Abstract:
Labu siam (Sechium edule) merupakan jenis tanaman sayuran yang dapat dimanfaatkan untuk berbagai jenis makanan, labu siam (Sechium edule) bukan tanaman asli Indonesia, melainkan berasal dari Thailand. Labu siam (Sechium edule) memiliki kandungan gizi yang cukup lengkap salah satunya flavonoid yang memiliki sifat hipoglikemik. Selain dapat diolah sebagai sayuran, labu siam (Sechium edule) juga dapat diolah menjadi biskuit. Penelitian ini telah dilakukan di Laboratorium Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Tadulako. Jenis penelitian ini adalah penelitian eksperimental laboratoris dengan menggunakan uji statistik uji t berpasangan didasarkan pada analisis kadar glukosa darah tikus (rattus norvegicus) dalam 3 waktu intervensi (perlakuan). Hasil analisis uji laboratorium kandungan: analisis kadar glukosa darah tikus (Rattus norvegicus) diperoleh rerata kadar glukosa sebelum intervensi (108,80 mg/dL), rerata kadar glukosa hari ke-7 intervensi (105,60 mg/dL), dan rerata kadar glukosa hari ke-14 intervensi (96,40 mg/dL). Perbedaan kadar glukosa dalam tiga waktu perlakuan: sebelum dengan setelah hari ke-7 (p value = 0,016), sebelum dengan setelah hari ke-14 intervensi (p value = 0,002) dan setelah intervensi hari ke 7 dengan setelah intervensi hari ke-14 (p value = 0,007). Penelitian ini menyarankan kepada masyarakat agar lebih memanfaatkan buah labu siam (Sechium edule) karena kandungan-kandungan gizi yang terdapat pada buah labu siam (Sechium edule) sangat bermanfaat bagi kesehatan, khususnya bagi penderita diabetes melitus.

Abstract:
Sechium edule shows a wide biological diversity. Hybrids and varieties of Sechium edule have been created, producing fruits of different shapes, sizes, colors, tastes, and textures. These hybrids and varieties have been used as food and in traditional medicine. In this research, the antiproliferative activity of the Madre Negra™ genotype of Sechium edule var. amarus silvestrys on the MCF7 breast cancer cell line was assessed. Different extracts in hexane, methanol, and dichloromethane were obtained to perform a bio-guided study. The dichloromethane extract showed the largest significant inhibitory activity (p < 0.05). This extract was separated into 92 fractions, which were reduced to eight fractions by chromogenic identity. Of the eight fractions, two recorded significant antiproliferative activity (p < 0.05) on MCF7, an even higher activity than the total extract. The active metabolites were identified as flavonoids, tannins, and terpenes, of which cucurbitacins I, B, D, and E stood out. The present paper can be considered as preliminary results of our research work.

Penelitian bertujuan untuk mengetahui umur simpan biskuit dari campuran tepung labu siam (Sechium edule) dan tepung kacang hijau (Vigna radiata). Sampel yang digunakan dalam penelitian ini adalah biskuit yang terbuat dari tepung labu siam (Sechium edule) dan kacang hijau (Vignaradiata). Penelitian ini dilaksanakan pada bulan Juni-September 2020 yang diawali dengan pembuatan biskuit dari formulasi tepung labu siam dan kacang hijau. Analisis data untuk pendugaan umur simpan biskuit menggunakan metode Arrhenius. Hasil penelitian menggunakan metode ASLT. Berdasarkan hasil yang diperoleh, umur simpan biskuit dari campuran tepung labu siam (Sechium edule) dan formulasi kacang hijau (Vignaradiata) paling tinggi diperoleh pada tepung labu siam 100% pada suhu 25 oC selama 177 hari. Kacang hijau 100% tertinggi diperoleh pada suhu 25 oC selama 176 hari. Tepung labu siam 50% dan kacang hijau 50% tertinggi diperoleh pada suhu 25 oC selama 187 hari. Tepung labu siam 75% tertinggi dan kacang hijau 25% diperoleh pada suhu 25 oC selama 170 hari. Tepung labu siam 25% dan kacang hijau 75% tertinggi diperoleh pada suhu 45oC selama 195 hari.

Labu Siam (Sechium edule) mengandung flavonoid yang diketahui memiliki aktivitas hipotensi. Tujuan penelitian ini adalah untuk mengetahui adanya pengaruh pemberian ekstrak buah labu siam sebagai antihipertensi. Metode penelitian ini adalah eksperimen semu (quasy experiment) dengan desain equivalent time-sample design yang dilakukan dengan cara memberikan perlakuan dan diukur tekanan darah pre-test dan post-test. Subjek penelitian diberikan ekstrak labu siam dengan dosis 100 mL/hari selama 7 hari. Hasil uji statistik menunjukkan terdapat perbedaan nilai tekanan darah sistolik dengan nilai signifikan p=0,001 dan nilai tekanan darah diastolik dengan nilai signifikan p=0,009 (α=0,05). Sehingga dapat disimpulkan bahwa labu siam (Sechium edule) dapat menurunkan nilai tekanan darah sistolik dan diastolik pada pasien hipertensi.

Abstract:
A novel species of powdery mildew, Neoerysiphe sechii sp. nov., has been proposed based on host, morphological characteristics, and gene sequences. The new species was found on chayote (Sechium edule) and a wild species, chayotillo (S. mexicanum), in Veracruz, Mexico. This novel species was identified as a member of Neoerysiphe, based on the morphological characteristics of the anamorph: catenescent conidia, lobed appressoria, and Striatoidium-type conidia. No teleomorph was observed. Neoerysiphe sechii is distinguished by foot cells that are frequently cylindrical with a slight widening at the base, a feature not observed previously in other species in the genus. An Erysiphales-specific primer was designed for the 5′ end of the ITS region of the nuclear rDNA. Independent and concatenated phylogenetic analyses with our rDNA sequences and sequences available in GenBank for Neoerysiphe spp. confirmed that the powdery mildew specimens found in Sechium spp. form an independent and strongly supported lineage. This is the first report of a Neoerysiphe species causing powdery mildew on members of Cucurbitaceae, and it is the second species reported from Mexico after N. cumminsiana. Neoerysiphe sechii is found on cultivated and endemic hosts in Mexico, suggesting that the fungus itself may be endemic.

In this research, the drying kinetics and equilibrium conditions during osmotic dehydration of chayote (Sechium edule, Sw.) were studied using 20, 30 and 40 % (w/v) sucrose solutions. A thermostatic bath was built to allow the experimental evaluation of osmotic dehydration kinetics and equilibrium moisture content determination. The experiments were carried out at three different temperatures (30 ?C, 40 ?C and 50 ?C) in order to evaluate the influence of temperature and sucrose concentration. Chayote fruits were washed and submerged in sodium hypochloride solution for ten minutes, for sanitization. Afterwards, the fruits were dried and cut into slices of about 0.5 cm thickness. Then, the slices were put into hypertonic sucrose solutions during preestablished time intervals. After osmotic dehydration, the solution was discarded and the slices were softly dried with absorbent paper to remove surface moisture. The moisture content was determined gravimetrically. Long time experiments up to 96 h were performed to provide information about the equilibrium moisture content. An specific equilibrium isotherm was selected by statistical methods. The correlation that better described equilibrium moisture content behavior was the Smith sorption isotherm, adapted to osmotic environment. The Overhults model was used to describe osmotic dehydration kinetics and estimate the drying constants k and n. Based on the results obtained in this research, it could be demonstrated that the equilibrium moisture content is strongly influenced by sucrose concentration and that k is a parameter dependent on the temperature and sucrose concentration, whereas n may be considered constant for the given experimental setup.

Excess fat accumulation in the liver increases the risk to develope progressive liver injuries, the lighter will cause fatty liver, the serious one will also cause steatohepatitis, liver cirrhosis even emergence of hepatocarcinoma. Fatty liver may have implication abount metabolic syndrome, such as obesity, hypertension and diabetes. Therefore, develop the pharmacological or natural agents to reduce fat accumulation in the liver and has already become an important thing in research for prevent or treat more severe forms of fatty liver disease. The Sechium edule shoots has already been verified and contained abundant polyphenol, and polyphenol has been known to decrease serum lipid, cholesterol and prevent atherosclerosis. However, the effect of Sechium edule shoots on modulating hepatic lipid metabolism are unclear. This study was to investigate the effects and mechanisms of Sechium edule shoots polyphenol extracts (SPE) in reducing hepatic lipid accumulation, and used oliec acid and high fat diet (HFD) to induce fat accumulation of HepG2 and an animal experimental model respectively. We found that the Sechium edule shoots water extracts (SWE) could regulate serum and hepatic lipid contents (e.g. triglyceride and cholesterol) and reduce the rate of body weight in HFD-induced rat. Furthermore, SWE and SPE through the AMPK (AMP-activaing protein kinase) signaling pathway could decrease numerous lipogenic relative enzymes, such as FAS, HMG-CoA reductase, SREBPs and increase the expression of CPT-I and PPARα proteins, which are critical regulators of hepatic lipid metabolism. These observations suggested that Sechium edule shoots have potentiality in developing the health food for preventing and remedying fatty liver.

RIPs are a group of plant enzymes that can inhibit polypeptide-chain elongation by inactivating the ribosomes. The molecular mechanism of the inhibition is identified as a specific N-glycosidase activity that hydrolytically cleaves the N-glycosidic bond of adenine at position 4324 (A4324) of 28S rRNA. RIPs are divided into two groups: (1) type I RIPs, single-chain polypeptide with molecular mass ranging from 25 to 30 kDa; (2) type II RIPs, heterodimer proteins consisting of an A-chain with enzymatic activity equivalent to the type I RIPs and a lectin-like B-chain. By binding to the D-galactose of the cell-membrane receptors, the B-chain of type II RIPs facilitates the A-chain to enter the cytoplasm and irreversibly inactivate ribosomes. A ribosome-inactivating protein (RIP), sechiumin, was purified from the seeds of edible gourd, Sechium edule Swartz, with an apparent relative molecular mass of 27 kDa. It inhibits the protein synthesis of rabbit reticulocyte lysate strongly with an IC50 of 0.8 nM, but has a much lower inhibitory effect on that of intact HeLa cells, with an IC50 of 5500 nM. Sechiumin has a highly specific RNA N-glycosidase activity towards 28S rRNA, as the A-chain of abrin does. It suggests that sechiumin is one of the type I RIPs The cDNA of sechiumin has been cloned and expressed by pET expression system in Escherichia coli. The sechiumin cDNA has 951 nucleotides, encoding a protein with 285 amino acids. The amino acid sequence deduced from the cDNA reveals that the first 21 N-terminal amino acid residues constitute a signal peptide and the 19 C-terminal amino acid residues were proposed to be removed by the post-translational modification as found in other type I RIPs, trichosanthin. Sechiumin has nearly 60 % homology to several type I RIPs purified from Cucurbitaceae family. Two amino acid residues, Glu160 and Arg163, found at the putative active site of sechiumin are known to be catalytically active in ricin and abrin. The recombinant sechiumin was obtained as an insoluble protein, and the preparation of active soluble form was achieved by renaturing the denatured protein. Site-directed mutagenesis was utilized to identify the potential active site of sechiumin with the mutants E160Q, R163L and E160Q.R163L, respectively. The IC50 of E160Q and R163L was found to decrease 150-fold and 457-fold to that of wild type re-sechiumin, respectively, by using rabbit reticulocyte lysate protein translation inhibition assay, while E160Q.R163L decreased 1222-fold. It suggested that Glu160 and R163 might play an important role in active site. Deletion of four residues from the C-terminus of sechiumin, decreased its protein biosynthesis inhibitory activity moderately. Deletion of seven residues from the C-terminus of sechiumin caused a great loss in its activity due to the removal of a highly conserved hydrophobic residue, Leu239. N-terminal deletion analysis revealed that the mutations of sechiumin with the deletion of the first 4 or 6 amino acid residues from N-terminus including the firstβ-sheet which contains a highly conserved hydrophobic residue, Phe4, led to the inactivation of the RIP. The results indicate that the two residues located either from N- or C-terminus of sechiumin are dispensable for its inhibitory activity of protein biosynthesis. The polypeptides with these deletions generated from this study may be useful for the preparation of valuable immunotoxins.

Ribosome-inactivating proteins (RIPs) are a group of related proteins that are widely distributed throughout the plant kingdom and share an unusual property of being able to inactivate eukaryotic ribosomes. A new type I ribosome- inactivating protein was purified from the seeds of Sechium edule Swartz by gel-filtration chromatography, ion exchange chromatography with CM52 cellulose column and FPLC Mono S column.The purified new type I RIP was designated as SECHIUMIN. The homogeneity of sechiumin was shown by SDS/PAGE analysis and the results indicated that sechiumin consists of a single polypeptide chain with a molecular weight of about 34,500 Dalton. From the results of IEF analysis and PAS staining, sechiumin was considered to be a basic (pI 8.4) glycoprotein. This protein inhibited the protein synthesis of a rabbit reticulocyte lysate with an IC50 (concentration causing 50% inhibition) of 1 nM and had much less effect on protein synthesis of Hela cells with IC50 of 5000-6000 nM. After rat liver ribosomes were treated with 1 nM sechiumin, the total rRNAs were isolated and treated with aniline, and a new rRNA fragment could be clearly observed , just like other RIPs , by PAGE. From these data, it was concluded that sechiumin is one of the type I ribosome- inactivating proteins.

Chayote (Sechium edule) is a fruit that is said to be a nutritional and have healing properties. 90% of its weight is made up of water which makes this fruit highly perishable. Convective drying is an excellent option to reduce the loss of this product as well as prolong its shelf life. Thus, the main objective of this work was to assess the physicochemical characteristics of different chayote samples (slices, cubes and strips) during its drying and rehydration process. Results suggest that cubes represent an alternative for commercial use as an additive in foodstuffs due to their size.Keywords: chayote; convective drying; rehydration process; physiochemical properties.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole.
