The aim of the present study was to evaluate anatomopathology, hemogram and blood serum components of commercial layers experimentally inoculated with SGcobScbiA, which is a Salmonella Gallinarum (SG) strain it shows attenuation of the virulence in previous research and it was compared with high virulence SGNalr and SGcobS strains in order to show if SGcobScbiA has potential to be use as a vaccine against SG and S. Enteritidis wild strains. 280 commercial layers were divided into 4 groups (G); G1 (SGcobS), G2 (SGNalr), G3 (SGcobScbiA) and G4 (control group). With exception of G4, all the other groups received 0,2 mL of their respective strain containing about 108 CFU/mL of the inoculum with five days of age. Birds were sacrificed 24 hours before (1DBI) and 24 hours after the inoculation (1DAI), and three (3DAI), five (5DAI), seven (7DAI) ten (10DAI), and fifteen (15DAI) days after the administration of the inoculum, slaughtering ten birds at a time in each group. Birds were submitted to euthanasia and blood samples were collected in order to make the hematological and blood serum components test. Samples of liver, spleen, thymus, bursa of Fabricius, kidneys and heart were collected for the histological test. The birds inoculated with SGcobS strain had similar behavior when compared with that ones who received SGNalr strain, however some different responses in the hematological and blood serum components were found. On the other hand, the birds inoculated with SGcobScbiA strain had similar behavior when compared with the control group, however, lower alterations in some parameters were found. Further studies must be done to verify if these alterations will not interfere in the performance of the vaccinate birds with SGcobScbiA strain.

Abstract:
This study analyses how Stargate SG-1 perpetuates dominant representations of whiteness, and how whiteness is used as a marker of racial identity in American popular culture. The popular science fiction television show Stargate SG-1 continually uses the nonwhite alien to juxtapose the seeming superiority of the white human, with white Americans acting as trusted gatekeepers for the entire planet. Whiteness becomes almost invisible and normative as the alien “other” requires assistance or containment enacted through SG-1’s adventures “off-world”. I also examine the representation of superior white aliens as an extension of these dominant white discourses. It is through the study of the constructed nature of “race” that whiteness is made visible. As represented in Stargate SG-1 whiteness discourses contribute to and reflect “common sense” constructions of race within U.S. society. This examination of Stargate SG-1 illuminates how negotiations of whiteness are constructed within United States dominant cultural discourses as a means to exclude the “other”.

The Topographic Sheet of Guapiara 1: 50000 (SG-22-X-B-II-2) is located in the southernmost part of the state of São Paulo and comprises the central portion of the Mantiqueira Province, the central-southern belt of the Ribeira Belt. The local geological framework involves rocks from the metavulcanosedimentary sequence of the Açunguí Supergroup, meso to neoproterozoic metamorphosed age and deformed in the neoproterozoic, associated with granitoid rocks generated during the collisional episodes related to the Brasilian Cycle and to the formation of the Gondwana Supercontinent. In the region, eight main lithostratigraphic units were mapped: metasediments of the Água Clara Formation and the Votuverava and Itaiacoca Groups of meso-neoproterozoic age, neoproterozoic granite bodies representing the Três Córregos Granite, granite neoproterozoic bodies of the Capão Bonito Granite, sedimentary rocks of Itararé Group, basic effusives rocks associated with the Serra Geral Formation and recent sediments of the Quaternary. The structuralmetamorphic framework is dominantly determined by the final neoproterozoic tectonic arrangement, evidenced in the epimetamorphic rocks by a structural evolution of the polyphase type. The primary structures S0, due to the intense processes of transposition of ductile foliations and milonite deformation, are sparsely preserved in some areas, with predominance only of the relic gradation bedding of S0, in less deformed areas, to a tectonic banding in the regions of Shear Zone. Four deformational phases and three main metamorphic phases are recorded: the Dn phase, low to medium angle and preferential direction NE / SW, which is subparallel to the compositional bandage S0; The Dn + 1 phase, from low to medium angle and preferential direction NW / SE; The phase Dn + 2, of high angle and preferential direction NE / SW, this being the most penetrative and the main phase and the Dn + 3 phase, of high angle and preferential direction NW / SE. The associated metamorphic events are characterized by the metamorphism M1, the metamorphic apex, of the regional progressive type in the high greenschist to low amphibolite facies; M2, contact type under low pressure conditions and low to medium temperature, with the formation of hornfels and local skarns; M3, regressive regional metamorphism in low greenschist facies.

Chiccone, Christopher Louis. "Automated SG-DBR Tunable Laser Calibration Optimized for Optical Coherence Tomography Applications." DigitalCommons@CalPoly, 2011. https://digitalcommons.calpoly.edu/theses/519.

Full text
Abstract:
SG-DBR lasers look to solve many problems associated with present OCT sources by being cost effective, smaller in size, more robust, and by operating at faster repetition rates. Swept Source Optical Coherence Tomography (SS-OCT) requires a tunable laser source that exhibits linear frequency sweeps, large frequency spans, and high repetition rates. This work accomplishes this by using four synchronized waveforms sent to the input of a Sampled Grating-Distributed Bragg Reflector (SG-DBR) laser. Three mirrors control the wavelength, while an internal semiconductor optical amplifier controls the laser output power. In dealing with this complicated tuning mechanism, a manual sweep calibration is too time-consuming. This thesis demonstrates an efficient method for automating the calibration of tunable SG-DBR lasers by implementing a gain medium voltage sensing algorithm, as opposed to the previous inefficient manual efforts. Experimental OCT tests are also performed by utilizing a Mach-Zehnder interferometer as a device under test to verify the accuracy of the laser calibration methodology. The OCT response to a single reflection event is measured over a range of repetition rates. A method to reduce these spurious display responses caused by wavelength stitching imperfections is implemented through a self-generating optical clock.

Abstract:
This thesis consists of two essays. Both essays are related to customer base concentration. In the first essay, I examine the impact of customer base concentration on selling, general, and administrative (SG&A) costs stickiness. With a sample of 65,549 firm-year observations from 9,460 unique firms on the US market from 1980 to 2012, I find that concentrated customer base accentuates SG&A costs stickiness, even after controlling for related variables such as asset intensity, employee intensity, firm size and firm age. Further findings indicate that the positive association between customer base concentration and SG&A costs stickiness is enhanced if supplier firms are in durable industry (SIC 3400-3990), or have more relationship-specific investments, but weakened if the duration of the relationship between a supplier firm and its customer base is longer. The findings are robust to alternative measures of major variables and to alternative testing models. In the second essay, I examine the impact of customer base concentration on investment efficiency. With a sample of 57,852 firm-year observations from 12,123 unique firms on the US market from 1980 to 2013, I find that concentrated customer base is positively associated with investment efficiency improvement. The findings are evidenced by the testing results that concentrated customer base strengthens the positive association between investment activities and investment opportunities. From shareholders’ perspective, concentrated customer base also strengthens the contribution of investment expenditure to shareholders’ value. In addition, I find that the positive association between customer base concentration and investment efficiency is influenced by some salient features of the supplier-customer relationship. Generally, such association is more pronounced if the supplier firm’s industry share is larger; if the supplier firm relationship-specific investment is greater; and if the supplier-customer relationship is longer. The findings are robust to a number of sensitivity tests. Keywords: customer base concentration; SG&A costs stickiness; investment efficiency; agency problem; information asymmetry

Sparks, Tim. "Integrated surface water-groundwater modelling : linking surface water and groundwater using DIVAST-SG." Thesis, Cardiff University, 2007. http://orca.cf.ac.uk/54617/.

Full text
Abstract:
Surface-water and groundwater are two resources both requiring careful management and protection. Computer modelling of both has long been used as an aid to their management. Historically they have been modelled separately, as their behaviour is represented by different mathematical equations. However, in reality, they are a linked resource each affects the other. DIVAST is a two-dimensional hydrodynamic and water quality numerical model developed for estuarine and coastal modelling. The original model enables the simulation of problems such as pollution and flooding in surface waters. In this study the existing model is extended to allow the modelling of groundwater as well as surface water in the same model. Chapters 1-5 introduce the problem, review some existing models, and then derive, discretise, and implement the equations for surface water and groundwater flow into the new model. Chapters 6-10 test the new model against analytical solutions, laboratory data, field data, and an existing groundwater model (MODFLOW). The outcome is a new version of the DIVAST model, known as DIVAST-SG (Depth Integrated Velocities And Solute Transport in Surface water and Groundwater). It simulates interactions between two-dimensional surface water and groundwater, in addition to the facilities of the original code. The equations are solved within one model, avoiding coupling problems. It is successfully tested against analytical solutions, laboratory studies and field data, and compared to an existing groundwater code, where it successfully models a gravel aquifer adjacent to tidal surface water. A framework is laid for continuing this work to produce a pseudo 3-D surface- water / groundwater code. In addition, novel techniques are pioneered in the laboratory, where open cell foam is used in a tidal flume to represent a porous aquifer adjacent to a river, and a highly detailed dataset of groundwater field data is compiled in the course of the work.

Swept Source Optical Coherence Tomography (SS-OCT) is a medical imaging technique that requires high repetition rate, widely-tunable coherent laser sources. Sampled grating distributed Bragg reflector (SG-DBR) lasers are proven in telecom applications and are expected to fulfill the requirements for SS-OCT at a significantly lower cost than alternative solutions. Constructed entirely on a semiconductor substrate, SG-DBR lasers require four synchronized waveforms to modulate the output wavelength and intensity. Because of this unique tuning mechanism, there are a number of systematic and noise sources that can affect the quality of the OCT point-spread function (PSF). Based on these noise sources, software is developed to simulate the waveforms in an SS-OCT system and determine the factors that limit width of the PSF central peak and the broadband skirts. Design curves are then created to specify the requirements to obtain a given performance. Next, experimental tests are performed on a JDS Uniphase C-band SG-DBR laser to assess its performance limitations. Finally, by comparing theory and experiment, recommendations are made on acceptable systematic and random noise errors induced in the PSF.

Abstract:
Microwave signal generation is demonstrated using a single-chip fast wavelength-tunable SG-DBR laser. Microwave signals are established by a delayed self-heterodyne technique. The optical frequency of the laser is square-wave modulated between two closely spaced wavelengths. These two wavelengths are made time coincident using a fiber based Mach-Zehnder interferometer. From the interferometer, the difference frequency is detected and amplified. The optical difference frequency takes on the form of a microwave signal that can be seen on an electrical spectrum analyzer. The hardware structure of the high-speed wavelength switching SG-DBR package is shown. The wavelength switching architecture and temperature robustness of the laser will also be presented. Microwave signals up to 17 GHz have been generated by frequency modulating the phase section of the laser. Measured from the microwave signal spectral width, laser linewidth values as narrow as 10 MHz have been achieved for low back mirror current inputs. Linewidth results with respect to device DC bias conditions are also presented. Time resolved frequency step measurements have shown inherent thermal transients of approximately 200 ns upon wavelength switching. From the square wave switching profile, switching times of approximately 24 ns were achieved.

ABSTRACT – Salmonella Gallinarum (SG) is a host-restrict pathogen that causes fowl typhoid, a severe systemic disease that is one of the major concerns to the poultry industry worldwide. When infecting the bird, SG makes use of evasion mechanisms to survive and to replicate within macrophages. In this context, phoPQ genes encode a two-component regulatory system (PhoPQ) that regulates virulence genes responsible for adaptation of Salmonella spp. to antimicrobial factors such as low pH, antimicrobial peptides and deprivation of bivalent cations. Herein, we aimed to investigate the role of the mentioned genes to SG. Thus, a phoPQ-depleted SG strain (SG ∆phoPQ) was constructed and its virulence assessed in twenty-day-old laying hens susceptible to fowl typhoid. SG ∆phoPQ did cause neither clinical signs nor mortality in birds orally challenged, being non-pathogenic. Furthermore, this strain was not recovered from livers or spleens. On the other hand, chickens challenged subcutaneously with the mutant strain had discreet to moderate pathological changes and also low bacterial counts in liver and spleen tissues. These findings show that SG ∆phoPQ is attenuated to susceptible chickens and suggest that both genes are important during chicken systemic infection by SG.

Abstract:

Abstract:
Metallic quantum clusters are stable structures that can exhibit many useful magnetic, chemical, and optical properties. Developing clusters for specific applications requires accurate methods for characterizing their physical and chemical properties. Most cluster characterization methods are ensemble-based measurements that can only measure the average values of the cluster properties. Single cluster measurements improve upon this by yielding information about the distribution of cluster parameters. This investigation describes the initial results on a new approach to detecting and characterizing individual gold nanoclusters (Au25(SG)18) in an aqueous solution with nanopore-based resistive pulse sensing. We also present a new application where the clusters are shown to increase the mean residence time of polyethylene glycol (PEG) molecules within an alpha hemolysin (αHL) nanopore. The effect appears over a range of PEG sizes and ionic strengths. This increases the resolution of the peaks in the single molecule mass spectrometry (SMMS) current blockade distribution and suggests a means for reducing the ionic strength of the nanopore solute in the SMMS protocol.

In the current context, a country's economic growth strongly depends on its success at an international level. Among the dynamics imposed by globalization, new protagonists and new strategic partnerships where the language spoken plays a critical role, emerges. Therefore, States have been giving growing attention to the implementation of measures that allow for the exploration of opportunities in the economies that share the same language and culture. A greater coherence in the promotion of international presence and the use of indigenous resources is the source of added value that creates wealth, increases productivity and contributes to reaching full-employment. This thesis assesses the relevance of the spoken language in the internationalization of an economy and studies the case of a specific Portuguese company.

Abstract:
The meteorological observations from satellites in the microwave domain are currently limited to 190 GHz. The next generation of European operational Meteorological Satellite (EUMETSAT Polar System-Second Generation-EPS-SG) will carry an instrument, the Ice Cloud Imager (ICI), with frequencies up to 664 GHz, to provide unprecedented measurements in the sub-millimeter spectral range, aiming to improve the characterization of ice clouds over the globe. To prepare this upcoming satellite-borne sub-millimeter imager, during this thesis, scientific efforts have been made on four complementary aspects. Realistic radiative transfer simulations have been performed from 19 to 700 GHz, for real meteorological scenes, covering a large variability of clouds in Europe. The goal was two fold, first to better understand the sensitivity of the microwave to sub-millimeter waves to the cloud frozen phases, and second, to create a robust training database for a statistical cloud parameter retrieval. The Atmospheric Radiative Transfer Simulator (ARTS) is coupled with atmospheric profiles from the Weather Research and Forecasting (WRF) model, for twelve diverse European mid-latitude scenes. The single scattering properties of the hydrometeors (cloud ice, snow, graupel, rain, and cloud water) are carefully selected, especially for the frozen phases, and compatibility with the micro- physics in WRF is tested: the Discrete-Dipole Approximation (DDA) is adopted for snow particles. The resulting simulations have been systematically compared with coincident satellite observations from imagers and sounders up to 200 GHz. The agreement between simulations and observations shows the good quality of the simulated training database, at least up to 200 GHz

Abstract:
The presente research was undertaken in the \"Morro do Setuva\" region and neighborhoods, countryside of Bocaiúva do Sul - Paraná, emphasizing geological mapping. In this ares outcrops the Atuba Complex and the Setuva, Capiru and Votuverava formations. The area was mapped in 1:50.000 scale. Eleven associations were mapped and three of them are related to Atuba Complex Domain: bended biotite-hornblende gneisses association (Pca3). The Capiru Formation Domain is represented by five lithological associations: silt-sand meta-rhythmites (Mc1), massive dolomitic meta-limestone (Mc2), fine-coarse quartzites (Mc4) and red phyllite (Mc5). These associations suggested marine regressive sequence to the sedimentation in the shallow carbonate platform. The Setuva Formation Domain corresponds to the following association: sericite schists (Ms1), fine-coarse quartzite and milonitics dolomitic meta-limestones (Ms3). Capiru and Setuva sedimentation basin was originated by an extensional event that produced an initial rift, which reached passive continental margin condition with a stable platform. In the Votuverava Formation Domain occurs meta-pelites (Mv1) and meta-basites (Mv2), Mv1 and Mv2 assigned different tectonic-sedimentary cycles from the ones that were deposited in Capiru and Setuva formations; the Lancinha Shear Zone is the tectonic limit between this domain and the others. The Atuba Complex is the local basemed of Capiru and Setuva formations and with them the complex has contacts through thrust shear zones with WNW-ESE transport. This event was responsible for a wide deformation of Setuva Formation and it had originated tectonofacies. A transpressive event, with major shear zones and associated folding, was recognized, the \"Antiforma do Setuva\" was rhe most important fold register. The metamorphism affected the Votuverava, Capiru and Setuva formations is green schist, chlorite zone. In the Atuba Complex was detected a development under high, medium and low polymetamorphism conditions.

This master’s dissertation describes all the work carried out to numerically compare the aerodynamic performance of a regular flapped airfoil to a morphing flapped airfoil of the Rotating Rib style. Based on works previously done, the main purpose of this thesis is to validate experimental results retrieved in wind tunnel. Computational Assisted Design tools (CAD), to model the geometry of the wings’ profiles based on the ones already built, Computational Fluid Dynamics tools (CFD), through numerical calculation, and postprocessing tools, for data analyses, were used. To better define which model to use a comparison between the many available models in the tools with experimental data previously validated and revalidated in other studies. The turbulence model used was the k-ε Realizable with Standard Wall Functions, as it revealed itself as the closest to the experimental data. So a two-dimensional analysis was made, with a steady-state flow and a pressure-based solver with an absolute velocity formulation. The mesh created is a type C with an O type central zone of high resolution for a better definition of the boundary layer. The results show significant improvements concerning the morphing wing, regarding the lift and drag coefficients, which may lead to the improvement of the aircraft performance.

Abstract:
Temporal gravimeter observations, used in geodesy and geophysics to study variation of the Earth’s gravity field, are influenced by local water storage changes (WSC) and – from this perspective – add noise to the gravimeter signal records. At the same time, the part of the gravity signal caused by WSC may provide substantial information for hydrologists. Water storages are the fundamental state variable of hydrological systems, but comprehensive data on total WSC are practically inaccessible and their quantification is associated with a high level of uncertainty at the field scale. This study investigates the relationship between temporal gravity measurements and WSC in order to reduce the hydrological interfering signal from temporal gravity measurements and to explore the value of temporal gravity measurements for hydrology for the superconducting gravimeter (SG) of the Geodetic Observatory Wettzell, Germany. A 4D forward model with a spatially nested discretization domain was developed to simulate and calculate the local hydrological effect on the temporal gravity observations. An intensive measurement system was installed at the Geodetic Observatory Wettzell and WSC were measured in all relevant storage components, namely groundwater, saprolite, soil, top soil and snow storage. The monitoring system comprised also a suction-controlled, weighable, monolith-filled lysimeter, allowing an all time first comparison of a lysimeter and a gravimeter. Lysimeter data were used to estimate WSC at the field scale in combination with complementary observations and a hydrological 1D model. Total local WSC were derived, uncertainties were assessed and the hydrological gravity response was calculated from the WSC. A simple conceptual hydrological model was calibrated and evaluated against records of a superconducting gravimeter, soil moisture and groundwater time series. The model was evaluated by a split sample test and validated against independently estimated WSC from the lysimeter-based approach. A simulation of the hydrological gravity effect showed that WSC of one meter height along the topography caused a gravity response of 52 µGal, whereas, generally in geodesy, on flat terrain, the same water mass variation causes a gravity change of only 42 µGal (Bouguer approximation). The radius of influence of local water storage variations can be limited to 1000 m and 50 % to 80 % of the local hydro¬logical gravity signal is generated within a radius of 50 m around the gravimeter. At the Geodetic Observatory Wettzell, WSC in the snow pack, top soil, unsaturated saprolite and fractured aquifer are all important terms of the local water budget. With the exception of snow, all storage components have gravity responses of the same order of magnitude and are therefore relevant for gravity observations. The comparison of the total hydrological gravity response to the gravity residuals obtained from the SG, showed similarities in both short-term and seasonal dynamics. However, the results demonstrated the limitations of estimating total local WSC using hydrological point measurements. The results of the lysimeter-based approach showed that gravity residuals are caused to a larger extent by local WSC than previously estimated. A comparison of the results with other methods used in the past to correct temporal gravity observations for the local hydrological influence showed that the lysimeter measurements improved the independent estimation of WSC significantly and thus provided a better way of estimating the local hydrological gravity effect. In the context of hydrological noise reduction, at sites where temporal gravity observations are used for geophysical studies beyond local hydrology, the installation of a lysimeter in combination with complementary hydrological measurements is recommended. From the hydrological view point, using gravimeter data as a calibration constraint improved the model results in comparison to hydrological point measurements. Thanks to their capacity to integrate over different storage components and a larger area, gravimeters provide generalized information on total WSC at the field scale. Due to their integrative nature, gravity data must be interpreted with great care in hydrological studies. However, gravimeters can serve as a novel measurement instrument for hydrology and the application of gravimeters especially designed to study open research questions in hydrology is recommended.

Abstract:

Abstract:
In this project, the performance aspects of a new early generation 1310 nm Sampled-Grating Distributed Bragg Reflector (SG-DBR) semiconductor laser are investigated. SG-DBR lasers are ideal for Source Swept Optical Coherence Tomography (SS-OCT), a Fourier-Domain based approach for OCT, necessitating a tunable wavelength source. Three internal sections control the frequency output for tuning, along with two amplifiers for amplitude control. These O-band SG-DBR devices are now being produced in research quantities. SG-DBR lasers have been produced at 1550 and 1600 nm for some times. Fundamental questions regarding the performance of the 1310 nm devices must be quantified. Standard metrics including the laser linewidth, amplitude modulation and frequency modulation responses are characterized. The intrinsic electrical parasitics of the laser diode segments and packaging are also investigated. In addition, testing fixture including a Thermal Electric Cooler (TEC) controller is for the characterization task. Measurements of these key metrics are essential to the enhancement of future devices, aiding in the optimization of more mature products.

Abstract:
We grow AB₂ random hyperbranched polymer structures in different ways and using different simulation methods. In particular we use a method of ad hoc construction of the connectivity matrix and the bond fluctuation model on a 3D lattice. We show that hyperbranched polymers split into two universality classes depending on the growth process. For a “slow growth” (SG) process where monomers are added sequentially to an existing molecule which strictly avoids cluster–cluster aggregation the resulting structures share all characteristic features with regular dendrimers. For a “quick growth” (QG) process which allows for cluster–cluster aggregation we obtain structures which can be identified as random fractals. Without excluded volume interactions the SG model displays a logarithmic growth of the radius of gyration with respect to the degree of polymerization while the QG model displays a power law behavior with an exponent of 1/4. By analyzing the spectral properties of the connectivity matrix we confirm the behavior of dendritic structures for the SG model and the corresponding fractal properties in the QG case. A mean field model is developed which explains the extension of the hyperbranched polymers in an athermal solvent for both cases. While the radius of gyration of the QG model shows a power-law behavior with the exponent value close to 4/5, the corresponding result for the SG model is a mixed logarithmic–power-law behavior. These different behaviors are confirmed by simulations using the bond fluctuation model. Our studies indicate that random sequential growth according to our SG model can be an alternative to the synthesis of perfect dendrimers.

Abstract:
Motion artefacts due to cardiac and respiratory motion present a daily challenge in cardiac Magnetic Resonance Imaging (MRI), and many different motion correction procedures are used in clinical routine imaging. To reduce motion artefacts further, patients are required to hold their breath during parts of the data acquisition, which is physically straining – especially when done repetitively. Self-Gating (SG) is a method that extracts cardiac and respiratory motion information from the MRI data in the form of signals, called SG signals, and uses them to divide the data into the specific cardiac and respiratory phases it was acquired from. This method both avoids motion artefacts and allow for free-breathing acquisition. This project’s goal was to find a method for extracting cardiac and respiratory SG signals from MRI data. The data was acquired with a golden angle radial acquisition method for 3-dimensional (3D) scans. Extraction of the raw signal was tested for both raw k-space data and high temporal resolution image series, where the images were reconstructed using a sliding window reconstruction. Filters were then applied to isolate the cardiac and respiratory information, to create separate cardiac and respiratory SG signals. Thereafter trigger points marking the beginning of the cardiac and respiratory cycles were generated. The trigger points were compared against ECG and respiratory trigger points provided by the MR scanner. The conclusion was that the SG signals based on k-space data was functional on the scans from the evaluated subjects and the most effective choice of the two options, but image based SG signals may prove to be functional after further studies.

Abstract:
An antinomy is a contradiction between a principle and its opposite, where there is a compelling case for accepting both. This thesis adopts the antinomy of local-state government in Australia as its central conceptual theme, describing it with the following defensible, but contradictory principles that:¶ · Australian local governments are statutory agencies of Australia’s state governments, with no power or authority beyond that which is ascribed to them by the states (the outside-in principle); and¶ · Local governments in Australia are independent agencies whose authority and interests transcend their regulatory powers by nature of their attachment to their local area (the inside-out principle).¶ The central conceptual theme of the antinomy of local-state government shapes the overall thesis, as well as providing the focus for its introduction and conclusion. The thesis induces elements of the antinomy and structures much of its discussion around these key issues. It does not try to prove or resolve the antinomy. Instead the thesis uses the concept to explore and develop its second complex theme - the practical and applied experience of Australian local governments (LGs) as they attempt to deliver beneficial environmental outcomes. The great bulk of the substantive work presented in the thesis focuses on descriptions and analyses of LGs’ environmental work and the contexts within which they do it. The thesis contends that the local-state antinomy underpins many problems facing Australian LGs as they attempt to deliver beneficial environmental outcomes. Four research questions are addressed. They are:¶ · How can Australian LG capacity to deliver beneficial environmental outcomes be understood?¶ · Within this capacity, what are the environmental outcomes now being achieved by Australian LGs?¶ · How can Australian local government extend its capacity to deliver beneficial environmental outcomes? And¶ · What are the implications of the local-state antinomy on Australian LG capacity to deliver beneficial environmental outcomes?¶ This thesis reviews literature on Australian LG, LG environmental work, and the methods that are appropriate in investigating these questions. The overall thesis uses scientific, grounded theory and action research methods and draws on ideas from symbolic interactionism. Parts of the thesis also use environmental risk assessment, gap analysis techniques, case study and comparative analysis. The goal of generating grounded theories led to a strong focus on the development and exploration of analytical categories and the relationships between them. One such category summarises the relationship between LG and state government (SG), whereby LGs are identified as the inside sphere of government, while the SG is one of several outside spheres. Environmental efforts that impact between the spheres are described in relation to their source and impact, using this terminology, so that inside-out initiatives are driven by LGs but impact more broadly, and outside-in initiatives are driven by states but impact on local areas.¶ Two extensive studies are presented, each stemming primarily from one side of the local-state antinomy. The first is a quantitative, statewide study of local (and state) government implementation of the Queensland Environmental Protection Act. That process is considered a predominantly outside-in environmental initiative, in that LG interest and authority for that work stem directly from a SG statute. For simplicity, this is referred to as an outside-in study. That study involved the development and application of the Comparative Environmental Risk Assessment Method, that enabled the assessment of the environmental and other outcomes from the Queensland legislation.¶ The outside-in study is complimented by comparative case studies that mostly reflect inside-out environmental initiatives as they are defined and described by LGs. Again, this required the development of innovative research methods, specifically a comparative case study method. 34 case studies gathered from different types of LGs across Australia are presented, each representing an attempt by LG to deliver beneficial environmental outcomes.¶ In answer to the research questions, LG capacity to deliver environmental outcomes can be understood when the antinomy is examined through the research methods and analytical categories developed and presented here. LGs are delivering significant beneficial environmental outcomes, both as agents of SGs and through their own initiatives. Improving LG capacity to deliver environmental outcomes primarily requires a respect for LG perspectives, and for LG priorities, which inherently include a focus on their own local areas. State governments can build effective partnerships between the spheres and enhance LG environmental capacity by recognising and supporting LG’s own priorities, while assisting their engagement with broader strategic objectives.

Abstract:
Currently, in the period of Poststructualism literature is widely approached as a cultural and social phenomenon and the aims of literary studies as the illuminating of social codes, ideology etc. embedded in literature. This study evaluates the contribution of specific critics working within the so-called "New paradigm". The concept "New paradigm" is used as an umbrella term covering a number of distinct developments, referring to literature as a system of literary "actions" rather than as a canon of books. In recent years several Afrikaans critics have embraced or adapted current theories in the domains of Deconstruction, Ideology Criticism, Literary, Sociology, Recepticm Aesthetics, Semiotics and Feminism. This study explores and evaluates the application of such theoretical paradigms to their work in the field of practical criticism. This study also explores the relationship between current theories of literature and their antecedents. The notion of current literary theory and practice not only as a broadening of scope and content of the notion of the "literary" but on the other hand as a liberation of theory and practice from an over-deterministic thrust (i.e. the approach of literature as no more than a set of social documents) is also evaluated in this study. A twofold aim of this study can thus be defined: a. to determine the extent to which the shift from the "old" to "new" theories was made necessary by the developing literary and social scene; b. to examine the links beween theory and practice in the work of specific, key figures in Afrikaans. Is it possible for theory and practice to be separated? Is there an important consistence in the relationship between the two in the work of the figures chosen?

Abstract:
This thesis deals with issues around the implementation of an AMM (Automated Meter Management) system within the Czech electricity distribution system. The theoretical section of the thesis provides a historical context and looks at the development of the electricity sector. The thesis then goes on to discuss issues regarding the competitiveness of distribution companies in terms of IT/ICT use. This section defines the elements of the Smart Grid concept, in particular the AMM system. The chapters dealing with the conditions for implementing the system in the Czech Republic and pilot projects abroad make up the major sources for the practical section. The goal of the practical section is to come up with a project aim for the AMM system implementation trial project in Prague. The proposed project aim is divided into a number of separate tasks whose results are obtained using a detailed analysis of available sources and the conclusions of the theoretical part of the thesis. The thesis's main benefit is in forming a basis for the AMM implementation trial project in PREdistribuce, Inc. distribution networks and providing a comprehensive overview of the development and current state of the electricity sector.

Fernandes, Margareth.

Abstract:
Actually, chronic lymphocytic leukemia (CLL) can be divided in two subsets: one with somatically mutated immunoglobulin heavy-chain variable-region genes (MIgVH) and other with unmutated sequences. (UMIgVH). Some studies have shown that CD38 expression in CLL cells are correlated with IgVH mutational status. However, the value of CD38 as surrogate IgVH mutational status is controversial. Recent studies, have found that Zap-70 protein tyrosine kinase expression is strongly associated with the mutational status IgVH. The aim of this study was to evaluate the Zap-70 and CD38 expression, for flow cytometry, in CD19+ LLC cells and correlate with the Binet’s staging system, treatment-free survival (TFS) and a overall survival (OS). Zap-70 and CD38 was evaluated, in 144 CLL patients that was classified in A, B and C Binet’s staging system: 59 (41%) in stage A, 38 (26%) in B and 47 (33%) in C. We observed low Zap-70 and CD38 expression in stage A patients than in stage B and C cases. When we analyzed the TFS in stage A patients Zap-70+ and CD38+ patients showed shorter TFS than Zap-70- and CD38-. Then we observed that the OS of Zap-70+ and CD38+ patients was, also, shorter than Zap-70- and CD38- cases. However, statistical differences was not found when Zap-70 and CD38 expression was correlated with stage A, B or C Binet’s staging system. To understand the associated Zap-70 and CD38 expression, we divided the CLL patients in two subgroups (Zap-70-/CD38 - and Zap-70+ or CD38+). We observed that CD38+ or Zap-70+ was associated Binet’s staging system, once most of stage B (74%) and C (66%) patients are Zap-70+ or CD38+. However, stage A patients, Zap-70+ or CD38+, showed shorter OS than Zap-70-/CD38-. These differences were not observed in stage B and C patients. Shorter TFS was also observed in the Zap-70+ or CD38+ stage A patients. These results suggest that combined analysis of Zap-70 and CD38 can be used to evaluate stage A patients to observe the clinical evolution of the disease. Nevertheless, other studies must be carried to confirm the clinical use of these markers.
