Abstract:
The filamentous fungus Aspergillus niger is an industrially exploited protein expression platform, well known for its capacity to secrete high levels of proteins. To study the process of protein secretion in A. niger, we established a GFP-v-SNARE reporter strain in which the trafficking and dynamics of secretory vesicles can be followed in vivo. The biological role of putative A. niger orthologues of seven secretion-specific genes, known to function in key aspects of the protein secretion machinery in Saccharomyces cerevisiae, was analysed by constructing respective gene deletion mutants in the GFP-v-SNARE reporter strain. Comparison of the deletion phenotype of conserved proteins functioning in the secretory pathway revealed common features but also interesting differences between S. cerevisiae and A. niger. Deletion of the S. cerevisiae Sec2p orthologue in A. niger (SecB), encoding a guanine exchange factor for the GTPase Sec4p (SrgA in A. niger), did not have an obvious phenotype, while SEC2 deletion in S. cerevisiae is lethal. Similarly, deletion of the A. niger orthologue of the S. cerevisiae exocyst subunit Sec3p (SecC) did not result in a lethal phenotype as in S. cerevisiae, although severe growth reduction of A. niger was observed. Deletion of secA, secH and ssoA (encoding SecA, SecH and SsoA the A. niger orthologues of S. cerevisiae Sec1p, Sec8p and Sso1/2p, respectively) showed that these genes are essential for A. niger, similar to the situation in S. cerevisiae. These data demonstrate that the orchestration of exocyst-mediated vesicle transport is only partially conserved in S. cerevisiae and A. niger.

Abstract:
This study assessed the differences between immigrants and natives in terms of combined effects of sugar-sweetened beverage (SSB) or snack consumption and preventive behaviors for severe early childhood caries (SECC) through a large-scale survey of 31,565 native and 1046 immigrant child–parent pairs in Taiwan. Children aged 3–5 years underwent dental examinations, and parents completed structured questionnaires. Immigrants had a significantly higher SECC prevalence than native children (32.3% vs. 19.4%). A combined effect of SECC was observed in native children who did not receive assistance when brushing teeth at night before sleep and those who consumed SSBs more than four times weekly (adjusted odds ratio (aOR) = 4.8). Moreover, native children who did not use dental floss and who consumed snacks more than four times weekly had an aOR of 4.1 for SECC. The combined effects of children with immigrant parents who did not receive assistance when brushing their teeth at night before sleep and those who consumed snacks more than four times weekly had an aOR of 8.2 for SECC. The results suggest the necessity of cross-cultural caries prevention programs for immigrants. Parents must limit children’s SSB and snack intake, and implement preventive measures to reduce SECC development.

Abstract:
ABSTRACT The Gram-positive pathogen Staphylococcus aureus secretes various proteins into its extracellular milieu. Bioinformatics analyses have indicated that most of these proteins are directed to the canonical Sec pathway, which consists of the translocation motor SecA and a membrane-embedded channel composed of the SecY, SecE, and SecG proteins. In addition, S. aureus contains an accessory Sec2 pathway involving the SecA2 and SecY2 proteins. Here, we have addressed the roles of the nonessential channel components SecG and SecY2 in the biogenesis of the extracellular proteome of S. aureus. The results show that SecG is of major importance for protein secretion by S. aureus. Specifically, the extracellular accumulation of nine abundant exoproteins and seven cell wall-bound proteins was significantly affected in an secG mutant. No secretion defects were detected for strains with a secY2 single mutation. However, deletion of secY2 exacerbated the secretion defects of secG mutants, affecting the extracellular accumulation of one additional exoprotein and one cell wall protein. Furthermore, an secG secY2 double mutant displayed a synthetic growth defect. This might relate to a slightly elevated expression of sraP, encoding the only known substrate for the Sec2 pathway, in cells lacking SecG. Additionally, the results suggest that SecY2 can interact with the Sec1 channel, which would be consistent with the presence of a single set of secE and secG genes in S. aureus.

Abstract:
ABSTRACT The majority of secretory proteins are translocated into and across hydrophobic membranes via the universally conserved Sec pore. Accessory proteins, including the SecDF-YajC Escherichia coli membrane complex, are required for efficient protein secretion. E. coli SecDF-YajC has been proposed to be involved in the membrane cycling of SecA, the cytoplasmic bacterial translocation ATPase, and in the stabilizing of SecG, a subunit of the Sec pore. While there are no identified archaeal homologs of either SecA or SecG, many archaea possess homologs of SecD and SecF. Here, we present the first study that addresses the function of archaeal SecD and SecF homologs. We show that the SecD and SecF components in the model archaeon Haloferax volcanii form a cytoplasmic membrane complex in the native host. Furthermore, as in E. coli, an H. volcanii ΔsecFD mutant strain exhibits both severe cold sensitivity and a Sec-specific protein translocation defect. Taken together, these results demonstrate significant functional conservation among the prokaryotic SecD and SecF homologs despite the distinct composition of their translocation machineries.

Abstract:
This study carried out tensile tests at definite elongation, tensile and shear tests on 4 admixture-modified styrene-acrylic emulsion-based cement composites (SECCs), and measured the strength, deformation, and energy consumption indexes of test specimens, so as to investigate the influences of coalescing agent, plasticizer, silane coupling agent, and nanometer aluminium oxide on the bond, tensile, and shear mechanical properties of the test specimens. Additionally, the Field Emission Scanning Electron Microscope (FE-SEM) test and Mercury Intrusion Porosimetry (MIP) test were conducted on the composite material specimens, to analyze the microscopic mechanism of different admixtures in modifying the mechanical properties of the SECC. The results suggested that the addition of coalescing agent, plasticizer, silane coupling agent, and nanometer aluminium oxide improved the bond, tensile and shear properties of the SECC specimens to various degrees. Of them, the coalescing agent promoted the mutual cross-linking of organic polymers with inorganic products, and optimized the transition interface to enhance the comprehensive mechanical properties of the test specimens; by contrast, nanometer aluminium oxide developed secondary hydration reaction with the inorganic products, and refined the pore structure to modify the mechanical properties of test specimens. Therefore, both of them achieved significant modification effects. Typically, the optimal bond properties of FFAMC, PLMC, SCAMC, and NAMC test specimens were achieved at the coalescing agent, plasticizer, silane coupling agent, and nanometer aluminium oxide addition amounts of 4%, 1.5%, 3%, and 1%, respectively. Besides, the improving effects of different admixtures on the tensile property of SECC specimens followed the order of coalescing agent > nanometer aluminium oxide > plasticizer > silane coupling agent, with the optimal addition amounts of 4%, 1.5%, 1%, and 2%, respectively. In addition, the improving effects of different admixtures on the shear performance of SECC specimens followed the order of coalescing agent > nanometer aluminium oxide > silane coupling agent > plasticizer, with the optimal addition amounts of 4%, 1.5%, 1%, and 1%, respectively.

Abstract:
Two new temperature-sensitive alleles of SEC3, 1 of 10 late-acting SEC genes required for targeting or fusion of post-Golgi secretory vesicles to the plasma membrane in Saccharomyces cerevisiae, were isolated in a screen for temperature-sensitive secretory mutants that are synthetically lethal with sec4-8. The new sec3 alleles affect early as well as late stages of secretion. Cloning and sequencing of the SEC3 gene revealed that it is identical to profilin synthetic lethal 1 (PSL1). The SEC3 gene is not essential because cells depleted of Sec3p are viable although slow growing and temperature sensitive. All of the sec3 alleles genetically interact with a profilin mutation, pfy1-111. The SEC3 gene in high copy suppresses pfy1-111 and sec5-24 and causes synthetic growth defects with ypt1, sec8-9, sec10-2, and sec15-1. Actin structure is only perturbed in conditions of chronic loss of Sec3p function, implying that Sec3p does not directly regulate actin. All alleles of sec3 cause bud site selection defects in homozygous diploids, as do sec4-8 and sec9-4. This suggests that SEC gene products are involved in determining the bud site and is consistent with a role for Sec3p in determining the correct site of exocytosis.

Abstract:
Early childhood caries (ECC) is recognized as an infectious disease. The first step in its development is primary infection by the bacterium S. mutans which has been identified as the primary etiologic factors in dental caries. Lactobacilli were also found to play a role in the progression of the disease. However, the underlying mechanism of immune response to caries is still unclear. The purpose of this study was to assess the level of cariogenic bacteria namely S. mutans and lactobacilli in caries free children, and children with SECC and their corresponding mothers. The study also aims at correlating the children's levels to their mothers. Sixty children and their mothers attending the dental clinic in King Abdulaziz University participated in our study. Their age ranged from 3 – 5 years. The study groups consisted of 30 children with SECC and a control group comprising of 30 caries free children. Children together with their mothers were examined and their caries level was recorded. Stimulated saliva was collected from each participant for bacterial, immunological assessment, and Lactobacilli counts in each sample were determined. Children with SECC had higher levels of S. mutans and Lactobacilli than caries free children. The mothers of children with SECC had a statistically higher count of Lactobacilli than caries free children's mothers. However, the difference was not statistically significant with respect to their S. mutans counts. A significant relationship exists among the mother- child pair in the SECC group with respect to S. mutans level in saliva.

CICERCHIA, ANDREA. "Giustizia di antico regime: il Tribunale criminale dell'Auditor Camerae (secc. XVI-XVII)." Doctoral thesis, Università degli Studi di Roma "Tor Vergata", 2010. http://hdl.handle.net/2108/1372.

Full text
Abstract:
This research aims to review the implementing guidelines of the government of justice within the papal state machinery during the centuries of old regime. In particular it was taken into analysis the long evolutionary parable of the criminal jurisdiction of an ordinary bench for the city of Rome and of a central one for the whole territory of the State: the Criminal Court of Auditor Camerae, autonomous from 1485 from the Apostolic Camera, endowed with large civil and criminal competences on clergy and laity of the Curia. Its vast competences on Chambers connoted it, since the beginning of the 16th century, as the main organ in the provision of civil justice in Rome; both the characteristics of papal power (spiritual and temporal) led the Court to extend – in the field of Commerce and of non-compliance to the papal bulls – its jurisdiction, with the power to impose excommunications and interdicts. The impossibility to divide these skills and the complexity of the procedures in criminal matter have so inclined historiography to overshadow the dynamics in favor of a broader consideration of the civil aspect (indeed corroborated by a wider conservation of the civil fund, compared to the criminal one). Therefore the present research’s purpose is to highlight the procedure in criminalibus of Justice exerted by Auditor Camerae. Specifically, it was developed a cross-study of two chronological plans concerning the development of the Court: on the one hand, it has been made some order in the official law, with reference to a centuries-old route that from 1485 could embrace the entire legal and institutional development until the first half of the eighteenth century. The sources used in this context were found, as well as through the official bullarum, also in various funds kept in the Vatican Secret Archives and in the collections of announcements and edicts issued by the Court; on the other hand, the present research aims to approach an investigation able to weave these normative sources with those ones preserved in the fund of the criminal court (preserved in the State Archives of Rome) in order to deepen the analysis of the institutional structure - in particular that of the judges of officer of the police and notaries - and record the inevitable deviation from what was the effective exercise of justice; in the latter case, the attempt was to focus the investigation on that period, between the sixteenth and seventeenth century, defined as the real apogee of the Court. What emerges, ultimately, from this research is the image of a complex judicial organism that especially in the late sixteenth and early next century - particularly around the years of the reform of Paul V (1612) - was found to play an important role even from the criminal justice’s point of view and not just in roman jurisdiction but in the whole Papal State’s territorial context. Are so defined some institutional aspects of a Court (which has not been specifically examined by the most recent historiographical analysis), which make it possible to trace broader considerations about the entire government of justice in the territories of the Church between the sixteenth and seventeenth century.

Abstract:
The thesis focuses on the relationship between a male religious community, the Congregation of the Oratory of Rome of the 16th and 17th centuries, and healthcare, connecting social history, religious history and history of medicine. The first part centers on the involvement of the Oratorians in the urban charitable network, underlining their contribution in the development and the administration of a series of hospitals, but also their practical commitment in the spiritual and bodily care of the sick. The second part concentrates on the health professionals who attended the Oratory, framing their relationship with the fathers within a broader network of social and political connections in the Papal Rome. Also, stressing the differences between apothecaries, barber-surgeons and physicians, the analysis considers their involvement in the religious life of the community and the exploitation of their humanistic and anatomical expertise. The third part studies the complex relationship of the priests with their own bodies and healthcare. It highlights the weight of bodily conditions for the acceptance of new members in the Oratory, and the links between aesthetics, social identity and medicine, but it takes into account also the ambiguous functions of food – intended as a tool both for asceticism and healthcare – and the concrete functioning of the medical organization of the community. Moreover, a chapter analyses the circulation of medical knowledge, highlighting the role of the library, the collection and exchange of medical recipes and the importance of the Oratorian apothecary shop, while another chapter concentrates upon the therapeutic voyages and the practice of thermalism. The last part, focusing on the Roman plague of 1656-57, analyses how the Congregation coped with the epidemic outbreak and development, both in the house and in the city, in order to underline their fragile and ambiguous balance between charity and health protection.

Abstract:
Between the Middle Ages and the end of the eighteenth century, taxation in Europe was very different from that of modern society. It was characterised by the predominance of indirect taxes over direct, with the latter being substantially irregular. The studies published in these proceedings of Study Week promoted by the Fondazione Datini examine the comparative evolution of the European fiscal systems and fiscal policies and the effects of taxation on the economy. The wealth of specific and original contributions, as well as the attempts at structural organisation, reveal the distinct vivacity of European and western historiography, offering an overall and articulated picture of the European tax systems in the pre-industrial age.

Abstract:
AbstractIn this chapter I survey key theories and concepts that provide substance to the workings of Transformative Social-Ecological Innovation (TSEI). A number of relevant theories and concept have already been mentioned in the previous chapters, such as Social Contract theory (Sect. 10.1007/978-3-030-67130-3_3#Sec1), and in Sects. 10.1007/978-3-030-67130-3_3#Sec8 and 10.1007/978-3-030-67130-3_3#Sec13, including resilience theory and social-ecological systems (Sect. 10.1007/978-3-030-67130-3_3#Sec8), quintuple helix innovation model (Sect. 10.1007/978-3-030-67130-3_3#Sec13), as well as institutional change and the structure-agency debate (Sect. 10.1007/978-3-030-67130-3_3#Sec13), and several economic theories (Sects. 10.1007/978-3-030-67130-3_3#Sec2 and 10.1007/978-3-030-67130-3_3#Sec3). In this chapter I will start with providing a conceptual discussion and definition on Transformative Social-Ecological Innovation (Sect. 4.1), and devote more attention to various theories and approaches that are relevant for TSEI, such as transition studies (Sect. 4.2), institutional design principles for governing the commons (Sect. 4.3), design principles from nature (Sect. 4.4), complex adaptive systems (Sect. 4.5), adaptive, reflexive, and deliberative approaches to governance, management, and planning (Sect. 4.6), social learning, policy learning, and transformational learning (Sect. 4.7), shared value, multiple value creation, and mutual gains approach (Sect. 4.8), effective cooperation (Sect. 4.9), transdisciplinary cooperation, living labs, and citizen science (Sect. 4.10), and the art of co-creation: approaches, principles and pitfalls (Sect. 4.11).

Abstract:
Abstract In this chapter, solutions to the wave equation that satisfies the boundary conditions within three-dimensional enclosures of different shapes are derived. This treatment is very similar to the two-dimensional solutions for waves on a membrane of Chap. 10.1007/978-3-030-44787-8_6. Many of the concepts introduced in Sect. 10.1007/978-3-030-44787-8_6#Sec1 for rectangular membranes and Sect. 10.1007/978-3-030-44787-8_6#Sec5 for circular membranes are repeated here with only slight modifications. These concepts include separation of variables, normal modes, modal degeneracy, and density of modes, as well as adiabatic invariance and the splitting of degenerate modes by perturbations. Throughout this chapter, familiarity with the results of Chap. 10.1007/978-3-030-44787-8_6 will be assumed. The similarities between the standing-wave solutions within enclosures of different shapes are stressed. At high enough frequencies, where the individual modes overlap, statistical energy analysis will be introduced to describe the diffuse (reverberant) sound field.
