Baban Tengah Hamlet, Mulyorejo Village, Silo District is a unique village. However, in its uniqueness there are several problems. Seen in the educational aspect, elementary school institutions are very important because the first foundation of children is to become the nation's next generation. However, along with the development of the children experience moral degradation. As in Baban Tengah hamlet, there are minors who have committed criminal acts by drinking alcohol and smoking. This is of course a problem in the world of education. From this, it is necessary to preserve culture and habituation of positive activities in order to form and enhance student character with various formal, informal and non-formal learning processes. This research use desciptive qualitative approach. As for the collection of data and information in this study using observation techniques, interview documentation and triangulation. The results of this study indicate the concept of local traditions, the results of local traditions, supporting and inhibiting factors in improving the character of students. Dusun Baban Tengah Desa Mulyorejo Kecamatan Silo merupakan desa yang unik. namun di dalam keunikannya terdapat beberapa permasalahan.Terlihat pada aspek pendidikan, lembaga sekolah dasar menjadi sangat penting karena pondasi pertama anak menjadi generasi penerus bangsa. Akan tetapi seiring berkembangnya zaman anak mengalami degradasi moral. Seperti halnya di dusun Baban Tengah terdapat anak di bawah umur sudah melakukan tindakan kriminal dengan meminum-minuman keras dan merokok. Hal ini tentu menjadi permasalahan dalam dunia pendidikan. Dari hal tersebut diperlukan pelestarian budaya dan pembiasaan aktivitas positif guna pembentukan dan meniingkatkan karakter siswa dengan berbagai proses pembelajaran formal, informal maupun non formal. Penelitian ini menggunakan pendekatan deskriptif kualitatif. Adapun dalam pengumpulan data dan informasi dalam penelitian ini menggunakan teknik observasi, wawancara dokumentasi dan triangulasi. Hasil penelitian ini menunjukkan konsep tradisi lokal, hasil tradisi lokal, faktor pendukung dan penghambat dalam meningkatkan karakter peserta didik.

Abstract:

Entomopathogenic Nematodes (EpNs) that serve as biological control agents include Steinernema spp and Heterorhabditis spp. EPNs Heterorhabditis indicus (Bromo Isolate) has a high toxicity against larvae Lepidoptera and Coleoptera and was successfully developed for mass production in Biological Control Laboratory of Jember University. These nematodes will be formulated as solid and liquid biopesticides. This research aims: to find EPNs local isolates from smallholder coffee plantations; to identify local and in vivo and in vitro cultured EPNs isolates; to determine the pathogenicity of EpNs local isolate against Coffee Berry Borrer (CBB) Hypothenemus hampei Ferr and to formulate EpNs in vitro culture as a liquid and solid formula. These formula were tested for the patogenicity and viability of EpNs to the larvae of H. hampei, Tenebrio molitor and Galleria melonella. The results showed that the pathogenecity of EpNs isolate to the larvae of CBB in Silo was 30% after 24 hours and 90% after 48 hours of in vivo inoculation. However, the mortality of CBB larvae was only 10% by liquid spraying on the coffee berry. The viability was 524 IJ (Infective Juvenile) on liquid formula packed on polyurethane sponge, and this was higher than that on solid formula (330 IJ).

Abstract:

Using of silage and haylage of forage legumes in ruminant nutrition and promotion of promoting proper forage conservation techniques should be an important strategy in livestock production in our country. Forage legumes are difficult to ensile, so it is necessary to apply the starter culture of selected strains of lactic acid bacteria that support the ensiling process and prevent bacterial butyric fermentation and thus contribute to the preservation and improvement of silage and haylage quality. In this paper, the influence of bacterial inoculant ?Silko for alfalfa? on the quality of silage and haylage of alfalfa in two separate trials is presented. The inoculant is a combination of homofermentative lactic bacteria Lactobacillus plantarum and Pediococcus spp. The first-cut alfalfa in the second year was used for silage and haylage. The silage was examined in mini-silos in the laboratory, and the haylage at the cattle farm where the plant material was cuts were collected in experimental silo bags. The treatments were control (untreated silage, i.e. haylage) and silage, i.e. haylage treated with inoculant ?Silko for alfalfa? (rate of 5 ml t-1 fresh material). The silages were analyzed after 90 days, and haylage after 40. The inoculant ?Silko for alfalfa? has been found to maintain the nutritive value of silage and haylage and to improve their chemical, energy and fermentation parameters relative to the control. Since ?Silko for alfalfa? positively affects the correct lactic acid fermentation of silage and haylage and contributes to a lesser loss of nutritional value and energy it is expected that it can promote a high level of productivity of ruminants, and thus contribute to the growth of profit in livestock production.

Isolation can affect our well-being negatively. To prevent the spread of the infection COVID-19, many workers, including university lecturers, are required to work from home. In order to maintain high levels of well-being and team cohesion, academics at the University of Derby Online Learning initiated a virtual huddle to briefly socialise and check on their colleagues’ well-being every morning. This piece of field notes reports the context (COVID-19 in the United Kingdom), the details of this morning socialization, the first-hand experience of attending this huddle, and possible applications. Perceived positive impacts of our huddles include better well-being, cultivating compassion in team culture, and enhanced team cohesion. These advantages can be also useful in student supervision, wider socialization with colleagues to counter the silo mentality, and other occupational sectors. Our field notes will be helpful for lecturers and other types of employees who work collaboratively yet in isolation during this uncertain and challenging time of crisis.

Ginger (Zingiber officinale) is a rhizome plant that is very popular as a spice and medicinal ingredient. The use of ginger itself varies depending on culture. While in Indonesia, ginger roots and rhizomes are widely used for treatment. Starting from crushed, made into tea, until smeared for healing then the use of ginger root is very thick in the community environment. Seeing how popular and the various benefits that exist in the ginger plant make masangka in Harjomulyo Village, Silo District, Jember Regency, especially the women of the Al-Hasan Muslimatan Group held an instant ginger powder production activity. This is because Harjomulyo Village is a very strategic plantation area for ginger cultivation so that this plant is quite abundant in the Harjomulyo Village area. The problem that arises that is the marketing of instant ginger production has not been effective. This is due to the absence of special media as a means of promotion / marketing. Until now marketing techniques have only been carried out manually by word of mouth so that not many people know that abundant ginger plants in Harjomulyo Village are produced to become instant ginger ready to drink or as a mixture of drinks or other food ingredients. Therefore there is a need for training and assistance related to production and marketing activities in the hope that it can improve the packaging design to be more attractive so that it has a high selling value; use social media as a new means of marketing instant ginger products for women from the Al-Hasan Muslimatan Group Harjomulyo Village; using Instagram as a marketing tool; as well as using the Android-based "Follower Chief" application to automate marketing using Instagram social media.

Aim: We investigated the role of induced endothelial cells (iECs) in mesenchymal stem cells (MSCs)/iECs co-culture and assessed their osteogenic ability on silk fibroin nanofiber scaffolds. Methods: The osteogenic differentiation was assessed by the ALP assay, calcium assay and gene expression studies. Results: The osteogenic differentiation of the iECs co-cultures was found to be higher than the MSCs group and proximal to endothelial cells (ECs) co-cultures. Furthermore, the usage of isogenic iECs for co-culture increased the osteogenic and endothelial gene expression. Conclusion: These findings suggest that iECs mimic endothelial cells when co-cultured with MSCs and that one MSCs source can be used to give rise to both MSCs and iECs. The isogenic MSCs/iECs co-culture provides a new option for bone tissue engineering applications.

Corn was inoculated withLactobacillus plantarumandPropionibacterium freudenreichiisubsp.shermaniieither independently or as a mixture at ensiling, in order to determine the effect of bacterial additives on corn silage quality. Grain corn was harvested at 32–37% of dry matter and ensiled in a 4 L laboratory silo. Forage was treated as follows: bacterial types: B0 (without bacteria-control), B1(L. plantarum), B2 (P. freudenreichiisubsp.shermanii), and B3 (combination ofL. plantarumandP. freudenreichiisubsp.shermanii). Each 2 kg of chopped forage was treated with 10 mL of bacterial culture and allowed to ferment for 27 days. The first experiment determined the most suitable wavelength for detection of bacteria (490 nm and 419 nm for B1 and B2, resp.) and the preferable inoculation size (1 × 105 cfu/g). The second experiment analysed the effect of B1 and B2 applied singly or as a mixture on the fermentation characteristics and quality of corn silage.L. plantarumalone increased crude protein (CP) and reduced pH rapidly. In a mixture withP. freudenreichii, the final pH was the lowest compared to other treatments. As a mixture, inclusion of bacteria resulted in silage with lower digestibility than control. Corn silage treated withL. plantarumorP. freudenreichiieither alone or mixed together produced desirable silage properties; however, this was not significantly better than untreated silage.

Can an integrated territorial approach successfully do away with the silo structure that marks public action through the hybridisation of sectoral logics? Drawing from various strands of research, as well as an assessment of multiple studies on the impact of integrated territorial approaches on local social development, this article develops an analytical framework to address this question. We argue that integration takes place according to four regimes, whose dynamics range from the simple juxtaposition of sectoral organisations to a hybridisation of their organisational logics. The regimes we identify are operational networking, interstitial effervescence, collaborative accommodation and institutional convergence. Each emerges from an interaction between the specific dynamics of each experience in a given milieu and supra-local socio-institutional processes, which generate new ways of conceiving and organising the coordination of public and collective action at the local level. Points for practitioners This article puts into perspective the virtues of the integrated approach as an antidote to public administration silos. An integrated approach to local action only produces the expected effects if each public agency agrees to transform its organisational culture and to direct its action according to the evolution of local ecosystem processes of change in the milieu as a whole.

Abstract:

The present study has been one of the first attempts to thoroughly examine the effects of different kefir sources on fermentation characteristics, aerobic stability, and microbial communities of alfalfa silages. The effects of commercial kefir (CK) and homemade kefir culture (HK) applied with untreated a common control (CON) and three different application doses (5.0, 5.7, and 6.0 log cfu g−1) on wilted alfalfa and stored at an ambient temperature of 25–30 °C are studied. After 45 days of ensiling, fermentation characteristics and aerobic stability of silages were measured, and bacterial diversity was investigated by 16S ribosomal RNA gene sequencing using the GenomeLab™ GeXP platform. Both CK and HK accelerate more lactic acid production and reduced ammonia nitrogen concentration. Factor analysis of kefir sources suggests that the addition of kefir improves the aerobic stability of silages, even the initial water-soluble carbohydrate (WSC) content is inadequate via its antimicrobial effect on yeast and mold formation. Enterococcus faecium, Pediococcus pentosaceous and Lactobacillus brevis were dominant bacterial species among the treated groups at silo opening, while Lactobacillus plantarum and Lactobacillus brevis became dominant bacterial species after 7 days of aerobic exposure. In conclusion, the application of kefir on alfalfa silages improves fermentation quality and aerobic stability even with low WSC content.

Purpose – The purpose of this paper is to inquire how large multinational firms can develop and implement knowledge-sharing measures that move their corporate strategy towards the open innovation paradigm, since open innovation becomes increasingly important as source for competitive advantage. Design/methodology/approach – We review the literature on open innovation and combine it with a single case study of one multinational firm that is gradually implementing its open innovation strategy. We pay special attention to the development and usage of a collaborative IS infrastructure that is deployed to create a culture of openness and to support knowledge networking amongst the workforce. Findings – The in-depth case study demonstrates that managers have to balance a complex interplay of human and IT components to make open innovation happen. Measures taken to foster openness and knowledge exchange inside the firm include developing managerial innovation capabilities, creating communities and networks around strategic topics and leveraging the adoption of the collaborative IS infrastructure through piloting use of cases in innovation projects. Research limitations/implications – The findings of this case study remain limited to the characteristics of large firms in multinational markets. Practical implications – This article offers valuable insights for corporate strategists, IT specialists and change managers who want to open up corporate innovation. We present a range of institutional measures that help to overcome silo mentalities and knowledge-sharing barriers and establish an open innovation culture within large firms operating in multinational markets. Originality/value – Complementing previous research, this article highlights how large firms can use a combination of strategic, cultural and technological measures to bring open innovation from strategic vision to organisation-wide reality. We identify in addition factors which either inhibit or foster the implementation of knowledge sharing and open innovation practices inside large firms.

ObjectiveThere is an ongoing challenge of effective integration between primary and social care in the United Kingdom; current systems have led to fragmentation of services preventing holistic patient-centred care for vulnerable populations. To improve clinical outcomes and achieve financial efficiencies, the barriers to integration need to be identified and addressed. This study aims to explore the unique perspectives of frontline staff (general practitioners and practice managers) towards these barriers to integration.DesignQualitative study using semistructured interviews and thematic analysis to obtain results.SettingGeneral practices within London.Participants18 general practitioners (GPs) and 7 practice managers (PMs) based in London with experience of working with social care.ResultsThe study identified three overarching themes where frontline staff believed problems exist: accessing social services, interprofessional relationships and infrastructure. Issues with contacting staff from other sectors creates delays in referrals for patient care and perpetuates existing logistical challenges. Likewise, professionals noted a hostile working culture between sectors that has resulted in silo working mentalities. In addition to staff being overworked as well as often inefficient multidisciplinary team meetings, poor relationships across sectors cause a diffusion of responsibility, impacting the speed with which patient requests are responded to. Furthermore, participants identified that a lack of interoperability between information systems, lack of pooled budgets and misaligned incentives between managerial staff compound the infrastructural divide between both sectors.ConclusionIn this study, primary care staff identify intangible barriers to integration such as poor interprofessional relationships, in addition to more well-described structural issues such as insufficient funding and difficulty accessing social care. Participants believe that educating the next generation of medical professionals may lead to the development of collaborative, instead of siloed, working cultures and that change is needed at both an interpersonal and institutional level to successfully integrate care.

Abstract New Silk Roads and their economic, political and cultural aspects are new components of our modernity. As such, they raise questions and call for new studies. They are best addressed within interdisciplinary approaches exploring a wide range of subjects across a variety of geographical and historical settings. The cultural facets of the new developments (meanings, values, norms and their multiculturalism) cannot be examined out of their economic and political contexts. To make sense of the New Silk Road(s) phenomenon, connections among different geographical locations must be studied alongside the links between the past and the present. For this reason, I consider my manuscript to be suitable for publication in Open Cultural Studies. New Silk Roads can also be seen as a species of globalisation, and I hope that my contribution will propel academic discussions in the field of global studies, seeking to provide answers to such queries as: Are we witnessing the rise of a new globalisation and a new global order? How can they be related to the present and past ones? Do we need new global theories to grasp them or are the existing frameworks still adequate?

Purpose Despite the growing scholarly interest in service innovation and its associated benefits for organizations, research into the barriers to developing new services remains scant. In addition, most of these studies have been mainly conducted at the firm level, failing to incorporate macro forces in the industry. To fill this gap, this study aims to investigate major industry trends as well as organizational attributes that affect organizations’ capability in designing innovative services. Design/methodology/approach A qualitative approach was used. In all, 21 semi-structured interviews with senior executives in different banks were conducted to collect data. Data were analyzed through thematic analysis. Findings The results of this study suggest that privatization, technological shortcomings, legislative inefficiencies and deposit orientation instead of market orientation were major industry trends that affect service innovation. Furthermore, ambiguity in knowledge management regimes, silo mentality and the absence of a collaborative organizational culture, growing need to focus on human capital and risk aversion were the main organizational attributes that should be addressed for effective service innovation. Practical implications Service innovation is a strategic asset for organizations. To effectively leverage the benefits, managers should have an end-to-end approach toward the subject. The findings of this study would inform managers of different obstacles in the development of new services from an organizational and industry-wide perspective. Based on the outputs of the interviews and a thorough review of the literature, managerial implications are presented. Originality/value This study is one of the few studies that explores service innovation challenges at both macro and micro levels of analysis, providing a more holistic view of the phenomenon in the context of service organizations.

Abstract:

Improving the quality of raw milk for the production of cheese in the autumn-winter period is one of the important tasks of biotechnology. It can be solved by improving the quality of feed, as they directly affect the functional and technological properties of raw milk. Amaranth silo obtained from the green mass of amaranth by canning using the developed starter culture was used in the diet of lactating cows. Scientific and economic experience was carried out on the basis of peasant farms II Korovnikov, Khokholsky district of Voronezh region. Milk was obtained from two groups of dairy cows of the Holstein-Friesian breed with an average productivity of 4200 kg of milk, selected on the basis of analogues. The diets of the experimental and control groups in accordance with the accepted experimental scheme included canned food from the green mass of amaranth and corn. Cows were fed for 105 days and evaluated the functional and technological properties of raw milk. The results of the study of the functional and technological properties of raw milk obtained from cows, in the diet of which in the autumn-winter period included high-protein canned food from amaranth, are presented. It was found that the replacement of traditional corn silage with amaranth contributed to an increase in the most important components of raw milk for cheese production: mass fraction of protein – by 0.2% (including mass fraction of casein fraction by 0.13%) and mass fraction of fat – by 0.38%. Veterinary health indicators of raw milk have also improved. In the experimental group, there was a decrease in the number of mesophilic aerobic and facultative anaerobic microorganisms (KMAFAnM) in relation to the control by 5.6%, which correlated with a decrease in the acidity of milk.

Under appropriate culture conditions, mesenchymal stem cells (MSC), also called more properly multipotent mesenchymal stromal cells (MMSC), can be induced toward differentiation into different cell lineages. In order to guide stem cell fate within an environment resembling the stem cell niche, different biomaterials are being developed. In the present study, we used silk fibroin (SF) as a biomaterial supporting the growth of MMSC and studied its effect on chondrogenesis of canine adipose–derived MMSC (cADMMSC). Adipose tissue was collected from nine privately owned dogs. MMSC were cultured on SF films and SF scaffolds in a standard cell culture medium. Cell morphology was evaluated by scanning electron microscopy (SEM). Chondrogenic differentiation was evaluated by alcian blue staining and mRNA expression of collagen type 1, collagen type 2, Sox9, and Aggrecan genes. cADMMSC cultured on SF films and SF scaffolds stained positive using alcian blue. SEM images revealed nodule-like structures with matrix vesicles and fibers resembling chondrogenic nodules. Gene expression of chondrogenic markers Sox9 and Aggrecan were statistically significantly upregulated in cADMMSC cultured on SF films in comparison to negative control cADMMSC. This result suggests that chondrogenesis of cADMMSC could occur when cells were grown on SF films in a standard cell culture medium without specific culture conditions, which were previously considered necessary for induction of chondrogenic differentiation.

The objective of this study was to evaluate cell adhesion and proliferation on the hydroxyapatite (HAp)-coated silk fibroin (SF) fabric. Nano-scaled sintered HAp particles were covalently coated on SF chemically modified by graft polymerization. After the fabrication of the HAp/SF composite, mesenchymal cells (MCs) derived from EGFP-expressing transgenic rat bone marrow were seeded on the composite and cultured for 10 days. Fluorescence and scanning electron microscopy (SEM) revealed that the cells adhered and actively proliferated on the composites comparable to those on tissue culture polystyrene (TCPS) dishes. The results suggest that the composites are suitable for mesenchymal cell culture scaffolds and useful materials for regenerative medicine.

There is a need for character education so that the younger generation can be relied upon in the future according to the nation's culture, such as being responsible for mutual cooperation, mutual assistance and good manners. Reog Kendhang's art proves that local culture can increase the sense of community nationalism and be able to become social controls so as not to fall into bad lifestyle. Purpose The purpose of this paper is to describe the development of a digital tool in an English county striving towards a vision of integrated information that is used to underpin an increasingly integrated future of health and social care delivery. Design/methodology/approach It discusses the policy context nationally, the origins and implementation of the initiative, the authors’ experiences and viewpoint highlighting key challenges and learning, as well as examples of new work undertaken. Findings In all, 12 health and care organisations have participated in this project. The ability for local commissioners and providers of services to now understand “flow” both between and within services at a granular level is unique. Costs are modest, and the opportunities for refining and better targeting as well as validating services are significant, thus demonstrating a return on investment. Key learning includes how organisational development was equally as important as the implementation of innovative new software, that change management from grass roots to strategic leaders is vital, and that the whole system is greater than the sum of its otherwise in-silo parts. Practical implications Data linkage initiatives, whether local, regional or national in scale, need to be programme managed. A robust governance and accountability framework must be in place to realise the benefits of such as a solution, and IT infrastructure is paramount. Social implications Organisational development, collaborative as well as distributed leadership, and managing a change in culture towards health and care information is critical in order to create a supportive environment that fosters learning across organisational boundaries. Originality/value This paper draws on the recent experience of achieving large-scale data integration across the boundaries of health and social care, to help plan and commission services more effectively. This rich, multi-agency intelligence has already begun to change the way in which the system considers service planning, and learning from this county’s approach may assist others considering similar initiatives.

AbstractThe advancement of human retinal pigment epithelial cell (hRPE) replacement therapy is partly dependent on optimization of cell culture, cell preservation, and storage medium. This study was undertaken to search for a suitable storage temperature and storage medium for hRPE. hRPE monolayer sheets were cultured under standard conditions at 37°C and then randomized for storage at six temperatures (4, 16, 20, 24, 28, and 37°C) for 7 days. After revealing a suitable storage temperature, hRPE sheets were subsequently stored with and without the silk protein sericin added to the storage medium. Live/dead assay, light microscopy, pH, and phenotypic expression of various proteins were used to assess cell cultures stored at different temperatures. After 7 days of storage, hRPE morphology was best preserved at 4°C. Addition of sericin to the storage medium maintained the characteristic morphology of the preserved cells, and improved pigmentation and levels of pigmentation-related proteins in the cultured hRPE sheets following a 7-day storage period at 4°C.

Esta tarea ha sido abordada tanto desde la academia como desde la antropología aplicada, con proyectos para el rescate del patrimonio cultural. Es más, en parte por nuestra labor, los propios pueblos indios tienden a identificar patrimonio cultural, cultura e identidad. En el artículo se argumenta en torno a la necesidad de superar esta visión folclórica de la cultura y la identidad, para transitar a la revaloración de las lógicas implícitas, como parece hacerlo la propuesta del buen vivir. FOLKLORIC IDENTITY AND RECLAIMING GOOD LIVING Cultural relativism has prescribed respect for other cultures practically since its beginning. In spite of this, at least since the last four decades, following the Barbados Meeting, a large number of anthropologists have taken on the revaluing of ethnic cultures as part of our work. Both academia and applied anthropology have approached this task with projects aiming to retrieve cultural heritage. Furthermore, it is in part due to our work as anthropologists that indigenous peoples have tended to identify cultural heritage, culture and identity. This article addresses the need to overcome a folkloric vision of culture and identity and thus move toward revaluing the implicit logic, as supported by the buen vivir ‘good living’ proposal.

Abstract:

Duodenal aspirates are not commonly collected, but they can be easily used in detection of small intestinal bacterial overgrowth (SIBO). Proton pump inhibitor (PPI) use has been proposed to contribute to the development of SIBO. We aimed to determine the yield of SIBO-positive cultures detected in duodenal aspirates, the relationship between SIBO and PPI use, and the clinical outcomes of patients identified by this method. In a retrospective study, we analyzed electronic medical records from 1263 consecutive patients undergoing upper endoscopy at a tertiary medical center. Aspirates were collected thought out the third and fourth portions of the duodenum, and cultures were considered to be positive for SIBO if they produced more than 100,000 cfu/mL. Culture analysis of duodenal aspirates identified SIBO in one-third of patients. A significantly higher percentage of patients with SIBO use PPIs than patients without SIBO, indicating a possible association. Similar proportions of patients with SIBO improved whether or not they received antibiotic treatment, calling into question the use of this expensive therapy for this disorder.

Silk is a symbol of Chinese traditional garment culture, it represents the characteristics and spirits of Chinese civilization; Cheongsam is an integrated epitomization of modern Chinese civilization and western culture, and is an acknowledged quintessence of traditional Chinese garment. No matter cheongsam chooses the silk or silk chooses the cheongsam, it is the excellent match of unique material and classical style that accomplishes the elegant appearance and cultural connotations. However, in reality, cheongsam culture has been deducted but failed to come to any agreements, and presents awkward cheongsam phenomenon. This paper ascribed and explained cheongsam phenomenon, discussed the real situation and culture passing-down in terms of modern garment culture.
