Abstract:
Abstract We have generated and characterized a Drosophila cyclin E hypomorphic mutation, DmcycEJP, that is homozygous viable and fertile, but results in adults with rough eyes. The mutation arose from an internal deletion of an existing P[w+lacZ] element inserted 14 kb upstream of the transcription start site of the DmcycE zygotic mRNA. The presence of this deleted P element, but not the P[w+lacZ] element from which it was derived, leads to a decreased level of DmcycE expression during eye imaginal disc development. Eye imaginal discs from DmcycEJP larvae contain fewer S phase cells, both anterior and posterior to the morphogenetic furrow. This results in adults with small rough eyes, largely due to insufficient numbers of pigment cells. Altering the dosage of the Drosophila cdk2 homolog, cdc2c, retinoblastoma, or p21CIP1 homolog dacapo, which encode proteins known to physically interact with Cyclin E, modified the DmcycEJP rough eye phenotype as expected. Decreasing the dosage of the S phase transcription factor gene, dE2F, enhanced the DmcycEJP rough eye phenotype. Surprisingly, mutations in G2/M phase regulators cyclin A and string (cdc25), but not cyclin B1, B3, or cdc2, enhanced the DmcycE JP phenotype without affecting the number of cells entering S phase, but by decreasing the number of cells entering mitosis. Our analysis establishes the DmcycE JP allele as an excellent resource for searching for novel cyclin E genetic interactors. In addition, this analysis has identified cyclin A and string as DmcycEJP interactors, suggesting a novel role for cyclin E in the regulation of Cyclin A and String function during eye development.

Abstract:
The effect of surface roughness of virgin specimens (prior roughness) made of low-alloyed high strength steel on their fatigue life after the case-hardening treatment was studied by rotating bending tests of virgin and nitrided samples. In a whole range of S-N curves, the fatigue strength of virgin samples after lathe-turning machining (high roughness) was naturally much higher than that of samples after grinding (low roughness). As expected, the fatigue strength of nitrided specimens was higher than that of virgin samples when averaged through the whole fatigue life range. When distinguishing the rough and smooth nitrided samples, the low-cycle fatigue strength of rough samples revealed to be lower than that of the smooth ones and vice a versa in the high-cycle region. This could be explained by the fact that, in the low cycle region, the cracks in the rough samples nucleated from deep surface defects while, in the smooth specimens, they nucleated from shallow defects (or as fish eyes) which prolonged their initiation stage. Almost all cracks in the high cycle region initiated as fish-eyes but the extent of nitrided layers in the rough specimens was slightly higher than that in the smooth specimens. Therefore, the fish-eye centers were shifted further to the interior of the rough specimens which increased their fatigue life.

Models of motor guidance that dynamically adjust to the availability and quality of sensory information are based on the observation that dexterous tasks are routinely performed using various combinations of visual and tactile inputs. However, a dynamic neural system that acquires and processes relevant visual and tactile information remains relatively uncharacterized in humans. In this study, whole-brain functional magnetic resonance images were acquired during a dexterous manipulation task, compression of the end caps of a slender spring prone to buckling, to investigate the neural systems associated with motor guidance under four visual and tactile guidance conditions: (1) eyes closed (no visual input), smooth end caps, (2) eyes closed, rough end caps, (3) eyes open and watching hand, smooth end caps, and (4) eyes open and watching hand, rough end caps. Performance of the dexterous task remained constant in all conditions. Variations in the two levels of visual input resulted in modulation of activity in the middle and inferior occipital gyrii and inferior parietal lobule, and variation in the two levels of tactile input during the task resulted in modulation of activity in the precentral (primary motor) gyrus. Although significantly active in all conditions, cingulate gyrus, medial frontal gyrus, postcentral gyrus, and cerebellum activities were not modulated by levels of either visual or somatosensory input, and no interaction effects were observed. Together, these data indicate that a fine-tuned motor task guided by varying visual and tactile information engages a distributed and integrated neural complex consisting of control and executive functions and regions that process dynamic sensory information related to guidance functions.

Abstract:
Purpose. The objective of the present study was to obtain and investigate the tissue samples taken from the anterior chamber angle of the children of different gestational age under the normal conditions and in the presence of congenital glaucoma; the secondary objective was to carry out the histological study of the pieces of tissues removed from the glaucoma patients of different gestational age undergoing hypotensive filtration surgery (sinusotrabeculectomy). Materials and methods. The anterior chamber angle histological preparations from a total of five enucleated eyes were available for the intravital investigation of the trabecular meshwork (two of them were obtained from the full-term infants and one from a premature baby). Results. The histological study of the structures of the anterior chamber angle of the enucleated eyes has demonstrated the presence in all the cases of cornea, Descemet’s membrane, trabecular meshwork, Schlemm’s canal, and iris. The morphological structure of the anterior chamber angle associated with congenital glaucoma shows up some peculiar features compared with the normal one, such as its block by the permanent fibrous membrane which penetrates deep into the trabecular meshwork that, in its turn, is formed from the rough and thickened tissue. The histological picture of the fetal eye at different gestational age exhibits the open anterior chamber angle, the well developed porous trabecular meshwork, and fragmentation of Descemet’s membrane near the trabecular meshwork. No signs of fibrous degeneration of the trabecule have been apparent. Conclusion. The intravital investigation of the trabecular meshwork has demonstrated some peculiar features that differ depending on the gestational age in birth. In the full-term babies, it is hypoplastic and covered with the rough fibrous membrane at certain places. Its histological picture is similar to that of the preparations of the enucleated eyes. In the premature babies, the trabecular meshwork is fibrous and characterized by enhanced cellularity; it undergoes fibrous degeneration that becomes apparent after the birth of the premature child (it is supposed to be related to the proliferative processes inherent in such babies).

Abstract:
Background. No consensus has been reached regarding the management of central serous chorioretinopathy (CSCR). We aimed to investigate the efficacy of navigated laser treatment for chronic CSCR and changes in the morphology of the retinal pigment epithelium (RPE). In this retrospective observational study, 19 patients with subjective symptoms admitted to the Nanjing Medical University Affiliated Eye Hospital were included between January 2021 and August 2021. All patients underwent visual acuity and optical coherence tomography (OCT) examination during follow-up. Fluorescein angiography (FA) was performed at baseline and at the final follow-up to confirm the dye leakage sites. The mean logMAR BCVA at baseline and at the end of follow-up was 0.49 ± 0.24 and 0.24 ± 0.15 , respectively. The mean logMAR BCVA 3 months after treatment was significantly better than that before treatment ( p = 0.002 ). Significant improvements were observed in central macular thickness (CMT) and subfoveal choroidal thickness (SFCT) after navigated laser photocoagulation ( p < 0.0001 ). Subretinal fluid was completely resolved in 13 (68%) of 19 eyes at 3 months. RPE lesions on OCT images showed RPE detachment in 17 eyes (61.5%), small protrusion of the RPE layer in 5 eyes (7.5%), and a rough RPE layer in 4 eyes (31%). After laser treatment, 9 points (35%) showed retinal detachment, and 10 (38.5%) regions showed an irregular RPE layer. An irregularly protruded RPE layer was also observed in different regions of the leakage spot in 5 (19%) eyes, and RPE defects were seen in 2 (7.5%) eyes. Navigated laser photocoagulation for chronic CSCR can achieve substantial anatomical and visual improvement. OCT outcomes may provide new information to facilitate understanding of the mechanism of chronic CSCR. Navigated laser photocoagulation should be chosen as an optimal treatment option in patients with chronic CSCR who cannot afford photodynamic therapy.

The body, slightly flattened on the sides, is covered with fairly large scales. The eyes are large, the iris is red. Abstract:
We describe a genetic locus rough deal (rod) in Drosophila melanogaster, identified by mutations that interfere with the faithful transmission of chromosomes to daughter cells during mitosis. Five mutant alleles were isolated, each associated with a similar set of mitotic abnormalities in the dividing neuroblasts of homozygous mutant larvae: high frequencies of aneuploid cells and abnormal anaphase figures, in which chromatids may lag, form bridges, or completely fail to separate. Surviving homozygous adults are sterile, and show cuticular defects associated with cell death, i.e., roughened eyes, sparse abdominal bristles, and notched wing margins. The morphological process of spermatogenesis is largely unaffected and motile sperm are produced, but meiocyte aneuploidy is common. The nature of the observed abnormalities in mitotic cells suggests that the reduced fidelity of chromosome transmission to the daughter cells is due to a failure in a mechanism involved in assuring the proper release of sister chromatids.

L’utopie est la suivante : elle débute par l’injonction amoureuse de Pablo Neruda, ‘‘je veux faire avec toi ce que le printemps fait avec les cerisiers’’, à la suite de laquelle, j’entame une petite collection dépareillée qui met en scène et met ensemble : pièces détachées, éléments brillants, particules rougissantes ou ternies et corps inconnus, qu’il me faudra déployer et marier dans les moindres détails, de novembre 2010 à novembre 2015 pour constituer ce que j’appelle : ‘‘la plastique du corps amoureux’’. J’observerai les effets des images, des corps, de l’œil de l’artiste et celui du regardeur, sous la contrainte du rougissement. Il ne sera pas nécessaire de chercher l’unité mais de bien faire le dessein d’hypothèses amoureuses éparses. Car si la plastique a le pouvoir de faire émerger une forme, celle d’un corps amoureux, ne serait-elle pas une tentative de s’exiler de la forme et de sa propre chair ? Faut-il parler d’informité quand on en vient au couple tant la limite est fragile lorsqu’on s’attaque aux contours ? Que fait-on une fois qu’il n’y a plus d’amour ? Un déséquilibre dans les mécanismes de défense de la cornée peut entraîner une invasion par des micro-organismes (MO) et générer une kératite infectieuse (KI), ou encore un syndrome sec. Les KI sont la première cause de cécité monoculaire. Le syndrome sec, également fréquent, est sujet à de nombreuses études cliniques pour l’évaluation de nouveaux traitements. L’objectif de cette thèse est de mettre au point des méthodes d’imagerie pour le diagnostic et le suivi des pathologies de la surface oculaire au travers de deux projets innovants : 1/ le projet FLUOCOR consistant à développer une solution complète de diagnostic rapide in vivo des MO les plus souvent responsables des KI afin de débuter immédiatement le traitement adapté et améliorer le pronostic visuel final. Cette solution comprendra des BioMarqueurs fluorescents (BMfs) spécifiques des agents infectieux, fluorescents dans le rouge ou le proche infrarouge non éblouissant et un nouvel instrument optique de détection. Des BMfs basés sur le couplage de BODIPY innovant avec des molécules ciblant les MO ont été obtenus. Des tests in vitro ont montré une bonne spécificité mais les tests ex vivo sur cornées humaines ont montré une fixation non spécifique sur les cellules épithéliales. Pour pallier ce manque de spécificité, le couplage de ces BODIPY sur des anticorps dirigés contre les MO est en cours. 2/ le projet FLUOSCOPE qui a permis de développer une nouvelle stratégie d’imagerie de la surface oculaire pour le suivi du syndrome sec, de la méthode d’instillation des colorants jusqu’au traitement d’images, en passant par la conception d’un instrument optique désormais industrialisé par les laboratoires Théa
An imbalance in the defence system of cornea can result in several diseases, like infectious keratitis (IK) in case of invasion by microorganisms (MO) or dry eye in case of insufficient tear quality and/or quality. IK are the first cause of unilateral blindness worldwide. Sicca syndrome, also frequent, is the subject of numerous clinical trials assessing new treatments. The aim of the PhD Thesis work is to develop new imaging methods for the diagnosis and follow-up of diseases of the ocular surface. Two innovative project were conducted: 1/ the FLUOCOR project consists in developing a complete solution of rapid in vivo etiologic diagnosis of IK, for the most frequent and/or severe infections to allow rapid starting of the most adapted treatment in order to improve the final visual prognosis. This solution will comprise fluorescent biomarkers (excitable by sustainable non blinding red or near infrared lights) specific to the target MO, and a new optical imaging device for their detection. Biomarkers based on new BODIPY coupled with molecules targeting the MO were obtained with a good in vitro specificity. Nevertheless, on ex vivo human corneas they stained superficial epithelial cells. In order to overcome this difficulty, the coupling of BODIPY on specific antibodies targeting the MO is ongoing; 2/ the FLUOSCOPE project that allowed development of a new strategy of ocular surface imaging for the diagnosis and follow up of sicca syndrome. Abstract:
Drosophila melanogaster (the fruit fly) is a widely used model organism for the study of human disease condition and genetic pathways due to the high degree of gene conservation between fly and human. The compound eyes of Drosophila comprise of a repetitive arrangement of ommatidia with a honey-comb like an appearance. Any mutation or genetic alteration disturbs the normal development of compound eye and results in 'rough eye' condition. Such rough eye conditions are used to study effects of the disease gene and in the identification of the modifiers that rectify the disease condition. Typically, phenotyping of the rough eye is carried out from the optical microscope and scanning electron microscopic observations of the corneal lens. However, this method is qualitative and very subjective. In order to arrive at a quantifiable parameter, we explore nanoscale morphology, elastic modulus and adhesion force of the corneal lens using atomic force microscope (AFM) and field-emission scanning electron microscope (FESEM). These methods have the advantage of the ease and less time consuming to obtain a large number of data for statistically meaningful evaluation. Though AFM has emerged as an important tool in biology for its high-resolution imaging and mechanical property testing, the latter is not used widely compared to former due to high variability in the measurement. Therefore, parameters bringing variability in measurement of corneal lens properties like sample preparation, aging, usage of solvent, the location of indent are identified and a testing procedure has been drawn for reliable and repeatable measurements. Rough eyes caused by different genetic mutants of Drosophila and human tau disease condition are tested to bring out their corneal lens characteristics. Effect of different levels of a tau protein, same levels of tau isoforms and effect of tau disease modifiers in corneal lens properties are studied. From this study, we identified corneal lens morphologies of tau disease condition is distinctly different from the normal fly condition. Adhesion force to be depended on the levels of the tau protein and isoform. Elastic modulus found to be lower in the tau disease condition and dependent on the type of mutant tau, levels of transcripts and isoform. Under disease suppressor condition, the normal formation of nanoscale morphology and elastic modulus close to that of wild type is observed. Study with genetic mutants of Drosophila enables to identify the molecular player involved in causing distinct nanoscale morphologies and molecules involved in bringing the changes in elastic modulus. Drosocrystallin, Wingless and Vein molecules found to play roles in corneal lens characteristics. Based on these studies, we establish that AFM can be used as a quick tool in identifying quantifiable parameters that measure the physical variations in the corneal lens. In addition, the effect of disease protein, evaluation of disease status, determination of the modifiers, the study of molecular players in controlling the nanoscale morphology, corneal lens strength, identification of genetic players under a disease condition can be made quickly

Abstract:
Horror films in Hong Kong cinema have eschewed terror in favour of comedy, where supernatural beings take the form of hopping vampires, wandering spirits and underworld demons rendered in latex masks and movie slime. This chapter explores the comic presentation of these subjects in Hong Kong horror, where the self-reflexive exposure of the cinematic machinery of costume and special effects appear to put it at odds with the spectral affectivity of the Hong Kong ghost story. This chapter returns to two classic films from the mid-1980s, A Chinese Ghost Story (Tsui Hark 1987) and Rouge (Stanley Kwan 1988), films from the ‘second wave’ period long noted to carry ‘Hong Kong’ as a subject of concern in the run up to the British handover of 1997, and revisits their historical positioning in the light of more recent post-1997 incarnations such as Visible Secret (Ann Hui 2001), My Left Eye Sees Ghosts (Johnnie To 2002), and Rigor Mortis (Juno Mak 2013).

Cat-scratch disease (CSD) is a worldwide zoonoses caused by infection with the bacterium, Bartonella henselae. The formal description of the disease by Debré in 1950 (Debré et al. 1950) corresponds to the most frequently diagnosed form of the disease. Cats are the main reservoir for B. henselae and transmission is via Ctniocephalides felis. Humans usually become infected after being scratched or bitten by a cat and is most frequently seen in children and young adults.CSD is a self-limiting illness which often begins with a small papule developing at the site of cat scratch or bite within 3-14 days of the infection. Nearby lymph nodes, usually neck, axillary or groin, become swollen and can persist for several months. It may take up to 7 weeks for the enlarged lymph nodes to appear and individuals may not recall any cat scratch or bite. In healthy cases antibiotics are not indicated.About 5-10% of patients may develop other forms of CSD including eye infection characterised by conjunctivitis and swollen lymph nodes, rash, liver and spleen enlargement, and more rarely encephalitis. Immunosuppresed patients may develop more severe disease, such as bacillary angiomatosis.General advice for preventing CSD includes avoiding rough play with cats, particularly kittens. Cat scratches and bites should be washed immediately with water and soap and cats should not be allowed to lick open wounds.

Like W. C. Handy and Langston Hughes, Zora Neale Hurston was a translator: she sought textual analogies—words on a page--for the bittersweet lyricism, dynamism, and bold self-declarations found in blues music made by Black people in the rural South of the early Twentieth Century. She was also, like both men, a migrant to the urban North, a key figure of the Harlem Renaissance. A biographical as well as literary-critical exploration, this chapter focuses on Hurston’s two best-known works: Mules and Men (1935), a folklore study, and Their Eyes Were Watching God (1937), a cornerstone of the African American literary tradition. Both works vividly evoke the rough but vital blues culture of rural Florida, offering us Black spaces of self-making through the eyes of a Black female participant-observer. Both texts also force readers to confront the presence of scarifying, sometimes deadly violence within that juke-joint world. Hurston, this chapter argues, uses the novel to rewrite the folklore study, offering us a questing and indomitable young woman, Janie Crawford, who earns her way into the blues and lives out her destiny with the help of Tea Cake, a passionate, adventurous, and mercurial young bluesman.

Just when spring appears to have arrived, a late-season storm blows down from the north. Despite the overcast sky, the temperature at first is beguilingly warm. Rain starts to fall, then changes to sleet as the temperature drops. “Rough winds do shake the darling buds of May,” indeed. The sleet becomes graupel—crusty, rounded pellets of snow—and then wet flakes. Blobs of slush fall from overhanging branches and float briefly down the creek before melting and dispersing. Pulses of wind and snow gust in as birds shelter silently among the densely needled branches of big spruce trees. The moose that has spent the winter around the beaver meadow lies calmly in a protected spot at the base of a spruce. A foot of snow obliterates the newly green shoots of grass. I see no outward indication of it, but perhaps, in the warm darkness of the lodge, the beaver kits have been born. They start small, only about a pound at birth, but they are born fully furred, with open eyes and incisors erupted, almost ready to get down to the business of chewing branches. Each year’s litter is born in May or early June. Usually two to four kits are born, but a litter can be a single kit or as many as eight kits. Baby food for beavers is herbaceous vegetation, which the kits start eating within two weeks. By the end of July or early August the kits will be weaned and able to forage on their own. A varied diet of vegetation allows them to reach a weight of 10 to 16 pounds by the time the ice returns. The first year is a grace period for the new kits. Unlike the yearlings, the kits do not help maintain the lodge or cache food against the lean days of winter. They simply get to enjoy life and explore the wondrous new world into which they have been born. Diverse human observers watching this exploration by young beavers have interpreted their activities as an expression of joy.

As he propped himself up on his charpoy, he told me, his eyes adjusted to the darkness of his tiny room, tucked away in a slum near New Delhi’s prosperous Vasant Kunj neighborhood. It was, he recalls, pitch black. Kumar’s inability to see enhanced his other senses. His ears picked up a chorus of mournful howls from the stray dogs roaming the streets some distance away. A few seconds later, he began to take account of a gentle rumble of snoring. Bandhana, his wife, was still asleep beside him. The next stimulation to hit him, he said, was that of smell: they had saved leftovers from dinner in a small dish, placed on the floor in one corner of the bedroom. (There was no kitchen, no fridge, no cupboards.) The smell of stale dal and starchy rice lingered in the air, mingling with the stench of urine from the latrine right outside. The apartment’s only toilet had no working flush, just a mug and a bucket with which to pour water. At night, Kumar’s father—who slept in the next room—would get up to urinate in the dark, sitting on his haunches and peeing all over the cramped toilet floor. Kumar was only twenty-two years old but looked much older. He was developing a hunch. His gray shirt and trousers—both made of the same rough, fraying cotton—seemed to fall over his limbs like oversized bags. Life was wearing him down. Kumar was working sixteen-hour shifts driving a three-wheeled auto-rickshaw, seven days a week. Ever since he had gotten married and brought Bandhana to join him in the city, his expenses had ballooned. His father, meanwhile, was working less and less, his health and eyesight beginning to fail him. Asha Ram Kumar worked as a chauffeur for a family nearby in Vasant Kunj. He had recently bumped their car into a lamppost. Any day now, Kumar told me, his father would lose his job.

In the design of kansei (emotional) quality, one of the important issues is to extract causal relations between physical design attributes and the customer’s emotional responses. Without such relations, a designer has to rely on his/her own sense that may be different from the customer’s. In this paper, we propose a new method for extraction of logical rules consisting of combinations of design attributes that explain a customer’s emotional judgment towards product appearance. In the method, we apply a reduct calculation in rough set theory to derive alternatives of causal rules between design attributes and emotional judgments, and use the customer’s eye gaze features for refining the rules. We extract two types of visual attentions (VA), i.e., a single visual attention (SVA) and a combinational visual attention (CVA), by using the proposed gaze features. To demonstrate the effectiveness of the method, we conducted a sensory evaluation experiment using a car-interior design as a case study. In the experiment, multiple participants evaluated impressions of multiple design samples by selecting from a set of words. During the experiment, we recorded the participants’ eye gaze movements as coordinates on a screen, and asked them to vocalize aloud what they were thinking. After an evaluation of each design sample, we conducted a retrospective interview. From the results, we confirmed that the estimated SVA and CVA significantly covered the vocalized thoughts and statements made in the retrospective interview. The estimated VA reduced 53% of the erroneous causal rules and improved the quality of the rules. We found a case where two participants making the same emotional judgments have implicitly different points of view when evaluating the same design sample. Most conventional causality analysis has been unsuccessful in finding such diversity of points of view.

Vehicles with better usability have become increasingly popular due to their ease of operations and safety for driving. However, the way how usability of in-vehicle system user interface is studied still needs improvement. This paper concerns how to use advanced computational, neurophysiology- and psychology-based tools and methodologies to determine affective (emotional) states and behavioral data of an individual in real time and in turn how to adapt the human-vehicle interaction to meet the user’s cognitive needs based on this real-time assessment. Specifically, we set up a set of neuro-physiological equipment that is capable of collecting EEG, facial EMG (electromyography), skin conductance response, and respiration data and a set of motion sensing and tracking equipment that is capable of eye ball movement and objects that the user interacts. All hardware components and software is integrated into a cohesive augmented sensor platform that can perform as “one coherent system” to enable multi-modal data processing and information inference for context-aware analysis of affective and cognitive states based on the rough set inference engine. Meanwhile subjective data is also recorded for comparison. A usability study of in-vehicle system UI is shown to demonstrate the potential of the proposed methodology.
