Abstract:
Cells have the ability to respond and adapt to environmental changes through activation of stress-activated protein kinases (SAPKs). Activation of the p38 SAPK is critical to elicit the early gene expression program required for cell adaptation to stress. Alternative splicing (AS) is a crucial mechanism for gene regulation that has been shown to be modulated in response to a wide range of extracellular stimuli. However, the mechanisms behind AS regulation and the functional consequences of differential isoform expression in stress conditions remain largely unknown. In this study, we identified novel associations between p38 and the splicing machinery, providing mechanisms by which p38 regulates AS. Furthermore, characterization of protein isoforms produced by p38-dependent AS give an insight into the biological relevance of this regulation in adaptation to environmental stresses. Altogether, the results of this thesis highlight a role for SAPK signaling pathways in adaptation to environmental changes through AS modulation.
Les cèl·lules tenen l'habilitat de detectar i respondre a les fluctuacions en el seu entorn a través de l'activació de les proteïnes quinasa activades per estrès (SAPKs). L'activació de la SAPK p38 és essencial per activar el programa d'expressió gènica necessari perquè les cèl·lules s’adaptin als estímuls extracel·lulars. El processament alternatiu del pre-ARNm és un mecanisme de regulació gènica crucial que es regula en resposta a diferents canvis en l'ambient. No obstant, els mecanismes moleculars pels quals es regula, així com la funció de les diferents isoformes de les proteïnes que s'expressen en resposta a estrès no es coneixen. En conjunt, els resultats presentats en aquesta tesis proporcionen una nova visió dels mecanismes pels quals p38 modula el processament alternatiu del pre-ARNm en resposta a estrès i posen en evidència la importància d'aquest mecanisme per l'adaptació de la cèl·lula a canvis en l'ambient extracel·lular.

Abstract:
Le génome est sans cesse menacé dans sa structure par des stress génotoxiques d’origine endogène (stress oxydant, blocage de la réplication…) ou exogène (irradiations, produits chimiques, métaux lourds…). La voie de réponse aux dommages de l’ADN coordonne un réseau d’événements en cascades qui incluent les points de surveillance du cycle cellulaire, la réplication/réparation/recombinaison de l’ADN et la mort cellulaire programmée. Ces voies collaborent pour assurer la transmission fidèle du génome et empêcher la prolifération des cellules qui aurait accumulé des altérations génétiques. En général, des défauts dans une de ces voies entraînent un changement de sensibilité aux agents génotoxiques, une instabilité génétique et une prédisposition au cancer. La recombinaison homologue (RH) est une voie essentielle pour la réparation de l’ADN ; elle joue un rôle fondamental dans le maintien de la stabilité du génome. Le stress oxydant, résultant d’une augmentation de la concentration intracellulaire de ERO, est une des causes majeures de dommages aux lipides, aux protéines et à l’ADN et pour cela, il représente un défi pour la survie cellulaire et pour la stabilité du génome. Les ERO apparaissent physiologiquement lors de la respiration cellulaire ou résultent d’un stress environnemental, comme l’exposition aux radiations UV ou agents chimiques oxydants. Elles contribuent à certains processus comme la croissance cellulaire, l’activité et au repliement des protéines, la sénescence et la mort cellulaire programmée. Il est important de souligner qu’un état redox altéré est souvent associé à un fonctionnement anormal des cellules, comme il est observé pour les cellules cancéreuses et les cellules sénescentes. L’objectif de ce projet a été d’analyser l’interface entre les voies DDR et SAPK (Stress activated protein kinase) évolutivement conservées et d’en étudier les conséquences sur la stabilité du génome en utilisant comme organisme modèle la levure à fission. Nos résultats montrent que la voie SAPK joue un rôle sur la RH en promouvant la phosphorylation de Rad52, une protéine impliquée dans différentes sous-voies de la RH. Nous avons aussi montré que Rad52 est phosphorylée sur différents acides aminés, parmi lesquels certains sont les cibles de kinases inconnues qui n’ont aucun lien avec la voie SAPK. Nous avons observé que Rad52 est phosphorylée soit après un stress oxydant, soit dans des cellules génétiquement sujettes à des perturbations de la RH. Nous avons identifié deux sites de phosphorylation de la protéine Rad52, dont un seul est dépendant de la voie SAPK. En étudiant la phosphorylation de Rad52 dans les cellules invalidées pour la voie SAPK ou mutées pour un des sites de phosphorylation de Rad52, nous avons pu montrer que la RH est modulée par la voie SAPK même en absence de insulte externe. Notre travail ouvre le chemin vers une nouvelle compréhension des mécanismes fondamentaux du maintien de l’intégrité du génome
Genomes are routinely submitted to injuries from either endogenous stress (oxidative stress, DNA replication block…) or from exogenous sources (radiations, chemicals, heavy metals…). The DNA damage response (DDR) coordinates a network of pathways including cell cycle checkpoints, DNA replication/repair/recombination, and programmed cell death, ensuring faithful genome transmission and preventing from the proliferation of cells bearing genetic alterations. Defect in one of these pathways generally results in altered sensitivity to genotoxins, genetic instability and cancer predisposition. Homologous recombination (HR) is an essential DNA repair pathway playing pivotal role to maintain genome stability.Oxidative stress, resulting from increased intracellular concentration of ROS, is one of the major causes of lipid, protein and DNA damage, and therefore a challenge for cell survival and genome stability. ROS are generated physiologically as by-products of cellular respiration, or as result of environmental stresses, such as exposure to solar UV radiations or to oxidant chemicals, and they actively participate in processes such as cellular growth, protein activity and folding, senescence and programmed cell death. It is noteworthy that an altered redox homeostasis is often associated to abnormally functioning cells, such as cancer and senescent cells. The aim of this project was to study the interface between two major evolutionarily conserved pathways, DDR and SAPK (Stress activated protein kinase) and the consequences on genome stability using fission yeast as a model organism. We report data showing that SAPK pathway impinges on HR by promoting phosphorylation of Rad52, a key protein involved in all sub-pathways of HR. We also revealed that Rad52 is phosphorylated at multiple different sites some of which are substrate for unidentified kinases unrelated to the SAPK pathway. Rad52 phosphorylation occurs either after oxidative stress or in cells genetically prone to HR perturbation. We identified two sites of phosphorylation, one of which is dependent on functional SAPK pathway. By studying Rad52 phosphorylation in cells mutated in the SAPK pathway or mutated at the Rad52 site of phosphorylation, we showed that HR is modulated by SAPK even in the absence of external insults. Our work pave the way to a novel understanding of fundamental mechanisms required for genome integrity maintenance

Abstract:
Control of cell cycle by Stress Activated Protein Kinases (SAPKs) is an essential aspect for adaptation to extracellular stimuli. In Saccharomyces cerevisiae, the activation of the Hog1 SAPK, results in a delayed transcription of the G1 cyclins CLN1,2 and the stabilization of the B-type cyclin inhibitor SIC1, therefore postponing entry into S phase. The results displayed here, show, by a combination of mathematical modelling and quantitative in vivo experiments, that, before Start, the control of G1-S transition is mainly exerted by inhibiting expression of cyclins, both G1 (CLN1,2) and S phase (CLB5,6) cyclins. On the other hand, after Start, it is the phosphorylation and stabilization of Sic1 by Hog1 that becomes imperative to prevent inadequate firing of replication before adaptation. Therefore, we found that there is a distinct temporal role for Sic1 and cyclins on the G1 regulation by a SAPK in response to stress. We have also found that Hog1 induces a G2 delay, by down-regulating CLB2 transcription and phosphorylating Hsl1 to promote Hsl7 delocalization and subsequent accumulation of Swe1, an inhibitor of Clb1,2-Cdc28, and thus postponing anaphase. Altogether, we demonstrate novel Systems Biology approaches are useful to better understand how an intracellular signalling pathway incises on cell cycle control, beyond a mechanistic description, as well as showing how a single MAPK modulates different cell cycle checkpoints to improve cell survival upon stress.
El control del cicle cel·lular per Proteïna Cinases Activades per Estrès (SAPKs) es un aspecte essencial per a l'adaptació als estímuls extracel·lulars. A Saccharomyces cerevisiae, l'activació de la SAPK Hog1, resulta en un retardament de la transcripció de les ciclines de G1 (CLN1,2) i l'estabilització del inhibidor de les ciclines del tipus B, SIC1, i per tant posposa l'entrada en fase S. Els resultats que aquí s'exposen, mostren, mitjançant la combinació de modelatge matemàtic i experiments quantitatius in vivo, que, abans d'Start, el control de la transició es duu a terme principalment inhibint l'expressió de les ciclines, tant les de G1 (CLN1,2) com les de la fase S (CLB5,6). Per altra banda, després d'Start, la fosforilació i estabilització de Sic1 per part de Hog1 esdevé un fet necessari per a prevenir la iniciació de la replicació abans d'adaptar-se. Per tant, hem descobert aquí que la regulació de Sic1 i les ciclines juguen un paper diferent segons el moment en que apareix l'estrès. Hem descrit també, que Hog1 produeix una parada a G2 a través de la inhibició de la transcripció de CLB2 i la fosforilació d'Hsl1, la qual promou la deslocalització d'Hsl7 i la subsegüent estabilització de Swe1, un inhibidor específic de Clb1,2-Cdc28, i d'aquesta manera es posposa l'entrada en Anafase. Tot plegat, demostra que l'ús d'aproximacions pròpies de la Biologia de Sistemes és útil per a entendre de quina forma una via de senyalització intracel·lular incideix sobre el control del cicle cel·lular, mes enllà de la pura descripció de la mecànica del sistema. D'aquesta forma, proposem que una sola MAPK modula distints punts de control del cicle cel·lular per millorar la probabilitat de supervivència en front de l'estrès osmòtic.

Abstract:
This thesis describes the cloning and characterisation of a novel human gene and its protein products, which have been designated SAPK- and Ras-interacting protein (SinRIP). SinRIP shares identity with JC310, a partial human cDNA that was previously identified a candidate Ras-inhibitor (Colicelli et al., 1991, Proc Natl Acad Sci USA 88, p. 2913). In this study, it was shown that SinRIP is a member of an orthologous family of proteins that is conserved from yeast to mammals and contains proteins involved in Ras- and SAPK-mediated signalling pathways. Comparison of this family of proteins showed that human SinRIP contains a potential Ras-binding domain (RBD; residues 279-354), a PH-like domain (PHL; 376-487), and a highly conserved novel region designated the CRIM (134-265). Several other potential targeting sites, such as nuclear localisation signals and target sites for kinases, were identified within the SinRIP sequence. The human SinRIP gene is unusually large (>280 kbp) and is located on chromosome 9 at 9q34. SinRIP mRNA was detected in a wide variety of tissue-types and cell lines by RT-PCR, and the SinRIP sequences in the EST database were derived from an diverse array of tissues, suggesting a widespread or ubiquitous expression. Northern blot analysis revealed the highest levels in skeletal muscle and heart tissue. However, the steady-state levels of SinRIP mRNA vary greatly from cell to cell, and SinRIP expression is likely to be regulated at multiple post-transcriptional levels. It was shown that SinRIP mRNA is likely to be translated inefficiently by the normal cap-scanning mechanism, due to the presence of a GC-rich and structured 5’-UTR, which also contains upstream ORFs. Alternative polyadenylation signals in the SinRIP 3’-UTR can be used, resulting in the expression of short and long SinRIP mRNA isoforms. Several potential A/T-rich regulatory elements were also identified in SinRIP mRNA, which may target specific SinRIP mRNA isoforms for rapid degradation. Importantly, it was shown that SinRIP mRNA is alternatively spliced, resulting in the production of distinct SinRIP protein isoforms. Three isoforms, SinRIP2-4, were definitively identified by RT-PCR and full-length cloning. The SinRIP isoforms contain deletions in conserved regions, and are likely to have biochemical characteristics that are different to full-length SinRIP1. SinRIP2 is C-terminally truncated and lacks the PHL domain and part of the RBD, and relatively high levels of SinRIP2 expression arelikely to occur in kidneys. The RBD is disrupted in SinRIP3, but all other domains are intact, and RT-PCR analyses suggest that SinRIP3 is present in some cells at levels comparable to SinRIP1. A rabbit polyclonal antiserum against SinRIP was generated and detected endogenous SinRIP proteins. Using the anti-SinRIP antibody in immunoblots, multiple SinRIP isoforms were observed in most cell types. SinRIP1 and another endogenous SinRIP protein, likely to be SinRIP3, were detected in most cell lines, and appear to be are the major SinRIP proteins expressed in most cells. The subcellular localisation of both recombinant and endogenous SinRIP proteins was investigated by immunofluorescence assays and biochemical fractionation. Recombinant SinRIP1 protein was found in the cytoplasm and associated with the plasma membrane. In contrast, the SinRIP2 protein was predominantly nuclear, with only low-level cytoplasmic staining observed. The endogenous SinRIP proteins, likely to comprise these and other SinRIP isoforms, were found in both the nucleus and cytoplasm. SinRIP1 interacted with GTP-bound (active) Ras, but not GDP-bound (inactive) Ras, in an in vitro assay, and also co-localised with activated H- and K-Ras in cells. The binding profile observed is typical of Ras-effectors, and SinRIP did not inhibit signalling by the Ras proteins, suggesting that it is not likely to be a Ras-inhibitor. It was also shown that SinRIP1 and SinRIP2 both interact and colocalise with c-Jun NH2- terminal kinase (JNK). Abstract:
Colchicine and paclitaxel are two common drugs used in chemotherapy to halt tumor growth. In the present study IAR 20 cells were treated for 24 and 48 hr with colchicine and paclitaxel alone, in combination or no drug which served as a control. Through the use of Western blotting, we determined that the treatments affected expression due of several proteins including bcl-2, bax, p53 and caspase-8. The changes observed in protein expression due to the treatments correlated to the photomicrographs of the cells in culture and cell viability, indicating that the drugs were activating and initiating apoptosis. Interestingly, morphological changes such as membrane blebbing and cell swelling (indicators of apoptosis) were observed in the treated cultures and even more important the combined treatment yielded both changes in morphology. Also, activity assays were performed to study the effects the treatments had on the activities of SAPK/JNK and caspase-3, known activators of apoptosis. High activities of SAPK/JNK and caspase-3 in 48 hr treatments directly influenced cell viability in that the treatments with the highest activities yielded the lowest cell numbers, indicating that apoptosis was occurring. Based on these findings it was concluded that combined treatments of colchicine and paclitaxel are not advantageous in hepatocytes and could provide some insight into the treatment of liver cancer. Additionally, it appeared the drugs were initiating apoptosis in a p53 independent manner.

Stress activated protein kinase (SAPK) interacting protein 1 (Sin1) is a member of a recently characterized gene family, conserved from yeast to humans. The gene copy number is strictly conserved (one Sin1 gene per genome), and the protein may be expressed ubiquitously in mammalian tissues. The Sin1 family has been implicated in several different signal transduction pathways. Originally identified as a partial cDNA and candidate Ras inhibitor, recent functional studies have revealed interactions with an interferon (IFN) receptor subunit (IFNAR2), and the SAPK JNK. Interactions have also been described between the yeast orthologues and the phosphatidylinositol kinase TOR2. Collectively, these data suggest that Sin1 has an important cellular role, and this study has investigated possible functions for this protein. As human Sin1 proteins have no paralogues within the genome, secondary structure homology was used to identify major domains within the protein. Four major domains within human Sin1 were deduced: an N-terminal domain containing a functional nuclear localization signal, a functional nuclear export signal, and a coiledcoil region; the conserved region in the middle that is likely to be a ubiquitin-like β-grasp protein binding domain; a Ras binding domain; and a pleckstrin homology-like domain that targets Sin1 to the plasma membrane and lipid rafts in vivo. Full and partial length EGFP constructs were used to examine the localization of human Sin1, and several isoforms derived from alternative splicing. All isoforms localized to the nucleus and nucleolus. Beyond this, Sin1α and Sin1ϒ had cytoplasmic staining, while Sin1 and Sin1β were also found at the plasma membrane and lipid rafts. Both the N-terminal domain and the conserved region in the middle were found to contribute to nuclear localization. Comparative genomic analysis between human, mouse, rat, dog, and chicken Sin1 genes revealed a number of conserved intronic regions, and the putative functions of these were predicted. Additionally, a putative promoter module within a CpG island and encompassing the transcription start site was predicted in all species. The human CpG island was found to have promoter activity in HEK293 cells. Using bioinformatics, genes that may be co-regulated with Sin1 were identified. These genes contained the Sin1 promoter module, and were found to co-express in large scale gene expression studies. Most of these genes were directly involved in the cellular response to pathogen infection, suggesting a conserved role for Sin1 in this pathway. As espécies de Candida são patógenos oportunistas e as infecções nas cavidades bucais causadas por este gênero são comuns em pacientes com fatores predisponentes, entre eles imussuprimidos, diabéticos e usuários de próteses dentais. No Brasil o índice de endentados é elevado sendo necessária a utilização de próteses bucais, que são locais favoráveis à colonização de microrganismos e o desenvolvimento de biofilme. A utilização de antissépticos como medida complementar na higiene oral é cada vez mais difundida. O objetivo deste trabalho foi detectar em amostras de C. albicans e C. não-albicans, isoladas da saliva de portadores de próteses bucais, os fatores de virulência, proteinase e biofilme e genes relacionados à proteinase (SAP1, SAP3) e à adesão (ALS1 e ALS3) e avaliar a atividade antifúngica in vitro de três formulações comerciais à base de clorexidina 0,12%, cetilpiridínio 0,07% e 0,075% frente aos isolados de Candida e biofilmes. Foram coletadas amostras de saliva de 70 pacientes usuários de próteses bucais. As leveduras foram isoladas em meio de CHROMagar® Candida e identificadas pela metodologia clássica e a confirmação de C. albicans por biologia molecular (PCR). Leveduras do gênero Candida foram isoladas de 32 (45,7%) amostras. Destas foram identificadas: C. albicans (29), C. krusei (5), C. glabrata (4), C. tropicalis (3) e C. parapsilosis (2), totalizando 43 leveduras entre monoculturas e culturas mistas. Os genes pesquisados foram detectados somente em amostras de C. albicans, os genes ALS2, ALS3 e SAP3 foram identificados em 86% dos isolados e o SAP1 em 93%. A atividade de proteinase entre os dos isolados de C. albicans foi de 72,4% e entre das espécies de C. não-albicans 35,7%. A de formação de biofilme sobre placas de 96 poços, e após coloração com XTT, demonstrou que a maioria dos isolados de Candida e todas as culturas mistas desenvolveram biofilmes em 24 horas. A atividade de três antissépticos frente as leveduras foi de <=1,25%, equivalente a uma diluição de 1:80 do antisséptico. Concluímos os três antissépticos também foram eficazes frente aos biofilmes de monoculturas e culturas mistas, as cepas de C. albicans continua sendo a espécie prevalente na cavidade bucal, os genes SAP1, SAP3, ALS2 e ALS3 estão presentes na maioria dos isolados de C. albicans, nestas amostras a maioria foram proteinase e biofilme positivos. A atividade de proteinase foi superior nas espécies de C. albicans em comparação com C. não-albicans. Os biofilmes das espécies de Candida quando formados em culturas mistas se desenvolvem mais em comparação com a formação de biofilme em monocultura. As três formulações de antissépticos foram eficazes contra todos isolados de leveduras em forma planctônica e na maioria dos biofilmes.
Candida species are opportunistic pathogens and infections in the oral cavity caused by these yeasts are common in patients with predisposing factors, such as diabetic or immunocompromised individuals with dental prosthesis. In Brazil, the number of toothless patients is high and the use of dental prosthesis is common, which represents a niche for colonization and formation of microbial biofilms in the oral cavity. The use of mouthwashes to aid in the oral hygiene has increased. The objective of this research was to isolate C. albicans and C. non-albicans from saliva of dental prosthesis users and to evaluate the isolates for the presence of virulence factors, proteinase production, biofilm formation and to search for genes encoding for proteinases (SAP1, SAP3) and adhesion factors (ALS1 and ALS3). Another objective was to evaluate in vitro the antifungal activity of three commercial mouthwashes containing 0.12% chlorhexidine and 0.07% or 0.075% cetylpyridinium, against planktonic Candida sp. cultures and in biofilms. Saliva samples of 70 patients with dental prosthesis were collected, the yeasts were isolated on CHOMagar® Candida and identified by classical biochemical tests combined with molecular test (PCR) for confirmation of C. albicans. Yeasts of Candida genus were isolated from 32 samples (45.7%) and were identified as C. albicans (29), C. krusei (5), C. glabrata (4), C. tropicalis (3) and C. parapsilosis (2), with a total of 43 yeasts, including mono and mixed cultures. The genes studied were detected only in C. albicans: ALS2, ALS3 and SAP3 genes were detected in 86% of the isolates and SAP1 in 93%. The proteinase activity among the isolates of C. albicans was 72.4% and among the species of C. non-albicans it was 35.7%. Assay done with cultivation in 96-wells microplates and staining with XTT, showed the majority of Candida isolates and all the mixed cultures were able to form biofilms within 24h. The activity of the three mouthwashes towards the yeasts was < 1.25%, equivalent to a dilution of 1:80 of the formulation. The three mouthwashes tested were also active against mono or multispecies biofilms. In conclusion, it was shown C. albicans is the most prevalent species. The genes SAP1, SAP3, ALS2 and ALS3 are present in the majority of C. albicans isolates, which were also positive for proteinase and biofilm formation. There was more proteinase activity in C. albicans in comparison with C. non-albicans. The biofilms of Candida species grew better in mixed cultures than in monoculture. The three commercial mouthwashes formulations were effective against all the yeast isolates in planktonic growth and they also inhibited the majority of yeasts in biofilm. Future work is needed to completely elucidate the role of Candida species in biofilms of oral cavity. It is also interesting to propose studies to standardize sensitivity tests for mono and multi-species biofilms.

Abstract:
Regulation of transcription by Stress Activated Protein Kinases (SAPKs) is an essential aspect for adaptation to extracellular stimuli. In mammals, the activation of the p38 SAPK results in the regulation of gene expression through the direct phosphorylation of several transcription factors. However, how p38 SAPK regulates the proper gene expression program of adaptation to stress as well as the basic mechanisms used by the SAPK remains uncharacterized. The results displayed in this manuscript show that the p38 SAPK plays a central role in the regulation of gene expression upon stress, as up to 80% of the upregulated genes are p38 SAPK dependent. Moreover, we also observed that a specific set of genes were upregulated in response to each specific stimuli, and just a small set of genes were commonly up-regulated by several stresses, which involves mainly transcription factors. In addition, we observed that, to proper regulate gene transcription, the p38 SAPK is recruited to stress-induced promoters via its interaction with transcription factors. Additionally, p38 activity allows the recruitment of RNA polymerase II and the MAPKK MKK6 to stress-responsive promoters. The presence of active p38 SAPK at open reading frames also suggests the involvement of the SAPK in elongation. Altogether, the results showed in this manuscript establish the p38 SAPK as an essential regulator in the transcriptional response to stress, as well as define new roles for p38 in the regulation of transcription in response to stress.
La regulación de la transcripción por las Proteínas Quinasas activadas por Estrés (SAPKs) es un aspecto esencial para la adaptación a los estímulos extracelulares. En mamíferos, la activación de la SAPK p38 da lugar a la regulación de la expresión génica a través de la fosforilación de varios factores de transcripción. Sin embargo, cómo p38 SAPK regula el programa de expresión génica de adaptación al estrés así como los mecanismos utilizados por la SAPK permanece sin caracterizar. Los resultados presentados en este manuscrito muestran que p38 SAPK juega un rol central en la regulación de la expresión génica en respuesta a estrés, ya que hasta el 80% de los genes inducidos son dependientes de p38 SAPK. También observamos que en respuesta a cada tipo de estrés se induce un grupo de genes específicos, y sólo hay una pequeña respuesta de genes comunes a los diferentes tipos de estrés la cual engloba principalmente factores de transcripción. Además, hemos observado que para regular la transcripción, p38 se recluta a los promotores de respuesta a estrés a través de su interacción con factores de transcripción. Asimismo, la actividad de p38 permite el reclutamiento de la RNA Polimerasa II y de la MAPKK MKK6 a los promotores inducidos por estrés. La presencia de p38 activa en las regiones codificantes sugiere su participación durante la elongación. En conjunto, los resultados mostrados en este manuscrito establecen a p38 como un regulador esencial de la transcripción en respuesta a estrés, así como definen nuevas funciones de p38 en la regulación de la transcripción en respuesta a estrés.

En el llevat Saccharomyces cerevisiae els canvis en les condicions osmòtiques del medi extracel.lular són sensades per la MAP cinasa Hog1, la qual permet dur a terme l'adaptació cel.lular mitjançant la modulació de l'expressió gènica, de la traducció i de la progressió del cicle cel.lular. A l'inici d'aquest projecte de tesi, els mecanismes pels quals Hog1 controla l'expressió gènica no eren del tot coneguts. El nostre objectiu va ser caracteritzar el mecanisme molecular pel qual Hog1 modula la transcripció en resposta a estrès osmòtic. Hem aconseguit demostrar que el reclutament de Hog1 als promotors sensibles a estrès osmòtic per part del factor de transcripció és essencial per al reclutament i activació de la RNA polimerasa II, mecanisme que podria estar conservat en les cèl.lules eucariotes. També hem identificat noves activitats remodeladores de cromatina implicades en la resposta gènica a osmoestrès mediada per Hog1. Vàrem realitzar un cribatge genètic per identificar mutacions que provoquessin osmosensibilitat i una reducció en l'expressió de gens de resposta a estrès osmòtic. Aquest cribatge ens va permetre identificar nous reguladors de la transcripció mediada per osmoestrès: la histona deacetilasa Rpd3 i els complexes SAGA i mediador. Els nostres resultats permeten, doncs, definir un important paper per a Rpd3, SAGA i mediador en la inducció gènica mediada per Hog1, i han estat importants per assolir una millor visió de com les cinases activades per estrès regulen la iniciació de la transcripció.
In Saccharomyces cerevisiae, changes in the extracellular osmotic conditions are sensed by the HOG MAPK pathway, which elicits the program for cell adaptation, including modulation of gene expression, translation and cell-cycle progression. At the beginning of this PhD Project, the mechanisms by which Hog1 was controlling gene transcription were not completely understood. Our main objective was to characterize the molecular mechanisms by which the Hog1 MAPK modulates transcription upon osmostress. We have shown that anchoring of Hog1 to osmoresponsive promoters by the transcription factor is essential for recruitment and activation of RNA polymerase II, a mechanism that might be conserved among eukaryotic cells. In addition, we identified novel chromatin modifying and remodelling activities involved in the Hog1-mediated osmostress gene expression. We performed a genome-wide genetic screening searching for mutations that render cells osmosensitive and displayed reduced expression of osmoresponsive genes. Rpd3 histone deacetylase, SAGA and Mediator complexes were identified as novel regulators of osmostress-mediated transcription. Thus, our results define a major role for Rpd3, SAGA and Mediator in the Hog1-mediated osmostress gene induction, and have been important to achieve a better view of how a SAPK regulates transcription initiation.

As infecções fúngicas se tornaram um grande problema de saúde pública, especialmente em ambientes hospitalares. Candida spp. são os principais patógenos em infecções fúngicas invasivas. A produção de aspartato proteases secretadas (Sapp) por Candida spp. pode estar relacionado com o aumento do número de infecções e resistência aos medicamentos. A produção de proteases é codificada por uma família de 10 genes SAP1-10, sendo SAP2 o gene mais comumente expresso em Candida albicans. A expressão de SAP4 está associada à produção de enzimas Sap4p e formação de hifas que podem contribuir para a invasão dos tecidos hospedeiros e destruição de macrófagos em processos infecciosos. Este trabalho avaliou a expressão dos genes SAP2 e SAP4 em C. albicans ATCC 10231 cultivadas na presença ou ausência de macrófagos e expostas a concentrações subinibitórias de fluconazol e anfotericina B. Candida albicans foi cultivada em ágar Sabouraud Dextrose a 37ºC por 24 h, e posteriormente em caldo carbono levedura base enriquecido com soro-albumina bovina (YCB-BSA), e na ausência ou presença de concentrações subinibitórias de fluconazol e anfotericina B. A linhagem celular monocítica humana de origem leucemia (THP-1) foi cultivada em meio RPMI-1640 suplementado com 10% de soro fetal bovino, penicilina (100 U/ml) e estreptomicina (100 μg/ml) a 37ºC, sob atmosfera de 5% de CO2 durante 10 dias. Para a indução da diferenciação das células, 106 monócitos foram cultivados em meio RPMI-1640 suplementado e forbol-12-miristato-13-acetato (PMA) 100 nM durante 48 h. Foram utilizadas amostras de 5x106 C. albicans cultivadas na presença ou ausência de 106 macrófagos e expostas ou não a concentrações subinibitórias de antifúngicos. A amostras foram cultivadas em placas de cultura celular com 6 poços por 1 h a 37 °C, sob atmosfera de 5% de CO2. O RNA total foi extraído das amostras utilizando o reagente TRIzol®, após a purificação do RNA com DNase I, este foi convertido em cDNA, e, em seguida, a quantificação do gene SAP4 foi realizada por Reação em Cadeia de Polimerase quantitativa (qPCR) utilizando ACT1 como gene normalizador. Candida albicans cultivada na presença de macrófagos apresentou regulação positiva na expressão de SAP2 e SAP4 (p < 0,01), respectivamente, na ordem de 4,83 e 10,34 vezes maior se comparada com amostras da levedura cultivada sem contato com macrófagos. E ainda, a exposição de C. albicans a concentrações subinibitórias de fluconazol aumentou a expressão de SAP2 e diminuiu a expressão de SAP4, já a exposição da levedura a concentrações subinibitórias de anfotericina B diminuiu a expressão de SAP2 e SAP4. Os genes SAP2 e SAP4 são relacionados como fatores de virulência de C. albicans e sua expressão pode ser regulada pelo contato com fagócitos ou exposição a antifúngicos. Portanto, compreender a expressão desses genes na patogênese fúngica pode ajudar na pesquisa e desenvolvimento de novos medicamentos para o tratamento da candidíase, contribuindo assim para a redução da incidência de morbidade e mortalidade associada a infecções fúngicas.
Fungal infections have become a major public health problem especially in hospital settings. Candida spp. are considered the main pathogen in invasive fungal infections. The production of secreted aspartic proteinases (Sapp) by Candida spp. can be related to the increase in the number of infections and drug resistance. The production of proteinases is encoded by a family of 10 genes known as SAP1-10, of which, SAP2 is the most commonly expressed gene in Candida albicans. The expression of SAP4-6 is associated to the production of Sap4-6p enzymes and hyphae formation which can contribute to the invasion of host tissues and destruction of macrophages during infectious processes. This study evaluates SAP2 and SAP4 gene expression in C. albicans ATCC 10231 grown in the presence or absence of macrophages and exposed to subinibitory concentrations of fluconazole and amphotericin B. C. albicans was grown in Sabouraud Dextrose Agar for 24 h at 37ºC and after in yeast carbon base more 0.2% bovine serum albumin (YCB-BSA) in the absence or presence of subinibitory concentrations of Fluconazole and Amphotericin B. The human monocytic leukemia cell line (THP-1) was grown in RPMI-1640 Medium supplemented with 10% fetal bovine serum, penicillin (100U/ml) and streptomycin (100μg/ml) at 37 °C and atmosphere of 5% CO2 during 10 days. For the induction of cell differentiation, 106 cells were seeded in RPMI-1640 medium supplemented and phorbol 12-myristate 13-acetate (PMA) 100 nM for 48 h. The samples of 5x106 C. albicans were cultured in the presence or absence of 106 macrophages and these was exposed or not to subinibitory concentrations of antifungals. All samples were incubated in culture test plates with 6 flat-botton wells for 1 h at 37 °C and atmosphere of 5% CO2. Total RNA was extracted from the samples using TRIzol® reagent, after RNA purification with DNaseI it was converted to cDNA, and then, the relative quantification of the SAP4 gene was performed by Real-Time Polymerase Chain Reaction (qPCR) using ACT1 gene as normalizing endogenous gene. Candida albicans grown in the presence of macrophages showed upregulation in the expression of SAP2 and SAP4 (p < 0.01), respectively, in the order of 4.83 and 10.34 fold higher as compared to yeast samples grown without contact with macrophages. Concerning exposure to antifungal agents, C. albicans exposed to subinhibitory concentrations of amphotericin B showed downregulation in expression of SAP2 and SAP4. However, the yeast's exposure to subinhibitory concentrations of fluconazole caused upregulation in SAP2 expression and downregulation in SAP4 expression. SAP2 and SAP4 genes are related to virulence factors of C. albicans and its expression can be regulated by contact with phagocytes or exposure to antifungals. Therefore, understanding the expression of these genes in fungal pathogenesis may aid in research and development of new drugs for treating candidiasis, thus contributing to reducing the incidence of morbidity and mortality associated with fungal infections.

Abstract:
During this study, an attempt was made to explore the opinions and attitudes of members of the South African Police Service (SAPS) towards issues of diversity before, immediately after and three months after participating in diversity training workshops presented by the SAPS Training Division during 1997 in Port Elizabeth. The aim of this thesis was not to assess the diversity training itself, but to discover if the training, as currently presented, in any way influenced the attitudes of participants. In order to meet this goal, literature and empirical studies were conducted. The literature study sets the theoretical foundation pertaining to the history of the SAPS and the attitudes and prejudices of and diversity among SAPS members. During the empirical research phase, a non-probability purposive sampling procedure was adopted. Four of a range of diversity workshops presented by the Training Division of the SAPS during 1997 were selected for the purpose of this study. An internal SAPS process was used to nominate members to attend the workshops. The researcher requested the participants in each of the four workshops to complete a self-administered questionnaire before as well as after the workshop concerned. Immediately after each of the four workshops, a short interview was held with each attendee. To explore the stability of any change evident from responses on the questionnaires completed after the workshops, the attendees were again requested to complete the same questionnaire three months later. To increase the validity of any conclusion that attitudinal change was related to the workshop, a control group was used. This study has revealed that a cross-spectrum of SAPS members of both sexes and diverse racial backgrounds believe that various forms of discrimination exist within the SAPS. It confirmed that the diversity training presented by the SAPS Training Division is a useful instrument to heighten members' awareness of the different norms and customs of other cultural/ethnic groups and of the necessity that the SAPS should be constituted of a cross-spectrum of racial groups reflecting the South African society. Finally, based on the research findings, recommendations were made involving management and its supportive services and diversity training.

A entidade gestora do sistema de atribuição de produtos de apoio, o Instituto Nacional para Reabilitação, identificou a necessidade de possuir uma ferramenta que permita efetuar análises e extrair diversa informação estatística sobre os processos de atribuição de qualquer produto, instrumento, equipamento, tecnologia e software usado por uma pessoa com deficiência, vulgo, produtos de apoio. O presente projeto é a concretização do acima referenciado, desenvolvimento de um Data Warehouse e de relatórios, como uma ferramenta de Business Intelligence. Inicialmente foi efetuado um estudo aprofundado das regras do processo de negócio e do modelo relacional, que suporta o sistema de gestão de base de dados operacionais. Foram efetuadas várias reuniões com os representantes da entidade gestora e um estudo dos requisitos do projeto, através da criação de queries ao sistema fonte de dados. Após refinamento dos requisitos da informação estatística e conclusão da fase de análise, que inclui-o atividades como identificação de fontes, destino de dados e modelização, foi desenvolvido o Data Warehouse. Trabalho envolvendo os inerentes processos de extração, transformação e carregamento, seguido dos testes, automatização de processos e colocação em produção. O Data Warehouse, desenvolvido permitiu concretizar a implementação da infraestrutura de análise, efetuado com a ferramenta de Business Intelligence e assim atingir o objetivo final, possuir informação estatística, atualizada e precisa.
The managing body responsible for allocating disability equipment, the National Institute for Rehabilitation (Instituto Nacional para Reabilitação), has identified the need for a tool that allows the analysis and extraction of all the statistical information related to the process of granting disability equipment. In order to achieve this objective, the current project developed a Data Warehouse and reporting system using a Business Intelligence tool. The first step was to conduct an in-depth study focusing on the business process rules, and the relational model that supports the database management system. In order to determine the project’s requirements, various meetings were held with the representatives of the managing body, as well as data source queries. The Data Warehouse was created after refining the requirements of the statistical information and the completion of the analysis step, which included tasks such as source identification, data destination, and model-building. This involved the extraction, transformation and loading of the data, which was followed by testing, process automation, and the production stage. After loading the Data Warehouse, the data is organised in a way that allows the deployment of the analysis infrastructure, via the Business Intelligence tool, in order to achieve the final goal – to have updated and accurate statistical information.

Com o lançamento e utilização de novas ferramentas para contextos educativos que pretendem realçar a importância da comunicação e colaboração em ambientes de aprendizagem, surgem novos desafios que é necessário investigar para encontrar soluções adequadas. No caso deste estudo, procura-se compreender e encontrar soluções adequadas para a problemática da sobrecarga de informação que pode ocorrer num espaço em que todos os elementos de uma comunidade têm a possibilidade de partilhar conteúdo e comunicar entre si. Nestes contextos, é possível que a informação disponível possa atingir uma quantidade que o utilizador não tem a capacidade de assimilar na sua totalidade e, muitas vezes, originando que perca informação relevante para si. Uma possível solução para este desafio passa pela integração de sistemas de recomendação transversais a essas novas ferramentas, potenciando o seu desenvolvimento baseado no próprio utilizador e também na sua interação com a comunidade, promovendo o desenvolvimento de novas formas de partilha e de interação entre os utilizadores. Neste trabalho apresentam-se as principais etapas da conceção, desenvolvimento e avaliação da integração de um sistema de recomendações numa plataforma social para contexto educativo, o SAPO Campus.
With the advent of new educational tools for learning contexts with the purpose of highlighting the importance of communication and collaboration in learning environments, new challenges should be taken into account in order to discover proper solutions. With this investigation, we seek to understand and find a suitable approach to the information overload problem that can occur in places where all community members have the ability to communicate and share content. It is possible, in these contexts, for the available information to reach such an amount that the user will not have the ability to fully assimilate it, which often causes the loss of the track of data, which is importante to the user. A possible solution for this challenge implies the integration of transverse recommendation systems in these tools, enhancing their growth based on the user and his interaction with the community and promoting the emergence of new methods for him to share and interact with others. This study presents the main steps of the design, development and assessment of the process for integrating a recommendation system in SAPO Campus, a social platform for the educational context.

The theoretical part describes the corporate social responsibility from a historical and a current perspective. Above all, the thesis puts emphasis on the social pillar, namely the activities of the company in relation to its stakeholders, particularly employees and volunteering. Volunteering is an important part of corporate social responsibility. The questionnaire and subsequently its statistical evaluation answers the question whether the students currently studying in two economic universities consider SAP as a responsible company and how they relate to volunteerism.

Corruption and unethical management is a legitimate global concern as it can have a direct and substantial impact on the longevity and the success of an organisation, and a country. As a result much attention has been given to unethical behavior and organisational ethical violations over the last decade. Recent events have indicated that ethics management is an organisational concern with mammoth implications for organisations. The aim of this study was to investigate the development of strategies for the promotion of ethics and the impact of unethical conduct on an organisation. A direct link exists between employee behaviour and the organisational environment. The desired ethical environment is negatively affected if the employees cannot distinguish between right and wrong and do not act ethically when confronted with ethical dilemmas. Proposed ethics strategies may also be impacted on by the unethical behaviour of the leadership of organisations, compliance with legislation and poorly managed ethics strategies. The main research problem in this study was to identify what strategies could be adopted to create an ethical culture within an organisation. To achieve this objective a model was developed, proposed and supported with literature and the research study. A research questionnaire was designed to obtain the relevant data that was distributed to previously identified respondents. The results indicated that the majority employees were of the view that the organisational environment and leadership was not ethical and that the majority of employees were not actively pursuing an ethical environment. Findings indicated that the majority of employees felt that contributing factors to achieve an ethical environment was present, but ethic strategies were ineffective and poorly managed. The proposed study model, when implemented successfully, would allow the organisation the effective application of ethic interventions.

A web moderna, ou web 2.0, assenta nos princípios de abertura e participação dos seus utilizadores. A natureza voluntária do uso de serviços da web 2.0, aliada à dependência da participação por parte dos seus utilizadores, leva a uma forte concorrência entre serviços semelhantes na web. Esta concorrência leva à procura de novas formas de diferenciação entre serviços. Neste contexto surge a indústria de gamification, que procura transferir elementos de videojogos a outros contextos para aumentar o envolvimento dos utilizadores. Contudo, o discurso desta indústria recente é alvo de fortes críticas de profissionais de game design. Neste trabalho de investigação apresenta-se tanto o discurso da indústria de gamification como as suas críticas. Assumindo que a abordagem atual da gamification assenta numa compreensão limitada dos videojogos, parte-se para a elaboração de um novo quadro concetual que possa guiar o desenho da interação em ambientes web. Esta abordagem fundamenta-se num levantamento bibliográfico da teoria do game design. O quadro concetual resultante é usado no desenho e desenvolvimento de um serviço de social bookmarking no Sapo Campus UA, uma plataforma de serviços web 2.0 para contextos educativos, com o objetivo explícito de aumentar a participação dos seus utilizadores na aplicação. A utilidade do quadro concetual é avaliada com sessões de teste com utilizadores do público-alvo do serviço. Os resultados obtidos indicam que o game design pode enriquecer o desenho da interação na web através da criação de ciclos de ação com resultado claro e feedback positivo.
The modern web, or web 2.0, is centred on the principles of openness and user participation. The voluntary use of web 2.0 services, together with the dependency on user participation, results in a strong competition. This situation gave rise to the gamification industry, which aims to bring elements of video games to other contexts in order to increase user engagement. However, professional game designers strongly criticize the discourse of this young industry. This dissertation presents the gamification industry's discourse and its major critics. Assuming that the current approach to gamification only holds a limited understanding of videogames, we seek to build a new conceptual model to be used in web interaction design. This approach is based in a review of game design literature. The resulting conceptual model is used in the design and development of a social bookmarking service for the Sapo Campus UA project, a web 2.0 platform for higher education, with the specific goal of increasing user participation. The modelʼs usefulness is evaluated through user tests with potential future users. The results of this study indicate that game design can assist web interaction design in the creation of user activity cycles with clear results and positive feedback.

Abstract:
A novel solution method based on Mono-implicit Runge-Kutta methods has been fully developed and analysed for the numerical solution of stiff systems of ordinary differential equations (ODE). These Backward Runge-Kutta (BRK) methods have very desirable stability properties which make them efficient for solving a certain class of ODE which are not solved adequately by current methods. These stability properties arise from applying a numerical method to the standard test problem and analysing the resulting stability function. This technique, however, fails to show the full potential of a method. With this in mind a new graphical technique has been derived that examines the methods performance on the standard test case in much greater detail. This technique allows a detailed investigation of the characteristics required for a numerical integration of highly oscillatory problems. Numerical ODE solvers are used extensively in engineering applications, where both stiff and non-stiff systems are encountered, hence a single code capable of integrating the two categories, undetected by the user, would be invaluable. The BRK methods, combined with explicit Runge-Kutta (ERK) methods, are incorporated into such a code. The code automatically determines which integrator can currently solve the problem most efficiently. A switch to the most efficient method is then made. Both methods are closely linked to ensure that overheads expended in the switching are minimal. Switching from ERK to BRK is performed by an existing stiffness detection scheme whereas switching from BRK to ERK requires a new numerical method to be devised. The new methods, called extended BRK (EBRK) methods, are based on the BRK methods but are chosen so as to possess stability properties akin to the ERK methods. To make the code more flexible the switching of order is also incorporated. Numerical results from the type-insensitive code, SARK, indicate that it performs better than the most widely used non-stiff solver and is often more efficient than a specialized stiff solver.

Abstract:

Sapa Heat Transfer AB (SHT AB) är en av tre delar inom koncernen Sapa Group, som ägs av det norska företaget Orkla ASA. SHT AB har produktion både i Sverige (Finspång) och i Kina (Shanghai). Företaget är en världsledande leverantör och utvecklare av aluminiumband som används i olika typer av värmeväxlare, främst inom bilindustrin.

I flera steg av produktionen, som innefattar omsmältning, varm- och kallvalsning samt skärning, uppkommer ett skrotfall av aluminium. Detta skrot tas omhand och återanvänds som råvara i företagets omsmältverk. SHT AB har idag inget automatiserat IT-stöd för att prognostisera skrotavfallet, dessa beräkningar görs istället med hjälp av Excel. Då skrotfallet beräknas används ett schablonvärde för processens utbyte, som motsvarar genomsnittet för samtliga legeringskombinationer i produktion. Eftersom ett medelvärde används blir prognosen missvisande för vissa legeringar. Detta då utbytet varierar mycket, vilket gör att det både finns de legeringskombinationer som har betydligt högre utbyte än genomsnittet och de som har betydligt lägre. SHT AB har idag inte heller någon koppling mellan faktisk orderstock och kortsiktig prognostisering av skrotfall.

Syftet med detta examensarbete är att ta fram förslag på hur Sapa Heat Transfer AB kan och bör utveckla sin prognostisering av skrotfall. Därigenom ska Sapa Heat Transfer AB få bättre precision i både långsiktig och kortsiktig skrotfallsprognos, och på så vis förbättra råvaruplaneringen.

För att förbättra den långsiktiga skrotfallsprognosen måste nedbrytningen av prognosen för legeringskombinationer analyseras. En förbättring av nedbrytningen förväntas innebära en förbättrad långsiktig prognos. Förbättrad innebär i detta fall att skrotfallsprognosen bättre ska följa den långsiktiga prognosen för försäljning av legeringskombinationer. Precisionen hos skrotfallsprognosen blir givetvis beroende av precisionen hos denna prognos. En förändring av nedbrytningen medför dock att kapacitetsplaneringen för omsmältverkets ugnar påverkas.

Åtgärdsförslaget som tas fram för den långsiktiga prognostiseringen är tänkt att förbättra denna genom att schablonvärdet som används ersätts med varje legeringskombinations specifika prognostiserade utbyte. Data som behövs för att genomföra en sådan prognos finns tillgänglig i SHT AB:s databaser, och med hjälp av prognostiseringsmetoden exponentiell utjämning kan utbytet prognostiseras. Genom att använda denna metod med ett lågt värde på utjämningskonstanten α, fås en prognos där extremvärden inte ger kraftiga utslag på prognosen.

Den kortsiktiga skrotfallsprognosen, som idag bygger på en nedbrytning av den långsiktiga prognosen, kan förbättras genom att istället kopplas till den faktiska orderstocken. Om denna koppling görs baseras skrotfallsprognosen mer på känd information istället för på en annan prognos. En koppling till orderstocken kan dock bara användas för kortsiktig skrotfallsprognostisering då orderstocken inte sträcker sig flera månader framåt.

Åtgärdsförslaget som tas fram för den kortsiktiga skrotfallsprognosen utgår från att en schablonmässig prognos först tas fram för de skrotgrupper som ska prognostiseras. I denna används en individuell prognostiserad skrotfallsfaktor som tas fram med exponentiell utjämning för respektive skrotgrupp. Skrotfallsfaktorn är baserad på historisk data för planerat material in till processen och invägt skrot till smältverket. Med hjälp av skrotfallsfaktorn, data för hur mycket material som är planerade in i processen kommande veckor och med hänsyn till att skrot faller under hela ledtiden, kan ett förväntat skrotfall för kommande vecka/veckor beräknas.

För att få en bättre precision än hos den schablonmässiga prognosen, kan sedan prognosen för de skrotgrupper som är mest intressanta utvecklas och anpassas. I förslaget har förenklingen gjorts att skrot väntas falla symmetriskt över ledtiden vilket inte stämmer helt med verkligheten. Därför bör prognosen också utvecklas så att hänsyn tas till att det exempelvis faller förhållandevis mycket skrot i slutet av ledtiden.


Sapa Heat Transfer AB (SHT AB) is one of three components in the corporate group Sapa Group, owned by the Norwegian company Orkla ASA. Sapa Heat Transfer AB has production in Sweden (Finspång) and China (Shanghai). The company is a world leading supplier and developer of aluminum strips used in various types of heat exchangers, primarily in the automotive industry.

In several stages of the production, which includes remelting, hot and cold milling and cutting, a part of the processed material becomes scrap metal. This scrap metal is collected and reused as raw material in the remelting factory. SHT AB currently has no automated IT support to forecasting this scrap metal; these calculations are, in the current situation, made with the help of Excel. In the calculation of scrap metal the company uses a standard value, corresponding to the average yield for all alloy combinations in the production. As a mean value is being used for all alloy combinations, the forecast becomes misleading for some alloys. The yield varies widely depending on the alloy, which means there are alloy combinations that are above the average yield and some that are below. In the current situation SHT AB has no link between actual orders and the short-term forecast of scrap metal.

The purpose of this thesis is to develop proposals on how Sapa Heat Transfer AB can and should develop its forecasting of scrap metal. This will help them getting a better accuracy in their forecasting and thereby improving the raw material planning.

In order to improve the long-term forecast of scrap metal the decomposition of the forecast for the alloy combination had to be analyzed. An improvement of the decomposition will mean an improvement of the precision for the long-term prognosis of scrap metal. A change in the decomposition will also influence the capacity planning in the remelting factory.

The action proposal for the long-term forecasting will result in a better accuracy of the forecast by replacing the mean value for the yield by a specific forecasted yield of each alloy combination. The data needed to carry through such a forecast is available in SHT AB´s databases, and by using the forecasting method exponential smoothing the yield can be forecasted. By using this method a forecast will be obtained that, with a low value of α, will not take such great impact from extreme values that sometimes occur.

The current short-term scrap metal forecast, based on a decomposition of the alloy combination forecast, can be improved by a link to the actual backlog. If this link is established the scrap metal forecast will be based on known information instead of forecasted data. A link to the backlog can only be used for a short-term forecast due to the length of the awaiting orders.

The action proposal to improve the short-term forecast assumes that a standardized forecast is developed for the scrap groups to be projected. The forecast should use an individually forecasted scrap factor produced with exponential smoothing for each scrap group. The scrap factor is based on historical data for the material planned to go into the process and the weighed scrap delivered to the remelting factory. With this scrap factor, data on how much material that is planned into the process the coming weeks and with consideration of scrap falling throughout the lead time the expected amount of scrap for the coming week / weeks can be calculated.

To get a better precision compared to the standardized forecasts, the forecasts of the most interesting scrap groups should be developed and adapted. They can be given individual values of the smoothing constant α for the prediction of the scrap factor and the way the lead time is being considered can be adjusted. In the proposal the simplification that scrap is expected to fall symmetrically distributed over the lead time has been done. In reality this is not the case. Therefore, the forecast should also be developed to take into account that, for example, more scrap is falling in the end of the lead time.

Sapa är en av de största tillverkarna i världen av aluminiumprofiler. Sapa Building System AB är ett dotterbolag till Sapa group och är en av de största leverantörerna i Europa på byggsystem. Några av deras produkter är bakdörrarna RD 3010 och RD 3020 för lastbilar, släp och trailers som från början var ämnade som sidodörrar. Som bakdörrar är de överdimensionerade då sidodörrar hade högre hållfasthetskrav. I dagens läge väger och kostar de mer än nödvändigt där av önskar Sapa Building System AB en nyutveckling av bakdörrarna.

Målet är att omkonstruera bakdörrarna RD 3010 och RD 3020 så att de blir lättare, smidigare och billigare genom att konstruera en ny kantskoning till Sapa Building System AB:s befintliga aluminiumlämprofiler.

Arbetet har resulterat i att ritningsunderlag för nya profiler har tagits fram där man valt att limma ihop dörrarnas delar istället för att nita. Detta kan i sin tur leda till att kostnad, vikt och ledtid reduceras.


Sapa is one of the three largest manufactures of aluminum profiles in the world. Sapa Building System AB is a subsidiary to Sapa group and is one of the largest suppliers of building systems in Europe. Some of their products are the rear doors RD 3010 and RD 3020 for trucks and trailers which were from the beginning meant as side doors. As rear doors they are over dimensioned because side doors have higher requirements. As there are no requirements today they weigh and cost more than necessary. Sapa Building System AB wants a new development of the rear doors.

The purpose with this work is to develop new aluminum profiles for the company’s rear doors so that cost, weight and assembly time is lowered.

One of the reasons that Sapa Building System AB could offer this thesis was that they considered it may be profitable to have outsiders develop their products, someone that does not have the “sapa-way-thinking” in their mind. A person that has a different perspective and a different angle of approach of the problem and can provide innovative solutions.

The goal is to reconstruct the rear doors RD 3010 and RD 3020 so that they will be lighter, easier to handle and cheaper by developing a new frame for the existing aluminum profiles that Sapa Building System AB has today.

As a result drawings for new profiles has been made where the doors is assembled with glue instead of using rivets. This may lead to a reduction of price, weight and assembly time.

ENGLISH ABSTRACT: This study was undertaken to determine whether a lack of effective service delivery at the South African Police Service at Delft was due to a lack of community participation and development in Delft. Data was collected through a plurality of research methodologies, including participatory action research by means of interviews with role players and participative observation; a literature study; a diachronical study, application of relevant legislation as a guideline as well as input gained by means of discussion with various experts. The collected data was analysed in relation to the theme, the objective of the study and the research hypothesis. The following observations were made as a result of the study: • The local police station SAPS Delft, by acting as a catalyst, has assisted in the establishment of many structures in the community; and • The erecting of a new police station facilitated community participation and development. The study, having considered the observations and drawn conclusions, has offered a number of recommendations namely: • The erecting of a new police station should take place simultaneously with the development of the township where it is situated. By this means all role players will participate. • The local government and relevant national government departments are of the utmost importance when it comes to addressing the root causes of the problems in the community or when putting alternatives in place. The location of a police station should be such that its convenience and accessibility will result in enhanced and sustainable service delivery, provided that the community capitalises on it. Finally, to ensure success, there must be education and training of both the police and the community, facilitated by the SAPS, government departments and NGOs.
AFRIKAANSE OPSOMMING: Die studie is onderneem om vas te stel of die gebrek aan effektiewe dienslewering by Delft Suid-Afrikaanse Polisie Diens te wyte is aan die gebrek aan gemeenskapsdeelname en -ontwikkeling in Delft. Data is ingesamel deur middel van ‘n pluraliteit van navorsingsmetodologië, insluitend die volgende: deelnemende aksie navorsing deur onderhoude met rol spelers en deelnemende waarneming; ‘n literatuurstudie; ‘n diakroniese studie, toepassing van relevante wetgewing as ‘n riglyn asook insae verkry deur besprekings met ‘n verskeidenheid kenners. Die ingesamelde data is geanaliseer in verhouding tot die tema en die doelwitte van die studie en is vergelyk met die navorsingshipotese. As escolas portuguesas do ensino não superior estão dotadas com infraestruturas e equipamentos que permitem trazer o mundo para dentro da sala de aula, tornando o processo de ensino e de aprendizagem mais rico e motivador para os alunos. A adoção institucional de uma plataforma que segue os princípios da web social, o SAPO Campus (SC), definida pela abertura, partilha, integração, inovação e personalização, pode ser catalisadora de processos de mudança e inovação. O presente estudo teve como finalidade acompanhar o processo de adoção do SC em cinco escolas, bem como analisar o impacto no processo de ensino e de aprendizagem e a forma como os alunos e professores se relacionam com esta tecnologia. As escolas envolvidas foram divididas em dois grupos: o primeiro grupo, constituído por três escolas onde o acompanhamento teve uma natureza mais interventiva e presente, enquanto que no segundo grupo, composto por duas escolas, foram apenas observadas as dinâmicas que se desenvolveram no processo de adoção e utilização do SC. No presente estudo, que se assume como um estudo longitudinal de multicasos, foram aplicadas técnicas de tratamento de dados como a estatística descritiva, a análise de conteúdo e a Social Network Analysis (SNA), com o objetivo de, através de uma triangulação permanente, proceder a uma análise dos impactos observados pela utilização do SC. Estes impactos podem ser situados em três níveis diferentes: relativos à instituição, aos professores e aos alunos. Ao nível da adoção institucional de uma tecnologia, verificou-se que essa adoção passa uma mensagem a toda a organização e que, no caso do SC, apela à participação coletiva num ambiente aberto onde as hierarquias se dissipam. Verificou-se ainda que deve implicar o envolvimento dos alunos em atividades significativas e a adoção de estratégias dinâmicas, preferencialmente integradas num projeto mobilizador. A adoção do SC foi ainda catalisadora de dinâmicas que provocaram mudanças nos padrões de consumo e de produção de conteúdos bem como de uma atitude diferente perante o papel da web social no processo de ensino e aprendizagem. As conclusões apontam ainda no sentido da identificação de um conjunto de fatores, observados no estudo, que tiveram impacto no processo de adoção como o papel das lideranças, a importância da formação de professores, a cultura das escolas, a integração num projeto pedagógico e, a um nível mais primário, as questões do acesso à tecnologia. Algumas comunidades construídas à volta do SAPO Campus, envolvendo professores, alunos e a comunidade, evoluíram no sentido da autossustentação, num percurso de reflexão sobre as práticas pedagógicas e partilha de experiências.
Portuguese schools are currently fully equipped with resources and facilities that make it possible to bring the world into the classroom, thus enhancing both the teaching and learning dynamics, as well as increasing students’ motivation. The institutional adoption of platforms, such as SAPO Campus (SC), that follow the core principles of the social web, and are defined by openness, sharing, integration, innovation and personalization, can become the driving force for change and innovation within schools. This study’s main goal was to monitor how five different schools adopted and implemented the SC platform, as well as analyse its impact on the teaching and learning processes and how students and teachers responded to this technology. The schools included in this research were divided into two groups: the first group consisted of three schools in which there was a more direct and proactive intervention regarding the adoption and use of the platform, whereas the second group was made up of two schools in which these processes were only observed, without the direct involvement of researchers. Considering this was a longitudinal survey focusing on multiple cases, different data processing techniques, such as descriptive statistics, content analysis and Social Network Analysis (SNA) were used throughout the study. The data collected was then triangulated and validated making it possible to analyse the impact of SC and how it was used. These impacts can be placed at different levels: those concerning the institution, teachers and students. Regarding technology appropriation, at an institutional level the platform’s endorsement sends out a message to the organization, calling out for collective engagement and participation within an open environment, where hierarchies become less apparent. It also became clear that students must be engaged in significant activities, as the platform’s use strives when dynamic strategies are used, especially within the scope of stimulating projects. SC’s adoption was also a catalyst for dynamics that changed consumption and production patterns and fostered a different attitude towards the role social web plays in the teaching and learning process. The results also point towards several elements that were observed throughout the study as having had an impact in the platform’s adoption. These elements include the role of leaderships, the importance of teacher training, school culture, integration into a pedagogical project and, at a more basic level, questions regarding the access to technology. Some communities that emerged from and around SAPO Campus and that involved teachers, students and the community, have become self-sufficient, having based their growth on the reflection on pedagogical practices and the sharing of experiences.

Galärvarvet är på många sätt en utmärkt plats för en ny och framträdande kulturbyggnad i Stockholm. Samtidigt utgöt operans komplexa och omfattande program en stor utmaning. I detta projekt löses konflikten mellan program och plats meddels en långsmal byggnad som bevarar naturvärden och rörelsemönster.
Galarvarvet is in many ways an great site for a new and prominent cultural building in Stockholm. At the same time the complexity and size of the program of an opera poses a great challenge. In this project the conflict between program and site is solved with a stretched out building that preserves environmental values and movements.
