual environment. I believe the dataset could have been cut down to about 10 percentof the size and still achieve the research goal. This shows not just the possibility of teaching aneural network a good policy in stochastic environments with supervised machine learning, but alsothat it can draw accurate (enough) conclusions to imitate human behavior when driving a car.

SMAC est adapté aux problèmes de conception, problèmes qui se caractérisent par la construction d'une solution. L'accent est alors mis sur la démarche permettant à l'expert d'aboutir à une solution. Les connaissances sont memorisées à l'aide d'une représentation intermédiaire, basée sur un hypermedia, permettant ainsi de définir une interface homme/machine conviviale. La définition de la représentation des connaissances s'appuie sur un modèle de l'expertise ainsi que sur un modèle du mode de raisonnement de l'expert. Ce dernier est affiné par la prise en compte d'études théoriques et expérimentales du processus de conception. Les connaissances statiques, ou connaissances du domaine, sont memorisées sous forme d'objets ; quant aux connaissances dynamiques, elles sont enregistrées sous forme de problèmes avec des situations associées. Of all of the cancer patients throughout the world, approximately 50% of them are affected to some degree by cachexia. This syndrome involves significant skeletal muscle wasting, loss of adipose tissue and overall decrease in body weight in patients, particularly those with lung, pancreatic and gastric cancers. Cancer-induced cachexia is characterized by the presence of increased cytokines, notably TNF-α, IL-1β and IL-6. Most patients suffering of cancer-induced cachexia experience increased toxicity in response to chemotherapy, leading to fewer rounds of treatment and thus impeding the patients’ chances for recovery. More research into effective treatments for cancer-induced cachexia would therefore be indispensable.
The inhibitor of apoptosis proteins (IAPs) have emerged as important cancer targets, primarily because of their roles as caspase inhibitors and regulators of NF-κB signalling. Small molecule IAP antagonists known as Smac mimetic compounds (SMCs) are currently in stage I/II clinical trials. They function by targeting cIAP1 and cIAP2 (and to a lesser extent, XIAP) resulting in a cytokine mediated death response in cancer cells. SMCs induce the production of TNF-α, a cytokine with which SMCs can potently synergize. However, limited efficacy occurs in some cancer cell lines (presumably because TNF-α cannot be induced in an autocrine fashion) and an exogenous source of the cytokine, such as that induced by using an oncolytic virus, is required. Notably, TNF-α (initially known as “cachectin”) is known to play a significant role in the induction of skeletal muscle atrophy. We therefore wanted to examine the effects of TNF-α induction by SMC and oncolytic virus co-treatment on both tumour regression and skeletal muscle in tumour bearing mice.
We investigated the effects of SMC treatment on Lewis Lung Carcinoma (LLC) and B16F10 melanoma cell lines, both of which have been shown to be established cachectic cancer cell lines. Our in-vitro analysis of LLC and B16F10 cells revealed that LLC cells are sensitive to SMC and TNF-α co-treatment whereas B16F10 cancer cells remain resistant. SMC treatment, in combination with an oncolytic virus, VSVΔ51, increased tumour regression and survival time in LLC tumour bearing mice. Based on findings from previous studies, we investigated the role of cellular FLICE-like inhibitory protein (c-FLIP) in the resistance of the B16F10 melanoma cell line to SMC treatment. We were able to determine that the down-regulation of c-FLIP sensitizes the B16F10 cells to SMC and TNF-α induced cell death.
In extending these findings, we found that SMC treatment alone can cause skeletal muscle wasting in the tibialis anterior muscle of LLC tumour bearing mice. However, the atrophic response was observed to be minimal as documented by a slight but significant decrease (approximately 10%) in muscle fibre cross-sectional area. Moreover, no biochemical evidence of muscle atrophy, as visualized by changes in the expression of myosin heavy chain (MHC) and Muscle RING Finger protein 1 (MuRF1), was found. Regardless, we speculate that the impact of SMC treatment on muscle wasting would be transient and reversible, and propose that the benefits of such a combination immunotherapy would greatly outweigh the risks.

MUC4 is a heterodimeric mucin glycoprotein expressed in the epithelia of tissues. Previous studies in our laboratory demonstrated that MUC4 protein expression is regulated by exogenous growth factors and that MUC4 is found in complex with the receptor tyrosine kinase ErbB2. MUC4 protein expression in airway epithelia was evaluated using molecular biology techniques. The impact of the protein on ErbB2 activation was evaluated post mechanical wounding of airway epithelia, and upon MUC4 RNA silencing. MUC4 levels were increased with exposure to the differentiating agent retinoic acid and decreased upon exposure to epidermal growth factor, a proliferative agent. In the absence of MUC4, ErbB2 phosphorylation was diminished. These results support the hypothesis that MUC4 expression is enhanced during differentiation of epithelia. Furthermore these findings provide evidence for an additional level of ErbB regulation in airway injury and subsequent epithelial wound healing.

Before presenting the Sheet Moulding Compound (SMC) process, which is the primarily focus of this work, a literature survey is carried out to deal with fibre reinforced polymer composites in general. Then the first part of this work is presented and is primarily focused on experimental visualisation of the flow during mould closure of SMC. Circular plates are manufactured with industry scale equipment at close to production conditions. Special attention is given to the advancing flow front, for which the full complexity is captured by means of continuous high resolution close-up monitoring. From the experimental visualisation of the flow front, three phases are defined, namely squish, flow, and boiling. During the initial phase, squish, outer layers do not remain outer layers, the actual flow is very complex and air is likely to be entrapped. The governing process parameters during this phase are mould temperature, mould closing speed and amount of preheating in the mould. During the second phase, flow, the flow is stable and seemingly viscous. During the last phase, boiling, bubbles are observed in the low pressure region at the flow front, favouring the void content both internally and on the surface. Based on a chemical analysis including mass spectrometry and thermogravimetry, the gas is probably styrene. In the second part it is investigated if an inverse modelling approach by proportional regularisation can be applied to mimic the pressure distribution during compression moulding of SMC. The process is simulated with Computational Fluid Dynamics and the mastered parameter, the viscosity of the SMC, is allowed to vary as a function of time. A grid refinement study of two ways to model the process and for three fictitious pressure scenarios yields that the suggested approach work very well and that the numerical errors can be minimised as desired. Finally a validation process is carried out showing that to get quantitative agreements of the whole pressure field more advanced viscosity models must be used. In order to verify the inverse modelling system have to important errors are studied. Firstly the error between calculated and experimental pressure, secondly the discretisation error due to solving the problem for many small volumes. Both have to be minimized and the later is studied with Richardson's extrapolation. SMAC mimetic compounds (SMCs) are small molecule antagonists of the Inhibitor of Apoptosis (IAP) family of proteins. Binding of SMCs to the IAPs results in the sensitization of cancer cells to apoptosis in the presence of death ligands, such as tumour necrosis factor alpha (TNFα). I hypothesize that type I interferon (IFN) stimulation in cancer cells and in immune cells leads to the production of TNFα, which can then synergize with SMCs to kill cancer cells. The combined treatment of SMC and IFNα induces tumour regression in mice, and this effect is completely abrogated upon treatment with TNFα-neutralizing antibody. The synergistic effects are mediated by tumour cells and by contribution of immune cells, particularly macrophages and dendritic cells, as the systemic depletion of phagocytic innate immune cells results in an increase in tumour volume following combination treatment. The characterization of immune cell contribution will aid in the translation of the SMC combination therapy into clinical applications for the treatment of cancer.

La morphogénèse des ovocoques, dont fait partie le pathogène humain Streptococcus pneumoniae, implique des processus d’élongation et de division associés à la synthèse de la paroi bactérienne. Le composant majeur de cette paroi est le peptidoglycane, un polymère de sucre réticulé par des chaines peptidiques, qui confère la forme de la bactérie et est essentiel à sa survie. La synthèse de peptidoglycane nécessaire à l’élongation et la division bactérienne est effectuée par des complexes protéiques appelés respectivement « élongasome » et « divisome ». Les mécanismes d’assemblage et l’activité de ces complexes dans la cellule bactérienne restent encore non élucidés. Pour imager l’activité des complexes de synthèse du peptidoglycane in vivo à l’échelle du nanomètre, j’ai développé une méthode faisant appel à des dérivés de D-amino acides, à la chimie click et à la microscopie de localisation de molécules uniques (dSTORM ou direct Stochastic reconstruction microscopy). Cette méthode a permis d’obtenir des images à une résolution d’environ 20 nm, révélant des aspects inattendus de la synthèse du peptidoglycane et remettant en question le rôle de certaines protéines dans la morphogenèse du pneumocoque. En combinant ces observations avec les données de la littérature, un modèle simplifié de la morphogénèse des ovocoques est proposé<br>The morphogenesis of ovovcocci, which include the human pathogen Streptococcus pneumoniae, involves elongation and division processes associated with cell wall synthesis. The main component of the cell wall is the peptidoglycan, a polymer made of glycan chains cross-linked by peptide chains, which confers the bacterial shape and is essential for cell survival. Peptidoglycan synthesis required for cell elongation and division is performed by large protein complexes called “elongasome” and “divisome”, respectively. The assembly mechanisms and activity of these complexes in the bacterial cell remain mysterious. To image the activity of the peptidoglycan synthesis complexes in vivo at the nanoscale, I developed a method combining D-amino acid derivatives, click chemistry and single-molecule localization microscopy (dSTORM or direct Stochastic reconstruction microscopy). This method allowed obtaining images at a resolution of about 20 nm resolution, revealing unexpected features of peptidoglycan synthesis and challenging the role of some proteins in pneumococcus morphogenesis. Small molecular Inhibitor of Apoptosis (IAP) antagonists, known as Smac mimetic compounds (SMCs), are a novel class of anti-cancer drugs currently undergoing clinical trials. SMCs were designed to mimic the function of the pro-apoptotic protein, Smac, which directly depletes cells of cIAP1 and cIAP2, and consequently renders tumour cells sensitive to death in the presence of proinflammatory ligands such as TNFα. The Korneluk lab recently reported that SMCs synergize with the attenuated oncolytic virus Vesicular stomatitis virus (VSVΔ51) by eliciting an enhanced immune response in mice, such that the combined therapy is vastly superior to stand-alone therapies. To improve on this SMC-mediated synergistic response, I generated variants of TNFα-armed VSVΔ51. Due to high ectopic expression of TNFα in infected cells, a five times lower viral dose of TNFα-armed VSVΔ51 combined with SMC treatment was sufficient to improve the survival rate as compared to SMC and VSVΔ51 co-therapy. This improved synergistic response is attributed to a bystander effect whereby the spread of TNFα from infected cells leads to the death of neighbouring, uninfected cells in the presence of a SMC. In addition, the double treatment induced vasculature collapse in solid tumours, revealing another mechanism by which cytokine-armed VSVΔ51 in combination with a SMC can induce cancer cell death. This approach demonstrates great potential for engineered oncolytic virus and SMCs as a new combination immunotherapy for cancer treatment.

This thesis presents the results of a combination of new observations with the Parkes telescope of neutral hydrogen (HI) in the Small Magellanic Cloud (SMC) with an Australia Telescope Compact Array aperture synthesis mosaic. The data are used to study the HI distribution and mass, the velocity field and rotation curve of the SMC, as well as to probe the 3-D structure of the SMC. A kinematic study of the HI data reveals the existence of three supergiant shells which were previously undetectable in the ATCA data alone. The HI spatial power spectrum has been investigated over a range of contiguous scale sizes wider than those previously achieved in any other galaxy. This thesis also demonstrates that the infrared data obtained with the Infrared Astronomical Satellite for the SMC can be successfully reconstructed with much higher resolution using the Pyramid Maximum Entropy algorithm. The new infrared (IR) data are used to study the integrated IR spectrum, the dust temperature and dust column density in the SMC. The high resolution HI and IR data enable an investigation of the spatial correlation of dust and gas and the assumption of the dust and gas being well-mixed in the ISM. The spatial power spectrum of the dust column density shows that, as with the HI power spectrum, there is no preferred scale size for dust clouds. The remarkable similarity of the spatial power spectra for the HI and dust column density distributions suggests a unique hierarchical structure organisation for the ISM in the SMC. Such an organisation is likely to be governed by the Kolmogorov type turbulence and could be described by fractal nature with the volume fractal dimension of 2.4.<br>Doctor of Philosophy (PhD)

I materiali compositi più diffusi sono quelli a matrice polimerica (PMC, Polymer Matrix Composites) con fibre di rinforzo, largamente utilizzati per la loro capacità di conciliare ottima resistenza meccanica con elevata leggerezza.
Nel presente elaborato di tesi sono state studiate le caratteristiche meccaniche di materiali compositi a matrice resinosa, epossidica, rinforzati con fibre di carbonio chopped, ovvero fibre spezzate e disposte in modo del tutto casuale all’interno della matrice, mediante analisi microstrutturale e prove di trazione.
Viene descritto il processo di produzione delle piastre di materiale composito ottenuto per SMC (Sheet Moulding Compound) da cui sono stati ricavati i provini.
Lo studio a livello microstrutturale è stato possibile grazie all’inglobamento nella resina di alcune sezioni dei provini, le cui superfici sono state esaminate al microscopio acquisendo una quantità di immagini tale da permettere la ricostruzione della superficie stessa tramite software ed il calcolo percentuale delle porosità tramite SolidWorks.
La caratterizzazione meccanica è stata eseguita utilizzando la macchina per le prove di trazione presente nell’hangar della sede di Forlì della Scuola di Ingegneria e Architettura dell’Università di Bologna: la preparazione dei provini è basata sull’applicazione di tabs di alluminio.
I provini in materiale composito sono stati forniti in quattro differenti tipologie riguardanti la pressione a cui sono stati prodotti: 25, 50, 100 e 150 bar.
Lo scopo dell’elaborato è stabilire la pressione ottimale di produzione dei provini, a cui il materiale composito mostra le migliori proprietà meccaniche, in particolare la più alta resistenza a carico di trazione.
Le prove sono state condotte su provini a tre diverse lunghezze, per diversificare le modalità di stress meccanico.
I risultati sono stati poi analizzati per stabilire quale valore di pressione di processo conferisce le migliori caratteristiche meccaniche al materiale.

Software testing is an important process for ensuring the quality of the software. As the complexity of the software increases, traditional means of manual testing becomes increasingly more complex and time consuming. In most embedded systems, designing software with as few errors as possible is often critical. Resource usage is also of concern for proper behavior because of the very nature of embedded systems. To design reliable and energy-efficient systems, methods are needed to detect hot points of consumption and correct them prior to deployment. To reduce testing effort, Model-based testing can be used which is one testing method that allows for automatic testing of model based systems. Model-based testing has not been investigated extensively for revealing resource usage anomalies in embedded systems. UPPAAL SMC is a statistical model checking tool which can be used to model the system’s resource usage. Currently UPPAAL SMC lacks the support for performing automatic test generation and test selection. In this thesis we provide this support with a framework for automatic test generation and test selection using mutation analysis, a method for minimizing the generated test suite while maximizing the fault coverage and a tool implementing the framework on top of the UPPAAL SMC tool. The thesis also evaluates the framework on a Brake by Wire industrial system. Our results show that we could for a Brake-by-wire system, simulated on a consumer processor with five mutants, in best case find a test case that achieved 100% mutation score within one minute and confidently identify at least one test case that achieved full mutation score within five minutes. The evaluation shows that this framework is applicable and relatively efficient on an industrial system for reducing continues resource usage target testing effort.

Neuroblastoma is the third most common pediatric cancer. Dinutuximab is a recently approved monoclonal antibody targeting GD2, a ganglioside ubiquitously present on neuroblastoma. Recent studies have shown that αGD2 therapy activates PD1-PDL1 signalling, resulting in the inhibition of its full therapeutic potential. The PD1-PDL1 signalling axis is a cellular checkpoint that inhibits immune responses. The blocking of this interaction has been successful in the treatment of numerous cancers, including in combination with anti-GD2 therapy. The Inhibitor of apoptosis (IAP) proteins are commonly upregulated in cancers and prevent cell death through the inhibition of caspases and through the control of NF-κB activity. Smac mimetic compounds (SMCs) have been designed to target IAP activity, thereby promoting cancer cell death. Here, I used the SMC, LCL161, to improve αGD2 antibody treatment against a GD2+ syngeneic neuroblastoma mouse model. I found that murine cell lines NXS2 and N2a were resistant in vitro to LCL161-mediated apoptosis, despite expressing apoptotic components often silenced in neuroblastoma. In vivo, I observed a slight delay in tumour growth induced by LCL161 and I confirmed an in vivo anti-angiogenic effect of LCL161 through ultrasound imaging and necropsy evaluation. I then combined LCL161 and αGD2 antibody (clone ME361-S2a) treatment and reported a delay in NXS2 subcutaneous tumour growth, which was further potentiated with the addition of an αPD-L1 antibody. With optimization, there is potential for SMCs to be used in combination with αGD2 therapy in GD2+ cancers like neuroblastoma.

Inhalatory anesthetics affect synaptic transmission, but little is known about their actions in the central nervous system.
Dopamine is a catecholaminergic neurotransmitter that has important functions in the central nervous system and it is possible that its release can be modulated by inhalatory anesthetics. The study of inhalatory anesthetics effects in the neurotransmitter release could provide further information about the mechanisms that contribute with the actions of these agents during anesthesia. In this study, cortical slices of rat brain were labeled with [3H] dopamine to observe the effects of sevoflurane and halothane in the release of this neurotransmitter. Dopamine release was significantly increased in the presence of sevoflurane (0,46 mM) and halothane (0,048 mM) were incubed. This effect was independent of extracellular or intracellular calcium, and it was not affected by TTX (blocker of voltage dependent sodium channels) or reserpine (an blocker of vesicular monoamine transporter). These data suggest that dopamine release induced by anesthetics is independent of the exocytotic process and that this release would be mediated by the dopamine transporter. Experiments using low temperature or by decreasing sodium concentration in the incubation media indicated that dopamine release induced by anesthetics was considerably reduced. The same effect was also observed when dopamine and noradrenaline transporter inhibitors were used (GBR12909 and nisoxetine, respectively). Ouabain, Na+/K+ ATPase pump inhibitor, is known for stimulating dopamine release through the membrane transporter. In its presence, the evoked response by anesthetics was decreased. Nevertheless, further experiments demonstrated that halothane and sevoflurane did not inhibit the Na+/K+ ATPase pump. In conclusion, our study strongly suggests that halothane and sevoflurane increase dopamine release in cortical slices of rat brain and that this release is mediated by the dopamine transporter present in the plasma membrane.<br>Os anestésicos inalatórios parecem afetar a transmissão sináptica, mas suas ações no sistema nervoso central permanecem obscuras. A dopamina é um neurotransmissor catecolaminérgico que exerce importantes funções no sistema nervoso central e, assim , é possível que a sua liberação possa ser modulada pelos anestésicos inalatórios. Dessa forma, a investigação dos efeitos dos anestésicos na liberação deste neurotransmissor pode fornecer informações adicionais em relação aos mecanismos que contribuem com as ações destes agentes durante a anestesia. No presente estudo, fatias de córtex cerebral de ratos foram marcadas com [3H]dopamina objetivando-se estudar o efeito dos anestésicos inalatórios, sevoflurano e halotano, na liberação deste neurotransmissor. O sevoflurano (0,46 mM) e o halotano (0,048 mM) aumentaram significativamente a liberação de [3H]dopamina em fatias de córtex cerebral de ratos. Esse efeito foi independente do cálcio extra e intracelular, além de não ter sido afetado pela presença da TTX (bloqueador dos canais de sódio sensíveis a voltagem) e reserpina (inibidor do transportador vesicular das monoaminas). Esses dados sugerem que a liberação de dopamina induzida pelos anestésicos é independente dos processo exocitótico e que, possivelmente, essa liberação pode ser via transportador de dopamina. Experimentos realizados em baixa temperatura ou com diminuição do íon sódio no meio de incubação indicaram que a liberação de dopamina induzida pelos anestésicos foi significativamente reduzida. Esse efeito também foi observado quando utilizou-se inibidores do transportador de dopamina e noradrenalina (GBR12909 e nisoxetina, respectivamente). A ouabaína, um inibidor da bomba Na+/K+ ATPase, é conhecida por estimular a liberação de dopamina via transportador de membrana. Na presença desta drogra a resposta evocada pelos anestésicos foi reduzida. No entanto, experimentos adicionais mostraram que os anestésico halotano e sevoflurano não inibem a bomba de Na+/K+ ATPase. Em conclusão, este estudo indica que o sevoflurano e o halotano aumentam a liberação de dopamina em fatias de córtex cerebral de ratos principalmente a partir de transportadores de membrana desse neurotransmissor.

Tourette syndrome (TS) is a neuropsichyatric disorder characterized by tics and familiar inheritance. Several research groups are investigating the genetic aspects of TS, however, frequently the findings are not replicated in subsequent studies. Study of gene Slit and trk-like family member 1 (SLITRK1) has shown correlation with TS in three non-related patients. The family of SLIT proteins and their receptors, the robo family, are involved in axonal guidance in the crossing over at the spinal cord. The objective of our study was to further elucidate the putative role of Slit-Robo pathway in TS. The role of SLITRK1 gene in TS remains uncertain, since only 0.6% of patients in only one study were presented with changes in this gene. We have chosen genes that are related to the Slit-Robo pathway and present in regions pointed as suspicious in linkage association studies. Among the genes selected for our study were SLIT2, SLITRK1, ROBO3 and ROBO4. Our study is based in families with one or more affected individuals. All patients were evaluated by an experienced neuropsychiatrist. To replicate Abelson et al. (2005) findings, we used DHPLC to investigate the codifing region of SLITRK1 and subsequent sequencing of altered samples. Other genes were studied with tagSNPs. All data was analized with common statistical programs, as TRANSMIT and TDT-UNPHASED. We have found mutation in the SLITRK1 gene in one TS patient. We failed to find association among ROBO3 and ROBO4 genes and TS. Our study found association among SLIT2 gene polimorfisms and TS. In conclusion, some Slit-robo pathway genes are associated with TS, reinforcing the importance of proteins and genes involved in the neurodevelopment process for future investigation<br>A Síndrome de Gilles de la Tourette (ST) é uma entidade neuropsiquiátrica caracterizada pela presença de tiques e com importante componente hereditário. Muitos grupos vêm estudando os aspectos genéticos da ST, mas frequentemente os dados não se sustentam em estudos posteriores. Recentemente, foi descrita a presença de mutação do gene Slit and Trk-like family member 1 (SLITRK1) como gene correlacionado com a ST em três pacientes não-relacionados. A família das proteínas SLIT e seus receptores, os ROBO, aparentemente estão envolvidos no direcionamento axonal em relação ao cruzamento da linha média na medula vertebral. O objetivo do nosso estudo foi esclarecer se a via slit/robo está geneticamente associada à Síndrome de Tourette. Considerando que a participação nessa doença ainda é incerta, visto que apenas 0,6% dos pacientes de um único estudo apresentam alterações no gene SLITRK1, inicialmente procuramos estabelecer o papel do gene da SLITRK1 na nossa amostra de crianças com diagnóstico de Síndrome de Tourette, e posteriormente selecionamos outros genes correlacionados com SLITRK1 e que são codificados em regiões cromossômicas que apresentam linkage com a Síndrome de Tourette. Foram escolhidos para estudo os genes SLIT2, SLITRK1, ROBO 3 e ROBO4. Nossa amostragem é constituída por famílias de trios e famílias com um ou mais membros afetados com ST. A replicação do estudo de Abelson e colaboradores (2005) foi feita utilizando análise por DHPLC da região codificadora de SLITRK1 e posterior sequenciamento das amostras que apresentaram alterações na cromatografia. Os demais genes estudados foram avaliados a partir da genotipagem de tagSNPs. Todos os dados foram analisados utilizando-se testes estatísticos, como TDT e análise de haplótipos ambos realizados no UNPHASED e capazes de avaliar a associação e o poder do estudo. Como resultado, foram confirmados os achados de alteração no gene da SLITRK1 em paciente com ST. O estudo dos genes ROBO3 e ROBO4 não encontrou associação com a ST. Dois polimorfismos do gene da SLIT2 apresentam desvio de transmissão em pacientes com ST. E foi encontrada 9associação entre dois tagSNPs do gene da SLIT2 e a ST. Dessa forma, os genes SLIT2 e SLITRK1 são genes possivelmente relacionados com ST e devem ser continuamente investigados

Conhecidos há aproximadamente 2500 anos, os transtornos do humor continuam a dominar o interesse da saúde pública. Cerca de 5% dos pacientes depressivos não respondem a qualquer medida farmacológica e/ou psicoterápica. Para esses pacientes, a eletroconvulsoterapia (ECT) constitui uma importante oportunidade de melhora.Indução de convulsões na forma de ECT tem sido usada no tratamento de desordens psiquiátricas por mais de 60 anos. As principais indicações diagnósticas incluem depressão, mania, catatonia e esquizofrenia. Devido às dificuldades na identificação dos mecanismos de ação da ECT, têm-se usado a estimulação eletroconvulsiva (EEC) aplicada experimentalmente a animais com o intuito de obter dados que contribuam para explicar alguns efeitos terapêuticos da ECT. Há relatados de alteração nos níveis das proteínas DARPP-32 (fosfoproteína regulada por AMPc e dopamina) e NCS-1 (sensora neuronal de cálcio 1) em pacientes com transtornos neuropsiquiátricos. Neste trabalho foram avaliados os níveis de expressão das proteínas DARPP-32 e NCS-1 em quatro regiões cerebrais (striatum, córtex, hipocampo e cerebelo) de ratos submetidos ao choque eletroconvulsivo agudo e crônico. A estimulação eletroconvulsiva aguda gerou aumento na expressão de DARPP-32 no córtex. É interessante notar que nessa área as alterações foram observadas logo após a realização do estímulo e após 24 horas sendo essa sustentada até às 48 horas. Nas outras áres avaliadas (striatum, hipocampo e cerebelo) não foram observadas alterações significativas. Tais achados corroboram a ausência de eficácia dessa terapêutica de modo agudo usualmente. A estimulação crônica provocou alterações significativas em todas as áreas estudadas, o que está de acordo com a utilização de repetidas sessões desta técnica na clínica. A estimulação eletroconvulsiva aguda gerou somente alterações pontuais nos níveis de NCS-1 no córtex (diminuição em 48 horas), hipocampo (diminuição em 03 horas) e cerebelo (aumento em 3 horas). A estimulação crônica gerou modificações importantes no striatum e córtex, cerebelo. No striatum, aumento é notado logo após do último estímulo e a partir de 03 horas até às 24 horas. No córtex e cerebelo, o aumento é evidente de 12 horas até às 48 horas. Tais dados, assim como os obtidos para DARPP-32, mostram uma maior eficiência da terapia crônica em relação à aguda e uma dinâmica temporal que favorece a aplicação da técnica num intervalo de 2-3 dias (tempo pelo qual manteve-se os níveis elevados das proteínas).

The high affinity choline transporter (CHT1) is responsible for choline uptake, which is derived from acetylcholine degradation, in the synaptic cleft of cholinergic neurons. CHT1 activity is very important since cholinergic neurons have very low ability to synthesize acetylcholine de novo. Although CHT1 is mainly a plasma membrane transporter, it is found predominantly recycling between intracellular vesicles and the cell surface. Cellular trafficking seems to play an important role in the transporter activity. In the present experiments, we identified proteins that might interact with CHT1, using yeast two hybrid system. We found that CHT1 interacts with BRI3, Flotillin, SEC14like and MAP1A. These proteins participate in cellular trafficking. To further characterize these interactions we chose SEC14like, a protein that is homologous to yeast SEC14, which is essential in promoting protein transport through the yeast Golgi complex. SEC14like also plays a crucial role in phosphatidylinositol and phosphatidylcholine synthesis and metabolism. SEC14like function in humans is poorly understood. We used co-immunoprecipitation, confocal microscopy and choline uptake to further identify the importance of CHT1-SEC14like interaction. Co-immunoprecipitation assays showed that the two proteins form complexes in mammal cells. Using confocal microscopy, we found that the pattern of distribution of CHT1 was altered in cells that overexpressed SEC14like. Choline uptake analysis suggested that SEC14like overexpression modified transporter activity, decreasing 30% the rate of choline transport. All these data suggest a new possibility of regulation of cholinergic activity by control of the cellular trafficking of CHT1.<br>O transportador de colina de alta afinidade (CHT1) é responsável por recaptar a colina presente na fenda sináptica, proveniente da degradação da acetilcolina (ACh), em neurônios colinérgicos. Sua atividade é de grande importância uma vez que neurônios colinérgicos têm uma baixa capacidade para síntese de novo de colina. Embora seja um transportador de membrana plasmática, o CHT1 encontra-se em reciclagem contínua entre vesículas intracelulares e a membrana plasmática em modelos celulares neuronais. O tráfego celular parece ter um importante papel na regulação da atividade desse transportador. Em nosso trabalho procuramos identificar proteínas que poderiam interagir com o CHT1 através do sistema de duplo híbrido em leveduras. Verificamos a interação do transportador com BRI3, Flotilina, SEC14like e MAP1A. Essas proteínas têm em comum a participação em eventos de tráfego celular. No intuito de caracterizar de maneira mais profunda essas interações, escolhemos a proteína SEC14like para dar continuidade aos nossos experimentos. SEC14like é homóloga à proteína SEC14 de levedura, cuja função é essencial para o transporte de proteínas através do complexo de Golgi de leveduras, além de ter papel importante na síntese e metabolismo de fosfatidilinositol e fosfatidilcolina. Em humanos, SEC14like é uma proteína ainda pouco descrita. Utilizamos co-imunoprecipitação, microscopia confocal e experimentos de transporte de colina para delinear com mais clareza a relevância da interação CHT1-SEC14like. Através de ensaios de co-imunoprecipitação, verificamos que essa interação ocorre em células de mamíferos. Observamos também, utilizando microscopia confocal que o padrão de distribuição do transportador foi modificado em células que superexpressam SEC14like. A análise da captação de colina, sugere que a superexpressão de SEC14like, altera a atividade do transportador, diminuindo em torno de 30% a taxa de transporte de colina. Esses dados sugerem uma nova possibilidade de regulação da atividade colinérgica, envolvendo uma proteína de importância no tráfego celular e o CHT1.

The vesicular acetylcholine transporter (VACht) is responsible for acetylcholine
(Ach) storage in synaptic vesicles, an important step for cholinergic transmission. Our
goal was to study the role of VAChT in cholinergic tonus maintenance through the
generation of mice presenting alterations in VAChT gene by homologous
recombination..
One of used strategies was the insertion of a neomycin resistance cassette in
untranslated 5 region of VAChT gene. This genetic modification generated mice
HET HOM
presenting reduced expression of VAChT (VAChT Knockdown). KD and KD
present a 45% and 65% decrease in VAChT protein expression, respectively.
Accumulation of not released Ach in brain of KD was observed and it is not related
with increased expression or activity of ChAT (choline acetyltransferase) or CHT1
(High affinity choline transporter). Electrophysiological analysis at neuromuscular

and KD
junction showed a decrease in quantal size distribution for both, KD
specially in the second one.
We produced mice in which VAChT gene was flanqued by LoxP sites to

Este trabalho teve como objetivo geral investigar uma possível associação entre
cinco polimorfismos genéticos funcionais em duas doenças bucais de etiologias diferentes
em pacientes brasileiros. Para isto, a tese foi dividida em três artigos.
No primeiro, foi avaliada a associação entre os polimorfismos genéticos IL-1B
+3954 (C/T) e 5HTTLPR com a Síndrome da Ardência Bucal (SAB). Para este estudo foi
extraído o DNA de trinta pacientes com SAB e trinta e um controles. Não observamos
diferença estatística entre os grupos quanto à distribuição do polimorfismo 5-HTTLPR
(P=0.60), por outro lado, um aumento significante do genótipo heterozigoto, CT, foi
encontrado nos pacientes com SAB (P=0.005). Concluímos então que há uma associação
entre o genótipo alto produtor de IL-1 e SAB, embora mais estudos são necessários para
identificar o papel desta citocina na etiologia da SAB.
Em seguida, estudamos a associação entre o polimorfismo IL-1B +3954 (C/T) e a
EUR. Sessenta e dois pacientes com EUR e 62 voluntários saudáveis foram genotipados
para o polimorfismo IL-1B +3954. Um aumento significante da presença do genótipo
heterozigoto, CT, foi observado nos pacientes com EUR (p= 0.01). Estes resultados
sugerem que há uma associação entre o genótipo que confere uma produção aumentada
de IL-1 e EUR.
Por último, avaliamos pela primeira vez, através análise multivariada, a associação
de polimorfismos genéticos nos genes IL-B, IL-6, IL-10 e TNFA e EUR. Sessenta e quatro
pacientes com EUR econtroles participaram deste estudo. Concluímos que existe uma
associação entre os genótipos heterozigotos dos genes IL-1 e TNFA (p= 0.03 e p=0.04,
respectivamente) e EUR na população estudada.

Leucoplasia bucal é a lesão cancerizável mais comum da cavidade bucal com índice de transformação maligna variando de 1 a 18%. Atualmente, os principais parâmetros para predição de malignização em leucoplasias são as características clínicas associadas com os achados histopatológicos. Contudo, esses indicadores não conseguem determinar com precisão quais lesões irão sofrer transformação
maligna, sendo necessária a descoberta de marcadores biológicos com essa função. Estudos recentes têm demonstrado que alterações moleculares no gene supressor de tumor WWOX, localizado no cromossomo 16, na região q23.3q24.1, associado ao sítio de fragilidade FRA16D, estão presentes em diversas neoplasias malignas, incluindo o carcinoma de células escamosas. No presente
trabalho, foi realizada uma análise molecular do gene WWOX em 23 casos de leucoplasias da cavidade bucal. Através da técnica de nested RT-PCR, a transcrição do RNA mensageiro estava alterada em 26% dessas lesões. Pela técnica de imunoistoquímica, uma redução da expressão da proteína Wwox foi observada em 26% dos casos em relação ao controle. Todas as lesões que mostraram alteração do WWOX apresentavam displasia epitelial, sendo 5 amostras com displasia moderada/acentuada e 3 com displasia discreta. Esses
resultados mostram que alteração no gene WWOX é um evento genético inicial e pode contribuir para o processo de carcinogênese na boca.

Leishmania amazonensis é um dos principais agentes etiológicos em um amplo espectro de formas clínicas de leishmanioses e apresenta uma ampla distribuição geográfica nas Américas, que coincide com áreas comuns de transmissão de outras espécies de Leishmania. Neste trabalho, a eficácia protetora induzida pela imunização com o polímero formado por dois peptídeos sintéticos selecionados pelas técnicas de Phage Display e Spot synthesis foi avaliada em camundongos BALB/c contra a infecção desafio com L. amazonensis. Os peptídeos foram selecionados utilizando anticorpos da classe IgG purificados a partir de um pool de amostras de soro de cães com leishmaniose visceral. Foram obtidos três clones de bacteriófagos expressando peptídeos de interesse que apresentaram maior especificidade em relação aos anticorpos IgGs purificados. Os peptídeos sintéticos foram sintetizados pelo método de Spot synthesis em membranas de celulose e testados em experimentos de screening. Dois peptídeos (denominados de 11H e 12A) foram reconhecidos pela técnica de Spot synthesis e foram sintetizados nas suas formas solúveis; após, foram conjugados covalentemente com glutaraldeido, originando um polímero de aproximadamente 30 kDa e nesta forma, usados como imunógeno. A imunização com o polímero foi capaz de induzir uma elevada resposta imune Th1 antes da infecção. Após o desafio, uma produção elevada de IFN-?, baixos níveis de IL-4, IL-10 e anticorpos específicos ao parasita foram observados nestes animais. Em contraste, camundongos dos grupos controle exibiram baixos níveis de IFN ? e níveis elevados de IL-4, IL-10 e anticorpos antiparasita. Dessa forma, o polímero dos dois peptídeos mostrou-se como um bom candidato à vacina contra a Leishmaniose Tegumentar Americana.

The vesicular acetylcholine transporter (VAChT) is the protein responsible for packaging cytoplasmic acetylcholine (ACh) into the synaptic vesicle of cholinergic neurons. The cytosolic C-terminal region of VAChT is important in intracellular trafficking events and targeting to secretory organelles. In this study, we evaluate the existence of trafficking signals in this region. Using confocal microscopy and GFP (green fluorescent protein) as a molecular label, the intracellular trafficking of several VAChT constructs was observed. Our results suggest that amino acids 471-490 contain important signals for endocytosis, trafficking, endoplasmic reticulum (ER) export and the targeting of VAChT to synaptic like microvesicles (SLMVs). In the region comprehended by residues 481-485 seems to exist a signal for endoplasmic
reticulum export since the mutation of these amino acids retains the transporter in the ER. Mutation of the di-leucine motif (L485 L486) relocates the transporter to the soma plasma membrane. Protein-protein interaction experiments demonstrated that the C-terminal region of VAChT interacts with subunit of clathrin adaptor complex AP2 in a di-leucine motif dependent manner. To evaluate the role of clathrin in the
trafficking and endocytosis of VAChT, we used the AP180-C construct. When AP180-C is overexpressed the clathrin-mediated endocytosis is blockade. These experiments demonstrated that clathrin mediated endocytosis is an essential event for VAChT trafficking. In addition, our results showed that the amino acids involved in the interaction with the clathrin endocytic machinery are present in the sequence between residues 471-490 of the C-terminal region of VAChT. In our next step, we
tried to identify proteins that interact with VAChT and contribute in its trafficking and sorting to synaptic vesicles. For this reason, we performed a screening in a human brain cDNA library. The interaction between VAChT and SEC14like was detected in this system and was confirmed by co-immunoprecipitation. SEC14 is implicated in vesicle trafficking and in phosphatidylinositol and phosphatidylcholine transfer between membranes. Co-localization experiments suggest not only that VAChT and SEC14like co-localize but also that the subcellular distribution of VAChT appears to be altered in the SEC14 presence. Further studies must be performed to verify the implications of SEC14like overexpression on VAChT trafficking. Our results strongly suggest that: 1- there is an ER export signal between amino acids 481-485; 2- the di-leucine motif is responsible for VAChT interaction with AP2; 3- the clathrin dependent
endocytosis has a fundamental role in endocytosis and trafficking of the transporter; and 4- the overexpression of SEC14like seems to interfere with VAChT trafficking.<br>O transportador vesicular de acetilcolilna (VAChT) é a proteína responsável pelo empacotamento de acetilcolina (ACh) citoplasmática em vesículas sinápticas de neurônios colinérgicos. A região C-terminal do transportador, que é voltada para o citosol, é importante nos eventos de tráfego intracelular e direcionamento para organelas secretórias. Neste estudo, nós procuramos identificar os sinais de tráfego
presentes nessa região. Utilizando a GFP (proteína fluorescente verde) como etiqueta molecular e a microscopia confocal, o tráfego intracelular de diversas construções do VAChT, com mutações e deleções, foi avaliado. Nossos resultados sugerem que os aminoácidos 471-490 contém sinais importantes para a saída do retículo endoplasmático, endocitose, tráfego e direcionamento do VAChT para synaptic like microvesicles (SLMVs). Entre os resíduos 481-485 parece existir um sinal de exportação do retículo endoplasmático, uma vez que a mutação destes aminoácidos levou à retenção do transportador neste compartimento. A mutação do motivo di-leucina, presente nas posições 485 e 486, leva à um acúmulo do transportador na membrana plasmática. Experimentos de interação proteína-proteína mostraram que a região C-terminal do VAChT interage com a subunidade do complexo adaptador de clatrina AP-2, de uma maneira dependente do motivo baseado em leucina. Com o intuito de avaliarmos o papel da clatrina no tráfego e endocitose do VAChT, utilizamos a construção AP180-C que, quando superexpressa, bloqueia a endocitose de proteínas que dependem de clatrina. Estes experimentos demonstraram que a endocitose mediada por clatrina é um evento essencial para o tráfego do VAChT. Além disso, nossos resultados também demonstraram que os aminoácidos envolvidos na interação com a maquinaria endocítica estão presentes na sequência compreendida entre os resíduos 471-490 da região C-terminal do transportador. Nosso próximo passo foi tentar identificar proteínas que interagem com o VAChT e auxiliam no seu tráfego e direcionamento para as vesículas sinápticas. Para isso, realizamos a varredura de uma biblioteca de cDNA de cérebro humano. A interação entre VAChT e SEC14like foi detectada neste sistema, e confirmada através de co-imunoprecipitação. SEC14 é uma proteína envolvida com tráfego de vesículas e na transferência de fosfatidilinositol e fosfatidilcolina entre membranas. Experimentos de co-localização, sugerem não apenas que VAChT e SEC14 se co-localizam, como também que a distribuição subcelular do transportador parece estar alterada na presença de SEC14. Outros estudos ainda precisam ser realizados para avaliar a implicação da superexpressão de SEC14like no tráfego do VAChT. Os resultados deste trabalho sugerem fortemente que, existe um sinal de exportação do retículo endoplasmático entre os aminoácidos 481-485, o motivo baseado em leucina é o responsável pela interação entre o VAChT e AP-2, a endocitose mediada por clatrina tem um papel fundamental na endocitose e tráfego do transportador e que a superexpressão de SEC14like parece interferir no tráfego do VAChT.

Cellular prion protein (PrPsen ou PrPc) is expressed in a wide variety of cells, specialy neurons. This protein is rich in alpha helix structures, but it can assume an alternative conformational isoform known as PrPres (or PrPsc). PrPres presents mainly beta sheet structure and gains the ability to form aggregates as well as is protease resistant. PrPres is able to propagate by converting PrPsen molecules to the alternative conformation isoform PrPres in a new biologic mechanism, not yet fully understood. Then, PrPres molecules form aggregates that accumulates in the brain causing Prion Diseases, a group of neurodegenerative disorders very rare and fatal. PrPsen and PrPres internalization and subsequent intracellular trafficking may give important sights about the physiological role of PrPsen and the mechanisms of PrPres infection and propagation between cells. In order to pursue these mechanisms, we used a GFP-tagged version of the mouse cellular prion protein. This construction allowed us to check PrPsen distribution and follow its endocytosis and traffic in a living neuronal cell line, SN56. SN56 cells expressing GFP-PrPc showed fluorescence signal distributed at the
plasma membrane and in intracellular compartments localized close to the nucleus, similarly to endogenous protein. In response to copper treatment, GFP-PrPc was internalized and directed to a perinuclear region, suggesting that Cu2+ may have an important role in PrPsen physiology. We also examined the steady state distribution of a
N-terminal deletion mutant of PrPsen (Ä32 121) tagged with GFP. This mutant showed an accumulation at the plasma membrane and weak labeling of the perinuclear region suggesting an endocytosis deficiency. This mutant did not show internalization after copper treatment suggesting that N-terminal region may be involved with PrPsen
endocytosis. Organelle specific markers suggest that the perinuclear region labeled with GFP-PrPc is composed of recycling endosomes and Golgi. We also evaluate the mechanism involved in GFP-PrPc endocytosis and our data suggests that dynamin I is important to both, constitutive and copper induced GFP-PrPc endocytosis. Cells expressing a C-terminal fragment of AP-180, a protein that recruits clathrin to the plasma membrane, showed 75% inhibition of clathrin dependent
endocytosis. In these conditions, constitutive endocytosis of GFP-PrPc was not altered and the copper induced internalization showed a partial inhibition. These data suggest xvi that GFP-PrPc may present a complex endocytosis mechanism involving clathrin dependent and independent pathways. The clathrin independent mechanism does not involve caveolae, since we failed to detect caveolin, a caveolae marker, in SN56 cells. We also examined PrPres internalization and intracellular trafficking by labeling several PrPres strains with the fluorescent dye alexa568. SN56 cells were able to take up Chandler PrPres from aggregates and distribute this fluorescent protein in vesicles inside the cell body and neuritis. However, those cells were not able to internalize PrPres of other strains, like 263K and 87V, with the same efficiency seen for Chandler
PrPres. Moreover, cells treated with Chandler PrPres showed formation of new PrPres molecules, suggesting the establishment of a sustained infection, while cells treated with either 263K or 87V PrPres did not show PrPres formation. These observations suggest that uptake and intracellular trafficking of PrPres coincide with sustained infection in
SN56 neuronal cells. Thus, the mechanisms and pathways taken by PrPres may be important to the establishment of a sustained infection.
Chandler PrPres positive vesicles spread in the cell body and neuritis co-localized with late endosomes and lysosomes markers (dextran and lysotracker) and failed to colocalize with markers of clathrin dependent endocytosis (transferrin) or lipid raft endocytosis (cholera toxin), suggesting that those vesicles are directed to acidic organelles after internalization probably using a clathrin and raft independent
mechanism. Chandler PrPres positive vesicles did not colocalize with GFP-VAChT, a marker for sinaptic vesicles, in neuritis or cell body.
Overexpression of a constitutively active GTP-bound Rab7 fused to GFP (GFPRab7 Q67L) showed that Chandler PrPres localizes in late endosomes and lysosomes in the cell body and neuritis. Expression of Rab7 and mutants in another cell line persistently infected with PrPres (N2a) showed alteration of new PrPres formation. The results with infected SN56 and N2a expressing Rab7 mutants suggest that trafficking in late endosomes and lysosomes may play an important role in sustained PrPres formation.<br>A proteína prion celular (PrPsen ou PrPc) é uma proteína expressa em vários tipos celulares, especialmente em neurônios. Esta proteína apresenta uma estrutura rica em alfa hélices, no entanto pode sofrer uma mudança de conformação secundária gerando uma isoforma conhecida como PrPres (ou PrPsc). Esta isoforma apresenta uma estrutura predominante de folhas beta e adquire a capacidade de se associar formando amilóides, bem como resistência a tratamento com proteases. Através de um mecanismo biológico ainda pouco esclarecido, PrPres é capaz de promover a conversão de moléculas de
PrPsen para PrPres. A formação e deposição de agregados insolúveis de PrPres no sistema nervoso estão relacionados a uma série de doenças neurodegenerativas raras e fatais. A internalização e tráfego intracelular destas isoformas podem revelar informações importantes sobre o papel fisiológico de PrPsen, bem como os mecanismos de infecção e propagação de PrPres entre células. Interessados nestes mecanismos, nosso grupo construiu uma versão da proteína prion celular ligada a green fluorescent protein (GFP) para monitorarmos a distribuição, endocitose e tráfego desta proteína em células
neuronais SN56 vivas. Células SN56 expressando esta proteína de fusão (GFP-PrPc) apresentaram a fluorescência distribuída pela membrana plasmática e em compartimentos intracelulares próximos ao núcleo, de maneira similar a proteína endógena. GFP-PrPc foi internalizada e direcionada para uma região perinuclear na presença de cobre, indicando um papel importante deste íon na fisiologia de PrPc. Um mutante de deleção N-terminal (aminoácidos 32 a 121) ligado a GFP apresentou maior acúmulo na membrana plasmática e menor concentração na região perinuclear, sugerindo uma deficiência na endocitose. Além disso, o cobre não foi capaz de induzir a
endocitose deste mutante, indicando que a região N-terminal é importante para a endocitose constitutiva e induzida por cobre de PrPc. Experimentos de dupla marcação indicaram que a região perinuclear positiva para GFP-PrPc era constituída de endossomas de reciclagem e Golgi. O mecanismo de endocitose de GFP-PrPc também foi avaliado e a endocitose constitutiva e induzida por cobre de GFP-PrPc foi dependente de dinamina I. Células expressando o fragmento C terminal da proteína AP180, que compete com a proteína endógena responsável pelo recrutamento de clatrina para a membrana plasmática, xiv
apresentam 75% de inibição da endocitose dependente de clatrina. Nestas células, a endocitose constitutiva de GFP-PrPc não sofreu alteração e a induzida por cobre apresentou uma inibição parcial, sugerindo que o mecanismo de endocitose de GFPPrPc é complexo, podendo envolver tanto um mecanismo dependente como
independente de clatrina. Provavelmente, o mecanismo independente não envolve caveolae, já que não pudemos detectar caveolina, um marcador para caveolae, em células SN56. Nós também avaliamos a internalização e tráfego de PrPres usando diferentes cepas de PrPres conjugadas à molécula fluorescente alexa568. Células SN56 foram capazes de processar agregados da cepa de PrPres Chandler de camundongo distribuindo a fluorescência em vesículas espalhadas pelo citoplasma e neuritos. Outras cepas de PrPres, 263K e 87V, não foram processadas e internalizadas com a mesma eficiência
em células SN56. As células incubadas com Chandler PrPres foram capazes de produzir novas moléculas de PrPres, indicando que estavam infectadas persistentemente, enquanto as cepas 263K e 87V não foram capazes de infectar estas células. Estes dados mostram que a endocitose e tráfego de PrPres foram coincidentes com a infecção das
células, sugerindo que as vias utilizadas por PrPres para internalizar e trafegar na célula podem ter importância neste processo. As vesículas de Chandler PrPres colocalizaram com marcadores para endossomas
tardios e lisossomas (dextran e lisotracker) e não colocalizaram com marcadores da via clássica de clatrina (transferrina) ou vesículas provenientes de endocitose por lipid rafts (toxina da cólera) indicando que após internalização provavelmente por um mecanismo independente de clatrina ou rafts, Chandler PrPres é direcionado para
organelas acídicas. A expressão do mutante constitutivamente ativo de Rab 7 (GFPRab7 Q67L) confirmou que PrPres está presente em endossomas tardios e lisossomas no corpo e neuritos. Em neuritos, Chandler PrPres não colocalizou com GFP-VAChT, um marcador de vesículas sinápticas. Experimentos com outra linhagem celular (N2a)
persistentemente infectada por PrPres indicaram alteração na formação de PrPres na presença de mutantes de Rab 7. Estes dados com Rab7 em SN56 e N2a infectadas sugerem que o tráfego de PrPres pelos endossomas tardios e lisossomas pode ter uma participação no processo de formação de novas moléculas de PrPres.

Neuronal calcium sensor-1 (NCS-1) is the most ancient protein of EF-hands super
family which is a group of proteins that binds calcium. It was shown that NCS-1
has many functions such as its capacity of inhibiting dopamine receptor D
2
desensitization and its capacity to activate phosphatidylinositol 4-kinase (PI4K).
The dopamine and cAMP regulated phosphoprotein-32kDa (DARPP-32) has a
central role in modulation of transduction of dopaminergic signaling. When
phosphorylated at Threonine 34, DARPP-32 turns into a potent inhibitor of protein
phosphatase-1 (PP-1) and consequently modulates the signaling integration.
Recently it was demonstrated upregulation of NCS-1 and DARPP-32
downregulation in prefrontal (PFC) cortex of schizophrenic patients. In our study
we have investigated the role of NCS-1 on cAMP/PKA intracellular signaling
pathway modulation. We also investigated if chronic treatment with typical and
atypical antipsychotics modulates NCS-1 and DARPP-32 expression, both in vitro
and in vivo (in rat model). We used pheochromocytome PC12 wild-type cells (WT)
and overexpressing NCS-1 (Clone). Our results showed that Clone PC12 cells
have reduction in DARPP-32 expression, cAMP levels, pDARPP-32(Thr34) and
pCREB(Ser133). However, there was no alteration in D
, Calcyon, pDARPP-

32(Thr75), Akt and pAkt(Ser473) in these same cells. WT and Clone PC12 cells
were treated with specific dopaminergic agonists and antagonists and we show
that alteration in pDARPP-32(Thr34) levels in Clone PC12 cells is independent of
dopaminergic signaling. Also, we demonstrated that both PC12 cells and different
brain regions of rats treated chronically with typical and atypical antipsychotics did
not present alteration in NCS-1 and DARPP-32 expression. Therefore, we
observed that NCS-1 modulates DARPP-32 expression and cAMP/PKA signaling

Uma de suas característcas é a presença da cauda miristoil N -terminal que pode se inserir em membranas lipídicas. NCS-1 apresenta diversas funções, como inibir a dessensibilização do receptor dopaminérgico D e ativar 2 PI4K. A fosfoproteína regulada por dopamina e AMPc-32kDa (DARPP-32) tem um papel importante na modulação da transdução da sinalização dopaminérgica. Quando fosforilada na Treonina 34, DARPP-32 inibe a proteína fosfatase-1 (PP-1) e consequentemente modula a integração das sinalizações. Utilizamos células PC12 selvagem e superexpressando NCS-1, sendo que estas últimas apresentaram redução dos níveis de expressão de DARPP-32 e nos níveis de AMPc, pDARPP-32(Thr34) e pCREB(Ser133). típicos e atípicos, não apresentaram alterações nos níveis de NCS-1 e DARPP-32. Portanto, observamos que NCS-1 modula expressão de DARPP-32 e altera a sinalização da via AMPc/PKA independentemente da dopamina.

Calcium influx through voltage sensitive calcium channels (VSCCs) plays an
important role on the nociceptive transmission in the dorsal horn of the spinal cord.
The intrathecal administration of VSCCs blockers, such as -conotoxin-MVIIA (-
CTx-MVIIA) has analgesic effect, but it narrows the therapeutic window, presenting
many adverse effects. The Brazilian spider Phoneutria nigriventer venom contains a
variety of substances with predominantly neurotoxic action, among them the Ph1
which was recently described as a potent neurotransmission blocker for interacting
with N-type voltage sensitive calcium channels. In this study, we verify the analgesic
potential of Ph1 by its intrathecal administration in acute and persistent pain
models in rodents. The Ph1 produced a prolonged antinociception and with great
effectiveness in the hot plate model in mice when compared to -CTx-MVIIA.
However, Ph1 or -CTx-MVIIA did not produce adverse motor effects in mice.
Ph1 was more effective and powerful than -CTx-MVIIA when assessed in rats,
not only in preventing the nociception induced by formalin, but particularly in
reversing the persistent inflammatory nociception previously installed. Furthermore,
the analgesic action of both toxins was related to inhibition of pro-nociceptive
neurotransmitter release evoked by calcium in the spinal cord, such as glutamate.
When the adverse effects were evaluated in rats, we verified that Ph1 had a
therapeutic index higher than -CTx-MVIIA. However, both toxins were able to
produce blood pressure alterations. In addition, the treatment with Ph1 toxin is
able to reduce the acute nociception induced by surgical incision, or chronic, caused
by arthritis or by sciatic nerve injury in rodents. Finally, the recombinant Ph1 toxin

expressed in Escherichia coli showed an antinociceptive activity similar to native
toxin. Thus, the present study demonstrated that Ph1 toxin presented analgesic
effects in different models of pain, suggesting that this toxin may have a potential to
be used as a drug for the control of the persistent pain.<br>O influxo de cálcio através de canais de cálcio regulados por voltagem (CCSVs)
medeia a informação nociceptiva no corno dorsal da medula espinhal. A
administração intratecal dos bloqueadores de CCSVs, tais como a -conotoxina-
MVIIA (-CTx-MVIIA), tem efeito analgésico, mas estreita janela terapêutica,
apresentando muitos efeitos adversos. O veneno da aranha brasileira Phoneutria
nigriventer contém uma variedade de substâncias com ação predominantemente
neurotóxica, dentre elas a toxina Ph1 que foi recentemente descrita como uma
potente bloqueadora da neurotransmissão por interagir com canais de cálcio
sensíveis a voltagem tipo N. Neste estudo, verificamos o potencial analgésico da
Ph1, administrada por via intratecal, em modelos de dor aguda e persistente em
roedores. A Ph1 produziu antinocicepção prolongada e com grande eficácia no
modelo da placa quente em camundongos, quando comparada com a -CTx-MVIIA.
Além disso, a Ph1 ou a -CTx-MVIIA não produziram efeitos adversos motores
em camundongos. Quando avaliadas em ratos, a Ph1 foi mais efetiva e potente
do que a -CTx-MVIIA não somente em prevenir a nocicepção induzida pela
formalina, todadvia especialmente em reverter a nocicepção inflamatória persistente,
previamente instalada. Além do mais, a ação analgésica de ambas as toxinas foi
relacionada com a inibição da liberação de neurotransmissores pró-nociceptivos
evocados por cálcio na medula espinhal, como o glutamato. Quando os efeitos
adversos foram avaliados em ratos, verificamos que a Ph1 teve um índice
terapêutico maior que a -CTx-MVIIA. Porém, ambas as toxinas foram capazes de
produzir alterações na pressão arterial. Entretanto, o tratamento com a toxina Ph1
é capaz de reduzir a nocicepção aguda induzida por incisão cirúrgica ou crônica

causada por atrite ou por lesão do nervo ciático em roedores. Por fim a toxina
recombinante Ph1 expressa em Escherichia coli mostrou uma atividade
antinociceptiva similar à toxina nativa. Assim, o presente estudo demonstrou que a
toxina Ph1 apresentou efeitos analgésicos em diferentes modelos de dor,
sugerindo que esta toxina pode ter potencial para ser usado como uma droga para o
controle da dor persistente.

Epidemiologic studies suggest that the methylenetetrahydrofolate reductase
(MTHFR) C677T polymorphism may modify the risk of both colorectal cancer
(CRC) and breast cancer (BC). The C677T polymorphism is associated with the
expression of a thermolabile enzyme with decreased activity that influences the
levels of methyl-donor molecules. Considering that methylation abnormalities
appear to be important in the pathogenesis of CRC, we have examined the
relationship between the genotype at the MTHFR C677T polymorphism,
hypermethylation of the promoter region of five genes (DAPK, MGMT, hMLH1,
INK4a ARF
p16
and p14 ) and microsatellite instability (MSI) in 106 patients with CCR. In
195 BC cases, the genotype C677T was correlated with clinical parameters of the
patients and stage of disease. The MTHFR C677T polymorphism was assessed by
using polymerase chain reaction-restriction fragment length polymorphism (PCR-
RFLP). The analysis of MTHFR 677 TT genotype between cases and controls did
not reveal any statistically significant difference. We did not show statistically
significant differences in the genotypic frequencies of the MTHFR C677T
polymorphism with or without hypermethylation of one or multiple loci. However,
our study found a significant excess of 677TT individuals among patients with CRC
with MSI. This strong association appears to be independent of the methylation
status of hMLH1 or of the biogeographical genomic ancestry of the patients (typed
utilizing indels). An explanation for this mechanism between MSI, 677TT genotype
and CCR is not clear. We did not find statistical difference in the frequency of 677
TT genotype between controls and BC cases. In BC patients, no frequency
differences of 677 TT genotype were observed among smoking habit, age, and
menopausal status at the initial diagnosis. However, a strong statistical association
was found between TT genotype and BC alcohol drinkers and BC stage IIIA. Our
results suggest that the low activity of the MTHFR, and the powerful folate
antagonistic action of alcohol, could increase the risk for BC. Besides the
association between severity of disease and TT genotype, in this study, BC stage is
linked to temporality of diagnosis and does not reflect tumor aggressiveness.
Additional studies should be carried out to clarify the relationship among alcohol
drinking, stage, and. MTHFR C677T polymorphism. The preventive use of folate for

associação entre o genótipo TT e a presença de MSI. Essa associação parece ser
independente do status de metilação do promotor do gene hMLH1, ou da
ancestralidade genômica dos pacientes (genotipados através da análise de indels).
Em relação ao CM, não foi encontrada associação estatística entre genótipo TT e
CM. Não houve diferença estatística das freqüências genotípicas da C677T com
relação ao tabagismo, à idade e status hormonal (pré ou pós-menopausa), no
momento do diagnóstico. No entanto, foi encontrada uma forte associação
estatística entre o genótipo TT e pacientes com CM etilistas e pacientes
diagnosticadas no estadio IIIA. Nossos resultados sugerem que os efeitos
deletérios advindos da baixa atividade da MTHFR, associados à potente ação
antagonista do folato, induzida pelo álcool, podem aumentar o risco para CM.
Apesar de encontrarmos associação entre estadio avançado da doença e genótipo
TT, o estadiamento neste estudo pode estar ligado à temporalidade do
diagnóstico, não refletindo a agressividade tumoral. Estudos adicionais focados na
relação do álcool, estadiamento para CM e polimorfismo da MTHFR devem ser
realizados. O uso preventivo de folato, para pacientes etilistas portadoras do
genótipo TT, deve ser considerado, com o objetivo de reverter os efeitos do
genótipo 677TT no CM.

A depressão é uma doença de elevada prevalência, que determina sofrimento, incapacidade e desordem social e eleva a taxa de mortalidade de doenças associadas, como a doença arterial coronariana.
A população idosa é especialmente comprometida pela doença, apesar de ser frequentemente subdiagnosticada em decorrência das manifestações próprias nesta faixa etária. Nestes pacientes, ela se associa frequentemente à incapacidade cognitiva e outros comprometimentos funcionais. A depressão de início tardio é um subtipo especial de depressão, cujo início ocorre após os 60 anos de idade. Ela apresenta etiologia e características diferentes da depressão de início precoce.
Inúmeros estudos apontam o papel da serotonina nos distúrbios do humor e alterações em sua síntese poderiam levar à depressão maior. A TPH2 é uma enzima limitante da síntese deste neurotransmissor. Assim, alterações moleculares no gene TPH2 foram consideradas como associadas aos fenômenos comportamentais incluindo transtornos do humor, suicídio, traços de personalidade e dependência química por álcool e drogas.
Em 2005, Zhang et al. demonstraram que os níveis de 5-HT foram reduzidos em 80% quando a mutante Arg441His era expressa em células PC-12, indicando uma perda significativa da função da enzima mutante. Além disso, este SNP funcional foi observado em nove indivíduos dentre 87 portadores de depressão maior refratária ao tratamento medicamentoso e com idade superior a 60 anos. Entretanto, vários grupos, após estudarem mais de 5000 indivíduos de diversos grupos étnicos, não conseguiram encontrar a variante acima descrita. Estes grupos haviam incluído em suas amostras indivíduos com depressão grave, necessitando eletroconvulsoterapia, como os descritos por Zhang et a. (2005). Porém, nenhum grupo estudou pacientes idosos com o objetivo de analisar se a variável em questão encontrava-se associada à depressão de início tardio.
Após analisarmos o exon 11 de 161 indivíduos idosos (100 portadores de depressão de início tardio e 61 controles) não observamos a presença da variante descrita. Sendo assim, concluímos que a variante descrita por Zhang é uma forma rara e que, provavelmente, esteja relacionada à origem étnica da população estudada.

Twelve dairy cows were collected between three weeks to parturition to three weeks after. They was in a free-stall in Inhaúma city, Minas Gerais estate between October and December of 2012. The metabolic profile was compounded with serum non-esterified fatty acids (AGNE), beta-hydroxybutyrate (BHBA), cholesterol (COL), total protein (TP), albumin (ALB), calcium, phosphorus, magnesium as well as the activity serum enzymes aspartate transaminase (AST) and lactate dehydrogenase (LDH). In a group of six animals, four sites were evaluated for blood collection - jugular, mammary and coccygeal veins and tip of tail - to BHBA measurement and comparison of the method of analysis of the electrochemical handset with the laboratory method by spectrophotometry by chemical moist. There were differences in serum NEFA and BHBA between prepartum and postpartum (p <0.05). There was a decrease in the concentration of COL with the approaching birth with subsequent increase (p <0.05). Serum concentrations of minerals, PT and ALB did not differ (p> 0.05). The enzymatic activity of AST and LDH were higher in the postpartum period (p <0.05). Serum BHBA measure in the samples collected from tip of tail, jugular and coccygeal did not differ (p> 0.05) being higher than in the mammary vein (p <0.05). There was no difference between the methods of analysis (p> 0.05). The metabolic profile is an important monitoring tool. The handset is efficient in determining BHBA and can be used blood samples from the jugular and coccygeal veins and from the tip of tail.<br>Estudou-se um grupo de doze vacas da raça Holandesa, três semanas pré-parto até três pós-parto, em sistema free-stall, localizado em Inhaúma, Minas Gerais, no período de outubro a dezembro de 2012. Avaliou-se o perfil metabólico através da concentração sérica de ácidos graxos não esterificados (AGNE), beta hidroxibutirato (BHBA), colesterol (COL), proteína total (PT), albumina (ALB), cálcio, fósforo, magnésio bem como a atividade sérica das enzimas aspartato transaminase (AST) e lactato desidrogenase (LDH). Em um grupo de seis animais, avaliaram-se quatro locais para colheita de sangue - veias jugular, mamária e coccígea e ponta de cauda - para mensuração de BHBA e comparação do método de análise eletroquímico do aparelho portátil com o método laboratorial por espectrofotometria pela química úmida. Houve diferença nas concentrações séricas de AGNE e BHBA entre o pré-parto e pós-parto (p<0,05). Observou-se diminuição na concentração de COL com a aproximação do parto com posterior aumento (p<0,05). As concentrações séricas dos minerais, PT e ALB não apresentaram diferenças (p>0,05). A atividade enzimática de AST e LDH foram maiores no período pós-parto (p<0,05). A concentração sérica de BHBA, obtida através das amostras coletadas da ponta de cauda, e veias jugular e coccígea, não diferiram (p>0,05) sendo mais altas do que na veia mamária (p<0,05). Não houve diferença entre os métodos de análise (p>0,05). A avaliação do perfil metabólico é uma importante ferramenta de monitoramento. O aparelho portátil é eficiente na determinação de BHBA, podendo-se utilizar amostras de sangue das veias jugular e coccígea e da ponta de cauda.

This paper aimed at studying the canine population dynamics and evaluate its population management strategies in Itabirito county, Minas Gerais State, from 2007 to 2011. Data were collected through two cross-sectional studies applied at intervals of one year, 10% of the canine population, randomly sampled systematically in all neighborhoods of the urban area, and by obtaining secondary data from the Zoonosis Service Control and the Department of animal protection in the county. The impact of population density in canine sterilization was performed by means of mathematical modeling. The results revealed the existence of 5.2 dogs per capita, 0.47 dogs per property (2.25 to 1.85) dogs per property with their presence. The dogs were present in 72.8% of the surveyed properties, which 42.09% contained two animals and 93.86% had physical barriers blocking them. The average age of the animals was 3.5 and the ratio male / female was (1,1105:1 - 0,98:1). The birth rate was 19.39%, the mortality rate was around 20.86% and abandonment dog´s rate was over 3.6%. The average number of pups born alive was 3.7 per pregnancy and neonatal mortality was 31.4%. The proportion of sterilized animals was (15% - 21.78%). As for the breed, 56.41% of the dogs had not defined breed, 41.71% were considered as small size and 66.67% were aimed at companionship. In additional, 47.46% of the dogs were welcomed by donation and 42.38% never left to wander. Regarding reproductive control, around 17% of the dog´s population that had come from all the neighborhoods in the urban area, were sterilized through the years 2007 to 2011. The annual rates of sterilization used in the resident population were not effective in promoting a significant decrease in canine population density over ten years, however in the presence of abandoning the impact of this strategy on canine density was higher. The selective collection of dogs on the streets was done humanely, but the number of dogs collected was lower compared to the total number of animals in the city, and the euthanasia of these animals were the most frequent destination. The campaign promoted by the animal protection entity presented a significant number of adoptions, but was inefficient as tracking dogs after adoption. The dog´s population was in equilibrium, and this may has been associated with interventions promoted by man. The rates of sterilization, euthanasia and adoption presented close values and together may have influenced the equilibrium in the dog´s population. However, the abandonment problem of these animals was detected and It was influenced by social, economic and cultural conditions, that leaded to the implementation of educational activities in the county as a strategy to increase the level of the guard in charge of the dog´s population and reduce the abandoned dogs and provide sustainability for management actions in canine population in Itabirito.<br>O objetivo deste trabalho foi estudar a dinâmica populacional canina e avaliar as estratégias de manejo populacional aplicadas nesta população no município de Itabirito, MG, durantes os anos de 2007 a 2011. Os dados foram obtidos por meio de dois estudos transversais aplicados com intervalos de um ano, em 10% da população canina, amostrada de forma aleatória sistemática em todos os bairros da zona urbana do município. Foram analisados ainda dados secundários provenientes do Serviço de Controle de Zoonoses e da entidade de proteção animal do Município. O impacto da esterilização na densidade populacional canina foi realizado por meio de modelagem matemática. Verificou-se a existência de 5,2 cães por habitante, 0,47 cão por imóvel e (2,25 - 1,85) cães por imóvel com a presença deles em 2010 e 2011 respectivamente. Os cães estiveram presentes em 72,8% do total de imóveis pesquisados, dos quais 42,09% possuíam dois animais. A maioria (93,86%) possuíam barreiras físicas que continham os animais. A idade mediana dos animais foi 3,5 anos e a razão macho/fêmea (1,11:1 - 0,98:1) em 2010 e 2011 respectivamente. A taxa de natalidade foi 19,39%, a taxa de mortalidade 20,86% e a taxa de abandono foi de 3,6%. A média de filhotes nascidos vivos por gestação foi de 3,7 e a mortalidade neonatal foi de 31,4%. A proporção de animais esterilizados foi 15% - 21,78% em 2010 e 2011 respectivamente. Quanto à raça, 56,41% dos cães não tinham raça definida, 41,71% foram considerados de porte pequeno e 66,67% tinham como finalidade a companhia. Quanto à forma de aquisição, 47,46% dos cães foram recebidos de doação e 42,38% nunca saíam para passear. Em relação ao controle reprodutivo, aproximadamente 17% da população foi esterilizada durante os anos de 2007 a 2011, sendo esses animais provenientes de todos os bairros da zona urbana do Município. As taxas de esterilização anuais empregadas na população domiciliada não foram eficazes para promover uma diminuição significativa na densidade populacional canina, ao longo de dez anos, porém o impacto dessa estratégia na densidade canina foi maior na presença do abandono. O recolhimento seletivo de cães nas ruas foi realizado de forma humanitária, porém o número de cães recolhidos foi baixo em relação ao número total de animais no Município, sendo a eutanásia desses animais o destino mais frequente. A campanha de adoção promovida pela entidade de proteção animal possui números expressivos de adoção, mas é ineficiente quanto ao monitoramento dos cães após a adoção. A população canina se encontra em equilíbrio, e esse fato pode estar associado às intervenções promovidas pelo homem. As taxas de esterilização, eutanásia e adoção obtidas mostraram valores próximos e juntas podem ter influenciado a situação de equilíbrio que a população canina se encontrava, durante o período estudado. Entretanto, foi detectado o problema de abandono desses animais, influenciados por condições sociais, econômicas e culturais da população humana. Diante do cenário encontrado, recomendou-se a implantação de ações educativas no Município para dar sustentabilidade às ações de manejo populacional canino, para aumentar o nível da guarda responsável da população e diminuir o abandono dos cães no Município.

Was conducted a descriptive study with the objective of to describe the occurrence of canine visceral leishmaniasis, in the Northwest Regional in Belo Horizonte, from 2006 to 2011. Data regarding with the canine control were obtained from the Municipal Secretariat of Health. From a total of 151.725 tests, 86.1% were negative, 7.4% were positive, 2,1% were undetermined and 3.8% were monitor. The expansion of the disease along the period was evident. There was an increase of the coverage of census surveys and a decrease in the tests requested by residents, followed by decreasing of the canine prevalence in areas covered in the period. The undetermined results also have declined, while serologies "monitor" have increased. The seroconversion rate among dogs undetermined was 54.3%, occurring in 104 days on average, with a median of 54 days. It was found that 98, 1% of the samples were collected on filter paper and the laboratory diagnosis was delivered in 18.1 days on average, after the blood collection. The infected dogs removal happened about 51.7 days on average, with median of 39 days. The positive dogs withdrawal was 86.6%, which was negatively impacted by refusals, which represented 7.8% over the period, ranging from 1.9% to 15.6%. It is necessary to intensify the activities of health education in order to alert citizens about the dogs importance in the infection transmission dynamics and thereby increase the population adherence to the Visceral Leishmaniasis Control Program. The database of the Information System for Zoonosis Control of Belo Horizonte was conducive to study the expansion and the combat of the disease in the city. Thus, it is recommended that it receives periodic maintenance, with quality monitoring, so that further studies might be performed.<br>Foi realizado um estudo descritivo com o objetivo de descrever a evolução do programa de vigilância e controle da leishmaniose visceral canina bem como a dispersão dos cães positivos, na Regional Noroeste de Belo Horizonte, de 2006 a 2011. Para tanto, foram utilizados dados referentes ao controle do reservatório urbano, obtidos do Sistema de Informação e Controle de Zoonoses da Secretaria Municipal de Saúde. De um total de 151.725 exames, 86,1% dos cães foram considerados negativos, 7,4% positivos, 2,1% indeterminados e 3,8% monitorar. A expansão da soropositividade canina ao longo da série temporal foi evidente. Verificou-se aumento da cobertura de inquéritos sorológicos censitários e decréscimo da solicitação de exames pelos munícipes, seguido de diminuição da soroprevalência da infecção em cães. Os resultados indeterminados também apresentaram redução, ao contrário do que ocorreu com os resultados monitorar. A taxa de soroconversão entre os cães indeterminados foi de 54,3%, ocorrendo em 104 dias em média, com mediana de 54 dias. Constatou-se que 98, 1% das amostras foram coletadas em papel filtro e que o resultado do diagnóstico laboratorial foi disponibilizado para o distrito sanitário em 18,1 dias em média, após a coleta sanguínea. Desta à remoção do cão infectado, transcorreram 51,7 dias em média, com mediana de 39 dias. A retirada de cães positivos foi de 86,6%, a qual foi influenciada negativamente pela elevação das recusas, que representaram 7,8% no período, variando de 1,9% a 15,6%. O banco de dados do Sistema Informatizado de Controle de Zoonoses se mostrou propício à realização de estudos de dispersão e combate à doença no município. Assim, recomenda-se que o mesmo receba manutenções periódicas e monitoramento da qualidade, a fim de que outras análises possam ser futuramente realizadas. Sugere-se a intensificação das ações de educação em saúde com o intuito de conscientizar o cidadão sobre a importância do cão na dinâmica da transmissão da leishmaniose visceral e, desse modo, aumentar a adesão da população às atividades de controle preconizadas pelo programa.
