Abstract:

Rossi, Nicolo. "Sintesi di PNA a libertà conformazionale ridotta." Master's thesis, Alma Mater Studiorum - Università di Bologna, 2014. http://amslaurea.unibo.it/7345/.

Full text
Abstract:
The present work started a research project aimed at the synthesis of conformationally “locked” PNA (Peptide Nucleic Acids) monomers. Compared to classic aeg-PNA, this structural modification would result in an improvement in the pairing properties with natural nucleic acids, due to entropic variations in the process. Specifically, an attempt was made to build a PNA monomer around a β-lactam ring. That ring could be imagined as obtained by linking the methylene groups in α position of both the nucleobase and the carboxyl function. These structural properties would imply pre-organization of the final oligomer, improving the pairing process in biological systems. The first step of this work was the investigation of the Staudinger reaction for the ciclization of the lactam ring, and in particular the activation method of the carboxylic group of the nucleobase derivatives. Use of triazine chloride led to the synthesis of the adenine-based β-lactam-PNA. Attempts to synthesize the same monomer based on cytosine, guanine and thymine were unsuccessful, so alternative methods for carboxylic group activation were investigated. Conversion of carboxylic acids to acyl chlorides led to a partial result: despite the method worked well with analogues of the final reactants, it didn’t worked with substrates needed for lactam based PNAs. Search for a valid activation process continued involving carbonyl diimidazole, Mukayama reagent, and LDA (with methylester derivative of nucelobase) without good results. Last, it was investigated a different synthetic approach by first synthesizing a proper backbone with a chlorine in the β- lactam ring. This chlorine ring should undergo substitution by a nucleobase anion to give the desired PNA monomer. Unluckily also this synthetic route didn’t lead to the desired monomers.

Abstract:

Abstract:
Four captivity narratives set in the Great Lakes region during the second half of the eighteenth century feature scenes in which Native Americans present the authors with books. These book presentations were symbolic interactions, in which the Indians affirmed their recognition of the value of books to the colonists. When the adopted captive James Smith lost his books, he feared for his life; by finding them and restoring them to him, his Kahnawake Mohawk kin paradoxically enabled his immersion in their society. For the diplomat Thomas Morris, who was detained by Miamis, and Thomas Ridout and Charles Johnston, who were both captured by Shawnees, their books facilitated their participation in secular literary culture. For Morris and Ridout, especially, the books furnished striking allegorical parallels to their experiences.

Full text
Abstract:
Designing complex internal cooling passages is extremely difficult without detailed information about the flow behavior due to the presence of separated flow zones and strong secondary flows. This paper describes a new approach to designing internal flow passages called Rapid Iterative Design Using Experimentation (RIDUE). RIDUE utilizes rapid prototyping (RP) manufacturing to quickly build an accurate model of a complex internal flow passage and magnetic resonance velocimetry (MRV) to measure the full three dimensional velocity field. Because both techniques offer very fast turnaround, it is feasible to conduct iterative design with a cycle time of 1–2 days. RIDUE is demonstrated using a generic turbine blade internal cooling passage with four serpentine channels. Two channels have rectangular cross-section and two are square. In each channel, two of the four walls have ribs (also called turbulators) angled at 45 degrees to the main flow direction. Two models based on the generic geometry are studied, each with different turbulator cross-sections, one square and one rounded. Both models were built using a stereo lithography apparatus. MRV provided three-component velocity vectors for flow at a Reynolds number of 10,000 based on the hydraulic diameter of the first passage. Sample vector fields are presented to illustrate the detail with which the flow can be investigated. Although little difference is seen in the flows between the two models, it is demonstrated that through its use of RP processes and the MRV measurement technique, RIDUE is a viable technique for modern internal passage design.

Abstract:
Conservation and enhancement of ruined fortified systems in Tanaro Valley (Piedmont)The contribution reports on a research concerning the preparatory investigation of some fortified systems of Tanaro Valley (Piedmont) in view of their restoration. The research highlighted the strong connection between the castles and the memory of events and characters that were linked to these architectures; the relationship between landscape and ruins that, today, represents a potential resource for the territory; the willingness of local authorities to improve knowledge, protection and enhancement of this heritage; the need to investigate theoretical and methodological issues and define the proper procedures for the conservation, restoration, safeguard, maintenance and management of a widespread and vulnerable heritage. The study, taking advantage of interdisciplinary contributions, used innovative instruments for the survey and monitoring of this heritage, as well as graphic and digital restitution techniques to propose to the scientific community –and communicate to the native population– the interventions of restoration, conservation, enhancement and management of these fortified systems. The research demonstrates that particular attention has to be paid to these less-known historical architectures that can however contribute, as effectively as the most famous and eye-catching fortification systems, to the political and social development and to the historical identity whose recognition, preservation and valorization are strongly hoped for.

Abstract:
The physical and conceptual result of the great changes which affect our society is a city in ‘stand-by’. Even if the XXI century is considered to be an ‘urban century’, many Western cities are shrinking and lack the resources which are necessary to complete planned masterplans. By postponing the transformation of the city to an indefinite future, the number of vacant spaces is increasing. This situation reduces the city into an amorphous landscape that also reflects the social problems concerning a wide number of citizens. In this uncertain scenario, the temporary project becomes an effective tool to open the urban space toward its use as well as to avoid a further degradation of the landscape. Through interim practices the 'forced breaks', to which the city is constrained, can be transformed into moments of experimentation, becoming an opportunity to reflect on the means and methods of the project itself. This work proposes a reflection on the concept of time based on the idea of real duration introduced by Bergson. 'Activating' usually unused sequences of time, the interim project works as a dynamic means of urban regeneration based on the empowerment of local communities. Finally, the aim of this paper is to summarize the features that make the intermediate project able to trigger a transformation of the city based on a new way to participate, attempting to strengthen the resilient capacity of the people involved. For this purpose several European projects are collected and classified into four dimensional categories, showing the adaptability of the temporary project at all scales and levels of planning.
