Full textThe association between podoplanin and cellular proliferative activity has been demonstrated but is still source of debate. The aim of this study was to investigate the expression of podoplanin in ameloblastomas and remnants of odontogenic epithelium (ROE) from dental follicles of unerupted teeth and verify the relation between the podoplanin expression and proliferative activity of the odontogenic cells. Thirty-three patients submitted to the incisional biopsy or resection of ameloblastoma and 32 patients submitted to the extraction of unerupted tooth were selected for analysis. Archived paraffin-embedded ameloblastomas and dental follicles specimens were sectioned and stained with anti-human podoplanin and anti-Ki-67 antibodies. The podoplanin expression by odontogenic epithelial cells was evaluated using a score method and the Ki-67 labelling index was obtained by the percentage of positive odontogenic cells (at least 500 cells/ameloblastoma and all ROE cells). All ameloblastomas showed podoplanin expression in ameloblast-like cells of the epithelial islands. Weak or absent immunostaining was observed in the central cells resembling stellate reticulum. Podoplanin expression in ROE was mainly found in the cells in contact with the connective tissue. Membranous expression of podoplanin in ameloblastomas was stronger than its expression in ROE (!=0.001). Statistically significant difference between cytoplasmic and membranous expression of podoplanin in ROE was observed (!=0.001). The index of cellular odontogenic proliferative activity, verified by Ki-67 expression, was highest in ameloblastomas when compared with ROES (!<0.001). No statistically significant correlation between podoplanin and cellular odontogenic proliferative activity in ameloblastomas and dental follicles was found (!>0.05). These results support the evidence that there is no connection between podoplanin expression and odontogenic cellular proliferative activity in ameloblastomas and reinforce that the exact role of this protein in benign odontogenic tumor needs to be elucidated.

Full textThe yolk sac is one of the extra-embryonic membranes which plays an important role in early embryonic survival and serves as source of nutrition during the period where in the placenta is not completely true formed. The yolk sac is a likely source of stem cells, which have first blood cells during development in mammals, the red blood cells, which express transcription factors that specify these hematopoietic cells to their destination. This study aimed to identify and characterize hematopoietic stem cells from the yolk sac of bovine embryos at different stages of pregnancy, which are collected at a local slaughterhouse. For a description of the macroscopic and cellular culture of yolk sac cells, are as follows bovine embryos were divided into groups of gestational age: Group I (25 to 29 days), Group II (30 to 34 days), Group III (35 to 39 days ), Group IV (40 to 44 days) and Group V (45 to 50 days) which stayed longer in culture and showed the formation of cell clusters, different from groups IV and V (40-45 days) in few that remained days in culture and showed no cell clumps. This divergence is related to gestational age (45 to 50 days), during which begins regression of the yolk sac. In flow cytometry groups I, II and III (25 to 39 days) had similar characteristics, high expression of hematopoietic markers (CD34, CD90 and CD117). For the groups IV and V (40 to 50 days) it is observed a decrease in expression of CD117 and CD34 (hematopoietic markers) and in group V were increased expression of CD45 (leukocyte marker), confirming that these cells are not keeping Undifferentiated cells are shown to be resistant to cryopreservation, capable of forming colonies in methylcellulose matrix showed the formation of colonies after 14 days in culture morphology and to blood cells (lymphocytes and monocytes) was confirmed by cytology cell. In gene expression was low GATA3 gene expression, different levels of expression between the groups for the marker and RUNX1 ANXA5. Our most significant findings confirmed the isolation and identification of hematopoietic cells from the bovine embryo yolk sac, therefore, it is feasible and an effective way of obtaining stem cells for future applications in cell therapy and gene.

As Lobato modified his texts a lot, especially the ones produced in the beginning of his career as a children's writer, we add, in our study, a comparison between some editions of O Saci: 1st (1921), 3rd (1928), 6th (1938) and 10th (definitive edition, 1947)

Full textThe main goal of this study was to describe morphology and the early gestational development of 15 to 70 day-old bovine embryos obtained by natural mating and 35 day-old bovine embryos obtained by in vitro fertilization (IFV) technique. One hundred and forty-one concepts originated by natural mating and seven by IVF technique were used. All concepts were dissected, macroscopically measured and photographed. Extraembryonic membranes were cut in 5 cm2 fragments and fixed in 4% paraformoldehyde for light and Scanning Electron Microscopy (SEM) and in 2.5% glutaraldehyde for Transmition Electron Microscopy (TEM). Ali membranes showed different stages of development during analyzed periods. The beginning of allantois macroscopic vascularization, the attempt of fusion between allantois and chorium membranes and the effective development of the first cotyledons were observed in 30-to-40 day-old embryos with 1.9± 0.24 cm of Crown-Rump (CR) length. The average CR, the embryos and gestational sac weight, the chorion and amnion length increased during gestation. The allantoic epithelia showed a cellular dimorphism with 0.9 em CR, however, the maturation has not happened until the foetal length of 7.2 em CR. The trophoblast showed different levels of mononucleate cells and binucleate giant cells. The yolk sac persisted until 70 days of gestation and the epithelium seemed functionally activated until 50 days of gestation. The CR and yolk sac size were the only measured macroscopic parameter showingdifference between natural mating and FIV concepts. Yolk sac size presented average values of 5.53 cm and 1.07 cm for natural mating and FIV concepts, respectively. Therefore, it is necessary to analyze a higher number of FIV embryos to corroborate that observed yolk sac size difference. All results obtained in this study suggest the existent of an active vitelline placenta, important to the gestational maintenance and temporarily establish between the transitory choriovitelline placenta and the permanent chorioallantoic placenta.

Full textThe yolk sac is the main source of embryo nutrition during the period that true placenta are not still formed and responsible by hematopoieses because from it were developed the blood cells, as a part of embryo primitive circulatory system. We aimed to characterize the transitional area between the yolk sac and the primitive intestine by light microscopy, transmission electron microscopy and detection of germ cell lines by Oct-4 immunohistochemistry. Were done in all embryos serial sections to permit the identification for all structures. Were possible identify the yolk sac connection with primitive intestine composed by many blood islet corresponding to relation with yolk and mesenchymals cells. Inside the vessels were identified hemangioblasts. The layer corresponding to mesenchyme is thin and with elongated cells and the vitellinic membrane endoderm is composed by large, uni or binucleate endodermic cells. This shape is similar to a gland related to their architecture, vessels islets and tissue layers. Primitive intestine epithelium begins their differentiation with stratified columnar cells and undifferentiated mesenchymals cells. This transitional area is thin related to their lumen with irregular shape and constituted by delicate lumen basement and hemangioblast. Inside the mesenchyme there are many smaller capillaries observed with hemangioblast in it. Embryos and fetus submitted to immunohistochemistry and were verified positive cells like metanefrons, gonads, hepatic parenchyma and yolk sac.

O presente trabalho refere-se ao SACI - Sistema de Apoio na Avaliação de Distúrbios do Crescimento Infantil, que se propõe a realizar o diagnóstico precoce de distúrbios associados à falência do crescimento infantil, em crianças com até 2 anos. O SACI explora o raciocínio probabilístico em sistemas especialistas, tendo sido desenvolvido para a plataforma Windows, no ambiente Delphi 5, utilizando as DLL's da shell Netica para esse ambiente de programação. O SACI permite que o diagnóstico seja realizado, de forma a disponibilizar os dados relacionados à evolução do crescimento da criança graficamente, e que sejam investigadas as suspeitas de perda de peso e altura da criança, por meio de evidências a serem observadas. Na avaliação realizada junto a médicos pediatras, especialistas do domínio de conhecimento, verificou-se que o SACI é de fácil utilização, apresentando resultados satisfatórios às diversas consultas realizadas.

INTRODUÇÃO: A correção endovascular do aneurisma de aorta abdominal (EVAR) modificou o tratamento desta patologia, reduzindo a mortalidade e as complicações, á curto prazo quando comparada à cirurgia aberta. Ainda assim, os pacientes necessitam de seguimento rigoroso a fim de reconhecer as possíveis complicações. O crescimento do diâmetro do saco aneurismático pós-EVAR está relacionado ao risco de rotura ou necessidade de reabordagem, sendo que alguns fatores pré-operatórios podem prever esse aumento. OBJETIVO:Identificar se os sintomas pré-tratamento EVAR podem ser um fator preditivo para a continuidade da expansão do saco aneurismático após tratamento, no seguimento em 12 meses. MATERIAL E MÉTODOS: Estudo retrospectivo realizado através da coleta de dados do prontuário dos pacientes em seguimento na Faculdade de Medicina de Botucatu e que preencheram critérios de inclusão. Os pacientes foram separados em dois grupos de estudos: G1- Pacientes assintomáticos quanto a presença do aneurisma de aorta infrarenal; G2- Pacientes sintomáticos quanto a presença do aneurisma de aorta infrarenal. Todos os grupos foram acompanhados por 12 meses e avaliados quanto ao crescimento do saco aneurismático após o procedimento endovascular, através de AngioTC e Duplex Scan e coleta do exame “proteína C Reativa” para avaliação inflamatória relacionada ao EVAR. RESULTADOS: Foram 9 estudados 112 pacientes. A faixa etária apresentou uma média de 68,6 anos. 80% eram do sexo masculino e 95% brancos. A maioria dos pacientes eram hipertensos e fumantes, 74,1% e 67% respectivamente. No total houve 22,3% de endoleak e o aumento do saco aneurismático foi de 26,8%. Avaliando todos os pacientes, ocorreu diminuição do diâmetro do AAA em média de 0,8cm, entre o pré e pós-operatório. Observou se que 25,3% dos assintomáticos e 30% dos sintomáticos tiveram crescimento do saco aneurismáticos. A presença de endoleak ocorreu em 19% dos assintomáticos e 41,2% dos sintomáticos. No total, 40 % dos pacientes que evoluíram com aumento do saco aneurismático apresentavam endoleak. CONCLUSÃO: Os pacientes que tiveram dor no pré-operatório, portanto, sintomáticos, apresentaram uma taxa numérica maior do crescimento do saco aneurismático em relação ao grupo que não apresentava sintomas, mas este achado não foi estatisticamente significante. A presença do endoleak não foi a única causa do crescimento dos AAAs. Não houve diferença do PCR colhido no pré-operatório em relação ao crescimento do AAA em ambos os grupos. A hipertensão arterial foi a comorbidades mais frequente na nossa casuística.
BACKGROUND: The endovascular repair of abdominal aortic aneurysm (EVAR) modified its treatment, reducing the mortality and complications in short-term compared to open repair. Even though, patients need strict follow-up to identify possibly complications. The sac aneurysm enlargement after EVAR is related to the risk of rupture or reintervention, with some preoperative factors can predict this enlargement. OBJECTIVE: Identify if the AAA symptons, especially pain, are predictors of abdominal aortic aneurym sac enlargement after EVAR. METHODS: Retrospective study through the gathering of data about the patientes in follow-up in Botucatu School Medicine. The patients were separated in two groups: G1 – assymptomatics in the preoperative; G2 – syptomatic in the preoperative. Both groups were followed-up for 12 months to the sac enlargement through AngioCT, duplex scan and Reative-C-Protein. RESULTS: 112 patientes were analysed. The age average was 68,6 years. 80% were male and 95% white. Most of them were hypertensive and smoker, 74,1% and 67% respectively. There was 22,3% of endoleaks and 26,8% sac enlargement ratio. 25,3% of the assymptomatics and 30% of the symptomatics had sac enlargement. 19% of the assymptomatics compared with 41,2% of the symptomatics had endoleak. 40% of the patients with sac enlargement had endoleaks. CONCLUSION: The symptomatics preoperative patients has a higher risk compared with assymptomatic group. The endoleak is not the only 11 reason for the AAA sac enlargement. There was no difference between the groups G1 and G2 related to AAA sac enlargement and Reative-CProtein. Hypertension was the most prevalent comorbidity in our study.

Full textMurine rodents are well investigated in regard to placentation, but data for other groups such as cricetids including New World mice or Sigmodontinae are sparse. Recently the placenta and fetal membranes are regarded to be promising sources for obtaining stem cells. In particular, the yolk sac is structurally diverse and shows important functions throughout gestation. For this reason, this study aims to describe the chorioallantoic and yolk sac placentation in Necromys lasiurus. In addition, the potential of the yolk sac as a source for mesenchymal stem cells that are not well studied so far will be evaluated. In total, 10 individuals from early gestation to near term were investigated by means of histology, immunohistochemistry and electron microscopy as well as cell culture and differentiation, flow citrometry and immunocytochemistry. The discoidal chorioallantoic placenta was organized in a labyrinth zone, junctional zone, and decidua. The labyrinth was most import for maternal-fetal exchange processes. It possessed a hemotrichorial barrier of about 2.41 µm thickness near term. The junctional zone included syncytial areas and cytotrophoblasts. Trophoblast giant cells were located between the junctional zone and decidua as well as in the lateral margins of the placenta. The morphology of the inverted visceral yolk sac varies according to its location and relationship with the placenta and uterus. When cultured, the adherent cells of the yolk sac formed fibroblastoid colonies (92.13%) and expressed mesenchymal stem cells markers, and some hematopoietic precursor cells markers. They showed successful osteogenic, adipogenic, and chondrogenic differentiation and did not develop tumors when transferred to nude mice. Thus, these cells resulted as stem cells with promising therapeutic values for cell therapy.

Full textThe yolk sac is an embryonic attachment present in all animal species, which function is to keep the primary development of the fetus until the placenta assumes this function. Moreover, it plays important roles as: nutrition of the embryo, protein synthesis, phagocytic activity, transfer of materials and hematopoiesis. The use of embryonic stem cells generates many discussions because of the ethical and religious issues associated with the obtaintion of these cells. Also the relationship of low plasticity of adult stem cells motivates researchers to explore other sources of stem cells. Thus, cells from tissues such as the embryonic anexes call more attention, due to its ease of access, rapid growth and good plasticity. Our group has shown that cells of the yolk sac has good ability to proliferate and are multipotent. Several researchers have shown that the period in which the cells are obtained can be critical for differentiation. Therefore, in this project we aimed to establish and characterize stem cells from dog yolk sac in two stages of pregnancy, in order to analyze comparatively the expression profile of mesenchymal, hematopoietic and plurypotency markers, and study their behavior and plasticity potencial in vitro. In this study we observed that yolk sac cells obtained from pregnancies at 40 and 50 days adhered to plastic, had fibroblast like-morphology, good cell proliferation and differentiated into adipocytes and osteocytes and expressed some mesenchymal markers, therefore suggesting to be a mesenchymal stem cell. Also, they did not lead to tumor formation when injected into immunosuppressed mice, nude.
