Full textA diversity of processes related to growth, development and adaptation to environmental conditions are regulated by signal transduction pathways. The SUCAST Project (Sugarcane Signal Transduction) (Souza et al., 2001) aims to identify, characterize and associate putative functions to sugarcane signal transduction components using bioinformatic tools and to evaluate their expression profiles using cDNA microarrays. BLAST searches conducted on sequences stored in the SUCEST databank - The Sugar Cane EST Project (Vettore et al., 2003) - revealed more than 3,500 putative genes related to signaling in sugarcane. The protein kinases were anaçyzed for the presence of conserved domains and classified according to a phylogenetic analysis of the predicted catalytic domain. The phylogenetic approach indicated six characteristic groups for protein kinases and four mains groups for receptor-like kinases. The expression patterns of SUCAST components were evaluated in six organs (root, flower, leaf, lateral bud, first and fourth internodes), in response to different stimuli (treatment with the phytohormones abscisic acid and methyljasmonate, drought, endophytic bacteria interaction, attack by Diatraea saccharalis and phosphate deficiency) and in sugarcane cultivars contrasting for sucrose accumulation. The tissue profiling experimenta showed 217 differentially expressed genes in at least one of the six organs analyzed and 153 genes of ubiquitous expression. A total of 179 differentially expressed genes were obtained in response to phytohormones and environmental stimuli, in at least one of the conditions analyzed were obtained. Comparisons between high and low sucrose (brix) cultivars led to the detection of 51 differentially expressed genes in at least one of the samples analyzed (leaves +1 and internodes at different maturing stages). The expression patterns were also analyzed in time-course experiments of phosphate deficiency, drought and phytohormone treatments by Self-Organizing Maps (SOM) clusterization. Quantitative PCR results confirmed 72% of the microarray expression data in sugarcane organs for 25 selected genes and 80.5% of the 36 expression profiles selected in response to phytohormones and environmental stimuli. Additionally, real-time PCR reactions were carried out to evaluate the expression levels of five genes in individuals contrasting for sucrose accumulation and 57% of the results obtained were in agreement with the microarray data. All data generated were integrated and compiled into the SUCAST databank (http://www.sucest-fun.org). The knowledge generated adds to the comprehension of the function of SUCAST components, indicating targets to be explored in the improvement of sugarcane cultivars.

Full textIntroduction: Spondyloarthritis is a group of diseases with clinical, laboratory and image similar. Analysis of clinical manifestations of spondyloarthritis in patients with and without the HLA-B*27 were performed, but the results revealed great heterogeneity. Objectives: The aim of this study was to evaluate the clinical manifestations of spondyloarthritis according to the classification criteria of the European Study Group Spondyloarthropathy (ESSG) in the presence and absence of the HLA-B*27. Patients and methods: From a total of 156 patients with clinical suspicion referred for the investigation of gene HLA-B*27, 73 were diagnosed with spondyloarthritis according to the criteria of ESSG. The HLA-B*27 were identified using commercial kits (Dynal ReliTM SSO HLA-B Typing Kit, Invitrogen). Clinical data were collected from medical records of patients. The results were compared using the chi-square or Fisher exact test. The values of Odds Ratio (OR) and confidence interval of 95% were also calculated. The p-value equal to or less than 0.05 was considered significant. Results: Of the 73 selected patients, 53 (72.6%) were male and 20 (27.4%) were female. The mean age was 49.4 and did not differ between genders (p = 0337). The spondyloarthritis found among the 73 patients were: ankylosing spondylitis (n = 47; 64.4%), psoriatic spondyloarthritis (n = 9; 12.3%), undifferentiated spondyloarthritis (n = 9; 12.3%), spondyloarthritis enteropathica (n = 6; 8.2%) and reactive arthritis (n = 2; 2.7%). The average age of onset was equal to 39.1 (± 11.7) and did not differ between genders (p = 0.9057). Of the total, 35 (47.9%) patients were HLA-B*27 positive and 38 (52.1%) were negative. This gene was positively associated with ankylosing spondylitis (OR: 5.37, 95% CI: 1813-15905, p = 0.003) and negatively with spondyloarthritis enteropathica (OR: 0.07, 95% CI: 0003-1301, p = 0.025). The sacroiliitis was associated with the presence of the gene (OR: 10 552, 95% CI: 1260-88256, p = 0.014) and intestinal injury absence (OR: 0.195, 95% CI: 0038-0978, p = 0.048). Conclusions: The HLA-B * 27 was associated with ankylosing spondylitis, enteropathic but not to spondyloarthritis. The radiological signs of sacroiliitis prevailed in patients positive for HLA-B*27, while intestinal involvement was associated with the absence of this gene. especially in cases of dystrophic scoliosis. In both cases, studies with larger samples are needed to assess whether these trends are evident.
Introdução: Espondiloartrite é um grupo de doenças com características clínicas, laboratoriais e imagenológicas semelhantes. Análises das manifestações clínicas das espondiloartrites em pacientes com e sem o gene HLA-B*27 foram realizadas, mas os resultados revelaram grande heterogeneidade. Objetivos: O objetivo deste trabalho foi avaliar as manifestações clínicas das espondiloartrites utilizadas nos critérios de classificação do European Spondyloarthropathy Study Group (ESSG) na presença e na ausência do gene HLA-B*27. Casuística e método: De um total de 156 pacientes com suspeita clínica, encaminhados para investigação do gene HLA-B*27, 73 tiveram diagnóstico de espondiloartrites confirmado, de acordo com os critérios do ESSG. O gene HLA-B*27 foi identificado com o uso de kits comerciais (Dynal ReliTM SSO HLA-B Typing Kit, Invitrogen). Os dados clínicos foram colhidos dos prontuários médicos dos pacientes. Os resultados foram comparados com o uso dos testes Qui-quadrado ou o teste exato de Fisher. Os valores de Odds Ratio (OR) e intervalo de confiança a 95% também foram calculados. O valor p igual ou menor que 0,05 foi considerado significante. Resultados: Dos 73 pacientes selecionados, 53 (72,6%) eram do sexo masculino e 20 (27,4%), femininos. A média de idade foi igual a 49,4 e não diferiu entre os sexos (p=0.337). As espondiloartrites encontradas entre os 73 pacientes foram: espondilite anquilosante (n=47; 64,4%), espondiloartrite psoriásica (n=9; 12,3%), espondiloartrite indiferenciada (n=9; 12,3%), espondiloartrite enteropática (n=6; 8,2%) e artrite reativa (n=2; 2,7%). A média de idade de início dos sintomas foi igual a 39,1 (±11.7) e não diferiu entre os sexos (p=0,9057). Do total, 35 (47.9%) pacientes eram HLA-B*27 positivo e 38 (52.1%), negativos. Este gene foi associado positivamente à espondilite anquilosante (OR: 5.37; IC 95%: 1.813-15.905; p=0.003) e negativamente à espondiloartrite enteropática (OR: 0.07; IC 95%: 0.003-1.301; p=0.025). A sacroiliíte se associou à presença do gene (OR: 10.552; IC 95%: 1.260-88.256; p=0.014) e o comprometimento intestinal à ausência (OR: 0.195; IC 95%: 0.038-0.978; p=0.048). Conclusões: O gene HLA-B*27 foi associado à espondilite anquilosante, mas não à espondiloartrite enteropática. Os sinais radiológicos de sacroiliíte prevaleceram nos pacientes positivos para o gene HLA-B*27, enquanto o comprometimento intestinal foi associado à ausência deste gene.

Resumo: A Síndromede Saethre-Chotzen(SSC,Acrocefalossindactiliatipo 111) é uma heredopatia autossômica dominante, com alta penetrância e expressividade variável, sendo craniossinostose um de seus principais sinais clínicos. Mutações nos genes FGFR1-FGFR3 e Twist são causas conhecidas de craniossinostoses, a primeira por aumento da expressão e a última por haploinsuficiência. O gene Twist foi mapeado na região 7p21 e algumas deleções e translocações balanceadas, bem como mutações no exon 1 deste gene, têm sido descritas nas formas de craniossinostose associadas a SSC. Quatro famílias brasileiras com ACS 111foram avaliadas do ponto de vista dismorfológico e rastreadas para mutações no gene Twist. O protocolo de investigação incluiu exame dismorfológico dirigido, estudo de imagem (radiografia de crânio, tórax, coluna vertebral, tomografia computadorizada de crânio e ultrassonografia abdominal), estudo citogenético com bandamento G e seqüenciamento direto de DNA dos produtos de PCR amplificados para o exon 1 do gene Twist, nos indivíduos que apresentavam ao menos um sinal clínico desta condição. Ao exame, observou-se importante variabilidade de características faciais, incluindo assimetria facial (20/24), braquicefalia (16/24), orelhas com acentuação do ramo da hélice (15/24) e implantação baixa (13/24), hipoplasia malar (13/24), ponte nasal proeminente (13/24), implantação baixa dos cabelos na fronte (12/24), ptose palpebral (12/24).Achados adicionais incluiram sindactilia cutânea parcial dos artelhos (18/24), clinodactilia do 50 dedo (13/24), háluces alargados (13/24). Somente em umafamília observou-se sindactilia cutânea total, entre o 20 e 30 artelhos. Sete indivíduos apresentaram radiografia de crânio com craniossinostose bicoronal, um, craniossinostose unicoronal e dois com forame parietal bilateral. O sinal da prata batida apareceu nos oito casos. A radiografia de tórax em dois indivíduos mostrou fusão parcial dos 10 e 20 arcos costais à esquerda e na radiografia de coluna lombo-sacra dois casos apresentaram espinha bífida lombar. O exame de cariótipo dos propósitos de cada família foi normal e o estudo molecular nãodetectou alterações no exon 1 do gene Twist. Esse estudo reforça a importância da avaliação dismorfológica em SSC e sugere que o emprego de outras técnicas de biologia molecular poderia contribuir para elucidar o papel do gene Twist nos achados clínicos desta casuística. Além disso, os dismorfismos detectados poderiam, ainda, ser decorrentes de outros genes envolvidos na regulação do desenvolvimento craniofacial e de membros
Abstract: Saethre-Chotzensyndrome(acrocephalosyndactyly type 111)is an autosomal dominant craniosynostosis condition, with high penetrance and variable expressivity. Mutations in the Fgfr1-Fgfr3 and Twist genes are known to cause craniosynostosis, the former by constitutive activation and the latter by haploinsufficiency. The Twist gene maps to 7p21 and mutations in the gene have been reported in the SCS form of craniosynostosis. The aims of this study were characterize the dysmorphological variability and mutations in exon 1 of Twist gene using direct sequencing in four Brazilian famílies presenting SCS. Twenty-four patients were included in our study, diagnosed as having features of Saethre-Chotzen syndrome. The phenotypic characteristics of ali patients were inventoried. Also, a DNA test had been performed and their genotype was noted. Facial features, present were facial asymmetry (20/24), brachycephaly (16/24), small ear with prominent crura (15/24), maxillary hypoplasia (13/24), lowset ears (13/24), and ptosis of the eyelids (12/24), lowset frontal hairline (12/24), ocular hypertelorism (11/24). Additional findings included partial hands and feet cutaneous syndactyly (18/24), clinodactyly (13/24), and broad great toes (13/24); in one family was observed total cutaneous syndactyly in feet. Skull X-rays were abnormal in 8 patients in which it was performed;partiaI fusionof 1st and 2ndribs wasdetectedin 2 individuaisand 2 had lumbar spina bifida. Chromossomal analysis on GTG-banding were normal. The analysis was carried out by direct DNA sequencing of PCR amplified products for exon 1 in the proband of each family. No Twist mutations were found. In conclusion, this four SCS famílies may have mutations in other genes of the same developmental pathway. This study reinforces the importance of the dysmorphological evaluation in patients with craniosynostosis, as well as ali their famílies, especially ACS 111,in which inter and intrafamilial variability make the diagnosis more difficult

Full textThe cold shock proteins belong to a family of proteins presenting a highly conserved domain, called cold shock domain (CSD). They are involved in various cellular processes, including adaptation to low temperature, nutritional stress, cell growth and stationary phase. In C. crescentus, a non-pathogenic α-proteobacteria, the cold shock proteins CspC and CspD have two CSDs and they are induced during stationary phase. This study aims to determine the transcription factors and cellular signals involved in cspC gene regulation in C. crescentus. In the present study we scanned a library of 4000 mutant clones with the Tn5 transposon, from which seven mutants were identified presenting reduced expression of cspC: CCNA03569 (hypothetical protein); CCNA02510 (polysaccharide deacetylase); CCNA01594 (ribosomal protein L11 methyltransferase); CCNA02186 (acetolactate synthase 3 regulatory subunit); CCNA00084 (bifunctional phosphoribosylaminoimidazolecarboxamide formyltransferase/IMP cyclohydrolase); CCNA03616 (sulfite reductase (NADPH)) e CCNA01448 (fructose 1,6-bisphosphatase II). Expression assays in the presence of a conditioned medium, showed that cspC and cspD are apparently not induced in response to increased cell density. The phenotype of the mutant cspC was evaluated as to oxidative stress resistance in the stationary phase. The results showed that the DcspC strain is highly sensitive to hydrogen peroxide and superoxide but is not sensitive to organic hydroperoxide. The absence of cspC probably is compensated by dps, since the expression of this gene is increased in DcspC strain. In contrast, the transcritpion of katG is decreased, but the activities of KatG and SodB are not affected in the cspC mutant. Under conditions of stress caused by hydrogen peroxide, sucrose or salt, cspC expression is not affected. The sigma factors sigmas SigT and SigU and transcription regulator Fur are not involved in the regulation of cspC, but cspC expression is increased in DsigJ and decreased in DoxyR strains, respectively. It was also determined that cspC shows a positive autoregulation, which may occur via stabilization of its own mRNA.

Full textThe wide spectrum of candidiasis and its clinical importance encourage the research with the purpose of clarifying the mecanisms of pathogenicity and identification of virulence factors of Candida sp. Therefore, the aim of this study was to verify through phenotypic and molecular assays the adhesion, enzymatic activity e genetic variability of clinical C. albicans and C. tropicalis isolates. The adhesion ability to the extracellular matrix glycoproteins laminin and fibronectin was evaluated using the ELISA technique (Enzyme-linked imunosorbent assay). The research of proteases was carried out in agar plate containing bovine albumin and through a quantitative method in buffer solution containing hemoglobin. The presence of ALS2, ALS3, SAP1, SAP3 and PLB1 was verified using polimerase chain reaction (PCR) and intra and interespecies polimorphisms through Random Amplified Polymorphic DNA (RAPD) technique. All C. albicans and C. tropicalis isolates binded to immobilized laminin and fibronectin. Ca33 and Ct13 isolates had relative adhesion index significantly higher than the other isolates for both glycoproteins (p < 0,001). Protease activity was observed in all isolates of C. albicans using either the semi-quantitative or quantitative assay. The protease activity of C. tropicalis was better detected through the quantitative assay. The amplification of genes related to adhesion (ALS2 and ALS3), proteases (SAP1 and SAP3) and phospholipase (PLB1) activity using PCR was observed in all C. albicans strains. PCR amplification products were not observed in C. tropicalis isolates for the researched genes. The genetic variability by RAPD revealed an heterogeneous population in both species. Nevertheless, C. tropicalis presented higher genetic variability than C. albicans strains.

O vírus da cinomose canina (VCC) é um vírus RNA, que pertence ao gênero Morbillivírus e família Paramyxoviridae. A capacidade de resposta imune, assim como sua virulência são fatores críticos para a invasão viral dos tecidos epiteliais e do sistema nervoso central (SNC). O VCC é o maior responsável pelas encefalites em cães, acometendo diversas idades. O objetivo desse estudo foi detectar o VCC nos cães com sinais neurológicos encaminhados para necropsia no Laboratório de Patologia Veterinária da Universidade Federal de Mato Grosso (LPV-UFMT). Durante um período de um ano, 85% (68/80) dos cães necropsiados tinham lesões microscópicas compatíveis com encefalomielite causada pelo VCC. Desses, 67.6% (46/68) foram confirmadas positivas através da imuno-histoquímica (IHQ). Microscopicamente, as lesões do SNC foram classificadas em encefalite desmielinizante aguda em 15.2% (7/46) dos cães, em subaguda em 73.9% (34/46) e crônica em 10.8% (5/46) dos cães. O cerebelo foi principal órgão a apresentar marcação positiva na IHQ (97.8%). O VCC é responsável pelos sinais neurológicos em cães principalmente abaixo de um ano de idade. A cinomose demonstrou sua relevância dentro da população canina de Cuiabá, sendo necessário ao nosso entendimento, caracterizar a estirpe viral relacionada à região.
Canine distemper virus (CDV) is a RNA virus classified under the genus Morbillivirus within the family of Paramyxoviridae. The time of onset of the immune response and, likely, also the virulence of the virus are critical factors in the extent of viral invasion of epithelial tissues and of the central nervous system. The CDV is the most responsible of encephalitis in dogs from different ages. In this study, the aim was to detected CDV in dogs with neurologicals signs referred for necropsy at the Laboratory of Veterinary Pathology, Federal University of Mato Grosso (LPV-UFMT).Over a period of 1 year, 85% (68/80) of the dogs necropsied had microscopic lesions compatible encephalomyelitis by CDV. Which 67.6% (46/68) were confirmed by immunohistochemistry (IHC). Microscopically, the CNS lesions were classified demyelinating encephalitis in 15.2% (7/46) to acute, 73.9% (34/46) in subacute and 10.8% (5/46) to chronic. The cerebellum (97.8%) was the main target organ to verify positivity in the IHC. Canine distemper virus is a pathogen responsible for neurological clinical signs in dogs mainly under one year of age. Distemper demonstrated its relevance within the canine population of Cuiabá, being necessary to our understanding, characterizing the viral strain related to the region.

Full textBy using a new concept, which is based on Paraconsistent Logic (LPA) and has not yet been applied for classification, this work aims at constructing an intelligent system for Music Genre Classification (MGC). This topic, that is emergent in the literature, has received an increasing attention from the scientific community due to its applicability, emphazising both a commercial potential to commercialize multimedia content on the Internet and data mining tasks involving music signals. By adopting a database formed by samples of songs, which represent different styles of music, such as jazz, bolero, bossa nova, forró, salsa and sertanejo, and a discriminative paraconsistent classifier, a supervised procedure is used to solve the problem. The system is divided in two modules. The first extracts features from the music files, based on the concepts of time-frequency analysis and crictical bands of the human ear. On the other hand, the second implements the proposed classifier, which allows an efficient treatment of contradictions in such a way that is more similar to the human brain. The results obtained, when compared with existing approaches used to MGC, demonstrate how LPA is suitable for this purpose. Additionally, this is the original contribution to the state-of-the-art: the use of LPA for MGC, an inexistent approach up to date.

This research aims to conduct a descriptive analysis of the performance of LIBRAS (Brazilian Sign Language) / Portuguese translator interpreter (TILSP), in the journalism genre and in the sphere of television activity, from the work experience of the author of this research as an interpreter in this kind of discourse. For this, we chose as corpus a public domain television journalistic production, consisting of an edition cut of Programa Sentidos with the libras interpretation during it. The show is broadcasted by a cable television channel and then is available on the internet. We question which verbal-visual elements contribute and / or interfere with the libras interpretation of the journalistic genre, in the television sphere; how these elements affect the translations choices and lexical and syntactic construction in the libras interpretation from this genre, and which sense effects are produced from the interference of these elements in the process of Portuguese-LIBRAS interpretation. In this sense, The Circle of Bakhtin will serve as theoretical foundations of our research, and the comprehension of the Bakhtinian concepts of exotopy, chronotope, speech genres, enunciation, concrete utterance, text, discourse, authorship and extraverbal situation are the guidelines for the corpus analysis, which was transcribed and described from an exotopic movement shift, from the analyzed subject to the researcher, in a way that the researcher could look at himself taking into consideration the linguistic and extralinguistic elements of the target language in the act of translation/interpretation. The target language, LIBRAS, was transcribed using the Sign Language transcription system ELAN (EUDICO Language Annotator), developed by Max Planck Institute for Psycholinguistics and used on the research group Estudos da Comunidade Surda: Língua, Cultura e História (Deaf Community Studies: Language, Culture and History) in the University of São Paulo and in the course de Letras/Libras (Language Studies/Libras) of the Federal University of Santa Catarina.The analysis showed that the verbal-visual elements and images of all the reports are factors of libras linguistic marks modification, as look and body direction, and that these images are crucial for the negotiation of discursive meanings from this sphere at the time of interpretation. The incorporation of shapes and outlines of the images during signaling TILSP showed us that the whole verbal-visual component of that sphere, not only interferes with the interpretation of sign language, but also contributes to the realization of TILSP interpretive strategies aiming the transmission of senses brought by the enunciators discursive project in this genre. From the analysis we propose a theoretical and methodological possibility of referral to TILSP training and working from an enunciative-discursive perspective, adopting the analysis of Programa Sentidos as an orientation for this proposal. To this end, we used as guidelines some television journalistic productions, taking into account the possibility of future insertions of TILSP in this sphere of discourse production and realization of interpretative practices in the journalistic genre, suggesting the expansion of its operations in other genres circulating in this sphere
Este trabalho tem por objetivo realizar uma análise descritiva da atuação do tradutor intérprete de libras/português (TILSP) no gênero jornalístico e na esfera de atividade televisiva a partir da experiência de atuação do autor desta pesquisa como intérprete nesse gênero do discurso. Para isso escolhemos como corpus uma produção telejornalística, de domínio público, constituída por um recorte de uma edição do Programa Sentidos, veiculado por meio de um canal de TV a cabo, disponibilizado na internet posteriormente, com interpretação para a libras durante a exibição. Questionamos que elementos verbo-visuais colaboram e/ou interferem na interpretação da libras no gênero jornalístico, esfera televisiva; como esses elementos afetam as escolhas tradutórias e a construção lexical e sintática na interpretação da libras a partir desse gênero; e quais efeitos de sentidos são produzidos a partir da interferência desses elementos no processo de interpretação do português para a libras. Nesse sentido, fundamentamos a pesquisa na teoria do Círculo de Bakhtin compreendendo os conceitos de exotopia, cronotopo, gêneros do discurso, enunciação, enunciado concreto, texto, discurso, autoria e situação extraverbal como fios condutores da análise do corpus que foi transcrito e descrito, a partir de um movimento exotópico de deslocamento de sujeito analisado para o de pesquisador, de forma que o pesquisador olhe a si mesmo, considerando os elementos linguísticos e extralinguísticos da língua alvo no ato tradutório/interpretativo, a libras, por meio do sistema de transcrição de língua de sinais ELAN (EUDICO Language Annotator) desenvolvido pelo Max Planck Institute for Psycholinguistics e utilizado no grupo Estudos da Comunidade Surda: Língua, Cultura e História na Universidade de São Paulo e no curso de Letras/Libras da Universidade Federal de Santa Catarina. A análise mostrou que os elementos verbo-visuais e a totalidade das imagens das reportagens são fatores de modificação das marcas linguísticas da libras como direção do olhar e do corpo e que essas imagens são decisivas para a negociação de sentidos discursivos provenientes dessa esfera no momento da interpretação. A incorporação das formas e delineios das imagens durante a sinalização do TILSP nos mostraram que a totalidade verbo-visual, constituinte dessa esfera, não só interfere na interpretação da língua de sinais, mas também colabora para que o TILSP realize estratégias interpretativas objetivando a transmissão dos sentidos instaurados pelo projeto discursivo dos enunciadores desse gênero. A partir da análise propomos uma possibilidade de encaminhamento teórico metodológico para a formação e atuação do TILSP a partir de uma perspectiva enunciativo-discursiva, adotando a análise do Programa Sentidos como norteador desta proposta. Para tanto, utilizamos como fio condutor algumas produções telejornalísticas considerando a possibilidade de futuras inserções dos TILSP nessa esfera de produção do discurso e a realização de práticas interpretativas a partir do gênero jornalístico, sugerindo a expansão de sua atuação em outros gêneros circulantes nessa esfera

Objetivou-se com este estudo estimar par?metros gen?ticos e fenot?picos para probabilidade de prenhez nas idades 15, 19, 23, 27, 31, 35, 39 e 43 meses (PP_15, PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 e PP_43), em novilhas da ra?a Sindi, via modelo de limiar em regress?o aleat?ria utilizando os polin?mios ortogonais de Legendre de segunda, terceira e quarta ordem, considerando a homogeneidade e heterogeneidade de vari?ncia residual. Os componentes de covari?ncia foram estimados por abordagem Bayesiana, utilizando o programa THRGIBBS3F90. Foram inclu?dos os efeitos fixos de criador, fazenda do criador, ano de nascimento, esta??o de nascimento e como covari?vel (de efeito linear) foi considerado o peso da vaca na idade em que ela concebeu. Al?m desses efeitos para todas as idades estudadas, foram considerados os efeitos aleat?rios gen?ticos aditivos, de ambiente permanente e residual. O melhor modelo para a an?lise RRTM foi o LEG_4441 que obteve as seguintes estimativas de herdabilidade 0,34; 0,41; 0,45; 0,41; 0,37; 0,32; 0,32; 0,33 para PP_15, PP_19, PP_23, PP_27, PP_31, PP_35, PP_39, PP_43, respectivamente. As estimativas das correla??es gen?ticas obtidas entre PP_15 com PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 e PP_43 foram 0,1; -0,39; -0,65; -0,83; -0,93; -0,81; -0,71, respectivamente. As estimativas das correla??es fenot?picas para PP_15 com PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 e PP_43 foram respectivamente 0,12; -0,09; -0,17; -0,22; -0,26; -0,25; 0,40. A an?lise que utilizou modelo LEG_4441 foi a mais indicada para estimar os par?metros gen?ticos para a probabilidade de prenhez em diferentes idades em novilhas da ra?a Sindi, indicando que a prenhez precoce possui variabilidade gen?tica para ser inclu?da como caracter?stica alvo de sele??o em programas de melhoramento, com potencial para se obter ganhos gen?ticos satisfat?rios.

Our aim with this study was estimating genetic and phenotypic parameters for pregnancy probability at 15, 19, 23, 27, 31, 35, 39 and 43 month old (PP_15, PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 and PP_43) in Sindhi heifers, by using threshold model in a random regression framework, adopting Legendre orthogonal polynomials of second, third and fourth order, considering homogeneity and heterogeneity of residual variance. Covariance components were estimated by Bayesian approach using THRGIBBS3F90 program. The model included the fixed effects of herd, herd of the farm, year of birth, birth season and as a covariate (with linear effect) was considered the weight of the cow at the age when she calved. In addition to these effects for all ages studied, additive genetic, permanent environmental and residual random effects were considered. The best model for the analysis was the RRTM LEG_4441 which obtained the following heritability estimates: 0.34; 0.41; 0.45; 0.41; 0.37; 0.32; 0.32; 0.33 to PP_15, PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 and PP_43, respectively. Genetic correlation estimates between PP_15 with PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 and PP_43 were 0.1; -0.39; -0.65; -0.83; -0.93; -0.81; -0.71, respectively. Phenotypic correlation estimates for PP_15 with PP_19, PP_23, PP_27, PP_31, PP_35, PP_39 and PP_43 were respectively 0.12; -0.09; -0.17; -0.22; -0.26; -0.25; 0.40. The analysis by using LEG_4441 model was the most appropriate to estimate genetic parameters for pregnancy probability at different ages in Sindhi heifers, indicating that early pregnancy has genetic variability to be included as a goal trait selection in breeding programs, with the potential to achieve satisfactory genetic gains.

Full textAn important topic in biological sequences analysis is gene finding, i.e. the identification of protein coding regions. This identification allows the posterior research for meaning, description or biological categorization of the analyzed organism. Currently, several methods combine pattern recognition with knowledge collected from training datasets or from comparison with genomic databases. Nonetheless, the accuracy of these methods is still far from satisfactory. New methods of DNA sequences processing and genes identification can be created through search-by-content such sequences. The periodic pattern of DNA in protein coding regions, called three-base periodicity, has been considered proper of coding regions. Digital signal processing techniques supply a strong basis for regions identification with three-base periodicity. In this work, we present a bioinformatics pipeline, basic concepts of the genomic identification and digital signal processing methods used for protein coding regions identification. We introduce a new method for identification of these regions, based on a newly proposed transform, called Modified Morlet Transform. We present some obtained experimental results from synthetic and real DNA sequences. The main contributions consist of the bioinformatics pipeline development for genoma projects and the creation of a method for protein coding regions identification where the three-base periodicity is latent. The method presents superior performance and important advantages in comparison to traditional method based on the short time Fourier transform.

Wavelet coding is an efficient technique to overcome the multipath fading effects, which are characterized by fluctuations in the intensity of the transmitted signals over wireless channels. Since the wavelet symbols are non-equiprobable, modulation schemes play a significant role in the overall performance of wavelet systems. Thus the development of an efficient design method is crucial to obtain modulation schemes suitable for wavelet systems, principally when these systems employ wavelet encoding matrixes of great dimensions. In this work, it is proposed a design methodology to obtain sub-optimum modulation schemes for wavelet systems over Rayleigh fading channels. In this context, novels signal constellations and quantization schemes are obtained via genetic algorithm and mathematical tools. Numerical results obtained from simulations show that the wavelet-coded systems derived here have very good performance characteristics over fading channels
A codifica??o por matrizes wavelets tem se mostrado um m?todo eficiente para combater o desvanecimento, fen?meno que causa flutua??es na intensidade do sinal transmitido em um canal de comunica??o sem fio, devido ? propaga??o por m?ltiplos percursos. Mas como os s?mbolos codificados pelo sistema wavelet s?o n?o-equiprov?veis, os esquemas de modula??o influenciam de maneira fundamental o desempenho desse sistema. Por isso se torna essencial um m?todo eficaz para a obten??o desses esquemas de modula??o de forma a otimizar o desempenho do sistema wavelet, principalmente quando se emprega matrizes wavelets de grandes dimens?es. Esse trabalho aborda o projeto de esquemas de modula??o para um sistema de transmiss?o sem fio baseado na codifica??o por matrizes wavelets em canais com desvanecimento Rayleigh plano. Para o projeto desses esquemas de modula??o s?o criados novas constela??es e esquemas de quantiza??o. O projeto desses esquemas de modula??o ? guiado por um algoritmo gen?tico. We investigated the syndromes of the Sini decoction pattern (SDP). To obtain a common profile for SDP, we distributed questionnaires to 300 clinical traditional Chinese medicine practitioners. According to the survey, we concluded 2 sets of symptoms for SDP: (1) pulse feels deep or faint and (2) reversal cold of the extremities. Then we collected 24 individuals, and extracted their total mRNA of peripheral blood mononuclear cells for cDNA microarray experiments. 12 samples were used as the training set to identify the biomarkers for distinguishing the SDP and non-SDP groups. The remaining 12 samples were used as the test set. The test results indicated that the gene expression profiles of the identified biomarkers could effectively distinguish the 2 groups.

Full textA terapia génica envolve a transferência de material genético terapêutico para células alvo, pela introdução de genes funcionais que substituam ou complementem aqueles que se encontram defeituosos, com o objectivo de tratar ou prevenir uma ampla gama de doenças, hereditárias ou adquiridas. No entanto, para o seu sucesso é necessário um sistema de entrega de material genético eficiente, capaz de proteger o DNA da degradação por nucleases e com o mínimo de toxicidade e imunogeneicidade que permita uma expressão genética estável e duradoura. Durante os últimos anos, têm sido desenvolvidos uma ampla gama de vectores, que têm sido divididos e caracterizados como vectores virais e não virais. Os vectores virais apresentam maior eficiência na transferência de material genético, tanto in vitro como in vivo, no entanto apresentam algumas limitações como a reduzida capacidade de empacotamento genético e o facto de conduzirem a respostas inflamatórias/imunológicas indesejáveis que consequentemente limitam administrações subsequentes. Por sua vez, os vectores não virais apresentam algumas vantagens sobre os vectores virais, nomeadamente, um perfil imunológico mais seguro, uma produção mais fácil, uma maior capacidade de empacotamento genético e a sua reduzida toxicidade. Contudo, o uso de vectores não virais é limitado devido às suas eficiências de transfecção relativamente baixas, marcadas pela baixa translocação nuclear, e consequentemente reduzida expressão genética. Vários esforços têm sido realizados no sentido de ultrapassar a barreira nuclear, um dos maiores passos limitantes no desenvolvimento de sistemas de entrega genética não virais eficazes. Uma das estratégias passa pela incorporação de sinais de localização nuclear em complexos poliméricos, uma vez que estes péptidos catiónicos ao serem reconhecidos pelas importinas permitem um transporte genético eficaz para o núcleo através dos complexos de poros nucleares, aumentando assim a entrega nuclear do DNA, e por consequente, a sua expressão genética. Neste contexto, o objectivo deste trabalho foi a caracterização e optimização de vectores não virais, baseados em polímeros como o quitosano e o ácido hialurónico, que foram escolhidos devido às suas notórias propriedades de biocompatibilidade, biodegradação e ausência de toxicidade. Péptidos baseados em sinais de localização nuclear endógenos, pertencentes á família dos IGFBP, derivados nomeadamente do IGFBP-3 e IGFBP-5, foram avaliados com o intuito de melhorar a translocação nuclear sem comprometer o perfil imunológico bastante baixo dos vectores não-virais. Várias estratégias foram testadas para avaliar a eficiência de transfecção e a expressão genética mediada por poliplexos de quitosano e ácido hialurónico em células HEK293T: nomeadamente a co-administração, a co-complexação e a ligação covalente dos péptidos IGFBP aos poliplexos de quitosano. Os nossos resultados mostraram que as nossas formulações, com ou sem sulfato de sódio, péptidos IGFBP ou ácido hialurónico, e independentemente da estratégia usada, originaram poliplexos com um intervalo de tamanhos entre 250 nm e 750 nm e carga de superfície positiva, caracterizados através de medições no Zetasizer. Os nossos poliplexos foram ainda capazes de complexar o DNA de forma eficaz, conforme analisado através de ensaios de electroforese em gel de agarose. Posteriormente à caracterização dos poliplexos, os péptidos IGFBP foram ainda avaliados quanto á sua citotoxicidade em dois períodos de incubação, 24 horas e 72 horas. Os ensaios de viabilidade celular não mostraram qualquer citotoxicidade para ambos os péptidos IGFBP para as várias concentrações testadas nos dois períodos de tempo testados. Após estes resultados, e uma vez que os poliplexos apresentaram características desejáveis quanto ao seu tamanho, carga de superfície e complexação eficiente do DNA, estes foram avaliados quanto à sua eficácia através de ensaios de transfecção in vitro, analisados posteriormente por microscopia de fluorescência e citometria de fluxo. A eficiência de transfecção revelou-se ser dependente da concentração dos péptidos IGFBP e variar consoante o método de entrega utilizado. Nos ensaios de transfecção, utilizando o método de co-administração dos poliplexos com os péptidos IGFBP, nenhum aumento na eficiência de transfecção foi observado. Nos métodos em que estratégias como a co-complexação e a ligação covalente dos péptidos IGFBP aos poliplexos de quitosano foram usadas, um aumento significativo da eficiência de transfecção foi conseguido, no entanto, apenas para os poliplexos associados ao IGFBP-3. Uma possível explicação para estes resultados é o facto de as acessibilidades e/ou afinidades para com as subunidades das importinas diferirem entre os péptidos IGFBP-3 e IGFBP-5, o que consequentemente, poderá levar a níveis de translocação diferentes entre os péptidos, no entanto, ainda não é claro qual o mecanismo. Foi ainda possível verificar que combinando polímeros como o quitosano e o ácido hialurónico, que os poliplexos resultantes renderam um aumento significativo da eficiência de transfecção para ambos os péptidos, IGFBP-3 e IGFBP-5, o que poderá ser explicado por uma possível modificação no enrolamento das cadeias do quitosano aquando da adição do ácido hialurónico. Na sua globalidade os resultados obtidos demonstraram que, apesar de ser ainda necessário uma optimização da eficiência de transfecção, poliplexos co-complexados com péptidos IGFBP são de facto bons candidatos a sistemas de entrega genética não virais. Os poliplexos com dois polímeros combinados, quitosano e ácido hialurónico, foram os que revelaram maiores eficiências de transfecção, para ambos os péptidos, IGFBP-3 e IGFBP-5. Futuramente, seria interessante não só expandir a gama de concentrações de péptidos IGFBP testadas como também o tipo de linhas celulares, compreendendo ainda uma optimização das formulações dos poliplexos.
Gene therapy entails the transfer of therapeutic genetic material into specific cells; however, their success requires an efficient gene delivery system, which allows a stable gene expression. Nuclear import is considered the major limiting step in the development of effective non-viral gene delivery systems; the incorporation of NLS that can mediate nuclear intake can be used as a strategy in order to enhance the internalization of DNA into the nucleus. In this work, an endogenous NLS peptide, based on IGFBP, namely IGFBP-3 and IGFBP-5, was evaluated in order to ameliorate nuclear translocation without compromising the fairly low immunological profile of non-viral vectors. Several strategies were tested to determine their effect in chitosan and acid hyaluronic polyplex-mediated transfection efficiency in HEK293T cells: co-administration, co-complexation, and covalent ligation to chitosan polyplexes. Our results show that our vectors are capable of an effective DNA complexation and present size and surface charge appropriated for gene delivery applications. Transfection efficiency is concentration dependent and varies with the delivery method employed. Co-complexation and covalent ligation of IGFBP peptides to chitosan polyplexes yielded a 2-fold increase in transfection efficiency associated with the use of IGFBP-3 peptides. The incorporation of acid hyaluronic yielded a significant increase in transfection efficiency to both peptides. Despite of the improvements in transfection efficiency it needs to be further improved, these results indicate that polyplexes co-complexed with IGFBP peptides are good candidates for non-viral gene delivery systems and would be interesting to expand the range of tested IGFBP peptides concentrations as well as the type of cell lines used.
