Abstract:
ABSTRACT Strains of Streptococcus mutans lacking DnaK or GroEL appear not to be isolable. To better distinguish the roles played by these chaperones/chaperonins in the physiology of S. mutans, we created a knockdown strategy to lower the levels of DnaK by over 95% in strain SM12 and the level of GroEL about 80% in strain SM13. Interestingly, GroEL levels were approximately twofold higher in SM12 than in the parent strain, but the levels of DnaK were not altered in the GroEL knockdown strain. Both SM12 and SM13 grew slower than the parent strain, had a strong tendency to aggregate in broth culture, and showed major changes in their proteomes. Compared with the wild-type strain, SM12 and SM13 had impaired biofilm-forming capacities when grown in the presence of glucose. The SM12 strain was impaired in its capacity to grow at 44°C or at pH 5.0 and was more susceptible to H2O2, whereas SM13 behaved like the wild-type strain under these conditions. Phenotypical reversions were noted for both mutants when cells were grown in continuous culture at a low pH, suggesting the occurrence of compensatory mutations. These results demonstrate that DnaK and GroEL differentially affect the expression of key virulence traits, including biofilm formation and acid tolerance, and support that these chaperones have evolved to accommodate unique roles in the context of this organism and its niche.

Abstract:
Marine actinomycetes are being explored to discover potential actinomycetes that produce antifungal compounds. In a previous study, marine actinomycetes isolates from the mangrove ecosystem were found to inhibit growth of the phytopathogenic fungi Colletotrichum siamense KA. In this study, the three of these isolates with the highest antagonistic activity—SM11, SM14, and SM15—were evaluated for their antifungal activity using antibiosis assay. The fermentation was performed in SCB:PDB medium (1:1) for 6, 9, and 12 days. The results showed that SM14 was the strongest potential isolate; it inhibited the growth of C. siamense KA on average up to 64.90% for 12 days on PDA filtrate medium. Molecular identification showed SM14 was closely related to Streptomyces sanyensis, but had differences in morphological and biochemical characteristics compared to SM11 or SM15. This indicated that the three isolates were different strains and may challenge further research on identifying and analyzing their antifungal compounds.

Abstract:
This paper reports the first direct molecular evidence that X chromosome loss during determination of male aphids (XO) is random. Clonal and sexual females, and males, of the species Sitobion near fragariae were screened using three polymorphic microsatellite loci. Two loci, Sm10 and Sm17, showed the same heterozygous genotypes in all three aphid morphs. The third, Sm11, was heterozygous for the same two alleles in clonal and sexual females, but of the 25 males screened 11 showed the ‘160’ allele and 14 showed the ‘156’ allele. This result indicates X-linkage of locus Sm11, with random loss of the X chromosome during the formation of male embryos. The possible implications of this result are discussed with respect to aphid sex determination, recombination and chromosome evolution.

Smooth muscle myosin heavy chain (SMHC) isoforms, SM1 and SM2, are the products of alternative splicing from a single gene (P. Babij and M. Periasamy. J. Mol. Biol. 210: 673-679, 1989). We have previously shown that this splicing occurs at the 3'-end of the mRNA, resulting in proteins that differ at the carboxyterminal (R. Nagai, M. Kuro-o, P. Babij, and M. Periasamy. J. Biol. Chem. 264: 9734-9737, 1989). In the present study we demonstrate that additional SMHC isoform diversity occurs in the globular head region by isolating and characterizing two distinct rat SMHC cDNA (SMHC-11 = SM1B and SMHC-5 = SM1A). Sequence comparison of the two clones reveals that they are completely identical in their coding regions, except at the region encoding the 25/50 kDa junction of the myosin head, where the SM1B isoform contains an additional seven amino acids. This divergent region is located adjacent to the Mg(2+)-ATPase site, and differences in this region may be of functional importance. Ribonuclease protection analysis demonstrates that the corresponding SM1B and SM1A mRNA messages are coexpressed in all smooth muscle tissues; however, the proportion of the two mRNA present differs significantly between tissues. The SM1A-type mRNA predominates in most smooth muscle tissues, with the exception of intestine and urinary bladder, which contain greater proportions of the SM1B message. The differential distribution of these two isoforms may provide important clues toward understanding differences in smooth muscle contractile properties.

Two Schistosoma mansoni cDNA clones 30S and 1H were identified by immunoscreening of sporocyst lambdagt11 library and by random sequencing of clones from lambdaZap libraries, respectively. Clone 30S was one of 30 clones identified by an antibody raised against tegument of 3-h schistosomules. The clone was found to encode an 81 amino-acid protein fragment. It was expressed in Escherichia coli as a fusion protein of calculated molecular mass of about 35 kDa with C-terminus of Schistosoma japonicum glutathione-S-transferase (Sj26; about 26 kDa). The recombinant fusion protein was specifically recognized by serum of rabbits immunized with irradiated cercariae. Clone 1H is one of 76 expressed sequence tags derived from an adult worm library. It encodes the complete sequence of a tegumental membrane protein, Sm13. The 104 amino-acid open reading frame encodes a protein with a calculated molecular mass of about 11.9 kDa. Clone 1H was expressed in E. coli as an insoluble fusion protein with Sj26 of about 40 kDa. In Western blots, the fusion protein was recognized by serum from rabbits vaccinated with irradiated cercariae but not by preimmune rabbit sera. The cloning, characterization and expression of those proteins are therefore potentially usefull for vaccine development.

Water scarcity is abiotic stress that is becoming more prevalent as a result of human activities, posing a threat to agriculture and food security. Recently, endophytic bacteria have been proven to reduce drought stress and increase crop productivity. Here, we explored the efficacy of seed endophytic bacteria in maize crops under water deficit conditions. For this purpose, twenty-seven endophytic bacteria have been isolated from three distinct maize cultivars seeds (Malka 2016, Sahiwal Gold and Gohar-19) and evaluated for desiccation tolerance of −0.18, −0.491, and −1.025 MPa induced by polyethylene glycol (PEG) 6000. The nine isolates were chosen on the basis of desiccation tolerance and evaluated for maize growth promotion and antioxidant activity under normal and drought conditions. Results showed that drought stress significantly decreased the growth of maize seedlings. However, isolates SM1, SM4, SM19, and SM23 significantly improved the root and shoot length, plant biomass, leaf area, proline content, sugar, and protein content under normal and drought conditions. Antioxidant enzymes were significantly decreased at p-value < 0.05 with inoculation of seed endophytic bacteria under drought conditions. However, further experiments of seed endophytic bacteria (SM1, SM4, SM19, and SM23) should be conducted to validate results.

ABSTRACT Bacillus anthracis spores are enclosed by an exosporium comprised of a basal layer and an external hair-like nap. The filaments of the nap are composed of trimers of the collagen-like glycoprotein BclA. The attachment of essentially all BclA trimers to the exosporium requires the basal layer protein BxpB, and both proteins are included in stable high-molecular-mass exosporium complexes. BclA contains a proteolytically processed 38-residue amino-terminal domain (NTD) that is essential for basal-layer attachment. In this report, we identify three NTD submotifs (SM1a, SM1b, and SM2, located within residues 21 to 33) that are important for BclA attachment and demonstrate that residue A20, the amino-terminal residue of processed BclA, is not required for attachment. We show that the shortest NTD of BclA—or of a recombinant protein—sufficient for high-level basal-layer attachment is a 10-residue motif consisting of an initiating methionine, an apparently arbitrary second residue, SM1a or SM1b, and SM2. We also demonstrate that cleavage of the BclA NTD is necessary for efficient attachment to the basal layer and that the site of cleavage is somewhat flexible, at least in certain mutant NTDs. Finally, we propose a mechanism for BclA attachment and discuss the possibility that analogous mechanisms are involved in the attachment of many different collagen-like proteins of B. anthracis and closely related Bacillus species.

Abstract:
As a solid waste, the amount of residual sludge produced by the municipal wastewater treatment process is escalating. How to dispose it properly is attracting much attention in society. Herein, solidifying residual sludge using magnesium oxychloride cement (MOC) is promising for converting it into building materials. Various factors of mass ratio (RW/S) of liquid to solid, molar ratio (Rn) of MgO to MgCl2 in MOC, mass ratio (Rm) of residual sludge to MOC, the mass concentration of Na2SiO3 (DNa2SiO3), and dosage of fly ash (DF) influenced the unconfined compression strength (RC) of the as–obtained MOC–solidified residual sludge, and it was characterized using SEM and XRD analysis. The results show that the value of RC for MOC–residual sludge solidified blocks increased initially and then decreased as Rn and Rm increased, respectively, for 60–day curing. At 10–day curing, equilibrium RC was reached at all RW/S values except 1.38, and at 60–day curing, RC decreased with RW/S increasing. The maximum RC of 60 days of 20.90 MPa was obtained at RW/S = 0.90, Rn = 5.0, and Rm = 1.00. Furthermore, adding Na2SiO3 or fly ash in the solidifying process could improve RC. The water resistance test showed that SM13 and NF5 samples exhibited good alkaline resistance after immersion for 7 and 14 days in an aqueous solution with pH = 7.0–11.0. The water resistance of MOC–residual sludge solidified blocks decreased with increase in immersion duration in aqueous solutions. The fly ash could also help improve water resistance of MOC–solidified residual sludge in neutral and basic aqueous solutions. This work provides an important theoretical basis and possibility for the efficient disposal and comprehensive utilization of residual sludge through solidification/stabilization technology using MOC from the perspective of mechanics and water resistance.

In present investigation karyotype of mitotic chromosomes were determined, two species of the genus Spilanthes, namely Spilanthes acmella (L.) Murray and Spilanthes calva DC. from plants transplanted in earthen pots collected from different localities from Central Nepal in my home garden. The somatic chromosome number determined in present study were 2n=36 for both species, Spilanthes acmella (L.) Murray and Spilanthes calva DC. The chromosomes total length ranged from 0.6 to 1.6 µm in Spilanthes acmella (L.) Murray and 0.4 to 2.1µm in Spilanthes calva DC. The relative length ranged from 2.3 to 12.1 µm in the species Spilanthes calva and 2.0 to 8.5 µm in the species Spilanthes acmella. The Karyotype formula were M16+m2+sm16+st2 in Spilanthes calva DC. and M22 +sm14 in Spilanthes acmella (L.) Murray cytologically obtained by using 2% aceto-orcein. The karyotype of the two species shows considerable variation exists in morphology of chromosome and structure. Sesuai Peraturan Pemerintah (PP) NO. 50 tahun 2012 tentang SMK3 maka semua perusahaan wajib menerapkan SMK3 termasuk perusahaan X yang mengelola terminal BBM. Untuk itu perusahaan X patuh pada regulasi yaitu menerapkan SMK3. Tujuan penelitian ini adalah untuk melihat seberapa jauh perusaha an x dalam menerapkan SMK3 telah sesuai dengan PP No. 50 diatas. Metode penelitian dilaksanakan secara deskriptif kualitatif. Hasil penelitian menunjukkan bahwa perusahaan X telah menerapkan sepenuhnya SMK3 dengan konsisten sesuai kaidah SMK3 yang ada di PP Nomor 50. Kesimpulan bahwa perusahaan X sudah menerapkan SMK3 dan ditunjukkan dengan bukti nyata mendapat hasil gold berdasarkan penilaian korporat perusahaan X.

Research data collected by distributing to 28 respondents in PT. PULAUINTAN. Experts are the project managers. The results of the study, personal compliance analysis method resulted in high adherence or 64.58%. Furthermore, the performance analysis of SMK3 implementation, the analysis result shows the performance value of SMK3 is or 59%. Then with the results of high compliance analysis and performance is then there is need for improvement action performance of SMK3 implementation to improve the implementation of SMK3. PP no.50 tahun 2012 menerangkan bahwa SMK3 wajib dilaksanakan untuk seluruh industri maupun jasa. The Himana Condotel merupakan salah satu proyek konstruksi gedung dengan resiko kecelakaan yang cukup tinggi, maka dilakukan analisis terhadap tingkat penerapan SMK3. Analisis yang dilakukan adalah deskriptif observasiona dengan subjek sample dari pihak manajemen dan pihak pekerja. Analisis uji beda menggunakan Mann Whitney U test dan studi literature SMK3 yang tertuang pada PP no. 50 tahun 2012 untuk mengetahui sejauh mana timgkat penerapan SMK3 kontraktor dalam merencanakan, menerapkan serta memeriksa SMK3 pada proyek The Himana Condotel. Hasil dari penelitian ini adalah tingkat penerapan SMK3 pada proyek The Himana Condotel yaitu merencanakan SMK3 dengan rata-rata nilai dari subjek manajemen 90,94 ± 3,431 dan dari unsur subjek pekerja sebesar 87,53 ± 3,417 masuk dalam tingkat penerapan menuaskan. Menerapkan SMK3 dengan rata-rata nilai dari subjek manajemen 82,38 ± 0,754 dan dari unsur subjek pekerja sebesar 81,90 ± 2,338 masuk dalam tingkat penerapan menuaskan, dan memeriksa SMK3 dengan rata-rata nilai dari subjek manajemen 99,87 ± 0,020 dan dari unsur subjek pekerja sebesar 95,76 ± 4,987 masuk dalam tingkat penerapan memuaskan.

Abstract The Drosophila gene for snRNP SmD3 (SmD3) is contained in reverse orientation within the first intron of the Ornithine Decarboxylase Antizyme (AZ) gene. Previous studies show that two closely linked P elements cause the gutfeeling phenotype characterized by embryonic lethality and aberrant neuronal and muscle cell differentiation. However, the exact nature of the gene(s) affected in the gutfeeling phenotype remained unknown. This study shows that a series of P inserts located within the 5′-untranslated region (5′-UTR) of SmD3 or its promoter affects only the expression of SmD3. Our analysis reveals that the gutfeeling phenotype associated with P elements inserted in the 5′-UTR of SmD3 results from amorphic or strongly hypomorphic mutations. In contrast, P inserts in the SmD3 promoter region reduce the expression of SmD3 without abolishing it and produce larval lethality with overgrown imaginal discs, brain hemispheres, and hematopoietic organs. The lethality of these mutations could be rescued by an SmD3+ transgene. Finally, inactivation of AZ was obtained by complementing with SmD3+ the deficiency Df(2R)guf lex47 that uncovers both SmD3 and AZ. Interestingly, AZ inactivation causes a new phenotype characterized by late larval lethality and atrophy of the brain, imaginal discs, hematopoietic organs, and salivary glands.

ABSTRACT The Sm16/SmSLP/SmSPO-1 (Sm16) protein is secreted by the parasite Schistosoma mansoni during skin penetration and has been ascribed immunosuppressive activities. Here we describe the strategy behind the design of a modified Sm16 protein with a decreased aggregation propensity, thus facilitating the expression and purification of an Sm16 protein that is soluble in physiological buffers. The Stokes radii and sedimentation coefficients of recombinant and native proteins indicate that Sm16 is an approximately nine-subunit oligomer. Analysis of truncated Sm16 derivatives showed that both oligomerization and binding to the plasma membrane of human cells depend on multiple C-terminal regions. For analysis of immunomodulatory activities, Sm16 was expressed in Pichia pastoris to facilitate the preparation of a pyrogen/endotoxin-free purified protein. Recombinant Sm16 was found to have no effect on T-lymphocyte activation, cell proliferation, or the basal level of cytokine production by whole human blood or monocytic cells. However, Sm16 exerts potent inhibition of the cytokine response to the Toll-like receptor (TLR) ligands lipopolysaccharide (LPS) and poly(I:C) while being less efficient at inhibiting the response to the TLR ligand peptidoglycan or a synthetic lipopeptide. Since Sm16 specifically inhibits the degradation of the IRAK1 signaling protein in LPS-stimulated monocytes, our findings indicate that inhibition is exerted proximal to the TLR complex.

Pada program ini dilakukan survei lokasi, wawancara sebelum dan sesudah seminar. Materi seminar adalah penjelasan mengenai SMK3, penerapan SMK3 dalam masa pandemi di industri kecil dan menengah. This research aimed to know Non-Specific LBP prevalence in housekeeping employees in Badung regency. Design of this research is cross-sectional descriptive with quantitative and qualitative data collection techniques. The variables measured were SMK3, Non-Specific LBP, Age, and Working Period. The results showed that housekeeping employees at hotels applying SMK3 had a tendency to avoid Non-Specific LBP complaints compared to those not applying SMK3. ABSTRACT The Sm14 antigen of Schistosoma mansoni was cloned and expressed in Mycobacterium bovis BCG as a fusion with the Mycobacterium fortuitum β-lactamase protein under the control of its promoter, pBlaF*; the protein was localized in the bacterial cell wall. The rBCG-Sm14 strain was shown to be relatively stable in cultured murine and bovine monocytes in terms of infectivity, bacterial persistence, and plasmid stability. The immunization of mice with rBCG-Sm14 showed no induction of anti-Sm14 antibodies; however, splenocytes of immunized mice released increased levels of gamma interferon upon stimulation with recombinant Sm14 (rSm14), indicating an induction of a Th1-predominant cellular response against Sm14. Mice immunized with one or two doses of rBCG-Sm14 and challenged with live S. mansoni cercaria showed a 48% reduction in worm burden, which was comparable to that obtained by immunization with three doses of rSm14 purified from Escherichia coli. The data presented here further enhance the status of Sm14 as a promising candidate antigen for the control of schistosomiasis and indicate that a one-dose regimen of rBCG-Sm14 could be considered a convenient means to overcome many of the practical problems associated with the successful implementation of a multiple-dose vaccine schedule in developing countries.

We have investigated epitaxial superconducting SmBa2Cu3O7 (Sm123) films grown by pulsed-laser deposition on single-crystal SrTiO3 substrates. The deposition temperature plays an important role in determining the superconducting properties of Sm123 films. The superconducting transition temperature increases with the deposition temperature whereas the transition width decreases at deposition temperatures in the range of 700–875 °C. A Sm123 film deposited at 850 °C exhibits a transition temperature above 93 K with a transition width less than 0.5 K. Even though Sm123 films exhibit a higher transition temperature than Yba2Cu3O7 (Y123), the Sm123 shows lower critical current density at liquid-nitrogen temperature. The nominal critical current density of Sm123 film is less than 1 MA/cm2 at 75.4 K. Nevertheless, the Sm123 films have less anisotropy and stronger pinning characteristics compared to Y123. They are also much smoother with fewer particulates, as revealed by scanning electron microscopy.

Berbagai permasalahan kecelakaan dan penyakit akibat kerja di perusahaan merupakan alasan yang kuat diperlukannya penerapan Sistem Manajemen Kesehatan dan Keselamatan Kerja (SMK3) yang benar. Namun dilain sisi banyak perusahaan masih belum menerapkan SMK3. Tidak adanya Sistem Manajemen K3 mengakibatkan belum terkelolanya kegiatan K3 secara benar dan masih rendahnya kesadaran karyawan untuk bekerja secara aman dan sesuai prosedur. Data kecelakaan kerja di CV. Putra Abadi Langkat (PAL) masih belum terdokumentasi dengan baik dan di temukan kecelakaan seperti tertimpa, terjepit, dan terpotong. Oleh karena itu Pengabdian Masyarakat ini diharapkan dapat membantu CV. PAL dalam menerapkan SMK3. CV. PAL diharapkan berkomitmen untuk menjaga standar Keselamatan dan Kesehatan Kerja dalam setiap aktivitasnya. Untuk itu perusahaan akan menerapkan SMK3 yang terintegrasi dengan manajemen perusahaan. Adapun penerapan Sistem Manajemen K3 tersebut melalui tahapan sebagai berikut : kebijakan dan kepemimpinan, perencanaan, implementasi dan operasi, pemeriksaan, serta tinjauan manajemen. Abstract Recurrent mutations in SMC3, encoding a cohesin subunit, have been identified in acute myeloid leukemia (AML) and other myeloid malignancies by our group and others. SMC3 mutations are heterozygous in AML patients. Missense, nonsense, and splice site mutations have been observed across all domains of SMC3. Given the breath of mutations, it is important to determine whether these represent recurrent loss-of-function mechanisms, or if some might have dominant negative effects. To determine the impact of Smc3 deletion on hematopoiesis, we studied both Smc3 haploinsufficient and Smc3 deficient mice as models of loss-of function and dominant negative phenotypes respectively. The Smc3 haploinsufficient mouse model has a lacZneo gene trap inserted in intron 3-4 of Smc3, which leads to a premature transcription stop and therefore produces a truncated and dysfunctional protein. The homozygous Smc3trap allele is embryonic lethal. The Smc3trap/+mice have an early growth defect, although their body weight catches up to wild type mice after 6 weeks of age. We found no difference in spleen weights, peripheral blood counts, and bone marrow (BM) compositions between Smc3trap/+ and wild type mice. The Smc3trap/+ BM cells formed similar number of colonies as wild type cells when plated in methylcellulose in vitro and lost self-renewal capabilities after replating for two weeks. Competitive repopulation assay in vivo showed neither advantage nor disadvantage for the Smc3trap/+BM cells (n=10). Thus, Smc3trap/+BM cells have normal colony forming capacity in vitro and normal homeostatic feedback in vivo. Further, we generated Smc3 conditionally deficient mice by removing the gene-trap cassette, which retains the loxP sites flanking exon 4 (Smc3fl), and crossing these mice with either Vav1-Cre+/- or ERT2-Cre+/- to delete the allele (Smc3fl/+/Vav1-Cre+/- is constitutively haploinsufficient in hematopoietic cells, whereas Smc3fl/+/ERT2-Cre+/-is only haploinsufficient when induced with tamoxifen). We characterized both models by serial replating assays, flow cytometry assays for hematopoietic stem/progenitor cells (HSPCs), and BM lineage in vitro and found no difference in these mice compared to the Smc3fl/+control. In contrast to the Smc3fl/+/Mx1-Cre+/- mice (Viny et al. JEM 212 (11): 1819-1832), we observed a significant competitive disadvantage for the Smc3fl/+/ERT2-Cre+/-BM cells (p<0.0001, n=10), most pronounced in Gr1+ myeloid cells in vivo (p<0.0001), implying Smc3 haploinsufficiency alters hematopoiesis in those mice in vivo. We characterized the effects of homozygous Smc3 loss on hematopoiesis in the inducible Smc3fl/fl/ERT2-Cre+/- mice by treating mice with tamoxifen at 6 weeks of age (Smc3fl/fl/Vav1-Cre+/- is embryonic lethal). Deletion of Smc3 led to rapid bone marrow failure and 100% lethality with a median survival of 8 days (n=4, 2 independent experiments). At the time of death, we observed severe reduction in the sizes of spleen (Sp) and thymus (Thy), in total number of BM, Sp, and Thy cells, and in white blood counts, lymphocytes, monocytes, and platelets. The Smc3 deficient BM cells had decreased levels of Smc3 by Western blot. The impact of Smc3 deletion on HSPC functions in vivo was assessed by a competitive repopulation assay of Smc3fl/fl/ERT2-Cre+/-BM cells (p<0.0001, n=10). Recipient mice were treated with tamoxifen after 6-week engraftment. After tamoxifen-mediated deletion, Smc3 deficient cells were rapidly outcompeted in vivo, indicating complete loss of HSPC functions. Collectively, these results suggest that Smc3 is necessary for normal hematopoiesis and for HSPC functions. The AML-associated SMC3 mutations are therefore unlikely to be dominant negative because the complete loss of Smc3 is incompatible with hematopoiesis. Berdasarkan Peraturan Pemerintah Republik Indonesia Nomor 50 Tahun 2012, Sistem Manajemen Keselamatan dan Kesehatan Kerja (SMK3) adalah bagian dari sistem manajemen perusahaan secara keseluruhan dalam rangka pengendalian risiko yang berkaitan dengan kegiatan kerja guna terciptanya tempat kerja yang aman, efisien dan produktif. PT. PLN (Persero) Sektor Mahakam sebagai salah satu perusahaan dengan potensi bahaya dan resiko kecelakaan yang tinggi telah memasukan SMK3 ke dalam sistem manajemen perusahaan meskipun sejauh ini belum ada peninjauan ulang dan pengukuran tentang dukungan tenaga kerja terhadap penerapan SMK3. Tujuan penelitian ini untuk mengetahui faktor yang berhubungan dengan persepsi tenaga kerja tentang SMK3 pada tenaga kerja PT. PLN (Persero) Sektor Mahakam Unit PLTGU Tanjung Batu. Desain penelitian yang digunakan adalah survei analitik dengan metode kuantitatif dan pendekatan cross sectional dengan besar sampel sebanyak 47 tenaga kerja. Penelitian ini menggunakan uji korelasi chi square dengan CI=95%. Hasil penelitian menunjukkan bahwa ada hubungan pengetahuan (p=0.029), sikap (p=0.002), dan pelatihan K3 (p=0.028) dengan persepsi tentang SMK3, sedangkan variabel pengawasan (p=0.109) tidak berhubungan dengan persepsi tenaga kerja tentang penerapan SMK3. Sikap menjadi faktor yang paling berhubungan dengan persepsi tenaga kerja tentang SMK3.

Spliceosome assembly during pre-mRNA splicing requires the correct positioning of the U1, U2, U4/U6, and U5 small nuclear ribonucleoprotein particles (snRNPs) on the precursor mRNA. The structure and integrity of these snRNPs are maintained in part by the association of the snRNAs with core snRNP (Sm) proteins. The Sm proteins also play a pivotal role in metazoan snRNP biogenesis. We have characterized a Saccharomyces cerevisiae gene, SMD3, that encodes the core snRNP protein Smd3. The Smd3 protein is required for pre-mRNA splicing in vivo. Depletion of this protein from yeast cells affects the levels of U snRNAs and their cap modification, indicating that Smd3 is required for snRNP biogenesis. Smd3 is structurally and functionally distinct from the previously described yeast core polypeptide Smd1. Although Smd3 and Smd1 are both associated with the spliceosomal snRNPs, overexpression of one cannot compensate for the loss of the other. Thus, these two proteins have distinct functions. A pool of Smd3 exists in the yeast cytoplasm. This is consistent with the possibility that snRNP assembly in S. cerevisiae, as in metazoans, is initiated in the cytoplasm from a pool of RNA-free core snRNP protein complexes.

Penelitian ini bertujuan untuk mengetahui penerapan SMK3 pada beberapa daerah dan perusahaan-perusahaan di Provinsi Papua serta kendala apa saja yang terjadi dalam penerapan SMK3 itu sendiri. Penelitian ini menggunakan penelitian kuantitatif dengan dengan menggunakan kuesioner untuk melihat penerapan Sistem Manajemen Keselamatan dan Kesehatan Kerja (SMK3) pada perusahaan-perusahaan di Provinsi Papua dengan bantuan program komputer Microsoft Exel dalam pengolahan data. Hasil pegolahan data diketahui bahwa penerapan pada beberapa kabupaten di provinsi Papua seperti Kota Jayapura 71% dan Kabupaten Nabire 69% tingkat penilaian penerapan SMK3 baik. Sedangkan untuk Kabupaten Mimika 91% dan Kabupaten Merauke 86% termasuk kategori tingkat penilaian penerapan Memuaskan. Demikian halnya pada beberapa perusahaan di provinsi Papua, penerapan SMK3 termasuk kategori tingkat penilaian penerapan baik. Pada perseroan terbatas (PT) adalah 71% dan persekutuan komanditer (CV) adalah 79%. Namun masih terdapat beberapa kendala dalam penerapan SMK3 di provinsi Papua di antaranya adalah masih banyak pekerja yang mengganggap bahwa K3 kurang penting dan kurangnya personil/petugas/ahli K3 dalam perusahaan-perusahaan di provinsi Papua.

Abstract Hematopoietic stem cells (HSCs) are undifferentiated, self-renewing, pluripotent cells that have the capacity to differentiate into all mature lineage-specific cells in adult blood. Adult HSCs can remain in a quiescent state for a prolonged time, and quiescence is a fundamental characteristic of HSCs in adult bone marrow. Thus, the cell cycle must be precisely regulated. Yin Yang 1 (YY1) is a multifunctional transcription factor and Polycomb Group Protein (PcG) that is important for embryonic development, adult hematopoiesis, cell proliferation and maintaining higher-order chromosomal structure. We have generated YY1 conditional knockout mice (Yy1f/f Mx1-Cre) and showed that Yy1 deficient HSCs fail to self-renewal and had disrupted HSCs quiescence. Stem cell factor (SCF)/c-Kit signaling, a critical regulatory pathway in HSC development, is significantly downregulated in Yy1-/- HSCs. Interestingly, YY1 regulation of HSCs self-renewal and quiescence is independent on its PcG domain/function. Instead, YY1 occupied at Smc3 promoter area and repressed Smc3 expression. In Yy1-/-HSCs, Smc3 expression was upregulated. SMC3 is a core component of cohesin protein complex and plays critical roles in HSC self-renewal, myeloid differentiation and leukemogenesis. To further dissect the underlying mechanisms by which YY1 regulates SMC3 expression in HSCs, we have generated conditional knockout mice with YY1 homozygous deletion and SMC3 heterozygous deletion (Yy1f/f Smc3f/+Mx1-Cre). In Yy1-/- Smc3+/- bone marrow cells, SMC3 expression was normalized to the wild-type level. In adult bone marrow cells, YY1 physically interacted with cohesin complex proteins through its zinc finger domain. By analyzing the YY1, SMC1A and SMC3 ChIP-Seq database, our study showed that YY1 and cohesin co-occupied at promoter areas of genes that are critical for cell metabolism. Evidence from previous study showed that impaired metabolism, including increased reactive oxygen species (ROS) and decreased mitochondrial function, can cause defect in stem cell self-renewal and quiescence. In Yy1-/- Smc3+/-HSCs, cell quiescence was restored although HSC self-renewal was still impaired, indicates that YY1 and SMC3 may control HSC cell quiescence via regulating genes critical for cell metabolism. Our study identified YY1 as the first transcription factor that regulates expression of cohesin complex component SMC3. We are currently further dissecting underlying mechanisms and functional significances of metabolic pathways regulated by YY1-SMC3 axis in HSCs. The purpose of this research is to explore the implementation, evaluation process, impact and expectation of SMK3 in Primary Health Care Bendilwungu Tulungagung Method. Based on the type of this research is a qualitative research to describe the implementation of SMK3 at Primary Health Care Bendilwungur. Another supporting instrument uses an instrument in the form of a list of questions containing semi-structured questions. The research informants numbered ten and four. Research Triangulation at the Puskesmas. Other tools used are friends, field notes, cellphones, then documented in the form of transcripts (field notes). Data processing was carried out by means of content analysis. Furthermore, it is reported and presented in a descriptive description in relation to the theory. Results. The results of the study indicate that the application of SMK3 in the Primary Health Care Bendilwungu has been running from the end of 2018 as indicated by the existence of the SK Head of the Puskesmas as a guideline and legal umbrella in implementing the implementation of SMK3 in the Puskesmas. In the process of implementing and implementing SMK3 in the Puskesmas there are basic obstacles namely that there is no calm of K3 and SMK3 professionals and the joining of the SMK3 Implementation Unit with Patient Safety and the lack of knowledge and understanding of K3 and SMK3 to several Puskesmas employees. Conclusion. The Primary Health Care Bendilwungu needs to improve its discrepancies in the application of SMK3 and provide training and guidance on K3 and SMK3 to employees so as to always improve the OHS work standard supervision at the Primary Health Care Bendilwungu and make K3 a work culture.

Occupational safety and health (OSH) issues in the Indonesian forestry sector have not been addressed properly even though activities in this sector pose a high risk of OSH disorders. The standard instrument (applies on a national scale) to ensure OSH protection in each business units is the Sistem Manajemen K3 (SMK3, Government Regulation Numbered 50 Year 2012). At present, not many forest management units in Indonesia are willing to implement integrated SMK3, because of the assumption that SMK3 is a cost-centered activity. This study aims to present an overview of SMK3 implementation cost (for 64, 122, and 166 criteria) in a Forest Management Unit (FMU) X, as well as analyzing financial capacity of the FMU X to allocate sufficient funds for the SMK3 implementation. The results showed that the Acacia mangium plantation in the FMU X is a feasible business, indicated by the Net Present Value (NPV) of IDR377,690,545, Benefit-Cost Ratio (BCR) 3.96, and Internal Rate of Return (IRR) 25.02%. Further analysis shows that FMU X has good financial ability to support any cost required in implementating SMK3. It is the best for the FMU X to implement 166 criteria (advanced level) of SMK3 (at a cost of IDR704,598/ha/year, NPV of IDR376,393,262, BCR 3.92, and IRR 24.75%).

ABSTRACT Yeast Smt3 and its vertebrate homolog SUMO-1 are ubiquitin-like proteins (Ubls) that are reversibly ligated to other proteins. LikeSMT3, SMT4 was first isolated as a high-copy-number suppressor of a defective centromere-binding protein. We show here that SMT4 encodes an Smt3-deconjugating enzyme, Ulp2. In cells lacking Ulp2, specific Smt3-protein conjugates accumulate, and the conjugate pattern is distinct from that observed in a ulp1ts strain, which is defective for a distantly related Smt3-specific protease, Ulp1. The ulp2Δ mutant exhibits a pleiotropic phenotype that includes temperature-sensitive growth, abnormal cell morphology, decreased plasmid and chromosome stability, and a severe sporulation defect. The mutant is also hypersensitive to DNA-damaging agents, hydroxyurea, and benomyl. Although cell cycle checkpoint arrest in response to DNA damage, replication inhibition, or spindle defects occurs with normal kinetics, recovery from arrest is impaired. Surprisingly, either introduction of aulp1ts mutation or overproduction of catalytically inactive Ulp1 can substantially overcome theulp2Δ defects. Inactivation of Ulp2 also suppresses several ulp1ts defects, and the double mutant accumulates far fewer Smt3-protein conjugates than either single mutant. Our data suggest the existence of a feedback mechanism that limits Smt3-protein ligation when Smt3 deconjugation by both Ulp1 and Ulp2 is compromised, allowing a partial recovery of cell function.

ABSTRACT A variety of transcription factors are targets for conjugation to the ubiquitin-like protein Smt3 (also called SUMO). While many such factors exhibit enhanced activity under conditions that favor conjugation, the mechanisms behind this enhancement are largely unknown. We previously showed that the Drosophila melanogaster rel family factor, Dorsal, is a substrate for Smt3 conjugation. The conjugation machinery was found to enhance Dorsal activity at least in part by counteracting the Cactus-mediated inhibition of Dorsal nuclear localization. In this report, we show that Smt3 conjugation occurs at a single site in Dorsal (lysine 382), requires just the Smt3-activating and -conjugating enzymes, and is reversed by the deconjugating enzyme Ulp1. Mutagenesis of the acceptor lysine eliminates the response of Dorsal to the conjugation machinery and results in enhanced levels of synergistic transcriptional activation. Thus, in addition to controlling Dorsal localization, Smt3 also appears to regulate Dorsal-mediated activation, perhaps by modulating an interaction with a negatively acting nuclear factor. Finally, since Dorsal contributes to innate immunity, we examined the role of Smt3 conjugation in the immune response. We find that the conjugation machinery is required for lipopolysaccharide-induced expression of antimicrobial peptides in cultured cells and larvae, suggesting that Smt3 regulates Dorsal function in vivo.

For that PT. Mujur Lestari makes implementation and Monitoring to minimize the risk of work accidents for employees. The sample used in this study were employees of PT. Mujur Lestari, totaling 81 people. The data collection method used a questionnaire while the analytical method used was multivariate analysis. The results showed that there was an influence between knowledge on the Occupational Health and Safety Management System (SMK3) at PT. Mujur Lestari with a tcount greater than ttable, the effect of applying the Occupational Health and Safety Management System (SMK3) with a tcount greater than ttable, the effect of Monitoring on the Occupational Health and Safety Management System (SMK3) with a greater tcount from ttable and there is no relationship between the implementation and Monitoring of the Occupational Safety and Health Management System (SMK3). The conclusion shows that the knowledge variable has a positive and significant effect on the Occupational Safety and Health Management System (SMK3), the application variable has a positive and significant impact on the Occupational Safety and Health Management System (SMK3) and the Monitoring variable has a positive and significant effect on the Occupational Safety and Health Management System. Hasil peninjauan ini menunjukan bahwa Sistem Manajemen Keselamatan dan Kesehatan Kerja (SMK3) pada pekerjaan proyek tersebut tidak berjalan sebagaimana mestinya, baik dari kebijakannya, organisasi, perencanaan dan pengaplikasian, pengukuran kinerja, peninjauan ulang, maupun evaluasinya. The amount of risk from various potential hazards in the hospital so that efforts are needed to control and minimize the occurrence of work accidents, one of which is by implementing SMK3. This study aims to analyze the factors that influence nurses on the implementation of the occupational health and safety management system at Langsa City Hospital. The research design used an analytic survey with a cross sectional design. The sample of this study was 82 people used was simple random sampling. Data collection methods are primary data, secondary data and tertiary data. The research data were analyzed by univariate, bivariate and multivariate using logistic regression tests. The results showed that there was an effect of knowledge on the application of SMK3 (p = 0.002), there was an effect of attitude on the application of SMK3 (p = 0.010), there was an effect of action on the application of SMK3 (p = 0.005), there was an effect of perception on the application of SMK3 (p = 0.028) ), there is an effect of infrastructure on the implementation of SMK3 (p = 0.000), there is an effect of leadership support on the implementation of SMK3 (p = 0.021) and the most dominant variable is the action variable. Based on the research results, it can be concluded that there is an influence of knowledge, attitudes, actions, perceptions, infrastructure and leadership support on the implementation of SMK3 at Langsa Hospital. The suggestion in this research is that the hospital can adopt rifaskes innovation that can help the best health services for nurses.

Abstract:
This study aims to explore the anti-bacterial and toxicity activities from a rare actinobacterium isolated from mangrove, Mempawah District, West Kalimantan. The mangrove mud sample from Mempawah district was inoculated on ISP4 agar using a pour plate method. After 4 days of incubation, a colony of suspected actinobacterium was appeared, then isolated and coded as SM1P. SM1P was characterized based on morphological and biochemical traits and identified as a genus of Streptroporangium then called Streptroporangium sp. SM1P. Streptroporangium sp. SM1P was carried out anti-bacterial assay on both ISP1 agar and ISP4 agar media using the cross-streak method for the solid-state fermentation. The result showed that Streptroporangium sp. SM1P could inhibit Streptococcus sp. and Salmonella typhi on ISP1 agar and treptococcus sp., Escherichia coli, Vibrio cholerae, Staphylococcus aureus and Salmonella typhi on ISP4 agar. Streptroporangium sp. SM1P was cultivated on ISP1 broth and extracted using ethyl acetate, then evaporated to obtain crude extract. The crude extract was used for anti-bacterial assay (well-diffusion method for liquid-state fermentation) and toxicity assay (brine shrimp lethality test). The crude extract was active against 2 of the test bacteria (Streptococcus sp. and E. coli). The best medium and state fermentation for anti-bacterial assay were ISP4 agar with the condition of solid-state fermentation. The extract SM1P prepared on ISP1 broth showed toxic activity based on LC50 (106.094 µg/mL). Therefore, Streptroporangium sp. SM1P have a potential source to explore secondary metabolites having anti-bacterial and toxicity activities.

Posttranslational modifications of proteins play critical roles in the control of cellular differentiation, development, and environmental adaptation. In particular, the covalent attachment of the small ubiquitin-like modifier, SUMO, to target proteins (sumoylation) regulates cell cycle progression, transcription, nucleocytoplasmic transport, and stress responses. Here we combine proteomic, molecular, and cellular approaches to examine the roles of sumoylation in the major fungal pathogen of humans, Candida albicans. Using an N-terminally FLAG-tagged SUMO, 31 sumoylated proteins were identified in C. albicans with roles in stress responses (e.g., Hsp60, Hsp70 family members, Hsp104), the cytoskeleton and polarized growth (e.g., Tub1, Cct7, Mlc1), secretion, and endocytosis (e.g., Lsp1, Sec24, Sec7). The output from this proteomic screen was entirely consistent with the phenotypes of C. albicans mutants in which the single SUMO-encoding locus (SMT3) was inactivated or down-regulated. C. albicans smt3/smt3 cells displayed defects in growth, morphology, cell separation, nuclear segregation, and chitin deposition, suggesting important roles for sumoylation in cell cycle control. Smt3/smt3 cells also displayed sensitivity to thermal, oxidative, and cell wall stresses as well as to the antifungal drug caspofungin. Mutation of consensus sumoylation sites in Hsp60 and Hsp104 affected the resistance of C. albicans to thermal stress. Furthermore, signaling via the cell integrity pathway was defective in C. albicans smt3/smt3 cells. These observations provide mechanistic explanations for many of the observed phenotypic effects of Smt3 inactivation upon C. albicans growth and environmental adaptation. Clearly sumoylation plays key roles in fundamental cellular processes that underpin the pathogenicity of this medically important fungus.
