This work is devoted to understanding the formation of supported lipid bilayers (SLBs) on curved surfaces as a function of lipid properties such as headgroup charge/charge density and alkyl chain length, and nanoparticle properties such as size and surface characteristics. In particular, the formation of SLBs on curved surfaces was studied by varying the size of the underlying substrate SiO2 nanoparticles with size range from 5-100 nm. Curvature-dependent shift in the phase transition behavior of these supported lipid bilayers was observed for the first time. We found that the phase transition temperature, Tm of the SLBs first decreased with decreasing the size of the underlying support, reached a minimum, and then increased when the size of the particles became comparable with the dimensions of the lipid bilayer thickness; the Tm was above that of the multilamellar vesicles (MLVs) of the same lipids. The increase in Tm indicated a stiffening of the supported bilayer, which was confirmed by Raman spectroscopic data. Moreover, Raman data showed better lipid packing and increased lateral order and trans conformation for the SLBs with increasing the curvature of the underlying support and decrease of the gauche kinks for the terminal methyl groups at the center of the bilayer. These results were consistent with a model in which the high free volume and increased outer headgroup spacing of lipids on highly curved surfaces induced interdigitation in the supported lipids. These results also support the symmetric lipid exchange studies of the SLBs as a function of the curvature, which was found to be slower on surfaces with higher curvature. Further, the effect of surface properties on the formation of SLBs was studied by changing the silanol density on the surface of SiO2 via thermal/chemical treatment and monitoring fusion of zwitterionic lipids onto silica (SiO2) nanoparticles. Our findings showed that the formation of SLBs was faster on the surfaces with lower silanol density and concomitantly less bound water compared to surfaces with higher silanol density and more bound water. Since the two SiO2 nanoparticles were similar in other respects, in particular their size and charge (ionization), as determined by zeta potential measurements, differences in electrostatic interactions between the neutral DMPC and SiO2 could not account for the difference. Therefore the slower rate of SLB formation of DMPC onto SiO2 nanoparticles with higher silanol densities and more bound water was attributed to greater hydration repulsion of the more hydrated nanoparticles. Lastly, we have investigated the effect and modulation of the surface charge of vesicles on the formation of SLBs by using different ratios of zwitterionic and cationic DMPC/DMTAP lipids. Through these studies we discovered a procedure by which assemblies of supported lipid bilayer nanoparticles, composed of DMPC/DMTAP (50/50) lipids on SiO2, can be collected and released from bilayer sacks as a function of the phase transition of these lipids. The lipids in these sacks and SLBs could be exchanged by lipids with lower Tm via lipid transfer.

Full textIn the electricity production, one central key to reduce damages and losses due to wetness in steam turbines is the study of liquid films. These thin films are created by the deposition of droplets and are highly sheared. This film may then be atomized into coarse water. At the moment, no comprehensive and validated model exists to describe this phenomenon. A 2D model based on a integral formulation associated with closure laws is developed to represent this film. Compared to classical Shallow-Water equation, the model takes into account additional effect : mass transfer, droplet impact, shearing at the free surface, surface tension, pressure gradient and the rotation. The model properties (hyperbolicity, entropy, conservativity, linear stability, Galilean invariance and rotational invariance) has been analyzed to judge the pertinence of the model. A new 2D code is implemented in a free module of the code EDF Code Saturne and a finite volume method for unstructured mesh has been developed. The verification of the code is then carried out with analytical solutions including a Riemann problem. The model, which degenerates into classical Shallow-Water equations for the case of a falling liquid film on a inclined plane, is validated by the experiment of Liu and Gollub, 1994, PoF and compared to reference models (Ruyer-Quil and Manneville, 2000, EPJ-B et Lavalle, 2014, PhD thesis). Another study depicts a sheared film under low-pressure steam turbine conditions and is validated by the experiment of Hammitt et al., 1981, FiI. Lastly, the code film is coupled to 3D steam data around a fixed blade of a BP100 turbine, from Blondel, 2014, PhD thesis. This industrial application shows the feasibility of liquid film's simulation in real steam turbine condition

Full textDespite numerous works, the causal link between wind and waves is still a controversial subject. This is due, among others, to the multi-scale nature of a realistic ocean surface and to wave breaking, which changes its topology. In this thesis, such problems are studied from a theoretical perspective, using a phenomenological model linking the spectral and averaged properties of wall-bounded turbulence through the geometry attached eddies.The first part of the thesis revisits this phenomenological model by questioning its underlying assumptions and, in particular, reveals inconsistencies in the models used for the energy redistribution between turbulence components (the Rotta model). The phenomenological model is then used to study the coupling between long wind-waves (of order 10m) and turbulence. Results indicate that the deformation of attached eddies, induced by this interaction, could explain some of the variability in momentum fluxes for a given mean wind. Finally, the study of the coupling between turbulence and short breaking waves is approached by defining a roughness sublayer, in which the properties of the attached eddies depend solely on the speed of the dominant breaking fronts for a given wind. These two studies from the basis of a new paradigm to study the multi-scale coupling between the turbulent and wave spectra. This would allow accounting for the influence of environmental parameters on momentum and heat fluxes, and opens new paths both from a theoretical perspective and for the analysis of experimental data

Abstract The aim of this article is to study Kieran G. O’Grady’s paper "Desingularized moduli spaces of sheaves on a K3" in 1998, where the author constructs the moduli space of rank two torsion-free semistable sheaves on a non-singular K3 surface with c1 = 0 and c2 = c a even number not less then 4. This moduli space is denoted by Mc, which is a G.I.T. quotient from the Quot-scheme and is singular. By using Kirwan’s method of successive blow ups of the strictly semistable loci with reductive stabilizer, one can obtain a desingularization Mcc of Mc. What’s surprising is that when c = 4, there is a Mori extremal divisorial contraction of Mc4 so that the outcome is a hyperk¨ahler manifold Mf4. Moreover, the natural map from Mf4 to M4 is a morphism and hence a simplectic desingularization of M4. The hyperk¨ahler manifold Mf4 is not birational/deformation equivalence to another two typical constructions of HK manifolds: the Hilbert schemes of points and Kummer varieties.
