Abstract:

The retailers’ profitability and the consumers’ satisfaction depend on finding the optimal location for a retail store. When considering the stores’ spatial distribution, business potential can be understood and a squandering planning of resources can be avoided. In this paper we identify the spatial patterns of retail stores located in the traditional commercial centers of twelve large -and medium-sized Greek cities, aiming to explain why such patterns exist. The type of retail activities was determined using the image of the ground-floor stores provided by the Google Street View (GSV) service and thus 7322 stores were recorded in a geodatabase as point features. The results reveal that the retail stores’ distribution has a clustered and random spatial pattern at least in one city, where the high population density and the increase in rental prices of premises for professional activities constitute the factors that form these spatial patterns respectively.

Abstract:

Purpose This study aims to measure the association between local retail grocery provision and private residential rental prices in England. Renting is an important sector of the housing market in England and local grocery provision is an important aspect of service provision and consumers are known to be highly sensitive to the branding of this type of retailing. Design/methodology/approach This research uses a novel data source from a property rental Web platform to estimate a hedonic model for the rental market. These models incorporate information on the nature of the properties and their neighbourhoods, with an emphasis on how different retail brands are associated with rental prices. This retail brand is captured on two scales: the provision of local branded convenience stores and the provision of larger stores. Findings The study finds clear differentials in how the local grocery brand is associated with rental prices. When controlling for commonly explored confounding factors, “Luxury” retailers such as Waitrose and Marks and Spencer are associated with higher rental prices, while “Discounter” retailers are associated with lower rental prices. This finding has many implications, particularly in relation to potential price changes in an already challenging housing market for many people. Research limitations/implications This is an observational study and as such only associations (not causation) can be implied by these findings. Originality/value The focus of this research is on the private residential property market, an important market in England but one that has enjoyed less scrutiny than the sales or socially rented markets. Rather than using general accessibility to retail, this research has differentiated the association by the retail brand and store size, two very important aspects of consumer choice.

Abstract:

Managing inventory in a multi-level supply chain structure is a difficult task for big retail stores as it is particularly complex to predict demand for the majority of the items. This paper aims to highlight the potential of machine learning approaches as effective forecasting methods for predicting customer demand at the first level of organization of a supply chain where products are presented and sold to customers. For this purpose, we utilize Artificial Neural Networks (ANNs) trained with an effective second order algorithm, and Support Vector Machines (SVMs) for regression. We evaluated the effectiveness of the proposed approach using public data from the Netflix movie rental online DVD store in order to predict the demand for movie rentals during an especially critical for sales season, which is the Christmas holiday season. In our analysis we also integrated data from two other sources of information, namely an aggregator for movie reviews (Rotten Tomatoes), and a movie oriented social network (Flixster). Consequently, the approach presented in this paper combines the integration of data from various sources of information and the power of advanced machine learning algorithms for lowering the uncertainty barrier in forecasting supply chain demand.

Feminists have been concerned about the debasement of women in sexually explicit material. The purpose of this study was to determine the extent of domination and sexual inequality in x-rated videocassettes through a content analysis of 45 widely available x-rated videocassettes. The sample was randomly drawn from a list of 121 adult movie titles widely available in family videocassette rental stores in southern California. Over half of the explicitly sexual scenes were coded as predominantly concerned with domination or exploitation. Most of the domination and exploitation was directed by men toward women. Specific indicators of domination and sexual inequality, including physical violence, occurred frequently. The growth of the videocassette rental industry and the popularity of x-rated films, coupled with the messages these films convey, is a cause for concern.

The tenant mix layout of shopping malls affects shopper consumption behaviour and the performance of malls. The main function of the tenant mix layout is to increase store sales by increasing footfall. However, although existing studies have shown the importance of the spatial clustering effect and the physical information about tenants, the authors of those studies did not properly consider both the spatial clustering effect and the physical information about tenants at the meantime. Through this study, we aimed to maximize the spillover effect of the stores in the shopping centre while considering both the spatial clustering effect and physical information about tenants. Therefore, we present a problem called the tenant mix problem, which is to determine the optimal tenant configuration scheme for existing shopping centre space segmentation to maximize the rental income of a shopping centre. To solve this problem, a nonlinear integer optimization model with defined characteristics was proposed and solved using a genetic algorithm. A shopping centre case study is also presented to verify the performance of the model.

Currently, all major IT and telecom firms are busy trying to stimulate non-contractual complementary developments around their own core competences and offerings. But little has been done to explain the logic, strengths, and weaknesses of non-contractual innovation. The literature on open-platform leadership recognises the importance of non-contractual innovation, but only within the limited confines of a normative approach based on two implicit assumptions: that a platform's core and periphery are sharply and easily differentiated and that platforms are always grown and orchestrated from a monolithic core. Through analysis of two cases of decentralised open innovation: the emergence of video rental stores and the emergence of desktop-publishing systems. I argue that these assumptions do not apply to all open platforms. I conclude that by forcing a hierarchical framework onto the analysis, the normative approach underplays the role of non-contractual innovation and turns a blind eye to the radically self-organised and unforeseeable nature of some platforms' success.

The human needs for livelihood have become important part as an effort to maintain its survival. Therefore, one of the job opportunities that can be done is to work in the informal sector. The informal sector also appears in public spaces, the example is at low-cost apartment building (rusunawa), such as food stalls and grocery stores in corridors, playgrounds or parking areas of rusunawa. This study looks at the development of the function of the rusunawa as a residence into an informal economic space in the Rusunawa in the City of Surabaya. This study used a qualitative descriptive method, by connecting the correlation between the public space and the informal economy space (logic model analysis and cross-case synthesis). So that it can provide knowledge about the planning and development of rusunawa in the future, as policy guidelines regarding the planning and development of rusunawa. <p class="JGI-AbstractIsi">This study is conducted to see the potential demand with the characteristics of the campus environment, industrial environment, and tourism environment on internet fixed broadband use, where the objective of this survey is limited to households, individuals and businesses. Analysis is carried out in a descriptive manner based on the results of qualitative discussions by experts which is reinforced by the findings of the survey results. The survey results show that the internet utilization by the public (households, businesses) is mostly for communication and entertainment, so that the cellular internet is considered adequate for public's internet need. This is relevant to the public’s opinion that 87.9% of the internet needs are met when using cellular phone. There are 3 (three) perceptible reasons of why respondents choose not to subscribe to Fixed broadband internet, namely: 1) There is no necessity for subscription, 2) Lack of Knowledge/Information related to Broadband Fixed internet, and 3) Expensive Prices. The potential demand for each characteristic is always there, however, the most dominant one comes from the businesses, such as cafes/eateries, food stores, gift shops, photocopying businesses, vehicle rental services, travel businesses, etc.<strong></strong></p>

Purpose Relying on the theories of substitutes for leadership and psychological empowerment, this study aims to explore how perceptions of customer positive feedback can substitute for managers’ transformational leadership in driving frontline employees’ psychological empowerment and, in turn, task performance. Design/methodology/approach The authors tested the research hypotheses with frontline employees working in 17 equipment rental stores. Employees completed a questionnaire about customer positive feedback, transformational leadership and psychological empowerment, and supervisors completed a separate questionnaire about employees’ task performance. A total of 178 employee-supervisor dyads formed the final sample of the study. Findings The results provided support for our hypotheses. Psychological empowerment fully mediated the relationship between transformational leadership and task performance. Moreover, customer positive feedback moderated the indirect relationship between transformational leadership and task performance such that it was significant and positive only when customer feedback was low. Originality/value This paper contributes to the service marketing literature by showing that customer positive feedback can substitute for managers’ leadership in helping frontline employees feeling more in control of their work and psychologically empowered. Another useful contribution for practitioners is that customers may have a positive impact on frontline employees’ motivation state, which past research has little explored.

Abstract:

The objective of the project was to encourage health promotion through education, outreach, and community-based training. The people attending health fairs (n = 467), community events (n = 469), and Kindergarten classes (n = 241) were the study participants. Hands-on training was offered at homebuilding retail stores (n = 25). U.S. Department of Housing and Urban Development (HUD)’s online visual training was given to realtors (n = 220), and inspectors, contractors, and Do-It-Yourself (DIY) workers (n = 75). Training workshops were attended by home-buyers and rental home owners at the Neighborhood Association Meetings (n = 91). The impact of training was evaluated by pre- and posttests. Nearly, 90% of the participants (n = 25) reported the hands-on training was useful. At posttest after the HUD online training, 59.4%, 67.9%, 65.1% of the participants (n = 220) identified soil, car batteries, and paint as sources of lead in the environment, respectively. Nearly 70% identified lead as a poison in the environment while 77.5% and 47.2% demonstrated two behaviors which help prevent lead poisoning. A total of 62.3%, 48.1%, and 58.5%, at posttest identified three complications or illnesses—behavioral, physical, and psychological, respectively. The home owners are required to get permission from the City for housing repair. In coordination with the federally funded housing repair or lead abatement programs, the trained inspectors are authorized to certify the renovation or repair works. These outreach activities were successful in improving the knowledge of the community people on lead poisoning prevention.

Metrics to quantify environmental factors including classification of urban or rural county status; the number of SNAP-authorized stores, food pantries and recreational facilities; average fair market housing rental price; and natural amenity rank were collected from government websites and data sets covering the years 2012–2016 and used as covariates in mixed multiple linear regression modelling.SettingThirty-seven Indiana counties, USA, 2012–2016.SubjectsSNAP-Ed eligible adults from households with children (n328).ResultsNone of the environmental factors investigated were significantly associated with changes in household food security in this exploratory study.ConclusionsSNAP-Ed improves food security regardless of urban or rural location or the environmental factors investigated. Expansion of SNAP-Ed in rural areas may support food access among the low-income population and reduce the prevalence of food insecurity in rural compared with urban areas. Further investigation into policy, systems and environmental factors of the Social Ecological Model are warranted to better understand their relationship with direct SNAP-Ed and their impact on diet-related behaviours and food security.

CONTEXT AND OBJECTIVES: Themes like alcohol and drug abuse, relationship difficulties, psychoses, autism and personality dissociation disorders have been widely used in films. Psychiatry and psychiatric conditions in various cultural settings are increasingly taught using films. Many articles on cinema and psychiatry have been published but none have presented any methodology on how to select material. Here, the authors look at the portrayal of abusive use of alcohol and drugs during the Brazilian cinema revival period (1994 to 2008). DESIGN AND SETTING: Qualitative study at two universities in the state of São Paulo. METHODS: Scenes were selected from films available at rental stores and were analyzed using a specifically designed protocol. We assessed how realistic these scenes were and their applicability for teaching. One author selected 70 scenes from 50 films (graded for realism and teaching applicability > 8). These were then rated by another two judges. Rating differences among the three judges were assessed using nonparametric tests (P < 0.001). Scenes with high scores (> 8) were defined as "quality scenes". RESULTS: Thirty-nine scenes from 27 films were identified as "quality scenes". Alcohol, cannabis, cocaine, hallucinogens and inhalants were included in these. Signs and symptoms of intoxication, abusive/harmful use and dependence were shown. CONCLUSIONS: We have produced rich teaching material for discussing psychopathology relating to alcohol and drug use that can be used both at undergraduate and at postgraduate level. Moreover, it could be seen that certain drug use behavioral patterns are deeply rooted in some Brazilian films and groups.

Background and objectives: Lead poisoning is a preventable environmental health hazard. Although the prevalence of lead poisoning is declining, the rates are disproportionately high in selected communities. This community-based participatory research (CBPR) program aimed to enhance people’s awareness on lead poisoning prevention through community outreach and educational interventions in Mississippi. Methods: Secondary data of 42,372 children obtained from the Mississippi State Department of Health were analyzed to identify the most affected communities in Mississippi. Community-based outreach and education activities were carried out in the most affected areas to increase population awareness on lead poisonig prevention. Results: Hands-on training was offered to 25 participants at homebuilding retail stores. Of them, 23 (92%) reported the hands-on training was very useful or useful. Among 91 home-buyers and rental home owners who attended workshops offered by the Neighborhood Association, 90% mentioned that the training was useful or very useful. An online visual training was given to 220 realtors, and 75 inspectors, contractors, and Do-It-Yourself (DIY) workers. At posttest, 59.4%, 67.9%, 65.1% of the realtors, inspectors, contractors and DIY workers (n = 295) identified soil, car batteries and paint as sources of lead in the environment, respectively. A total of 62.3%, 48.1% and 58.5%, at posttest, identified three complications - behavioral, physical and psychological, respectively. The mean posttest score was significantly higher than the pretest scores (7.47 ± 2.07 vs. 6.60 ± 1.68, p = 0.04, respectively). Abstract:

The COVID-19 pandemic seriously impacted almost the fields of the Vietnam economy in the shortterm and long-term, including the retail industry. While, retailing is regarded as one of the most strongly-impacted fields, but it also expresses the quick capability of recovery in comparison to other sectors. This study has been carried out to evaluate and analyze holistically the major effects of the pandemic on the Vietnam retail industry with nine critical impacts: the number of shoppers decreases; great volatility in the rental market; closed shops, maximum staff cuts; retail sales decline seriously; the explosion of e-commerce and home delivery services; change the campaign and decrease marketing budget; invest in building new online sales channels; convert consumption from offline to online and increase stockpiles of goods in the epidemic. Based on these analyses, the author recommends several significant suggestions for the Vietnam government including (1) directly support by in cash for the closed-stores during the pandemic for the traditional retailing; (2) quickly implement liquidity support packages and offer credit packages with preferential interest rates for the modern retailers; (3) solving problems of labor shortages in businesses that produce essential products; (4) increasing the essential stockpile source and manage efficiently the market supply. While some suggestions are delivered for retail businesses to overcome the recession and boost growth after the pandemic: (1) understand customers' problems; (2) reshape business strategy; (3) keep in touch with stakeholders and (4) expand new sales channels. The above suggestions are recommended on the basis of the bright outlooks from both macro factors and the internal retailing market.

In relation to the introduction of quarantine restrictions and an emergency situation to counteract COVID-19, there are potential prospects for the development of online retail in Ukraine. The aim of the article is to study the main problems, disadvantages and advantages of e-commerce and highlight further trends in retail development through the use of Internet resources. The results of the analysis. The development of e-commerce is based on attracting more and more users to the Internet, so it is not surprising that the part of turnover from e-commerce in GDP and the part of e-component in retail correlates with the Internet penetration rate, the part of the population aged 14 to 74 that are regular Internet users. Thus, in the leading countries of Western Europe, this figure is approaching 100%, while in Ukraine it is only about 63%. In the course of an investigation we identified the role and positive impact of domestic marketplaces on the development of E-commerce in various segments of the economy. Marketplaces, which are a kind of platform for uniting different sellers and buyers by interests, give impetus to the growth of positive dynamics of supply and demand, sales of goods and services, in fact, without leaving home - for consumers, and without spending significant rental costs offline stores - for sellers. The positive gradation of e-commerce growth has been affected by quarantine restrictions due to the COVID-19 pandemic. Thus, the need for self-isolation and social distancing, temporary suspension of offline shops of non-food products and establishments in various fields of services (entertainment centers, catering establishments) on the basis of government requirements, resulted not only in increasing the popularity of existing marketplaces and social networks, but forced many retailers to start or develop existing online stores alongside traditional sale outlets. The advantages of e-commerce are determined: speed and availability of obtaining information about the product, its price, characteristics; efficiency and convenience of trade operations; reduction of operating costs; ensuring long-term communications with customers; development of Internet marketing through a possible variety of tools used; the possibility of using modern analytical tools to study the market and evaluate the effectiveness of marketing activities; increasing openness to consumers and increasing their loyalty. The main disadvantages of E-commerce are highlighted as insufficient security of consumer data and unreliable payment systems; quality of communication and service; instability of the national currency; imperfect legal framework; high delivery prices. All these problems have their basis and depend on many factors, first of all - on the level of sustainable development of the economy as a whole. Conclusions and direction for further research. Despite of the challenges of the time, the urgent needs of consumers in goods and services, e-commerce currently has a positive development trend and considerable prospects for further expansion of the market. The development of e-commerce should take place with the simultaneous simplification and regulation of ways to consolidate legal relations of contractors, methods and models of promotion of goods and services over the Internet, which clearly requires additional research. The past few decades have introduced us to the Digital Natives, a generation born in the 1980s, who have been familiar with Information and Communication Technology (ICT) and the myriads of ICT products since the day they were born. The last decade has also made us familiar with numerous disruptive innovations and technologies that have now been so pervasive in our daily lives and have ‘threatened’ many established practices on how we conduct our lives and businesses, such as ride-sharing (Uber, Grab, etc.), accommodation-sharing (AirBnb), and the blockchain technology that has been applied in more and more aspects of life, with Bitcoin as one of its applications in the crypto currency sector. Those changes and disruptions have not spared the higher education (HE) sector. Nowadays HE institutions need to take into account the characteristics of the Digital Natives, and various disruptive innovations and technologies if they want to remain relevant and stay ahead of the competition in the sector. All aspects of HE – including academic libraries, are not immune to these changes and disruptions. In the past, academic libraries have tended to play their roles as the storehouse of information and the provider of space for individual learning. The advent of the Digital Natives and disruptive technologies have led some in the HE sector to argue that academic libraries would soon lose their relevance since nowadays information can be accessed digitally from anywhere and at any time. People who believe this line of thinking have given some examples. One of them is the demise of Blockbuster (movie rental) stores in the United States (US) with the emergence of online video/movie streaming services. However, many forget that libraries, including academic libraries, also have their social functions besides their traditional roles as an information storehouse and individual learning space. Due to their social functions, libraries have even been viewed as one of the most democratic spaces that societies have. This is especially true in the case of public libraries, but also – to some extent – true for academic libraries, where they also perform their functions as community hubs.

Abstract:

Tissue nitric oxide (NO) levels increase dramatically during ischemia, an effect that has been shown to be partially independent from NO synthases. Because NO is stored in tissues as S-nitrosothiols and because these compounds could release NO during ischemia, we evaluated the effects of buthionine sulfoximine (BSO; an intracellular glutathione depletor), light stimulation (which releases NO, decomposing S-nitrosothiols), and N-acetyl-l-cysteine (a sulfhydryl group donor that repletes S-nitrosothiols stores) on the changes in outer medullary NO concentration produced during 45 min of renal artery occlusion in anesthetized rats. Renal ischemia increased renal tissue NO concentration (+223%), and this effect was maintained along 45 min of renal arterial blockade. After reperfusion, NO concentration fell below preischemic values and remained stable for the remainder of the experiment. Pretreatment with 10 mg/kg nitro-l-arginine methyl ester (l-NAME) decreased significantly basal NO concentration before ischemia, but it did not modify the rise in NO levels observed during ischemia. In rats pretreated with 4 mmol/kg BSO and l-NAME, ischemia was followed by a transient increase in renal NO concentration that fell to preischemic values 20 min before reperfusion. A similar response was observed when the kidney was illuminated 40 min before the ischemia. The coadministration of 10 mg/kg iv N-acetyl-l-cysteine with BSO + l-NAME restored the increase in NO levels observed during renal ischemia and prevented the depletion of renal thiol groups. These results demonstrate that the increase in renal NO concentration observed during ischemia originates from thiol-dependent tissue stores.

On the basis of evidence that changes in the extracellular concentration of calcium effectively modulate renin secretion from renal juxtaglomerular cells, our study aimed to determine the effect of calcium influx activated by depletion of intracellular calcium stores on renin secretion. For this purpose we characterized the effects of the endoplasmatic Ca2+-ATPase inhibitors thapsigargin (300 nM) and cyclopiazonic acid (20 μM) on renin secretion from isolated perfused rat kidneys. We found that Ca2+-ATPase inhibition caused a potent inhibition of basal renin secretion as well as renin secretion activated by isoproterenol, bumetanide, and by a fall in the renal perfusion pressure. The inhibitory effect of Ca2+-ATPase inhibition on renin secretion was reversed within seconds by lowering of the extracellular calcium concentration into the submicromolar range but was not affected by lanthanum, gadolinium, flufenamic acid, or amlodipine. These data suggest that calcium influx triggered by release of calcium from internal stores is a powerful mechanism to inhibit renin secretion from juxtaglomerular cells. The store-triggered calcium influx pathway in juxtaglomerular cells is apparently not sensitive to classic blockers of the capacitative calcium entry pathway.

Brown adipose tissue (BAT) is a specialised fat store that is metabolised by the newborn lamb to ensure effective adaptation to the cold challenge of the extra-uterine environment. Increasing BAT reserves therefore has the potential to increase neonatal thermogenesis and survival. It is established that arginine supplementation can increase fetal BAT stores but the biological mechanisms involved are unclear. The objective of this study was to test the hypothesis that increased fetal BAT stores resulting from maternal arginine supplementation is mediated by activation of the thermogenic program. Brown adipose tissue was collected from fetuses of ewes supplemented with arginine from 100 to 140 days of gestation. Increased peri-renal fat stores in fetuses from arginine-supplemented ewes was associated with an increase in uncoupling protein 1 (UCP-1) and PRD1-BF-1-RIZ1 homologous domain containing protein-16 expression, but not proliferator-activated receptor gamma or proliferator-activated receptor gamma-co-activator-1α in BAT. The activity of UCP-1 is regulated by hormones including cortisol and thyroid hormones. Cortisol level in fetuses from supplemented sheep was 68% greater than those from control ewes, indicating that cortisol may control upregulation of UCP-1 expression in the ovine neonate. The DNA and RNA concentration in BAT of both groups suggest that increased peri-renal fat stores is not associated with an increase in cell number or number of ribosomes, but rather an increase in the size of individual fat cells. Collectively, these results indicate that maternal arginine supplementation during mid to late gestation improved the thermoregulatory ability of lambs and this could potentially increase their survival in early life.

Diagnosis and treatment of renal stones during pregnancy is a complex problem. Risks to the fetus from ionising radiation and interventional procedures need to be balanced with optimising clinical care for the mother. Management of such patients requires a clear understanding of available options, with a multidisciplinary team approach. In this review, we discuss the role of different diagnostic tests including ultrasound, magnetic resonance urography, and computerized tomography. We also provide an update on recent developments in the treatment of renal stones during pregnancy. Expectant management remains first-line treatment. Where definitive treatment of the stone is required, new evidence suggests that ureteroscopic stone removal may be equally safe, and possibly better than traditional temporising procedures.

Abstract:

The regulation of film exhibition in Iceland has closely shadowed the history of cinema exhibition itself. Although regulation practices have undergone various shifts and realignments throughout the twentieth century, they retained certain core concerns and a basic ideological imperative having to do with child protection and child welfare. Movies were thought to have a disproportionate impact on children, with „impressionable minds“ often being invoked. Their interior lives and successful journey towards maturity were put at risk each and every time they encountered unsuitable filmic materials. Thus, while assuming that adults could fend for them-selves among the limited number of theaters in Reykjavík, children were a whole another matter and required protection. Civic bodies were consequently formed and empowered to evaluate and regulate films. But even in the context of fairly rigorous surveillance and codification, the turn taken by regulatory authorities in the 1980s strikes one as exceptional and unprecedented. The Film Certification Board (TFCB) was, for the first time, authorized to prohibit and suppress from distribution films deemed especially malignant and harmful. Motivating this vast expansion of the powers of the regulatory body were concerns about a variety of exploitation and horror films that were being distributed on video, films that were thought to transgress so erroneously in terms of on-screen violence that their mere existence posed a grave threat to children. Two years after finding its role so radical-ly enlarged, TFCB put together a list of 67 „video-nasties“, to borrow a term from the very similar but later moral panic that occurred in Britain. Police raids were conducted and every video store in the country was visited in a nation-wide effort to remove the now illegal films from rental stores. This article posits that the icelandic nasties list can be viewed as something of a unique testament to the extent to which the meaning, aesthetic coherence and the affect of cultural objects is constructed in the process of reception, while also main-taining that the process of reception is thoroughly shaped by historical discourses, social class, embedded moral codes and a social system of values, as well as techno-logical progress. in what amounts to a perfect storm of moralizing, political games-manship and the sheer panic of a certain segment of the population, the governing institutions in iceland managed in the span of months to overturn constitutionally protected rights to free speech and privacy, as well as undermine central principles of the republic. Two decades would pass before these setbacks were recuperated, and then only on a legal and institutional level. While analyzing the history of the icelandic video nasties, the article also attempts to grapple with and articulate the symbolic register of the ban, how it speaks to the status of cinema in Iceland at the close of the twentieth century, and what ideological strains, morals and/or values were being put into play and funneled into this particular debate. Then, to close, the role of the most notorious of the nasties, Cannibal Holocaust (Ruggero Deodato, 1980), is examined in the context of media coverage and parliamentary debates at the time.

Experiments addressed the hypothesis that afferent and efferent arterioles differentially rely on Ca2+ influx and/or release from intracellular stores in generating contractile responses to AVP. The effect of Ca2+ store depletion or voltage-gated Ca2+ channel (VGCC) blockade on contractile responsiveness to AVP (0.01–1.0 nM) was assessed in blood-perfused juxtamedullary nephrons from rat kidney. Depletion of intracellular Ca2+ stores by 100 μM cyclopiazonic acid (CPA) or 1 μM thapsigargin treatment increased afferent arteriolar baseline diameter by 14 and 21%, respectively, but did not significantly alter efferent arteriolar diameter. CPA attenuated the contractile response to 1.0 nM AVP by 34 and 55% in afferent and efferent arterioles, respectively ( P = 0.013). The impact of thapsigargin on AVP-induced afferent arteriolar contraction (52% inhibition) was also less than its effect on the efferent arteriolar response (88% inhibition; P = 0.046). In experiments probing the role of the Ca2+ influx through VGCCs, 10 μM diltiazem evoked a 34% increase in baseline afferent arteriolar diameter and attenuated the contractile response to 1.0 nM AVP by 45%, without significantly altering efferent arteriolar baseline diameter or responsiveness to AVP. Combined treatment with both diltiazem and thapsigargin prevented AVP-induced contraction of both vascular segments. We conclude that Ca2+ release from the intracellular stores contributes to the contractile response to AVP in both afferent and efferent arterioles but is more prominent in the efferent arteriole. Moreover, the VGCC contribution to AVP-induced renal arteriolar contraction resides primarily in the afferent arteriole.

The early distal tubule (EDT) of the frog nephron, similar to the thick ascending limb in mammals, mediates the transepithelial absorption of NaCl. The continued absorption of NaCl in the face of varying Na+ load is maintained by coordination of the activity of ion-transporting proteins in the apical and basolateral membranes, so-called pump-leak coupling. Previous studies identified intracellular Ca2+, originating from an intracellular Ca2+ store, as playing a key role in pump-leak coupling in the EDT (Cooper GJ, Fowler MR, and Hunter M. Pflügers Arch 442: 243–247, 2001). The purpose of the experiments described in this paper was to identify the intracellular Ca2+ storage pools in the renal diluting segment. Store Ca2+ movements were monitored by the fluorescence of mag-fura 2 in permeabilized segments of frog EDTs. The presence of both ATP and Ca2+ was required to maintain store Ca2+ content. Removal of either of these substrates resulted in a passive leak of Ca2+ from the stores. The uptake of Ca2+ into the store was sensitive to the SERCA inhibitor 2,5-di( tert-butyl) hydroquinone, whereas Ca2+ release from the store was stimulated by IP3 but not cADPR. Store Ca2+ was insensitive to the mitochondrial ATP synthase inhibitor oligomycin, and, under conditions that energized Δψm, the complex 1 inhibitor rotenone and the protonophore FCCP. Ionomycin was able to mobilize store Ca2+ following exposure to IP3. These results suggest that the endoplasmic reticulum is a dominant Ca2+ store in the frog EDT. A second pool, sensitive to ionomycin but not IP3, may overlap with the IP3-sensitve pool. The data also rule out any contribution by mitochondria to EDT Ca2+ cycling.

Abstract. Early diabetic nephropathy exhibits renal glomerular hyperfiltration and an increase in renal plasma flow. The hyperfiltration is a dysfunctional state that may arise from a hyperglycemic-induced hypocontractility of glomerular mesangial cells that may be associated with depressed Ca2+signaling events. The present study was designed to determine the effects of acute (minutes) and chronic (days) elevated glucose levels on endothelin-induced calcium signaling with a particular emphasis on the potential influence on stores and store-operated Ca2+influx (SOCI ; also called capacitative calcium entry) in glomerular mesangial cells. Primary cultures of rat mesangial cells were grown in either high (30 mM) or normal (5 mM) glucose-containing media and tested in the presence of either high (30 mM) or normal (5 mM) glucose levels. Intracellular calcium levels were monitored with the calcium-sensitive fluorophore fura-2 before and after treatment with either endothelin-1 (10 nM), to induce typical Ca2+signals, or the endoplasmic reticulum (ER) Ca-ATPase inhibitor thapsagargin (1 μM), to unload ER Ca2+stores. Both acute and chronic exposure to high glucose levels depressed the endothelin-induced calcium signal. However, neither release of Ca2+from stores nor SOCI were depressed by high glucose levels. In contrast, an endothelin-induced calcium entry pathway (likely receptor-operated calcium influx), separate from SOCI, was markedly depressed in the presence of both acute and chronic high glucose levels. The depressant effect of high glucose was rapidly (minutes) reversible upon returning to normal glucose levels. It is concluded that high glucose levels depress endothelin-induced calcium signaling in rat mesangial cells by inhibiting non-SOCI Ca2+entry pathways, namely the receptor-operated Ca2+influx pathway. The glucose-induced alterations in the receptor-operated calcium influx pathway may, in part, contribute to the depressed contractile state of glomerular cells during periods of hyperglycemia.
