Abstract:
Several peptide fragments of streptokinase (SK) were prepared by incubating SK with immobilized human plasmin (hPlm) and purified by h.p.l.c. with a reverse-phase phenyl column. The N-terminal sequences, amino acid compositions and molecular masses of these peptide fragments were determined. The SK peptide fragment of 36 kDa consisting of Ser60-Lys387 (SK-p), was the only peptide fragment that could be tightly bound to immobilized hPlm. Another three large SK peptide fragments, SK-m, SK-n and SK-o, with molecular masses of 7 kDa, 18 kDa and 30 kDa, and consisting of Ile1-Lys59, Glu148-Lys333, Ser60-Lys333 respectively, were also obtained from the supernatant of the reaction mixture. The purified SK-p had high affinity with hPlm and could activate human plasminogen (hPlg) with a kPlg one-sixth that of the native SK. SK-o had low affinity with hPlm and could also activate hPlg, although the catalytic constant was less than 1% of the native SK. SK-n, as well as SK-m, which is the N-terminal 59 amino acid peptide of the native SK, had no activator activity. However, SK-m could enhance the activator activity of both SK-o and SK-p and increase their second-order rate constants by two- and six-fold respectively. It was concluded from these studies that (1) SK-o, the Ser60-Lys333 peptide of SK, was essential for minimal SK activator activity, (2) the C-terminal peptide of SK-p, Ala334-Lys387, was essential for high affinity with hPlm, and (3) the N-terminal 59-amino-acid peptide was important in maintaining the proper conformation of SK to have its full activator activity.

Abstract:
Streptokinase (SK) is a bacterial protein used clinically as a thrombolytic agent in humans. Administration of SK causes a rapid increase in the frequency of anti-SK T cells and the titre of specific anti-SK antibodies that, on subsequent administration of SK, may neutralize the activity of the drug or elicit allergic-type reactions. By locating and modifying the immunogenic T-cell epitopes within the SK protein, it is possible that an agent with reduced immunogenicity but equal efficacy may be produced. We have investigated the T-cell epitopes within SK using nine non-overlapping, recombinant peptide fragments of SK. We investigated the proliferative T-cell response of peripheral blood mononuclear cells obtained from patients before and 6 days after administration of SK for myocardial infarction. We also examined the response of cultured anti-SK T-cell lines derived from patients 6 days after treatment with SK. Before administration of SK, peripheral blood mononuclear cells from six of nine patients showed a proliferative response to SK. The response was significantly higher 6 days after administration of SK (P = 0.0004). Cultured T-cell lines showed similar proliferative responses to clinical-grade SK and recombinant SK. Marked differences in T-cell responses were apparent in response to each recombinant SK fragment (P = 0.04). The mean proliferative response exceeded background to only two peptides, peptide 2 (P = 0.04) and peptide 3 (P = 0.009). Peptide 3, representing amino acids 100–150 of mature SK, was recognized preferentially in the majority of assays. Marked variation in the T-cell response to SK following treatment with this agent was observed between subjects. Despite these differences, peptides 2 and 3 induced T-cell proliferation at a level significantly above background in the majority of subjects. These epitopes may represent a region of enhanced immunogenicity within SK.

SummaryChimeric 59D8-SK was designed to confer fibrin-selectivity to streptokinase by fusion of the Fab fragment of anti-fibrin antibody 59D8 to the N-terminus of streptokinase (SK: Ile1-Lys414). It was expressed in a mouse hybridoma cell line and purified by affinity chromatography on a 59D8-antigen column. Chimeric 59D8-SK is a disulfide-linked heterodimer composed of an antibody light chain (Mr 27,000) and a N-glycosylated chimeric heavy chain (Mr 90,000). The fibrin targeting by 59D8 increased plasma clot lysis by 2-fold, but connecting 59D8 to SK has provided 59D8-SK several unique properties: (i) 59D8-SK activated human Glu-plasminogen with a significant lag period that coincided with limited proteolysis of 59D8-SK similar to that observed for wild-type SK. In a kinetic study, both gave very similar kinetic parameters for the activation of Glu-plasminogen even though 59D8-SK was N-glycosylated in its SK portion; (ii) 59D8-SK was relatively inactive in human plasma, compared to SK, but it became activated in the presence of clots; (iii) 59D8-SK lysed clots slowly but completely whereas SK lysed clots rapidly but incompletely. Even though the mechanism behind these new properties is not fully understood, they are characteristics of a second-generation plasminogen activator.

Abstract:
Abstract The catabolism of streptokinase (SK) and polyethylene glycol derivatives of SK (PEG-SK) were studied in mice. The clearance and catabolism of SK:plasmin (SK:Pm) and PEG-SK:Pm activator complexes were also investigated. Native 125I-SK cleared rapidly (t1/2 = 15 minutes) from the circulation, with the majority of the ligand accumulating in the liver and gastrointestinal (GI) tract and a substantial fraction also localizing in the kidneys. SK, which was removed from the plasma by the liver, was secreted into bile and then the GI tract. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) demonstrated that 125I-SK recovered from liver and bile was homogeneous and of the same molecular weight (mol wt approximately 50,200) as native SK. PEG-125I-SK cleared slowly (t1/2 greater than 200 minutes), with more than 80% of the preparation localizing in liver and GI tract. The PEG-125I-SK secreted into the bile was also intact. The bile containing 125I-SK was incubated with stoichiometric amounts of plasminogen and electrophoresed under nondenaturing conditions. This study demonstrated that the secreted SK was able to form SK:Pg complexes. SDS-PAGE also showed activation of 125I-Pg that was incubated with recovered bile containing the SK. 125I-SK:Pm catabolism was also studied. In these experiments, the mol wt approximately 42,000 fragment obtained when SK is cleaved by plasmin was found in the bile. This fragment of 125I-SK was not recovered as part of a complex with plasmin, consistent with our previous observations that catabolism of SK:Pm involves transfer of the plasmin to plasma proteinase inhibitors while SK is catabolized independently. By contrast, when PEG-125I-SK:Pm was injected into mice, only intact PEG-125I-SK was found in the bile, consistent with our previous observations that the PEG derivatization blocks its degradation by plasmin.

Resumo: Os sk-splines são uma generalização natural dos splines polinomiais, os quais foram introduzidos e tiveram sua teoria básica desenvolvida por Alexander Kushpel nos anos de 1983-1985. Estas funções são importantes em várias aplicações e seu espaço é gerado por translações discretas de uma única função núcleo. Neste trabalho, estudamos condições necessárias e suficientes para a existência e unicidade de sk-splines interpolantes de funções periódicas. Além disso, estudamos a aproximação de funções de determinadas classes por sk-splines nos espaços Lp. Como aplicação estudamos a aproximação de funções infinitamente diferenciáveis e finitamente diferenciáveis por sk- splines
Abstract: The sk-splines are a natural generalization of polynomial splines. They were introduced and their basic theory developed by Alexander Kushpel between 1983 and 1985. These functions are important in many applications and the space of sk-splines is the linear span of shifts of a single kernel K. In this work, we study necessary and sufficient conditions for the existence and uniqueness of sk-splines interpolants of periodic functions. Furthermore, we study the approximation in several classes of functions by sk-splines in the Lp spaces. As an application we study the approximation of infinitely and finitely differentiable functions by sk-splines

The work is divided into theoretical and practical part. The theoretical part contains a brief look on the Bankruptcy and Restructuring Act in Slovakia, with the main focus being on the restructuring process. The practical part is divided into two parts, first one describes restructuring of Váhostav - SK, a. s. The second part contains a critical view on the Slovak Bankruptcy and Restructuring Act, which is compared to the Czech Insolvency Act, where the main focus is still on the process of reorganization.

Abstract:
The SK 60 Aircraft has been a workhorse for the Air Force’s training of future pilots since the late 1960s. The aging SK60 aircraft is going out of style, and the system has merely a few years of total running time left. A replacement for the SK 60 needs to be implemented in the coming years, and the Armes Forces Flying School has been commissioned to conduct flight tests on suitable replacement systems. This study will conduct a system analysis of two aircrafts mentioned in the Flying Schools report. The systems are mainly valued on their suitability as a replacement for the SK 60 from a training perspective. The systems are also valued based on the possibility of expanding the military utility of each system such as their ability to carry out attack operations. Based on the sample the Pilatus Aircraft PC -21 and BAE Systems Hawk MK 120 aircrafts were chosen for evaluation. The two planes are different yet comparable which makes them suitable for comparison in this study. Hawk MK 120 was deemed as the most suitable replacement for the SK 60. The MK 120 appeared as slightly more favorable than the PC-21 after valuation and appraisal, both from an educational point of view but also with regard to its ability for extended military purposes.

The content of this diploma thesis is an urban study with the architectural details of the former Bata Industrial Park in Partizánske, and a particular conversion program for the building. My goal was to find a suitable concept for converting building no. 61 with respect to the city, to the people living in it, and to reintegrate into the life of a building that is empty today, unused and dilapidated. The goal was not to forcefully fill the functions of the building, but in successive steps and phases to find a solution how to reuse the building and to point to the factory as a element of the city's foundation and also to highlight the unique architecture of the Baťa period.

Titel: Intersport & Skiljebo SK. Ett vinnande samarbete?

Problem: Sponsring är en viktig marknadsföringskanal för näringslivet och en viktig inkomstkälla för det svenska föreningslivet. Det är därför viktigt att relationen mellan sponsor och sponsortagare fungerar och att sponsorerna får ut vad de förväntar sig av samarbetet. Intersport och Skiljebo SK (SSK) har sedan många år tillbaka ett sponsrings- och samarbetsavtal. Intersport sponsrar SSK med utrustning och pengar mot att de får agera som återförsäljare/leverantör till föreningen samt att de hoppas få föreningens medlemmar som lojala kunder. De vet i dagsläget inte det exakta resultatet av samarbetet och är intresserade av att ta reda på om föreningens medlemmar är lojala kunder hos dem. De vill också stärka relationen till föreningen och deras medlemmar. För att kunna ta reda på hur de kan stärka relationen till SSK och medlemmarna i föreningen måste undersökningen visa hur det ser ut idag.

Syfte: Syftet med undersökningen är att beskriva och analysera relationerna mellan Intersport, SSK och SSK:s medlemmar utifrån attributen kommunikation, förtroende och engagemang. Dessa tre attribut är mycket viktiga beståndsdelar i en relation. Vidare är syftet att ta reda på om samarbetet leder till en ömsesidig lojalitet mellan Intersport och SSK respektive en köplojalitet mellan Intersport och SSK:s medlemmar.

Metod: För att kunna beskriva och analysera relationerna och för att ta reda på om det finns en lojalitet används ett något annorlunda angreppssätt där vi frågade alla tre inblandade parter. Det är alltså en triad av relationer där varje relation undersöks utifrån ovan givna attribut. För att få fram svar på hur det förhåller sig i relationerna används intervjuer för Intersport och SSK. För att få fram SSK:s medlemmars åsikter görs en enkätundersökning.

Resultat: Undersökningen visar att det finns brister i relationerna och att köplojaliteten mellan SSK:s medlemmar och Intersport är lägre än förväntat. Den ömsesidiga lojaliteten mellan Intersport och SSK som förening är dock mycket hög.

Title: Intersport and Skiljebo SK. A winning collaboration?

Problem: Sponsorship is an important marketing channel for the business world and an important income source for the Swedish sports associations. It is therefore important that the relationship between the sponsor and the sponsorship receiver is working and that the sponsors get what they expects of the collaboration. Intersport and Skiljebo SK (SSK) has since many years back a sponsor and collaboration deal. Intersport is giving SSK club equipment and money so they in return can be the retailer and supplier to the club and their members. Intersport hope this collaboration will lead to loyal SSK customers. Intersport does not know the exact result of the collaboration so they are interested to found out if the club members are loyal customers or not. Intersport wants to strengthen the relationship with the club and their members. To be able to do this they have to found out, with the help of this research, how the situation looks today.

Purpose: The purpose with the research is to describe and analyze the relationships between Intersport, SSK club and their members on the basis of the three attributes communication, trust and commitment. These three attributes are all very important parts in a relationship. The purpose is also to found out if the collaboration is leading to a mutual loyalty between Intersport and SSK club and respectively a buying loyalty between Intersport and SSK club members.

Method: To be able to describe and analyze the relationships and to see if there is a loyalty, a rather unusual approach is used were, we ask all three involved parties. It is a triad of relationships where every relation is investigated on the basis of the before mentioned attributes. Interviews were done with Intersport and SSK club to get an answer how the relationship is working. A survey was done to see what opinion SSK club members had about the relationships.

Result: The research show that there are some flaws in the relationships and the buying loyalty between SSK members and Intersport is not as high as Intersport maybe expected. The mutual loyalty between Intersport and SSK club is however very high.

The purpose of my Master's Thesis is to determine the value of the SK Slavia Praha a.s. as of 30.06.2013. I focus on comparing differences between valuation of classical manufaturing company and valuation of sport club in theoretical part. I describe suitable methods for valuation of company and apparatus, that I chose to valuation of club of course in theoretical part. I use financial analysis to analyse main indicators, which affect the value of company in practical part. I use yield and property method for valuation. Property method is more meaningful due to the fact that company does not have profit in recent years. Sports club in the Czech Republic are not traded on stock market unfortunately, so we can't compare result with anything. On the other hand Slavia changed owners recenty unfortunately, so estimate value could be based on the price which current owner bought it, but this value is only for comparison, relative to situation which club was at that time.

The aim of this thesis is to identify and analyse factors affecting the attendance of domestic football matches of SK Dynamo club České Budějovice. The main emphasis is placed on working with fans. The theoretical part relates to the topic and focuses on the determining factors of demand for sport events, the sport product theory and phenomenon of a fan. The practical part introduces the club SK Dynamo České Budějovice more closely and analyses individual factors affecting demand for the club's matches and defines the significance of these factors. A chapter on working with the fans follows. At first the elements of sport product are characterised and secondly the club marketing elements. The practical section chapters are underpinned by the results of a survey which was carried out among the fans of SK Dynamo České Budějovice. The discourse and conclusion summarize the analysed results, evaluates working with fans and last but not least outlines possible steps for improvements of attendance and working with club's fans.

Abstract:
1967 tog svensk idrott beslutet att avskaffa amatörreglerna, vilket innebar att idrottare nu kunde ta emot pengar för sitt idrottsutövande. Det dröjde dock fram till slutet på 1980-talet innan svensk fotboll på allvar professionaliserades och där hela spelartrupper heltidsanställdes av fotbollsklubben. I och med TV:s och medias ökade inblandning och intresse i fotbollen har sporten kommersialiserats allt mer och idag kommer stora delar av fotbollsklubbars intäkter från TV, reklam och sponsring. 1999 tog Riksidrottsförbundet ett beslut som gav idrottsföreningar tillåtelse att starta ett aktiebolag (IdrottsAB) som arbetar jämsides med den ideella föreningen, dock med begränsningen att ägandet i aktiebolaget till 51 % måste innehas av den ideella föreningen och dess medlemmar. I svensk fotboll har trenden med IdrottsAB inte tagit den fart som först förutspåddes, med tre allsvenska klubbar 2012 som valt vägen att bolagisera verksamheten. En av dessa tre klubbar är Örebro SK som 2006 bildade ÖSK Elitfotboll AB efter att 2004 ha blivit tvångsnedflyttade till Superettan på grund av ett negativt eget kapital. Syftet med den här studien är att undersöka hur bolagiseringen har påverkat ÖSK, vilka för- och nackdelar som upplevs med IdrottsAB, om fokus på det ekonomiska tagit överhand, om bolagisering hotar det ideella engagemanget och vilka för- och nackdelar som ses med 51-procentsregeln. Denna studie drivs med en kvalitativ ansats där vi valt att intervjua fem personer med koppling till ÖSK. Respondenterna ansåg att den största anledningen till att klubben valde att bolagisera var dels att ett aktiebolag ger ett proffsigare intryck samt att reglerna som gäller för aktiebolag är betydligt tydligare än de som gäller för ideella föreningar. Detta är även de största fördelarna tillsammans med ett större fokus på marknadsintäkter. Delade meningar råder om huruvida momspliktigheten varit positiv eller negativ för klubben. Den stora nackdelen anses vara poliskostnader vid matcharrangemang som faktureras aktiebolag men inte ideella föreningar. Respondenterna påpekar att bolagiseringen varit betydelsefull för klubben, om beslutet inte tagits hade vägen tillbaka till Allsvenskan varit mycket längre. Vad gäller 51-procentsregeln råder det delade meningar om ett avskaffande av den vore positiv eller negativ för svensk fotbolls konkurrenskraft internationellt. Abstract:
Human plasminogen was activated better by streptokinase (SK) in the presence of fibrin or its degradation products(potentiating agents). Such potentiating agents must be mixed with SK and plasminogen simultaneously. The addition of potentiating agents to the mixture of SK and plasminogen did not result in any enhancement of the activator activity of SK-plasminogen complex. Since potentiating agents such as fibrin bind to lysine binding sites (LBS) of plasminogen, and SK was known to bind to light chain side of plasminogen, it appears that a trimolecular complex of SK, plasminogen and a potentiating agent is a better enzyme than a complex of SK and plasminogen. The effectiveness of the potentiation was in the order of fibrin> SK-potentiator(early FgDP)> fibrinogen >fragment D >fragment E, when Glu-plasminogen was activated by SK. When Lys-plasminogen was used, fragment E was more effective than fragment D, thus in the order of fibrin> SK potentiator> fibrinogen>fragment E >fragment D. Such enhancement of the activation of plasminogen by fibrin or potentiating agents is more significant in the presence of lower concentration of SK. Increase in the concentration of SK did not result in much enhancement of the activation of plasminogen. Kinetic analyses indicated that kcat of the trimolecular complex (SK, plasminogen and potentiating agent) was higher than SK-plasminogen complex, without change in Km. These results suggest that a trimolecular complex of SK, plasminogen and potentiating agent converts plasminogen to plasmin more effectively than a dimole-cular complex of SK and plasminogen. Considering the fact that the plasma concentrations of thrombolytic agents such as SK or UK were low even after drip infusion of their large dose Intravenously, the potentiation of the activation of plasminogen by SK in the presence of fibrin maybe significant in the mechanisms of thrombolytic therapy.

Abstract:
We identified a patient who exhibited paradoxical propagation of thrombus coincident with the administration of intracoronary streptokinase (SK) that was mediated by anti-SK antibodies. The patient had not been treated with SK in the past and had a plasma SK-neutralizing capacity of 160 U/ml. Using platelet-rich plasma (PRP) obtained from the patient, we found that SK initiated spontaneous platelet aggregation and secretion in vitro. Aggregation was specific for SK and not induced by urokinase or tissue plasminogen activator. In 14 of 15 controls, no platelet aggregation was observed in PRP with addition of SK. The addition of plasma or purified IgG from our index case to the PRP of all 14 controls supported SK-induced aggregation. This aggre-gatory response was not inhibited by aprotinin. Using purified proteins in a washed platelet system, we found that platelet aggregation was dependent on the presence of SK, specific anti-SK IgG, plasminogen, and platelets. These data demonstrate that anti-SK antibodies can promote platelet aggregation, presumably by binding to platelet-bound plasminogen-SK complexes. These data also imply that some individuals may possess anti-SK antibodies that are capable of inducing platelet aggregation in vivo and, thereby, promoting clot propagation or thromboembolic complications. In the absence of adequate, specific screening, this observation argues for the use of nonimmunogenic thrombolytic agents in the emergent setting.

Fibrinolytic activity of APSAC (anisoylated plasminogen-strep-tokinase activator complex) 30 U intravenous bolus (n = 10) was compared with intravenous (IV) streptokinase (SK). 1.500.000 IU (n = 14), SK 500.000 IU (n = 6), and intracoronary SK 250.000 IU (n = 20) given for acute myocardial infarction ≤; 4 hours) and following pretreatment with heparin (10.000 IU bolus).Plasminogen activator levels (fibrin plate) were highest for ASPAC group and correlated with the dosages of SK*. Levels also directly correlated to the duration of the activity (> 10 mm2), from 3 hours for SK 250.000 IU up to 18 hours for APSAC*.Plasmin activity (fibrin plate) raised acutely with all regimens and was maintained (> 5 mm2) during 3 hours (SK 250.000 IU), 12 hours (SK 500.000 IU), 16 hours (SK 1.500.000 IU) and 18 hours (APSAC). Reductions of plasminogen, α2 antiplasmin and fibrinogen (Clauss) were found accordingly most depressed after APSAC and significantly less after SK 250.000 IU (p < 0.05).The thrombolytic efficacy was judged on the patency rate assessed from post interventional (4 hours) angiogram in larger groups treated with intracoronary SK 250.000 IU : 28/35 (80 %), IV SK 500.000 IU : 29/41 (72 %), IV SK 1.500.000 : 18/25 (72 %), APSAC : 8/10 (80 %).To conclude, fibrinolytic activity of APSAC was best compared to the intensity and duration of IV SK 1.500.000 IU. Its thrombolytic efficacy appears to be in the range of the other regimens.* p < 0.01 rank test
