Full textEquine infectious anemia virus (EIAV) is a non-primate lentivirus infecting equids worldwide that causes a persistent infection characterized by recurring febrile episodes. Equine infectious anemia evolves mainly to an asymptomatic stage, which is an example of immunity management of lentiviral infection. This disease remains without therapeutic solutions and in France, the law requires mandatory euthanasia to control the contaminations. In this PhD, we had two main objectives. The first was to explore antiviral treatments to offer other solutions to prevent the spread of the disease. We tested the efficacy of eighteen antiretroviral drugs, known to impair human immunodeficiency virus type 1 (HIV-1) replication, in an in vitro equine cell EIAV infection model. Our results identified thirteen FDA-approved drugs with a significant antiviral effect against EIAV. These compounds target viral enzymes such as the reverse transcriptase, the protease and the integrase, indicating the similarities between HIV-1 and related EIAV. The second objective was to decipher virus-host protein-protein interactions, thanks to a yeast-two hybrid screening assay. We identified fifty-one host proteins interacting with five different EIAV proteins. Interestingly, some of those interactions were already known for other lentiviruses, but for other proteins, our study is the first evidence of their interaction with a lentivirus. Some of these cellular interactors were further functionally characterized in EIAV infection set-up with gene silencing. These cellular interactors bring a better understanding of EIAV lentiviral infection cycle in the host cell. They are also possible targets for compounds, possibly impairing the infection via targeting of a host pathway rather than the virus directly. In conclusion, our results contribute to a better understanding of the lentiviral infection cycle, with EIAV as a model, and also provide therapeutic strategies against this non-treated animal disease

Full textCampylobacteriosis is the most prevalent bacterial foodborne gastroenteritis reported in the European Union and is mainly associated to consumption of poultry meat. Reducing the intestinal colonization of broilers by Campylobacter from 2 to 3 log10 CFU/g could decrease human cases incidence by 76 to 100%. Vaccination of poultry could be a potential strategy but despite many studies, no efficient vaccine is available yet. The aim of this project was to identify new vaccine antigens against Campylobacter using the reverse vaccinology strategy and to assess their immune and protective powers against the avian intestinal colonization. Based on their sub-cellular localization, immunogenicity, B-epitopes density and their sequence conservation among C. jejuni and C. coli strains, fourteen antigens were selected. Six out of them were produced and in vivo tested according to an optimized avian vaccine protocol. Four antigens showed intestinal load decreases from 2 to 4.2 log10 CFU/g correlated with the induction of specific humoral responses. Vaccine candidates’ immunogenicity and the protective efficiency of two antigens were observed again. These first results highlight the interest and reliability of the reverse vaccinology. The assessment of these new antigens vaccine potential needs to be continued and deepened in next experiments

Full textAnimal models of osteoarthritis are useful to evaluate the potential of osteoarthritis therapeutics at the preclinical stage of development. In this thesis, the dog is used as a model of naturally-occurring (i.e. companion animal) and experimentally induced (i.e. by surgical transection of the cranial cruciate ligament) osteoarthritis. The peak of the vertically-oriented ground reaction force, which is measured during kinetic gait analysis, is proposed to be an indicator of structural and functional benefits in these models of osteoarthritis. In a canine model of naturally-occurring osteoarthritis, the threshold of the minimal detectable change in peak vertical force was determined. An improvement in the locomotor disability can now be identified according to the measurement error (noise) of the peak vertical force. This allows the identification of responders when the peak vertical force is used as an outcome measure of functional benefits. A retrospective analysis later determined that current therapeutic approaches provided a responder rate of 62.8% with an effect size of 0.7 in dogs with naturally-occurring osteoarthritis. This analysis also determined that the therapeutic response is favored in cases of severe locomotor disability. In a canine model of osteoarthritis induced by surgical transection of the cranial cruciate ligament, the peak vertical force demonstrated an inverse relationship with different types of structural changes, as evaluated upon magnetic resonance imaging. The sensitivity of the peak vertical force to detect structural benefits on the subchondral bone was also shown in this model using an antiresorptive agent (i.e. tiludronate). The experiments conducted in dogs with naturally-occurring osteoarthritis further validate findings from clinical trials in which the peak vertical force is used as an outcome measure of functional benefits. The practice of an evidence-based medicine is then expected. The experiments conducted in dogs with surgically-induced osteoarthritis support the recording of the locomotor disability, being in line with the level of the structural changes. By performing gait analysis in addition to structural evaluations, it is expected to establish the impact of structural benefits on joint discomfort This thesis suggests that a platform for preclinical investigations, which combines the canine model of osteoarthritis induced by surgical transection of the cranial cruciate ligament and a clinical trial in dogs with naturally-occurring osteoarthritis, offers the opportunity to discern structural benefits having functional impacts. A better prediction of outcomes for human clinical trials is expected by using the peak vertical force.
