Fluorescence microscopy is a tool of primary interest in biomedical research as it allows to selectively visualize particle dynamics within the cell. Hence, there is a high demand for algorithmic tools capable of automatically analyzing raw microscopic data. After a presentation of the theory and techniques surrounding particle dynamics, fluorescence microscopy, tracking methods and motion classification, we present in this thesis a new mapping method based on spatiotemporal kernel estimators that robustly estimate intracellular diffusion and drift from tracking data. We evaluate it in an extensive set of experiments using simulated, real, 2D and 3D data and show that our method provides precise and accurate diffusion and drift maps while outperforming existing methods. As such, it allows biologists to study intracellular dynamics of specifically tagged particles with a wider range of acquisitions and fluorescence microscopy techniques. The laser shock wave generation is a novel process becoming more and more common. The shock waves are used to generate mechanicals effects in the sample. The laser absorption results in the creation of a plasma at the surface. This plasma during its expansion creates a shock wave propagating through the sample. This work aims to study the various phenomena involved in the laser-matter interaction. In the field of laser generated shock waves, two different processes exist: the Laser Shock Peening (LSP) and the LASer Adhesion Technique (LASAT). The new challenge deals with the limitations of those processes and the solutions to be setting up to improve them. Some ideas concerning the confinement improvement as water substitution or thermal coatings optimization are suggested in this work. Moreover, the use of double delayed laser pulses allows, for LASAT, the location of main tensile stresses near interfaces. However, for LSP, some aspects dealing with the profitability linked to the peening rate are investigated here. Last but not least, whether the very short (0 to 1000 ns) or very long delays (from 200µs to 200ms), the study of the laser-matter interaction phenomena allows to overcome some limitations for both laser shock processes

Liquid-gas and liquid-vapour two-phase flows are encountered in a large range of industrial applications. They are observed in multiple configurations, in terms of dimension and orientation and then have various characteristics. In the literature, it can be distinguished two limit cases of the two-phase flow linked to its confinement: micro and macro-scale. Nonetheless, characterisation of the micro-to-macro scale transition is still a main issue for the comprehension of two-phase flow. In addition, thermal behaviours of flow boiling are affected by the nucleation phenomenon, which is strongly influenced by the geometry of the application and the saturation conditions. The main objective of the thesis is to contribute to the comprehension of adiabatic and diabatic two-phase flows in mini-scale, where the general behaviours are not well definite. In the present study, the effects of confinement and orientation of two-phase flows on flow patterns, pressure drops and heat exchanges are studied and compared with observations and models of the literature. An 1.6 mm inner diameter inclinable test section was installed on the test bench of Charnay (2014), which enabled to study R-245fa flow boiling. This test section enables to visualize the flow directly in the evaporator and the simultaneous acquisition of the pressure drops and the outside wall temperature field. A sapphire tube, heated by a transparent ITO coating, is unemployed to insure the transparency. The temperature field is obtained by an IR image processing. Previously, tests were led on the section of Charnay (2014) in adiabatic conditions. The present test section was used for two series of tests, led in both adiabatic and diabatic conditions. It appeared a strong effect of confinement and orientation on the flow patterns and pressure drops. The effect of the orientation is likely the same that one observed in macro-scale. In addition, the orientation slightly affects heat exchanges in comparison with the uncertainties of the IR dispositive. Finally, it is observed an effect of orientation on dryout flow pattern. In this configuration, the temperature field evolution with time is synchronized with the dynamic evolution of the two-phase flow

Afin de mieux comprendre les phénomènes physiques mis en jeu dans ce régime d'interaction particulier, une modélisation originale du procédé a été développée. Un code numérique traitant les processus de claquage dans l'eau de confinement permet dans un premier temps de déterminer les caractéristiques (intensité crête et durée à mi-hauteur) de l'impulsion laser transmise à travers la fenêtre de confinement. Un modèle hydrodynamique auto-consistant traitant les plasmas confinés (plasmas froids et denses, corrélés et partiellement dégénérés) utilise ensuite ces paramètres pour calculer les chargements mécaniques et thermiques induits à la surface de la cible traitée. Pour terminer, ces chargements sont utilisés en entrée du code aux éléments finis ABAQUS afin de simuler les contraintes résiduelles d'origine mécanique et thermique induites par le traitement.
Les résultats de ces simulations ont été validés par comparaison avec différentes mesures expérimentales réalisées pour des conditions d'irradiation laser (longueurs d'ondes de 1064 nm et de 532 nm, durées d'impulsion de 3 ns et 10 ns) typiques des conditions opératoires réelles utilisées au niveau industriel.
Ces résultats montrent que les petites taches focales permettent de limiter fortement le chauffage de la cible par le plasma confiné, et donc de s'affranchir des effets thermiques induits par le traitement. Ils ouvrent donc de nouvelles perspectives quant à la réalisation du traitement par choc laser sans utiliser de revêtement thermo-protecteur. Par ailleurs, ils permettant d'expliquer les résultats obtenus avec la configuration de traitement développée par Toshiba (très petites taches focales, haute cadence, pas de revêtement protecteur), qui demeuraient incompris jusqu'alors.
