This dissertation is about fairness and the role it plays in political and personal morality. Specifically, I investigate when it is appropriate to rely on considerations of fairness to draw substantive conclusions about what we should do. In Chapter 1 ("Numbers, Fairness and Beneficence") I discuss the "numbers problem," the problem of explaining why you should save more people rather than fewer when forced to choose. Existing non-consequentialist approaches to the problem appeal to fairness to explain why. I argue that this is a mistake and that we can give a more satisfying answer by appealing to requirements of beneficence or generosity. In Chapter 2 ("Fairness, Distributive Justice and Global Justice") I discuss justice in the distribution of resources, both within states and across different states. On one influential view, it is always unjust for one person to have less than another through no fault of her own. State borders, on this account, have no importance in determining which distributions are just. I show that an alternative approach is needed. I argue that distributions of wealth are only unjust in so far as they issue from unfair treatment. It follows that not all inequalities in the distribution of goods are unjust. I use these results to explain how state borders do play a role in determining which inequalities are unjust, since some of these inequalities issue from unfair treatment of citizens by the state. In Chapter 3 ("Contractualism, Politics and Morality") I discuss Rawls' contractualist theory of social justice and Scanlon's extension of it to provide a theory of "rightness", or morality more generally.
(cont.) I argue that while there is some justification for adopting a contractualist theory of social justice, this justification does not support a contractualist theory of rightness. This is because social justice is centrally a matter of cooperative fairness whereas rightness is not.

In this thesis, I investigate the nature of metaphysics and the role it plays in our broader theoretical pursuits. In doing so, I defend it against various criticisms and offer a novel conception of why metaphysical disputes are important. In Chapter One I address the general question of when disputes are defective and when they are worth taking seriously. I first criticize one popular way of answering this question that appeals to the difference between verbal and factual disputes. Verbal disputes involve divergence in what the participants mean by their terms and some think that metaphysical disputes are defective in this way. I argue that this approach fails because the verbal/factual distinction is incapable of doing the work this view requires of it. I then offer an alternative view where the status of a dispute depends on its role in our theorizing. Worthwhile (or, as I call them, significant) disputes are those with appropriate connections to the rest of our theorizing while defective (or insignificant) disputes are insular, with no implications for anything beyond themselves. In Chapter Two I apply the framework developed in the previous chapter to a pair of ontological disagreements: those over the existence of concrete possible worlds and coincident material objects, like a statue and its clay. The question is whether ontological disputes like these have the requisite theoretical connections to render them significant. I argue that they do. I then address some general reasons for doubting their significance, arguing that they are not compelling. In Chapter Three I contrast my approach with some other recent defenses of metaphysics, with particular focus on the views developed by Theodore Sider. Metaphysics is, on these views, an inquiry into the world's fundamental structure. I argue that this approach is unsatisfactory because it cannot guarantee that metaphysical disputes are significant in the way I describe. It thus threatens to render metaphysics irrelevant to our other theoretical activities, undermining many of its legitimate successes, like the role theorizing about metaphysical modality played in the development of modal logics applicable in many different fields.

Full textThis doctoral thesis emphasizes the significance of informational conditions in studying economic decisions. The first chapter concerns access to accurate probabilistic information in the domain of medical interventions. We estimate that, in many developed countries, there appears to be demand for governments to grant citizens free access to impartial reviews of medical evidence, as provided in Cochrane Reviews. For these countries, we estimate that this demand can be met at low costs. The second chapter concerns the communication of such information and examines the facilitating effect of natural frequencies on the derivation of posterior probabilities, as delineated by Gigerenzer and Hoffrage (1995). In a meta-analysis, we clarify concepts and disentangle the effects of 15 study characteristics. We find that in the simplest study design, 4 percent correct solutions when presented with conditional probabilities and 24 percent when presented with natural frequencies. The final two chapters examine the satisficing class of strategies for uncertain decision environments in which agents lack a full probabilistic description of the decision problem. According to Simon (1955), satisficing strategies use aspiration levels to terminate search for suitable alternatives. The third chapter describes how in economics, satisficing is modeled as a preference structure or as a decision rule that yields choices inferior to utility maximization, whereas in cognitive science, satisficing strategies use aspiration levels to solve inference problems. We explain the divergence, noting that they refer to risky and uncertain environments, respectively. The final chapter examines satisficing in an applied setting, studying taxi drivers' shift termination behavior. We find that drivers' hourly earnings are very uncertain and drivers’ behavior is best predicted by simple satisficing strategies that terminate shifts when reaching an aspired shift income or shift duration.

Full textAs fosfatidiletanolaminas constituem a segunda classe de fosfolípidos mais abundantes nos organismos. Elas estão presentes nas membranas biológicas e nas lipoproteínas. As alterações estruturais dos fosfolípidos, ocorrem devido ao stress oxidativo e podem manisfestar-se em alterações das suas propriedades e funções. Já são conhecidas algumas condições fisiopatológicas, nas quais os fosfolípidos oxidados estão envolvidos, por exemplo sinalização celular, resposta imunitária, apoptose e doenças relacionadas com o envelhecimento. Por esse motivo, o interesse no estudo dos fosfolípidos oxidados e suas funções tem crescido nos últimos anos. Contudo, a maioria dos estudos realizados, focam a oxidação das fosfatidilcolinas, tendo sido dedicada pouca atenção a outras classes de fosfolipídos, como as fosfatidiletanolaminas. As fosfatidiletanolaminas, podem ainda sofrer outras modificações, devido ao grupo amina livre presente na cabeça polar, como por exemplo a glicação. As fosfatidiletanolaminas glicadas já foram detectadas em condições de hiperglicemia, em pacientes diabéticos, e tem correlação com a hemoglobina glicada. Sabe-se que a glicação de biomoléculas, pode aumentar as modificações oxidativas, que por sua vez, podem ser responsáveis pelo estado inflamatório, existente na diabetes mellitus. Tanto o stress oxidativo, como a inflamação estão relacionados com a diabetes e as suas complicações. A espectrometria de massa tem sido utilizada como uma importante tecnologia na detecção e caracterização de modificações oxidativas de fosfolípidos. Assim, neste trabalho pretendeu-se estudar as modificações oxidativas induzidas em fosfatidiletanolaminas glicadas, e os seus efeitos biológicos nos monócitos e células dendríticas do sangue periférico. Pretendeu-se ainda, estudar as alterações que ocorreram nas espécies de fosfatidiletanolaminas do fígado de ratos diabéticos. Os resultados obtidos permitiram identificar vários produtos de oxidação de fosfatidiletanolaminas glicadas, nomeadamente novos produtos formados pela oxidação da cabeça polar glicada. A oxidação na cabeça polar glicada foi, ainda, confirmada pela realização de experiências com spin traps combinadas com espetrometria de massa. Posteriormente, as fosfatidiletanolaminas oxidadas, glicadas e glicoxidadas demonstraram ter efeitos pró-inflamatórios, confirmados pelo aumento da estimulação monócitos e de células dendríticas, expresso no aumento do número de células produtoras de citocinas em comparação com o estado basal. As diferentes modificações de fosfatidiletanolaminas induziram estímulos distintos nos dois tipos de células. Sendo as fosfatidiletanolaminas glicadas e as glicoxidadas, os compostos que induziram um maior estímulo. Estes resultados sugeriram que as fosfatidiletanolaminas glicadas e as glicoxidadas podem estar associadas com o estado inflamatório que decorre da hiperglicemia crónica. Ainda, a avaliação do perfil lipídico de extratos de fígado de ratos diabéticos demonstrou que a hiperglicemia induz inúmeras alterações das espécies de fosfatidiletanolaminas e das espécies de outras classes de fosfolípidos, em simultâneo com sinais de lesão hepática. Em conclusão, este trabalho demonstra a relação existente entre, a hiperglicémia, o stress oxidativo, a glicação e oxidação de fosfatidiletanolaminas e ainda a inflamação e compliações diabéticas. Portanto a contribuição da lipidómica é importante para compreender os efeitos prejudiciais da hiperglicemia não controlada, e por isso, merece ser mais explorado.
Phosphatidylethanolamines (PE) are the second most abundant phospholipids found in biological membranes and in lipoproteins. In cases of oxidative stress, phospholipids may undergo structural changes that lead to alterations of phospholipids properties and functions. Oxidized phospholipids are known to be involved in various physiopathological conditions, such as signaling of inflammation, immune response, apoptotic events and in age related diseases. Because of such implications, the interest in the study of this subject is increasing. However, most of the studies performed only focused the role phosphatidylcholines and little attention has been paid to other phospholipid families, like phosphatidylethanolamines. Also, phosphatidylethanolamines can undergo other modifications, due to the free amino group present at polar head, such as glycation. Glycation of phosphatidylethanolamines was already reported to occur under hyperglycemia, in diabetic patients and to have correlation with glycated hemoglobin. The glycation of biomolecules increase oxidative modifications, which can be responsible for the low inflammatory state, typical of diabetes mellitus. Both oxidative stress and inflammation are related to diabetes and can be associated with diabetic complications. Mass spectrometry has been used as an important tool in the detection and characterization of these phospholipids’ oxidative modifications. So work aimed to study the oxidative modifications induced in glycated phosphatidylethanolamines, and their biological effects in blood monocytes and dendritic cells. It also aimed to evaluate the alterations of liver PE species under hyperglycemia in diabetic rats. The results allowed identifying various oxidation products of glycated phosphatidylethanolamines, including some new products with oxidation in polar head and in glucose. Oxidation in glycated polar head was confirmed by spin trapping experiments combined with mass spectrometry. Furthermore the oxidized, glycated and glycoxidized phosphatidylethanolamines showed pro-inflammatory effects, confirmed by the increase monocytes and dendritic cell stimulation with increased frequency of cytokine-producing cells, relative to the basal level. The different PE modifications induced different stimulus in the different cells, being the glycated and glycoxidized compounds the ones that induced higher stimulus. The results suggested that glycated and glycoxidized phosphatidylethanolamines could be associated with inflammatory status present in chronic hyperglycemia. In addition, the evaluation of lipid profile of liver extracts from diabetic rats showed that under hyperglycemia occurred changes in PE and other phospholipid species accompanied by signs of hepatic damage. In conclusion, this work reenforces the network between hyperglycemia, oxidative stress, PE glycation and oxidation, inflammation and diabetic complications. Thus the contribution of lipidomic to understand deleterious effects of uncontrolled hyperglycemia is a promising field of research that deserves to be further explored.

This thesis explores the ways in which LGBTQ+ individuals in Philadelphia ascribe significance to various places based on oral history interviews and additional primary source material collected initially for the National Park Service Northeast Regional Office’s LGBTQ+ Heritage Initiative. By examining stories from LGBTQ+ individuals of places that matter most to them in Philadelphia, this thesis argues that historic preservationists can expand their definition of significance to include personal testimony and broaden their practices to better engage the communities whose histories they seek to preserve.

Full textFör att undersöka vad medveten närvaro kan betyda i dialektisk beteendeterapi (DBT) meningskoncentrerades nio intervjuer. Medveten närvaro hade en positiv betydelse för samtliga deltagare. Det centrala för deltagarna var färdigheterna observera och en sak i taget. Färdigheterna hjälpte patienterna att stanna upp i ett negativt händelseförlopp. Behandlarna menade att medveten närvaro hjälpte vid stressrelaterade situationer. Samtliga deltagare underströk betydelsen av färdighetsträningen i grupp och praktiska övningar. Det fanns ett tydligt behov av att förenkla teorin kring medveten närvaro i DBT samt att medveten närvaro behövde få ett större utrymme. Om medveten närvaro kan hjälpa patienter i DBT att stanna upp i ett destruktivt händelseförlopp så är det en verkan av stor betydelse för patienterna såväl som för anhöriga och sjukvården.

Full textDuring photosynthesis sunlight is absorbed by photosynthetic pigments and converted into organic compounds, such as carbohydrates. Photosynthesis needs to be highly regulated, since both too much and too little light are harmful to plant. If too little light is absorbed, a plant cannot store enough energy, which will have effects on growth and fitness of the plant. With too much light absorbed, a dangerous side reaction of photosynthesis, the production of reactive oxygen species can happen. These reactive oxygen species can damage the proteins in the chloroplast and the lipids of the chloroplast.
To avoid the production of reactive oxygen species, plants have evolved many mechanisms, which act on different time-scales and different levels of organization. As a first measure, when the absorbed light is exceeding the capacity for its utilization, is to switch the light-harvesting antenna from efficient light harvesting to energy dissipation. This process is called feedback de-excitation (FDE). The protein PsbS is essential for this process as well as a functioning xanthophylls cycle with the enzyme violaxanthin de-epoxidase (VDE).
I have investigated the effects of plants with changes in their ability to dissipate excess excitation energy in the model plants species Arabidopsis thaliana. Three genotypes with either increased or decreased capacity for FDE were used during my experiments. The first genotype over-expresses the PsbS gene, having approximately two-fold increased amounts of PsbS and FDE. The second is a PsbS deletion mutant with no PsbS protein and no FDE. The third genotype cannot perform the conversion of violaxanthin to zeaxanthin, because the enzyme VDE is missing. This mutant has some FDE left.
Arabidopsis thaliana is an annual plant, which flowers only once in its lifetime. Therefore, when counting the seeds produced an estimation of fitness can be made from the amount of seeds produced. This was done during my experiments and shown that FDE is a trait and that plants with increased FDE have a higher fitness and vice versa.
This was also the case for a collection of plants lacking a single protein from the light harvesting antenna. All of these genotypes had a fitness reduction, proving that their function is not redundant.
In an attempt to explain why the fitness is reduced in plants with altered FDE, photosynthetic measurements, as well as a determination of the transcriptome and the metabolome was performed. Plants lacking FDE had higher levels of photoinhibition, leading both to lower rates of photosynthesis and to higher repair cost. This could in part explain the reduction in fitness. These plants also had major changes in their transcriptome and their metabolome. Primary metabolism was most effected, for example carbohydrate and amino acid metabolism. But there were also changes in secondary metabolism such as an up regulation of the biosynthesis of anthocyanins.

I primarily investigate the role that time plays in the constitution of objective knowledge. I discuss that since time is the necessary condition for objects to be given to our sensibility, without it any representation would be without a temporal order and perhaps would not make any sense at all. Kant claims that it is imagination that enables the connection between sensibility and understanding possible. After investigating the relation between imagination and time, I argue for the following thesis: what lies at the very heart of the possibility of the connection between sensibility and understanding must be time itself. Finally, I focus on the kinship between time and the self. I argue that when it comes to knowledge of the phenomenal world, time together with transcendental self stands as the condition of all human knowledge.
