Abstract:
Abstract In this paper, a high-precision, energy-saving dynamic zoom ADC consisting of a 6-bit SAR ADC and a second-level Σ − ∆ modulator (SDM) is proposed. Non-ideal factors, including slew rate (SR), limited bandwidth and DC gain of the op-amp, and kT/C noise, mismatch of DAC capacitors, are analyzed with a behavioral model in Simulink to guide actual circuit design more accurately. The ADC parameters are adjusted and optimized by model simulation. Because the DAC capacitors mismatch produces harmonic distortion, the DWA technique is used. A new digital combined circuit is proposed to minimize power consumption. To further improve its energy efficiency, a gain enhancement current mirror OTA is designed and the SAR ADC adopts upper plate sampling technique. The layout of zoom ADC occupies 0.856 mm2 in 65 nm techniques. It achieves 109.2 dB SNDR, 17.84 bits ENOB at 1 kHz signal bandwidth and consumes only 0.233 mW.

Abstract. An uncooled VGA Infrared Focal Plane Array (IRFPA) based on microbolometers with a pixel pitch of 25 μm for thermal imaging applications is presented. The IRFPA has a 16-bit digital video data output at a frame rate of 30 Hz. Thousands of Analog to Digital Converters (ADCs) are located under the microbolometer array. One ADC consists of a Sigma-Delta-Modulator (SDM) of 2nd order and a decimation filter. It is multiplexed for a certain amount of microbolometers arranged in a so called "cluster". In the 1st stage of the SDM the microbolometer current is integrated time-continuously. The feedback is applied using a switchable current source. First measurements of Noise Equivalent Temperature Difference (NETD) as a key parameter for IRFPAs will be presented.

Realisasi terhadap desentralisasi pendidikan di tingkat sekolah adalah diberikannya otonomi yang luas dalam pengelolaan sumber daya yang ada. Pemberdayaan sekolah, di samping untuk memenuhi tuntutan desentralisasi, juga ditujukan sebagai sarana peningkatan efisiensi, mutu, dan pemerataan pendidikan. Di Indonesia, bentuk otonomi sekolah tersebut akan dilaksanakan dalam konteks “School Based Management” (SBM), di mana selain memiliki otonomi yang luas, sekolah wajib mengikutsertakan masyarakat dalam pengelolaan, dan pengelolaan sekolah dilakukan dalam kerangka kebijakan nasional. Penelitian ini dikategorikan sebagai penelitian yang bersifat kualitatif-deskriptis yang menggunakan pendekatan studi kasus. Dan untuk mendapatkan data-datanya, diperlukan metode wawancara, observasi dan dokumentasi. Sedangkan dalam menganalisa digunakan analisa data SWOT Analisis (Strength, Weakness, Opportunity, Threat) yang mempertimbangkan faktor kekuatan, kelemahan, peluang dan ancaman. Dari penelitian yang dilakukan dihasilkan kesimpulan, pertama School Based Management (SBM) merupakan alternatif yang dapat diterapkan dalam mengelola sekolah/madrasah. Kedua, penerapan SBM pada beberapa sekolah unggulan ternyata amat bervariasi jika dilihat dari sudut strategi pelaksanaan dan hambatan yang dihadapi. Ketiga, hambatan yang dihadapi tersebut dalam melaksanakan SBM meliputi: rendahnya partisipasi masyarakat, rendahnya kualitas SDM dan faktor eksternal. Keempat, di samping hal yang dapat menghambat penerapan SBM, ada peluang yaitu SDM yang memadai, dukungan partisipasi masyarakat serta konteks sosial yang cenderung untuk menyekolahkan anaknya di madrasah.

Dusun Arjomulyo merupakan salah satu dusun di Desa Bangelan yang mempunyai potensi Sumber Daya Alam yang baik. Daerah ini memiliki berbagai potensi yang dapat dikembangkan. Mulai dari potensi tanah yang subur cocok untuk pertanian maupun perkebunan, peternakan hingga potensi wisata. Kegiatan ini bertujuan untuk mengembangkan beberapa potensi yang ada pada Dusun Arjomulyo Desa Bangelan ini. Program kerja kami di Dusun Arjomulyo antara lain : Pengembangan SDM dan SDA Dusun Arjomulyo Desa Bangelan untuk ekonomi unggul, penambahan fasilitas wisata Umbulan Tanaka Water Fall, pembuatan papan sejarah Tanaka Water Fall, pembuatan papan petunjuk. Pengambilan sampel dilakukan dengan teknik Snowball Sampling. Data diambil dari hasil wawancara dengan Bapak Budiono, S.Sos selaku Kepala Desa Bangelan, Bapak Samu’in selaku Kepala Dusun dan Bapak Sudar selaku Pengelola Wisata. Hasil penelitian kami menunjukkan dengan adanya potensi wilayah perkebunan dan wisata yang pada saat ini sedang dikembangkan oleh swadaya masyarakat Dusun Tanaka (Sekarang Arjomulyo) mempunyai peran penting dalam masyarakat serta dapat mendorong peningkatan ekonomi masyarakat Dusun Arjomulyo Desa Bangelan. Simpulan dari kegiatan dan kamian ini adalah dengan dilakukannnya pengembangan SDM dan SDA Dusun Arjomulyo Desa bangelan untuk Ekonomi Unggul serta penambahan fasilitas penunjang yang terdapat pada Wisata Umbulan Tanaka Water Fall.

Tujuan penelitian ini adalah untuk mengetahui Sport Development Index Kabupaten Mandailing Natal Provinsi Sumatera Utara. Metode penelitian ini adalah penelitian kuantitatif dengan pendekatan deskriptif. Penelitian dilaksanakan di kabupaten Mandailing Natal Provinsi Sumatera Utara. Waktu penelitian dimulai dari bulan Januari 2019 s/d Februari 2019. Kegiatan tersebut dimulai dari pengajuan ijin, tahap pengumpilan data, dan tahap pengolahan data. Teknik pengumpulan data SDI dilakukan dengan metode cluster sampling. Jadi dalam penelitian ini diambil secara random 3 kecamatan yang ada di Kabupaten Mandailing Natal dengan kategori kecamatan yang maju, kecamatan yang menengah dan kecamatan yang rendah, hal ini ditentukan berdasarkan Indeks Pembangunan Manusia Indonesia. Berdasarkan hal itu terdapat 3 kecamatan yang dipilih yaitu masing–masing kecamatan diambil 3 kelurahan dimana setiap kelurahan di ambil 30 orang, dengan pembagian 15 laki – laki dan 15 perempuan sehingga dalam satu kecamatan jumlah sampel terdapat 30 orang dan jika ada 3 kecamatan maka terdapat 90 orang sampel dalam penelitian ini. Hasil penelitian ini adalah : Indeks ruang terbuka kabupaten Mandailing Natal diperoleh 0,221 dengan kategori rendah. Indeks SDM kabupaten Mandailing Natal diperoleh 0,00081 dengan kategori rendah. Indeks partisipasi masyarakat kabupaten Mandailing Natal diperoleh 0.0011 dengan kategori rendah. Indeks kebugaran kabupaten Mandailing Natal diperoleh 0,134 dengan kategori rendah. Kesimpulan dalam penelitian ini Indeks Pembangunan Olahraga Kabupaten Mandailing Natal rendah. Rendahnya kesempatan untuk beraktivitas olahraga disebabkan oleh semakin berkurangnya lapangan dan fasilitas untuk berolahraga, serta kesadaran SDM terhadap kesehatan, lemahnya koordinasi lintas lembaga dalam hal penyediaan fasilitas umum untuk lapangan dan fasilitas olahraga bagi masyarakat umum dan tempat pemukiman. Pariwisata mempengaruhi munculnya pengusaha-pengusaha diberbagai sektor pendukung kegiatan wisata, seperti UMKM guna untuk memenuhi kebutuhan wisatawan.Salah satu UMKM yang ada di Desa Gumantar adalah UMKM Molah Mudi dengan jenis produk berupa kopi khas Gumantar, kripik pisang, kripik nangka, kripik apel, kripik bongol pisang dan krupuk jambu mete, namun mengalami penurunan jumlah produksi, pada tahun 2018 rutin memproduksi produk setiap bulannya dengan jumlah yang stabil, kemudian pada tahun 2019 hanya memproduksi sebanyak 60 bungkus kopi dan tahun 2020 hanya memproduksi produk sesuai dengan perintaan konsumen. Penelitian ini bertujuan untuk mengetahui hubungan kompetensi SDM terhadap produktivitas UMKM Molah Mudi di wisata Adat Desa Gumantar.Metode penelitian yang digunakan adalah metode kuantitatif dengan jenis penelitian observasional. Populasi sebanyak 21 orang. Hasil penelitian ditemukan bahwa42.8% SDM tidak tahu tentang produksi produk, 47.6% SDM tidak terampil dan 38.2% mampu dalam memperoduksi produk 42.9% SDM tidak kompeten dan 61.9% SDM tidak produktiv. Ada hubungan pengetahuan (knowledge) SDM, keterampilan (skill) SDM dan kemampuan (ability) SDM dengan produktivitas UMKM Molah Mudi di wisata Adat Desa Gumantar dengan nilai P value lebih kecil dari nilai sig 0.05. sehingga dapat disimpulkan bahwa Ada hubungan kompetensi SDM dan produktivitas UMKM Molah Mudi di wisata Adat Desa Gumantar dengan nilai P value 0.025 lebih kecil dari nilai sig 0.05

Jenis penelitian yang digunakan dalam penelitian ini adalah jenis penelitian deskripsi kualitatif. Pengumpulan data dilakukan melalui observasi, wawancara dan dokuemntasi. Wawancara dilakukan kepada sejumlah informan yang telah ditentukan yaitu yang memiliki kompetensi yang berkaitan dengan tujuan penelitian. Hasil penelitian menyimpulkan, bahwa Perencanaan Kegiatan yang dilakukan oleh Pemerintah Desa belum maksimal dilaksanakan sebagaimana mestinya karena tidak melibatkan masyarakat miskin dalam menentukan skala prioritas usulan yang akan dikerjakan; Sumber Daya Manusia (SDM) dan keahlian yang dimiliki oleh Tim Pelaksana Kegiatan Desa (TPKD) belum memenuhi syarat menjadi pelaksana kegiatan Desa; Pemanfaatan Sumber daya alam (SDA) serta pemanfaatan tenaga kerja lokal oleh pemerintah desa dalam pembangunan infrastruktur berupa pembangunan jalan belum dilaksanakan. Abstract:

Background:

Companies of today commonly search to gain competitive advantages throughout different forms of co-operation, one of which is referred to as Supply Chain Management. Although little research has been assigned to the topic of how to manage and control this type of relation, lately a growing number of academics has been arguing that ABC is an appropriate mean of controlling this type of relationships.

Purpose:

The purpose of this thesis is to investigate to what degree the ongoing debate on the use of ABC as a mean of control within SCM correspond to the present theories related to ABC and SCM respectively.

Research method:

The thesis takes it’s starting point within the literature, meaning that the empirical information consists of published articles in the ongoing debate. This information is put in relation to the original theories concerning both topics.

Conclusions:

The results shown in the thesis in some content agrees with the ongoing debate referring to the positive aspects that could result from using ABC as a mean of control within SCM. However the main result of the study is the discovery of a lack of discussion concerning the problems that implementing ABC within SCM might implicate, a fact that we believe contributes to the mainly positive view on using ABC within Supply Chain Management.


Bakgrund

Företag av idag ingår i allt större utsträckning i någon form av samarbete för att på så vis skapa sig konkurrensfördelar på marknaden, en av dessa samarbetsformer går under beteckningen Supply Chain Management. Hur styrningen i denna typ av relation bör se ut är ett relativt outforskat ämne, dock finns det i dagens forskningsdebatt flera forskare som förespråkar ABC-kalkylering som ett lämpligt styrinstrument för denna typ av relationer.

Syfte

Syftet med uppsatsen är att undersöka hur väl den pågående forskningsdebatten gällande användandet av ABC som styrverktyg inom SCM överensstämmer med befintlig teoribildning inom de respektive forskningsområdena.

Genomförande

Studien tar sin utgångspunkt i litteraturen, med detta avses att den empiri som studeras består av publicerade artiklar i den aktuella debatten. Informationen ställs sedan i relation till den existerande teoribildningen inom ABC respektive SCM.

Slutsats

De resultat som framkommer ur studien visar på att den aktuella debatten i vissa avseenden har rätt i de positiva effekter ett ABC-system skulle kunna generera inom SCM. Men vad studien huvudsakligen kommer fram till är att det i forskningsdebatten saknas en ordentlig problematisering av ämnet, vilket enligt oss också förklarar den övervägande positiva syn som föreligger till användandet av ABC inom SCM.

Abstract:
Les protéines de la membrane plasmique jouent un rôle fondamental dans la détection des informations véhiculées par le milieu extracellulaire et l’adaptation des cellules aux variations de l’environnement. Elles font l’objet d’une régulation fine qui permet de moduler leur présence à la membrane et de contrôler les voies de signalisation en aval. Dans ce contexte, les arrestines qui constituent une superfamille de protéines adaptatrices, se sont imposées comme des régulateurs clés depuis la découverte des β-arrestines et arrestines visuelles, spécifiques des eucaryotes supérieurs, et de leur rôle dans la régulation des récepteurs couplés aux protéines G hétéro-trimériques, jusqu’à l’identification plus récente de nouveaux membres apparentés, présents des mammifères jusqu’aux protistes, et partageant un rôle commun de régulation de cargos membranaires. Ce travail de thèse porte sur la caractérisation fonctionnelle de deux représentants du clan arrestine de l’amibe Dictyostelium discoideum, les protéines AdcB et AdcC. Ces deux protéines partagent une même organisation multimodulaire, spécifique aux Dictyostélides, qui associe au cœur arrestine, un domaine putatif C2 de type calcium-binding et deux modules SAMs, respectivement aux extrémités N- et C-terminales des protéines. Nous avons établi que ces domaines apportent des fonctions spécifiques à ces arrestines en leur conférant la capacité de lier des lipides anioniques in vitro en réponse au calcium à travers leur module C2, et de former des structures homo- et hétéro-oligomériques via leurs domaines SAMs. En dépit de ces similarités, AdcB et AdcC présentent un comportement différent in cellulo dans la mesure où seul AdcC transloque à la membrane plasmique en réponse à une élévation du calcium cytosolique, provoquée par la stimulation des cellules par les chimioattractants AMPc et acide folique ou le calcium lui-même. Integral proteins of the plasma membrane play a major role in the detection of environmental cues and in the adaptation of cells to variations of their environment. Regulatory mechanisms modulate their presence at the cell surface and control the signaling cascades activated in response to their stimulation. In this context, members of the arrestin revealed to be key regulators, since the discovery of β- and visual arrestins and their well-described role in the regulation of G-protein coupled receptors in complex organisms, and the more recent identification of arrestin-related proteins, present from mammals to protists and sharing functions in membrane cargo trafficking. This work aims at the functional characterization of two arrestin-related proteins of the social amoeba Dictyostelium discoideum, the AdcB and AdcC proteins. These two members of the arrestin clan share a similar multimodular organization, specific to Dictyostelids, with a putative N-terminal calcium-binding type C2 domain and two C-terminal SAM domains surrounding the arrestin module. We showed that the C2 domain confers calcium-dependent binding properties to anionic lipids in vitro and that the SAM domains allow the self-association and hetero-interaction of the two proteins in complexes of high molecular weight. Despite these similarities, AdcB and AdcC harbor a distinct behavior in vivo as only AdcC translocates to the plasma membrane in response to an intracellular calcium rise triggered by the chemoattractants acid folic and cAMP or extracellular calcium. In parallel, a phenotypic characterization of adcB and adcC single or double null mutants and a search for partners were conducted, that open new avenues for future research on these adaptor proteins

Dans le but d’améliorer la disponibilité des services fournis par un récepteur, la conception d’un récepteur GNSS permettant de recevoir plusieurs signaux de toutes les bandes simultanément semble être la solution. Une architecture à sous échantillonnage RF optimisée de type SDR (Software Defined Radio) comportant un étage RF intégrable et reconfigurable et un étage de traitement numérique avec une implémentation logicielle du traitement en bande de base est défini pour ce récepteur GNSS, tout en répondant aux exigences des spécifications des standards GNSS : des réseaux radio cellulaires : GPS, Glonass, Galileo, Beidou. Un choix des composants discrets suite au dimensionnement system est effectué et ceci pour installer un prototype de validation expérimental. Ensuite nous nous s’intéressons à la caractérisation de la chaine RF afin d’étudier les limitations causés par la non linéarité et d’étudier la stabilité du prototype proposé. Un étage de traitement numérique des signaux IF, capturés à la sortie de l’ADC, est implémenté sous Matlab. In order to improve the availability of services provided by a receiver, designing a GNSS receiver to collect multiple signals from all bands simultaneously seems to be the solution. An optimized software-defined RF (SDR) sub-sampling architecture with an integral and reconfigurable RF stage and a digital processing stage with a software implementation of the baseband processing is defined for this GNSS receiver, while meeting the requirements GNSS standards specifications: cellular radio networks: GPS, Glonass, Galileo, Beidou. Many discrete components are selected after system dimensioning. Thus, experimental validation prototype is installed. Then we are interested in the characterization of the RF front-end in order to determine the limitations caused by the nonlinearity and to study the stability of the proposed prototype. A stage of digital processing of the IF signals, captured at the ADC output, is implemented under Matlab software. Chez les spores de bactéries, le photoproduit le plus abondant formé dans l'ADN irradié par les UV est un dimère de thymines appelé Photoproduit des spores (SP, 5-(a-thyminyl)-5,6-dihydrothymine). Au début de la germination, ce photoproduit est spécifiquement réparé par une enzyme, la Spore Photoproduct Lyase (SPL), régénérant les deux résidus thymine originaux. Cette enzyme est une protéine Fe-S qui appartient à la famille des « Radical-SAM ». Les protéines de cette famille d'enzymes possèdent un centre [4Fe-4S], coordiné par 3 cystéines conservées organisées selon le motif CxxxCxxC, et utilisent la SAdénosylméthionine comme cofacteur. Elles fonctionnent toutes selon un mécanisme radicalaire, initié par la formation du radical 5'-désoxyadénosyle issu de la coupure homolytique de la S-Adénosylméthionine par le centre [4Fe-4S] réduit. Dans ce travail, nous avons effectué une caractérisation biochimique et spectroscopique des SPL de Clostridium acetobutylicum et Bacillus subtilis. Par ailleurs, nous avons synthétisé un substrat minimum sous la forme d'un dinucléoside monophosphate appelé SPTpT, pour lequel une caractérisation structurale par RMN a été réalisée. Le SPTpT est reconnu et efficacement réparé par l'enzyme, ce qui nous a permis d'obtenir de nouvelles informations sur le mécanisme enzymatique de réparation. Enfin, la séquence primaire des SPL contient une 4e cystéine conservée, essentielle à la réparation, mais qui n'est pas impliquée dans la coordination du centre [Fe-S]. The major photoproduct of UV-irradiated DNA from bacterial spores is 5-(a-thyminyl)-5,6-dihydrothymine, a unique thymine dimer called Spore Photoproduct (SP). At the beginning of germination, this photoproduct is specifically repaired by the Spore Photoproduct Lyase (SPL), regenerating the two thymine monomers. SPL belongs to a family of iron-sulfur enzymes named « Radical-SAM ». In this family, proteins have a [4Fe-4S] cluster, chelated by three cysteines highly conserved constituting the “Radical-SAM” motif CxxxCxxC, and use S-Adenosylmethionine as a cofactor. They all use a radical based mechanism, initiated by 5'-deoxyadenosyl radical formation, derived from the homolytic cleavage of S-Adenosylmethionine by the reduced [4Fe-4S] cluster. Here, we have characterized, by biochemical and spectroscopic methods, the recombinant SPL from Clostridium acetobutylicum or Bacillus subtilis. Furthermore, we have synthesized a novel substrate called SPTpT under the form of a dinucleoside monophosphate. The structural characterization of this compound, using NMR spectroscopy, has been carried out. Moreover, this substrate is recognized and efficiently repaired by this enzyme, providing a convenient way to investigate the repair mechanism. Finally, the sequence of all SPL contains a fourth well-conserved cysteine. Site-directed mutagenesis shows that this residue is strongly involved in the repair mechanism but not in the cluster formation. The role of this cysteine 141 in the catalytic mechanism has been studied

Several studies have shown insufficient security in air traffic communication. The increase in air traffic in recent years also increases the need for improved security. CPDLC is used to communicate in text over the data link and ADS-B determines the position of an aircraft. These techniques are looked upon and simulated in the thesis. By proving that ADS-B and CPDLC messages can be transmitted and received with cheap and easy access equipment we want to show the simplicity in performing an attack. The thesis state a foundation to simulate attacks in the future so further work concerning the lack of security can be done.

Il existe un certain nombre de situations au cours desquelles l'intégrité du génome cellulaire est mise en danger. Ceci se produit notamment lors de mouvements de gènes (translocation, éléments mobiles), lors d'infections virales parfois intégratives (AAV, HBV, HPV) ou lors d'infections rétrovirales. En effet, le cycle de réplication des rétrovirus nécessite une étape d'intégration du génome viral dans l'ADN génomique de la cellule infectée. Malgré les nombreuses études menées sur les infections par le VIH, les cancers viro-induits ou non et les thérapies géniques basées sur les rétrovirus, aucune donnée n'est actuellement disponible sur les modifications cellulaires induites par de telles perturbations chromosomiques. L'objectif du projet était d'identifier des mécanismes cellulaires induits par des atteintes à l'intégrité du génome, plus précisément par l'insertion de molécules d'ADN non cellulaire dans les chromosomes. L'intégration de matériel génétique additionnel a été induite par des vecteurs lentiviraux dérivés du VIH-1. Les modifications cellulaires uniquement dues à l'intégration ont été isolées par comparaison de vecteurs intégratif et non intégratif. Des cellules primaires du derme humain ont été sélectionnées pour l'étude. Les temps post infection et les doses virales les plus adéquates ont été sélectionnés grâce à des expériences de cinétiques d'intégration couplées à une quantification des ADN viraux intégrés. L'analyse des modifications cellulaires induites par l'intégration de l'ADN étranger a portée sur l'ensemble du transcriptome et sur l'ensemble du protéome cellulaire. Dans le but d'effectuer une analyse transcriptomique, des puces à ADN, représentant le génome humain complet, ont été utilisées. Cette étude a démontré une forte répression transcriptionnelle induite par l'intégration. De plus, toutes les fonctions cellulaires sont perturbées par le processus. Finalement, une classification par interactions et fonctions biologiques a mis en évidence cinq processus cellulaires majoritairement affectés par l'intégration de l'ADN étranger, qui correspondent au cycle et à la mort cellulaire, au remodelage et à la réparation de la chromatine et à la réponse immunitaire ou au stress. Dans le but de compléter cette analyse transcriptomique, une étude protéomique a été réalisée. Les protéines cellulaires ont été séparées sur des gels bi-dimensionnels. Parmi les neuf protéines identifiées en spectrométrie de masse, certaines appartiennent au cytosquelette et d'autres aux mécanismes de réponse au stress. L'intégration d'un ADN étranger au sein du génome provoque donc bien des perturbations cellulaires. L'intégration de matériel génétique additionnel ne concernant pas seulement les rétrovirus, les données obtenues lors de cette étude pourront permettre (i) de développer des stratégies de défenses contre les rétrovirus ou contre les autres maladies caractérisées par des atteintes à l'intégrité du génome, (ii) d'évaluer les risques encourus par l'intégration d'un vecteur thérapeutique dans le génome cellulaire lors des thérapies géniques ou des expériences de transfert de gènes.

Abstract:
A detailed row by row investigation of major lateral tooth cusp mineralisation, together with the concomitant development of the superior epithelial tissue surrounding the teeth of the chiton Acanthopleura hirtosa has been undertaken using a combination of light microscopy, and scanning and transmission electron microscopy. A holistic approach has been adopted that encompasses observations over a range of spatial scales, from whole radula mineralisation processes to those occurring within individual tooth cusps at various stages of development. In addition, mineralisation in radulae from freshly collected animals has been compared to that of animals maintained for extensive periods within a newly developed iron limited system, which restricts radula mineralisation without impeding the formation of the organic matrix. An evaluation of the iron limitation technique has revealed that maintaining specimens of A. hirtosa within an iron poor environment results in a significant departure from the normal pattern of mineralisation in these animals. As a consequence of iron limitation, there is an obvious increase in the number of unmineralised tooth rows in addition to associated alterations in structure and composition at all stages of tooth development. In normal specimens of A. hirtosa, the onset of mineralisation in the tooth cusps occurs following the prior accumulation of iron at the junction zone and the sudden accumulation of iron containing granules in the cusp epithelium at tooth row 13. The superior epithelium surrounding the tooth cusps undergoes a series of developmental changes leading up to, and following, the onset of mineralisation. In particular, the abundance of mitochondria within the apical cusp epithelium increases, presumably in order to provide the ideal conditions of pH, and thus solubility, needed for the supersaturation of iron and its nucleation at row 13. Once mineralisation has commenced, the microvilli attached to the cusps develop rapidly, and are suggested to do so in order to facilitate the transport of iron, and thereby ensure that a high concentration gradient of this element into the cusps is maintained. The delivery of iron into the cusps occurs from two fronts, the first from the superior epithelium via the posterior surface, and the second from the junction zone via an internal pathway situated along the lepidocrocite boundary between the magnetite and core regions of the tooth. The existence of a plume of elements between this internal mineralisation pathway and the junction zone, provides the first direct evidence that the junction zone is involved in the storage and release of elements for cusp mineralisation. Data from iron limited radulae also indicate that iron continues to be deposited at the junction zone in preference to the superior epithelium or cusps, despite the disruption of mineralisation, highlighting the importance of this region in the mineralisation process. Iron reinstatement experiments have also shown that the internal pathways of iron delivery within the organic matrix remain viable, despite prolonged periods of iron limitation. In addition, the reinstatement of iron has revealed that the plumes, situated between the junction zone and internal mineralising pathway of the cusp, stem from the centre of the plate like junction zone, directly above the stylus canal, a tube like cavity situated within the styli of each major lateral tooth. An in depth study of the stylus canal has revealed that cells within the canal are remarkably similar to those of the epithelium surrounding the cusps, suggesting that this structure may also be involved in the delivery of ions to the junction zone. The stylus canal is shown to be present in the major lateral tooth cusps of 38 chiton species distributed worldwide, and is therefore likely to be a feature common to all chitons. The presence of the canal, and indeed its absence from the bases of all remaining non iron mineralised teeth, irrespective of chiton species, also points strongly to a functional relationship between the stylus canal and tooth cusp mineralisation.

Electrochemical behavior of electrolytic manganese dioxide (EMD), chemically prepared battery grade manganese dioxide (BGM), titanium dioxide (TiO2), lithium iron phosphate (LiFePO4) and lithium manganese phosphate (LiMnPO4) in aqueous lithium hydroxide electrolyte has been investigated. These materials are commonly used as cathodes in non-aqueous electrolyte lithium batteries. The main aim of the work was to determine how the electroreduction/oxidation behavior of these materials in aqueous LiOH compares with that reported in the literature in non-aqueous electrolytes in connection with lithium batteries. An objective was to establish whether these materials could also be used to develop other battery systems using aqueous LiOH as electrolyte. The electrochemical characteristics of the above materials were investigated by subjecting them to slow scan cyclic voltammetry and determining the charge/discharge characteristics of Zn/cathode material-aqueous LiOH batteries. The products of electroreduction/oxidation were characterized by physical techniques using X-ray diffraction (XRD), scanning electron micrography (SEM), X-ray photoelectron spectroscopy (XPS), secondary ion mass spectrometry (SIMS), Thermogravimetric analysis (TG) and infra-red spectrometry (IR). The reduction of ã-MnO2 (EMD) in aqueous lithium hydroxide electrolyte is found to result in intercalation of Li+ into the host structure of ã-MnO2. The process was found to be reversible for many cycles. This is similar to what is known to occur for ã-MnO2 in non-aqueous electrolytes. The mechanism, however, differs from that for reduction/oxidation of ã-MnO2 in aqueous potassium hydroxide electrolyte. KOH electrolyte is used in the state-of-art aqueous alkaline Zn/MnO2 batteries. Alkaline batteries based on aqueous KOH as the electrolyte rely upon a mechanism other than K+ intercalation into MnO2. This mechanism is not reversible. This is explained in terms of the relative ionic sizes of Li+ and K+. The lithium-intercalated MnO2 lattice is stable because Li+ and Mn4+ are of approximately the same size and hence Li+ is accommodated nicely into the host lattice of MnO2. The K+ ion which has almost double the size of Li+ cannot be appropriately accommodated into the host structure and hence the K+ -intercalated MnO2 phase is not stable. Chemically prepared battery grade MnO2 (BGM) is found to undergo electroreduction/oxidation in aqueous LiOH via the same Li+ intercalation mechanism as for the EMD. While the Zn/BGM- aqueous LiOH cell discharges at a voltage higher than that for the Zn/EMD- aqueous LiOH cell under similar conditions, the rechargeability and the material utilization of the BGM cell is poorer. The cathodic behavior of TiO2 (anatase phase) in the presence of aqueous LiOH is not reversible. In addition to LiTiO2, Ti2O3 is also formed. The discharge voltage of the Zn/TiO2- aqueous LiOH cell and material utilization of the TiO2 as cathode are very low. Hence TiO2 is not suitable for use in any aqueous LiOH electrolyte battery. LiFePO4 (olivine-type structure) as a cathode undergoes electrooxidation in aqueous LiOH forming FePO4. However the subsequent reduction forms not only the original LiFePO4 but also Fe3O4. Thus the process is not completely reversible and hence LiFePO4 is not a suitable material for use as a cathode in aqueous battery systems. LiMnPO4 (olivine-type structure) undergoes reversible electrooxidation in aqueous LiOH forming MnPO4. The charge/discharge voltage profile of the Zn/MnPO4-aqueous LiOH cell, its coulombic efficiency and rechargeability are comparable to that of the cell using ã-MnO2. EMD and LiMnPO4 both have the potential for use in rechargeable batteries using aqueous LiOH as the electrolyte. Recommendations for further developmental work for such batteries are made.

AbstractThe purpose of this study is to investigate the factors that contribute to the decline in employee performance at ABC Company, specifically work ability, organizational culture, work environment, and motivation. This type of research employs quantitative methods in applied business research. The sample size was 58 ABC company employees who are still active and have worked for the company for at least three years. To test the hypotheses in this study, a PLS-based SEM approach was used. PLS testing was done in two stages: measurement model test and structural model test. A measurement model test was performed to assess the validity and reliability of each indicator’s constructs. Meanwhile, the structural model test was performed to see if there is an influence between variables/correlations between constructs as assessed by the PLS t-test. This study demonstrates that work ability has a significant impact on employee performance. In the meantime, organizational culture, work environment, and motivation have no effect on employee performance. Motivation cannot mediate the relationship between employee performance and work ability, organizational culture, and work environment.

AbstractCell-free protein synthesis (CFPS) enables the development of antibody conjugates, such as fluorophore conjugates and antibody-drug conjugates (ADCs), in a rapid and straightforward manner. In the first part, we describe the cell-free synthesis of antibodies containing fluorescent non-canonical amino acids (ncaa) by using pre-charged tRNA. In the second part, we describe the cell-free synthesis of antibodies containing ncaa by using an orthogonal system, followed by the site-specific conjugation of the fluorescent dye DyLight 650-phosphine. The expression of the antibodies containing ncaa was analyzed by SDS-PAGE, followed by autoradiography and the labeling by in-gel fluorescence. Two different fluorescently labeled antibodies could be generated.

Abstract Equipment manufacturers have developed peripherals for their tools that add soft defect localization (SDL) capability to existing optical beam tools, in many cases providing excellent results. However, these upgrades add significant cost to the tool. This paper presents the design considerations for a simple adapter that was developed in house to add SDL capability to optical beam induced resistance change (OBIRCH) tool, including resolution of some unexpected problems. This solution represents a simple, low cost method to add SDL testing capability to the OBIRCH tool and can also be used in conjunction with OBIC and XIVA tools with little or no modification. An early example of the SDL results provided by this adapter is also presented.

En levande landsbygd kräver verksamheter som kompletterar lantbruket. Odling av flodkräfta i dammar skulle kunna vara en mycket större verksamhet än vad som nu är fallet. Om det dessutom utvecklas en odlingsmetodik i växthusmiljöer skulle flodkräfta kunna odlas i betydligt större omfattning, inte minst för att ta fram livskraftiga yngel för utplantering. En sådan verksamhet skulle då kunna bedrivas även i de kallare delarna av landet, vilket inte är möjligt idag eftersom flodkräftans naturliga reproduktion begränsas av de kalla somrarna. Vår stora konsumtion av kräftor gör att importbehovet är omfattande. Vi importerar varje år frysta och kokta kräftor till Sverige till ett värde av närmare 450 Mkr. Den årliga konsumtionen av kräftor i landet uppgår till ungefär ½ kg per person och år, vilket motsvarar 5 000 ton. Det säljs kräftor som är fångade i Sverige (huvudsakligen signalkräfta) för ca 300 Mkr per år. Yrkesfisket på signalkräfta i de stora sjöarna omsätter ca 25 Mkr per år. Sammantaget importeras drygt hälften av de kräftor vi äter i Sverige. Eftersom det är förbjudet enligt svensk lag att importera levande kräftor, även för att hålla i akvarier, är odling och leverans av färska flodkräftor inom landet en framtidsverksamhet. Det som saknats tidigare är framförallt en saklig och heltäckande handledning hur en flodkräftodling kan etableras och hur bästa resultat fås ur odlingsverksamheten. Ett EU-beslut som gäller signalkräfta stärker dessutom våra ambitioner att slå vakt om flodkräftan. Beslutet innebär att signalkräftan finns på EU:s svarta lista över invasiva arter och signalkräfta får därmed inte spridas. EU:s förordning om invasiva främmande arter gäller från 1 januari 2016. Därmed finns det ytterligare anledning att gynna odling av flodkräfta i Sverige framöver. Våra ambitioner att bevara och utveckla förekomsten av denna rödlistade art, som flodkräfta utgör, kräver att odling i småvatten utökas. I det nationella åtgärdsprogrammet som syftar till att bevara flodkräfta lyfts odling av flodkräfta i dammar fram som en viktig åtgärd för artens bevarande. Detta faktum delas också med våra grannländer Finland, Danmark och Norge, där flodkräfta, precis som i Sverige är hotad av kräftpest, som främst sprids av signalkräftor. Denna handledning kan användas för att närmare undersöka möjligheterna att etablera en kräftodling och innehåller ett antal steg som leder fram till hur odlingen kan utformas. Den ger även information och stöd för de som kommit igång med flodkräftodling i dammar men som vill utveckla verksamheten eller använda den som ”felsökningsmanual” i fall något inte fungerar. Denna handledning bör därför även vara av intresse för etablerade odlare av flodkräfta med kommersiell verksamhet. Handledningen är rikligt illustrerad med foton och diagram. Det finns även annan ny information att tillgå i form av ett utbildningspaket som är relevant: ”Kräftodlingens ABC – kunskapsunderlag för flodkräftodling i dammar”. Denna handledning är en vidareutveckling av Kräftodlingens ABC och omfattar även inomhusodling och växthusodling av flodkräfta samt kopplingar till flera undersökningar som gjorts inom projektet. Allt i syfte att få fram så mycket referensmaterial som möjligt som kan användas som stöd vid andra odlingar. Här finns också mer ingående information om hur fångster utvärderas för att bedriva ett hållbart fiske på kräftor. Här finns också information om produktion och förväntningar i olika typer av odlingar som kan utgöra underlag för investeringsbeslut. Projekten har därför samverkat till stor del när båda dessa nya underlag tagits fram

A pandemia de COVID-19 teve impacto profundo nos jovens na América Latina e no Caribe (ALC). Desde 2020, os jovens da ALC enfrentam muitos desafios, como adaptação aos ambientes virtuais de aprendizagem, perdas e depressão, o desemprego e muito mais, sem sinais claros de alívio. Embora as medidas sociais e de saúde pública instituídas pelos governos sejam necessárias para retardar a transmissão da COVID-19, em grande parte não consideraram as necessidades da população jovem. Com poucos apoios, os jovens tiveram que enfrentar a pandemia sozinhos. À medida que a resposta à pandemia evolui, surgem questões-chave para profissionais e governos, tais como: Que lições podem ser aprendidas a partir das perspectivas dos jovens sobre a resposta à COVID-19 até agora? E como podemos envolver melhor os jovens como parte da preparação e resposta à pandemia agora e no futuro? Este resumo baseia-se na literatura cientifica e cinzenta que explora como a COVID-19 afeta os jovens, bem como na literatura que descreve a resposta à pandemia na ALC e em outras regiões, e apresenta considerações sobre como envolver os jovens, vendo-os não apenas como uma parte da população afetada, mas também como parceiros na resposta. Destina-se ainda a orientar os atores humanitários, profissionais de saúde pública, defensores da juventude, profissionais de envolvimento da comunidade e outros envolvidos na resposta à COVID-19. Por fim, o resumo também contribui para a base de evidências existente sobre o impacto da COVID-19 nos jovens. Essas lições são úteis para fortalecer a preparação e as respostas programáticas aos surtos. Os jovens são classificados como indivíduos com idades entre os 10 e 24 anos. Principais considerações são compartilhadas para adolescentes (10-19 anos) e jovens (15-24 anos). Barbados e Brasil foram escolhidos como estudos de caso devido ao seu grande número de jovens (compreendendo pouco menos de 20% da população em ambos os países), bem como suas diferentes respostas nacionais à COVID-19, apesar de enfrentarem desafios semelhantes durante a pandemia. The shoot apical meristem establishes plant architecture by continuously producing new lateral organs such as leaves, axillary meristems and flowers throughout the plant life cycle. This unique capacity is achieved by a group of self-renewing pluripotent stem cells that give rise to founder cells, which can differentiate into multiple cell and tissue types in response to environmental and developmental cues. Cell fate specification at the shoot apical meristem is programmed primarily by transcription factors acting in a complex gene regulatory network. In this project we proposed to provide significant understanding of meristem maintenance and cell fate specification by studying four transcription factors acting at the meristem. Our original aim was to identify the direct target genes of WUS, STM, KNAT6 and CNA transcription factor in a genome wide scale and the manner by which they regulate their targets. Our goal was to integrate this data into a regulatory model of cell fate specification in the SAM and to identify key genes within the model for further study. We have generated transgenic plants carrying the four TF with two different tags and preformed chromatin Immunoprecipitation (ChIP) assay to identify the TF direct target genes. Due to unforeseen obstacles we have been delayed in achieving this aim but hope to accomplish it soon. Using the GR inducible system, genetic approach and transcriptome analysis [mRNA-seq] we provided a new look at meristem activity and its regulation of morphogenesis and phyllotaxy and propose a coherent framework for the role of many factors acting in meristem development and maintenance. We provided evidence for 3 different mechanisms for the regulation of WUS expression, DNA methylation, a second receptor pathway - the ERECTA receptor and the CNA TF that negatively regulates WUS expression in its own domain, the Organizing Center. We found that once the WUS expression level surpasses a certain threshold it alters cell identity at the periphery of the inflorescence meristem from floral meristem to carpel fate [FM]. When WUS expression highly elevated in the FM, the meristem turn into indeterminate. We showed that WUS activate cytokinine, inhibit auxin response and represses the genes required for root identity fate and that gradual increase in WUCHEL activity leads to gradual meristem enlargement that affect phyllotaxis. We also propose a model in which the direction of WUS domain expansion laterally or upward affects meristem structure differently. We preformed mRNA-seq on meristems with different size and structure followed by k-means clustering and identified groups of genes that are expressed in specific domains at the meristem. We will integrate this data with the ChIP-seq of the 4 TF to add another layer to the genetic network regulating meristem activity.

Denne rapport præsenterer en række centrale resultater af et forskningsprojekt med fokus på daginstitutioners indsatser og betydning for pædagogisk personale, børn og familier under COVID-19 pandemien i Danmark. Forskningsprojektet har indsamlet data i perioden april 2020 til august 2020, og særligt i perioden fra midt april til juni, hvor alle landets daginstitutioner genåbnede efter cirka en måneds nedlukning af landets offentlige institutioner som følge af regeringens retningslinjer. Der er ligeledes indsamlet data i perioden august til oktober 2020, og igen i perioden januar til februar 2021. Der er således tale om tre empiriske nedslag hen over cirka et år af pandemiens forløb i relation til daginstitutionsledere og pædagogisk personale i landets daginstitutioner. De data, der ligger til grund for denne rapport, omfatter en spørgeskemaundersøgelse rettet mod daginstitutionsledere og en spørgeskemaundersøgelse rettet mod pædagogisk personale i daginstitutioner, geografisk fordelt over hele landet. Fælles for spørgeskemaundersøgelserne er et fokus på at udforske, hvordan daginstitutioner har arbejdet under COVID-19 pandemien, dels under selve nedlukningen i marts-april, hvor flere af landets daginstitutioner har fungeret som nødpasning, særligt for forældre med kritiske samfundsfunktioner, og dels under genåbningsfaserne, der officielt blev påbegyndt i midten af april og især har forløbet hen over forår og tidlig sommer 2020. Derudover er der ad to omgange foretaget en række forskningsinterview med daginstitutionsledere. I første omgang blev der foretaget 10 forskningsinterview i perioden august til november 2020, og i anden omgang er der foretaget 8 forskningsinterview med daginstitutionsledere fordelt på landets daginstitutioner omfattende perioden januar til februar 2021. Formålet med dette forskningsprojekt har været at kortlægge og udforske, hvordan daginstitutionsledere og pædagogisk personale i en række danske daginstitutioner for 0-6-årige børn løfter den store samfundsmæssige udfordring at håndtere COVID-19 pandemien på landets daginstitutioner.

Many pharmaceutical compounds are active at very low doses, and a portion of them regularly enters municipal sewage systems and wastewater-treatment plants following use, where they often do not fully degrade. Two such compounds, CBZ and LTG, have been detected in wastewater effluents, surface waters, drinking water, and irrigation water, where they pose a risk to the environment and the food supply. These compounds are expected to interact with organic matter in the environment, but little is known about the effect of such interactions on their environmental fate and transport. The original objectives of our research, as defined in the approved proposal, were to: Determine the rates, mechanisms and products of photodegradation of LTG, CBZ and selected metabolites in waters exposed to near UV light, and the influence of DOM type and binding processes on photodegradation. Determine the potential and pathways for biodegradation of LTG, CBZ and selected metabolites using a white rot fungus (Pleurotusostreatus) and ADP, and reveal the effect of DOM complexation on these processes. Reveal the major mechanisms of binding of LTG, CBZ and selected metabolites to DOM and soil in the presence of DOM, and evaluate the effect of this binding on their photodegradation and/or biodegradation. We determined that LTG undergoes relatively slow photodegradation when exposed to UV light, and that pH affects each of LTG’s ability to absorb UV light, the efficiency of the resulting reaction, and the identities of LTG’sphotoproducts (t½ = 230 to 500 h during summer at latitude 40 °N). We observed that LTG’sphotodegradation is enhanced in the presence of DOM, and hypothesized that LTG undergoes direct reactions with DOM components through nucleophilic substitution reactions. In combination, these data suggest that LTG’s fate and transport in surface waters are controlled by environmental conditions that vary with time and location, potentially affecting the environment and irrigation waters. We determined that P. ostreatusgrows faster in a rich liquid medium (glucose peptone) than on a natural lignocellulosic substrate (cotton stalks) under SSF conditions, but that the overall CBZ removal rate was similar in both media. Different and more varied transformation products formed in the solid state culture, and we hypothesized that CBZ degradation would proceed further when P. ostreatusand the ᵉⁿᶻʸᵐᵃᵗⁱᶜ ᵖʳᵒᶠⁱˡᵉ ʷᵉʳᵉ ᵗᵘⁿᵉᵈ ᵗᵒ ˡⁱᵍⁿⁱⁿ ᵈᵉᵍʳᵃᵈᵃᵗⁱᵒⁿ. ᵂᵉ ᵒᵇˢᵉʳᵛᵉᵈ ¹⁴C⁻Cᴼ2 ʳᵉˡᵉᵃˢᵉ ʷʰᵉⁿ ¹⁴C⁻ᶜᵃʳᵇᵒⁿʸˡ⁻ labeled CBZ was used as the substrate in the solid state culture (17.4% of the initial radioactivity after 63 days of incubation), but could not conclude that mineralization had occurred. In comparison, we determined that LTG does not degrade in agricultural soils irrigated with treated wastewater, but that P. ostreatusremoves up to 70% of LTG in a glucose peptone medium. We detected various metabolites, including N-oxides and glycosides, but are still working to determine the degradation pathway. In combination, these data suggest that P. ostreatuscould be an innovative and effective tool for CBZ and LTG remediation in the environment and in wastewater used for irrigation. In batch experiments, we determined that the sorption of LTG, CBZ and selected metabolites to agricultural soils was governed mainly by SOM levels. In lysimeter experiments, we also observed LTG and CBZ accumulation in top soil layers enriched with organic matter. However, we detected CBZ and one of its metabolites in rain-fed wheat previously irrigated with treated wastewater, suggesting that their sorption was reversible, and indicating the potential for plant uptake and leaching. Finally, we used macroscale analyses (including adsorption/desorption trials and resin-based separations) with molecular- level characterization by FT-ICR MS to demonstrate the adsorptive fractionation of DOM from composted biosolids by mineral soil. This suggests that changes in soil and organic matter types will influence the extent of LTG and CBZ sorption to agricultural soils, as well as the potential for plant uptake and leaching.
