Abstract. Rock mass classification is one of the empirical methods for slope stability analysis approach. Rock mass clasification systems that have been developed such as RSR “Rock Structur Rating” (Wickham et al., 1972), RMR “Rock Mass Rating” (Bienieawski, 1973, 1975, 1979, 1989), NGI Q-system (Barton et al., 1974) and the results of the development of the new "Rock Mass Clasification on Rock Slope Stability Assessment" (Ya-Ching Liu, Chao-Shi Chen, 2007) is a combination of the Analytical Hierarchy Process (AHP, Saaty, 1980) and Fuzzy systems. Delphi Method (FDM, Kaufmann and Gupta, 1988) this system consists of 3 grouping factors to assume slope stability consisting of geological, geometrical and environmental factors and the method developed by Ya-Ching Liu, Chao-Shi Chen, 2007 will be used in research this. The observation location used to use this method is spread over 7 mining blocks, namely block 67 - 73 which produces 2 classification characteristics of rock mass which are stable and tend to be unstable.Abstrak. Abstract:
Abstract Mechanism-guided drug repurposing can accelerate the development of therapeutic strategies for oral squamous cell carcinoma (OSCC), which has an unmet clinical need. We integrated cell line sensitivity data for 339 anti-cancer drugs with genome-wide CRISPR-Cas9 screen gene essentiality data for 21 unique OSCC cell lines, derived primarily from Asian patients. We identified a significant correlation between AZD5582 sensitivity (an antagonist of the inhibitor of apoptosis (IAP) family proteins), and dependency on several members of the NF kappa-B pathway (RNF31, MAP3K7 and IKBKG). Dependent OSCC lines are more sensitive to AZD5582, with nanomolar range half-maximal inhibitory drug concentrations (IC50). We confirmed the on-target specificity and potency of AZD5582 in vitro and in vivo. Furthermore, AZD5582 sensitivity was associated with high levels of tumor necrosis factor (TNF) and either functional apoptotic or necroptotic pathways. In summary, our approach of integrating pharmacological and CRISPR screen data in a unique cohort of OSCC cell lines data has enabled the identification of specific dependencies and drug repurposing candidates. Abstract Treating oral squamous cell carcinoma (OSCC) remains challenging due to limited knowledge of the critical genes that could affect cell survival in OSCC. We have previously utilized genome-wide CRISPR-Cas9 knockout functional screens to explore the genetic vulnerabilities of OSCC and identified Adenosine deaminase acting on RNA (ADAR1) as one of the top fitness genes. ADAR1 is an interferon-stimulated gene (ISG) that catalyses adenosine-to-inosine (A-to-I) editing of double-stranded RNA (dsRNA), which suppresses dsRNA sensing mechanism from triggering cell death. ADAR1 gene encodes for two isoforms, the constitutively expressed P110 and the interferon-inducible P150. Given that deleting ADAR1 severely impacts OSCC viability, we aimed to elucidate key molecular mechanisms underlying this gene dependency which may provide opportunities for developing better treatment strategies. Here, we validated ADAR1 dependency via competitive co-culture assay using single-guide RNA (sgRNA) knockout in selected OSCC cell lines. We also confirmed that sgRNA-mediated ADAR1 knockout resulted in prominent cell lethality through increased apoptosis and inhibition of colony formation. Cell lethality is evident in ADAR1 KO cells in conjunction with the phosphorylation of a dsRNA sensor, protein kinase R (PKR) which regulates cell death mechanism. In ADAR1-less dependent cells, IFN-β treatment increased the expression of ISGs including ADAR1, and sensitized the cells to ADAR1 KO-induced cell death. Overexpression of ADAR1-P150 but not ADAR1-P110 rescued cell lethality in ADAR1-depleted cells, suggesting that ADAR1-P150 is required for OSCC survival. We also demonstrate that the deaminase function of ADAR1 is important in conferring survival to a subset of OSCC lines. In addition to PKR, the melanoma differentiation-associated protein 5 (MDA5) dsRNA sensor is also important in mediating ADAR1 KO-induced cell lethality OSCC and co-deleting both PKR and MDA5 enabled the partial rescue of cell death compared to when each gene was knocked out individually. Collectively, our results uncover the essentiality of IFN-inducible ADAR1-P150 for OSCC survival, and reveal that the activation of PKR and MDA5 dsRNA sensing pathways underlie the cell lethality caused by ADAR1 deletion. AbstractDormancy in rice (Oryza sativa L.) seed is imposed by certain physical and chemical factors associated with its covering structures, i.e.hull and pericarp. The nature of these germination blocks, their mode ofaction, and processes regulating the release of dormancy are not fully understood. Of nine rice cultivars studied, Ching-shi 15, Stejaree 45, PTB10, and Mahsuri are weakly dormant, and Bansphul, Benaful, Kataktara, Dular, and N22 are dormant. Release of seed dormancy in rice by various treatments, oxidative processes and enzymic changes associated with dormancy, and parallelism between natural and artificially imposed dormancy patterns were examined. The influence of the hull in imposing dormancy was stronger and more prolonged than that of the pericarp. Application of GA3 was effective in inducing germination only in weakly dormant cultivars. Dormancy was completely released in all cultivars by subjecting the seeds to moist heat treatment, by removing the hull and pericarp, and by applying GA3 after dehulling. Dormant cultivars had higher O2 uptake rate and peroxidase activity and lower amylase and dehydrogenase activities than the weakly dormant ones. Hull removal substantially decreased peroxidase activity but enhanced amylase and dehydrogenase activities. Nonanoic acid (C90), a short-chain saturated fatty acid (SCSFA), when exogenously applied to non-dormant seeds imposed dormancy. Dry heat treatment or presoaking in 0.01 m KNO3 or 0.1 m H2O2 was very effective in releasing SCSFA-imposed dormancy. Amylase activity was greatly reduced by treatments with nonanoic acid (C90) or ABA. Considering earlier reports and results of the present study, it is proposed that seed dormancy in rice is regulated both by the presence of SCSFAs and ABA in the hull and the pericarp. The relative significance of these substances in cultivars of tropical and temperate origins and its implications in terms of ecogeographic adaptability are discussed.

Abstract Background: T-1301 is a novel small molecule multi-target kinase inhibitor, it also exhibited potent inhibitory activity against both wild type and mutant forms of various kinases, i.e., ABL1, FLT3, c-KIT, PDFGRA, RET, and TRKA. In in vitro studies, T-1301 effectively inhibited growth of acute myeloid leukemia (AML) as well as gastrointestinal stromal tumor (GIST) cells carrying mutations that are resistant to KIT-targeting tyrosine kinase inhibitors (TKIs) in clinical use. In animal studies, tumor regressions were observed in AML and GIST xenograft models, while growth inhibition were observed in various other types of solid tumor models. Method: This is an open label, multi-centered Phase 1 dose escalating study (NCT05156203). The primary objectives are to establish the maximal tolerated dose (MTD) and recommended Phase 2 dose (RP2D), and to assess the safety and tolerability of T-1301. The secondary objectives are to evaluate the pharmacokinetics (PK) and therapeutic response after receiving treatment. Eligible Patient should have histologically and cytologically confirmed advanced solid tumors (including lymphoma) with measurable or evaluable target lesion(s) per RICEST v1.1 and refractory to or without standard treatments, ECOG performance status 0-1, and adequate organ function. The dose escalation begins with accelerated titration and switch to the Bayesian Optimal Interval (BOIN) design when either one of the following events is observed in Cycle 1: the first instance of DLT, or any Grade 2 or higher drug-related adverse event (AE) in any patient. Then, at least 2 additional subjects are treated at the current dose level and the dose escalation will follow the Bayesian Optimal Interval (BOIN) design with cohorts of size 3-6.T-1301 is orally administered daily in a 28-day cycle (a 21-day consecutive dosing followed by a 7-day rest). The dosing schedule is once daily (QD) at the initial dose level and could be QD or twice daily starting with the second dose level according to the safety review committee (SRC) decision after reviewing the safety and PK data from each dose level. AEs are assessed according to CTCAE v5.0 and occurrence of dose limiting toxicity (DLT) is determined during the first cycle of treatment. The safety findings are reviewed by the SRC, which will determine the MTD and RP2D. Tumor response evaluation is performed after 2 cycles of treatment and assessed according to RECIST v1.1 criteria. For those who benefit from the study medication after 2 treatment cycles, intra-subject dose escalation for multiple times is allowed in extension study period. The Medieval China is a “Manuscript era”. The four divisions of Jing, Shi, Zi, Ji, and the documents of Buddhism and Taoism, all have to be considered based on this general background. For the first part of this paper, the keyword used in the comparison with “Buddhism” in the Medieval China is “Confucianism” rather than “Confucian classics”. Then, the concept and classification of Jiyi (collection of the lost parts of classics) are explained. The second part, starting from the newly published Lunyu Yi shu in Japan in 2020 and integrating with Jiang Zhou yi shulunjia yi ji and other manuscripts, is to analyze some Buddhist factors in the study of Confucianism Yi shu. The third part, from the East Asian Civilization sphere, explores the academic significance of Chinese Buddhism under the perspective of the integration of the Three teachings in the Middle Ages. Special attention will be paid to the Japanese written Guketsu getensho, and how the text form absorbed the Confucian thought will be analyzed. Through these cases, a preliminary conclusion about the relationship between the manuscript culture of Confucianism and Buddhism in the medieval China will be made.

Fringe belts were first recognized in Europe, primarily in relation to city walls. In China, fringe belts have been rarely studied, despite their very widespread occurrence. Yet China provides a highly complex world of urban morphological phenomena related to cultural settings substantially different from those in the West. In relation to both a long urban history and recent rapid processes of industrialization and urbanization, the fringe belts of Chinese cities deserve more in-depth research. To rectify this deficiency, this paper examines the developmental process and form of the fringe belts of Shangqiu (including both ancient Shangqiu and modern Shangqiu) as a central focus, using the basic methods of morphogenetic analysis. Since the Ming Dynasty the existence of fringe belts in Shangqui relates to double fixation lines (double city walls, the space between which is water for defence against invasion and flood). Since 1949, a new core developed outside ancient Shangqiu. In time, due to the alteration of the city’s organizational system and rapid expansion of modern Shangqiu, the whole of ancient Shangqiu, as well as its fringe belts, has become part of the fringe-belts system of modern Shangqiu. The development of the fringe belts of Shangqiu shows a different pattern from a concentric town such as Alnwick. This finding extends and refines the understanding of fringe belts. Scientific evaluation in China is going through a transition period. In dealing with the misuse and alienation of quantitative indicators, China had initiated a top-down reform. To help better understand the scientific evaluation policy in China and provide reference for further reform, this study investigated the evolution and characteristics of scientific evaluation in China from a bibliometric perspective. In total, 343 policy documents from 1978 to 2021 were collected. Based on the temporal distribution of policy documents and timeline of landmark policy documents, four phases were identified. Text mining technique and social network analysis were used to identify the shift of policy focus over different phases. The four phases were characterized from three aspects: (1) The time span and number of relevant policy documents; (2) The focus of policy document content; and (3) The collaborative network of ministries that release the policy documents.

Open source intelligence (OSINT) and science and technology intelligence (STI) are realized differently in the United States and China, China putting greater value on both. In the United States’ understanding, OSINT “enables” classified reporting, while in China it is the intelligence of first resort. This contrast extends to STI which has a lower priority in the U.S. system, whereas China and its top leaders personally lavish great attention on STI and rely on it for national decisions. Establishing a “National S&T Analysis Center” within the U.S. government could help to address these challenges.

This paper uses spatially granular surface urban heat island (SUHI) data to quantify the impact of COVID-19-related containment policies on economic output in the People’s Republic of China (PRC). Governments have adopted various policies and measures to control the spread of coronavirus disease (COVID-19). Results of the study suggest containment measures in the PRC decreased SUHI in locked cities marginally yet generated positive and negative spillover effects in unlocked cities, with positive effects dominating. The paper also notes that initial experiences helped inform the management of containment measures in the country.

Review question / Objective: The purpose of this systematic review is to evaluate the effect of acupuncture on SUI and the quality of life-based on the latest literature. Condition being studied: At least 25% of adult females in the world have urinary incontinence in some measure, of which more than half are stress urinary incontinence (SUI). SUI seriously affects the mental health of patients, but also leads to perineal rash, urinary tract infection, and other harms. The American Urological Association recommends pelvic floor muscle training (PFMT) as a conservative treatment for patients with mild to moderate SUI, but the cost of treatment is the main obstacle to its wide use of it. Acupuncture is one of the traditional therapies in ancient China, which is simple and cheap. Some systematic reviews and meta-analyses provide evidence for acupuncture in the treatment of SUI. Due to the quality of the study, these research results are not very reliable.

In recent years, mission-oriented approaches have received growing interest in science, technology and innovation (STI) policies against the background of two developments. First, while so-called “horizontal” or “generic” approaches to research, technology and innovation policies have largely been successful in improving the general innovation performance or the rate of innovation, there are perceived limitations in terms of insufficiently addressing the direction of technological change and innovation. Second, “grand societal challenges” emerged on policy agendas, such as climate change, security, food and energy supply or ageing populations, which call for thematic orientation and the targeting of research and innovation efforts. In addition, the apparent success of some mission-oriented initiatives in countries like China, South Korea, and the United States in boosting technological development for purposes of strengthening competitiveness contributed to boosting the interest in targeted and directional government interventions in STI. Against the backdrop of this renewed interest in mission-oriented STI policy, the OECD has addressed the growing importance of this topic and launched a project looking into current experiences with Mission-Oriented Innovation Policy (MOIP). The present study on MOIP in Austria was commissioned by the Austrian Federal Ministry for Climate Action, Energy, Mobility, Environment, Innovation and Technologiy (BMK) and comprises the Austrian contributions to this OECD project. The study aims at contributing Austrian experiences to the international debate and to stimulate a national debate on MOIP.

Sin embargo, las implicaciones de ese regreso han sido bastante diversas para cada región del mundo. Para América Latina ciertamente ha implicado una retórica menos agresiva y racista en comparación con la de la presidencia de Donald Trump. No obstante, cabe preguntarse si, más allá del tono, ha habido cambios de fondo en la política de Estados Unidos hacia la región. Aún más, es importante entender cuáles son los factores que han impulsado esos cambios o continuidades de la política estadounidense hacia América Latina, y si estos factores están aquí para quedarse o son coyunturales de esta administración. En ese sentido, este texto se concentra en dilucidar la política de Biden hacia América Latina y proponer una interpretación de los factores que la direccionan y la limitan. En particular, se ilustra cómo se ve moldeada por la competencia estratégica con China, por los cambios ideológicos en las presidencias latinoamericanas, y por las prioridades propias de la agenda doméstica estadounidense (como la migración y la transición energética).
