A study was carried out of the effect of a strong electrolyte (NaNO3) on the change in the electron spectra (in the visible region) of bromopyrogallol red in the presence of Septonex, sodium dodecyl sulfate and Triton X-100. It was confirmed that NaNO3 has a marked effect on the solubilization of the dissociated forms of the dye in Septonex, in contrast to solubilization by Triton X-100.

Abstract:
The effects of cationic tenside Septonex and the non-ionic tenside polyvinylpyrrolidone on the acidobasic and spectral properties of 3-pyridylfluorone have been studied. Data for the dependence A = f(pH*) were used to calculate protonization constants and molar absorption coefficients of the various acidobasic forms of 3-pyridylfluorone in solution with c(Septonex) = 5 mmol 1-1 or ρ(polyvinylpyrrolidone) = 1 g l-1 in 10% (v/v) methanol adjusted by sodium chloride to an ionic strength I 0.2. The results are compared with protonization constants of the reagent in solutions of the same ionic strength and methanol concentrations but without a surfactant. The effects of the tensides on the shape of absorption spectra of the various acidobasic forms of the reagent are also discussed.

Numerical analysis of absorbance data for the analytically significant UO22+ - Chromazurol B, UO22+ - Chromazurol S, and UO22+ - Eriochromcyanine R systems in the presence of the cationic surfactant SeptonexR revealed that also in micellar medium, complex equilibria establish and several complexes are formed in dependence on pH and concentration of uranyl ions, reagent, and surfactant. Based on the results, optimum conditions are found for the spectrophotometric determination of low concentration of uranium.

Various modified silica sorbents and unmodified silica were tested for the separation and preconcentration of 10-200 ng cm-3 from 50-1 000 cm3 sample volume of platinum group metals (PGMs), Pt(IV, II), Pd(II), Ir(IV), Rh(III), Os(VI), Ru(IV) and Au(III) in the form of ion associates of their chloro and bromo complexes with cationic surfactants. [1-(Ethoxycarbonyl)pentadecyl]trimethylammonium bromide (Septonex®), benzyl(dodecyl)dimethylammonium bromide (Sterinol®), trimethyl(tetradecyl)ammonium bromide or hexadecyl(trimethyl)ammonium chloride and 0.1 M HCl or 0.1 M HBr are suitable for the purpose. 100% recovery for Pd(II), Pt(IV) and Au(III) was obtained with octadecyl silicas Separon™ SGX C18, Separon™ SGX RPS and Phenyl™ silica in the presence of 0.006 M Septonex® or 0.003 M Sterinol®. The retained PGMs and Au(III) were eluted with acetonitrile and determined by inductively coupled plasma atomic emission spectrometry (ICP-AES) after evaporation of organic solvent in the presence of 0.1 M HCl. The enrichment factor on Separon™ SGX RPS reaches 100. A 1 000 : 1 excess of Cl-, NO3-, SO42-, Na+, K+, Ca2+, Mg2+, Fe3+ or Al3+ does not interfere with the preconcentration of PGMs and Au(III) on this sorbent. With the silica-based anion exchanger Separon™ SGX AX, the 100% recovery is observed for Ir(IV), Pd(II), Pt(IV) when the elution is performed with 5 M HCl or 5 M HNO3. Macroporous sorbents Amberlite™ XAD2 and Amberlite™ XAD4 are suitable for the retention of Au(III), Pd(II) and Pt(IV) from 0.1 M HCl and 0.006 M Septonex®. 100% recovery is achieved using acetonitrile as eluent but there is no preference of these sorbents to modified silica. In this way, the recovery of 0.1-0.2 μg cm-3 Au(III), Pd(II), Pt(IV) was also successfully tested with spiked matrices of soil extracts, power station dust samples and natural waters.

The combination of ion-pair extraction and differential pulse polarography is shown to be a method suitable for the determination of 10-7 mol l-1 concentrations of organic bases of quaternary ammonium compounds. Orange II (4-[2-hydroxy-1-naphtyl]azobenzenesulfonic acid) was found to be an appropriate polarographically active counter-ion. The proposed method was used for the determination of tetrapentylammonium bromide (as model compound), Septonex ([1-(ethoxycarbonyl)-pentadecyl]trimethylammonium bromide) and codeine.

The optical properties of bromocresol red, bromophenol blue and bromocresol green were studied spectrophotometrically in aqueous systems in the absence and in the presence of submicellar and micellar concentrations of cationic (Septonex, hexadecylpyridinium and hexadecyltrimethylammonium bromides), nonionic (Brij 35, Triton X-100) and anionic (sodium dodecylsulfate) tensides. The positions of the absorption maxima and molar absorptivities of the LH- and L2- acid-base species of the dyes depend on the kind and concentration of tenside, alkylpyridinium chain length, and kind and concentration of inert electrolyte.

Abstract:
The study of protein and surfactant interactions is of great significance in a number of applications, such as the cosmetic, food or pharmaceutical industries and many others. However, they require further study due to their compositional complexity and the limitations of current analytical approaches. In this thesis, the cationic surfactant septonex in combination with two differently charged proteins lysozyme and bovine serum albumin under different physiological conditions (temperature, surfactant concentration, environment and others) was selected to study the interactions. Characterization of protein-surfactant interactions is a very important but challenging task, therefore it is essential to use appropriate approaches to explore the nature of these interactions. In order to unify the information to provide rational models, calorimetric methods (DSC, ITC) and dynamic light scattering were used. Isothermal titration calorimetry monitors the evidence for the formation of the system of the mentioned substances and information on aggregation behavior, differential scanning calorimetry characterizes the thermal stability of proteins and dynamic light scattering made it possible to monitor changes in particle size. Both proteins have been proven to interact with positive septonex, even if the lysozyme molecule is also positively charged. However, significant differences were found between the two proteins. From the obtained results it is evident that the identical charge of the protein with the surfactant has an effect on the intensity of the measurement, although all measured interactions showed an endothermic character.

Abstract:
This thesis pursues monitoring of pH changes and description of properties in phase–separated hydrogels prepared by interaction of polyelectrolyte and oppositely charged surfactants. For the purposes of this work, all hydrogels were prepared by the interaction of hyaluronan with Septonex. All hydrogels were exposed to pH changes (4–9). Changes in pH were being monitored using spectroscopic indicators, methyl orange, methyl red, bromothymol blue and phenolphtalein. The pre– and post– pH changes in properties were compared using rheological methods. Additional information was gathered through thermogravimetric analysis. By these methods it was proved that the changes in the pH can modify the mechanical properties and partly the internal structure of the hydrogels. Rheological tests show, that from the application point of view, the most interesting hydrogels are observed at pH 9 which have the strongest bonds but have low permeability and hydrogels described at pH 7 which are much softer and are capable of absorbing large amounts of water.

Abstract:
This diploma thesis is focused on the study of physical and chemical properties of hyaluronan and cationic surfactant. As the cationic surfactant Septonex was used. The influence of the environment on the system, the effect of molecular weight of hyaluronan, and its concentration was studied. Then, the study of the influence and the effects of concentration of Septonex on the interaction of hyaluronan-surfactant followed. Different methods of measurement were chosen to characterize these ionokomplexes. The critical micelle concentration of the surfactant itself was measured, and then also with the addition of hyaluronan by spectrofluorimetry with fluorescent probe pyren. It was found, that hyaluronan forms gel with Septonex. On this basis, gels were prepared for three different molecular weights of hyaluronan – 300 kDa, 806 kDa and 1697 kDa. Gels were prepared in a ratio of hyaluronan – surfactant 1:1. In gels prepared in this way, the influence of environmental water and 0.15 M NaCl was studied and it was found that at 0.15 M NaCl clear gels are formed. Selected samples of the gels were then measured with oscillatory testing and the rheological behavior of gels of Septonex was studied. As the last method the turbidimetric measurement was chosen, which characterized the turbidity point in the gradual addition of Septonex to sodium hyaluronate solution. Again, the effect of the molecular weight of hyaluronan and its concentration in two environments - water and 0.15 M NaCl was evaluated. It was found that 0,15 M NaCl suppresses formation of turbidity and formation of precipitates.

Abstract:
The dissertation thesis deals with study of advanced steady-state and time-resolved fluorescence techniques, which can be used for study of micellar systems properties. Selected fluorescence techniques were used for characterization of Septonex and CTAB cationic micellar systems and theirs interactions with hyaluronan. Fluorescent probe pyrene was used for determination of critical micelle concentration (CMC) and micellar aggregation number of these surfactants. The changes of fluorescence behaviour of fluorescein and prodan were studied in wide concentration range of Septonex. Next chapter of thesis deals with study of Förster resonance energy transfer between perylene and fluorescein in Septonex and CTAB micellar solutions and the effect of hyaluronan addition to these systems. Also steady-state and time-resolved fluorescence anisotropy studies were used for research of the effect of hyaluronan addition to micellar solutions. The last chapter of this thesis is focused on photophysical behaviour of Prodan in different solutions (water, Septonex solutions below CMC, hyaluronan solution, Septonex micellar solution and Septonex micellar solution with hyaluronan), which was discussed on the basis of time-resolved emission spectra.

Abstract:
The diploma thesis deals with the preparation and characterization of catanionic complexes and their possible application to pharmaceutics and medicine. The catanionic complexes were prepared by mixing two oppositely charged surfactants with the concentration of 20 mmol·dm­3 in various volume ratios. Two systems were analyzed – CTAB + SDS and Septonex + SDS. Both systems were prepared in an aqueous and physiological environment. The turbidimetric analyses provided results of the intensity of turbidity of the prepared mixtures. The size and stability of the prepared particles were determined by DLS and ELS measurements. CTAB-rich samples showed significant viscosity changes and they were therefore characterized by rheological measurements. The results of this work show that the asymmetry of alkyl chains, surfactant selection, temperature, and ionic strength have a significant influence on the properties and self-assembly of surfactants in catanionic complexes.

Abstract:
The diploma thesis deals with resonance energy transfer in hydrogel matrix enviroment. In the theoretical part, all the important features of these systems and their components are summarized and all the principles for explaining the method used are described. The work also includes a short literary background research. In the experimental part, the resonance energy transfer was measured in micellar solutions and hydrogels in 0.15 M NaCl medium. The hydrogels were formed on the basis of interaction of the surfactant with the oppositely charged polyelectrolyte. As a positively charged surfactant, carbethopendecinium bromide (Septonex) was chosen and hyaluronan as the negatively charged polyelectrolyte. Perylene with fluorescein and perylene were selected as fluorescent pairs for RET. It has been found that increasing concentration of the acceptor molecule increases the intensity of RET, in other word the rate of acceptor binding to the donor. In the study of fluorescent pair of perylene with DiO was observed, that it was not appropriate to use fluorescence probe DiO at higher concentrations. Experiments were carried out to determine whether RET changes in the time from the point of mixing the donor and acceptor fluorescence probes together. This phenomenon has not been confirmed. In hydrogels, the effect of increasing surfactant concentration and molecular weight of hyaluronan was observed. The resulting analysis showed that the molecular weight of hyaluronan has a significant effect on RET efficiency in hydrogels.

This thesis is focused on diffusion of dyes in biopolymer-based hydrogels. These hydrogels are based on interaction between biopolymer-like electrolytes with oppositely charged surfactants. When polyelectrolytes interact with oppositely charged surfactants, micelle-like nano-containers can be formed. These nano-containers are able of binding hydrophobic compounds. In this study, combination of modified dextran (diethylaminoethyl dextran) with positive charge and oppositely charged sodium dodecylsulphate as surfactant was used for preparation of hydrogels. Next type of hydrogel was based on hyaluronan and positive charged surfactant Septonex (carbethoxypendecinium bromide). As a diffusion probes in hydrogels dyes Nile red and ATTO 488 were used. The diffusion of these dyes from aqeous solutions of NaCl or surfactants into hydrogels was monitored in time. Transport of dyes into structure of hydrogels was characterized by diffusion coefficients and structural parameters of hydrogels.

Abstract:
This diploma thesis deals with study of hydrogels formed by phase separation of hyaluronan with oppositely charged surfactants cetyltrimethylammonium bromide (CTAB) and Septonex. It follows the bachelor thesis and extends the knowledge about the detailed characterisation of the inner environment of the hydrogel by determining the diffusion behaviour of the fluorescent probes Atto 488 and Nile Red using fluorescence correlation spectroscopy (FCS) technique and its modified version dual-focus fluorescence correlation spectroscopy (2f-FCS). Compared results showed that both methods show similar values and probes specifically interact with CTAB but Atto 488 shows only weak interaction with Septonex compared to Nile Red. Additionally, these interactions were not affected by the molecular weight of hyaluronan. In conclusion, it was recommended to measure this type of hydrogel in a small depth of gel using a conventional method.

This thesis deals with the study of preparation and characterization of hyaluronan-micelle aggregates. The theoretical part deals with drug delivery systems, characterization of used materials and methods especially fluorescence spectroscopy, dynamic light scattering and turbidimetry. Methods of determination of measured data are summarized in the experimental section. The result section is divided into two subsections dedicated to different preparation methods of hyaluronan-surfactant complexes. Induced aggregates of hyaluronan with Septonex are characterized in terms of their origin and stability, and the results are compared with previously studied surfactants CTAB. In the second part are discussed so-called decorated micelles, their formation, properties and stability.
