Abstract:
Stumpy irrigation water availability is extremely important for sugarcane production in Pakistan today. This issue is rising inversely to river flow due to inadequate water distribution and an uneven rainfall pattern. Sugarcane growth faces a shortage of available water for plant uptake due to the low water–holding capacity of sandy loam soil, particularly under conventional flood irrigation methods. To address this problem, sugarcane clones were evaluated for their agronomic and physiological traits under conditions of low water availability in sandy loam soil. Ten cane genotypes, HSF–240, SPF–213, CPF–249, CP 77–400, S2008–FD–19, S2006–US–469, S2007–AUS–384, S2003–US–633, S2003–US–127, and S2006–US–658, were exposed to four levels of water deficit created through skip irrigations. These deficit levels occurred during the 9th, 11th, 13th, and 16th irrigations at alternate deficit levels between 2020 and 2022. Physiological data were collected during the tillering and grand growth stages (elongation) in response to the water deficit. The sugarcane clones S2006–US–658, S2007–AUS–384, and HSF–240 exhibited resistance to low water availability at both the tillering and grand growth stages. Following them, genotypes S2006–US–658, S2007–AUS–384, and HSF–240 performed better and were also found to be statistically significant. Clones susceptible to water deficit in terms of growth and development were identified as CP 77–400, S2008–FD–19, S2006–US–469, and S2003–US–633. These genotypes showed reduced photosynthetic rate, transpiration rate, stomatal conductance, relative water content, cane yield, and proline content under stressed conditions. Therefore, genotypes S2006–US–658, S2007–AUS–384, and HSF–240 were better performers concerning physiological traits under water deficit and sandy loam soil in both years. Moreover, a significant positive correlation was assessed between agronomic traits and photosynthetic rats. This study highlights that sugarcane can sustain its growth and development even with less irrigation frequency or moisture availability, albeit with certain specific variations.

Abstract:
Sugarcane is the crop that have the quality to sprout after its first harvesting, phenomenon known as ratooning. Among all other major crops, sugarcane ratooning is widely acceptable to farmers of the Punjab. The more the tillers grew, the higher are the chances of a greater number of stalks to be harvested. It is proven that more the number of stalks, the thinner is the diameter, finally results in more stalks and high production. Similarly, higher number of canes produce favorable ratoon. In this experiment, performance of Seven promising sugarcane varieties/clones along with the check variety CPF-246, were tested for their ratoon ability capacity at farmer’s field at Toba Tek Singh during the year 2017-18. It has been found that one of the varieties S2008-FD-19 produced that maximum ratoon yield, U2003-US-633 produced the maximum sugar recovery and S2003-US-133 produced maximum number of mill able canes. The observing of the current analysis also revealed that S2003-US-633 and S2003-US-133 have significant potential for boom sugar recovery through breeding programs and by improving the production techniques.

Las variedades de trigo harinero Cortazar S94, Bárcenas S2002, Urbina S2007 y Maya S2007 fueron liberadas para siembras en condiciones de riego en “El Bajío”. Estas variedades mantienen resistencia a diferentes razas de roya amarilla que se encuentran en el país; sin embargo, se desconocen las bases genéticas de su resistencia. Para determinar la genética de la resistencia, estas variedades se cruzaron con el progenitor susceptible ‘Avocet- YrA’, y la determinación de similitud de genes se realizó mediante pruebas de alelismo. Las cuatro variedades y las familias F3 de cada cruza se evaluaron durante verano de 2009 en el Campo Experimental ‘Valle de México’ (INFAP-CEVAMEX) Chapingo, México, bajo una epifitia artificial creada con el aislamiento MEX96.11. Con los resultados de la evaluación de familias F3 se determinó que la resistencia de planta adulta a roya amarilla en Cortázar S94, Bárcenas S2002 y Maya S2007 está condicionada por dos ó tres genes de efectos aditivos, mientras que en Urbina S2007 la resistencia está condicionada por tres o cuatro genes de la misma naturaleza. La ausencia de familias completamente susceptibles en las cruzas entre las variedades resistentes indica la existencia de al menos un gen de resistencia en común.

Topographical areas better adapted for sugar cane cultivation must be identified and preserved, for this study Indus valley river delta in South Punjab represents promising features for prosperous sugarcane cultivation (lower water pumping costs due to higher water tables, sugarcane being flood resistant crop in Pakistan. Eight sugarcane varieties and eight promising clones were grown on different locations in Punjab at farmer’s field using RCBD with three replications. The objective of this study is to evaluate varieties under different ecological zones. These clones were tested for their proficiency at four different locations during 2018-19. The “Thal” and Southern Punjab viz; 142/TDA Lalazar Layyah, Indus Sugar mills Rajanpur, Ashraf Sugar Mills Bahawalpur and Adam Sugar Mills Chishtian. The data on germination %, tillers / plants, Number of mill able canes, canes yield tones/ ha and commercial cane sugar (CCS) were recorded during the course of study. It is obvious from pooled mean of four locations that cane yield of clone S2002-US-133 gave 84.13 % higher cane yield as compared the check variety CPF 248 as far as CCS% is concerned, sugarcane clones S200-US-633 and S200-US-133 gave the higher sugar recovery that is 15.31 % & 15.23 % respectively more than check variety. Whereas S200-US-658 and HSF240 have the lowest CCS% 1.22 & 1.14 % less than check.

The activity of the cloned renal K+ channel (ROMK2) is dependent on a balance between phosphorylation and dephosphorylation. There are only three protein kinase A (PKA) sites on ROMK2, with the phosphorylated residues being serine-25 (S25), serine-200 (S200), and serine-294 (S294) (Z.-C. Xu, Y. Yang, and S. C. Hebert. J. Biol. Chem. 271: 9313–9319, 1996). We previously mutated these sites from serine to alanine to study the contribution of each site to overall channel function. Here we have studied each of these single PKA site mutants using the single-channel configuration of the patch-clamp technique. Both COOH-terminal mutations at sites S200A and S294A showed a decreased open channel probability ( P o), whereas the NH2-terminal mutation at site S25A showed no change in P o compared with wild-type ROMK2. The decrease in P o for the S200A and S294A mutants was caused by the additional presence of a long closed state. In contrast, the occurrence of the S25A channel was ∼66% less, suggesting fewer active channels at the membrane. The S200A and S294A channels had different kinetics compared with wild-type ROMK2 channels, showing an increased occurrence of sublevels. Similar kinetics were observed when wild-type ROMK2 was excised and exposed to dephosphorylating conditions, indicating that these effects are specifically a property of the partially phosphorylated channel and not due to an unrelated effect of the mutation.

Abstract:
La variedad Ibis M2016 es un de trigo harinero de gluten medio fuerte, que fue obtenida en el Programa de Mejoramiento Genético de Trigo del Campo Experimental Bajío por hibridación, mediante una cruza simple entre los genotipos Colibri y Finsi, su historia de selección es TC070080-2R-0C-0R-1CSE-0R. En parcelas de productores de El Bajío superó en rendimiento de grano a Cortazar S94, con 23.8% y a las variedades Alondra F2014, Luminaria F2012, Bárcenas S2002, Maya S2007 y Urbina S2007 con 3.4, 45.7, 41, 21.4 y 18.7%, respectivamente. Es moderadamente resistente a la roya amarilla, cuando Cortazar S94 alcanzó el 40% de daño por la enfermedad, Ibis M2016 presentó daños que van del 10 al 15%. La variedad se desarrolló del año 2007, cuando se realizó la cruza, en 2021 se liberó como variedad comercial. Ibis M2016 se puso a disposición de los productores para satisfacer la demanda de variedades de gluten medio fuerte, resistentes a royas y con rendimientos superiores al de las variedades comerciales. Ibis M2016 se recomienda para siembras en la región de El Bajío que presentó alturas de 1 500 a 1 800 msnm y comprende parte de los estados de Guanajuato, Michoacán, Jalisco y Querétaro.

Brucella spp. são bactérias gram-negativas, intracelulares facultativas que são patogênicas para muitas espécies de mamíferos causando a brucelose, uma zoonose difundida mundialmente. Por isso a busca de alternativas de controle mais eficientes se faz necessário como o desenvolvimento de novas cepas que possam ser testadas como potenciais imunógenos. Neste estudo realizou-se a deleção do gene virB10 da cepa S2308 de Brucella abortus gerando uma cepa knockout provavelmente incapaz de produzir a proteína nativa correspondente. O gene virB10 faz parte de um operon que codifica para um sistema de secreção do tipo IV, essencial para a sobrevivência intracelular e multiplicação da bactéria em células hospedeiras. A deleção foi realizada pela construção do plasmídeo suicida pBlue:virB10:kan e eletroporação deste em células eletrocompetentes de B. abortus S2308, ocorrendo a troca do gene selvagem pelo gene interrompido, com o gene de resistência a canamicina, por recombinação homóloga dupla. Camundongos BALB/c foram inoculados com as cepas S19, RB-51, ΔvirB10 de B. abortus e B. abortus S2308 selvagem; os resultados demonstraram que camundongos BALB/c inoculados com S19 e camundongos BALB/c inoculados com S2308 apresentaram queda mais rápida de linha de tendência, quando comparadas aos demais grupos, para recuperação bacteriana (RB) e peso esplênico (PE) respectivamente. Os grupos que receberam ΔvirB10 S2308 de B. abortus e RB-51 demonstraram comportamento semelhante para ambas as características. Na sexta semana após a inoculação, os resultados para RB (log de UFC ± desvio padrão) e PE (peso esplênico ± desvio padrão), respectivamente, mostraram: grupos inoculados com as cepas S2308 (4,44±1,97 e 0,44±0,11), S19 (1,83±2,54 e 0,31±0,04), RB-51 (0,00±0,00 e 0,20±0,01) e ΔvirB10 S2308 (1,43±1,25 e 0,19±0,03). Considerado o clearance bacteriano, todos os grupos diferiram estatisticamente do grupo que recebeu S2308 (p<0,0001), o grupo inoculado com ΔvirB10 S2308 de B. abortus foi semelhante ao grupo S19 (p=0,4302) e diferente do grupo RB-51 (p=0,0063). A avaliação da persistência revelou que o gene virB10 é essencial para a manutenção da virulência da bactéria. Os resultados obtidos possibilitarão que outras pesquisas sejam realizadas avaliando o potencial imunogênico desta cepa mutante.

In the previous work, two new nematic liquid crystal (NLC) mixtures, E7-2 and S200-2, were produced by adding eight LC monomers to two commercial LCs S200 and E7, respectively. At λ = 589 nm, the birefringence (Δn) characteristics of the two LC nematic mixtures E7-2 (Δn = 0.260) and S200-2 (Δn = 0.298) are greater than those of the commercial LC E7 (Δn = 0.224) and S200 (Δn = 0.266). The properties (TN-I, ε//, Δɛ, K11, and K33) of these four NLCs were measured. A double-layer metal loop arrays modulation structure based on metamaterial (MM) metal–dielectric–metal (MDM) was designed and fabricated for use in the THz frequency range. The results show that the LC mixtures E7-2 and S200-2 have greater modulation depth (MD) and less modulation insertion loss (IL) than E7 and S200 at THz frequencies. The results show that LC mixtures have significant potential for designing active tunable LC-based devices in the THz and visible light range.

Brucellosis is a globally distributed zoonotic disease affecting animals and humans, and current antibiotic and vaccine strategies are not optimal. The surface-exposed protein Omp25 is involved in Brucella virulence and plays an important role in Brucella pathogenesis during infection, suggesting that Omp25 could be a useful target for selecting potential therapeutic molecules to inhibit Brucella pathogenesis. In this study, we identified, we believe for the first time, peptides that bind specifically to the Omp25 protein of pathogens, using a phage panning technique, After four rounds of panning, 42 plaques of eluted phages were subjected to pyrosequencing. Four phage clones that bound better than the other clones were selected following confirmation by ELISA and affinity constant determination. The peptides selected could significantly inhibit Brucella abortus 2308 (S2308) internalization and intracellular growth in RAW264.7 macrophages, and significantly induce secretion of TNF-α and IL-12 in peptide- and S2308-treated cells. Any observed peptide (OP11, OP27, OP35 or OP40) could significantly inhibit S2308 infection in BALB/c mice. Moreover, the peptide OP11 was the best candidate peptide for inhibiting S2308 infection in vitro and in vivo. These results suggest that peptide OP11 has potential for exploitation as a peptide drug in resisting S2308 infection.

Abstract:
Recent decreases in costs, and improvements in performance, of silicon array detectors open a range of potential applications of relevance to plant physiologists, associated with spectral analysis in the visible and short-wave near infra-red (far-red) spectrum. The performance characteristics of three commercially available ‘miniature’ spectrometers based on silicon array detectors operating in the 650–1050-nm spectral region (MMS1 from Zeiss, S2000 from Ocean Optics, and FICS from Oriel, operated with a Larry detector) were compared with respect to the application of non-invasive prediction of sugar content of fruit using near infra-red spectroscopy (NIRS). The FICS–Larry gave the best wavelength resolution; however, the narrow slit and small pixel size of the charge-coupled device detector resulted in a very low sensitivity, and this instrumentation was not considered further. Wavelength resolution was poor with the MMS1 relative to the S2000 (e.g. full width at half maximum of the 912 nm Hg peak, 13 and 2 nm for the MMS1 and S2000, respectively), but the large pixel height of the array used in the MMS1 gave it sensitivity comparable to the S2000. The signal-to-signal standard error ratio of spectra was greater by an order of magnitude with the MMS1, relative to the S2000, at both near saturation and low light levels. Calibrations were developed using reflectance spectra of filter paper soaked in range of concentrations (0–20% w/v) of sucrose, using a modified partial least squares procedure. Calibrations developed with the MMS1 were superior to those developed using the S2000 (e.g. coefficient of correlation of 0.90 and 0.62, and standard error of cross-validation of 1.9 and 5.4%, respectively), indicating the importance of high signal to noise ratio over wavelength resolution to calibration accuracy. The design of a bench top assembly using the MMS1 for the non-invasive assessment of mesocarp sugar content of (intact) melon fruit is reported in terms of light source and angle between detector and light source, and optimisation of math treatment (derivative condition and smoothing function).

The Brucella AMOS PCR assay was previously developed to identify and differentiate specific Brucella species. In this study, an abbreviated Brucella AMOS PCR test was evaluated to determine its accuracy in differentiating Brucella abortus into three categories: field strains, vaccine strain 19 (S19), and vaccine strain RB51/parent strain 2308 (S2308). Two hundred thirty-one isolates were identified and tested by the conventional biochemical tests and Brucella AMOS PCR. This included 120 isolates identified as B. abortus S19, 9 identified asB. abortus strain RB51, 57 identified as B. abortus biovar 1, 15 identified as B. abortus bv. 2, 1 identified as B. abortus bv. 2 (M antigen dominant), 7 identified as B. abortus bv. 4, and 22 identified asB. abortus S2308 and isolated from experimentally infected cattle. The Brucella AMOS PCR correctly identified each isolate as RB51/S2308, S19, or a field strain of Brucella.

Quantitative measures of radiation-induced breast stiffness are required to support clinical studies of novel breast radiotherapy regimens and exploration of personalised therapy, however, variation between shear-wave elastography (SWE) machines may limit the usefulness of shear-wave speed (cs) for this purpose. Mean cs measured in four healthy volunteers’ breasts and a phantom using 2D-SWE machines Acuson S2000 (Siemens Medical Solutions) and Aixplorer (Supersonic Imagine) were compared. Shear-wave speed was measured in the skin region, subcutaneous adipose tissue and parenchyma. cs estimates were on average 2.3% greater when using the Aixplorer compared to S2000 in vitro. In vivo, cs estimates were on average 43.7%, 36.3% and 49.9% significantly greater (p << 0.01) when using the Aixplorer compared to S2000, for skin region, subcutaneous adipose tissue and parenchyma, respectively. In conclusion, despite relatively small differences between machines observed in vitro, large differences in absolute measures of shear wave speed measured were observed in vivo, which may prevent pooling of cross-machine data in clinical studies of the breast.

Daily administration of melatonin or S20098, a melatonin agonist, is known to entrain the free-running circadian rhythms of rats. The effects of the duration of administration on entrainment were studied. The animals demonstrated free-running circadian rhythms (running-wheel activity, body temperature, general activity) in constant darkness. Daily infusions of melatonin or S20098 for 1, 8, or 16 h entrained the circadian rhythms to 24 h. Two daily infusions of 1 h (separated by 8 h) entrained the activity peak within the shorter time interval. The entraining properties of melatonin and S20098 were similar and were affected neither by pinealectomy nor by infusion of 1- or 8-h duration. However, with 16-h infusion, less than half of the animals became entrained. Once entrained, the phase angle between the onset of infusion and the rhythms (onset of activity or acrophase of body temperature) increased with the duration of infusion. Before entrainment, the free-running period increased with the duration of infusion, an effect that was not predictable from the phase response curve.

ABSTRACT Brucella abortus is an intracellular pathogen that replicates within a membrane-bounded compartment. In this study, we have examined the intracellular pathway of the virulent B. abortus strain 2308 (S2308) and the attenuated strain 19 (S19) in HeLa cells. At 10 min after inoculation, both bacterial strains are transiently detected in phagosomes characterized by the presence of early endosomal markers such as the early endosomal antigen 1. At ∼1 h postinoculation, bacteria are located within a compartment positive for the lysosome-associated membrane proteins (LAMPs) and the endoplasmic reticulum (ER) marker sec61β but negative for the mannose 6-phosphate receptors and cathepsin D. Interestingly, this compartment is also positive for the autophagosomal marker monodansylcadaverin, suggesting that S2308 and S19 are located in autophagic vacuoles. At 24 h after inoculation, attenuated S19 is degraded in lysosomes, while virulent S2308 multiplies within a LAMP- and cathepsin D-negative but sec61β- and protein disulfide isomerase-positive compartment. Furthermore, treatment of infected cells with the pore-forming toxin aerolysin from Aeromonas hydrophila causes vacuolation of the bacterial replication compartment. These results are compatible with the hypothesis that pathogenic B. abortus exploits the autophagic machinery of HeLa cells to establish an intracellular niche favorable for its replication within the ER.

The etherification of glycerol with propylene over acidic heterogeneous catalysts, Amberlyst-15, S100, and S200 resins, produced mono-propyl glycerol ethers (MPGEs), 1,3-di- and 1,2-di-propyl glycerol ethers (DPGEs), and tri-propyl glycerol ether (TPGE). The propylation of glycerol over Amberlyst-15 yielded only TPGE. The glycerol etherification with 1-butene over Amberlyst-15 and S200 resins produced 1-mono-, 2-mono-, 1,2-di-, and 1,3-di-butyl glycerol ethers (1-MBGE, 2-MBGE, 1,2-DBGE, and 1,3-DBGE). The use of Amberlyst-15 resulted in the propylation and butylation of glycerol with higher yields than those obtained from the S100 and S200 resins. The PGEs, TPGE, and BGEs were evaluated as cold flow improvers and octane boosters. These alkyl glycerol ethers can reduce the cloud point of blended palm biodiesels with diesel. They can increase the research octane number and the motor octane number of gasoline.

The inhibition of squalene epoxidase by the allylamine antimycotic agents naftifine and compound SF 86-327 was investigated, with particulate enzyme preparations from the pathogenic yeasts Candida albicans and Candida parapsilosis and from rat liver. Both naftifine and compound SF 86-327 were potent inhibitors of the Candida epoxidases and showed apparently non-competitive kinetics with respect to the substrate squalene. The Ki values for naftifine and compound SF 86-327 in the C. albicans system were 1.1 microM and 0.03 microM respectively. The C. parapsilosis enzyme was slightly more sensitive to inhibition. Varying the concentrations of cofactors or the soluble cytoplasmic fraction (S200) had no effect on the inhibition. The epoxidase from rat liver was much less sensitive (Ki for compound SF 86-327 was 77 microM). The inhibition was also qualitatively different from that in Candida, being competitive with respect to squalene and also with respect to the S200 fraction. S200 fraction derived from C. albicans also antagonized the inhibition of the epoxidase from liver, but the liver S200 fraction did not affect inhibition of the Candida enzyme by compound SF 86-327. There was no evidence for an irreversible or mechanism-based inhibition of either the fungal or the mammalian epoxidase. The selective inhibition of squalene epoxidase was sufficient to account for the known antimycotic action of the compounds.

Su rendimiento superó en 16.8 y 18.6% a las variedades de trigos suaves Urbina S2007 y Maya S2007, liberadas en 2007 y hasta con 21% a Cortazar S94, la variedad más sembrada en El Bajío. obtuvo el título de obtentor, quedando protegida hasta el 18 de mayo de 2032; asimismo, está registrada y publicada en el catálogo nacional de variedades vegetales, con lo cual es posible considerarla en los programas de calificación y producción de semillas.
