Full textBackground: Chikungunya virus (CHIKV), Semliki Forest virus (SFV) and Sindbis virus (SINV) are mosquito-borne alphaviruses. CHIKV has caused important outbreaks of severe disease during the last decade. Alphavirus are enveloped viruses that enter cells via low-pH triggered fusion in the endocytic pathway. The envelope protein E1 is a class II fusion protein and is responsible for driving membrane fusion for entry. The E1 ectodomain is folded into three structured domains (I, II and III), arranged as a rod with domain I at the center and domains II and III at either side. Domain II is elongated and exposes a hydrophobic "fusion loop" at its distal end. The fusion loop inserts into the target membrane, and a subsequent conformational change brings the fusion loop into proximity with the viral-membrane-anchored segment of the protein, via a relocation of the C-terminal domain III and the downstream "stem" segment, which connects to the viral trans-membrane anchor at the C-terminal end. This results in a "hairpin" conformation of the protein that draws the two membranes together for fusion. Methods : We made the recombinant E1 ectodomain using a mutant of the fusion loop, to allow for solubility, and determined the crystal structures under acidic conditions, in their trimeric hairpin conformation. Results : Our work shows that the post-fusion trimers of E1 from the three alphaviruses studied adopt a strikingly different conformation in spite of their high sequence similarity, a change that could not have been predicted by homology modeling from the available structure of E1 from Semliki Forest virus truncated of part of the stem. They show that while the internal trimer core, composed of domain I and the domain II base, is maintained in the same conformation, and domain III flips to the sides of the trimer to form the hairpin in the same way, the downstream "stem" region and the domain II "tip" pack in a different way, resulting in three alternative forms: closed, intermediate, and open, depending on the virus. Conclusion : E1 is a very conserved protein in alphaviruses, and the available structure was believed to be a model for all alphaviruses. Our results highlight the importance of carrying out comparative structural studies, and help explain contradictory results in the use of domain III and the stem region for inhibition of the fusion step with different alphaviruses. These results will have an impact to better understand the conformational change and the role of the "stem" region for membrane fusion, and allow the development of fusion inhibitors in the future

Full textSyftet med studien är att förstå hur skolkuratorerna skapar sin yrkesroll utifrån sina erfarenheter. Studien är en kvalitativ intervjustudie med en fenomenlogisk ansats. I resultatet framgår det att skolkuratorerna värdesätter det enskilda samtalet med eleven. Samverkan på olika nivåer i samhället och kunskap om hur samhället fungerar beskrivs av skolkuratorerna som viktiga i arbetet med elever och föräldrar. Samarbetet i skolans arbetsgrupper har enligt skolkuratorerna en avgörande betydelse för handlingsutrymmet med eleverna. Det förebyggande arbetet med eleverna är enligt skolkuratorerna viktigt. Att använda hjälpmedel som till exempel en trollstav kan hjälpa till i det förbyggande arbetet med elever. Skolkuratorerna beskriver att de använder olika teorier och metoder. Valet av vilka av dessa som är mest lämpliga i en viss situation styrs av vad skolkuratorn anser vara lämpligt i en specifik situation. Dessa kan även tillämpas tillsammans med av skolkuratorn egna utvecklade metoder och hjälpmedel som till exempel trollstaven. I intervjuerna med skolkuratorerna beskriver de sin yrkesroll med entusiasm och professionalism. Det framgår även att de har olika syn på sitt arbete och sina arbetssätt. Vidareutbildning och möjlighet att ta del av vidare forskning uppskattas av skolkuratorerna. Kunskap om lagarna är enligt skolkuratorerna en viktig faktor för att känna sig trygg i sin egen yrkesroll. Det framgår också att skolkuratorerna ibland saknar någon att reflektera med trots att ett nätverk finns runt dem. De förväntningar som skolkuratorerna beskriver är att de vill ha en tydligare definierad lagstadgad skolkuratorstjänst, vilket de anser skulle tydliggöra förväntningarna på yrkesrollen.

Full textSyftet med arbetet var att åskådliggöra innebörden av att vara i matematiksvårigheter och elevers syn på hur de hanterar sin vardag. Undersökningen utgår från ett elevperspektiv och ambitionen var att elevens egna tankar och känslor skulle komma fram. Jag har använt mig av en kvalitativ ansats med observationer och semistandardiserade intervjuer. För alla eleverna i studien påverkas vardagen och skolan av att de har svårigheter i matematik. Det är allt från att planera sina fritidsaktiviteter till att veta när ett arbete i skolan ska vara inlämnat. Detta gör att föräldrar måste hjälpa till i denna planering i stor utsträckning även nu när barnen är tonåringar och borde klara sig själva lite mer. Eleverna själva är ofta inte medvetna om sina strategier för att klara de svårigheter som uppstår på grund av matematiksvårigheterna. Föräldrarna kan dock berätta mer om hur dessa strategier tar sig ut. Känslorna inför matematiklektionerna är ofta en uppgivenhet, ska de misslyckas idag igen och de känner sig stressade inför att kanske inte uppnå målen. Lärarens bemötande spelar här en stor roll och ett icke positivt bemötande gör att eleverna upplever att de är till besvär för läraren. Det leder till att eleverna får ett svagt självförtroende och olustkänslor inför matematiken. I jämförelse med andra studier så upplever eleverna i min studie inte att de blir stämplade till följd av att de får särskilt stöd, utan tvärtom så är det en lättnad att gå till stödgruppen.
This study focuses on pupil’s view how they handle mathematics learning disabilities in their daily life. The study is from pupil’s point of view and the ambition was to capture the pupil’s own thoughts and feelings. Observations and interviews were carried out with three pupils, in the age of 13, and their parents. The results show that all three of the pupils have difficulty in their daily life due to the mathematics learning disabilities. They have different problems from to plan their free time to plan when schoolwork should be submitted. The result of this is that the parents have to plan for them, even now when they are teenager’s and should be able to handle things like this by themselves. The pupils are not aware of the strategies they use to cope with the difficulties in mathematics. However, their parents are able to explain the strategies their children have developed. The emotions the pupils feel when they think about math class are that they feel dejected and they are under stress with thoughts as “will I fail again today”. It is very important how the teacher do talk to the pupils and this will affect the pupil’s self-confidence and they develop discomfort to mathematics. This study, unlike some other studies, does not show that pupils feel uncomfortable because they have special support. Instead they feel relief in visiting the support group.

Full textAim: To explore how nurses in medical wards evaluate their work and how they experience their psychosocial work environment. Method: A quantitative, comparative cross-sectional study. Questionnaires were distributed to 57 nurses on two medical wards at Akademiska hospital. Results: Thirtyfour nurses (60%) participated. Few nurses considered their work as calm and peaceful, and several nurses said they rarely or never had time to do the tasks that should be performed during a shift. The nurses generally stated that it’s very important to know patients' vital signs. The task that was most affected by high workload was to know patients' urine output, which only 17 nurses (50%) said they had time to check. The task that most nurses (n=28, 82%) said they had time for is to know patients' level of consciousness, followed by the physicians round (n=27, 79%). The task that most nurses said that they did not have time for was to give patients oral care (n=20, 59%), followed by knowing how much patients have eaten (n=16, 47%). Nurses who had worked under 2,5 years valued certain administrative and patient-related duties higher than nurses who had worked over 2,5 years.

Full textAn overarching perspective regarding the mapping of newly arrived pupils's needs characterizes the Swedish society and school system. This leads to a breakdown of pupils in so-called “Language Introductory Preparation Classes” which separates newly arrived pupils from pupils in ordinary classes and complicates integration. The purpose of this study is to capture the pupils`s perspective through a so-called narrative identity analysis aimed to investigate how 7 pupils from a Language Introduction- class at a high school in Stockholm, Sweden present their identities through their stories about their school situation. This has been implemented using interviews about the pupils' experiences and interpretations of events that has taken place at their school and class. Stories regarding the change of school system, the meeting with the class and future hopes have been presented in different ways by the students. The most prominent stories consist of ontological narratives based on the individual's subjective experiences and self-image, public stories about integration, and conceptual stories about racism. The students portray a similar picture of the roles of teachers and students at the school, a community with their fellow classmates, a struggle for recognition from the other students and the rulers of the school, as well as a hope for a happy ending and a resolution of the fight for recognition. The happy end also depicts an uncertainty about the future where an uncertainty regarding the ongoing struggle for recognition appears.

Full textThe cardiac type-2 ryanodine receptor (RyR2) encodes a Ca2+ release channel on sarcoplasmic reticulum (SR) membrane in cardiomyocytes, including sinoatrial node (SAN) myocytes, and releases Ca2+ required for contraction and SAN spontaneous rhythm. Its genetic defects are related to catecholaminergic polymorphic ventricular tachycardia (CPVT), which is a lethal heritable disease characterized by exercise/stress-induced syncope and/or sudden cardiac death. Interestingly, CPVT patients frequently present SAN dysfunction as bradycardia at rest.In a previous study, a novel CPVT-related RyR2 mutation (RyR2R420Q) in a Spanish family, associated with SAN dysfunction was reported. R420 is located at the N-terminal portion of the channel and seems to be an important site for maintaining a stable A/B/C domain of N-terminus in RyR2. As N-terminal mutation resultant RyR2 behaviour and SAN function are never analyzed before, we created the KI mice model bearing mutation R420Q to understand the underlying mechanism.In this thesis, we found increased Ca2+ release during diastole, indicating a gain-of-function effect of RyR2 N-terminal mutation R420Q. Interestingly, this defect may not be only an enhanced activity, as the Ca2+ sparks frequency was only slightly increased in KI, but also the closing mechanism, producing longer Ca2+ sparks. That is, the number of Ca2+ sparks is increased by the RyR2R420Q mutation, and meanwhile the amount of Ca2+ released in each Ca2+ spark is also dramatically enhanced. This increased Ca2+ release retards SR Ca2+ replenishment, disrupting the Ca2+ clock and the coupled clock, resulting in the slower SAN function. Thus favouring RyR stabilization in the closing state might not be an adequate therapy but accelerating its closure

The detention of foreign migrants sine permissum and without criminal charges is a migration control policy that, despite having a long history, it has increasingly expanded in all regions of the world since the decade of the seventies. Spain and Mexico have been no exception to this, which has been implemented with the so called internamiento and alojamiento, respectively. As a measure that affects personal liberty and puts under question the effectiveness of the principles of equality and non-discrimination, in this dissertation, after a critical review of the myths and realities of migration, plus the essential elements of equality, nondiscrimination, rights of foreigner and personal liberty in the applicable legal systems in each of the two countries under analysis; this immigration control policy is subjected to scrutiny in order to determine whether it can be considered permissible within the constitutional human rights systems of Spain and Mexico.

Full textIn the shadow of unemployment The purpose of this paper is to explore how unemployed single mothers who receive income support are experiencing this life, how it affects their identity and self-image and how they feel their lives affect their children. We conducted a qualitative study through interviews of five mothers who find themselves in this situation. From the interviews, we achieved results that indicate that these mothers feel that they are in vulnerable situations and perceive their economic situation was very difficult. The results also show that these mothers perceive themselves and their children are affected both socially and materially, and that their self-images also may become lowered because of that. A desire by mothers to be able to do more for their children than the economy permits has emerged. We have seen signs that the ambient views on these mothers have changed in different ways and that they can experience themselves and their children be excluded and stigmatized by their surroundings because of the life situation.

Full textA succesfully communicated message is dependent on a number of factors. One example of such factor is that the target group have confidence in the sender. What should one do if the target group even before the message is communicated, are sceptic towards the sender? With rhetoric and the ethos as a starting-point in this study, I examine how a public organisation can use ethos to build it’s credibility towards the target group. By focusing on the organisation’s competence, values and good-will and expressing all this in an informative text about the organisation in question, the expectation is that the tendency to use their product and services will increase among the target group. Theories of ethos serve as base for this study, as do qualitative interviews with the target group, the sender and one expert.

Full textThe aim of this study is to give insights in how teachers use their knowledge when they plan, conduct and evaluate their teaching of place value. The study is founded in Shulman’s six components of the process in teachers’ planning of teaching. The choice of method is semi structural qualitative interviews with teachers within the grades 1-3. The result shows that the teachers use different ways to teach and that they use their knowledge in different ways as they plan, execute and evaluate their lessons, including the basic knowledge they use and the adjustments they make in their education. It is also found that the teachers by most part follows Shulmans (1987) six components.

Full textThis qualitative study aimed to illustrate how men, that had or have had an eating disorder, expressed their experiences and thoughts about their eating disorders (ED’s), how they got sick, the treatment of their ED’s, and the recovery from the ED’s, in mass media. Method: A qualitative content analysis of mass media products in swedish. Results: The men in the study got an ED due to wanting control, relieve anxiety and/or being affected by ideals. The men felt loneliness and alienation in their ED and in the treatment of ED’s, because ED’s were considered a disease of women. Some men felt better and a sense of freedom after the recovery. Conclusion: Men with ED are an excluded group and they need more room in the society and within the treatment of ED’s.

Detta leder till att företagen måste få sina anställda att förebygga välfärdssjukdomarna genom utbildning och träning för att minska sjukfrånvaron på företaget. Syftet med uppsatsen är att granska företagets åtgärdssatsningar samt nyttjandegraden av dessa, för att därefter kunna förbättra nyttjandegraden av friskvårdssatsningarna genom ökad motivation och kommunikation från ledarna.

A presente dissertação procura avaliar em que medida a cooperação em ciência e tecnologia (C&T) entre a República Popular da China (RPC) com a República Argentina (RA) e com a República Federativa do Brasil (RFB) tem contribuído para o desenvolvimento de capacidades em ciência e tecnologia (C&T) e o fortalecimento dos complexos de inovação Triple Helix (universidade, indústria, Estado) nos respectivos países sul-americanos. A dissertação também procura avaliar até que ponto este tipo de cooperação sul-sul se diferencia da cooperação norte-sul. Para atingir estes objetivos foram analisados 185 tratados assinados pela RA e pela RFB com a RPC e com os Estados Unidos na área de cooperação C&T. Os tratados analisados foram aqueles assinados a partir do restabelecimento das relações diplomáticas na década de 1970 até 2016 e que aparecem disponibilizados nas páginas dos Ministérios das Relações Exteriores da Argentina e do Brasil. Nos tratados foram identificados os seguintes aspectos: áreas de interesses, investimentos, projetos conjuntos, processos de transferência de tecnologias, entre outros. Além da análise documental foram avaliados os resultados de dois programas de cooperação na área agropecuária o Laboratório Virtual (LABEX) da Embrapa em Beijing e o Centro Binacional Chino-Argentino de Ciência e Tecnologia de Alimentos. Constatou-se que existem algumas diferenças na lógica de cooperação sul-sul com a China e norte-sul com os EUA. No entanto, devido às diferenças em termos de capacidade científico-tecnológica entre os países sul-americanos e a China, a cooperação em C&T se diferencia apenas parcialmente da cooperação com os EUA. Os resultados da pesquisa de campo sobre a cooperação agropecuária mostraram que os impactos e realizações estão aquém das expectativas e dos discursos dos governos e documentos oficiais sobre a relação ganha-ganha com a China.
Abstract: This dissertation aims to assess the extent to which the cooperation in science and technology (S&T) between the People's Republic of China (PRC) and the Argentine Republic (RA) and the Federative Republic of Brazil (RFB) has contributed to the development of Science and technology (S&T) and the strengthening of Triple Helix innovation complexes (university, industry, State) in the respective South American countries. The dissertation also seeks to assess to what extent this type of South-South cooperation differs from North-South cooperation. To achieve these objectives, I analyze 185 agreements in S&T cooperation signed by RA and RFB with the PRC and the United States (USA). The agreements analyzed were those signed after the reestablishment of diplomatic relations in the 1970s until 2016 and which appear available on the pages of the Ministries of Foreign Affairs of Argentina and Brazil. In the agreements, the following aspects were identified: areas of interest, investments, joint projects, processes of technology transfer, among others. In addition to the documental analysis, I evaluated the results of two cooperation programs in the agricultural area: EMBRAPA's Virtual Lab-LABEX in Beijing and the Chinese-Argentine Binational Center for Food Science and Technology. In this dissertation, I conclude that there are some differences in the logic of the South-South cooperation with China and the North-South with the USA. However, due to differences in scientific-technological capacities between South American countries and China, S&T cooperation differs only partially from cooperation with the US. On the other hand, the results of cooperation fall far short of expectations of government speeches and official documents about the win-win relation with China.
