O poeta italiano Vittorio Sereni (1913-1983) é ainda inédito e pouco conhecido no Brasil. A fim de introduzir o autor ao público brasileiro, este trabalho tem como objeto de estudo seu livro de estreia, Frontiera, de 1941. A partir de um panorama que percorre de maneira sucinta o surgimento dos poetas modernos da Itália, e que situa o leitor acerca da posição ocupada por Vittorio Sereni nesse cenário, esta pesquisa investiga os caminhos cursados pelo poeta em sua juventude, no início de sua produção literária. Tal percurso, conforme se demonstrará, foi determinante para sua completa formação humana, cultural e intelectual, abrangendo, por exemplo, as aulas do filósofo Antonio Banfi na Universidade de Milão, a experiência como redator da revista \"Corrente di vita giovanile\", e a admiração e amizade com os poetas Eugenio Montale, Giuseppe Ungaretti e Umberto Saba. Ademais, este trabalho discute o polêmico tema do pertencimento ou não de Sereni ao movimento hermético italiano dos anos 30/40. Finalmente, por meio de análises de quatro poemas do livro selecionado, \"Inverno\", \"Inverno a Luino\", \"Strada di Zenna\" e \"Ecco le voci cadono\", serão especificadas algumas linhas temáticas centrais de Frontiera recorrentes em outras composições de sua primeira publicação.Italian poet Vittorio Sereni (1913-1983) is still unpublished and relatively unknown in Brazil. In order to introduce the author to the Brazilian public, this research studies his debut book, Frontiera, 1941. Through a brief overview of the history of the Italian modern poets, the present study investigates the path followed by Vittorio Sereni during his youth, at the beginning of his literary career. Such path, as will be shown later, was of the utmost importance for the poets humanitarian, cultural and intellectual development, and includes the philosophy lectures given by Antonio Banfi at the University of Milan, the time spent as a writer at the magazine \"Corrente di vita giovanile\" and the admiration of and friendship with fellow poets Eugenio Montale, Giuseppe Ungaretti and Umberto Saba. Moreover, this research addresses the controversial debate on whether Sereni integrated or not the Italian hermetic movement in the 1930s and 40s. Lastly, through the study of four poems from the selected book, \"Inverno\", \"Inverno a Luino\". \"Strada di Zenna\" e \"Ecco le voci cadono\", recurrent central themes in Frontiera will be presented and analyzed.

O Feijão Caupi (Vigna unguiculata (L.) Walp) é uma leguminosa com importante representatividade econômica e nutricional, especialmente no Brasil. Inibidores de serino proteases, como a tripsina, já foram descritos na espécie, assim como em outras plantas. No entanto, nesta espécie, ainda não foram identificados inibidores que apresentem atividade sobre a elastase de neutrófilos humana (HNE), protease envolvida em muitos processos patológicos, como na instalação e progressão da doença pulmonar obstrutiva crônica (DPOC). Nesse estudo, purificamos um inibidor a partir do extrato protéico de Vigna unguiculata que apresenta atividade sobre HNE. Inicialmente, foi realizado o processo de extração alcalina de proteínas, seguido de três passos cromatográficos distintos, utilizando as colunas Hitrap-Q (Troca-iônica), Source15RPC (Fase-Reversa) e ACE18 (Fase-Reversa). Essas etapas foram acompanhadas por testes de atividade inibitória, utilizando os substratos fluorogênicos Meo-Suc-Ala-Ala-Pro-Val-MCA (Elastase) e Z-Phe-Arg-MCA (Tripsina), além de ensaios da quantificação de concentração total de proteínas. Para determinar a massa do inibidor, foram utilizadas as técnicas de espectrometria de massa por MALDI-TOF e SDS-PAGE, o inibidor apresenta massa molecular de 10,99 KDa. O Ki para HNE foi determinado no valor de 9 pM. O inibidor não apresentou atividade inibitória sobre tripsina e trombina, porém foi observada atividade sobre subtilisina e quimotripsina. Estes dados indicam que o inibidor purificado trata-se de uma molécula ainda não caracterizada, devido às suas atividades inibitórias o nomeamos de Vigna unguiculata Elastase Inhibitor (VuEI).
The cowpea (Vigna unguiculata (L.) Walp) is a legume of important economic and nutritional representativeness, especially in Brazil. Serine protease inhibitors, such as trypsin, have been described in many species, as well as in other plants. In this specie an inhibitor with activity on human neutrophil elastase (HNE) has not yet been identified. This protease is involved in many pathological processes, such as the onset and progression of chronic obstructive pulmonary disease (COPD). We purified and characterized an inhibitor from the protein extract of Vigna unguiculata presenting activity towards HNE. Firstly, we performed the alkaline extraction procedure for proteins followed by three different chromatographic steps using Hitrap Q (ion exchange), Source15RPC (Reversed-Phase) and ACE18 (Reversed Phase) columns. These steps were followed by the inhibitory activity tests using fluorogenic substrates, MeO-Suc-Ala-Ala-Pro-Val-MCA (elastase) and Z-Phe-Arg-MCA (trypsin), and quantitation assays of protein concentration. To determinate the size of the molecule, we used MALDI-TOF mass spectrometry and SDS-PAGE. The molecular mass of the inhibitor was 10,99 kDa. The dissociation constant (Ki) toward HNE was 9 pM. HNE inhibitor showed no inhibitory activities toward trypsin and thrombin. However, the inhibitor presented activity toward subtilisin and chymotrypsin. These datas indicate that this molecule is a novel inhibitor to HNE and we named it Vigna unguiculata Elastase Inhibitor (VuEI).

La conception de la rhetorique de francis ponge peut paraitre contradictoire. Il y a en effet, une evolution significative du poete sur la question des "regles techniques" en matiere d'ecriture. Ponge preconise d'abord leur explicitation et engage meme une enquete sur les "questions rhetoriques" avec la revue "les cahiers du sud" puis il les declare inessentielles : les veritables regles relevant de l'indicible ne peuvent etre edictees. Pour comprendre ce changement de cap apparent, il faut examiner la position de ponge dans le paysage litteraire et plus largement dans le champ ideoloogique de l'apres-guerre. Son isolement le contraint, pour echapper a la fois au neo-academisme sous toutes ses formes et a l'histrionisme poetique toujours vivace, a inventer une nouvelle rhetorique paradoxale, "en acte", singulierement comprise dans le texte. A la fois art poetique et art de vivre, son lieu d'election se trouve chez lucrece, dans les "templa serena". On peut donc examiner la rhetorique pongienne "a l'oeuvre" et en faire emerger une figure inouie : l'indifference qui permet de concilier paradoxalement les antinomies et en particulier celles de l'analogie et de la difference, du meme et de l'autre. C'est ce travail de l'indifference qu'on peut lire a travers "les fictions rhetoriques" pongiennes qui "recitent" le noeud singulier des mots et des choses.

Memoria para optar al título de ArquitectoEl proyecto de Título consiste en el desarrollo de una propuesta de carácter cívico cultural en el Centro Histórico de la ciudad de La Serena, para revertir su obsolescencia generada por la expansión desmedida hacia la periferia. El polígono escogido forma parte de un conjunto de Patrimonio Industrial, donde funcionó la Ex Cervecería Floto-CCU, que actualmente se encuentra en estado de abandono, por lo que su condición no va de acuerdo a las dinámicas del sector ni con las proyecciones de crecimiento para el casco histórico, que además pertenece a la Zona Típica más extensa del país. Al contar con una gran superficie subutilizada, el terreno ofrece la oportunidad de poner en valor el patrimonio existente y a su vez dar cabida para la incorporación de nuevos programas que consoliden el carácter del lugar. Si bien el eje central del proyecto es la proposición del edificio consistorial dada por la actual dispersión de las dependencias municipales y la ausencia de un espacio representativo para la ciudadanía, también busca poner en valor el patrimonio existente, que hasta la fecha no se encuentra categorizado con interés histórico o patrimonial, por lo que su estado de vulnerabilidad es mucho mayor. Por medio de la intervención del conjunto, se otorga un plan de uso a los inmuebles a reutilizar, donde de acuerdo al análisis del área, se inserta un programa enfocado en el desarrollo de actividades cívicas y culturales, considerando, además, la relación entre los bordes de la manzana y su entorno. Hacia el norte y el oriente deberá enfrentarse principalmente con la actividad residencial dada por el Barrio patrimonial Almagro y su cercanía con la ribera sur del Río Elqui, mientras que hacia el sur y el poniente se encuentra vinculado con usos comerciales y la Plaza de Armas junto a edificios institucionales circundantes, donde el proyecto podrá conectarse a través de la apertura de sus bordes. Se propone una estrategia de recuperación a partir de una redefinición de las escalas del lugar, sus límites y relación con el tejido urbano, estableciendo diferentes etapas de desarrollo que permitan vincular de manera armónica el contexto urbano presente con los vestigios industriales del pasado, logrando una propuesta coherente con las nuevas necesidades de la ciudad y la región.

Esta dissertação constitui uma reflexão sobre a situação social, política e cultural da Inglaterra na década de 70, especialmente nos anos de 1972 e 1973, como descrita no romance Serena, de Ian McEwan. O objetivo principal desta pesquisa é apresentar como o momento histórico supracitado é narrado no discurso ficcional, a partir da perspectiva da personagem Serena, em comparação com o discurso histórico, de acordo com fontes selecionadas, que narram a história da Inglaterra no contexto da Guerra Fria, dos ataques terroristas do IRA e do funcionamento do Serviço Secreto doméstico inglês, chamado MI5.This dissertation constitutes a reflection about the social, political and cultural situation of England in the 70s, especially in the years of 1972 and 1973, as described in the novel Sweet Tooth, by Ian McEwan. The main objective of this research is to present how the historical moment mentioned above is narrated in the fictional discourse, from the perspective of the character, in comparison with the historical discourse and from a selection of sources, which narrate the history of England in the context of the Cold War, the terrorist attacks of the IRA and the functioning of the English domestic Secret Service, called MI5.

A dengue é um problema de saúde pública mundial, acometendo cerca de 50 milhões de pessoas por ano. O HCV é a principal causa de hepatite crônica e estima-se que 200 milhões de pessoas estejam infectadas no mundo. A terapia atual contra o HCV apresenta eficácia limitada e baixa tolerância, enquanto que para a dengue não existe um tratamento antiviral específico. Com o objetivo de explorar essa demanda, o presente trabalho descreve a síntese para a obtenção de quinze compostos peptideomiméticos inéditos, contendo o cerne rígido proveniente do isosorbídeo, como potenciais agentes inibidores da enzima NS3 protease de ambos os vírus. A estratégia de síntese dos peptideomiméticos consistiu inicialmente na obtenção de dez compostos inéditos, das séries 28a-c (derivada de oxazolonas) e 29a-g (derivada de aminoácidos N-protegidos). Os compostos foram inicialmente testados frente à inibição da protease do DENV-2, os quais não apresentaram resultados significativos. Entretanto, os resultados de inibição enzimática dos compostos frente à NS3/4A do HCV-1b foram mais satisfatórios, sendo o composto 28a (1,4:3,6-dianidro-5-[[(2Z)-2-(benzoilamino)-1-oxo-3-(2-tienil)-2-propen-1-il]amino] -2-deoxi-2-O-(fenilmetil)-D-iditiol, o mais ativo, com IC50 = 88μm. Estudos de docking molecular foram realizados para avaliar o modo de interação dos ligantes com a serina protease NS3/4A do HCV. Dentre os compostos sintetizados, 28a foi considerado protótipo para o desenvolvimento de novos compostos com potencial de inibição da enzima em questão. A partir de 28a, a nova série de compostos inéditos 42a-e foi proposta e obtida. Os resultados dos ensaios farmacológicos frente à NS3/4A do HCV-1b permitiram-nos inferir que a substituição do tiofeno de 28a pelo furano e 3-metiltiofeno em 42b e 42c, respectivamente, resultou em um aumento de cerca de 30% no perfil de inibição enzimática, com inibição de 70% dessa enzima viral. Dessa forma, podemos identificar esses dois novos compostos como protótipos para o planejamento de futuras moléculas potencialmente inibidoras da enzima serina protease do vírus da Hepatite C
Dengue fever is a worldwide public health concern, affecting approximately 50 million people per year. HCV is the main cause of chronic hepatitis and it is estimated that 200 million people are infected worldwide. Current therapy against HCV has limited efficacy and low tolerance, and there is no specific antiviral treatment for dengue fever. In order to exploit this demand, this work describes the synthesis for obtaining fifteen novel peptidemimetic compounds, isosorbide derivatives as potential inhibitory agents of the NS3 protease enzyme of both viruses. Initially the synthetic strategy of peptidemimetics consisted in obtaining ten novel compounds: 28a-c (derived from oxazolones) and 29a-g (derived from N-protected amino acids) series. The compounds were initially tested on the inhibition of protease DENV-2 , which results were not significant. However, the results of enzymatic inhibition of the compounds against the NS3/4A HCV-1b was more satisfactory, and the compound 28a (1,4 : 3,6- dianhydro -5 - [[( 2Z ) -2- ( benzoylamino) -1 -oxo- 3- (2- thienyl) -2 - propen -1- yl] amino] -2- deoxy -2- O- ( phenylmethyl ) -D- iditiol) , the most active , presenting IC50 = 88μm. Molecular docking studies were performed to assess the compounds mode of interaction with the serine protease NS3 / 4A HCV. Among the compounds synthesized, 28a was considered a prototype for the development of new compounds with a potential inhibition of the enzyme in question. Staring from 28a as a prototype the 42a-e serie was proposed and obtained. The results of pharmacological tests against the NS3/ 4A HCV -1b allowed us to conclude that substitution of the 28a furan thiophene and 3- methylthiophene in 42b and 42c respectively resulted in an increase of 30 % in the profile enzyme inhibition , with inhibition of 70 % of the viral enzyme . Thus , we can identify these new compounds as prototypes for planning future potentially molecule inhibitors of serine protease enzyme of the Hepatitis C virus

La traduzione poetica viene affrontata sul piano empirico dell'analisi testuale.
Una breve introduzione presenta le riflessioni più importanti sulla traduzione del testo poetico, da Benjamin e Steiner fino alle teorie più recenti di Meschonnic, Apel, Berman e Mattioli. Alla luce di queste teorie vengono analizzate le opere di due coppie di poeti e poeti-traduttori. Nel primo esempio troviamo il poeta svizzero (francofono) Philippe Jaccottet alle prese con l'intera opera di Ungaretti; nel secondo il rapporto travagliato di Vittorio Sereni con la poesia di René Char.
Oltre a indagare la natura problematica della traduzione poetica come pratica e come esperienza, questa tesi di Letteratura Comparata vuole presentare la traduzione come strumento ermeneutico e come meccanismo rienunciativo: il suo ruolo nella dialettica delle influenze e dell'evoluzione letteraria è da considerarsi infatti essenziale. La vocazione originariamente etica della traduzione è sfondo costante della trattazione.Poetic translation is faced on the empirical level of textual analysis. A short introduction presents some main theories about poetic translation, from Benjamin and Steiner till more recent experts: Meschonnic, Apel, Berman and Mattioli. Following these thoughts, the works of two couples of poets and translators are analyzed. In the first case Philippe Jaccottet will be dealing with the entire Ungaretti's poetic production; in the second example, the translating intent of Vittorio Sereni on René Char's poems is described. While investigating the complex nature of poetic translation as a practice and an experience, this thesis in Compared Literature tries to present translation as an hermeneutic tool and a re-enunciating mechanism: its role in the dialectic movement of influences and literary evolutions is considered as essential. The original ethical vocation of translation is a constant background of the research.

Así estructurada nuestra judicatura, funcionó sin mayores tropiezos. Pero las exigencias de una nación en pleno desarrollo, la misma configuración geográfica de Chile, la distancia existente entre los centros urbanos y el número creciente de infracciones a la Ley detectables en puntos de la capital con la consiguiente necesidad de una pronta, oportuna y eficaz aplicación de la Ley.

A poesia de Cristovam Pavia integra-se em duas das mais importantes revistas literárias da segunda metade do séc. XX - a Távola Redonda e a Árvore - deixando transparecer, contudo uma certa continuação com o lirismo presencista, numa tradição de modernidade. O mundo da infância é o mundo mais próprio ao 35 Poemas, tornando-se, assim, o núcleo temático do volume. O perfeito equilíbrio entre a forma e os temas revela-nos uma poesia natural, simples e por isso notável e inconfundível. A dissolução do eu na unidade total é a mensagem final da obra que se apreende de uma poesia crepuscular de forte significado simbólico.

O presente estudo almeja delinear a maneira pela qual autor e leitor se entrelaçam ao longo da narrativa metaficcional de Serena, de Ian McEwan, podendo tornar-se um só em determinados momentos. Para tanto, far-se-á uma análise com base no conceito de dialogismo de Bakhtin e dos pressupostos teóricos de Foucault, Barthes, Booth, Iser, Jauss, Eco e Piglia, que elucidam os papéis de autor e leitor em relação ao texto. The present study aims to analyze the way in which author and reader relates with each other along the metafictional narrative Sweet Tooth, written by Ian McEwan, and could become one at certain times. To this aim, there will be an analysis based on the concept of dialogism of Bakhtin and the theoretical assumptions of Foucault, Barthes, Booth, Iser, Jauss, and Eco and Piglia, which elucidate the roles of author and reader in relation to the text. Thus, we intend to understand in the narrative the various reading levels which sometimes overlaps herein are parallel to each other by setting up an investigation in which the analysis of the relationship between author and reader can be extended to every human relationship: by analyzing each other every time, the other and your own self.

This urban-scale building consists of ideas, form, function, structure, materials, details and human consideration, etc. None of them can be set aside. Stimulated and guided by knowledge, I developed forms and structures to fit for materials and construction methods. This project can wake up memories in local residents' deep minds. Resumo: O uso de toxinas isoladas de venenos como ferramentas moleculares na compreensão de diversos eventos fisiológicos e patológicos tem sido comprovadas por vários trabalhos na literatura. A serpente Crotalus durissus cascavella é encontrada nas áreas de caatinga do nordeste do Brasil, desde o Maranhão até norte do estado de Minas Gerais, e apesar de pouco estudada sua picada constitui um importante problema da saúde pública (Martins et al, 1998). A agregação platequetária é bem caracterizada para a convulxina isolada do veneno de Crotalus durissus cascavella e Crotalus durissus terrificus. O objetivo principal do projeto foi avaliar a atividade de agregação plaquetária induzida por outras frações biológicas e farmacologicamente importantes do veneno total de Cotalus durissus cascavella, que neste projeto foram a crotoxina e giroxina. Através de uma combinação de varias metodologias em HPLC como exclusão molecular, troca iônica e de fase reversa conseguimos isolar os principais constituintes da crotoxina (PLA2, crotapotina e as proteínas ¿trombina-like¿) e a L-aminoácido oxidase a partir da giroxina. Durante o fracionamento do veneno total em coluna de HPLC de exclusão molecular foram detectados dois picos de atividade serino protease, um na fração giroxínica e outro na fração crotoxínica, sendo que na fração crotoxínica encontrou-se uma nova protease até então não caracterizada e na fração giroxínica foram monitoradas a atividade L-aminácido oxidase. Através da cromatografia em HPLC de troca iônica em DEAE 5PW obteve-se a fração Laminoácido oxidase cujo grau de homogeneidade molecular foi confirmado por HPLC de fase reversa. Da fração crotoxínica foram obtidos três grupos principais de proteínas (PLA2, crotapotinas e proteases) e da fração proteolítica foram isoladas três isoformas principais denominadas de F201, F202 e F203, sendo a fração F202, a fração majoritária. F202 foi obtida com maior homegeneidade molecular com massa molecular de 28kDa e mostrou uma alta quantidade de ácido aspártico, ácido glutâmico e outros aminoácidos importantes como histidina, cisteína e lisina. Esta proteína mostrou alta especificidade para BApNA e mostrou um comportamento Michaelis-Menten com Vmáx estimado em 5,64 µM/min e um Km de 0,58mM para este substrato. Neste trabalho foi investigada a habilidade desta proteína em degradar fibrinogênio e observou-se que F202 clivou ambas as cadeias a e ß. A atividade enzimática assim como a agregação plaquetária foi inibida fortemente com a incubação com TLCK, um inibidor específico para serinoproteases. O N-terminal da seqüência de aminoácidos de F202 mostrou alta homologia com outras proteínas ¿trombina-like¿, mas foi significantemente diferente da ¿trombina-like¿ isolada da fração giroxina. Crotalus durissus cascavella apresenta uma fração menos estudada denominada giroxina que tem sido descrita como uma proteína ¿trombina-like¿ como relatado por Raw et al. (1986) e Alexander et al. (1988). Neste trabalho foi demonstrado que a giroxina é uma fração heterogênea composta de uma ¿trombina-like¿ e proteína LAO, a qual parece estar envolvida em várias atividades
Abstract: The use of the isolated toxins from poisons as molecular tools in the understanding of diverse physiological and pathological events has been proved by some works in literature. The serpent Crotalus durissus cascavella is found in the areas of Caatinga Northeast of Brazil, since Maranhão until North of the state of Minas Gerais, and in spite of it hasn¿t been studied a lot, its bite constitutes an important problem to the public health (Martins et al, 1998). The platelet aggregation is well characterized to the isolated convulxin of the poison of Crotalus durissus cascavella and Crotalus durissus terrificus. The main objective of the project was to evaluate the activity of platelet aggregation induced by gyroxin and crotoxin that are biological and pharmacological important fractions of the total poison of Cotalus durissus cascavella. Through a combination of various methodologies in HPLC -as molecular exclusion, ionic exchange and the reverse phase- we managed to isolate the main constituent of the crotoxin (PLA2, crotapotin and the thrombin-like proteins) and the L-amino acid oxidase from the gyroxin. During the fragmentation of the total poison in column of HPLC of molecular exclusion two peaks of serine protease were found: one in the gyroxin fraction and another one in the crotoxin fraction. In the crotoxin fraction a new protease in not characterized yet and in the gyroxin fraction was found the L-amino acid activity oxidase. Through the chromatography in HPLC of ionic exchange in DEAE 5PW that allowed to the attainment of the fraction L-amino acid oxidase whose degree of molecular homogeneity was confirmed by HPLC of reverse phase. From the crotoxin fraction three main groups of proteins (PLA2, crotapotin and proteases) were isolated, and from the named proteolyitic fraction three isoforms of F201, F202 and F203, noticing that the F202 fraction is the major one. The fraction F202 showed a high quantity of aspartic acid, glutamic acid and others amino acids very important as histidine, cysteine and lysine and so more molecular homogeneity could be obtained and with molecular mass of 28kDa. This protein whose behavior Michaelis-Menten with Vmáx measured in 5,64 µM/min and one Km de 0,58 mM to this substratum showed high specificity to BapNA. In this work was investigated the ability of this protein in degrading the fibrinogen and was observed that the F202 made the cleavage into both chains a and ß. The enzymatic activity as well as the platelet aggregation were strongly inhibited with the incubation with TLCK, a specific inhibitor to serine protease. The N-terminal of the amino acid sequence of F202 showed the high homology with other proteins thrombin-like, but it was significantly different from thrombin-like isolated fro m the gyroxin fraction. Crotalus durissus cascavella presents a fraction less studied named gyroxin that has been described as a protein thrombin-like as related by Raw et al. (1986) and Alexander et al. (1988). In this work was demonstrated that the gyroxin is a composed heterogeneous fraction of one thrombin-like and protein LAO, and this seems to be involved in some activities

This thesis is a monograph on the poetry of Vittorio Sereni [1913-83] which treats all four of his principal collections: Frontiera [1941], Diano d'Algena [1947], Gli st11lmenti umani [1965] and Stella vanabi/e [1981]. It focuses on his redefinition of the lyric'!' and his establishment of new and highly original modes for its construction in poetic space. The later poetry (post-1960) is presented as the place in which this revision of the poetic subject comes to the fore, and I demonstrate how Sereni's textual landscape is co-ordinated in such a way as to communicate the extraneous qualities of a subject exiled to a space outside of the securer domain oflyric verse. Importandy, Sereni's subject is exiled linguistically as much as physically, and I advocate an interdisciplinary approach to interpreting his poetry which bridges psychoanalysis and linguistics. My study is set apart from existing scholarship in making the text the focus of analysis, which permits a greater degree of insight into the specifically poetic characteristics of Sereni's crisis of identity and its expression in language. Two main dimensions of Sereni's poetry are revisited and reassessed. The first is the role of liminality, which I present as a condition of writing and as a sign of the presence of a desiring subject whose most desired object is the complete poem or total identity which eludes him; the second is his debt to tradition and to a whole series of authors, including Dante, Petrarch, Leopardi and Montale, who mediate his contact with a textual beyond which slips further and further from view. Deixis is the syntactical matrix Sereni uses to communicate these facets of 'desire-space' and it emerges as the binding unit between one tradition and the next as well as what measures the distance between them. I argue that Sereni's unique brand of experimentalism develops from this point and that he represents a unique moment in the history of twentieth-century Italian poetry in the way in which he adapts preexisting models oflyric discourse to new modes of expression.

Esta pesquisa empirica discute pontos de tensoes emergidos no dialogo interetnico em torno das questoes de saude indigena, dando enfase ao tema da saude mental. As tensoes foram analisadas segundo diferentes pontos de vista sobre a promocao da saude indigena, identificadas a partir da producao de um mapeamento que expressa os conteudos discursivos marcados por um grande divisor: de um lado a perspectiva ocidental, composta pelo Estado, compreendido atraves das Politicas Publicas de Saude Indigena; do Sistema Unico de Saude e sua assistencia a saude; pelos Conselhos Federal e Regional de Psicologia e pela Organizacao Mundial da Saude. Do outro lado esta a perspectiva dos indigenas, especialmente do povo Mbya Guarani, analisados com apoio da literatura antropologica, a partir da nocao de Teko Pora (Bem Viver); dos noticiarios produzidos e veiculados por coletivos indigenas; do conteudo narrativo contido no documentario Teko Rexa - Saude Guarani Mbya e dos discursos presentes nos noticiarios produzidos por entidades indigenistas. As nocoes de saude e saude mental discutidas por Gadamer (2006), bem como as nocoes de saude e de ethos refletidas por de Figueiredo (2008), referencial teorico-metodologico assumido nesta dissertacao fundamentam sobre a importancia do territorio nos processos de saude/doenca para os povos indigenas, bem como auxiliam a refletir sobre as tensoes presentes em ambas as perspectivas. Os dados foram analisados a partir da nocao de multiplicacao dialogica (Guimaraes, 2013) no ambito do construtivismo semiotico-cultural em psicologia. A partir da identificacao de tensoes presentes, foram levantadas questoes a respeito de como estruturar o cuidado em psicologia, refletindo sobre as possibilidades de seu transito entre diferentes perspectivas em saudeThis empirical research discusses points of tension emerging in the interethnic dialogue around indigenous health issues, emphasizing the theme of Mental Health. The tensions were analyzed according to different points of view on the promotion of indigenous health, identified from the production of a mapping that expresses the discursive contents marked by a great divide: on the one hand the western perspective, composed by the State, understood through Policies Public Health Indigenous; of the Unified Health System and its health care; by the Federal and Regional Councils of Psychology; and the World Health Organization. On the other side is the perspective of the indigenous people, especially the Mbya Guarani people, analyzed with the support of anthropological literature, based on the notion of Teko Pora (Bem Viver); of the news produced and transmitted by indigenous collectives; of the narrative content contained in the documentary \"Teko Rexa - Saude Guarani Mbya\" and the speeches present in the news produced by indigenist entities. In 1936 Olga Franzoni, Caproni’s girlfriend, dies of septicaemia. In 1943 Sereni is taken prisoner, thus beginning a two-year period of captivity. The impact of these episodes on Caproni’s and Sereni’s poetry has been thoroughly analysed, and to conceive of them as watersheds in the individual path of either poet thus comes as natural. However, what critics have less organically delved into is the possibility of considering these episodes as pivotal in shaping a more definite turning point, representing the moment from which Caproni’s and Sereni’s poetry becomes, in many respects, ‘posthumous’. Building on Giulio Ferroni’s idea of ‘postumo’, this study seeks to propose a critical lens through which to examine the oeuvre of both Sereni and Caproni. Through recourse to categories such as ‘end’ and ‘after’, I will explore the thematic implications of the nexus between poetry and experience for both Sereni and Caproni.

As calicreínas teciduais humanas (KLKs) compreendem uma família de 15 enzimas serina proteases (KLKs 1-15) amplamente encontradas nos tecidos humanos. Em diversas patologias como a dermatite atópica, psoríase, síndrome de Netherton, câncer de ovário, mama e testículos, as KLKs encontram-se em concentrações elevadas. Por exemplo, as KLKs 5 e 7 estão mais abundantemente expressadas na pele, na qual estão envolvidas com o processo de descamação da mesma, e também presentes em alguns tipos de carcinomas. Dessa forma, as KLKs 5 e 7 são consideradas importantes alvos terapêuticos para o tratamento de doenças onde elas encontram-se superexpressadas, enfatizando a existência de somente um fármaco comercialmente disponível como inibidor de KLK. Nesse contexto, o trabalho descreve a síntese de 3 séries de compostos peptideomiméticos, incorporando o cerne estatina e diferentes resíduos de aminoácidos, planejados como candidatos a inibidores das enzimas serina proteases do KLKs 5 e 7. Os compostos finais foram obtidos utilizando uma rota sintética eficiente tendo como reação-chave a formação da ligação peptídica entre o cerne estatina e cloridratos de aminoésteres, previamente sintetizados. Os compostos sintetizados foram identificados por técnicas de Ressonância Magnética Nuclear, Infravermelho e Espectrometria de massas de alta resolução e os produtos finais serão avaliados em testes in vitro de inibição das enzimas KLKs
Human tissue kallikreins (KLKs) comprise a family of 15 serine protease enzymes (KLKs 1-15) widely found in human tissues. In several pathologies such as atopic dermatitis, psoriasis, Netherton syndrome, ovarian, breast and testis cancer, KLKs are in high concentrations. For example, KLKs 5 and 7 are more abundantly expressed in the skin, in which they are involved in the desquamation process, and also present in some types of carcinomas. Thus, KLKs 5 and 7 are considered important therapeutic targets for the treatment of diseases where they are over expressed, emphasizing the existence of only one commercially available drug as a KLK inhibitor. In this context, the work describes the synthesis of three series of peptideomimetic compounds incorporating the statin core and different amino acid residues, designed as candidates for inhibitors of the serine protease enzymes of KLKs 5 and 7. The final compounds were obtained using an efficient synthetic route based on the reaction of formation of the peptide bond between the statin core and previously synthesized amino acid hydrochlorides. The synthesized compounds were identified by Nuclear Magnetic Resonance, Infrared and High Resolution Mass Spectrometry techniques and the final products will be evaluated in in vitro inhibition assays of the KLKs enzymes

Establecer la relación entre migrante y migrancia de los espacios urbanos en la narrativa peruana contemporánea. Identifica las características del protagonista migrante en Pálido, pero sereno. Tipifica los sujetos de la tradición literaria vinculada con la migrancia a la cual pertenece la mencionada novela. Utiliza una metodología estructural-funcional, basada en la lectura crítica de la novela para determinar los aspectos narrativos en la construcción de los personajes y la representación de los espacios. De igual forma, esta perspectiva permite establece las líneas directrices para ubicar la novela de Carlos Eduardo Zavaleta en una tradición literaria específica como es la tradición migrante. Es una investigación teórica con una leve base historiográfica, que aplica la técnica descriptivo-analítica y realiza el fichaje de las fuentes de información para su análisis e interpretación.Tesis

L'architecture de rene sergent (1865-1927) est peu connue. Cet artiste a etudie dans un contexte de commandes privees puisqu'il a commence comme collaborateur de sanson. Sa formation et ses tendances en font le meilleur representant de l'ecole neo-classique historiciste, a clientele de particuliers fortunes. L'evocation de l'oeuvre de ses confreres plus celebres comme les destailleur, parent, sanson et mewes, permet de cerner les differentes sensibilites de ce courant ainsi que ses fondements ideologiques, financiers et artistiques (premiere partie). La production de l'architecte est constituee d'un nombre appreciable de chateaux, crees ou remanies, et d'une majorite d'hotels particuliers parisiens, son programme de predilection. Les immeubles de rapport et professionels traduisent dans une moindre mesure la meme sensibilite neo-classique. Le catalogue raisonne de l'ensemble de sa production (seconde partie) permet de caracteriser l'evolution de son style depuis un neo-louis xiv composite jusqu'au neo-louis xvi epure des annees 1920. Le style mis au point est egalement analyse (troisieme partie) d'un point de vue purement architectural. Une attention particuliere est portee aux modalites d'imitation des architectes du xviiie siecle et surtout de a. J. Gabriel, obsession de sergent. Plus largement, le principe de l'imitation est etudie. Sergent's work includes some castles, built or restored, and many parisians private hotels, what he prefered. A pesquisa tem como objetivo mapear os rastros de oralidade pertencentes a uma comarca oral nas narrativas de Visconde de Taunay (1843-1899) e Hélio Serejo (1912-2007). A comarca oral é pautada nos estudos de Carlos Pacheco (1992), no qual se constatam marcas de uma cultura oral na ficção latino-americana. A ideia pachequeana é de que a ficção produzida na América Latina é firmada na oralidade e justificada historicamente pela cultura oral que a constitui. A comarca oral se constitui pelas etiquetas, normas e condutas sociais transmitidas oralmente, definindo o papel do indivíduo em relação ao grupo social a que pertence. O modo de contar dos narradores em Taunay e Serejo apontam os contextos do lugar de fala de cada um, bem como a ficcionalização da oralidade de um lócus específico: a região de fronteira do Brasil com o Paraguai. Com o narrador taunayniano verificou-se uma profusão de discursos que apontam a movência do autor em trânsito, ora dentro, ora fora da comarca, fazendo alternância de pronomes, num jogo de alteridade que faz sobressair o olhar pelo diferente, pelo novo, via estranhamento. Já no caso do narrador serejiano, há um diferencial: certo peso de carga afetiva devido ao forte traço da memória de pertencimento, já que o narrador extrai da sua vivência as vozes outrora auscultadas, conduzindo o registro escrito (ZUMTHOR, 1993).The research aims to map the orality traces belonging to a comarca oral in Visconde de Taunay (1843-1899) and Hélio Serejo (1912-2007) narratives. La comarca oral is guided in Carlos Pacheco´s (1992) studies, in which exhibit marks of an oral culture in Latin American fiction. The pachequeana idea is that the fiction produced in Latin America is signed on orality and historically justified by the oral culture. La comarca oral is constituted by labels, norms and social behaviors transmitted orally, defining the role of the individual in relation to the social group to which he/she belongs. The narrator style of telling in Taunay and Serejo works point out the context of the place of speech of each one, as well as the fictionalization of orality of a specific locus: the border region between Brazil and Paraguay. With taunayniano narrator there was a profusion of speeches pointing to mouvence of the author in transit, sometimes inside, sometimes outside the region, making switching pronouns, an otherness game that brings out the look for different, the new, by estrangement. In the case of serejiano narrator, there is a difference: right weight emotional charge due to the strong trait of belonging memory, as the narrator draws the voices once auscultated from his experience, leading the written record (ZUMTHOR, 1993).

La gestión de riesgos por desastres que nuestro país presenta en la actualidad no ha logrado superar los enfoques reactivos. Las medidas de preparación, prevención y mitigación necesarias no se encuentran adecuadamente coordinadas y arraigadas en el quehacer de los habitantes y las instituciones. Esto compromete el desarrollo de los territorios, aumenta los niveles de vulnerabilidad de las personas y las edificaciones, y asimismo, la exposición de aquellos ante diversos tipos de amenazas.
La presente Actividad Formativa Equivalente reinterpreta los contenidos de un proyecto de cooperación técnica desarrollado por la Secretaría Regional de Planificación y Coordinación de la Región de Coquimbo, profundizando las variables de análisis necesarias en una gestión preventiva de riesgos por desastres en función de escenarios más específicos, tal como el centro histórico de la ciudad de La Serena, el cual requiere identificar apropiadamente el contexto edificado y las actividades sociales económicas que lo condicionan.

Resumo: A proliferação celular comanda os processos de embriogênese e de crescimento do organismo, sendo essencial para a correta função de vários tecidos adultos. Apesar de ser importante para a homeostase do organismo, a sua desregulação compõe a força motriz do desenvolvimento tumoral. Somente nos últimos vinte anos começou a ser evidenciada a relação entre as vias de tradução de sinais estimuladas por fatores de crescimento e a reorganização da atividade metabólica, a qual precisa priorizar a biossíntese e o aumento da biomassa, processos essenciais para a divisão celular. Em células tumorais, o consumo de glutamina é aumentando concomitante ao aumento da atividade de glutaminase. Três isoenzimas de glutaminase são expressas na maioria dos tecidos (liver-type glutaminase, kidney-type glutaminase e glutaminase C), todavia pouco se sabe sobre a necessidade específica de cada uma delas para o metabolismo tumoral. Vários artigos recentes têm definido o papel da glutaminólise, ou metabolismo da glutamina e seus subprodutos, na ativação da mTOR. Neste sentido é uma hipótese válida imaginar que mTOR possa contra-regular glutaminase. Desta maneira, resolvemos investigar se mTOR atua na regulação da atividade de glutaminase. Para tanto, realizamos knockdown estável de PTEN em células MDA-MB 231 e verificamos que não o mesmo afetou os níveis protéicos de GAC e KGA, assim como não houve mudança na localização subcelular das isoformas. Cinética enzimática da fração mitocondrial desta linhagem revelou que o knockdown de PTEN levou à uma diminuição do KM da enzima sem alteração de Vmax. De acordo, o tratamento com rapamicina, inibidor da mTOR, elevou o KM para os níveis detectados nas células controles. A atividade de glutaminase de lisado total de MDA-MB 231, NIH 3T3, IMR90 e BJ5TA foi afetada pelo tratamento com rapamicina conforme julgado por ensaios de dose e tempo resposta. Mais, ensaios de privação de glicose, glutamina e de fatores de crescimento levaram à inibição de mTOR e concomitante redução da atividade de glutaminase. Somado a isso, o knockdown estável de TSC2 em MDA-MB 231 e BJ5TA, assim como o knockout de TSC2 em MEF, promoveu superestimulação de mTOR e foi capaz de aumentar a atividade de glutaminase. Dosagem de atividade de glutaminase de células MDA-MB 231 com knockdown de GAC, KGA ou GAC/KGA tratadas com rapamicina indicaram que mTOR possa agir em ambas as isoformas. Curioso foi que apenas células shGAC e shGAC/KGA apresentaram redução da fosforilação de S6K em Thr389 indicando que GAC ou o metabolismo de glutamina via esta isoforma, possa contra-regular mTOR. Em adição, na comparação entre PC3 e DU145, verificamos que DU145 apresentou maior expressão de GAC, maior consumo de glutamina, maior dependência de glutamina em seu crescimento, maior sensibilidade ao inibidor de glutaminase, BPTES, e por fim, se mostrou mais responsiva à metformina, ativador indireto de AMPK. A ativação de AMPK por metformina, um conhecido sensor de estresse energético, mostrou diminuir a atividade de glutaminase em célula de tumor de próstata, DU145, indicando uma potencial ação de AMPK na atividade de glutaminase
Abstract: Cell proliferation is crucial for embryogenesis and organism growth, being also essential for the proper function of several adult tissues. Although important for the homeostasis of the organism, its deregulation composes the driving force of tumor development. In the past twenty years the relationship between the processes of signal translation stimulated by growth factors and the reorganization of metabolic activity has become more evident. Growing cells need to prioritize the biosynthesis and biomass increase, processes essential for cell division. In tumor cells, the glutamine consumption is increased concurrently with the increasing in the glutaminase activity. Three glutaminase isoenzymes are expressed in most tissues (liver- type glutaminase, kidney -type glutaminase and glutaminase C), but not much is known about the necessity of each isoform for the tumor metabolism. Several recent papers have defined the role of glutaminolysis or glutamine metabolism in mTOR activation. So it is a valid hypothesis to speculate that mTOR can counter-regulate glutaminase. Thus, we decided to investigate whether mTOR can control glutaminase activity. To this end, we have made MDA - MB 231 cells stably knocked down for PTEN and verified no alteration in KGA and GAC protein levels, as well as there was no change on their subcellular location. Enzyme kinetics of the MDA-MB 231 mitochondrial fraction revealed that PTEN knockdown led to a decrease in the KM of the enzyme without changing Vmax. Accordingly, the treatment with rapamycin (mTOR inhibitor), led to an increase in KM back to the level detected in control cells. The glutaminase activity of MDA - MB 231, NIH 3T3, IMR90 and BJ5TA total cellular lysates was also affected by rapamycin treatment in a dose- and time-response fashion. Moreover, glucose, glutamine and growth factors deprivation promoted mTOR inhibition and concomitant reduction on glutaminase activity. Glutaminase activity of MDA-MB 231 cells knocked down for GAC, KGA or GAC/KGA and treated with rapamycin indicated that mTOR can regulate both isoforms. Curiously, it was only on GAC or GAC/KGA knocked down cells that we observed a decrease in S6K Thr 389 phosphorylation, which could indicate that GAC or the GAC dependent-glutamine metabolism is a specific mTOR counter-regulator. Accordling, stable TSC2 knockdown in MDA-MB 231 and BJ5TA, as well as TCS2 knockout in MEF cells, promoted overstimulation of mTOR and increasing on glutaminase activity. Moreover, a comparison between PC3 and DU145 revealed that DU145 has higher GAC expression, greater consumption of glutamine, is more dependent on glutamine for its growth, more sensitive to the inhibitor of glutaminase, BPTES, and more responsive to metformin, an indirect AMPK activator. The activation of AMPK by metformin, a known energy stress sensor, led to a decreased glutaminase activity in the prostate tumor cell line DU145 indicating a potential role of AMPK on glutaminase activity

Serina endopeptidases de insetos, principalmente tripsinas e quimotripsinas, estão envolvidas na digestão inicial de proteínas. Genes codificadores para estas enzimas estão organizados em famílias multigênicas tendo expressão diferencial de acordo com a dieta do inseto, estando envolvidos no desenvolvimento de resistência a diferentes metabólitos secundários vegetais. Para uma melhor compreensão desta interação, fez-se necessário o isolamento destas enzimas para insetos de diferentes ordens, bem como a caracterização de suas especificidades por duas abordagens: (a) caracterização cinética dos subsítios componentes do sítio de ligação de tripsinas e quimotripsinas, utilizando diferentes substratos, modificadores químicos e inibidores e (b) estudos estruturais por modelagem molecular, clonagem, expressão e cristalização destas enzimas de insetos. Além disso, estudos evolutivos por análise de distância possibilitaram uma caracterização inicial da interação insetoplanta. Estas determinações permitiram verificar que tripsinas de insetos apresentam diferenças de especificidade tanto dentre as diferentes ordens de insetos quanto em relação às tripsinas de vertebrados, sendo que as tripsinas da ordem Lepidóptera apresentam troca de especificidade primária hidrolisando preferencialmente substratos P1 Lys. Foram também observadas diferenças de hidrofobicidade para os subsítios caracterizados sendo que estes apresentam hidrofobicidades crescentes segundo o grau de complexidade dos insetos na sua escala evolutiva. A troca de especificidade e o aumento da hidrofobicidade podem permitir a hidrólise dos inibidores vegetais protéicos. A análise das sequências de tripsinas de insetos por Neighbor Joining (NJ) compõe uma árvore de distâncias topologicamente semelhante à árvore de relações filogenéticas determinadas por morfologia. A sobreposição de estruturas pré -determinadas de tripsina complexada a diferentes inibidores permite a identificação de posições de interação enzima-inibidor que justificam a classificação em grupos distintos de enzimas sensíveis ou resistentes a presença de inibidores na dieta de insetos. Da mesma forma: a caracterização da especificidade das quimotripsinas de insetos permitiu a separação de grupos distintos de quimotripsinas. Estes grupos são sustentados pela substituição do resíduo 59 em insetos polífagos que alimentam-se de plantas que contêm cetonas naturais reativas. Estas caracterizações demonstram a importância de um estudo detalhado da especificidade de serina endopeptidases possibilitando o desenho de moléculas apropriadas para inibição destas e desenvolvimento de estratégias de controle de insetos.Insect serine endopeptidases, mairily trypsin and chymotrypsin are involved in initial protein digestion. Genes that encode these proteins are members of complex multigene families and are differentially expressed according to insects diet , thus being involved with resistance to plant metabolites. Purification of trypsins from different insect orders and chymotrypsins, as well as, characterization of their specificity are essential to a better understanding of this interaction. Characterization relied on two approaches: (a) kinetic characterization of the binding subsities of trypsins and chymotrypsins using different substrates, chemical modification and inhibition assays and (b) study of protein structure by molecular modelling and cloning, expression and crystallization of these enzymes. Besides that, evolutionary studies performed through distance analysis, permitted the investigation of plantinsect interaction. These characterizations showed that insect trypsins, in terms of specificity, are quite different from vertebrate trypsins and among insect orders. Lepidopterans trypsins have a distinct primary specificity, since they hydrolyses preferentially P1 Lys substrates, and present a crescent subsite hydrophobicity, which is directly correlated with the evolutionary scale. Both, the specificity exchange and the crescent hydrophobicity can allow the hydrolysis of vegetal proteic inhibitors. The analysis of trypsin sequences in Neighbor-Joining (NJ) algorithm yield a distance tree that is coherent with morphological phylogenetic relationships. The superposition of predicted structures of trypsins-inhibitors complexes permits to observe amino acid residues of interaction between enzyme-inhibitor, which support the distinction of different groups between sensitive and insensitive trypsins to the presence of inhibitors on insect diet. Similarly, characterization of insect chymotrypsins according to their specificity allowed us to classify these enzymes into different groups. These groups are supported by residue 59 replacements in polyphagous insects, which feed on plants bearing natural reactive ketones. These studies show the irnportance of a detailed study of serine endopeptidases, which may help in the development of better insect control strategies.

Ce mémoire s’articule en deux parties. En ce qui concerne la première, il s’agit de fournir une nouvelle transcription ponctuée du manuscrit BnF NAF 1943, unique témoin, datant de la fin du XIVe siècle, du Roman d’Eledus et Serene. Ce manuscrit n’a été édité qu’une seule fois, en 1923, dans une thèse de John Revell Reinhard comportant plusieurs lacunes et présentant un apparat critique restreint. La seconde partie consiste en une étude générique du Roman d’Eledus et Serene. Bien que ce texte figure traditionnellement dans les listes de romans idylliques médiévaux, aucune étude ne s’est encore employée à statuer sur la légitimité de ce classement. Le genre même du roman idyllique est peu connu, même au sein de la communauté médiéviste, et est encore mal défini. Une première définition, que les critiques s’accordent à qualifier de « dépassée », a été établie en 1913 par Myrrha Lot-Borodine dans son ouvrage Le roman idyllique au Moyen Âge. Depuis lors, bien que plusieurs altérations ponctuelles aient été apportées à cette définition d’origine, aucun travail critique ne s’est encore penché sur l’élaboration d’une nouvelle définition du genre. Ce mémoire s’emploie donc à compiler les caractéristiques génériques qui ont été associées au roman idyllique médiéval par la critique, en diachronie, pour en établir une nouvelle définition, aussi complète et opératoire que possible. Finalement, une analyse du Roman d’Eledus et Serene à la lumière de cette définition et en fonction d’autres témoins du corpus idyllique vise à légitimer son appartenance au genre.

L'activite scientifique de serenus d'antinoe se situe entre le ive et le vie siecle de notre ere. Ses oeuvres fondamentales, la section du cylindre et la section du cone presentent ses propositions sur la theorie des sections cylindriques et ses propositions sur la theorie des sections coniques au travers d'un plan qui passe par le sommet. La comparaison des propositions de serenus avec celles d'apollonios nous permettra de deduire l'appartenance de la premiere partie de la section du cylindre a la tradition d'etude des sections coniques. Serenus connaissait le livre 1 des coniques d'apollonios, quelques notions du livre vi et les formulations des propositions du livre v. Par l'examen minutieux des oeuvres de serenus, de pappus et d'eutocius nous constaterons l'existence de plusieurs propositions equivalentes du point de vue des enonces, des methodes de demonstration et du langage utilise, fait qui nous conduit a penser que serenus faisait partie de la tradition d'exegese des sections coniques et d'etude de la geometrie des solides, comme c'est le cas pour les deux autres auteurs. Nous verrons que la transmission de l'oeuvre de serenus etait dependante de l'importance accordee selon les epoques a l'enseignement scientifique de haut niveau. La lecture des ouvrages de serenus s'inscrit dans un programme de lectures propedeutiques a la lecture de l'almageste. C'est a ce titre que l'etude de la section du cylindre et celle de la section du cone viennent apres la lecture des traites de la "petite astronomie", comme nous pouvons le remarquer par l'etude du contenu des manuscritsdans lesquelles nous trouvons les traites de serenus. Les arabes ont developpe une theorie des sections cylindriques, en se basant au depart sur l'oeuvre de serenus mais ensuite ils ont developpe des propositions dans des domaines inexplores par les mathematiciens grecs. Ils ont travaille sur des transformations geometriques, dans les traditions apollonienne et archimedienne, en traitant des problemes souleves au cours des mesures des aires non rectilignes et de l'etude des coniques.
