Abstract:
Abstract Regimen-related toxicity, one of the main reasons for discontinuation of chemotherapy in acute lymphoblastic leukemia (ALL), can not only be life-threatening but may affect the risk of relapse. In 246 children with newly-diagnosed ALL who were treated using a single protocol (St. Jude Total XIIIB), we used a candidate-gene approach to determine whether acute toxicities [gastrointestinal (stomatitis or diarrhea), hyperbilirubinemia, infection, or neurotoxicity] were related to 16 common polymorphisms in genes plausibly linked to the pharmacodynamics of the drug therapy. During the high-dose methotrexate consolidation therapy, only genotypes related to methotrexate disposition were associated with grade 3–4 gastrointestinal toxicity, adjusting for demographic factors and ALL risk group. Patients having the RFC AA/AG (p = 0.025) genotypes or those having the MTHFR 677 TT (p = 0.048) genotypes had a higher risk of the toxicity (OR = 10.4 and 3.2 respectively), than those with RFC GG or MTHFR CT/CC. In addition, the risk of hyperbilirubinemia during consolidation was higher among those with the lower activity UGT1A1 7/7 genotype than those with other UGT1A1 genotypes (OR = 12.2, p < 0.0001). Adjusting for time at risk over the entire therapeutic period, RFC AA/AG was again prognostic for gastrointestinal toxicity (p < 0.0001); the GSTP1 AA/AG genotypes (p = 0.021) and TPMT heterozygous genotypes (p = 0.047) were also risk factors. The RFC AA/AG genotype was also a modest risk factor for infection (p = 0.05). There were only weak predictors of neurotoxicity, with the MDR1 exon 26 TT genotype a risk factor (p = 0.025) for presumed methotrexate-related neurotoxicity over the entire therapeutic period. We conclude that germline polymorphisms influence the toxicity of antileukemic therapy and their identification may provide a tool for tailoring therapy in childhood ALL.

Master's thesis, 2017. http://hdl.handle.net/11110/1282.

Abstract:

Abstract:
This paper continues the pursuit of getting a deeper understanding regarding the transient stability of low-frequency AC railway power systems operated at 16 2/3 Hz synchronously to the public grid. The focus is set on the impact of different load models. A simple constant-current load model is proposed and compared to a previously proposed and studied load model in which the train’s active power is regulated. The study and comparison is made on exactly the same cases as and grid as with the already proposed and more advanced load model. The railway grid is equipped with a low-frequency AC high-voltage transmission line which is subjected to a fault. The study is limited to railways being fed by different distributions of RFC (Rotary Frequency Converter) types. Both AT (auto transformer) and BT (booster transformer) catenaries are considered. The RFC dynamic models are essentially Anderson-Fouad models of two synchronous machines coupled mechanically by their rotors being connected to the same shaft. The differences in load behaviour between the proposed constant-current load model and the previously proposed and studied voltage-dependent active power load model are analyzed and described in the paper.

Abstract:
In this study, a three-dimensional numerical investigation on the thermo-hydrodynamic performance of a newly proposed recharging microchannel (RMC) is carried out. In this new design, a straight microchannel separated into more than one small channels and each small channels having individual inlet and outlet. This design enhances the heat transfer and makes the temperature almost uniform across the length of the substrate. The comparison of fluid flow and heat transfer performance between this recharging microchannel (RMC), interrupted microchannel (IMC) and straight microchannel (SMC) with same hydraulic diameter and substrate length were conducted to explore the effect of geometrical configuration on the heat transfer enhancement. The results reveal that for the recharging microchannel, the average Nusselt number increases by 49–122%, while the total pressure drop increases by 15–89%, compared with the interrupted and straight microchannel for the Reynolds number ranging from 100 to 500. From the result, it is also observed that for the investigated Reynolds number range the recharging microchannel having the highest thermal performance compared to interrupted and straight microchannel with a maximum performance factor of 1.80. The outcome of this study indicates possible use of recharging microchannel heat sinks for high heat flux removal applications such as electronic cooling.
