Prompted by the investigative journalist Gitta Sereny's biography Albert Speer: His Battle with Truth, two recent productions, Esther Vilar's Speer and David Edgar's Albert Speer, have set out to explore the reputation of Hitler's architect and later Minister of Armaments and War Production, Albert Speer, the only leading Nazi to acknowledge his guilt at the Nuremberg Trials. The plays, like the biography, are concerned with the extent of Speer's knowledge of the ‘Final Solution’ during his career in the Nazi hierarchy, and consequently with the integrity of the stance he adopted at Nuremberg and thereafter – that is, of his claim of guilt by association and omission rather than by active participation. In her biography, Sereny claims that as a result of her association with Speer he eventually acknowledged his guilt to her, and was repentant. But Nick White believes that the evidence – much of it unearthed by Sereny herself – suggests otherwise, and that Sereny had failed to acknowledge that between 1978 and his death in 1981 Speer consistently deceived her about crucial aspects of this evidence. How successful are Vilar and Edgar in their quite different dramatic sifting, not only of the public persona of Speer, but also of the interpretation granted their subject by the biographer upon whom their plays, to a lesser and greater degree, depend? Nick White has taught at City University, London, and his PhD dissertation, ‘In the Absence of Memory? Jewish Fate and Dramatic Representation: the Production and Critical Reception of Holocaust Drama on the London Stage, 1945–1989’ (1998) has been followed by a companion volume of criticism, articles, and letters, The Critical Reception of Holocaust Drama on the British Stage, 1939–2000.

Mice that lack immunoglobulin A (IgA) were fully protected, suggesting that IgA may not be required for adaptive immunity in this model system.

ABSTRACT Several Shigella flexneri mutants with defects in aromatic amino acid and/or purine biosynthesis have been evaluated as vaccines in humans or in animal models. To be suitable as a vaccine, a mutant has to show virulence attenuation, minimal reactogenicity, and a good immunogenic potential in animal models. With this aim, we have constructed five S. flexneri 5 (wild-type strain M90T) mutants with inactivation of one or two of the loci purEK, purHD, and guaBA, governing early or late steps of purine biosynthesis. The mutants have been analyzed in vitro in cell cultures and in vivo in the Sereny test and in the murine pulmonary model of shigellosis. M90T guaBA, M90T guaBA purEK, M90T guaBA purHD, and M90T purHD purEK gave a negative result in the Sereny test. In contrast, in the murine pulmonary model all of the strains had the same 50% lethal dose as the wild type, except M90T guaBA purHD, which did not result in death of the animals. Nevertheless, bacterial counts in infected lungs, immunohistochemistry, and reverse transcription-PCR analysis of mRNAs for tumor necrosis factor alpha (TNF-α), gamma interferon (IFN-γ), interleukin-1β (IL-1β), IL-6, IL-12, and inducible nitric oxide synthase (iNOS) revealed significant differences among the strains. At 72 h postinfection, M90T guaBA purHD still induced proinflammatory cytokines and factors such as IL-1β, IL-6, TNF-α, and iNOS, along with cytokines such as IL-12 and IFN-γ. Moreover, in the absence of evident lesions in murine tissues, this mutant highly stimulated major histocompatibility complex class II expression, showing a significant ability to activate the innate immunity of the host.

ABSTRACT Construction of a stable Shigella sonnei vaccine has been complicated by the instability of the virulence phenotype caused by the spontaneous loss of the invasion plasmid. To select a suitable candidate for vaccine construction, 16 S. sonneistrains were screened for stability of the virulence phenotype. A stable strain, S. sonnei Mosely, was selected for further work. pΔvirG2, a deletion derivative of the virGgene in the sacB suicide vector pCVD442, was used to generate an S. sonnei virG deletion strain, WRSS1, which was invasive in HeLa cells but negative in the Sereny test. WRSS1 was found to be both immunogenic and protective in the guinea pig keratoconjunctivitis model.

O poeta italiano Vittorio Sereni (1913-1983) é ainda inédito e pouco conhecido no Brasil. A fim de introduzir o autor ao público brasileiro, este trabalho tem como objeto de estudo seu livro de estreia, Frontiera, de 1941. A partir de um panorama que percorre de maneira sucinta o surgimento dos poetas modernos da Itália, e que situa o leitor acerca da posição ocupada por Vittorio Sereni nesse cenário, esta pesquisa investiga os caminhos cursados pelo poeta em sua juventude, no início de sua produção literária. Tal percurso, conforme se demonstrará, foi determinante para sua completa formação humana, cultural e intelectual, abrangendo, por exemplo, as aulas do filósofo Antonio Banfi na Universidade de Milão, a experiência como redator da revista \"Corrente di vita giovanile\", e a admiração e amizade com os poetas Eugenio Montale, Giuseppe Ungaretti e Umberto Saba. Ademais, este trabalho discute o polêmico tema do pertencimento ou não de Sereni ao movimento hermético italiano dos anos 30/40. Finalmente, por meio de análises de quatro poemas do livro selecionado, \"Inverno\", \"Inverno a Luino\", \"Strada di Zenna\" e \"Ecco le voci cadono\", serão especificadas algumas linhas temáticas centrais de Frontiera recorrentes em outras composições de sua primeira publicação.
Italian poet Vittorio Sereni (1913-1983) is still unpublished and relatively unknown in Brazil. In order to introduce the author to the Brazilian public, this research studies his debut book, Frontiera, 1941. Through a brief overview of the history of the Italian modern poets, the present study investigates the path followed by Vittorio Sereni during his youth, at the beginning of his literary career. Such path, as will be shown later, was of the utmost importance for the poets humanitarian, cultural and intellectual development, and includes the philosophy lectures given by Antonio Banfi at the University of Milan, the time spent as a writer at the magazine \"Corrente di vita giovanile\" and the admiration of and friendship with fellow poets Eugenio Montale, Giuseppe Ungaretti and Umberto Saba. Moreover, this research addresses the controversial debate on whether Sereni integrated or not the Italian hermetic movement in the 1930s and 40s. Lastly, through the study of four poems from the selected book, \"Inverno\", \"Inverno a Luino\". \"Strada di Zenna\" e \"Ecco le voci cadono\", recurrent central themes in Frontiera will be presented and analyzed.

O Feijão Caupi (Vigna unguiculata (L.) Walp) é uma leguminosa com importante representatividade econômica e nutricional, especialmente no Brasil. Inibidores de serino proteases, como a tripsina, já foram descritos na espécie, assim como em outras plantas. No entanto, nesta espécie, ainda não foram identificados inibidores que apresentem atividade sobre a elastase de neutrófilos humana (HNE), protease envolvida em muitos processos patológicos, como na instalação e progressão da doença pulmonar obstrutiva crônica (DPOC). Nesse estudo, purificamos um inibidor a partir do extrato protéico de Vigna unguiculata que apresenta atividade sobre HNE. Inicialmente, foi realizado o processo de extração alcalina de proteínas, seguido de três passos cromatográficos distintos, utilizando as colunas Hitrap-Q (Troca-iônica), Source15RPC (Fase-Reversa) e ACE18 (Fase-Reversa). Essas etapas foram acompanhadas por testes de atividade inibitória, utilizando os substratos fluorogênicos Meo-Suc-Ala-Ala-Pro-Val-MCA (Elastase) e Z-Phe-Arg-MCA (Tripsina), além de ensaios da quantificação de concentração total de proteínas. Para determinar a massa do inibidor, foram utilizadas as técnicas de espectrometria de massa por MALDI-TOF e SDS-PAGE, o inibidor apresenta massa molecular de 10,99 KDa. O Ki para HNE foi determinado no valor de 9 pM. O inibidor não apresentou atividade inibitória sobre tripsina e trombina, porém foi observada atividade sobre subtilisina e quimotripsina. Estes dados indicam que o inibidor purificado trata-se de uma molécula ainda não caracterizada, devido às suas atividades inibitórias o nomeamos de Vigna unguiculata Elastase Inhibitor (VuEI).
The cowpea (Vigna unguiculata (L.) Walp) is a legume of important economic and nutritional representativeness, especially in Brazil. Serine protease inhibitors, such as trypsin, have been described in many species, as well as in other plants. In this specie an inhibitor with activity on human neutrophil elastase (HNE) has not yet been identified. This protease is involved in many pathological processes, such as the onset and progression of chronic obstructive pulmonary disease (COPD). We purified and characterized an inhibitor from the protein extract of Vigna unguiculata presenting activity towards HNE. Firstly, we performed the alkaline extraction procedure for proteins followed by three different chromatographic steps using Hitrap Q (ion exchange), Source15RPC (Reversed-Phase) and ACE18 (Reversed Phase) columns. These steps were followed by the inhibitory activity tests using fluorogenic substrates, MeO-Suc-Ala-Ala-Pro-Val-MCA (elastase) and Z-Phe-Arg-MCA (trypsin), and quantitation assays of protein concentration. To determinate the size of the molecule, we used MALDI-TOF mass spectrometry and SDS-PAGE. The molecular mass of the inhibitor was 10,99 kDa. The dissociation constant (Ki) toward HNE was 9 pM. HNE inhibitor showed no inhibitory activities toward trypsin and thrombin. However, the inhibitor presented activity toward subtilisin and chymotrypsin. These datas indicate that this molecule is a novel inhibitor to HNE and we named it Vigna unguiculata Elastase Inhibitor (VuEI).

La conception de la rhetorique de francis ponge peut paraitre contradictoire. Il y a en effet, une evolution significative du poete sur la question des "regles techniques" en matiere d'ecriture. Ponge preconise d'abord leur explicitation et engage meme une enquete sur les "questions rhetoriques" avec la revue "les cahiers du sud" puis il les declare inessentielles : les veritables regles relevant de l'indicible ne peuvent etre edictees. Pour comprendre ce changement de cap apparent, il faut examiner la position de ponge dans le paysage litteraire et plus largement dans le champ ideoloogique de l'apres-guerre. Son isolement le contraint, pour echapper a la fois au neo-academisme sous toutes ses formes et a l'histrionisme poetique toujours vivace, a inventer une nouvelle rhetorique paradoxale, "en acte", singulierement comprise dans le texte. A la fois art poetique et art de vivre, son lieu d'election se trouve chez lucrece, dans les "templa serena". On peut donc examiner la rhetorique pongienne "a l'oeuvre" et en faire emerger une figure inouie : l'indifference qui permet de concilier paradoxalement les antinomies et en particulier celles de l'analogie et de la difference, du meme et de l'autre. C'est ce travail de l'indifference qu'on peut lire a travers "les fictions rhetoriques" pongiennes qui "recitent" le noeud singulier des mots et des choses.

Memoria para optar al título de Arquitecto
El proyecto de Título consiste en el desarrollo de una propuesta de carácter cívico cultural en el Centro Histórico de la ciudad de La Serena, para revertir su obsolescencia generada por la expansión desmedida hacia la periferia. El polígono escogido forma parte de un conjunto de Patrimonio Industrial, donde funcionó la Ex Cervecería Floto-CCU, que actualmente se encuentra en estado de abandono, por lo que su condición no va de acuerdo a las dinámicas del sector ni con las proyecciones de crecimiento para el casco histórico, que además pertenece a la Zona Típica más extensa del país. Al contar con una gran superficie subutilizada, el terreno ofrece la oportunidad de poner en valor el patrimonio existente y a su vez dar cabida para la incorporación de nuevos programas que consoliden el carácter del lugar. Si bien el eje central del proyecto es la proposición del edificio consistorial dada por la actual dispersión de las dependencias municipales y la ausencia de un espacio representativo para la ciudadanía, también busca poner en valor el patrimonio existente, que hasta la fecha no se encuentra categorizado con interés histórico o patrimonial, por lo que su estado de vulnerabilidad es mucho mayor. Por medio de la intervención del conjunto, se otorga un plan de uso a los inmuebles a reutilizar, donde de acuerdo al análisis del área, se inserta un programa enfocado en el desarrollo de actividades cívicas y culturales, considerando, además, la relación entre los bordes de la manzana y su entorno. Hacia el norte y el oriente deberá enfrentarse principalmente con la actividad residencial dada por el Barrio patrimonial Almagro y su cercanía con la ribera sur del Río Elqui, mientras que hacia el sur y el poniente se encuentra vinculado con usos comerciales y la Plaza de Armas junto a edificios institucionales circundantes, donde el proyecto podrá conectarse a través de la apertura de sus bordes. Se propone una estrategia de recuperación a partir de una redefinición de las escalas del lugar, sus límites y relación con el tejido urbano, estableciendo diferentes etapas de desarrollo que permitan vincular de manera armónica el contexto urbano presente con los vestigios industriales del pasado, logrando una propuesta coherente con las nuevas necesidades de la ciudad y la región.

Esta dissertação constitui uma reflexão sobre a situação social, política e cultural da Inglaterra na década de 70, especialmente nos anos de 1972 e 1973, como descrita no romance Serena, de Ian McEwan. O objetivo principal desta pesquisa é apresentar como o momento histórico supracitado é narrado no discurso ficcional, a partir da perspectiva da personagem Serena, em comparação com o discurso histórico, de acordo com fontes selecionadas, que narram a história da Inglaterra no contexto da Guerra Fria, dos ataques terroristas do IRA e do funcionamento do Serviço Secreto doméstico inglês, chamado MI5.
This dissertation constitutes a reflection about the social, political and cultural situation of England in the 70s, especially in the years of 1972 and 1973, as described in the novel Sweet Tooth, by Ian McEwan. The main objective of this research is to present how the historical moment mentioned above is narrated in the fictional discourse, from the perspective of the character, in comparison with the historical discourse and from a selection of sources, which narrate the history of England in the context of the Cold War, the terrorist attacks of the IRA and the functioning of the English domestic Secret Service, called MI5.

Significant evidence suggests that serine/threonine-O-linked N-acetyl glucosamine0-(GlcNAc) modifications play a central role in the regulation of plant signaling networks. Forexample, mutations in SPINDLY,) SPY (an O-GlcNAc transferase,) OGT (promote gibberellin GA) (signal transduction and inhibit cytokinin responses. In addition, mutating both Arabidopsis OGTsSEC (and SPY) causes embryo lethality. The long-term goal of this research is to elucidate the mechanism by which Arabidopsis OGTs regulate signaling networks. This project investigated the mechanisms of O-GlcNAc regulation of cytokinin and gibberellin signaling, identified additional processes regulated by this modification and investigated the regulation of SEC activity. Although SPY is a nucleocytoplasmic protein, its site of action and targets were unknown. Severalstudies suggested that SPY acted in the nucleus where it modified nuclear components such as the DELLA proteins. Using chimeric GFP-SPY fused to a nuclear-export signal or to a nuclear-import signal, we showed that cytosolic, but not nuclear SPY, regulated cytokinin and GA signaling. We also obtained evidence suggesting that GA and SPY affect cytokinin signaling via a DELLA-independent pathway. Although SEC and SPY were believed to have overlapping functions, the role of SEC in cytokinin and GA signaling was unclear. The role of SEC in cytokinin and GA responses was investigated by partially suppressing SPY expression in secplants using a synthetic Spymicro RNA miR(SPY). The possible contribution of SEC to the regulation of GA and cytokinin signaling wastest by determining the resistance of the miR spy secplants to the GA biosynthesis inhibitor paclobutrazol and to cytokinin. We found that the transgenic plants were resistant to paclobutrazol and to cytokinin, butonlyata level similar to spy. Moreover, expressing SEC under the 35S promoter in spy mutant did not complement the spy mutation. Therefore, we believe that SEC does not act with SPY to regulate GA or cytokinin responses. The cellular targets of Spy are largely unknown. We identified the transcription factor TCP15 in a two-hybrid screen for SPY-interacting proteins and showed that both TCP15 and its closely homolog TCP14 were O-GlcNAc modified by bacterially-produced SEC. The significance of the interaction between SPY and these TCPs was examined by over-expressing the minwild-type and spy-4plants. Overexpression of TCP14 or TCP15 in wild-type background produced phenotypes typical of plants with increased cytokinin and reduced GA signaling. TCP14 overexpression phenotypes were strongly suppressed in the spy background, suggesting that TCP14 and TCP15 affect cytokinin and GA signaling and that SPY activates them. In agreement with this hypothesis, we created a tcp14tcp15 double mutant and found that it has defects similar to spyplants. In animals, O-GlcNAc modification is proposed to regulate the activity of the nuclear pore. Therefore, after discovering that SEC modified a nucleoporinNUP) (that also interacts with SPY, we performed genetic experiments exploring the relationship between NUPs and SPY nupspy double mutants exhibited phenotypes consistent with SPY and NUPs functioning in common processes and nupseeds were resistant to GA biosynthesis inhibitors. All eukaryotic OGTs have a TPR domain. Deletion studies with bacterially-expressed SEC demonstrated SEC'sTPR domain inhibits SEC enzymatic activity. Since the TPR domain interacts with other proteins, we propose that regulatory proteins regulate OGT activity by binding and modulating the inhibitory activity of the TPR domain.
