Abstract:
pSCL2 (120 kb), one of the linear plasmids found in Streptomyces clavuligerus NRRL3585, was isolated and partially sequenced. Computational analysis of the central region of pSCL2 revealed the presence of two open reading frames that appear to encode proteins highly homologous to RepL1 and RepL2, replication proteins from pSLA2-L, the large linear plasmid in Streptomyces rochei. The S. clavuligerus open reading frames were designated repC1 and repC2, encoding the proteins RepC1 (150 amino acids) and RepC2 (102 amino acids), respectively. The RepC and RepL proteins have identical translation features and very similar predicted secondary and tertiary structures. Functional analysis confirmed that RepC1 is essential for replication initiation of pSCL2, whereas RepC2 is dispensable but may play a role in copy number control. The RepC and RepL proteins do not show similarity to any other bacterial plasmid replication proteins. Three regions of DNA sequence, Box 1 (1050–850 bp), Box 2 (723–606 bp), and Box 3 (224–168 bp), located upstream of repC1, were also shown to be essential or very important for replication of pSCL2.Key words: pSCL2, Streptomyces clavuligerus, replication origin.

Abstract:
Overlapping disease aetiologies associated with multiple altered biological processes have been identified that change the endometrial function leading to recurrent implantation failure (RIF) and recurrent early pregnancy loss (REPL). We aimed to provide a detailed insight into the nature of the biological malfunction and related pathways of differentially expressed genes in RIF and REPL. Endometrial biopsies were obtained from 9 women experiencing RIF, REPL and control groups. Affymetrix microarray analysis was performed to measure the gene expression level of the endometrial biopsies. Unsupervised clustering of endometrial samples shows scattered distribution of gene expression between the RIF, REPL and control groups. 2556 and 1174 genes (p value < 0.05, Fold change > 1.2) were significantly altered in the endometria of RIF and REPL patients’ group, respectively compared to the control group. Downregulation in Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways of the differentially expressed genes (DEGs) in RIF and REPL including ribosome and oxidative phosphorylation pathways. Gene Ontology (GO) analysis revealed ribosomes and mitochondria inner membrane as the most significantly downregulated cellular component (CC) affected in RIF and REPL. Determination of the dysregulated genes and related biological pathways in RIF and REPL will be key in understanding their molecular pathology and of major importance in addressing diagnosis, prognosis, and treatment issues

El propósito de este trabajo fue evaluar el estrés social percibido y su relación con las lesiones deportivas, en jugadores de tenis. Sesenta y tres tenistas masculinos, con una media de edad de 31,62 años (DT=8,93), pertenecientes a Clubes Deportivos de la Provincia de Alicante (España), cumplimentaron instrumentos de estrés psicosocial y de lesiones sufridas durante el año anterior a la evaluación. Los resultados indican que los sucesos vitales experimentados se relacionan con algunas de las lesiones sufridas por los tenistas. Se han encontrado relaciones entre el grado de tensión psicológica experimentada, y la evaluación negativa de esta tensión, con el tipo (lesiones en muñecas, tobillos y esguinces), y la gravedad de las lesiones (lesiones leves).

In our ever changing understandment of city form, the interpretation and realization of space sometimes transcend the notion of time and the physicality of given boundaries. In the city of pastiche, a postindustrial condition has revealed a reading of the city that begins to question our temporal existence ... that we attempt to make a place of discourse, a place to create, contemplate and learn. This thesis proposes that this place be an extension of ourselves, of our city and beyond ... an urban oasis for the freedom of expression. In the advent of the intelligent age the place of pilgrimage becomes the shrine of information. The interface of information and knowledge to the civic arena anticipates a place for equity ... a collective intelligence. It is my intention to explore this dialectic condition in the context of a Center for Inter-Related Media for the city of Boston. The purpose of the Center will be to act as civic informant in the research and discernment of media upon our society. Abstract:
As architectures of the sea, cruise ships are unique, but under-explored spaces of travel and tourism. Characterised by their bounded, mobile and transient distinctiveness, a ship’s inner spaces are inhabited by an onboard community of people for purposes of work or leisure. Framed by the notion of cruise ships as imagined geographies, the primary focus of this study is to interpret and critically explore the discourses of popular films and what these reveal about cruise ships as distinctive socio-cultural spaces of travel and tourism. Embracing Michel Foucault and his influential work on discourse, knowledge and power, popular films are positioned as sites of knowledge production through which particular regimes of truth are constituted. The approach to the analysis of popular films in this study, acknowledges the multisensory nature of film and how such spatial constructs are experienced in and through the imagination, emotion and memory. Conceptualised as liminal spaces which can be temporarily inhabited and travelled through, my embodiments of Carry On Cruising [1962], Ship of Fools [1965] and Titanic [1997] are reflexive in their approach, enabling me to draw on my own experiences of living and working on cruise ships. My voyages with the three films that form the archive of data for the purposes of this study, reveals a uniform set of discourses that engender specific imaginings about travel and vacations at sea. Although different in genre, these films all contribute in varying ways to the notion of ships as hedonistic pleasure spaces of promiscuity and romantic pursuit. Through this archaeological endeavour my excavations also unveil a set of more nuanced and subtle discourses surrounding gender, employment practises, the spatial organisation of ships and shipboard rituals, all of which contribute to particular ways of seeing cruise ships and the cruise experience. In the context of tourism studies, this research makes a methodological contribution in its approach to visuality, discourse and film. Moreover this thesis contributes to a greater understanding of the discursive structures of popular films and what these reveal about the spatial constructs of passengers ships and their embodied cultural practices.

Abstract:
Taking as a starting point Pinter's statement that The more acute the experience, the less articulate its expression', this thesis offers a theorisation of that essential point beyond representation, through Lacan's objet petit a, the focal point of the subject's desire. It is this small object, unarticulated in language, unrepresented in the visual field, that is most acute for the subject, and more real than external reality. It is a structure applicable to poetry, psychoanalysis and film, and it is through Pinter's screenplays that this approach is made. Using previously unpublished material from the Pinter Archive, the progress of each screenplay is charted to find Pinter working towards just such a structure of desire for the central character within the narrative, and for the spectator. Chapter one outlines the basic premise of Lacanian theory and its relevance to the most recent writing on film. A direct link is established between Pinter and the Surrealists through Pinter's unpublished poem 'August Becomes', placing vision at the centre of being, and connecting Pinter, through the Surrealists, to Lacan. The construction of an object of desire is outlined in general terms within the screenplays, and the chapter concludes by identifying three different aspects of the object. The first two aspects are those of lack, which evokes desire: the object which is eternally lost, and can only be retrieved in fantasy or dream, and the object which, aligned to a real object in the external world, will change once that real object is achieved. The third aspect emerges when instead of a lack we encounter a fullness, which destroys the relationship with desire, and causes anxiety. Chapter two is a resume of all the screenplays to date in the light of this reading, while chapters, three four and five, offer a close reading of three screenplays: The Remains of the Day, The Handmaid's Tale and Victory, each of which offers a different aspect of the object as outlined above. In chapter six this approach is offered as a reading of Pinter's stage plays. Finally, a postscript outlines Pinter's latest screenplay, The Dreaming Child, which reinforces the subject of this thesis, that it is the object of desire which is more real, more acute than external reality. Throughout the screenplays Pinter can be seen to shape narrative and structure to create just such an acute, invisible object for his spectator, placing her in a vacillating relationship with desire.

Abstract:
In this study, I used a lens of performance theory to examine the creative collaborative processes of middle school students who composed digital videos. More specifically, I investigated the multiliteracies involved in a filmmaking camp and how students performed those literacies in ways that influenced the composition processes and the resulting texts. In order to study collaborative composition processes, I used ethnographic methods. In order to analyze data, I employed a mixed methodology of constant comparative analysis and dramaturgical analysis of interactions in three main informant groups in order to understand how students used multiple literacies to influence the composition processes and products. During these processes, students employed tactics and style to gain authority over designing group attention to their ideas. This resulted in an overall model of PAID Attention (paying attention, attracting attention, immersing attention, and designing attention). The use of influential literacies in this project was two-fold: students used literacies to influence texts, and as a result, those texts required the students' attention. Furthermore, when the students paid attention to the emerging task-at-hand, they were able to gain authority and agency for designing attention (to their texts by an audience) through influential performances of literacies. As found in this study, these patterns were not a solid package of cultural norms. Rather, the style emerged with the text and transformed with the different multiliteracies required during composition processes as students performed literacy knowledge. This study initiated an examination of influential literacy performances as the use of creative tactics during collaborative composition processes. I recommend further work examining multiliteracies as knowledge performances in a variety of settings in order to develop models to help students influence texts with their creative ideas and gain authority in collaborative groups.

The increasingly personal and ubiquitous capabilities of computing—everything from smartphones to virtual reality—are enabling us to build a brave new world in the digital realm. Despite these advances in the virtual world, our ability as end-users to transform the physical world still remains limited. The emergence of low-cost fabrication technology (most notably 3D printing) has brought us a dawn of making, promising to empower everyday users with the ability to fabricate physical objects of their own design. However, the technology itself is oblivious of the physical world—things are, in most cases, assumed to be printed from scratch in isolation from the real world objects they will be attached to and work with. To bridge this ‘gulf of fabrication’, my thesis research focuses on developing fabrication techniques with design tool integration to enable users to expressively create designs that can be attached to and function with existing real-world objects. Specifically, my work explores techniques that leverage the 3D printing process to create attachments directly over, onto and around existing objects; a design tool further enables people to specify and generate adaptations that can be attached to and mechanically transform existing objects in user-customized ways; a user-driven approach allows people to express and iterate structures that are optimized to support existing objects; finally, a library of ‘embeddables’ demonstrate that existing objects can also augment 3D printed designs by embedding a large variety of material to realize different properties and functionalities. Overall my thesis aspires to make fabrication real—enabling people to express, iterate and fabricate their designs that closely work with real-world objects to augment one another.

Abstract:
The proposition of a raw water quality index applying to conventional water treatment plants (RWQI) will allow evaluating the efficiency of the plant, not only concerning the water quality for public supply according to drinking water standard. Consequently, the natural water that presents high values of RWQI and WTPQI could not have the same efficiency of waters with low RWQI, meaning waters with lower quality with the same WTPQI value. The main objective of the proposition is to show the utility of this rate as a tool for systems of operation and production of potable water and for public information of the water quality as well. This research was made according the Delphi methodology, with help of specialists in the definition of weights and in the choice of quality parameters for the raw water to be used in theproposition of this index. Two questionnaires was sent to specialists. The first had the answers of weight and parameters, and the second had the feedback of the answers of the first one. Then the parameters scoring was structured, after the most relevant weights and parameters were determined according with the specialists opinion. This scoring was structured considering the impact occasioned in water treatment by the variation in parameters concentration. The optimum values of a parameter must have higher values in the parameter scoring, as low values must have lower values. The parameters scoring were determined after consulting bibliography, legislation and inquiring operators and researchers about the process of water supply potabilization. The last stage of this work was a mathematic formulation of the productory and somatory for aggregation of all variables to propose the RWQI and utilization of the rate in a conventional water treatment station. The productory was used to verify the influence of lower scoring parameters in rate values. The (RWQI) is an important information tool for the quality water sources like common stage that evaluates the performance from water treatment plants.
A proposição de um índice de qualidade da água bruta afluente a estações de tratamento de água convenciais (IQAB) permitirá avaliar a eficiência do tratamento de água em função não somente do cumprimento dos padrões de qualidade de água para abastecimento público ditados pela Portaria 518/04 (MINISTÉRIO DA SAÚDE, 2004) e do Índice de Qualidade de Estações de Tratamento de Água (IQETA), mas também em função das características da água bruta afluente a estação de tratamento. O principal objetivo da proposição deste índice é demonstrar sua utilidade como ferramenta capaz de auxiliar em sistemas de operação e produção de água potável, bem como para objeto de informação pública da qualidade da água em mananciais de abastecimento. O desenvolvimento da pesquisa foi feito através da metodologia Delphi com a opinião de especialistas para a definição dos pesos e escolha dos parâmetros de qualidade da água bruta a serem abordados para a proposição do índice. Foram enviados aos participantes dois questionários de pesquisa de opinião, o último contendo o feedback das respostas do primeiro para verificação dos pesos e parâmetros, visando um consenso nas opiniões. Após a definição dos pesos e dos parâmetros mais relevantes, foram desenvolvidas pontuações dos parâmetros, levando em consideração o impacto produzido no tratamento de água pelas variações nos valores do parâmetro. A pontuação dos parâmetros foi determinada através de consultas à literatura, questionamentos a operadores e pesquisadores sobre a potabilização das águas de abastecimento, bem como a legislação pertinente. A última etapa do trabalho foi a formulação matemática através de um produtório e somatório para agregação de todas as variáveis para a proposição do IQAB e aplicação do índice para uma estação de tratamento convencional. Por fim, a utilização do produtório foi aplicada para a observação da relevância dos parâmetros de pontuações mais baixas na interferência de valores do índice. O IQAB é uma importante ferramenta de informação da qualidade da água de mananciais como etapa rotineira na performance de sistemas de abastecimento.

Abstract:
Piano keyboard visualization was very popular right now, but there are very few virtual piano keyboard visualizations right now [1]. I was using unity to show the virtual piano keyboard and then they can play piano pieces by themselves or play a recording online [2]. After that you can listen and see how the recording pieces play it on the visual keyboard to give them a clear idea about how the songs played on a keyboard2 [3]. For those who played by themselves it can let them heard and know also when the visual piano play for them, they can tell if they have offbeat playing or they missing not. Piano4Play is an automated piano transcription and keyboard visualization system using AI and deep learning techniques. The user could upload a recorded piece of music, and our app would visualize the music on a digital piano keyboard. The user could see how the music is played visually in order to help piano beginners to see how the music will be played on piano in order to help them learn more quickly and easier, and advanced players could use the app to see whether they made any mistake when they are playing so they can get some improvement. Our app uses wav and MIDI files, repl, real-time database, google Collab and Unity.
