Abstract:
The crux in the locally linear embedding algorithm or LLE is the selection of embedding dimensionality and neighborhood size. A method of parameters selection based on the normalized cut criterion or Ncut for classification tasks is proposed. Differing from current techniques based on the neighborhood topology preservation criterion, the proposed method capitalizes on class separability of embedding result. By taking it into consideration, the intrinsic capability of LLE can be more faithfully reflected, and hence more rational features for classification in real-life applications can be offered. The theoretical argument is supported by experimental results from synthetic and real data sets.

Solvents play an important role in crystallization, a commonly used separation and purification technique in the pharmaceutical, chemical and food industries. They affect crystal properties such as particle size distribution, morphology, polymorphism etc. and therefore have consequences for the downstream processing of the solid material. Current solvent selection techniques for solution crystallization remain ad hoc and typically do not have a theoretical underpinning. Elucidation of the interactions between solvent and solute molecules and the mechanism underlying the solvent effects on each aspect of the crystal properties would be a major aid for the rational selection of solvents and also the development of crystallization processes. In this work we studied the effect of solvent on the polymorphism and morphology of organic crystals using molecular modeling techniques. The two most important contributions of this thesis are listed below. 1. We studied the self-assembly of solute molecules in solutions prior to nucleation for two polymorphic systems, tetrolic acid and glycine, using molecular dynamics simulations. We tested the existence of a link between the structure of the clusters formed in solution and the polymorphism of the crystals. Our results show that the link hypothesis succeeds in explaining the polymorph selection of tetrolic acid from different solvents. However it fails for glycine and thus should be used with caution. 2. We designed a computer-aided rational solvent selection procedure for improving the morphology of needle-like 2,6-dihydroxybenzoic acid form 2 crystal. We also experimentally grew 2,6-dihydroxybenzoic acid form 2 crystals using the solvent mixture suggested by computer simulations, which do exhibit reduced aspect ratios. This computer-aided selection procedure can not only quickly identify an effective solvent or solvent mixture, but also provide mechanistic understandings of the underlying chemistry. It can also be extended to improve the morphology of other needle-like organic crystals easily.

Guerreiro, A. "Rational analysis of physico-chemical parameters affecting recognition properties of molecularly imprinted polymers." Thesis, Cranfield University, 2006. http://dspace.lib.cranfield.ac.uk/handle/1826/11014.

Full text
Abstract:
Molecularly imprinted polymers (MIPs) are new type of materials with tailor made molecular recognition sites. Despite their enormous potential in separations, sensing and pharmaceutical industry, MIPs have so far failed to achieve significant commercial application. This is in part due to lack of understanding of MIPs at a molecular level, strong binding site heterogeneity and poor compatibility with aqueous solvents. This work is aimed at improving overall MIP performance and quality, with special focus on the improvement of compatibility with aqueous environment. Compatibility with aqueous solvents was achieved by (i) employing a molecular modelling step prior to polynerisation, (ii) by studying the ionic properties of the polymer and (iii) by analysis of the influence of different polymerisation conditions. The study of the pressure and magnetic field effects on polymers allowed us to produce materials with increased stability, lower binding site heterogeneity and higher affinity, as compared to standard imprinted materials. To minimise site heterogeneity imprinted nanoparticles with ca. 90 kDa were produced using a living polymerisation method. The particles with essentially uniform population of binding sites were obtained by employing affinity chromatography with immobilised template. As a model system variety of drugs were imprinted with the polymers being able to discriminate between closely related structural analogues in aqueous media. This work had substantial effect on success of project Pisarro - Piezoelectric Sensing Arrays for Biomolecular Interactions and Gas Monitoring where range of imprinted polymers were designed with high affinity to drugs of abuse. Currently 1 work is in progress aimed at application of these materials in commercial sensors for forensic use.

Abstract:
This thesis is a contribution to the research area concerned with selection of smoothing parameters in the framework of nonparametric and semiparametric regression. Selection of smoothing parameters is one of the most important issues in this framework and the choice can heavily influence subsequent results. A nonparametric or semiparametric approach is often desirable when large datasets are available since this allow us to make fewer and weaker assumptions as opposed to what is needed in a parametric approach. In the first paper we consider smoothing parameter selection in nonparametric regression when the purpose is to accurately predict future or unobserved data. We study the use of accumulated prediction errors and make comparisons to leave-one-out cross-validation which is widely used by practitioners. In the second paper a general semiparametric additive model is considered and the focus is on selection of smoothing parameters when optimal estimation of some specific parameter is of interest. We introduce a double smoothing estimator of a mean squared error and propose to select smoothing parameters by minimizing this estimator. Our approach is compared with existing methods. The third paper is concerned with the selection of smoothing parameters optimal for estimating average treatment effects defined within the potential outcome framework. For this estimation problem we propose double smoothing methods similar to the method proposed in the second paper. Theoretical properties of the proposed methods are derived and comparisons with existing methods are made by simulations. In the last paper we apply our results from the third paper by using a double smoothing method for selecting smoothing parameters when estimating average treatment effects on the treated. We estimate the effect on BMI of divorcing in middle age. Rich data on socioeconomic conditions, health and lifestyle from Swedish longitudinal registers is used.

Abstract:
The monograph presents a methodology for assessing the vulnerability of landscapes to external influences. A comparative analysis of the concepts of "stability", "sensitivity", "vulnerability" in relation to natural complexes. An overview of existing methods for assessing the vulnerability of natural complexes is presented. The author's method of assessing the vulnerability of landscapes to anthropogenic impacts is described. The methodology is based on: selection and justification of criteria for assessing the vulnerability of landscapes; preparation of a parametric matrix and gradation of assessment criteria in accordance with the developed vulnerability classes; calculation of weighting factors of vulnerability assessment parameters; selection of optimal territorial operational unit for landscape vulnerability assessment. The method is implemented in the GIS environment "Assessment of vulnerability of landscapes of the Kaliningrad region to anthropogenic impacts", created by the authors using modern geoinformation products. The specificity of spatial differentiation of different landscapes in terms of vulnerability to anthropogenic impacts at the regional and local levels is revealed. It is stated that the use of the methodology for assessing the vulnerability of landscapes to anthropogenic impacts and its integration into the system of nature management will ensure a balanced account of geoecological features and environmental priorities in territorial planning. It is of interest to specialists in the field of rational nature management, environmental protection, spatial planning.

Abstract:
In the proposed monograph, optimization models for managing limited resources in logical systems are considered. Such systems are primarily used by industrial enterprises, transport companies and trade organizations, including those that carry out wholesale activities. As a rule, the efficiency of these objects largely depends on how rational use of limited resources such as: consumer camera business, labor, vehicles, etc. In this paper, various approaches to managing such resources are considered both for deterministic models and for the situation when a number of model parameters are not specified exactly, that is, for stochastic models. In this case, it is proposed to evaluate the stability of models to the occurrence of various types of risk events, both by the structure of the solution and by the functionality. It is addressed to senior students, postgraduates and masters studying in the specialty "Management" and "Logistics", as well as specialists in the field of logistics systems modeling.

Abstract:
Ultra-low power turbine drives are used as energy sources in auxiliary power systems, energy units, terrestrial, marine, air and space transport within the confines of shaft power Ntd = 0.01...10 kW. They represent a set of input, output devices and an ultra-low power turbine (ULPT). The main objective of this article is to describe and to validate a new modified method for selecting rational parameters of two-stage axial ULPT with pressure stages that is based on the multi-objective optimization in conditions of initial data uncertainty. Using this method allows to design a high-performance turbine already at the preliminary design. This paper proposes an approach to the development of surrogate models for evaluating the integrated efficiency of two-stage ultra-low power turbine with pressure stages that is based on the use of existing mathematical models of turbine stage efficiency and mass. The paper describes the basic features of subroutine algorithm for parameters optimization and for efficiency criteria evaluating of a two-stage turbine. This method is intended for use at the preliminary design of a turbine drive. Results of optimization through this method can be applied at the stages of 3D gas-dynamic flow simulation, design and manufacturing of a turbine physical model. The method was tested at preliminary design of an air starter turbine for starting a diesel engine in cold climatic conditions. Validation of the method was carried out by comparing the results of optimization calculations and numerical gas-dynamic simulation in the Ansys CFX package. The results indicate a sufficient accuracy of the developed method for selection of two-stage turbine parameters.

Abstract:
Currently, the construction sector is facing requirements for higher quality and efficiency standards as well as objectives related to value creation. In construction, rational decision-making problems often involve creating a unique complex of criteria, which must consider multiple requirements and conditions. The paper presents the developed model for the selection of multi-layered external walls of a residential building. The model considers the following aspects and parameters: time, cost, quality, environmental conditions, aspects of sustainable development, and problemsolving benefits received from the assessment of alternatives and the development of the model. Game theory was used to calculate the defining rational decision. For the calculations was used game theory: Bayes, Wald’s rule, Savage criterion, Laplace’s rule. This study presents the developed model for the selection of multi-layered external walls of a residential building and calculation results based on game-theory rules, which are effective tools for rational decisionmaking.

Abstract:
The structural parameter estimation problem based on measured modal data is formulated as a multi-objective optimization problem in which modal metrics measuring the fit between measured and model predicted groups of modal properties are simultaneously minimized to obtain all Pareto optimal structural models consistent with the measured data. Equivalently, the multiple Pareto optimal models can be obtained by minimizing a single metric formed as a weighted average of the multiple metrics. The Pareto optimal models are obtained by varying the values of the weights. The optimal values of the parameters are sensitive to the values of the weighting factors. A Bayesian statistical framework is used to provide a rational choice of the optimal values of the weight factors based on the available data. It is shown that the optimal weight values for each group of modal properties are asymptotically, for large number of data, inversely proportional to the optimal prediction errors of the corresponding modal group. Two algorithms are proposed for obtaining simultaneously the optimal weight values and the corresponding optimal values of the structural parameters. The proposed framework is illustrated using simulated data from multi-DOF spring-mass chain structure. In particular, compared to conventional parameter estimation techniques that are based on pre-selected values of the weights, it is demonstrated that the optimal structural models proposed by the methodology are significantly less sensitive to large model errors or bad measured modal data, known to affect optimal selection.

Abstract:
This paper presents a method for selecting materials, cross-section shapes, and combinations thereof. The novelty of the method is the definition of Shape Transformers. These parameters are dimensionless measures of the geometric quantities of a cross-section. They describe the shape properties of a cross-section regardless of size and are thus invariant to any scaling imposed on the cross-section size. Shape Transformers are valuable with modelling the equation of mechanics and with the development of selection charts for optimum design. The rationale of the approach is that the fundamental equations of mechanics can be expressed by a product of four separable factors: the functional requirements, the material properties, the Shape Transformers, and the geometric quantities of a rectangle as defined by its cross-section size. This permits general expressions of performance indices to be derived for any scaling transformation. For example, indices for selecting materials and cross-sectional shapes that minimize the mass of beams are given for stiffness design. The last part of the paper illustrates how Shape Transformers facilitate a graphical exploration of performance data. The whole range of cross-sectional shapes can be visualized at a glance for each material. Lines of iso-performance enable efficiency comparison of materials and/or shapes for a given cross-section scaling. Shape Transformers assist design choices and give insight into optimum selection.

Abstract:
Offshore and subsea decommissioning will increase in the next five years or so as many producing fields are matured and cease production while the oil price continues to remain low. This emphasizes the need for a thorough decommissioning plan to ensure a safe and technically feasible solution while it is economically viable and safeguards the environment. Offshore and subsea decommissioning is commonly considered on a case-by-case basis using the Comparative Assessment (CA) process in which the best decommissioning solution is obtained. Health, Safety and Environmental (HSE) considerations are always paramount in any decommissioning process. The aim is to significantly reduce the long term risks to other benefactors of the sea while the associated short term risks to those responsible for decommissioning operations are minimized. A major part of any decommissioning project is subsea pipelines decommissioning (by “pipelines”, it is meant to include flowlines, trunklines and flexible too). There are a number of techniques available for decommissioning of subsea pipelines ranging from preservation for potential future use to full recovery or leaving in-situ. However, each subsea pipeline decommissioning technique should be considered on its own merit. Selection of each decommissioning technique depends on many parameters, inter alia, size of pipeline, type of pipeline (e.g. single pipe, pipe-in-pipe, piggyback), type of conveying fluid, operational environment (location), production history, Inspection, Repair and Maintenance (IRM) records, HSE considerations, connection to other facilities, technical feasibility (including potential use of advanced technologies), regulatory authorities requirements and socio-economic considerations. This paper will look at specifics of subsea pipelines decommissioning. It will examine the procedures to be undertaken from desk top activities (e.g. planning and CA) up to operational activities (e.g. pigging, flushing, cleaning, removal or leaving in-situ). Different scenarios are discussed and potential advantages and disadvantages of each scenario are presented. In addition, a guide is proposed for future pipelines decommissioning projects to follow a rational approach.

Gelb, Jr., Jack, Yoram Weisman, Brian Ladman, and Rosie Meir. Identification of Avian Infectious Brochitis Virus Variant Serotypes and Subtypes by PCR Product Cycle Sequencing for the Rational Selection of Effective Vaccines. United States Department of Agriculture, December 2003. http://dx.doi.org/10.32747/2003.7586470.bard.

Abstract:
Objectives 1. Determine the serotypic identities of 40 recent IBV isolates from commercial chickens raised in the USA and Israel. 2. Sequence all IBV field isolates using PCR product cycle sequencing and analyze their S 1 sequence to detennine their homology to other strains in the Genbank and EMBL databases. 3. Select vaccinal strains with the highest S 1 sequence homology to the field isolates and perform challenge of immunity studies in chickens in laboratory trials to detennine level of protection afforded by the vaccines. Background Infectious bronchitis (IB) is a common, economically important disease of the chicken. IB occurs as a respiratory form, associated with airsacculitis, condemnation, and mortality of meat-type broilers, a reproductive form responsible for egg production losses in layers and breeders, and a renal form causing high mortality in broilers and pullets. The causative agent is avian coronavirus infectious bronchitis virus (IBV). Replication of the virus' RNA genome is error-prone and mutations commonly result. A major target for mutation is the gene encoding the spike (S) envelope protein used by the virus to attach and infect the host cell. Mutations in the S gene result in antigenic changes that can lead to the emergence of variant serotypes. The S gene is able to tolerate numerous mutations without compromising the virus' ability to replicate and cause disease. An end result of the virus' "flexibility" is that many strains of IBV are capable of existing in nature. Once formed, new mutant strains, often referred to as variants, are soon subjected to immunological selection so that only the most antigenically novel variants survive in poultry populations. Many novel antigenic variant serotypes and genotypes have been isolated from commercial poultry flocks. Identification of the field isolates of IBV responsible for outbreaks is critical for selecting the appropriate strain(s) for vaccination. Reverse transcriptase polymerase chain reaction (RT-PCR) of the Sl subunit of the envelope spike glycoprotein gene has been a common method used to identify field strains, replacing other time-consuming or less precise tests. Two PCR approaches have been used for identification, restriction fragment length polymorphism (RFLP) and direct automated cycle sequence analysis of a diagnostically relevant hypervariab1e region were compared in our BARD research. Vaccination for IB, although practiced routinely in commercial flocks, is often not protective. Field isolates responsible for outbreaks may be unrelated to the strain(s) used in the vaccination program. However, vaccines may provide varying degrees of cross- protection vs. unrelated field strains so vaccination studies should be performed. Conclusions RFLP and S1 sequence analysis methods were successfully performed using the field isolates from the USA and Israel. Importantly, the S1 sequence analysis method enabled a direct comparison of the genotypes of the field strains by aligning them to sequences in public databases e.g. GenBank. Novel S1 gene sequences were identified in both USA and Israel IBVs but greater diversity was observed in the field isolates from the USA. One novel genotype, characterized in this project, Israel/720/99, is currently being considered for development as an inactivated vaccine. Vaccination with IBV strains in the US (Massachusetts, Arkansas, Delaware 072) or in Israel (Massachusetts, Holland strain) provided higher degrees of cross-protection vs. homologous than heterologous strain challenge. In many cases however, vaccination with two strains (only studies with US strains) produced reasonable cross-protection against heterologous field isolate challenge. Implications S1 sequence analysis provides numerical similarity values and phylogenetic information that can be useful, although by no means conclusive, in developing vaccine control strategies. Identification of many novel S1 genotypes of IBV in the USA is evidence that commercial flocks will be challenged today and in the future with strains unrelated to vaccines. In Israel, monitoring flocks for novel IBV field isolates should continue given the identification of Israel/720/99, and perhaps others in the future. Strains selected for vaccination of commercial flocks should induce cross- protection against unrelated genotypes. Using diverse genotypes for vaccination may result in immunity against unrelated field strains.
