Abstract:

Con sumo interés hemos leído el caso presentado por González Cruz et al., en el que se describe la presentación clínica del síndrome de enterocolitis inducida por proteínas alimentarias (SEIPA), así como su diagnóstico y tratamiento. Nos llamó la atención que los autores describan la prueba de parche como método de corroboración diagnóstica de esa enfermedad. La guía internacional más reciente no pudo realizar recomendación alguna respecto a su uso debido a los resultados contradictorios obtenidos en estudios clínicos. En el consenso español entre pediatras de atención primaria, gastroenterólogos pediatras y alergólogos pediatras no se recomienda la prueba de parche para la confirmación diagnóstica del SEIPA inducido por proteína de la leche de vaca, el desencadenante más común de esta entidad.

<p align="center"><strong>RESUMEN</strong></p><p>Las parroquias mencionadas en esta investigación son eminentemente agrícolas y han mostrado un crecimiento de su producción en los últimos años gracias al cultivo de tubérculos (yuca y malanga) destinados a la exportación, esta situación se complementa con la ejecución de proyectos de asfaltado de carreteras por parte del Gobierno Autónomo de Morona Santiago, por ello la investigación realizada se canalizó hacia la determinación del nivel de influencia de esta infraestructura vial sobre el crecimiento económico de las parroquias Sevilla Don Bosco y San Isidro entre los años 2011 y 2014, con el desarrollo de un modelo econométrico de crecimiento endógeno, que considera a la mano de obra y la inversión en insumos agrícolas como pilares de la producción y los kilómetros de vías asfaltadas, como resultado del análisis. La investigación determina, que la infraestructura la infraestructura vial Sevilla Seipa y Proaño San Isidro tuvo una influencia positiva en el crecimiento económico de las parroquias antes mencionadas, sin embargo, en contraste con la literatura económica analizada al respecto, no tuvo una influencia determinante debido a que la inversión privada fue fundamental para determinar sus posibilidades de producción y generar crecimiento económico.</p><p> </p><p><em> </em></p><p align="center"><strong>ABSTRACT</strong></p><p>The parishes mentioned in this research are predominantly agricultural and have shown economic growth recently based on in two products of exportation: cassava and malanga; this situation is complemented by the implementation of asphalt roads projects by the Morona Santiago´s Autonomous Government, so the research was oriented to determinate the influence level of road infrastructure over economic growth in the Sevilla Don Bosco and San Isidro´s parishes between 2011 and 2014, through the development of an econometric model of endogenous growth, which considers work force and investment in agricultural inputs as pillars of the production additionally paved road kilometers, as an enhancer factor of economic growth. The conclusion is about the road infrastructure Sevilla Seipa and Proaño San Isidro had a positive influence on the economic growth of these parishes, however, in contrast to the economic literature discussed, it didn´t have a great influence because the private investment was essential to determine the potential production and generates economic growth.</p>

Abstract:

BackgroundDifferent diagnostic interviews are used as reference standards for major depression classification in research. Semi-structured interviews involve clinical judgement, whereas fully structured interviews are completely scripted. The Mini International Neuropsychiatric Interview (MINI), a brief fully structured interview, is also sometimes used. It is not known whether interview method is associated with probability of major depression classification.AimsTo evaluate the association between interview method and odds of major depression classification, controlling for depressive symptom scores and participant characteristics.MethodData collected for an individual participant data meta-analysis of Patient Health Questionnaire-9 (PHQ-9) diagnostic accuracy were analysed and binomial generalised linear mixed models were fit.ResultsA total of 17 158 participants (2287 with major depression) from 57 primary studies were analysed. Among fully structured interviews, odds of major depression were higher for the MINI compared with the Composite International Diagnostic Interview (CIDI) (odds ratio (OR) = 2.10; 95% CI = 1.15–3.87). Compared with semi-structured interviews, fully structured interviews (MINI excluded) were non-significantly more likely to classify participants with low-level depressive symptoms (PHQ-9 scores ≤6) as having major depression (OR = 3.13; 95% CI = 0.98–10.00), similarly likely for moderate-level symptoms (PHQ-9 scores 7–15) (OR = 0.96; 95% CI = 0.56–1.66) and significantly less likely for high-level symptoms (PHQ-9 scores ≥16) (OR = 0.50; 95% CI = 0.26–0.97).ConclusionsThe MINI may identify more people as depressed than the CIDI, and semi-structured and fully structured interviews may not be interchangeable methods, but these results should be replicated.Declaration of interestDrs Jetté and Patten declare that they received a grant, outside the submitted work, from the Hotchkiss Brain Institute, which was jointly funded by the Institute and Pfizer. Abstract:

Here three new species of Seira from Madagascar are described and illustrated, all with Lepidocyrtus-like habitus, as short antennae, truncate unguiculus, and overall reduction of dorsal macrochaetae. The new species Seira lepidochaetosa sp. nov., Seira semicaerulea sp. nov. and Seira metafemurata sp. nov. can be distinguished from each other by color patterns and chaetotaxic elements. An identification key for these three species is also provided, including a discussion on truncate unguiculus on Seira species with dense and reduced macrochaetotaxy.

Abstract:

A análise de nutrientes na seiva vem destacando-se como ferramenta sensível no estudo do estado nutricional de plantas perenes, sendo útil também na avaliação da disponibilidade de nutrientes no solo para as plantas. Este experimento foi conduzido com o objetivo de avaliar a concentração de nutrientes na seiva de duas variedades de laranja, quando fornecidas cinco doses de N, P2O5 e K2O via fertirrigação. A seiva foi extraída mensalmente do ramo da brotação nova com éter etílico, durante duas safras consecutivas. Para compreender a dinâmica dos nutrientes na seiva, também foram feitas, simultaneamente, análises de folha e de solução do solo. Os valores de pH na seiva mantiveram-se constantes, em torno de 5,5, independentemente dos tratamentos. A análise da seiva mostrou-se sensível às variações da adubação, pois as concentrações de N e K na seiva aumentaram conforme as doses de nutrientes aplicadas e correlacionaram-se de forma positiva com os valores desses nutrientes na folha e na solução do solo. Alta concentração de K foi observada na seiva, em torno de 4,0 g L-1, sendo esta maior que todos os outros nutrientes analisados. Observou-se diferença significativa no teor de N-NO3 entre as duas variedades de copa estudadas: a variedade de copa Hamlin, de ciclo precoce, apresentou concentração na seiva cerca de 20% superior à variedade Valência, que é de ciclo tardio. Os resultados sugerem que a análise da seiva pode ser empregada como ferramenta auxiliar na avaliação do estado nutricional de plantas cítricas.

Abstract:

AbstractThe effect of Sepia 200c on the reproductive performance of Friesian cross dairy cows was assessed in a controlled trial from September 1989 until August 1990. Overall reproductive performance was monitored monthly and pregnancy diagnosis and cyclical status was determined. The cows were randomly allocated to 4 groups, untreated controls, placebo controls, Sepia-treated 24 to 48 hours postpartum and Sepia-teated day 14 postpartum.The trial was concluded in July 1990 when another treatment was employed to reduce declining herd reproductive status. Statistical analysis of the results was based on differences between the placebo control and the Sepia-treated, 24–48 hours postpartum, the pooled control groups and placebo control with Sepia day 14, and lastly the difference between the 2 Sepia-treated groups.The highest rates of periparturient disease occurred in the Sepia-treated groups, the highest single incidence rate of periparturient disease risk (cysts) found in the cows in the Sepia 24 to 48 hours postpartum group. Sepia day 14 produced the largest percentage of cows seen in first oestrus (45%), the largest number of observed heats (60%) in the cows without periparturient disease risk factors or recurrent traits and the lowest total mean value days to oestrus post-treatment.Sepia day 14 produced the largest percentage that held to first service, the lowest post-treatment period to first service, calving to conception interval, mean number of services and rectal examinations per conception, mean day calving interval and calving index. The results show that the effect of Sepia 200c administered to cows two weeks postpartum increased the numbers of cows observed in first oestrus, held to first service and lowered the calving to conception interval and calving index. The results of the Sepia day 14 postpartum group were also found to be compatible with the same group data reported in the previous study.

Abstract:

Variações diárias e sazonais do pH e composição mineral da seiva do xilema foram medidas em plantas de tomateiro com 40 dias, cultivadas em solução nutritiva e em casa de vegetação, visando avaliar os fatores que regulam sua variação. No primeiro experimento, a seiva foi coletada, sempre às 9:00 horas, em alguns meses do ano, em plantas da mesma idade. No segundo, a coleta da seiva ocorreu ao longo do dia, sempre na mesma planta. No terceiro, a seiva foi coletada em dois locais na planta, a 5 cm acima do colo e na base do pecíolo da 5ª folha. Na seiva, avaliaram-se o pH e as concentrações de Ca+2, K+, Mg+2, NH4+, NO3-, PO4-3 e SO4-. O pH variou de 5,3 a 6,4 no período experimental. pH acima de 6,0 ocorreu nos meses com maior temperatura média ambiente e o menor valor, 5,4, nos meses com menor temperatura ambiente. Potássio foi o cátion e nitrato o ânion encontrados em maior concentração na seiva.Considerando a variação diurna, uma maior concentração de nitrato, proporção cátions/ânions e pH ocorreram na seiva coletada à tarde. Considerando a variação sazonal, a maior concentração de nitrato e a proporção cátions/ânions ocorreram na seiva coletada nos meses com menor temperatura média e menores valores de pH. O pH da seiva exsudada do caule a 5 cm acima do colo foi menos ácido (pH 6,13) quando comparado ao pH da seiva exsudada no pecíolo da 5ª folha (pH 5,64). Os resultados sugerem que os fatores que regulam o pH da seiva influenciam diferentemente nas variações diurna e sazonal do pH.

Abstract:

Detailed and fully illustrated redescription of Seira pallidipes Reuter, 1895 is given based on specimens from the type locality. Examination of further specimens from the type locality of Seira pillichi Stach, 1930 has shown the identity of these two species justifying their synonymization: that Seira pillichi Stach, 1930 is a junior subjective synonym of Seira pallidipes Reuter, 1895 (syn. nov.). Variability in chaetotaxy is documented and discussed as well.

AbstractThis paper discusses the chronology of burial grounds containing specific Seima-Turbino type bronze weaponry (spears, knives, and celts). The “transcultural” Seima-Turbino phenomenon relates to a wide distribution of specific objects found within the sites of different Bronze Age cultures in Eurasia, not immediately related to each other. The majority of the Seima-Turbino objects represent occasional findings, and they are rarely recovered from burial grounds. Here, we present a new set of14C dates from cemeteries in western Siberia, including the key Asian site Rostovka, with the largest number of graves containing Seima-Turbino objects. Currently, the presented database is the most extensive for the Seima-Turbino complexes. The resulting radiocarbon (14C) chronology for the western Siberian sites (22nd–20th centuries cal BC) is older than the existing chronology based on typological analysis (16th–15th centuries BC) and some earlier14C dates for the Seima-Turbino sites in eastern Europe. Another important aspect of this work is14C dating of complexes within specific bronze objects—daggers with figured handles—which some researchers have related to the Seima-Turbino type objects. These items are mostly represented by occasional finds in Central Asia, however, in western Siberia these have been recovered from burials, too. The14C dating attributes these daggers to the end of the 3rd millennium cal BC, suggesting their similar timing to the Seima-Turbino objects. Further research into freshwater reservoir offsets in the region is essential for a more reliable reconstruction of the chronology of the Seima-Turbino phenomenon and the daggers with figured handles.

Com este trabalho objetivou-se analisar o comportamento do fluxo de seiva em espécies da floresta de transição Amazônia Cerrado e caracterizar a dependência do fluxo de seiva, em função do déficit de pressão de vapor da atmosfera (DPV). O fluxo de seiva foi medido utilizando-se sondas de dissipação térmica em 5 espécies diferentes. Os dados foram divididos em quatro estações. No período experimental as curvas de variação sazonal do fluxo de seiva evidenciam a ocorrência de picos no período úmido. Esta tendência sazonal do fluxo de seiva foi evidenciada pela relação entre valores diários de transpiração e do DPV. O valor limite do DPV nessas estações foi de 1 a 1,5 kPa. Com referência ao fluxo de seiva como representativo da taxa transpiratória das plantas, a redução do fluxo no período seco não confirma a hipótese da manutenção do processo de evapotranspiração. O fluxo de seiva nas espécies do estudo é influenciado mais pelas condições atmosféricas do que mesmo pela disponibilidade de água no solo, o que suscita um possível armazenamento da água no caule nos períodos mais secos quando a área foliar diminui mas a taxa transpiratória não é significativamente distinta daquela do período chuvoso.

Here we present a survey of Seira fauna from Iberian Peninsula and Canary Islands, including the description of three new species from Spain: Seira barrai sp. nov., Seira betica sp. nov. and Seira burgersi sp. nov., the latter with unprece-dented antennal morphology for Seira. Five species from Western Europe are redescribed and their dorsal detailed chaetotaxy is revealed for the first time: S. domestica (Nicolet), S. dinizi Gama, S. ferrarii Parona, S. mantis Zeppelini & Bellini and S. pini Jordana & Arbea, the first two from type locality, and the latter two based on analysis of the type material. A note on chaetal interpretation of the first abdominal segment for Seira is presented. An identification key to 31 species with known dorsal chaetotaxy from Western Palearctic region is also provided, including a discussion on dense macrochaetotaxy present in Palearctic and Oriental species and its reduction in species from others biogeographic regions.

AbstractThe results obtained from a study model using Sepia 200c in a herd of dairy cows led to an extended study. Overall reproductive performance was monitored monthly by a farm action list and the technique of palpation of the ovaries per rectum was used to determine pregnancy and cyclical status. A total of 101 cows were randomly treated with Sepia 200c on day 14 or 21 postpartum.Statistical analysis of the results was based on the differences between the untreated Control and Sepia-treated groups in periparturient disorders and pre- and post-service periods, and between the two Sepia-treated groups.In the pre-service period, a significant difference was found between the sepia-treated groups in the proportion of heifers calved, the number of assisted calvings and pre-service problems compared to Control. A difference of 9.9 total mean days to oestrus post-treatment was found between the Sepia-treated groups.During the post-service period, significant differences were found in the conception rate to first service, the percentage of cows in calf and total culled.A reduction (non significant) was found in the 21-day treatment group compared to control and 14-day treatment in the mean days calving to conception interval and the calving index. However, between the Sepia-treated groups a significant difference was found in total mean days calving to conception interval.The results of this study appear to demonstrate a difference in effect between the Sepia-treated groups and Control group. The study has been extended into a double blind placebo trial to find the effect of using a placebo and assess the use of Sepia given earlier postpartum on herd reproductive performance.

Seira boneti Denis, 1948, comb. nov. is examined and redescribed based on syntypes and by a newly discovered Chinese population. Lectotype and paralectotypes were designated, and the type locality of the species has been fixed to Câuda, near Nhatrang, Vietnam. The species was first described in the genus Lepidocyrtinus, but based on morphological and molecular evidence it is here transferred to Seira. For the phylogenetic placement of Seira boneticomb. nov., its mitogenome was included in a dataset comprising 19 species of Seirinae. Maximum Likelihood and Bayesian inferences clustered the species next to Seira sanloemensis Godeiro & Cipola, 2020 from Cambodia, forming a distinct Seira clade from the Old World, confirming the hypothesis of the existence of a different basal lineage of Seirinae in Southern Asia.

Infrared absorption spectroscopy is an effective tool for the detection and identification of molecules. However, its application is limited by the low infrared absorption cross-section of the molecule, resulting in low sensitivity and a poor signal-to-noise ratio. Surface-Enhanced Infrared Absorption (SEIRA) spectroscopy is a breakthrough technique that exploits the field-enhancing properties of periodic nanostructures to amplify the vibrational signals of trace molecules. The fascinating properties of SEIRA technology have aroused great interest, driving diverse sensing applications. In this review, we first discuss three ways for SEIRA performance optimization, including material selection, sensitivity enhancement, and bandwidth improvement. Subsequently, we discuss the potential applications of SEIRA technology in fields such as biomedicine and environmental monitoring. In recent years, we have ushered in a new era characterized by the Internet of Things, sensor networks, and wearable devices. These new demands spurred the pursuit of miniaturized and consolidated infrared spectroscopy systems and chips. In addition, the rise of machine learning has injected new vitality into SEIRA, bringing smart device design and data analysis to the foreground. The final section of this review explores the anticipated trajectory that SEIRA technology might take, highlighting future trends and possibilities.

Formins are a family of multidomain scaffold proteins involved in actin-dependent morphogenetic events. In Aspergillus nidulans, the formin SEPA participates in two actin-mediated processes, septum formation and polarized growth. In this study, we use a new null mutant to demonstrate that SEPA is required for the formation of actin rings at septation sites. In addition, we find that a functional SEPA::GFP fusion protein localizes simultaneously to septation sites and hyphal tips, and that SEPA colocalizes with actin at each site. Using live imaging, we show that SEPA localization at septation sites and hyphal tips is dynamic. Notably, at septation sites, SEPA forms a ring that constricts as the septum is deposited. Moreover, we demonstrate that actin filaments are required to maintain the proper localization pattern of SEPA, and that the amino-terminal half of SEPA is sufficient for localization at septation sites and hyphal tips. In contrast, only localization at septation sites is affected by loss of the sepH gene product. We propose that specific morphological cues activate common molecular pathways to direct SEPA localization to the appropriate morphogenetic site.

In the present study, Seira atlantica Negri, Pellecchia & Fanciulli, 2005 is recorded for the first time in Iran, up to date the species was known only from the type locality in Morocco. The specimens of S. atlantica were collected in alfalfa crops using insect net since the huge number of specimens were active underneath the skin of the newly emerged stems as well as in the crown of the plant. In plants holes created under the parenchyma of the stem surface were found, and subsequently small cavities were created under these holes. Therefore, it should be revealed if the species is introduced together with cultivars or is widely distributed in arid climate region. In the Middle-East eight Seira species have been registered, of which two distributed in Iran, Seira atlantica and Seira domestica Nicolet. Introduction.The Japanese floor-sitting Seiza is reflected of femoral artery pinched, which improved of tissues oxygenation and circulation. It might play role on body metabolism, also the nerves system.Methods: Study was conducted on a 20-year-old obese man performed routinely dzikr with Seiza sitting, so the time duration counted. Stomach circumference and body weight was monitoring twice weekly until 1 month.Results: The stomach circumference measurements of 114 cm observed improvement, as at week 1 (1.75%), week 2 (0.88%), week 3 (63%), and week 4 (3.51%) underwent 316 to 334 seconds weekly of Seiza sittings. A 100,4 kg of body weight showed gradually improvement: 0.55%, 0.35%, 0.25%, and 1.89% in each weeks.Discussion: The Seiza sitting might be relate to St36 Meridian points. It stimulated of parasympathetic system, digest and absorption, aliementary glands activated, also fat metabolism.Conclusions: The Seiza sitting during dzikr after prayer might improve of body metabolism.International Journal of Human and Health Sciences Vol. 07 No. 02 April’23 Page: 202-206

Background In Australia, the Socio-Economic Indexes for Areas (SEIFA), which includes the Index of Relative Socioeconomic Disadvantage (IRSD), captures the socioeconomic characteristics of areas. Because SEIFA rankings are relative to the country or state, the decile categorisations may not reflect an area’s socioeconomic standing relative to areas nearby. Aims The aim of the research was to explore whether IRSD rankings could be re-ranked to become locally sensitive. Data and methods Using existing SEIFA data to redistribute the membership of current decile IRSD groups, we tested three methods to re-rank all SA1 areas relative to the nearest areas capped at: (1) the nearest 99 neighbours, (2) a population threshold of 50,000 (3) a distance threshold of 10 km. Results The reclassification of SEIFA IRSD deciles was largest (up to 8 decile points of change) when comparing the nearest neighbour and population threshold local methods to current state-based rankings. Moreover, compared to using current national and state SEIFA IRSD rankings, the use of local rankings resulted in more evenly distributed deciles between cities, regional, and remote areas. Conclusions Because SEIFA IRSD rankings are used to allocate resources and health services, we encourage the combined use of a state and local ranking to refine areas considered the most disadvantaged.

Abstract B-cell epitope information is critical to immune therapy and vaccine design. Protein epitopes can be significantly affected by glycosylation, while no methods have considered this till now. Based on previous versions of Spatial Epitope Prediction of Protein Antigens (SEPPA), we here present an enhanced tool SEPPA 3.0, enabling glycoprotein antigens. Parameters were updated based on the latest and largest dataset. Then, additional micro-environmental features of glycosylation triangles and glycosylation-related amino acid indexes were added as important classifiers, coupled with final calibration based on neighboring antigenicity. Logistic regression model was retained as SEPPA 2.0. The AUC value of 0.794 was obtained through 10-fold cross-validation on internal validation. Independent testing on general protein antigens resulted in AUC of 0.740 with BA (balanced accuracy) of 0.657 as baseline of SEPPA 3.0. Most importantly, when tested on independent glycoprotein antigens only, SEPPA 3.0 gave an AUC of 0.749 and BA of 0.665, leading the top performance among peers. As the first server enabling accurate epitope prediction for glycoproteins, SEPPA 3.0 shows significant advantages over popular peers on both general protein and glycoprotein antigens. Steponu Seili raksturo dziļa un noturīga interese par latgaliešu grāmatniecību un neatlaidīga centība apkopot kultūras mantojumu. Savās mājās Makašānu pagasta „Kļovūs“ viņš izveidoja iespaidīgu latgaliešu rakstu bibliotēku ar teju pilnīgu grāmatu krājumu, plašu avīžu komplektu klāstu, kā arī seniem rokrakstiem un paša sakopotiem materiāliem par kultūras darbiniekiem. S. Seiļa arhīva materiālu daļa pirmoreiz nonāca Latvijas Nacionālajā bibliotēkā 1974. gadā no Latvijas PSR prokuratūras, kas iespieddarbus un rokrakstus bija izņēmusi no S. Seiļa mājām Rēzeknes rajona „Kļovūs“ kratīšanas laikā 1974. gada 1.–3. jūlijā. Pēc saņemto iespieddarbu un rokrakstu analīzes bibliotēka vienojās ar S. Seili par 54 rokrakstu glabājamo vienību paturēšanu tās kolekcijā un Reto grāmatu un rokrakstu nodaļā tika izveidots grāmatnieka personālarhīvs (RXA96), daļēji kompensējot īpašniekam rokrakstu vērtību ar samaksu par mikrofilmu izgatavošanu no viņam atpakaļ atdotajiem materiāliem. Personālarhīvā ietilpa Latgaliešu Skolotāju centrālās biedrības un citu kultūras organizāciju darbības materiāli, bet no paša S. Seiļa apcerējumiem un citu latgaliešu literātu manuskriptiem tika izgatavotas 16 vienību kopijas mikrofilmās. Ievērojot sabiedriski politisko situāciju, bibliotēkai bija jāpatur arī tie iespieddarbi, galvenokārt 20.–30. gadu periodiskie izdevumi, kas atbilda specfonda kritērijiem. Pārējie iespieddarbi un rokraksti, tai skaitā dienasgrāmatas, vēstules un pētījumi par personām, tika aizvesti uz „Kļoviem“ un nodoti to īpašniekam. Pārējos personālarhīva materiālus bibliotēka iegādājās 1980. gadā pēc S. Seiļa nāves no viņa mantiniekiem. Atstātajā garīgajā mantojumā ietilpa kā iespieddarbi, tā plašs rokrakstu arhīvs. Iespieddarbi, tai skaitā vairākas latīņu rakstu aizlieguma kontrafakcijas, atbilstoši bibliotēkas fondu organizācijas principiem tika iekļauti kopējā grāmatu krājumā. S. Seiļa personālarhīvam tika pievienotas vēl 254 rokrakstu vienības. Rokrakstu fonds papildinājās ar atsevišķiem izciliem drukas aizlieguma laika rokraksta grāmatniecības pieminekļiem, paša S. Seiļa un citu autoru jaunrades materiāliem, plašu korespondenci. Lasītājiem kļuva pieejama S. Seiļa uzkrātā informācija par latgaliskas izcelsmes ievērojamām personībām, kas apkopota „lietās“. Te var atrast ziņas vai par katru latgaliskas izcelsmes un latgaliešu kultūrā iesaistītu 20. gadsimta personību. Viņa vākumu pamatoti var uzskatīt par latgaliešu cilvēkarhīvu. 2000. gadā saskaņā ar līgumu bibliotēkas īpašumā pārgāja arī 13 mapes ar S. Seiļa dēla Pētera Seiļa deponētajiem materiāliem. These new records support that S. dowlingi has a wide distribution in Neotropical Region, and expanded to 34 the number of species of Seira Lubbock, 1870 found in Brazil. In addition, our revision excluded the record of Seira domestica (Nicolet, 1842) for Brazil.

Abstract:

Genetically engineered mouse models with altered oncogene or tumor suppressor gene activity have been utilized recently for carcinogen identification. The v -rasHa transgenic Tg.AC mouse, with its enhanced susceptibility to skin tumorigenesis, is thought to be well suited for examining the carcinogenicity of topically applied agents. Tg.AC mice were used to examine the carcinogenicity of SEPA 0009, a rationally designed organic molecule designed to enhance drug penetration through the skin. Fifty mg SEPA 0009/kg body weight, 1500 mg SEPA 0009/kg body weight, or the vehicle alone was applied daily to the skin of Tg.AC mice. Nontransgenic FVB/N mice were also treated with the vehicle alone or 1500 mg SEPA 0009. Daily application of a high-dose of SEPA 0009 caused severe and chronic irritation by 1 week that was maintained throughout the experiment. The irritation was accompanied by increased proliferation, increased apoptosis, and expression of the wound-associated keratin 6. High-dose SEPA 0009 induced squamous papillomas in Tg.AC, but not in nontransgenic mice, by 6 weeks. In mice treated with the high dose SEPA 0009, transgene expression was detected in papillomas at week 9, well after the onset of skin irritation and hyperplasia. In contrast, low-dose SEPA 0009 was not irritating to the skin and did not induce papillomas. Thus, SEPA 0009-induced tumorigenesis was associated with chronic and severe irritation. We propose that SEPA 0009-induced tumorigenesis in Tg.AC mice proceeds through an indirect mechanism that is secondary to cutaneous irritation.
