Several research lines show that sleep favors memory consolidation and learning. It has been proposed that the cognitive role of sleep is derived from a global scaling of synaptic weights, able to homeostatically restore the ability to learn new things, erasing memories overnight. This phenomenon is typical of slow-wave sleep (SWS) and characterized by non-Hebbian mechanisms, i.e., mechanisms independent of synchronous neuronal activity. Another view holds that sleep also triggers the specific enhancement of synaptic connections, carrying out the embossing of certain mnemonic traces within a lattice of synaptic weights rescaled each night. Such an embossing is understood as the combination of Hebbian and non-Hebbian mechanisms, capable of increasing and decreasing respectively the synaptic weights in complementary circuits, leading to selective memory improvement and a restructuring of synaptic configuration (SC) that can be crucial for the generation of new behaviors ( insights ). The empirical findings indicate that initiation of Hebbian plasticity during sleep occurs in the transition of the SWS to the stage of rapid eye movement (REM), possibly due to the significant differences between the firing rates regimes of the stages and the up-regulation of factors involved in longterm synaptic plasticity. In this study the theories of homeostasis and embossing were compared using an artificial neural network (ANN) fed with action potentials recorded in the hippocampus of rats during the sleep-wake cycle. In the simulation in which the ANN did not apply the long-term plasticity mechanisms during sleep (SWS-transition REM), the synaptic weights distribution was re-scaled inexorably, for its mean value proportional to the input firing rate, erasing the synaptic weights pattern that had been established initially. In contrast, when the long-term plasticity is modeled during the transition SWSREM, an increase of synaptic weights were observed in the range of initial/low values, redistributing effectively the weights in a way to reinforce a subset of synapses over time. The results suggest that a positive regulation coming from the long-term plasticity can completely change the role of sleep: its absence leads to forgetting; its presence leads to a positive mnemonic change
Diversas linhas de pesquisa demonstram que o sono favorece a consolida??o de mem?rias e o aprendizado. Tem sido proposto que o papel cognitivo do sono deriva de um redimensionamento global dos pesos sin?pticos, capaz de restabelecer homeostaticamente a capacidade de aprender coisas novas, apagando mem?rias durante a noite. Tal fen?meno seria t?pico do sono de ondas lentas ( slow wave sleep , SWS) e caracterizado por mecanismos n?o-Hebbianos, isto ?, independentes da atividade neuronal sincr?nica. Outra abordagem postula que o sono desencadeia tamb?m um realce de conex?es sin?pticas espec?ficas, levando a um entalhamento de certos tra?os mnem?nicos no ?mbito de uma matriz de pesos sin?pticos redimensionados a cada noite. Tal entalhamento ? entendido como a combina??o de mecanismos Hebbianos e n?o-Hebbianos, capazes respectivamente de aumentar e diminuir os pesos sin?pticos em circuitos complementares, levando ? melhoria seletiva de mem?rias e a uma reestrutura??o da configura??o sin?ptica ( synaptic configuration , SC) que pode ser crucial para a gera??o de novos comportamentos ( insights ). Os achados emp?ricos indicam que a indu??o de plasticidade Hebbiana durante o sono acontece na transi??o do SWS para o est?gio de movimento r?pido dos olhos ( rapid eye movement , REM), possivelmente devido ?s grandes diferen?as entre os regimes das taxas de disparos entre os estados e ? regula??o positiva de fatores envolvidos na plasticidade sin?ptica de longo prazo. Neste estudo, as teorias da homeostase e do entalhamento foram comparadas usando uma rede neural artificial ( artificial neural network , ANN) alimentada com potenciais de a??o registrados no hipocampo de ratos durante todo o ciclo sono-vig?lia. Na simula??o em que a ANN n?o aplicou mecanismos de plasticidade de longo prazo durante o sono (transi??o SWS-REM), a distribui??o pesos sin?pticos foram inexoravelmente re-escalada para uma media proporcional ? taxa de disparo das entradas, apagando eventualmente o padr?o de pesos sin?pticos inicialmente estabelecido. Em contraste, quando a plasticidade de longo prazo foi modelada durante a transi??o SWS-REM, o aumento dos pesos sin?pticos foi observado em toda a gama de valores iniciais, efetivamente redistribuindo os pesos de modo a refor?ar um subconjunto de sinapses ao longo do tempo. Os resultados sugerem que uma regula??o positiva proveniente da plasticidade de longo prazo pode alterar completamente o papel do sono: sua aus?ncia leva ao esquecimento, sua presen?a leva a uma mudan?a mnem?nica positiva

Dopamine (DA) is known to regulate both sleep and memory formations, while sleep plays a critical role in the consolidation of different types of memories. We believe that pharmacological manipulation of dopaminergic pathways might disrupt the sleep-wake cycle, leading to mnemonic deficits, which can be observed in both behavioral and molecular levels. Therefore, here we investigated how systemic injections of haloperidol (0.3 mg/kg), immediately after training in dark and light periods, affects learning assessed in the novel object preference test (NOPT) in mice. We also investigated the hippocampal levels of the plasticity-related proteins Zif-268, brain-derived neurotrophic factor (BDNF) and phosphorylated Ca2+/calmodulin-dependent protein kinases II (CaMKII-P) in non-exposed (na?ve), vehicle-injected controls and haloperidol-treated mice at 3, 6 and 12 hours after training in the light period. Haloperidol administration during the light period led to a subsequent impairment in the NOPT. In contrast, preference was not observed during the dark period neither in mice injected with haloperidol, nor in vehicle-injected animals. A partial increase of CaMKII-P in the hippocampal field CA3 of vehicle-injected mice was detected at 3h. Haloperidol-treated mice showed a significant decrease in the dentate gyrus of CaMKII-P levels at 3, 6 and 12h; of Zif-268 levels at 6h, and of BDNF levels at 12h after training. Since the mnemonic effects of haloperidol were only observed in the light period when animals tend to sleep, we suggest that these effects are related to REM sleep disruption after haloperidol injection
A dopamina (DA) regula tanto o sono quanto o processo de forma??o de mem?rias, enquanto o sono desempenha uma fun??o essencial na consolida??o de diferentes tipos de mem?rias. Acreditamos que a manipula??o farmacol?gica das vias dopamin?rgicas possa afetar o ciclo sono-vig?lia, acarretando d?ficits mnem?nicos, os quais podem ser observados tanto em n?vel comportamental quanto molecular. Dessa forma, no presente estudo, investigamos o efeito do haloperidol (halo - 0,3 mg/kg; i.p.), administrado imediatamente ap?s sess?es de treino no per?odo da manh? e da noite, sobre o desempenho de camundongos no teste de prefer?ncia por objetos novos (TPON). Observamos que esse tratamento causou um preju?zo mnem?nico apenas nos camundongos testados pela manh?. Devido ? maior ocorr?ncia de epis?dios de sono durante a fase clara dos roedores, acreditamos que esse d?ficit pode estar associado a uma cascata de plasticidade hipocampal, a qual envolve a express?o de diferentes mol?culas em per?odos distintos ap?s a exposi??o ao est?mulo mnem?nico inicial. Para testar tal hip?tese, realizamos imunohistoqu?mica (IHQ) em tecidos de animais na?ve n?o expostos (NV), bem como de animais expostos aos objetos com posterior inje??o de halo ou salina (ve?culo - VH), perfundidos 3, 6 e 12h ap?s o est?mulo inicial. Quantificamos a express?o hipocampal (CA1, CA3 e GD) das prote?nas CaMKII-P, Zif-268 e BDNF, atrav?s de densitometria e contagem de c?lulas (somente para Zif-268). Observamos um aumento parcial na express?o de CaMKII-P em animais VH 3h, na regi?o CA3. Quanto ao grupo halo, houve uma redu??o dos n?veis de CaMKII-P nos tr?s hor?rios analisados; de Zif-268 em 6h, tanto em CA1 quanto CA3; e de BDNF em 12h, apenas no GD. Considerando-se o melhor desempenho diurno na consolida??o de mem?rias, associado ? fun??o do halo na regula??o do sono, propomos que a redu??o de plasticidade sin?ptica e os d?ficits no processo mnem?nicoocorreram devido ? supress?o do sono REM mediada pelo tratamento com haloperidol

Full textNo-tillage systems (NTS) play a very important role in Brazilian agriculture. NTS are widespread through extensive areas with different climates, soil types and under different management production systems. NTS deeply alter the biological, physical and chemical properties of the soils, as compared to conventional tillage. Soil solution studies, evaluating nutrient concentration changes under management practices, are very important in soil fertility, plant nutrition and management. The effect of liming and three methods of application (one application of 4.5 t ha-1 incorporated, one application of 4.5 t ha-1 surface applied, and, 1.5 t ha-1 applied in each of three years) on the composition and ionic speciation of soil solution, during the implantation of NTS in a native pasture area until 0.8 m depth was studied. The effect of gypsum rate (3; 6 and 9 t ha-1) was also studied under the same conditions. Soil samples were extracted from a field experiment installed in1998 in Ponta Grossa/PR. The selected soil was classified as a dystrophic clayey Rhodic Hapludox. After the harvesting of soybeans in May of 2003, the samples were collected at the following depths: 0-0.05; 0.05-0.1; 0.1-0.2; 0.2-0.4; 0.4-0.6 and 0.6-0.8 m. Liming increased the pH of soil solution, but did not affect dissolved organic carbon (DOC) contents and ionic strength values. It was observed that pH was increased up to a depth of 0.2 m. Liming increased calcium and magnesium concentrations up to 0.2 m and 0.6 m depth, respectively, except for first layer with incorporate lime. However, only Mg showed an increase in activity. Gypsum application increased the SO4 -2 e and Ca concentration, and Ca activity through the studied profile. A negative effect of gypsum was the leaching of Mg and a reduction in its activity. The concentration of anions (Cl-, F-, NO3 -, PO4 -3) and Al were neither affected by gypsum or by lime. The main Al complexing agents in solution were organic anions and F-. A low free Al+3 concentration and activity was detected, with no differences management treatments. The Al in the solid phase was mainly complexed by organic matter. The Ca and Mg were complexed by organic anions, and the effect was more pronounced for Ca. The P (PO4 -3, HPO4 -3, H2PO4 -3) was main complexed for Al, while the SO4 -2 was with Ca and Mg mainly. Fe was the form Fe-OH and Fe-DOC. The elements Mn, K, N-NO3 - and Cl formed insignificant quantities ionic pairs in soil solution in whole layers.

Full textDans le cadre de cette thèse nous avons cherché à objectiver et caractériser l’intégration labio-manuelle dans la perception de la parole codée. Le poids accordé par le système perceptif aux informations manuelles, d’une part, et aux informations labiales, d’autre part, dépend-il de la qualité de chacune d’entre elles ?Varie-t-il en fonction du statut auditif ?Quand l’information auditive est disponible, comment le traitement de l’information manuelle est-il incorporé au traitement audio-visuel ?Pour tenter de répondre à cette série de questions, cinq paradigmes expérimentaux ont été créés et administrés à des adultes sourds et normo-entendants décodant la LPC.
Les trois premières études étaient focalisées sur la perception de la parole codée sans informations auditives. Dans l’étude n° 1, le but était d’objectiver l’intégration labio-manuelle ;l’impact de la qualité des informations labiales et du statut auditif sur cette intégration a également été investigué. L’objectif de l’étude n° 2 était d’examiner l’impact conjoint de la qualité des informations manuelles et labiales ;nous avons également comparé des décodeurs normo-entendants à des décodeurs sourds. Enfin, dans l’étude n° 3, nous avons examiné, chez des décodeurs normo-entendants et sourds, l’effet de l’incongruence entre les informations labiales et manuelles sur la perception de mots.
Les deux dernières études étaient focalisées sur la perception de la parole codée avec du son. L’objectif de l’étude n°4 était de comparer l’impact de la LPC sur l’intégration AV entre les sourds et les normo-entendants. Enfin, dans l’étude n°5, nous avons comparé l’impact de la LPC chez des décodeurs sourds présentant une récupération auditive faible ou forte.
Nos résultats ont permis de confirmer le véritable ancrage du code LPC sur la parole et de montrer que le poids de chaque information au sein du processus d’intégration est dépendant notamment de la qualité du stimulus manuel, de la qualité du stimulus labial et du niveau de performance auditive.

Full textThis paper seeks to present the experiences of three listeners, besides the author, childrens of deaf parents, discussing family relationships between parents and children, showing the differences in relation to families who have no deaf people at its core, demystifying prejudices and pre -concepts. Due to communication barriers and prejudice, many deaf people ended up being deprived of the right to be parents. In the reports presented in this thesis, there were times when it almost happened in the lives of the subjects interviewed for this research, but with strength and awareness of rights they had, these deaf parents would not allow such a thing to happen. The text begins with the history and formation of the author to then bring the authors covering topics such as deafness (Wrigley, 1996; Lacerda, 1998; Perlin, 1998; Moura, 2000; Sá, 2002; Quadros, 2004; Sacks, 2007), anthropology (Hall, 1992; Gomes, 2008), family relationships (Lacan, 1987) and autobiography (Ferraroti, 1991; Pineau, 2006; Demartini, 2008). The sign language of the deaf and educational history are also discussed in this paper. At the end are presented reports of life history made in the research field of literary form, with analyzes of testimonies collected by interviewees (deaf parents and hearing children) which showed to be son of deaf parents can be as normal as being the child of hearing parents.
Este trabalho busca apresentar as vivências de três pessoas ouvintes, além do próprio autor, filhas de pais surdos, discutindo as relações familiares entre pais e filhos, mostrando as diferenças em relação às famílias que não possuem pessoas surdas no seu âmago, desmistificando preconceitos e pré-conceitos. Devido as barreiras comunicacionais e pelo preconceito, muitos surdos acabavam sendo privados do direto de serem pais. Nos relatos apresentados nesta dissertação, houve momentos em que isso quase aconteceu na vida dos sujeitos entrevistados para esta pesquisa. Porém, com força e consciência dos direitos adquiridos, esses pais surdos não permitiram que tal coisa acontecesse. O texto inicia com a trajetória formativa e profissional do autor para, em seguida, trazer os autores que abordam temas como surdez (Wrigley, 1996; Lacerda, 1998; Perlin, 1998; Moura, 2000; Sá, 2002; Quadros, 2004; Sacks, 2007), antropologia (Hall, 1992; Gomes, 2008), relações familiares (Lacan, 1987) e autobiografia (Ferraroti, 1991; Pineau, 2006; demartini, 2008). A língua de sinais e a história educacional dos surdos também são discutidas neste trabalho. Por último, são apresentados os relatos de história de vida feitos na pesquisa de campo de forma literária, com as análises dos depoimentos colhidos pelos sujeitos entrevistados (filhos ouvintes e pais surdos) os quais evidenciam que ser filho de pais surdos pode ser tão normal quanto ser filho de pais ouvintes.

Full textThis dissertation analyzes how Ibn 'Atâ' Allâh al-Iskandarî deals, in his writings, with the notion of sin, highlighting its positive, or salvatory, aspect, taking into account the contributions of previous Sufi writers who played a role in his spiritual education. After an overview of the situation in Egypt in the early Mamluk era, this work tries to draw the portrait of Ibn 'Ata Allah by treating some less known dimensions of his personality and demonstrating the importance of his works in transmitting the spiritual teaching he received orally from his masters, the founders of the Shâdhiliyya. Concerning the concept of sin, it is first analyzed through a lexicographical study, before retracing the ways in which it was addressed by Muhâsibî, Makkî Qushayrî, Ghazâlî and Ibn 'Arabî. The latter’s influence on Ibn 'Atâ’ Allâh, often minimized in earlier works, is demonstrated in more than one respect, both in the way he describes Adam’s sin and in his general approach tending to transcend the existence of an immutable duality between sin and obedience

Full textIn this Master's thesis, we use the numerical methods such as molecular dynamics (LAMMPS's code) and kinetic-ART which is an on-the-fly off-lattice kinetic Monte Carlo algorithm that incorporates exactly all elastic effects. In the first, we compare a number of various algorithms used for sampling energy landscape of complex materials. The various algorithms chosen are those that use the Bell-Evans-Polanyi principle to progress on the potential energy surface.This study allowed us to understand the steps needed to escape a local minimum to another and to control research to quickly find the global minimum. This study also allowed us to understand the power of these methods on the kinetics of the structural evolution of complex materials. In the second part, we have developed a simulation tool (ReaxFF potential coupled with Kinetics-ART) able to study the first stages and oxidation process of silicon compare to the experimental time. To validate the system in place, we have tested the very first step of the silicon oxidation. The results obtained are in agreement with the literature. This tool will be used to understand the true oxidation process, the possible transitions of oxygen atoms at the silicon surface and the barrier associated with. Problem that are real challenges for the microelectronics industry.

Full textTo very few philosophers the history of the Western thought granted a place as significant as to Socrates: we quite naturally learn how to identify him as a hero of rationality and to recognize in him the very figure of the critical philosopher. In several respects, this representation of praise appears justified to us, although, from another point of view, it can make us sink in confusion, since our glance carries simultaneously, and like producing a contrast, on the image of Socrates obeying to the daimonion, his uncanny divine sign. How can we justify, starting from the Platonic corpus, both the engagement of Socrates with respect to rationality and his subordination to a seemingly irrational phenomenon? From this disconcerting question was born the present study which is thus devoted to the problem of the articulation between the relation of Socrates to the gods and his relation to the critical reason. More precisely, we sought to determine if there existed, on the epistemological level, a hierarchy between the daimonion and the method of rational investigation peculiar to Socrates, the elenchos. Such an exegetic study required, initially, a systematic and thorough analysis of the few passages related to the divine sign. We then adduced two paradigmatic solutions, that of G. Vlastos as well as that of T.C. Brickhouse and N.D. Smith. Lastly, we added to this second part besides a specific examination of Phaedrus and Timaeus, also a broad survey of the modes of divination, satisfying a triple aim: to make sense of the divine sign in comparison with the traditional art of mantic, to determine the role allotted by Plato to the reason in the divinatory process, and thus to be able to solve our principal question.

Full textἦhe paraἴle of the “merἵiful father” (δuke 1ημ11-32), being the core of the third gospel, tells us a great deal about Jesus, His message and interlocutors. The texts contains a unique and unrepeatable story with remarkable originality. Before drawing any conclusions, it is important to understand the sitz-im-leben of the text in order to comprehend its origin and central message. The text analysis and the social and religious aspects therein are taken in relation with the whole of the gospel of Luke, as the premises for focusing the characters in their respective motivations, features and distinctive behavior. Mercy is the main thread running through this text, despite being present only in the actions of the main character, the father. Moreover, mercy is the chief characteristic of the man telling the parable, Jesus. The two sons typify His interlocutors: the Pharisees and the Doctors of the Law (typified by the older son) and the sinners and the publicans (typified by the younger son). Nevertheless, the parable is not restricted to the exercise of mercy, instead it points to salvation, as announced in the final banquet.
