Abstract:
Perivascular lesions within the central nervous system (CNS) of rats with hyperacute experimental autoimmune encephalomyelitis (HEAE) contained large numbers of peripheral blood mononuclear cells (PBMC) and polymorphonuclear leukocytes (PMN), cells enzymatically capable of producing reactive nitrogen and oxygen intermediates (RNI and ROI), which, in excess, are mediators of tissue damage. PBMC and PMN isolated from the CNS and periphery of HEAE-affected rats secreted significantly (p less than 0.01-0.0001) elevated levels of ROI and RNI compared with that of similar cell populations from pertussis- and saline-treated control animals. Coincubation of systemically derived PBMC and PMN with antigen-stimulated myelin basic protein-specific T cell lines led to further increases in ROI and RNI output of between 15.3 and 83.1%, an effect that could be largely attributed to heat-labile, soluble products released by these T cell lines. Our studies suggest a putative neuropathological role for ROI and RNI in HEAE, which may be mediated via cytokines emanating from autoreactive T lymphocytes.

Abstract:
ABSTRACT Reactive oxygen intermediates (ROI) and reactive nitrogen intermediates (RNI) produced by activated macrophages participate in host defense against the facultative intracellular pathogensMycobacterium tuberculosis and Salmonella typhimurium. To survive within macrophages, such pathogens may have evolved ROI and RNI resistance mechanisms. ROI resistance pathways have been intensively studied. Much less is known about the mechanisms of resistance to RNI. To identify possible RNI resistance genes inM. tuberculosis, a mycobacterial library was expressed inS. typhimurium and subjected to selection by exposure to the NO donor S-nitrosoglutathione (GSNO) in concentrations sufficient to kill the vast majority of nontransformed salmonellae. Among the rare surviving recombinants was a clone expressingnoxR3, a novel and previously anonymous M. tuberculosis gene predicted to encode a small, basic protein. Expression of noxR3 protected S. typhimuriumnot only from GSNO and acidified nitrite but also from H2O2. noxR3 is the third gene cloned from M. tuberculosis that has been shown to protect heterologous cells from both RNI and ROI. This suggests diversity in the repertoire of mechanisms that help pathogens resist the oxidative and nitrosative defenses of the host.

Abstract:
Among the major antimicrobial products of macrophages are reactive intermediates of the oxidation of nitrogen (RNI) and the reduction of oxygen (ROI). Selection of recombinants in acidified nitrite led to the cloning of a novel gene, noxR1, from a pathogenic clinical isolate of Mycobacterium tuberculosis. Expression of noxR1 conferred upon Escherichia coli and Mycobacterium smegmatis enhanced ability to resist RNI and ROI, whether the bacteria were exposed to exogenous compounds in medium or to endogenous products in macrophages. These studies provide the first identification of an RNI resistance mechanism in mycobacteria, point to a new mechanism for resistance to ROI, and raise the possibility that inhibition of the noxR1 pathway might enhance the ability of macrophages to control tuberculosis.

Abstract:
ABSTRACT The contribution of reactive oxygen intermediates (ROI) and reactive nitrogen intermediates (RNI) to the killing of Listeria monocytogenes by macrophages activated by addition of spleen cells from listeria-immune mice plus specific antigen was examined. When macrophages were infected with L. monocytogenes and then spleen cells were added, there was not as big a difference in listericidal activity between macrophages cultured with normal spleen cells and those cultured with immune spleen cells as expected. In this culture system, RNI was mainly involved in the macrophage intracellular killing. In macrophages first activated and then infected, a significant level of enhanced killing was observed. Blockade of ROI production drastically affected the enhanced killing ability, while inhibition of RNI production had a negligible effect. Thus, the contributions of ROI and RNI to listericidal mechanisms of macrophages were different between macrophages activated at pre- and postinfection stages.

Abstract:
Tuberculosis remains one of the major infectious causes of morbidity and mortality in the world, yet the mechanisms by which macrophages defend against Mycobacterium tuberculosis have remained obscure. Results from this study show that murine macrophages, activated by interferon gamma, and lipopolysaccharide or tumor necrosis factor alpha, both growth inhibit and kill M. tuberculosis. This antimycobacterial effect, demonstrable both in murine macrophage cell lines and in peritoneal macrophages of BALB/c mice, is independent of the macrophage capacity to generate reactive oxygen intermediates (ROI). Both the ROI-deficient murine macrophage cell line D9, and its ROI-generating, parental line J774.16, expressed comparable antimycobacterial activity upon activation. In addition, the oxygen radical scavengers superoxide dismutase (SOD), catalase, mannitol, and diazabicyclooctane had no effect on the antimycobacterial activity of macrophages. These findings, together with the results showing the relative resistance of M. tuberculosis to enzymatically generated H2O2, suggest that ROI are unlikely to be significantly involved in killing M. tuberculosis. In contrast, the antimycobacterial activity of these macrophages strongly correlates with the induction of the L-arginine-dependent generation of reactive nitrogen intermediates (RNI). The effector molecule(s) that could participate in mediating this antimycobacterial function are toxic RNI, including NO, NO2, and HNO2, as demonstrated by the mycobacteriocidal effect of acidified NO2. The oxygen radical scavenger SOD adventitiously perturbs RNI production, and cannot be used to discriminate between cytocidal mechanisms involving ROI and RNI. Overall, our results provide support for the view that the L-arginine-dependent production of RNI is the principal effector mechanism in activated murine macrophages responsible for killing and growth inhibiting virulent M. tuberculosis.

Abstract:
Abstract Isoelectric focusing was used to separate proteins from cyst extracts of potato cyst nematode (PCN) populations. In a first set of assays, cyst extracts from standard populations of Globodera rostochiensis pathotypes Ro1, Ro2, Ro3, Ro2/3, Ro4, and Ro5, and G. pallida pathotypes Pa2 and Pa3, were loaded on isoelectric focusing gels. Gels were stained for superoxide dismutase (SOD), esterase, and glucose-6-phosphate isomerase (GPI). Twelve bands of SOD activity were detected, six (B1-B6) migrating towards the basic zone and the other six (A1-A6) migrating towards the acidic zone, starting from the loading point. A cluster analysis was carried out based on a data matrix that reported the presence or absence of SOD bands on the isozyme electrophoresis patterns (IEPs). Globodera spp. were clearly distinguished and, within G. rostochiensis, Ro2 and Ro4 shared a high level of similarity, respectively, with Ro3 and Ro5; moreover, Ro1 could be clearly distinguished from Ro2/3 and Ro4/5. Globodera pallida Pa2 and Pa3 also shared a high level of similarity. In contrast, esterase and GPI IEPs did not discriminate among G. rostochiensis standard pathotypes. Subsequently, 14 field populations of G. rostochiensis, five from Italy and nine from Venezuela, and three field populations of G. pallida, two from Italy and one from Chile, were assayed to obtain SOD IEPs. Italian populations had previously been identified at pathotype level by bioassays according to the generally accepted international test using different resistant potato cultivars and clones. The cluster analysis carried out on the SOD IEPs of all the populations tested formed four distinct groups within G. rostochiensis and only one within G. pallida. Pathotype identification of Globodera populations by SOD IEPs was not able to discriminate between bioassay standard couples Ro2/Ro3, Ro4/Ro5 and Pa2/Pa3. Therefore, three groups were assigned to Ro1, Ro2/3 and Ro4/5, and a fourth group to Pa2/Pa3. Four Venezuelan populations, not identified at pathotype level by bioassays, formed a distinct fifth group. By means of the method described herein, four additional unknown Venezuelan populations could be assigned to Ro1 group and one to Ro2/Ro3 group; one G. pallida population from Chile was assigned to Pa2/Pa3 group.

Abstract:
ABSTRACT The killing of blood-stage malaria parasites in vivo has been attributed to reactive intermediates of oxygen (ROI) and of nitrogen (RNI). However, in the case of the latter, this contention is challenged by recent observations that parasitemia was not exacerbated in nitric oxide synthase (NOS) knockout (KO) (NOS2−/− or NOS3−/−) mice or in mice treated with NOS inhibitors. We now report that the time course shows that Plasmodium chabaudi parasitemia in NADPH oxidase KO (p47phox−/−) mice also was not exacerbated, suggesting a minimal role for ROI-mediated killing of blood-stage parasites. It is possible that the production of protective antibodies during malaria may mask the function of ROI and/or RNI. However, parasitemia in B-cell-deficient JH −/− × NOS2−/− or JH −/− × p47phox−/− mice was not exacerbated. In contrast, the magnitude of peak parasitemia was significantly enhanced in p47phox−/− mice treated with the xanthine oxidase inhibitor allopurinol, but the duration of patent parasitemia was not prolonged. Whereas the time course of parasitemia in NOS2−/− × p47phox−/− mice was nearly identical to that seen in normal control mice, allopurinol treatment of these double-KO mice also enhanced the magnitude of peak parasitemia. Thus, ROI generated via the xanthine oxidase pathway contribute to the control of ascending P. chabaudi parasitemia during acute malaria but alone are insufficient to suppress parasitemia to subpatent levels. Together, these results indicate that ROI or RNI can contribute to, but are not essential for, the suppression of parasitemia during blood-stage malaria.

Abstract:
This study aims to analyse the effect of ratios of liquidity, solvency and activity on financial distress through ROE, ROA and ROI of PT Angkasa Pura II (Persero) as mediating variable. The decrease in the number of aircraft passengers due to restrictions on human movement due to Covid-19 pandemic affects airport revenues as indicated by deteriorating financial ratios and is thought to lead to financial distress. This study tries to detect the symptoms of financial distress in the state-owned enterprise engaged in airport services by looking at the direct and indirect effects of its financial ratios’ performance. By using secondary data from financial statements, this study tried to build a structural equation model to know the direct and indirect effect. The results of hypothesis testing indicated that the ratio of liquidity, solvency and activity had negative effects on the company’s ROE, ROA and ROI during the period 2001-2020, but only the activity ratio that had a significant effect. Meanwhile, the three ratios of liquidity, solvency and activity had direct negative but not significant on financial distress. Next, ROE, ROA, and ROI had a direct positive but not significant effect on the company’s financial distress during that period. The study concludes that there was no relationship between the three ratios to financial distress mediated by ROE, ROA and ROI of the company.

Abstract:
ABSTRACT Host control of Coxiella burnetii infections is believed to be mediated primarily by activated monocytes/macrophages. The activation of macrophages by cytokines leads to the production of reactive oxygen intermediates (ROI) and reactive nitrogen intermediates (RNI) that have potent antimicrobial activities. The contributions of ROI and RNI to the inhibition of C. burnetii replication were examined in vitro by the use of murine macrophage-like cell lines and primary mouse macrophages. A gamma interferon (IFN-γ) treatment of infected cell lines and primary macrophages resulted in an increased production of nitric oxide (NO) and hydrogen peroxide (H2O2) and a significant inhibition of C. burnetii replication. The inhibition of replication was reversed in the murine cell line J774.16 upon the addition of either the inducible nitric oxide synthase (iNOS) inhibitor NG-monomethyl-l-arginine (NGMMLA) or the H2O2 scavenger catalase. IFN-γ-treated primary macrophages from iNOS−/− and p47phox−/− mice significantly inhibited replication but were less efficient at controlling infection than IFN-γ-treated wild-type macrophages. To investigate the contributions of ROI and RNI to resistance to infection, we performed in vivo studies, using C57BL/6 wild-type mice and knockout mice lacking iNOS or p47phox. Both iNOS−/− and p47phox−/− mice were attenuated in the ability to control C. burnetii infection compared to wild-type mice. Together, these results strongly support a role for both RNI and ROI in the host control of C. burnetii infection.

Abstract:
Purpose Leveraging social action theory, social network theory and the notion of network externality, the purpose of this paper is to model two different return on investment (ROI) measures: the networked ROI which captures the network effect originating from a social media investment, and the discrete ROI which focuses social media discrete returns from individual users. Design/methodology/approach A field experiment was set up over a period of three months to test the effects of two variants of an advertisement campaign (a social vs a discrete ad) on the modeled networked and discrete ROIs. Findings The authors find that emphasizing discrete user actions leads to lower network gains, but higher monetary returns while the social action emphasis produces higher network gains, but lower monetary returns. The study further suggests that social action focus is preferable for brand promotion and engagement, whereas the discrete action focus is suitable for boosting sales and website traffic. Practical implications Several potential implications for social media researchers and marketers are also discussed. Originality/value The authors for the first time showed that that the social media returns are derived not only from individual actions taken by the user (e.g. likes and shares) but also from users’ social interdependencies and the additional exposure that results from network effects.

Abstract:
Aim. To assess external respiration (ER) and its relationship with the activity of enzymes involved in purine metabolism in patients with acute and chronic forms of pulmonary tuberculosis (TB).Materials and methods. In patients with acute and chronic TB, we assessed the activity of adenosine deaminase (ADA)-1, 2 in the blood serum (eADA), mononuclear cells, and neutrophils, the concentration of ecto-5’-nucleotidase (eNT5E) in the blood serum, the level of CD26 (dipeptidyl peptidase-4, DPPIV) in the blood serum and mononuclear cells, production of reactive oxygen intermediates (ROI) and reactive nitrogen intermediates (RNI) in mononuclear cells and neutrophils, as well as parameters of ER.Results. Patients with TB were found to have an increase in the concentration of eNT5E and eADA-2 activity in the blood serum, stimulated production of ROI in neutrophils, a decrease in the concentration of DPPIV (CD26) in mononuclear cells, and a fall in the production of RNI in mononuclear cells and neutrophils. In patients with chronic TB, a decrease in the activity of ADA-1 in mononuclear cells and a fall in the concentration of DPPIV (CD26) in the blood serum were noted. In patients with acute TB, a decrease in the activity of eADA-1 in the blood serum and ADA-1 in neutrophils, reduced production of ROI in mononuclear cells, and an increase in spontaneous production of ROI in neutrophils were revealed. Correlations were found between the parameters of ER and the concentration of eNT5E in the blood serum, spontaneous production of ROI in mononuclear cells and production of RNI in neutrophils in chronic TB, as well as between eADA-2 in the blood serum, ADA-1 in neutrophils, DPPIV (CD26) activity in mononuclear cells, and ROI and RNI production in mononuclear cells and neutrophils.Conclusion. The data obtained make it possible to associate regulation of external respiration with parameters of purine metabolism, in particular with the concentration and activity of enzymes responsible for generation and metabolism of adenosine, that determine its level outside cells and inside mononuclear cells and neutrophils, with expression of cofactor molecules, as well as with the duration of activation of cells in innate immunity, neutrophils, and monocytes/ macrophages, determined largely by the potential of adenosine regulation.

Abstract:
Abstract Systemic lupus erythematosus (SLE) is a protean autoimmune disease characterized by a wide array of intrinsic T cell defects. Reactive oxygen intermediates (ROI) modulate T cell activation, cytokine production, and proliferation at multiple levels. The purpose of this study was to measure T cell proliferation and ROI production in SLE versus healthy control T cells. CD4+ and CD8+ T cells were isolated by negative selection magnetic micro-beads. We characterized the memory phenotype and activation markers on the cells from both groups by flow cytometry. CD62Lhigh and CCR7high were the most abundant subsets in CD8+ T cells from healthy subjects. The frequency of CCR7highCD8+ T cells in SLE patients was significantly lowered compared to healthy subjects (p=0.0001). For proliferation and ROI studies, cells were stimulated with anti-CD3 and anti-CD28 antibodies. CFSE staining following activation revealed that SLE CD8+ T cells had decreased division in response to TCR-mediated stimulation as compared to healthy subjects. Both subsets of SLE T cells showed increased ROI in response to activation. Interestingly, among the SLE subjects, the maximum increase in H2O2 production was recorded in the CD8+ T cells of one patient with severe SLE disease. In conclusion, we demonstrate that T cells from SLE patients produce increased levels of ROI upon stimulation implicating ROI in SLE pathogenesis. Supported by NIH RO1-AR39501
