Abstract:
Two new temperature-sensitive alleles of SEC3, 1 of 10 late-acting SEC genes required for targeting or fusion of post-Golgi secretory vesicles to the plasma membrane in Saccharomyces cerevisiae, were isolated in a screen for temperature-sensitive secretory mutants that are synthetically lethal with sec4-8. The new sec3 alleles affect early as well as late stages of secretion. Cloning and sequencing of the SEC3 gene revealed that it is identical to profilin synthetic lethal 1 (PSL1). The SEC3 gene is not essential because cells depleted of Sec3p are viable although slow growing and temperature sensitive. All of the sec3 alleles genetically interact with a profilin mutation, pfy1-111. The SEC3 gene in high copy suppresses pfy1-111 and sec5-24 and causes synthetic growth defects with ypt1, sec8-9, sec10-2, and sec15-1. Actin structure is only perturbed in conditions of chronic loss of Sec3p function, implying that Sec3p does not directly regulate actin. All alleles of sec3 cause bud site selection defects in homozygous diploids, as do sec4-8 and sec9-4. This suggests that SEC gene products are involved in determining the bud site and is consistent with a role for Sec3p in determining the correct site of exocytosis.

Abstract:
The sac culture method in combination with 6% NZ-Amine A plus 1% yeast extract was found to be the optimal condition for staphylococcal enterotoxins A (SEA) and D (SED) production. This growth condition was tested for the production of unidentified staphylococcal enterotoxins (SEs). Twenty-one Staphylococcus aureus strains that previously showed an emetic response in monkeys but were negative for any of the identified SEs when cultured by the membrane-over-agar method were grown by the sac culture method. All 21 strains produced at least one known SE. One strain produced only enterotoxin C (SEC). Nine strains produced only SED. One strain produced both SEA and SED. Four produced both SEC and SED. One produced SEA, enterotoxin B (SEB), and SED. Two produced SEA, SEC, and SED. Three produced SEA, SEB, SEC, and SED. One of these strains, FRI-569, that produced only SED at a low level (10 ng/ml) was selected to confirm the production of an unidentified SE by the monkey feeding test. Emesis was induced in five out of six monkeys. The total amount of SED in the supernatant fluids fed to monkeys was only 0.5 μg, or 2.5% of the 50% emetic dose (20 μg). Production of an unidentified SE by strain FRI-569 was therefore confirmed. This optimal condition can be used to produce increased amounts of unidentified SEs for purification.

Abstract:
The avidin–biotin enzyme-linked immunosorbent assay (ELISA), reversed passive latex agglutination (RPLA) test, and the modified Ouchterlony precipitation test (MOPT) were compared in detecting enterotoxin production by Staphylococcus aureus strains. A total of 1015 strains isolated from human beings, animals, and foods were tested for staphylococcal enterotoxins A (SEA), B (SEB), and C (SEC). Of these, 495 (48.8%), 467 (46.0%), and 204 (20.1%) were classified as enterotoxigenic by the ELISA, RPLA test, and MOPT, respectively. The difference in the number of strains classified as enterotoxigenic by the ELISA and RPLA test was not significant (P ≥ 0.05; χ2), but both tests detected significantly (P < 0.001; χ2) more enterotoxigenic strains than the MOPT. The combined use of the three assay systems classified 258 (25.4%), 278 (27.4%), and 263 (25.9%) of 1015 strains tested as positive for SEA, SEB, and SEC, respectively. However, the three systems were all positive in only 29.1% of SEA-producing strains, 32.0% of SEB-producing strains, and 25.1% of SEC-producing strains. The MOPT was negative when the corresponding ELISA and RPLA test were positive (46.9% for SEA, 43.5% for SEB, and 40% for SEC); the RPLA test was negative when the corresponding Elisa was positive (10.5% for SEA, 15.5% for SEB, and 25.5% for SEC); and the ELISA was negative when the RPLA test was positive (13.6% for SEA, 9.0% for SEB, and 9.5% for SEC). All factors considered, the RPLA test appears most suitable for quantitatively screening large numbers of strains for staphylococcal enterotoxins. Of 504 food samples collected from cafeterias, 19 (3.8%) yielded strains of enterotoxigenic staphylococci, and 10 (52.6%), 4 (21.1%), 3 (15.8%), and 2 (10.5%) of these strains produced enterotoxins C (SEC), D (SED), B (SEB), and A (SEA), respectively. Moreover, SEA, SEB, and SEC were isolated from three hamburger samples. Of 181 food samples collected from four restaurants before the implementation of the hazard analysis and critical control point (HACCP) system, 7 (3.9%) were found to contain enterotoxigenic strains, and SED, SEC, and SEA were produced by 4 (57.1%), 2 (28.6%), and 1 (14.3%) of these strains, respectively. One meatball sample with SEC was detected in a restaurant. After the implementation of the HACCP system in four restaurants, neither enterotoxigenic staphylococci nor enterotoxins were detected in 196 studied samples.

Abstract:
Aim. To identify the frequency of occurrence of staphylococcal enterotoxins of the SEC and SEI type produced by Staphylococcus aureus strains isolated from patients with different nosology. As you know, the infection process in them proceeds with severe intoxication without vomiting and intestinal disorders.Materials and methods. 79 strains were studied (43 were isolated in case of pneumonia, 13 - in burns, 11 - in sepsis) S. aureus in the presence of SES and SEI using enzymelinked immunosorbent assay (ELISA).Results. It was found that 48.3% of S. aureus strains isolated from patients with pneumonia produced SEC and 72.1% - SEI. The frequency of occurrence of S. aureus strains producing SEC and SEI isolated in patients with burn infections was 23.0 and 15.4%, respectively. 36.4% of staphylococcal strains isolated in patients with sepsis produced SEC, 45.5% - SEI.Discussion. It was found that the proportion of S. aureus cultures producing SEC enterotoxins during sepsis is significantly higher than the strains producing SEB (5.4%) and much smaller than SEA (75.6%). A high percentage of SEI-positive strains was found compared to strains that produce the classic enterotoxins SEA, SEB and SEC isolated from pneumonia. In burn infections, the proportions of strains producing SEC and SEI were 15.4 and 23.0%, respectively, which is significantly lower than SEA (92.9%).Conclusion. The data obtained indicate the need to identify staphylococcus strains that produce both classic and newly discovered enterotoxins, which are crucial virulence factors leading to lethal sepsis, infectious endocarditis and toxic shock syndrome to eliminate them.

A total of 100 Jordanian clinical Staphylococcus aureus isolates was analysed for the presence of the enterotoxin genes sea, seb, sec, sed and see using multiplex PCR. Twenty-three isolates (23 %) were potentially enterotoxigenic. The prevalence of sea, sec and sea plus sec among the total clinical isolates was 15, 4 and 4 %, respectively. None of the isolates harboured sed, seb or see genes. S. aureus isolates were subjected to DNA fingerprinting by randomly amplified polymorphic DNA (RAPD) analysis to test whether isolates harbouring the toxin genes were genetically clustered. A total of 13 genotypes was identified at a 47 % similarity level. Genotypes I and V accounted for the largest number of enterotoxigenic isolates (19 %). This study has demonstrated the genetic diversity of Jordanian clinical S. aureus isolates and shown that the presence of the toxin genes is not genotype specific.

Staphylococcal food poisoning (SFP), caused by the contamination of staphylococcal enterotoxins, is a common foodborne disease worldwide. The aims of this study were: (1) to investigate classical staphylococcal enterotoxin genes, sea, seb, sec, sed, and see, among Staphylococcus aureus and coagulase-negative staphylococci (CNS) associated with bovine mastitis; (2) to determine the effect of temperature on the expression of classical staphylococcal enterotoxin genes in staphylococci in milk. The detection of classical staphylococcal enterotoxin genes was performed using S. aureus (n = 51) and CNS (n = 47). The expression of classical enterotoxin genes, including sea, seb, sec, and see, was determined during the growth of staphylococci in milk subjected to ultra-high-temperature processing at two different temperatures: 8 °C and room temperature. Classical staphylococcal enterotoxin genes were expressed more frequently in S. aureus (35.30%) than in CNS (12.77%). The sec gene was most frequently detected in S. aureus (29.41%) and CNS (6.38%). Moreover, the expression of sea and sec was significantly higher at room temperature than at 8 °C after 16 h of incubation (p < 0.05). These results emphasize the importance of maintaining the storage temperature of milk below 8 °C to reduce the risk of SFP.

The aim of this study was to investigate staphylococcal enterotoxins (SEs) by ELISA, and detect the five classical sea, seb, sec, sed, and see genes by real-time PCR in Staphylococcus aureus isolates from raw and pasteurized milk samples. Staphylococcus spp. were isolated from 98 out of 100 raw milk samples, and in 6 out of 100 pasteurized milk samples. On further biochemical tests, S. aureus was isolated in 48 samples (48%) of raw milk (n=100) and in one sample (1%) of pasteurized milk (n=100). Ten (10%) out of 100 raw milk samples were positive for at least one enterotoxin, and the most frequently observed SE was SEA (10%), followed by SEE (7%) and SEB (6%), but none of the isolates were positive for SEC and SED. At least one of the SEs gene types (sea, seb, sec, sed, see) was detected in 45 (93.8%; 45/48) S. aureus isolates from raw milk samples. sec, sea, seb, sed, and see genes were observed in 56.2%, 39.5%, 31.2%, 29.1% and 14.5% of strains respectively. The enterotoxin genes were the single type in 21 (46.7%) of the 45 isolates, there were two in 15 (33.3%), three in six (13.3%), four in two (4.4%), and one (2.2%) in all gene regions. The SE gene was not detected in the S. aureus (n=1) isolate from pasteurized milk. As a result of this study, the presence of enterotoxigenic S. aureus in raw milk was revealed, and it was pointed out that these SEs may contribute to cases of staphylococcal foodborne poisoning (SPF).

Staphylococcus aureus is an important cause of mastitis in cows. The ability of S. aureus strains to produce one or more enterotoxins in milk and dairy products is linked to staphylococcal food poisoning. To determine whether staphylococci causing bovine mastitis could cause human foodborne intoxication, the production of staphylococcal enterotoxins A through D (SEA, SEB, SEC, and SED) by 160 S. aureus isolates was evaluated with the use of a reverse passive latex agglutination enterotoxin kit. All S. aureus strains were isolated over a 9-month period from 2,343 routine submissions of a composite quarter collection of individual mastitic cows at 18 dairy farms in the San Joaquin Valley in California. Prior to enterotoxin detection, isolates were grown by a method that enhances the in vitro synthesis of enterotoxin. Twenty-two of 160 S. aureus isolates produced enterotoxin. Seven produced SEC, 12 produced SED, and 3 produced both SEC and SED. None of the isolates produced SEA or SEB.

L'ouvrage est divisé en trois parties: ce choix a été principalement motivé par l'exigence de fournir un cadre et un contexte historique aux évidences archéologiques et à l'entière problématique historique de la Sogdiane, notre domaine d'étude et d'activité. Le rôle géographique de cette région, située dans la vallée du fleuve Zeravs̆ān, a rendu inévitable de se reporter à l'autre région centre-asiatique de la Bactriane, tout aussi importante que celle-ci, géographiquement et historiquement proche de la Sogdiane. En raison d'une cohérence chronologique et historique des évènements, on a également dû évoquer d'autres zones géographiques et culturelles, forcément liées au coeur de l'Asie Centrale, c'est à dire le Plateau Iranien, la Chine, l'Inde et les terres originaires des grecs conduits par Alexandre le Grand. En outre, même si de façon peu approfondie, une section a été consacrée à un phénomène d'importance cruciale dans le domaine des études sur l'Asie Centrale et les steppes, à savoir le nomadisme pastorale, aussi bien pour ses aspects économiques et productives que pour son caractère de contact et déstabilisation par rapport aux société sédentaires, mais également pour son développement interne et progressif vers une organisation socio-politique et une confédération de type tribale. Pour des raisons pratiques, nous définirons la période historique du IIe siècle jusqu'au Ier siècle av. J. -C. "première phase nomadique", tandis que nous définirons "seconde phase nomadique" celle du IVe au Ve siècle ap. J. -C. , liée aux populations des Chionites, des Kidarites et des Hephtalites. La première partie présente les résultats des investigations archéologiques dans le site de Kotjepa (27 km sud-ouest de Samarcande) achevées par la Mission Archéologique Italo-Ouzbèk et de l'étude de la poterie entre les années 2008-2012. La deuxième aborde les évènements historiques et politiques de la fin du règne grec de Bactriane jusqu'à la création de la confédération nomadique des Hephtalites. La troisième partie, plus proprement archéologique, est un catalogue des principaux sites archéologiques et des cimetières nomadiques avec lesquels ils sont comparés les résultats de la fouille de Kojtepa: bien si la plus part des comparaisons n'a pas toujours fournir des réponses "positives", il a toutefois été décidé de présenter lui pour compléter le cadre historique tracée dans la première partie
The present work is divided into three distinct parts: this choice was motivated primarily by the necessity to provide a historical background to contextualize the archaelogical evidence that the whole issue concerning the historical Sogdiana, an area of our business on the fied. The geographical role of the region, centered in the fertile valley of the river Zeravšān, made it inevitable to refer to another, very important region of Central Asian, the region of Bactria, geographically contiguous and historically linked to Sogdiana. For similar reasons, in the name of chronological and historical coherence of events, reference was made to other geographical - cultural reality, inevitably connected to the heart of Central Asia, namely the iranian plateau, China, India and to the far land of Greeks of Alexander the Macedonian. Although not in the form too in-depth, one section is also dedicated to a phenomenon of crucial importance in studies on Central Asia and the steppe regions, the pastoral nomadism, both fot its economic and productive character and as point of contact and destabilization in relation to sedentary societies, and also for its internal development and the progressive evolution towards socio-political organizations of tribal confederation. For practical purposes, the historical period that goes from the 2nd century BC to the 1st century AD, was defined as "first nomadic phase", as well as the 4th and 5th centuries was defined as "second nomadic phase", in reference to the people of Chionites, Kidarites and Hephtalites. The first part of the work, presents the results of the field-work of the Uzbek-Italian Archaelogical Mission on the site of Kojtepa (Samarkand area) and the study of ceramic materials in order better interpret the role of the settlement, its history and its relations with other sites in the same region. The second part deals with the historical and political events from the end of the Greek kingdom of Bactria until the creation of the nomadic confederation of Hephtalites, and the third, more strictly archaelogical, describes the main archaeological sites related to each of the "historical steps" discussed above

How does one approach the human body when designing and use it as a tool for investigating to develop shape-expression in dress? For this research, the movement of the human body has been chosen as the base element to generate shapes from movement, to enable a form of visible objects. This has been accomplished by carefully studying the original movement of a human body through film. Looking att balance, direction and general shape. The new human body, which has been generated from this research, provides the viewer or creator a different perspective of the body in relation to the static body, which is regularly used in fashion design from the beginning of the design process. This research has been conducted to propose the importance of the human body as an active element in the beginning of the design process and to be viewed as a new platform of innovative design and artistic development in dress. In addition, to develop a contemporary understanding of the body’s role when designing and developing fashion design. Abstract:
Frutto dell’operato di maestranze specializzate ed espressione di un’edilizia di tenore elevato, i rivestimenti pavimentali costituiscono degli importanti indicatori archeologici, non solo come documenti di un sapere artigianale e di una cultura decorativa, ma anche come reperti-guida per una più ampia comprensione delle dinamiche insediative e socio-economiche di specifici ambiti territoriali. Il presente lavoro ha come oggetto lo studio complessivo dei mosaici e dei pavimenti di età antica rinvenuti nel territorio della Basilicata. Attraverso la raccolta e la razionalizzazione di tutta la documentazione esistente, edita ed inedita, ci si è proposti di realizzare un corpus organico che fosse funzionale ad una prima ricostruzione della cultura musiva dell’area indagata nella sua evoluzione storica, dal periodo ellenistico (metà IV sec. a.C.) alle soglie del Medioevo (fine VI sec. d.C.). Il lavoro comprende un totale di 324 pavimentazioni afferenti a diverse tecniche (cementizi decorati, tessellati, opus sectile, a commessi laterizi, a ciottoli e in tecniche miste) e provenienti sia da centri urbani (domus, impianti termale, edifici di culto, basiliche paleocristiane) sia – soprattutto – da contesti extraurbani (villae rusticae e complessi rurali). Ciascuna attestazione è stata oggetto di una schedatura sistematica che si è avvalsa dell’utilizzo di uno strumento di lavoro all’avanguardia. La ricerca, infatti, s’inserisce nel più ampio Progetto TESS di catalogazione dei pavimenti antichi della Penisola, coordinato al livello nazionale dal Dipartimento di Archeologia dell’Università di Padova (database consultabile on-line al sito: http://tess.beniculturali.unipd.it). La banca dati informatizzata ha costituito un ausilio fondamentale, non solo per un rapido e puntuale confronto con i dati relativi ad altre aree geografiche, ma soprattutto per l’organizzazione di un panorama documentario estremamente ampio e frammentario come quello lucano, composto da notizie e pubblicazioni di respiro locale o di più ampia diffusione nazionale. Lo spoglio degli archivi (ove autorizzato) e l’analisi autoptica dei rivestimenti ancora visibili hanno garantito la completezza delle informazioni, consentendo il recupero dei dati tecnici necessari per la creazione del catalogo. La ricca messe di disiecta membra è stata sistematizzata cercando innanzitutto di ricollocare – quando possibile – le singole attestazioni nel proprio contesto architettonico di pertinenza e di analizzarne in dettaglio i singoli elementi costituenti (aspetti decorativi, figurativi, tecnico-artigianali e conservativi). Per migliorare l’organizzazione della documentazione, si è inteso suddividerla in cinque macro-aree omogenee dal punto vista storico-topografico, che corrispondono ai cinque capitoli di cui si compone la prima parte della tesi. Nella seconda sezione, invece, si è cercato di rielaborare criticamente i dati esaminati e evidenziare le linee di tendenza più significative (topografiche, cronologiche, tecniche e decorative) per la ricostruzione di un primo quadro complessivo del patrimonio musivo del territorio indagato.

Il progetto di ricerca ha indagato la diffusione e le caratteristiche dei vetri in uso a Milano in età romana a partire dallo studio di un certo numero di reperti provenienti da scavi archeologici e tramite l’approfondimento di alcune tematiche relative ad aspetti produttivi e tecnologici. La ricerca ha preso spunto dallo studio dei materiali provenienti dagli scavi nei cortili dell’Università Cattolica (1986-2004), ed è stata estesa poi ad altri contesti di scavo per ampliare la base dei dati su cui ricostruire il quadro del vetro in uso nella città e delle problematiche ad esso inerenti. Il quantitativo di reperti individuati e la loro rappresentatività statistica permettono di tracciare un quadro dei materiali in uso nella vita quotidiana, le relazioni tipologiche tra alcuni gruppi di oggetti, vagliare le ipotesi sull’esistenza di una produzione locale e sui rapporti commerciali con altre aree, relativamente al periodo che va dall’inizio della diffusione dei recipienti in vetro (seconda metà del I sec. a.C.) fino al V sec. d.C. La ricerca si è avvalsa di analisi archeometriche ed ha enucleato anche alcuni temi di indagine specifica, in particolare sulle tecniche di lavorazione e di decorazione dei vetri a mosaico e dei manufatti con decori incisi.
Finds from recent archaeological excavation and analysis of older records are piecing together an increasingly detailed picture of the social context of Roman Milan: the material culture helps patch together a collage of the urban centre whose central position on the river plain North of the Po helped it become theatre to important events and intense industrial activity, attracting peoples of differing cultures and resources. This thesis studies Roman glass finds from Milan using the archaeological record and archaeometric analyses with the aim of reconstructing the framework of glass use in everyday life from the introduction of glass vessels in the mid-first century B.C. up to the fifth century A.D., considering relationship between individual groups of products, the hypothesis of a local glass industry and trade routes. The research, originating from post-excavation work on the courtyards of the Catholic University was extended to several unpublished Milanese excavations: the glass finds from these contexts, from both burial grounds and settlement sites, constitute a significant and statistically valid sample. Some in-depth technological examinations contribute to an understanding of certain questions still under debate: in particular on mosaic glass and engraved samples

Der Schwerpunkt liegt dabei auf Möglichkeiten der Erfassung derartiger Kompetenzen im Rahmen der psychologischen Diagnostik in Einrichtungen zur beruflichen Förderung von Menschen mit Lernbehinderungen. Dementsprechend ist der Workshop in erster Linie als anwendungsorientiertes Training für Fachkräfte in der beruflichen Bildung zu verstehen, welche die an der Universität Bamberg entwickelten diagnostischen Verfahren SEC-I und SEC-SJT (Inventar und Situational Judgement Test zur Erfassung sozial-emotionaler Kompetenzen bei jungen Menschen mit (sub-)klinischer kognitiver bzw. psychischer Beeinträchtigung) anwenden bzw. mit der Anwendung betraute Personen schulen möchten. Der Workshop umfasst sieben Themenbereiche, die je nach Bedarf flexibel zusammengestellt werden können: Theoretische Grundlagen und verschiedene Definitionen sozial-emotionaler Kompetenz, Grundlagen psychologischer Diagnostik, Potenzielle Schwierigkeiten beim Einsatz der diagnostischen Verfahren, Anwendung des Fragebogens zur Selbsteinschätzung bzw. des Situational Judgement Tests, Anwendung des Fragebogens zur Fremdeinschätzung und objektive Verhaltensbeobachtung. The modular workshop offers a science-based introduction to the concept of social-emotional competences. It focuses on the psychological assessment of such competences in in institutions specialized in the professional development of people with learning disabilities. As such, the workshop is primarily to be understood as an application-oriented training programme for professionals who work in vocational education and use (or teach the usage of) the assessment tools SEC-I and SEC-SJT (Inventory and Situational Judgment Test for the assessment of social-emotional competence in young people with (sub-) clinical cognitive or psychological impairment) which were developed at the University of Bamberg. The workshop comprises seven subject areas that can be flexibly put together as required: theoretical basics and definitions of social-emotional competence, the basics of psychological assessment, potential difficulties in its use, usage of the self-rating scale, the situational judgment test, the observer-rating scale, and objective observation of behaviour. The general aim of this workshop is to learn how to use and apply the assessment tools in practical settings.
