Abstract:
Abstract Nysius simulans (Stål) is a suctorial, fluid feeding herbivore that can transmit toxins and spread pathogens via saliva and is an economically important pest for soybean in South America. Currently, N. simulans in soybean is predominantly found in Argentina, but future changes in the distribution from both dispersal and range shifts due to climate change may affect soybean cultivation in southern South America. We developed a species distribution model to examine the distribution range of N. simulans. We compared the potential distribution of N. simulans under current and future projected climatic conditions in order to identify future areas of natural occurrence with ecological niche models using Maxent. Current records of N. simulans show that while the species is present in Argentina, and some areas of Brazil, Paraguay, Peru, and Uruguay, our models suggest that many new suitable areas will be available for N. simulans under climate change including other regions of Argentina, and southern Chile. Our results also predict potential future range shifts and distributions into Bolivia, but not Peru nor Brazil. In our model, seasonal trends in temperature were shown to have the greatest contribution to the potential distribution, whereas isothermality (i.e., temperature variability) was correlated to potential future distribution ranges. We conclude that current populations of N. simulans may be expanding its distribution range by diffusion (i.e., range expansion over generations at the margins of populations), and regions with potential future N. simulans distribution should be closely monitored.

Abstract:
AbstractWe analyze patterns of nucleotide variability at 15 X-linked loci and 14 autosomal loci from a North American population of Drosophila simulans. We show that there is significantly more linkage disequilibrium on the X chromosome than on chromosome arm 3R and much more linkage disequilibrium on both chromosomes than expected from estimates of recombination rates, mutation rates, and levels of diversity. To explore what types of evolutionary models might explain this observation, we examine a model of recurrent, nonoverlapping selective sweeps and a model of a recent drastic bottleneck (e.g., founder event) in the demographic history of North American populations of D. simulans. The simple sweep model is not consistent with the observed patterns of linkage disequilibrium nor with the observed frequencies of segregating mutations. Under a restricted range of parameter values, a simple bottleneck model is consistent with multiple facets of the data. While our results do not exclude some influence of selection on X vs. autosome variability levels, they suggest that demography alone may account for patterns of linkage disequilibrium and the frequency spectrum of segregating mutations in this population of D. simulans.

Abstract:
AbstractThe origins and divergence of Drosophila simulans and close relatives D. mauritiana and D. sechellia were examined using the patterns of DNA sequence variation found within and between species at 14 different genes. D. sechellia consistently revealed low levels of polymorphism, and genes from D. sechellia have accumulated mutations at a rate that is ∼50% higher than the same genes from D. simulans. At synonymous sites, D. sechellia has experienced a significant excess of unpreferred codon substitutions. Together these observations suggest that D. sechellia has had a reduced effective population size for some time, and that it is accumulating slightly deleterious mutations as a result. D. simulans and D. mauritiana are both highly polymorphic and the two species share many polymorphisms, probably since the time of common ancestry. A simple isolation speciation model, with zero gene flow following incipient species separation, was fitted to both the simulans/mauritiana divergence and the simulans/sechellia divergence. In both cases the model fit the data quite well, and the analyses revealed little evidence of gene flow between the species. The exception is one gene copy at one locus in D. sechellia, which closely resembled other D. simulans sequences. The overall picture is of two allopatric speciation events that occurred quite near one another in time.

This master’s thesis deals with the design of a robotic cell for the automation of woodworking machine tending. The task of the robot is to manipulate with the specified laminated chipboards of various sizes, before and after machining. The boards are brought to the cell on pallets in three possible placement variants. After a brief research part, conceptual designs of cell layout were created. The optimal and further elaborated design came as a result of selected evaluation criteria. The next part of the thesis consists of designs and selection processes of individual components. These were later used to create a simulation model in Tecnomatix Process Simulate software (Siemens), that helped to verify the functionality of the cell layout, design the control logic and acquire the time of the manipulation cycle used for informative calculation of annual production. In the end, the final design was evaluated from an economic point of view with an emphasis on the return of the initial investment.

Abstract:
In this thesis, called a simulation model of population growth, there was created the simulation model of a population growth in the Czech Republic to the year 2052. First there was constructed the projection through the component method used in demography. Deterministic values obtained by this method are then used in the simulation analysis. In the simulation analysis there was conducted an experiment with three variables which influence the population growth (the fertility, life expectancy/the coefficient of the decline of the probability of the death/ and the migration balance). With the support of the Crystal Ball, supporting program for Excel, which provides the possibility of the simulations, there was determined the stochastic character of the three variables. There are created the low, medium and high variant of the projections, as well as three other possible variants of population growth that might occur. These variations are called economic crisis, friendly migration policy and restrictive migration policy.

Abstract:
Altered surface and viscoelastic properties of mucus during respiratory diseases have a strong influence on its clearance by cilia and cough. Combined effect of the surface tension and viscosity with relatively unchanged storage modulus of simulated airway mucus (simulant) considered as the capillary number on displacement of 0.3mL simulant aliquot during simulated cough through a model adult human trachea at relatively low cough velocity (15±0.5m/s) of 0.3s duration was investigated. Displacement of simulant aliquots decreased significantly with capillary number (p<0.0001). The interactive effects of viscosity and surface properties may give insight into understanding mucus displacement at low cough velocities in patients with chronic obstructive pulmonary disease (COPD), cystic fibrosis (CF) and asthma.

Abstract:
This paper presents a drilling and coring device for the lunar exploration, which is possibly utilized to acquire the lunar regolith with a certain depth. The drilling device is composed of three components: rotary unit, percussive unit and penetrating unit. The rotary-percussion drill can work in two different operating modes: rotary mode and rotary-percussive mode, depending on the properties of cut object. In the relatively loose regolith, rotation and penetration can make the drill work in a well state. However, once rock is encountered in the drilling process, besides rotation and penetration, percussion must be launched to reduce the drilling power and the required penetrating force. Due to the indetermination of the lunar environment, it is not easy to control the coring drill to adapt to the encountered conditions. To obtain a high coring ratio with relatively low power, an intelligent drilling strategy is inevitably proposed to accomplish the drilling process control. Considering the lunar soil simulant should cover the possible composition of real lunar soil, simulant are classified into several levels based on the generalized drillability. For each level of drillability of lunar soil simulant, experiments are conducted to get the characteristics in frequency-domain of rotary torque output. The sampled characteristics of rotary torque output are utilized to train the object-recognition system based on Support Vector Machine (SVM). Information in all the levels of drillability of lunar soil simulant is stored in the object-recognition system as an expert system. To understand the properties of the drilling object, rotary torque is selected to identify the level of drillability of simulant in drilling process. Subsequently, once the level is obtained, drilling strategy is adjusted to adapt to the current level correspondingly in real time. Experiments are conducted to verify the intelligent drilling strategy successfully.

Abstract:
Experiments were conducted on the flow field start up behavior of a gelled waxy oil in a pipeline. A simulant fluid was used to mimic the low temperature rheology of crude oil. The break down of the gelled simulant fluid was studied during different startup conditions. It is shown that the “failure mode,” or manner and location in which the gelled simulant fluid breaks down, is closely related to both the temperature of the gel and the cooling time prior to pressurization. Flow visualizations indicate that for higher temperatures, and long cooling times, exists a weaker gel strength and failure occurs near the centerline of the pipe. Lower temperatures and long cooling times result in the breakdown of the gel at the pipe wall. Shorter cooling times result in a weak centerline gel strength, and results in gel failure near the centerline of the pipe. Pressure and temperature data were acquired at seven locations along the length of the test section, and these data were correlated to the velocity field, measured using Particle Image Velocimetry. Combined with rheology measurements, these data, allowed for shear stress estimates to be made. For the parameter ranges explored, the results exhibit three different failure modes, each associated with a different set of initial conditions. A critical temperature existed above which one failure mode was encountered and below which another failure mode was found. A third failure mode was associated with a cross-section that did not have a uniform radial temperature profile.

Abstract:
Tissue simulants are commonly used in medical procedure training and research to test the insertion and cutting mechanics of medical devices. Accurate representation of the forces and tissue properties is important for the efficacy of the training and research studies. This paper provides a quantitative method of determining the performance of tissue simulants. A force model was used to determine the three component forces: tearing, spreading, and friction forces, of a needle passing through six different skin simulants. Experiments were performed to determine the fracture toughness, shear modulus, friction between the simulant and needle, and the crack length in the simulant made by the needle. Polyurethane with Shore hardness 40 A was shown to be the best simulant by having the composition of forces most similar to porcine skin tissue: 39% and 61% for the tearing force, 18% and 32% for the spreading force, and 29% and 21% for the friction force for the polyurethane and porcine skin respectively.

Clearance of mucus by the beating action of cilia is the primary means of removing inhaled particulates and airway debris from airways in healthy people. However many pulmonary diseases are associated with impaired mucociliary clearance mechanisms. For these patients, cough is the default airway clearance mechanism. Unfortunately most pulmonary disease patients can only produce low expiratory flow rates and have difficulty coughing for this reason.
