Abstract:
Scurrula atropurpurea known as benalu is a medicinal plant that has been used for the treatment of various diseases such as antibacterial. Plants with ethnomedicine history use to cure pathogenic bacterial infections and their endophytic fungi is a promising source of antibacterial compounds. This study aimed to compared the antibacterial activity of S. atropurpurea leaves and their endophytic fungi. The secondary metabolites were isolated from the leaves of S. atropurpurea and their endophytic fungi by the chromatography method. The antibacterial activity test was carried out by Kirby Bauer method against Salmonella typhi (IPCCCB.11.669) and Escherichia coli (ATCC 25922) as Gram (-) and Bacillus subtilis (ATCC 6633) and Staphylococcus aureus (ATCC 25923) as Gram (+). The antibacterial compound from S. atropurpurea was determined by spectroscopy analysis as Quercetin-3-O-α-L-Rhamnopyranoside, while the antibacterial compound from endophytic fungi (strain BB1) as a lactone. Phylogenetic tree of strain BB1 has the highest homology with Neopestalotiopsis surinamensis strain CBS 450.74.

Abstract:
Objective: The aim of this study is to investigate antioxidant activity and phytochemical screening of ethanol extract, fractions of water, ethyl acetate, and n-hexane from mistletoe tea (Scurrula atropurpurea Bl. Dans).Methods: Simplicia extracted using soxhlet equipment with 96% ethanol. Fractionation was conducted using liquid-liquid extraction using a solvent of water, ethyl acetate and n-hexane. Screening of phytochemical and antioxidant activity was performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method using ultraviolet-visible spectrophotometry with ascorbic acid as standard. Phytochemical screening was conducted based on the method of Farnsworth.Results: The IC50 values of ethanol extract, water fraction, fraction of ethyl acetate, and n-hexane fraction were 21.92 ppm, 89.57 ppm, 14.08 ppm, and 162.09 ppm, respectively, whereas for ascorbic acid was 4.41 ppm. The ethanol extract and ethyl acetate fraction contained compounds were the same group, polyphenolic, tannins, flavonoids, monoterpenoid, steroids, triterpenoids, and quinones. Fraction of water contained compounds such as polyphenolic group, flavonoids, monoterpenoids, sesquiterpenoids, steroids, and triterpenoids. n-hexane fraction compounds contained steroids and triterpenoids.Conclusion: The ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction showed antioxidant activities. Benalu is one of the parasitic plants which have ataccked many collection plants species in Eka Karya Botanical Garden, Bali. Exploration and collection of these parasitic plants in this area are conducted. Four parasitic plants species, i.e. Dendrophthoe pentandra, Helixanthera cylindrica, Scurrula atropurpurea, and S. parasitica are recorded and they attack 32 collection plants species in Eka Karya Botanical Garden. Dendrophthoe pentandra is reported as the highest population species to parasiting collection plants species. While the Myrtaceae family and Syzygium genera are also reported as the highest parasited species.

Hypertension is a degenerative disease that become health problem in the world. S. atropurpurea BI. Dans and D. petandra are herbs that showed anti-hypertension activity. However, the potency of their combination onimmune system in hypertension condition have not been explored completely. The study aim to examine effect of S. atropurpurea BI. Dans and D. petandra combination on Cluster of Differentiation 4 (CD4), Cluster of Differentiation 8(CD8), interleukin-10 (IL-10), and tumor necrosis factor-α (TNF- α) level in hypertension rats. The rat was administrated orally with methanolic extract of S. atropurpurea BI. Dans and D. petandra (1:3) combination at dose of 50 mg/kgBW (P1), 100 mg/kgBW (P2) and 200 mg/kgBW (P3) for four weeks. Hypertension rats are induced with Deoxycorticosterone acetate (DOCA) and salt orally at the second weeks after herbs administration. Blood sample was collected from intra cardiac, furthermore, the level of IL-10 and TNF-α levels are examined using the Enzyme-linked immunosorbent assay (ELISA) method, whereas CD4 and CD8 levels are examined using flow cytometri. The data was analyzed using ANOVA test continued with LSD test (p < 0,05). The treatment groups (PI, P2, and P3) showed an increase of CD4 levels more and less 3 times,10 times, and 8 times respectively; an increase of CD8 level about 2 times, 3 times, and 2 times respectively compare to hypertension group (p<0.05). However, IL-10 level were increased only by P3 group (p<0.05). Meanwhile, the combination of herbsdecreased TNF-α level approximately 50-60% for three doses used (p<0.05). Combination of methanolic extracts of S. atropurpurea BI. Dans and D. petandra increase CD4, CD8 and IL-10 level and decreased TNF-α level on hypertension rat.

Abstract Mistletoes are hemiparasitic plants growing in the wild and cultivation plants such as sengon (Falcataria moluccana). Research aimed at inventing the diversity and infestation of mistletoes was conducted in cultivation areas of sengon in Malang Regency East Java Indonesia from September – December 2021. Data on mistletoes and their hosts was collected by exploratory and descriptive methods in ten districts and twenty villages. Purposive sampling was used and determined along tracks in line transects where sengon cultivation was found. Sample plots of 10x10 m were determined and made in each pattern of cultivation to enumerate the number and species of trees and mistletoes, and to measure the diameter of stems. The results showed that there were only two species of mistletoe discovered in sixteen cultivation patterns of sengon namely Scurrula atropurpurea (Blume) Danser and Viscum ovalifolium L. S. atropurpurea was the most dominant mistletoe in cultivation of sengon with parasite number of 906 individuals. The highest number of trees, infested trees, and mistletoes were obtained in the monoculture pattern of sengon. The highest severity index of mistletoe infestation was found in tangerine citrus + Sengon, i.e., 15. The average value of SI in all cultivation patterns was categorized as low, i.e., 5.28.

Mistletoes are aerial hemiparasitic plant that infesting and parasitizing on wild and cultivated plants. They are can be categorized as generalist and specialist mistletoes according to their host plants diversity. Shannon index of the host plants diversity can be used to determine host specificity of mistletoes. Study aimed to know and determine host plants and host specificity of mistletoes in Purwodadi Botanic Garden was conducted in 2013, 2017 and 2021. Data of mistletoes and their hosts species was collected by explorative and descriptive methods in 2013, 2017, and 2021. Recorded data by observation in the fields were species and species number of mistletoes and their hosts. Observation of the misteltoes used binoculars to ditermine the species of the mistletoes. Identification of the mistletoes species and their hosts was conducted directly in the garden and undirectly by making herbarium specimens and taking photographs. The results showed that there were five species of mistletoes infested 142 species, 82 genera and 36 families of host plants namely Dendrophthoe pentandra, Macrosolen tetragonus, Scurrula atropurpurea, Viscum articulatum, and Viscum ovalifolium. There were significantly different between the mistletoe species and their Shannon index of host plants . D. pentandra was the most generalist or the least specialist to host plants with the highest Shannon index value of 2.20±0.10, whereas S. atropurpurea was the least generalist or the most specialist with Shannon index value of 0.16±0.09.

Mistletoe tea is a parasitic plant that lives on its host, and this plant has potential as an herb. The tea plant parasite has several metabolites, namely alkaloids, flavonoids, terpenoids, glycosides, triterpenes, saponins, and tannins. The metabolite compounds produced by endophytic fungi have potential as herbs. Metabolite compounds are not only produced by mistletoe tea but are also produced by endophytic molds. This research aimed to isolate and characterize endophytic molds macroscopically and microscopically. This research uses an descriptive methods. Plant samples using mistletoe tea (Scurrula atropurpurea (Bl.) Dans) with the stages of sampling, making PDA media, isolation of endophytic molds, purification and macroscopic and microscopic characterization. Endophytic fungi that have been isolated were characterized by macroscopic and microscopic characterization. The results of the macroscopic and microscopic characterization research showed that seven isolates of mold were successfully isolated and characterized. Abstract:
The aim of this research was to investigate the potential effect of methanolic extract of Scurrula atropurpurea (MESA) in reducing oxidative stress through observation of superoxide dismutase (SOD) activity and malondialdehyde (MDA) concentration in mice. In this research, 25 male mice were divided into four groups with five replications. Group I (K1, without MESA) was considered as a control group, while Group II (K2), III (K3), and IV (K4) were given 1000, 2000, and 4000 mg MESA/kg BW, respectively. After 14 days, SOD activity and MDA concentration were measured spectrophotometrically. The SOD activity did not significantly increased (P>0.05) in group K1, K2, K3, and K4 with values were 19.469±2.072, 17.4±1.373, 20.53±3.411, and 23.87±2.528 U/ml, respectively. In contrast, MDA concentration was decreased (P>0.05) with values of 492.750±19.418, 492.750±19.418, 457.125±30.095, 454.625±19.080, and 475.875±37.659 ng/mL in groups K1, K2, K3, and K4, respectively. It can be concluded that the administration of MESA tend to increase SOD activity and decrease MDA concentration in this study, thus might be able to reduce oxidative stress in mice.

Mistletoe tea (Scurrula. atropurpurea (Bl.) Dans) and Mistletoe Mango (Dendrophthoe pentandra) are parasitic plants that live on tea and mango plants, very potential as medicines, containing several secondary metabolite compounds such as flavonoids (quercetin). Flavonoids can act as natural antioxidants that can protect the kidneys from free radicals. Damage to kidney function can indicate from creatinine, BUN, urea, and renal histopathology. The.purpose.of.this study was.to determine toxicity of. a combination of. Mistletoe tea. extract and mango mistletoe (EMBTBM) to damage kidney function in female Wistar rats for 28 days, using the true experimental design method. Data on creatinine, bun, urea, and renal histopathology data were analyzed using the ANOVA. test. The number of test animals is 20 female white rats divided into four groups; each group there are five rats. Group.1 a control, then groups 3, and 4. as. treatment. Based on the results of the study showed that statistically significant. between. all groups. was p> 0.05. Abstract Mistletoes are flowering plants in the Santalales order that parasitize the branches of trees and shrubs. The objective of this research was to reveal the diversity of mistletoe species and record their distribution within this study site. The study site was Dramaga Campus (240 ha), which consisted of a mosaic of tree plantation, experimental fields, buildings, and a housing complex. Field research was conducted in February-March 2021 by censusing all mistletoes, their distribution, and plant species parasitized by the mistletoes. There were 351 individual mistletoes found, consisted of 5 species: Dendrophthtoepentandra (55.2%), Viscum ovalifolium (32.3%), Scurrula atropurpurea (9.1%), Macrosolen chochinchinensis (2.8%), and Viscum articulatum (0.6%). These mistletoes were found mostly on branches or twigs (99.1%) and few on trunks (0.9%) of 49 tree species (25 families) having an average height of 14.9±4.2m, and diameter 34.11± 22.22cm. Tree species frequently parasitized were Falcataria moluccana (32.8%), followed by Samanea saman (12.5%), and Averrhoa carambola (7.7%). Most mistletoes were found in the open area (99.1%), including on trees between/among buildings and along roads/streets, mainly on the south and southeast part of the campus. The mistletoe attacked in this research location was included in the low category (79.4%).

Serangan benalu atau tumbuhan parasit tidak hanya dapat merusak estetika, namun juga menghambat pertumbuhan dan perkembangan, bahkan membunuh tanaman koleksi. Penelitian ini bertujuan untuk mengetahui keanekaragaman jenis benalu dan distribusi spasial dari sebaran benalu di Ecopark CSC-BG. Pengumpulan spesimen dan data, meliputi jenis dan jumlah benalu, jenis dan kondisi inang, habitat menggunakan metode jelajah di seluruh area ini. Ditemukan empat jenis benalu dari suku Lorantaceae, yaitu Dendrophthoe pentandra (L.) Miq., Scurrula atropurpurea (Blume) Danser, Macrosolen cochinchinensis (Lour.) Tiegh., dan Dendrophthoe sp. Benalu tersebut tersebar di bioregion Blok 1, Jawa-Bali, Kalimantan, Sumatra, dan Papua. Beberapa faktor ekofisiologi yang disarankan untuk diperhatikan dalam distribusi benalu adalah tinggi inang, ekspos cahaya pada tajuk bagian atas inang, kepadatan tanaman koleksi, keterbukaan atau ekspos cahaya matahari pada habitat. Indeks keanekaragaman (H’) dan indeks kemerataan (E’) benalu yang ditemukan menunjukkan H’ = 1,29 dan E’ = 0,93. Hasil analisis pola spasial, keberadaan benalu relatif merata di seluruh bagian Ecopark CSC-BG, dengan keragaman jenis tertinggi berada di bioregion Jawa-Bali, Sumatra, dan Kalimantan. Kesimpulan penelitian ini 4 jenis benalu yang ditemukan di Ecopark CSC-BG dengan nilai indeks keanekaragaman jenis benalu tergolong sedang dengan pola sebaran merata.AbstractThe Ecopark Cibinong Science Center-Botanic Gardens (CSC-BG), requires the right collection of plant maintenance strategies to support the health and sustainability of the collection. Attack by parasites or parasitic plants can not only destroy aesthetics, but also inhibit growth and development, and even kill the collected plants. This study aims to determine the diversity of parasite types and the spatial distribution of the distribution of parasites in Ecopark CSC-BG. Collection of specimens and data, including the type and number of parasites, types, and conditions of the host, habitat using the roaming method throughout this area. Four types of parasites were found from the Loranthaceae tribe, namely Dendrophthoe pentandra (L.) Miq., Scurrula atropurpurea (Blume) Danser, Macrosolen cochinchinensis (Lour.) Tiegh., and Dendrophthoe sp. These types of parasites are scattered in the Block 1 bioregion, Java-Bali, Kalimantan, Sumatra, and Papua. Some of the recommended ecophysiological factors to consider in the distribution of mistletoes were host height, light exposure to the upper canopy of the host, plant collection density, openness, or exposure to sunlight in the habitat. The diversity index (H') and evenness index (E') of mistletoes found were 1.29 and 0.93, respectively. The results of spatial pattern analysis, the presence of mistletoes was relatively evenly distributed throughout the garden, with the highest species diversity found in the Java-Bali, Sumatra, and Kalimantan Bioregions. This study concludes that there were 4 species of mistletoes found in Ecopark CSC-BG with a moderate diversity index and an even distribution pattern.

Abstract:
The objective of this study was to investigate the bioactivity, toxicity, and interaction of the prepared bioactive compounds from Scurrula atropurpurea and Dendrophthoe pentandra with angiotensin-converting enzyme (ACE) macromolecule via an in-silico route. The bioactivity was investigated through Way2Drug PASS Online program. The drug-likeness property and human intestinal absorption of the bioactive compounds were evaluated through absorption, distribution, metabolism, and excretion (ADME) analysis. The interaction of bioactive compounds with ACE was examined via a molecular-specific docking analysis. The crystal structure of ACE was evaluated using the protein database 1086 chain A. The results elucidated that the prepared compounds exhibited potential bioactive agents for vasodilation, vasoprotective, and cardioprotective applications based on PASS analysis. The ADME analysis also revealed that the flavonol 3-O-D-glucoside did not violate any of the Lipinski's rule and had high gastrointestinal absorption. Furthermore, the results also suggested that the flavonol 3-O-D-glucoside of mistletoes had a harmless LD50 but was not recommended to be used for a long period. The molecular docking revealed that the flavonol 3-O-D-glucoside had the lowest binding energy with a value of −8.3 kcal.mol−1, suggesting the potential inhibitor for ACE. Generally, the toxicity and molecular docking analysis showed the compounds inhibited the ACE macromolecule with a series of flavonol > kaempferol > casticin > quercetin > quercitrin > isoquercitrin. Therefore, the S. atropurpurea and D. pentandra from Indonesian natural resources open new excellent potency for traditional herbal medicines, especially to treat hypertension. Tea Benalu (Scurrula atropurpurea [Bl.] Dans.) and Mango Benalu (Dendrophthoe pentandra L. miq.) containssecondary metabolites with potential antioxidant potential. Antioxidants can inhibit the oxidation process and reduce Total Cholesterol, Triglycerides, Low-Density Lipoprotein (LDL) and increase High-Density Lipoprotein (HDL). The safety of the preparations for the combination of parasites of tea and mango leaves was tested to obtain toxic effects after repeated treatment tests over some time. The purpose of this study was to determine the toxic effects of a combination of methanolic extracts of a combination of tea parasites and mangoes in female rats (Rattus norvegicus) subchronically for 28 days by looking at the results of the clinical biochemical examination on lipid profiles, namely total cholesterol levels, triglycerides, high-density lipoproteins (28%). HDL), and Low-Density Lipoprotein (LDL). The method of this study was experimental with a combination dose of methanol extract of tea parasite leaves and mango parasite leaves at a dose of 250 mg / KgBB, 500 mg / KgBB and 1000 mg / KgBB with 5x replications each treatment. The subjects used white rats (Rattus norvegicus) female Wistar strains aged 6-8 weeks with a minimum body weight of 100 grams. The treatment in this study was in the form of each dose given 5 times a week for 28 days (subchronic toxicity test) orally. Lipid profile levels were analyzed using the one-way ANOVA test. The results showed that the level of control blood lipid profile with a treatment dose of 250 mg / KgBW, 500 mg / KgBW, and 1000 mg / KgBW was not significantly different. The toxicity test of combined extracts of tea leaves and mango parasite leaves of female rat Rattus novergicus on subchronic exposure 28 days with those doses showed no difference tangible between treatments of controls. Metode penelitian ini adalah eksperimental dengan perlakuan dosis kombinasi ekstrak metanol daun benalu teh dan daun benalu mangga dosis 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB dengan ulangan 5x tiap perlakuan. Subjek menggunakan tikus putih (Rattus norvegicus) betina strain wistar yang berumur 6-8 minggu dengan berat badan minimal 100 gram. Perlakuan dalam penelitian ini berupa setiap dosis diberikan 5 kali seminggu selama 28 hari (uji toksisitas subkronik) secara per-oral. Kadar profil lipid dianalisis menggunakan uji One-way ANOVA. Hasil penelitian menunjukkan bahwa kadar profil lipid darah kontrol dengan perlakuan dosis 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB tidak berbeda nyata. Miq., Macrosolen tetragonus (Blume) Miq., Scurrula atropurpurea (Blume) Danser, Viscum articulatum Burm.f., and Viscum ovalifolium Wall. ex DC. The population of the mistletoes is dynamics affected by genetic and environment. The study aimed to investigate the population dynamics of mistletoe species on Cassia fistula L. in Purwodadi Botanic Garden, Indonesia. Data collection was conducted by direct observation in the garden on the mistletoe species, parasite density, and host prevalence in four block locations and 28 trees of C. fistula. Data were analyzed by two-way ANOVA and tested by Least Significantly Difference (LSD) if there was a significant difference in blocks and year. The results showed two species of mistletoe found on C. fistula, i.e. D. pentandra and V. articulatum. Parasitic density was significant different and infested host prevalence also different in blocks and years. The highest parasite density per host plant was reached in block III.D in 2015,i.e. 7.00±2.95 and 2.28±0.85 individuals per plant for D. pentandra and V. articulatum, respectively. The highest infested host prevalence of 100.00 was reached in block III.D in 2015 and XVI.A since 2012.
