Full textThe development of superconcentrated aqueous electrolytes, namely Water-in-salt electrolytes (WiSE), from 2015 onwards has renewed the interest for aqueous-based Li-ion battery (LIB). Indeed, they were proposed to overcome issues related to safety and sustainability of common carbonate-based organic solvent while solving the poor performances of diluted aqueous electrolyte due to the narrow electrochemical stability window (ESW) of water (1.23 V). Such achievements are largely attributed to modification of the electrolyte structure upon increase in concentration that changes the physico-chemical properties and the interfacial reactivity. An inorganic LiF-based solid electrolyte interphase (SEI) was reported to be formed, opening the path for the use of low potential negative electrodes, further increasing the energy density of these batteries. This work aims to provide answers regarding the viability of WiSE in LIB. By conducting a systematic study of the impact of superconcentration on battery performances as function of the operating conditions, we demonstrate that the SEI is not able to prevent water reduction following the hydrogen evolution reaction (HER), neither during cycling nor during resting period, i.e. self-discharge. Indeed, the rates for water consumption calculated during cycling and resting period are found within the same order of magnitude, highlighting the SEI limitation to prevent water reduction although the surface is passivated. Determining the activation energies for HER during cycling and self-discharge, we suggest that self-discharge is more likely driven by water reduction than Li+ deintercalation. Eventually, LiF solubility measurements, gas chromatography tests and environmental scanning electron microscopy suggest that SEI instability is related to structural defects that cannot be self-passivated in WiSE. A presoaking step in organic electrolyte of an artificial Li/LiF layer reduces water consumption and thus confirms the need for the SEI to self-repair

Full textUnder de senaste decennierna har vindkraftsutbyggnaden ökat till havs, vilket innebär att det krävs tillförlitliga vindhastighetsuppskattningar över hav, särskilt i kustområden, där de flesta vindturbinerna kommer att placeras. Den här avhandlingen kommer att fokusera på Östersjön, vilket är ett relativt litet hav omgivet av landmassor, där land-hav-interaktion har en stor påverkan på vindmönstren över havet. Mesoskaliga modeller kan användas för att studera den lägre delen av den marina atmosfären. I den här avhandlingen används modellen "Weather Research and Forecasting" (WRF). I den första studien visar simuleringar att stabila förhållanden över havet är vanligt förekommande under sommar och vår, och sker i samband med en ökad förekomst av vindmaxima på låg höjd och ökad vindskjuvning under 200 m - alltså på höjder där vindkraftverk förekommer. Modellen, med flertalet gränskiktsparametriseringar, utvärderas för stabila fall mot flygplansmätningar, och resultaten visar att styrkan och höjden för vindmaxima är antingen över- eller underskattade oavsett parametrisering. I den andra studien undersöks advektiva fenomen. Simuleringar, flyplansobservationer och satellitbilder visar att avlånga rullvirvlar uppstår i det konvektiva gränsskiktet över land och advekteras flera tiotals kilometer över hav ut från kusten. Detta trots stabila förhållanden över havet, där den konvektiva turbulensen snabbt dör ut. Rullvirvlarna ger upphov till variationer i horisontell vindhastighet på flera meter per sekund över avstånd på några kilometer, vilket kan öka osäkerheten hos korttidsprognoser för vind när det är blåser från land. Sammanfattningsvis har det har visats att mesoskaliga processer i och ovanför det marina gränsskiktet har en stor inverkan på vindfältet åtminstone 100 km från kusten, och modeller behöver fortfarande förbättras för att kunna fånga dessa företeelser.

Full textThese recent years, Organic Field-Effect Transistor (OFET) development has significantly improved it performances and it stability. This was made possible, through a better understanding of the mechanisms governing charge transport in these devices. However, some phenomena remain unclear, in particular, at the interface between the semiconductor and the dielectric. Charge carrier trapping which is one of the main causes of charge transport disturbance in organic transistors, is one of them. So, this work aims to investigate such phenomena in pentacene-based transistors.Polar groups and particularly, hydroxyl groups, located at the insulator-semiconductor interface, are the main sources of charge carriers trapping in OFET. To prevent their presence, an OFET fabrication technology based on a passivating dielectric, poor of hydroxyl groups, calcium fluoride-based interfacial layer has been developed. Effect of this layer on pentacene-based transistors operation has been studied, as well as these devices aging under different storage atmosphere (in vacuum and in air) and under electrical stress.Thus, it has been highlighted that an interfacial layer of calcium fluoride with a too high thickness (around 5 nm) changes pentacene layer morphology which results in a quasi-disappearance of charge transport in pentacene in OFET configuration. Aging studies showed that under the effect of CaF2 interfacial layer, even with a very thin thickness (a few nanometers), a greater quantity of moisture is induced in pentacene layer probably due to the hygroscopic nature of calcium fluoride

Full textThis thesis is devoted to study the stabilization of some locally coupled systems. First, we study the stability of a one-dimensional coupled wave equations with two interior non smooth viscous dampings where we establish exponential stability. Second, we study the stabilization of a locally coupled wave equations with only one internal viscoelastic damping of Kelvin-Voigt type. Both the damping and the coupling coefficients are non smooth. Using a spectrum approach, we prove the non-uniform stability of the system. Next, using a frequency domain approach, combined with a piecewise multiplier technique and the construction of a new multiplier satisfying some ordinary differential equations, we show that the energy of the smooth solution of the system decays polynomially. Third, we investigate the energy decay of hyperbolic systems of wave-wave, wave-Euler Bernoulli beam and beam-beam types. Indeed, the two equations are coupled through boundary connection with only one localized non smooth fractional Kelvin Voigt damping. We establish a polynomial energy decay rate. Finally, we study the stability of a multidimensional system of two wave equations coupled by velocities with only one localized non-smooth Kelvin-Voigt damping. By using a spectral analysis, we prove the non uniform stability of the system. Further, using a frequency domain approach combined with a multiplier technique, we establish some polynomial stability results by considering different geometric conditions on the coupling and the damping domains. In addition, in the absence of any geometric condition, we establish two polynomial energy decay rates of the system on a square domain

Full textIn order to determine the impact of the trifluoromethyl group on the physico-chemical and biological properties of fluorinated peptides, we have decided to synthesize peptides incorporating α-trifluoromethyl amino-acids.Each enantiomer of α-Tfm-Alanine was prepared in an enantiopure form and in a large scale. The trifluoromethyle group placed in the α position deactivates its amine function so that its coupling needs harsh activation conditions to be achieved with mixed anhydride.The determination of the hydrophobicity of fluorinated peptides with an analytical method based on the measurement of indexes derived from retention times by RP-HPLC showed the dramatic influence of the trifluorométhyl group on the increase of the hydrophobicity.Fluorinated peptides/peptides interactions were studied for the inhibition of the aggregation of Aβ42 in patients suffering from Alzheimer's disease. The first results indicate a reduction of Aβ42 aggregation by a fluorinated peptide.Pepsine digestion of a fluorinated tetrapeptide showed a dramatic reduction of the rate of its cleavage in comparision with the peptide incorporating an alanine residu. The use of the sensitive 3-FABS 19F NMR detection method showed the recognition and the cleavage of a fluorinated peptide by trypsin which definites it as a substrate for trypsin

Full textThis PhD dissertation represents a first empirical assessment of the implications of the interaction between banking competition and securitization on efficiency, risk, and capital structure of US commercial banks United States. Precisely, the main objective of this PhD dissertation is to answer the following questions: 1) What is the impact of competition and securitization, separately and jointly, on bank efficiency? 2) Does securitization reduce bank risk under competitive pressure? 3) What are the implications of the interaction between competition and securitization on bank capital structure? The development of these questions is motivated by the rapid growth of securitization, the evolution of bank competition through various US bank regulations, the ongoing debate between proponents and criticizers of competition and securitization; and the novelty of these topics among academics and practitioners. After implementing a propensity score matching technique, the first chapter suggests that securitization significantly increases bank efficiency as measured by the Stochastic Frontier Approach (SFA). This positive impact appears to be more pronounced in competitive banking markets. In the second chapter, we use fixed-effects model through which securitization is found to have a negative impact on bank risk, particularly and surprisingly during the recent financial crisis, but only if banks highly securitize loans in competitive markets. The third chapter argues that banking competition could be considered a channel that explains how securitization significantly influences US commercial banks’ capital structure. Specifically, deeper investigations, introduced by the quantile regression, show that less-capitalized banks that highly securitize loans in competitive markets are more likely to increase their overall capital ratios, whereas a decrease in these ratios is reported for highly-capitalized banks. Providing several empirical and operational contributions as well as important managerial implications and policy recommendations, this PhD dissertation emphasizes the importance of considering a careful banking regulation on securitization that takes into account the heterogeneous responses of bank capitalization, the different reactions of various securitized loans, and the overall significant effect of US banking market structure

Full textIn 1979, the decision of the g5 mandated that each country worked toward a balance of foreign exchange. This implies that each is committed to a process of disinflation in order to improve the competitiveness and profitability of their firms. The governments adopt this strategy owing to the increasing disiquilibriums in both employment and prices that occurred during the seventies. Since then, the governments have led restrictive policies so as to curb inflation and restore competitiveness. If the disinflation is drastic, then gains of competitiveness noise exports and the shrinking of internal demand reduces imports. Mechanically, the balance of trade improves but does not put an end to the negative impact of output and employment. Moreover, the efficiency of this strategy restrains considerably within a fixed exchange rate zone, and all the more if everyone seeks gains of competitiveness through the fall of purchasing power. Additionally, when faced to asymmetrical shocks, the monetary policy in europe is far from being optimal. This is confirmed by the dogmatism that led the economic policy in the early mineties ; it reinforced the recession and induced a rise in unemployment in europe. The management of the unification of germany and the absence of coordination between economic policies led politicians to a resignation to unemployment and a proscription of social programs to reduce it. However, the level of unemployment indicates notably that economic activity is lower than what it should be. Eventually, the basic purpose of economic policies is to increase the production of wealth (i. E. Full employment) while monitoring a fair income share-out : finance must not threaten social cohesion

Full textThe objective of this thesis is to develop a new concept that consists to permanently encapsulate cosmetic active ingredients into mesoporous silica. The encapsulation of these active molecules improves their skin tolerance, optimizes their light stability while preserving their effectiveness and facilitates their formulation in cosmetic products. Thus, UV filters have been encapsulated in MCM-41 type mesoporous silica by using in-situ route because it seemed the most efficient route to achieve permanently encapsulation of large quantities of active. Two UV filters have been studied. One is lipophilic (Parsol MCX) and the other is a hydrophilic (Parsol HS). For all the active molecules studied, an appropriate encapsulation rate was obtained and the characterizations, particularly these performed by solid-state NMR, have shown that the encapsulation is effective within the pores which, coupled with the good stability of encapsulation in the case of hydrophilic actives, ensures minimal contact between the active and the consumer's skin. In contrast, a significant release of lipophilic UV filter was found. It was established that the physico-chemical properties of the synthesized materials differ significantly according to the lipophilic or hydrophilic nature of the encapsulated molecules. In all cases, the porous organization, the structural order and the morphology of the particles containing active ingredients vary significantly compared to the reference MCM-41 type mesoporous silica, synthesized without active ingredient. Thus, the presence of the active ingredient in the reaction medium has an influence on the structure and the texture of the synthesized materials, which is due to interactions between the actives molecules, the silicate species and the surfactant molecules in the reaction medium

Full textOur research has an empiric and theoretical origin; the empirical origin emerges from an exploratory study conducted in multinational organizations in Mexico. The interviews made to the managers participating in this study, make evident an interesting investigation concern: the existence of a stability demand to the application of organizational change. We therefore decided to explore this issue of investigation in another field which seams fertile to us in order to investigate it: We chose the small and medium size companies in Mexico. The theoretical origin of our dissertation is found in a revision of literature made regarding the concepts of change and stability in the organizations. This revision of literature allows us to affirm that the majority of research study changes and stability in separate manner, without studying its cohabitation. This revision of literature guides us to concentrate in the technological change. We can summarize this problem (empirical and theoretical) in the following question: Is there a demand for stability expressed by the managers when a technological change is applied? Why? To reply to these questions we divided our research into two phases. One in an extensive manner, in which we designed and applied a questioner, and another one more profound which implies the conduction of interviews directed to the middle managers and CEOs of small and medium size firms in Mexico. The results of our investigation are of a theoretical and empirical order. In the theoretical level, our questionnaires make evident three dimensions of our construct " stability demand " expressed by the managers. The components of this three dimensions are combined in different fashions according to the type of technological change implemented (we analyzed three) these results make us doubt the postulate of unidimensionality of the concepts of change and stability in organizations. In the empirical level our interviews make evident that during the conduction of a technological change, the managers play a double role: guarantor of stability and change agents. Full textThis thesis studies the moduli space of principal G_2-bundles over a smooth connected projective curve, where G_2 is the exceptional Lie group of smallest rank. The group G_2 is first introduced as the group of automorphisms of the complex algebra of the Cayley numbers. Other equivalent definitions are also proposed. We study the reductions and extensions that a principal G_2_bundle can admit, as well as the link between a principal G_2-bundle and its associated vector bundle in relation to the notion of (semi)stability. The moduli space of semistable principal G_2-bundles is analysed. We notably obtain a characterisation of its smooth locus, with an explicit decomposition of its singular locus into three connected componants. We also give an analysis of the Verlinde space of G_2 at level 1

Full textNous avons récemment découvert et étudié une nouvelle classe d'agents antimicrotubules appelés phényl 4-(2-oxo-3-alkylimidazolidin-1-yl)benzènesulfonates (PAIB-SOs) hautement actifs et sélectifs pour les cellules cancéreuses du sein. Leur mécanisme de sélectivité est basé sur la métabolisation des PAIB-SOs par le CYP1A1 en agents antimitotiques puissants appelés phényl 4-(2-oxo-3-imidazolidin-1-yl)benzènesulfonates (PIB-SOs). L'objectif principal de ma recherche était d'évaluer la période nécessaire à l’induction d’une activité cytotoxique significative de quelques-uns de ces PAIB-SOs prometteurs sur les cellules cancéreuses du sein. Dans un second volet, nous désirions étudier la stabilité métabolique et la cinétique de bioactivation par le CYP1A1 de 8 PAIB-SOs phare en vue d’en sélectionner 4 pour des essais sur des animaux. Premièrement, nos études ont montré que l'activité antiproliférative des PAIB-SOs est concentration et temps dépendante. Un temps de contact des PAIB-SOs étudiés avec les cellules cancéreuses du sein variant de 24 à 36 h est nécessaire pour observer une activité antiproliférative significative. Deuxièmement, nous avons confirmé que tous les PAIB-SOs évalués sont rapidement biotransformés en présence de CYP1A1 en PIB-SOs. Troisièmement, les expériences de stabilité hépatique des PAIB-SOs incubés avec des microsomes soit humains, de souris ou de rats ont montré que leur demi-vie et leur clairance intrinsèque varient selon la structure du PAIB-SO et l’espèce animale étudiée. La stabilité des PAIB-SOs incubés avec des microsomes de souris est plus faible que lorsqu’incubé avec des microsomes humains ou de rats. De plus, la stabilité métabolique de ces PAIB-SOs avec les microsomes humains ou de rats est similaire. Le criblage de notre chimiothèque a permis d’identifier les CEU-829, -938, -934 et -913 comme étant les PAIB-SOs métaboliquement les plus stables et générant les plus basses concentrations de PIB-SOs avec des demi-vies respectives de 22, 55, 31 et 41 minutes dans les microsomes humains, de 43, 52, 23 et 44 minutes dans les microsomes de rat et de 3,7, 20, 12 et 1,6 minutes dans les microsomes de souris. Nos études ont également mis en évidence les CEU-934 et -938 comme des molécules candidates prometteuses pour des études de pharmacocinétique et de pharmacodynamie chez la souris et les CEU-829 et -913 chez les rats.
We recently found and studied a new class of antimicrotubule agents named phenyl 4-(2-oxo-3- alkylimidazolidin-1-yl)benzenesulfonates (PAIB-SOs) that are highly active and selective to breast cancer cells. PAIB-SOs are the first antimicrotuble CYP1A1-dependent prodrugs and their mechanism of selectivity is based on the metabolization in human breast cancer cells of PAIB-SOs by CYP1A1 into potent antimitotics named phenyl 4-(2-oxoimidazolidin-1-yl)benzenesulfonates (PIB-SOs). The main objective of my research was to evaluate the period necessary to trigger an efficient antiproliferative activity on breast cancer cells and to study the metabolic stability and the kinetics of activation by CYP1A1 of 8 promising PAIB-SOs exhibiting high antiproliferative activity and selectivity toward breast cancer cells aiming to select 4 PAIB-SOs for animal studies. We first found that the antiproliferative activities of PAIB-SOs are concentration and timedependent. A contact time of the selected PAIB-SOs with breast cancer cells varying from 24 to 36 h is required to observe a significant antiproliferative activity. We also found that all PAIBSOs are rapidly metabolized in the presence of CYP1A1 into PIB-SOs. Finally, the hepatic stability experiments of PAIB-SOs using human, mouse or rat microsomes showed that the halflife and the intrinsic clearance depend on the structure of the PAIB-SO and the animal species studied. The stability of PAIB-SOs in mouse microsomes was weaker than in rat or human microsomes which were equivalent. Our screening program identified CEU-829, -938, -934 and - 913 as our most stable PAIB-SOs and generating the lowest quantity of PIB-SOs when incubated with human and rodent microsomes. In addition, they respectively exhibited half-lives of 22, 55, 31 and 41 minutes in human microsomes, 43, 52, 23 and 44 minutes in rat microsomes and 3.7, 20, 12 and 1.6 minutes in mouse microsomes. Altogether, our studies identified CEU-934 and - 938 as suitable candidates for further pharmacokinetic and pharmacodynamic evaluation in a mouse model and CEU-829 and -913 in a rat model.

Full textThe present dissertation is focused on the Baum-Connes conjecture for quantum groups. The main purpose of this work is the study of the Baum-Connes conjecture stability under some constructions of discrete quantum groups. In a first phase, we carry out a detailed and extensive study about the categorical reformulation of the Baum-Connes conjecture according to the results of R. Meyer and R. Nest. Next, we apply these techniques to the specific case of torsion-free discrete quantum groups. We carry out an exhaustive study of crossed products in order to handle them comfortably in connexion with the Baum-Connes conjecture. Notably, we give a proof of the universal property satisfied by a reduced crossed product by a discrete quantum group. We analyze as well some important properties for this dissertation. Let us mention in particular the associativity property of the crossed product with respect to a semi-direct product. Being inspired by the pionneer work of J. Chabert, we perform a generalization for discrete quantum groups of the invariance property of the Baum-Connes conjecture under the semi-direct product construction. Two permanence properties of own interest are studied as well. Namely, the torsion-freeness and the K-amenability. We observe that the torsion-freeness assumption forces a compact bicrossed product to be a torsion-free quantum semi-direct product, so that the corresponding Baum-Connes conjecture does not give any relevant information in this case. The general strategy used to accomplish such a generalization consists in defining a “decomposition” functor between the corresponding Kasparov categories in accordance with the semi-direct product operation. Thus, we observe that this strategy can be extrapolate to other (quantum) group constructions. Namely, to a quantum direct product. In this case, we state a connexion with the Künneth formula as pointed out by J. Chabert, S. Echterhoff and H. Oyono-Oyono for classical locally compact groups. The properties of torsion-frenness and K-amenability are also analyzed. It is known, thanks to R. Vergnioux and C. Voigt, that the strong Baum-Connes conjecture is preserved by divisible discrete quantum subgroups. The same is true for the strong torsion-freeness property, thanks to Y. Arano and K. De Commer. Here we show that both the usual Baum-Connes conjecture and the usual torsion-freeness property are preserved by divisible discrete quantum subgroups. The K-amenability property is analyzed too. A notably permanence property included in this dissertation is the invariance of the strong Baum-Connes conjecture under the free wreath product construction. For this, we carry out a complete classification of torsion actions of a quantum free product, which allows to give an appropriated formulation of the strong Baum-Connes conjecture for a free wreath product inspired by the pioneer work of C. Voigt. A major application is an explicit K-theory computation, in three relevant situations, for the Lemeux-Tarrago's compact quantum group which is monoidally equivalent to a free wreath product. Both this stability property for a free wreath product and the K-theory computations are part of a collaboration work with A. Freslon. To conclude, we question ourselves about the results obtained in order to suggest a list of questions, problems and goals that the author has encountered during the whole research period of the present dissertation and that are part of his future research projects

Full textBlack liquors are presently combusted in recovery boilers where the inorganic cooking chemicals are recovered and the energy in the organic material is converted to steam and electricity. A new technology, developed by Chemrec AB, is black liquor gasification (BLG). BLG has more to offer compared to the recovery boiler process, in terms of on-site generation of electric power, liquid fuel and process chemicals. A prerequisite for both optimization of existing processes and the commercialization of BLG is better understanding of the physical and chemical processes involved including interactions with the refractory lining. The chemistry in the BLG process is very complex and to minimize extensive and expensive time-consuming studies otherwise required accurate and reliable model descriptions are needed for a full understanding of most chemical and physical processes as well as for up-scaling of the new BLG processes. However, by using these calculated model results in practice, the errors in the state of the art thermochemical data have to be considered. An extensive literature review was therefore performed to update the data needed for unary, binary and higher order systems. The results from the review reviled that there is a significant range of uncertainty for several condensed phases and a few gas species. This resulted in experimental re-determinations of the binary phase diagrams sodium carbonate-sodium sulfide (Na2CO3-Na2S) and sodium sulfate-sodium sulfide (Na2SO4-Na2S) using High Temperature Microscopy (HTM), High Temperature X-ray Diffraction (HT-XRD) and Differential Thermal Analysis (DTA). For the Na2CO3-Na2S system, measurements were carried out in dry inert atmosphere at temperatures from 25 to 1200 °C. To examine the influence of pure CO2 atmosphere on the melting behavior, HTM experiments in the same temperature interval were made. The results include re-determination of liquidus curves, in the Na2CO3 rich area, melting points of the pure components as well as determination of the extent of the solid solution, Na2CO3(ss), area. The thermal stability of Na2SO3 was studied and the binary phase diagram Na2SO4-Na2S was re-determined. The results indicate that Na2SO3 can exist for a short time up to 750 °C, before it melts. It was also proved that a solid/solid transformation, not reported earlier, occurs at 675 ± 10 °C. At around 700 °C, Na2SO3 gradually breaks down within a few hours, to finally form the solid phases Na2SO4 and Na2S. From HTM measurements a metastable phase diagram including Na2SO3, as well as an equilibrium phase diagram have been constructed for the binary system Na2SO4-Na2S. Improved data on Na2S was experimentally obtained by using solid-state EMF measurements. The equilibrium constant for Na2S(s) was determined to be log Kf(Na2S(s)) (± 0.05) = 216.28 – 4750(T/K)–1 – 28.28878 ln (T/K). Gibbs energy of formation for Na2S(s) was obtained as ΔfG°(Na2S(s))/(kJ mol–1) (± 1.0) = 90.9 – 4.1407(T/K) + 0.5415849(T/K) ln (T/K). The standard enthalpy of formation of Na2S(s) was evaluated to be ΔfH°(Na2S(s), 298.15 K)/(kJ mol–1) (± 1.0) = – 369.0. The standard entropy was evaluated to be S°(Na2S(s), 298.15 K)/(J mol–1 K–1) (± 2.0) = 97.0. Analyses of used refractory material from the Chemrec gasifier were also performed in order to elucidate the stability of the refractory lining. Scanning electron microscopy (SEM) analysis revealed that the chemical attack was limited to 250-300 μm, of the surface directly exposed to the gasification atmosphere and the smelt. From XRD analysis it was found that the phases in this surface layer of the refractory were dominated by sodiumaluminosilicates, mainly Na1.55Al1.55Si0.45O4.

Full textThis thesis is devoted to the study of the stabilization of some distributed systems with dynamic boundary control. First, we consider the stabilization of the Rayleigh beam equation with only one dynamic boundary control moment or force. We show that the system is not uniformly (exponentially) stable. However, using a spectral method, we establish the optimal polynomial decay rate of the energy of the system. Next, we study the indirect stability of the wave equation with a fractional dynamic boundary control. We show that the decay rate of the energy depends on the nature of the geometry of the domain. Using a frequency approach and a spectral method, we show the non exponential stability of the system and we establish, different polynomial stability results. Finally, we consider the finite difference space discretization of the 1-d wave equation with dynamic boundary control. First, using a spectral approach, we show that the polynomial decay of the discretized energy is not uniform with respect to the mesh size, as the energy of the continuous system. Next, we introduce a viscosity term and we establish the uniform (with respect to the mesh size) polynomial energy decay of our discrete scheme

Full textThe Candida albicans genome displays a high tolerance to rearrangements, notably loss-ofheterozygosity (LOH) events. These events most often result from the repair of DNA double-strandbreaks (DSBs) and are known to play an important role in different aspects of C. albicans biology.To study the molecular mechanisms leading to LOH, we have combined an I-SceI meganuclease-dependent DSB-inducing system with a FACS-optimized reporter system of LOH. Our results show that expression of I-SceI leads to a dramatic increase in the frequency of LOH events,mainly gene conversion events. Characterization of cells having undergone a LOH led us to identify recessive lethal and deleterious alleles present in the heterozygous state in the genome of the C.albicans laboratory strain. These alleles influence the nature of the LOH events that are observed following a DSB. We also characterized the fidelity of the repair following an I-SceI-induced DNA DSB. This revealed unexpected complex recombination events occurring upon both break-induced replication and gene conversion with crossover repair events. In parallel, a collection of 564 over expression plasmids for genes involved in signaling pathways, genome integrity and cell wall integrity has been transformed in a C. albicans strainharboring the I-SceI-dependent DSB-inducing system and FACS-optimized LOH reporter. Analyses of the resulting transformants under conditions that allowed fro I-SceI expression or not, led to the identification of genes whose overexpression results either in an increase of the basal LOH rate or in areduction of the high LOH frequency associated to I-SceI expression
