Abstract:
This thesis deals with the removal of noise in speech recordings that have been recorded in an MRI environment. For this purpose, the Nvidia RTX Voice technology, the VST plug-in module Noisereduce and a self-designed method of subtractive de-noising of recordings are used. A program with a simple graphical interface in Python is implemented within the work to retrieve the recordings and then de-noise them using the proposed methods. The work includes measurements in a magnetic resonance environment with two microphones. The quality of the processed recordings is tested within the program using the STOI (Short-Time Objective Intelligibility Measure) method as well as the subjective analysis method within listening tests.

Abstract:
The focus of this thesis is on transferring data between mobile devices. It analyzes the resource consumption of using the Wireless LAN interface in some situations, and proposes and evaluates some new applications involving the transfer of voice and data files. Measurements show the high consumption of battery power due to the operation of a wireless network interface, even if it is not “actively” used to transfer data, while the memory consumption of the running applications is very limited. This thesis includes also an application to transfer files between two or more Pocket PCs: it is proposed as an addition to the VoIP application Minisip. Finally I have developed an application in order to explore the possibility to make a voice call by transferring real-time encoded text using UDP and/or RTP streams. This could be used together with speech-to-text and text-to-speech conversion applications at the end points to allow a “voice conversation” even on wireless links with very limited capacity, whereas a standard VoIP conversation would not be affordable.
Fokus i detta examensarbete ligger på överföring av data mellan mobila enheter. Det analyserar resursutnyttjandet vid användande av Wireless LAN-anslutning i några situationer, samt brukar analysen för att föreslå och utvärdera ett antal nya applikationer för överföring av röstsamtal och filer. Mätningar visar den höga konsumtion av batterikraft som anslutning till ett trådlöst nätverk gränsnitt, även då det inte används "aktivt" för dataöverföring, medan de applikationer som använts här behöver bara mycket begränsad minneskapacitet. I detta examensarbete ingår även ett program för överföring av filer mellan två eller flera mobila apparater, föreslaget att ingå i Minisip, en VoIP applikation. Dessutom föreslås och utvärderas en applikation som undersöker möjligheterna att vid röstsamtal överföra realtidskodad text via RTP och/eller UDP stream. Använd tillsammans med en röst-till-text- och text-till-röstomvandling i ändpunkterna möjliggör denna genomförande av röstsamtal även då mycket begränsade trådlösa anslutningar nyttjas.

Abstract:
This report presents an investigation of the possibilities to implement voice over IP (VoIP) in Sony Ericsson cellular phones. The results from this investigation show that it is partially possible to implement such a solution. The best option for doing so is to make use of the support for the Session Initiation Protocol and the Real-time Transport Protocol offered by the architecture. Another goal is to evaluate if Bluetooth is able to handle the requirements needed for the solution. The whole concept is proven by implementing a prototype. Measurements on this prototype show that Bluetooth will be able to handle the requirements of most IP-based voice communication, i.e., in respect to latency and bandwidth.

Abstract:
Wireless local area networks WLAN and Voice over IP technologies enable local low cost wireless telephony, while cellular networks offer wide-area coverage. The use of dual mode WLAN-cellular terminals should allow cost savings by automatically switching from GSM to WLAN networks whenever it is feasible. However, in order to allow user mobility during a call, a handover procedure for transferring a call between the WLAN interface and the cellular network should be defined. The decision algorithm that triggers such a handover is critical to maintain voice quality and uninterrupted communication. Information or measurements collected from the network may be used to anticipate when the connection will degrade to such a point that a handover is desirable in order to allow a sufficient time span for the handover’s successful execution. It is the delay in detecting when to make a handover and the time to execute it that motivates the need for a prediction. The goal of this thesis is therefore to present a method to predict when a handover should be made based upon network conditions. We selected a number of WLAN and VoIP software tools and adapted them to perform the measurements. These tools allowed us to measure parameters of the WLAN’s physical and link layers. Packet losses and jitter measurements were used as well. We have assumed that there is ubiquitous cellular coverage so that we only have to be concerned with upward handovers (i.e, from the WLAN to the cellular network and not the reverse). Finally we have designed and evaluated a mechanism that triggers the handover based in these measurements.

Förseningen i detekteringen när en handover ska ske och tiden för utförandet motiverar behovet av förutsägelse. Det här exjobbet introducerar en metod som förutsäger när handover ska börja baserade på nätverksförhållandena. Vi har valt några WLAN och VoIP-program och anpassat dem för att genomföra mätningarna. Programmen tillät oss att mäta WLANs parameter för fysiska och datalänksskikten. Pecket Loss och jitter-mätningar användes likaså. Vi antog att det fanns GSM tjänst på alla platser så att vi endast behövde göra uppg aende handover(t.ex. från WLAN till mobilt nätverk och inte tvärtom). Vi framkallade och testade en mekanism att starta handovern baserade på nätverksmätningarna.

Abstract:
A E3.5 jours de développement (E3.5), l’embryon murin se compose d’une monocouche de cellules externes correspondant au Trophectoderme (TE) et d’une masse cellulaire interne (MCI), hétérogène, constituée de deux sous-populations de cellules précurseurs : les cellules épiblastiques (Epi) et les cellules d’endoderme primitif (EPr). NANOG, marqueur épiblastique et GATA6, marqueur de l’EPr, sont co-exprimés à E3.5 dans la MCI puis adoptent une expression exclusive au sein de leur lignage respectif. La différenciation du lignage EPr nécessite l’expression de GATA6 et l’activation de la voie Récepteur Tyrosine Kinase (RTK) activée par le FGF (RTK-FGF) pour l’induction de gènes cibles de GATA6 tels que Sox17 et Gata4.Au cours de ma thèse, j’ai, dans un premier temps, étudié la relation GATA6/voie RTK-FGF lors de l’induction de l’expression des gènes de différenciation de l’EPr. J’ai utilisé des cellules souches embryonnaires murines ES sauvages ou mutantes pour Gata6 (ES Gata6-/-), dans lesquelles j’ai surexprimé différentes formes mutantes de Gata6 inactivées sur les différents résidus identifiés comme potentiellement phosphorylables par la voie RTK-FGF. Ainsi, j’ai analysé l’expression protéique des gènes Sox17 et Gata4 ainsi que des expressions ARN de ces cibles et d’autres gènes caractéristiques exprimés dans l’EPr dans les différentes conditions de surexpression des formes de Gata6 en absence ou présence d’inhibiteurs de la voie RTK-FGF. Ainsi, j’ai pu mettre en évidence que la transmission du signal s’effectue au travers de récepteur au FGF et qu’il existe une compensation entre les branches RTK-MEK-ERK et RTK-PI3K ciblant le résidu Sérine 37 de GATA6. Enfin, les résidus S34 et T509 sont nécessaires et les résidus S34, S37 et T509 semblent coopérer, au travers d’un mécanisme pour le moment non détaillé, pour l’induction des gènes cibles exprimés au sein de l’EPr.Dans un second temps, j’ai débuté la caractérisation phénotypique du rôle des facteurs Dickkopf1 (DKK1), un inhibiteur de la voie WNT/β-caténine, et NOGGIN, un inhibiteur de la voie des Bone Morphogenic Protein (BMP) lors de la différentiation de l’EPr en endoderme pariétal (EP) et viscéral (EV). A l’aide de modèles de souris KO pour DKK1 et NOGGIN, croisées en fond C57Bl6 pur, j’ai pu observer que l’expression d’OCT4 était maintenue au sein des embryons homozygotes mutants pour Dkk1 et double homozygotes mutants pour Dkk1 et Noggin. Cependant, le mécanisme potentiel de compensation ou de coopération de ces deux marqueurs n’est pour le moment pas détaillé précisément et mérite l’analyse d’un plus grand nombre d’embryons mutants
At E3.5 days of development (E3.5), mouse embryo consists of a monolayer of external cells corresponding to Trophectoderme (TE) and of an intern cell mass (ICM), heterogeneous, constituted by two subpopulations of precursory cells: epiblastic cells (Epi) and primitive endoderm cells (EPr). NANOG, an Epi marker and GATA6, a PrE marker, are co-expressed at E3,5 in the MCI and then adopt an exclusive expression within their respective lineage. EPr differentiation requires both expression of GATA6 and RTK pathway, activated by FGF ligand, in order to induce late markers Sox17 and Gata4 expression.First, I studied the relation GATA6/RTK during this process to understand the mechanism of induction of these target genes during final EPr differentiation. I used embryonic stem cells ES WT or Gata6 mutants (ES Gata6-/-), in which I transfected various Gata6 mutant constructions on different residues characterized as potentially phosphorylable by the RTK pathway. So, I analyzed protein expression of Sox17 and Gata4 target genes as well as RNA expression of characteristic genes expressed in the EPr in different inhibition conditions of RTK pathway. So, I was able to highlight that the transmission of the signal is made through the FGF receptor (FGFR1) and that there is compensation between RTK-MEK-ERK and RTK-PI3K pathways highlighted by later Gata6 overexpression of certain mutant forms. Finally, residue S34, S37 and T509 seems to cooperate, through a mechanism not detailed for the moment, for the induction of the EPr target genes.Then, I was interested to phenotypically characterize the role of Dickkopf1 (DKK1), an inhibitor of the WNT/β-catenin pathway, and NOGGIN, an inhibitor of the Bone Morphogenic Protein (BMP) pathway during the EPr differentiation in parietal endoderm (EP) and visceral (EV). Using models of mouse KO for Dkk1 and Noggin, met in pure background C57Bl6, I was able to observe that OCT4 expression was maintained within the Dkk1-/-, and Dkk1-/- Noggin-/- embryos. However, the potential compensation or cooperation mechanism of these two markers is not understanding well for the moment and deserves the analysis of a largest mutant embryos number

Abstract:
Les principaux virus entériques à l’origine de toxi-infections alimentaires collectives sont les norovirus génogroupes I et II (NoV GI, NoV GII) et le virus de l’hépatite A (VHA) responsables respectivement de gastro-entérites et d’hépatites. Ces virus sont transmissibles par la voie féco-orale directe ou via l’ingestion d’eaux ou d’aliments consommés crus ou peu cuits (coquillages, fruits et légumes). Le niveau de contamination virale des aliments souvent faible nécessite d’utiliser des méthodes de détection très sensibles. La plupart des virus entériques étant non cultivable, ces méthodes reposent sur la détection / quantification des génomes viraux par RT-qPCR ce qui ne permet pas de déterminer l’infectiosité des virus et limite l’appréciation du risque viral en santé publique. Les travaux de thèse visaient à proposer des méthodes moléculaires pour la détection, la quantification et le typage des virus entériques, à évaluer l’apport de nouvelles technologies moléculaires (comme la Digital RT-PCR (RT-dPCR) et la RT-PCR à haut débit) dans le cadre du diagnostic viral dans les aliments et enfin à développer des traitements précédant les réactions de RT-qPCR pour détecter des génomes issus de particules virales infectieuses. Une nouvelle technique d’extraction du VHA à partir de la laitue a été développée et évaluée équivalente à la technique de référence décrite dans les spécifications techniques publiées en 2013 (ISO/TS 15216-1 et 15216-2). Pour favoriser les études phylogénétiques dans le domaine alimentaire, 6 modèles moléculaires de RT-qPCR spécifiques des 6 sous-types humains du VHA (IA, IB, IIA, IIB, IIIA, IIIB) ont été développés et évalués pour le génotypage d’échantillons cliniques contaminés par le VHA. Ils peuvent être utiles pour tracer les sous-types du VHA dans des échantillons faiblement contaminés comme des matrices alimentaires, mais aussi permettre l'identification de co-infection de l'homme ou de souches de VHA recombinantes. La RT-dPCR en nanofluidique a été comparée à la RT-qPCR pour la quantification des génomes de NoV GI, NoV GII et VHA en présence de 2 contrôles de process (mengovirus et norovirus murin) dans des échantillons de laitues et d’eau embouteillée artificiellement contaminés. Un contrôle externe d’amplification a permis d’évaluer et de comparer l’inhibition des réactions de RT-qPCR et RT-dPCR. Les rendements d’extraction viraux se sont révélés significativement plus élevés après RT-dPCR qu’après RT-qPCR pour les NoV GI et mengovirus dans l'eau et pour les NoV GII et VHA dans les échantillons de laitue. De plus, les essais de RT-dPCR se sont avérés être plus tolérants à la présence de substances inhibitrices issues de laitues. La technologie qPCR en nanofluidique a également été utilisée afin de proposer une « puce » capable de détecter 20 virus entériques. Des limites de détection similaires ont été obtenues avec la qPCR et la dPCR. La qPCR nanofluidique a été trouvé moins sensible d’environ 1 à 3 log10 (du fait des faibles volumes (~nanolitre) d’échantillons analysés). Des prétraitements à base de monoazide +/- détergent à réaliser avant la RT-qPCR pour la détection de virus infectieux (VHA, rotavirus) ont été développés et évalués en réalisant des cinétiques d’inactivations thermiques. The main enteric viruses that cause foodborne outbreaks are noroviruses genogroupe I and II (NoV GI and NoV GII) and hepatitis A virus (HAV), respectively responsible for gastroenteritis and hepatitis. They are mainly transmitted via the faecal-oral route either by person-to-person contact or by ingestion of contaminated water, raw and undercooked food, particularly shellfish, soft fruits and vegetables. Viral contamination level is often low and requires sensitive methods of detection. As most enteric viruses are not cultivable, these methods are based on viral genome detection and quantification by real time RT-PCR. Such an approach provides no information regarding virus infectivity and therefore limits viral risk assessment in public health. These thesis works aim to propose molecular methods for enteric viruses detection, quantification and typing, also to evaluate new molecular technologies contribution (as Digital PCR and PCR high throughput) for food viral diagnosis and finally to develop treatments combined with RT-qPCR to only detect genomes from infectious viral particles. A new HAV extraction from lettuce method was developed and assessed as similar to the reference method which is described in the technical specifications published in 2013 (ISO/TS 15216-1; ISO/TS 15216-2). In order to facilitate phylogenetic analysis in food microbiology, six subtype-specific RT-qPCR assays for human HAV (HAV IA, IB, IIA, IIB, IIIA, IIIB) were developed and evaluated by testing HAV contaminated clinical samples genotyping. These assays may be particularly useful for accurately tracing HAV in low-level contaminated samples such as food matrices and moreover, to allow co-infection identification in human samples and/or HAV recombinant strains. Nanofluidic digital RT-PCR (RT-dPCR) was compared to RT-qPCR for NoV GI, NoV GII, and HAV genomes quantification, in presence of two process controls (mengovirus and murine norovirus) in artificially contaminated bottled water and lettuce samples. External amplification control allowed evaluating and comparing RT-qPCR and RT-dPCR assays inhibitions. Viral recoveries calculated by RT-dPCR were found to be significantly higher than by RT-qPCR for NoV GI and Mengovirus in water, and for NoV GII and HAV in lettuce samples. The RT-dPCR assay proved to be more tolerant to inhibitory substances present in lettuce samples. Nanofluidic PCR Array (PCR Array) has also been used in order to propose an array able to simultaneously detect 20 enteric viruses. Similar detection limits were obtained with qPCR and dPCR but PCR Array was found less sensitive of 1 to 3 log10 (due to the weak volumes (nanolitre) of analyzed samples). Pretreatments based on the use of monoazides +/- surfactant and to do before RT-qPCR were developed for discriminating between infectious and non-infectious particles of HAV and rotavirus. The aim of this study is to explore Saudi women’s experience of being healthy during the postpartum period (also called Nifas in Arabic). Little attention has been given to women’s experience of health during the postpartum period; however, it is essential to understand their experiences and understandings of health during this time if we are to develop healthcare programmes that meet their needs and improve outcomes during postpartum period healthcare. Little attention is given to postpartum women’s health in Saudi Arabia, and a postpartum woman with a normal vaginal childbirth is usually discharged from a maternity hospital 24-48 hours after the childbirth. Then, for the next 40 days, she is secluded within the home, cared for by others (usually her mother) and will only be seen by healthcare services (the obstetrician) a month after childbirth to receive their postpartum check-up. This sole consultation is limited to examining her episiotomy, a routine procedure for the majority of Saudi women. The aim of this study is to find out about women’s health practices during this hidden but important time for women’s health- the 40 days following child birth that are spent in the home. This is an interpretive phenomenological study that examines online forum discussions of Saudi Arabian women sharing experiences and seeking advice on their health during the postpartum period (28 threads), as well as a total of 12 in-depth, semi-structured interviews with women (n=7) and their carers (n=5) within their 40 days postpartum. Thematic analysis was used. The women described their experience of being healthy during the postpartum period. They believed that, during this time, women become vulnerable and open to certain illness, but they also perceived this period as an opportunity to achieve their optimal health and attractiveness. Some of the common themes that emerged were the challenges of confinement, achieving health, everyday threats and constraints, warnings and consequences, striving for normality, and becoming better than normal. However, key themes identified during interviews with postpartum women also included trust in various sources of knowledge, which included their carers, female relatives, friends, and online postpartum health discussions. The study also uncovered several strategies women used to follow health advice that were often hidden from their healthcare practitioners. In contrast, the findings from the online community discussions demonstrate that this was a forum where women could discuss issues which they did not feel able to discuss during interviews and in front of carers. These issues included the difficulties of judging expertise and advice, emotional and psychological health, husbands, and sexual activity. The study’s findings can be used to explain the postpartum women’s perception of health to healthcare providers, and the study can assist in understanding some of these women’s strategies to be healthy, including commonly followed traditional health practices. It highlights their difficulties in making sense of the large amount of traditional advice they are faced with, which covers every aspect of daily life during this period, from hygiene and food to preventing and healing episiotomies, but which also ignores key issues for these women (mental and sexual health). Finally, this study highlights Saudi women’s need for support and improved communication between these women and clinical services during the postpartum period; the frankness of the online discussions indicates that a reliable and confidential online health education forum might offer an effective way of providing this information and support.

L'oxyde de cobalt Sr2Co2O5+-x cristallise sous deux variétés cristallines différentes selon la configuration électronique du cobalt trivalent: la brownmillerite dérivée de la perovskite cubique (forme haute temperature) et une forme basse température lacunaire en oxygène dérivée de la perovskite hexagonale 2H dont les structures ont été précisées. Seule la forme brownmillerite a pu être oxydée par voie électrochimique en milieu alcalin, l'oxyde SrCo4+O3 de structure perovskite cubique est alors obtenu. Les études électrochimiques, cristallographiques et les propriétes physiques de ces matériaux ont permis d'expliquer ces phénomènes et l'absence d'oxydation de la forme basse température.

With its enormous potential Voice over Internet Protocol is one of the latest buzzwords in information technology. Despite the numerous advantages of Voice over IP, it is a major technical challenge to achieve a similar call quality as experienced in the ordinary Public Switched Telephone Network. This thesis introduces standardized Internet protocols for Voice over IP, such as Session Initiation Protocol (SIP), Real-time Transport Protocol (RTP), in its background chapter. In order to provide better Quality of Service (QoS) Voice over IP applications should support a feedback mechanism, such as the Real-time Control Protocol (RTCP), and use accurate timing information, provided by the Network Time Protocol (NTP). Additionally this thesis considers synchronization issues in calls with two and more peers. After a rather academic overview of Voice over IP, the open source real-time application “minisip”, a SIP user agent, and its operation and structure for handling audio streams will be introduced. Minisip was extended by an implementation of NTP and RTCP to provide a test platform for this thesis. A clear conclusion is that the addition of global time helps facilitate synchronization of multiple streams from clients located any where in the network and in addition the ability to make one-way delay measurements helps SIP user agents to provide better quality audio to their users.
Röst över IP, eller Internettelefoni baserad på “Voice over Internet Protocol” (VoIP), har med sin stora potential blivit ett av de senaste modeorden inom informationsteknologin. Vid sedan av ett antal fördelar med VoIP så innebär det en stor teknisk utmaning att uppnå en likadan samtalskvalitet som i det vanliga, fasta, telenätet. I den här uppsatsen beskrivs hur tjänstevalitet för VoIP kan förbättras genom att noggrant tidssynkronisera de (två eller flera) klienter som deltar i ett telefonsamtal. För detta krävs dels en återkopplingsmekanism, såsom “Real-time Control Protocol” (RTCP), samt en gemensam tidsuppfattning i de inblandade klienterna, vilket kan uppnås med hjälp av “Network Time Protocol” (NTP). Dessa protokoll, liksom de övriga Internet-standarder som VoIP baseras på (såsom “Session Initiation Protocol” (SIP) och “Real-time Transport Protocol” (RTP), beskrivs inledningsvis i uppsatsen. För studien har en SIP-klient baserad på öppen källkod använts (“Minisip”), och utökats med NTP och RCTP funktionalitet för att testa den föreslagna förbättringen av VoIP. En tydlig slutsats är att kännedom om en “global tid” möjliggör synkronisering av multipla ljudströmmar från klienter som befinner sig på olika nätverk. Möjligheten att mäta paketfördröjningen (envägs) bidrar också till en förbättrad ljudkvalitet.

Les cils et flagelles sont des organites cellulaires retrouvés des protozoaires aux mammifères. Une dérégulation de l’assemblage (ciliogenèse) ou de la fonction des cils, entraîne diverses maladies chez l’homme. Parmi les acteurs de la ciliogenèse, se trouvent les facteurs de transcription RFX. La recherche de gènes cibles de RFX chez la drosophile a permis d’identifier le gène Chibby (Cby), précédemment décrit comme un antagoniste de la voie de signalisation Wnt/wingless. Contrairement aux vertébrés, chez les invertébrés aucun lien n’est encore établi entre cil et voie wg. L’identification de cby comme cible de dRFX chez la drosophile suggère une fonction ciliaire de cby et permettrait l’établissement du lien cil/voie wg. CBY se localise à la zone de transition des cils des neurones sensoriels du système nerveux périphérique et aux centrioles des spermatides. Les drosophiles invalidées pour cby présentent un phénotype de non coordination, mais aucun phénotype de type wg. Ces mutants ont des défauts des cils sensoriels, ainsi que des défauts d’organisation des spermatides. De plus, chez les embryons, les protéines actrices du transport intra-flagellaire, NompB et CG11356, sont mal distribuées lorsque CBY est absente. Enfin, chez les mutants, la localisation de la protéine UNC est affectée dans les cellules germinales en fin de spermatogenèse. En conclusion, chez la drosophile, CBY est impliquée dans le tri protéique organisé à la base du cil de neurones sensoriels, de concert avec les autres protéines localisées à la zone de transition. Cilia and flagella are organelles found from protozoa to mammals. Deregulation of the assembly (ciliogenesis) or function of cilia, causes various diseases in humans. Among those involved in ciliogenesis are the RFX transcription factors. The search for RFX target genes in Drosophila identified the Chibby (CBY) gene, previously described as an antagonist of the Wnt / wingless pathway. Unlike in vertebrates, in invertebrates is still no link established between cilia and the wg pathway. The identification of CBY as a target of dRFX in Drosophila suggests a ciliary function of CBY and would allow the establishment of the link cilia / wg pathway. CBY is localized at the transition zone of cilia of sensory neurons of the peripheral nervous system and at centrioles in spermatids. Drosophila invalidated for CBY present a phenotype of uncoordination, but no wg phenotype. These mutants have defects in sensory cilia and defects in organization of spermatids. In addition, in embryos, the distribution of proteins involved in intra-flagellar transport, NompB and CG11356, is affected when CBY is absent. Finally, in the mutants, the localization of the UNC protein is affected in germ cells at the end of spermatogenesis. In conclusion, in Drosophila, CBY is involved in the protein sorting organized at the base of cilia of sensory neurons, with the other proteins located at the transition zone. In the testes, CBY is necessary for the maturation of spermatids. However, CBY is not involved in the regulation of the wg pathway

Le sulfure d'hydrogène (H2S) est un métabolite bactérien produit notamment par les bactéries sulfato-réductrices du côlon à partir des acides aminés soufrés, des sulfates/sulfites alimentaires et des sulfomucines. A fortes concentrations, le H2S est un gaz toxique, de par sa capacité à inhiber la cytochome c oxydase, et par conséquent, la respiration mitochondriale. A l'inverse, notre travail montre qu'à faibles concentrations, le H2S induit une énergisation mitochondriale des cellules épithéliales coliques humaines HT 29 Glc-/+ lui conférant aussi un rôle de substrat minéral énergétique. Dans ce travail, nous avons déterminé les concentrations de H2S permettant l'oxydation/détoxication de H2S par les cellules HT 29 Glc-/+ et celles provoquant une inhibition de la consommation d'oxygène. L'oxydation du H2S nécessite la coopération entre la " sulfide oxidation unit " et la chaîne respiratoire. La capacité des cellules HT29 Glc-/+ à oxyder le H2S est associée à la présence des transcrits codant les enzymes constituant la " sulfide oxidation unit " : la sulfure d'hydrogène quinone reductase (SQR), la dioxygenase ETHE1 et la thiosulfate transferase (TST). Nous avons démontré la priorité de l'oxydation du H2S sur les substrats carbonés . En effet, nos résultats suggèrent que les électrons venant de la SQR sont transférés au pool d'ubiquinone aux dépens de ceux venant du complexe I. Nos résultats démontrent que la SQR joue un rôle déterminant pour l'oxydation de H2S. De plus, la détoxication du H2S par les cellules HT29 Glc-/+ augmente au cours de la différenciation spontanée ou induite par un traitement au butyrate. L'augmentation de la détoxication de H2S au cours de la différenciation est associée à une augmentation de la réserve respiratoire soulignant l'importance de la chaîne respiratoire comme composante de la fonction de détoxication du H2S. En situation d'inhibition de la cytochrome c oxydase, la grande capacité des cellules coliques humaines à détoxiquer le H2S pourrait être en partie due à la présence d'un transfert réverse des électrons issus de l'oxydation de H2S de la SQR vers le complexe I. Outre le butyrate, le zinc un autre composé de la lumière colique, exerce un effet protecteur contre la toxicité cellulaire du H2S. Enfin, notre travail a mis en évidence une diminution de l'expression d'un gène codant pour une enzyme de la " sulfide oxidation unit " (la TST) dans le rectum comparée à différents segments du côlon, ce qui pourrait correspondre à des capacités de détoxication du H2S différente en fonctions des segments du gros intestin humain.

et une importante conductivité électrique. Ce travail est consacré à l'étude des performances
de films minces multicouches d'ITO obtenus par la voie sol-gel.
La microscopie électronique en transmission (MET) a permis d'analyser morphologie
et structure, et a mis en évidence pour la première fois la structure complexe de tels films solgel.
La MET et l'analyse par rétrodiffusion de particules α ont précisé la corrélation entre la
densité des multicouches, l'apparition de croûtes intermédiaires et la conductivité électrique
résultante.
Ce travail valide l'utilisation du recuit rapide pour la densification des films sol-gel en
comparaison avec un traitement classique.
Nous démontrons la potentialité de la voie sol-gel pour la croissance d'électrodes
d'ITO sur silicium poreux (SP). Les structures ITO/SP, pour lesquelles la photoluminescence
du SP est partiellement préservée, ont été étudiées en terme de diode Schottky.

L'objectif de notre thèse est d'évaluer le rôle de l'alcool et autres substances psychoactives dans la survenue et la gravité des traumatismes dus aux Accidents de la Voie Publique (AVP) en Côte d'Ivoire. Nous avons initié un recueil de données sur 893 victimes tuées ou blessées suite à un AVP, et admises aux urgences ou à l'Institut de Médecine Légale d'Abidjan. Ces données hospitalières ont été couplées aux données policières et aux résultats d'analyses toxicologiques. La méthode de dosage de l'alcoolémie par CPG-FID a démontré qu'elle répond parfaitement aux critères de performances analytiques recommandés en termes de fidélité, d'exactitude, de linéarité, de stabilité et de limite de quantification. Les niveaux d'alcoolémie chez les victimes au moment de l'AVP, sont élevés et compatibles avec un profil d'alcoolisation excessive. Plus de 75% des conducteurs alcoolisés ont une alcoolémie supérieure à 1g/L. Les prévalences d'alcoolémie au-delà de 0,8g/L sont de 41% chez les conducteurs de véhicules légers 4 roues, 35% chez les 2-roues, 17% chez les piétons et 13% chez les passagers. L'usage des autres substances psychoactives est moins fréquent chez les victimes (11%) et souvent associé à une alcoolémie supérieure à 0,8g/L. L'évaluation du risque de blessures graves (NISS9+) en fonction du degré d'alcoolisation suggère que les victimes avec une alcoolémie modérée ont un risque de blessures graves plus élevé en comparaison avec les victimes sobres (OR = 7,7; IC95% : 2,5-24,2). Au terme de cette thèse, nous proposons une procédure fiable de dosage de l'alcoolémie chez les victimes d'AVP et des mesures concrètes pour la prévention des traumatismes par AVP en Côte d'Ivoire
We implemented a data collecting process including 893 victims who died or were injured following a RTA, and were admitted to the Emergency Room or to the Forensic Institute in Abidjan. This hospital-based data was linked with police data and toxicological testing. The method developed in our laboratory for blood alcohol determination using GC-FID demonstrated a good compliance with analytical performance requirements in terms of repeatability, accuracy, linearity, stability and limit of quantification. Blood alcohol levels found in victims at the time of the RTA were compatible with an excessive alcohol intake. More than 75% of drivers with a positive alcohol test had a blood alcohol concentration greater than 1g/L. Prevalence of alcohol above 0.8g/L reaches 41% in light four-wheel drivers, 35% in two-wheel riders, 17% in pedestrian and 13% in passengers. The prevalence of psychoactive drugs was lower (11%) compared to alcohol and usually associated with blood alcohol level greater than 0.8g/L. The assessment of the risk of serious injury (NISS9+) associated to the level of blood alcohol content suggest that victims showing a moderate blood alcohol level are more at risk to sustain serious injury compared to sober victims (OR = 7.7; IC95% : 2.5 – 24.2). In conclusion, we make some practical proposals to strengthen and for facilitating the application of the current law concerning driving under the influence of alcohol. We also make recommendations for prevention of road traffic injuries in the Ivory Coast

The rapid growth of the Internet and the proliferation of its new applications pose a serious challenge in network performance management and monitoring. The current Internet has no mechanism for providing feedback on network congestion to the end-systems at the IP layer. For applications and their end hosts, end-to-end measurements may be the only way of measuring network performance. Understanding the packet delay and loss behavior of the Internet is important for proper design of network algorithms such as routing and flow control algorithms, for the dimensioning of buffers and link capacity, and for choosing parameters in simulation and analytic studies. In this thesis, round trip time (RTT), one-way network delay and packet loss in the Internet are measured at different times of the day, using a Voice over IP (VoIP) device. The effect of clock skew on one-way network delay measurements is eliminated by a Linear Programming algorithm, implemented in MATLAB. Distributions of one-way network delay and RTT in the Internet are determined. It is observed that delay distribution has a gamma-like shape with heavy tail. It is tried to model delay distribution with gamma, lognormal and Weibull distributions. It is observed that most of the packet losses in the Internet are single packet losses. The effect of firewall on delay measurements is also observed.

Používateľom tak bude poskytnutý moderný spôsob, ako prirodzene komunikovať cez internetový prehliadač. Hlavný dôraz je kladený na synchronizáciu medzi hlasovým asistentom a obsahom na webovej stránke. Synchronizácia je dosiahnutá obojsmerným prenosom hlasu a textových príkazov medzi klientom a serverom. Na to je použitá technológia WebRTC v kombinácií so signalizačným protokolom SIP. Práca sa zaoberá oblasťami ako VoIP telefonovanie, počítačové siete a strojové učenie(proprietárne rečové technológie od Phonexie). Benefitom nasadenia hlasového asistenta je zníženie nákladov na odchádzajúce hovory pre klientov, odľahčenie agentov na call centrách pri odpovedaní na často kladené otázky a zvýšenie záujmu zákazníkov vďaka použitiu nových technológií.

The master’s thesis focuses on the virtual local computer network for laboratory usage. It aims to propose and realize proper network connection in order to monitor expected data flow. Thanks to the network analysers (software ClearSight and hardware NetTool Series II) it plans to pursue in detail the used transmission protocols of TCP/IP layers. The most decisive feature happens to be the right choice of appropriate network components and their precise configuration. Consequently, the thesis formulates a proposal of a laboratory task for the needs of students, which is also closely related to the actual problems. The assignment of the task will serve the teachers as a test pattern for measurement. The results elaborated in the form of the model protocol should enable later comparison of the recorded data. Another part of the diploma thesis is the working-out of well arranged manuals for the network analysers involved.

In IEEE 802.11 WLANs, Link Adaptation mechanisms, which choose the transmission rate of each node, provoke unexpected and random variations on the effective channel capacity. When these changes are towards lower bitrates, inelastic flows, such as VoIP, can suffer from sudden congestion, which results on higher packet delays and losses. In this thesis, a VoIP codec adaptation algorithm is proposed as a solution, based on a cross-layer feedback from RTCP packets and the MAC layer, which can adapt the codecs of active calls to adjust them to the multirate scenario. A combination of this algorithm with a call admission control mechanism is also studied. The results show an important improvement in terms of the QoS of the already active flows as also in the total hotspot's capacity. Additionally, by defining a new Grade of Service related parameter, the Q-Factor, which captures the trade-off between dropping and blocking ratio and perceived speech quality, the codec adaptation algorithm can be tuned to achieve maximum capacity without severely penalizing any of those variables, and hence satisfying both technical and user quality requirements. Finally, a new QoS-enabled AP, which implements these enhancements is designed.
En las redes inalámbricas del estándar IEEE 802.11, los mecanismos de adaptación de enlace que eligen la tasa de transmisión de cada nodo, pueden provocar variaciones aleatorias e inesperadas en la capacidad efectiva del canal. Cuando estos cambios son hacia tasas de transmisión mas bajas, los flujos inelásticos, tales como los de VoIP, pueden de repente sufrir congestión, lo que se traduce en aumento de retrasos y pérdidas de paquetes. En esa tesis, se propone un algoritmo de adaptación de codificadores de voz como solución, basado en técnicas multinivel (cross-layer) que combinan el uso de información de diferentes capas, como los paquetes RTCP y la capa MAC, y que puede adaptar los codecs de las llamadas activas para ajustarlos al escenario "multi-rate". Adicionalmente, la combinación de este algoritmo con un mecanismo de control de admisión de llamadas (CAC) se ha estudiado. Los resultados muestran una importante mejora en términos de QoS de los flujos activos como también en la capacidad total del hotspot. Además, mediante la definición de un nuevo factor, el Q-Factor, que puede captar la compensación entre la tasa de corte y de bloqueo de llamadas y de la calidad percibida por esas, el algoritmo de adaptación de codecs se puede ajustar para lograr la máxima capacidad sin penalizar severamente ninguna de esas variables y así satisfacer los requisitos técnicos de calidad y los usuarios. Por último, un nuevo punto de acceso (AP) habilitado para ofrecer calidad de servicio, ha sido diseñado que lleva a cabo estas mejoras.

Le cancer de vessie représente la sixième cause de mortalité par cancer en France. Son incidence a augmenté ces 20 dernières années, mais les taux de survie restent inchangés. La carcinogénèse vésicale fait intervenir différents mécanismes moléculaires qui agissent en réseau comme c’est le cas dans de nombreux cancers. Le développement récent de nouveaux traitements prenant spécifiquement pour cible certaines voies de signalisation apportent de nouveaux espoirs thérapeutiques. Nous nous sommes intéressés dans ce travail à trois axes de recherche pour tenter d’identifier, dans les carcinomes urothéliaux, de nouveaux marqueurs pronostiques moléculaires et de nouvelles cibles thérapeutiques potentielles: l’angiogénèse, la voie de signalisation Hedgehog et les microARNs. Nous avons choisi la RT-PCR quantitative en temps réel à grande échelle permettant d’évaluer le niveau d’expression de nombreux gènes, avec une quantification précise et reproductible des transcrits. L’expression de ces gènes a été corrélée aux données de suivi clinique afin d’identifier de nouveaux biomarqueurs moléculaires prédictifs de l’évolution des tumeurs de vessie.Nous avons ainsi pu démontrer que les niveaux d’expression de certains de ces gènes variaient de façon significative dans les tumeurs de vessie, confirmant le rôle de l’angiogénèse dans la carcinogénèse urothéliale, et plus particulièrement de la voie du VEGF, et suggérant une implication majeure de la voie de signalisation Hedgehog et des microARNs. Par ailleurs, nous avons également pu identifier plusieurs biomarqueurs ayant une valeur pronostique en terme de survie globale dans les tumeurs infiltrantes. C’est le cas du VEGF, qui semble être un biomarqueur moléculaire particulièrement intéressant puisqu’il existe des thérapies ciblées spécifiquement dirigées contre ce ligand ou ses récepteurs avec plusieurs essais cliniques actuellement en cours dans le cancer de vessie. C’est également le cas d’une signature moléculaire associant 3 miARNs (miR-9, miR-182 et miR-200b) ayant une valeur péjorative dans les tumeurs infiltrantes, ouvrant la voie vers de nouvelles stratégies thérapeutiques.L’ensemble de ces études confirment l’intérêt majeur d’une meilleure compréhension des bases moléculaires de la carcinogénèse urothéliale vésicale débouchant sur l’utilisation rationnelle de nouvelles thérapies ciblées dans le cancer de vessie, avec l’espoir d’en améliorer la prise en charge et l’évolution
Bladder cancer is the sixth cause of cancer mortality in France and its incidence is increasing since the last 20 years, with no improvement in survival outcomes. Bladder carcinogenesis involves different molecular mechanisms such as in many cancers. The recent development of new targeted therapies targeting signaling pathways provides new therapeutic hopes.In this work, we choose to study three molecular pathways in order to identify new prognostic markers and new therapeutic targets in urothelial carcinoma: angiogenesis, Hedgehog signaling pathway, and microRNAs. Real-time quantitative RT-PCR was performed to measure simultaneously expression levels of several genes with precise and reproductible RNA quantification. Our results were correlated with clinical outcomes to identify new molecular markers associated with bladder cancer evolution and to guide the potential use of targeted therapies.We were able to demonstrate that expression levels of several transcripts differ significantly in bladder tumors as compared to normal bladder and that some of them may have a prognostic implication. This is the case of VEGF, which appears to be an interesting molecular marker since there are targeted therapies specifically targeting the pathway and several ongoing trials in bladder cancer. The Hedgehog pathway also appears to be altered in bladder tumors, with a ligand-dependent activation. Then, we were able to identify several deregulated microRNAs and describe a molecular 3 miRNA-signature (miR-9, miR-182 and miR-200b) having a prognostic value in muscle-invasive bladder tumors. All these studies confirm the major interest of molecular biology and new targeted therapies in the treatment of bladder cancer, with the hope of improving management and evolution

Este trabalho apresenta um sistema de conferência multiponto centralizada com controle de posse da palavra, que oferece comunicação por voz e por texto, desenvolvido para dar suporte a ferramentas de educação a distância baseadas em tecnologia web. Além do sistema de conferência multiponto, este trabalho introduz, também, conceitos de voz sobre IP (VoIP), codecs de áudio, tráfego de tempo real sobre redes IP, mecanismos de distribuição de pacotes em redes IP, conferências multimídia sobre redes IP, criação e controle de conferências e mecanismos para se controlar a transmissão de pacotes de voz em conferências multiponto, como detecção de atividade de voz (VAD) e controle de posse da palavra. O controle de posse da palavra ou moderação é uma metodologia para coordenar o acesso a recursos de mídia contínua, como os canais de áudio e vídeo em conferências, resolvendo eventuais conflitos e condições de corrida e garantindo o uso exclusivo dos mesmos por um participante da conferência. O controle de posse da palavra é apropriado para a dinâmica de uma sala de aula, onde na maioria das vezes, o professor exerce o papel de um moderador e de principal interlocutor. Além de regular o fluxo de informação na sala de aula virtual, o controle de posse da palavra também ajuda a reduzir a utilização dos recursos da rede. Como principal resultado deste trabalho, uma arquitetura para o sistema de conferência multiponto foi proposta, implementada e testada. Verificou-se que o sistema é escalável e que o controle de posse da palavra reduz o grau de interatividade entre os participantes da conferência, mas funciona eficazmente como mecanismo para reduzir a utilização dos recursos de rede. A comunicação adicional por texto ajuda a reduzir o impacto do controle de posse da palavra no grau de interatividade. Neste trabalho, o uso de uma topologia centralizada para a realização de conferências multiponto mostrou-se bastante vantajoso, principalmente porque o tráfego multicast não era suportado.
This work presents a centralized multipoint conference system with floor control, which offers voice and text communication capability, developed to support distance learning tools based on web technology. This work also brings concepts about voice over IP (VoIP), audio codecs, real-time traffic over IP networks, packet distribution mechanisms in IP networks, multimedia conferences over IP networks, establishment and control of conferences and mechanisms to control voice packets transmission in multipoint conferences, as Voice Activity Detection (VAD) and floor control. Floor control is a suitable methodology for coordinating access to continuous media resources, such as audio and video channels in conferences, solving resource conflicts and race conditions and guaranteeing their mutually exclusive use by one participant of the conference. Floor control is appropriate for the classroom’s dynamics, where the teacher, mostly, has the role of a moderator and main talker. Besides regulating the information flow in the virtual classroom, floor control helps reducing the need for network resources. As main result, an architecture for the multipoint conference system was proposed, implemented and tested. It was found that the system is scalable and that floor control reduces the interactivity among the conference’s participants, but works efficiently as a mechanism for reducing the need for network resources. Text communications help lowering the impact of floor control on interactivity. In this work, the deployment of a centralized topology for multipoint conferences showed to be very advantageous, mainly because multicast traffic was not supported.

Since the Voice over Internet Protocol (VoIP) was proposed, the issue of voice quality is always a big challenge. When more and more people use VoIP applications, the quality issue becomes critical. Basically, the measurement of voice quality is to test on the client-side. However, in the real network, it is unreasonable to control clients during the communication time. In recent years, because of the exhaustion of IP addresses, Network Address Translator (NAT) was proposed to solve the shortage of IP addresses. Nevertheless, NAT fails some peer-to-peer Internet applications, such as VoIP. So, VoIP applications need solutions for NAT traversal. There are lots of NAT traversal mechanisms suggested, such as static assignment, Virtual Private Network (VPN), and relay-based proxy server. RTP Proxy Server is a relay-based proxy server, which is the most popular one among these NAT traversal mechanisms. Now in most VoIP architectures, there exists a RTP Proxy Server for relaying RTP packets. In this thesis, we design a monitoring system, named RTP-M which works with RTP Proxy Server, to measure the VoIP quality. Additionally, RTP-M represents the measured result as graphs which are more intuitive for human beings. We hope that our RTP-M can provide administrators of VoIP service with the troubleshooting reference.

As the technologies of communication network and multimedia progess, the deveplopments of real-time multimedia application over. Internet becomes mature. Thus, people can communicate over Internet more lively.   The goal in this thesis to develop a real-time audio transfer system in Windows environment using object-oriented approach. In our system, user can transfer voice data over Internet. We also propose a simple bandwidth reservation mechanism on Ethernet using WCA.   Our audio transfer system was implement on Windows 95 /NT using Microsoft fundamental Class 4.0. The main functions of system include multi-use audio communication, user profile exchange and RTCP packet monitor. A network application interface based on Windows Sockets API 2.0 was designed as well.

With the progress of the network, peoples can communicate with each other easily. The popularization of the Broadband network makes many things that could be hard to achieve becoming possible. However, the threat of SPIT is likely to increase as the more flexible SIP multimedia standard becomes more popular. Spam over IP Telephony (SPIT) is expected to become a serious problem in near future. It has the potential to become an even bigger problem than email spam, because the callee will be disturbed by each received SPIT call. This paper describes how to based on SDES to achieve SRTP master key exchange on voice authentication, integrate SRTP and SIP to SRTP_UE and then use session key derived from master key to protect real-time voice communication from eavesdropping. A new SPIT prevention method that is effective and acceptable for the call participants because it does not affect the callee at call and limits the interaction with caller to an acceptable minimum. Using SRTP with DTMF to simulate ASR (Automatic Speech Recognition), meanwhile propose a system model for VoIP ID (It’s likely Citizen Digital Certificates) on authentication servers. The general concept may be applied for different data in the meanwhile, for instance, one idea would be to ask all clients to register a unique and valid mobile phone number for each VoIP ID. As foundation for building general SPIT prevention systems with this and other innovative methods, this paper proposes reference architecture for SPIT prevention systems.

VoIP technology is one of the important applications of the network. In addition to using traditional PSTN telephone, people can choose favorable VoIP telephone because of the Internet popularization and High speed Internet. RTP (Real-time Transfer Protocol) is the foundation of VoIP, and it is suitable for applications to manage real-time transmission of multimedia data. But RTP does not provide any confidentiality, the eavesdrop of VoIP is easy to realize. In this paper, we discuss how to design and implement an embedded SIP user agent that provide an fast secure encoding of RTP voice streaming. We use limited resources to implement RTP encoding mechanism to prevent anyone from eavesdropping .
