Abstract:
The mechanistic target of rapamycin kinase complex 1 (mTORC1) is a key signaling hub that acts as a central regulator of several cellular processes, including cell growth and metabolism. The activation of mTORC1 occurs at the lysosomal surface via a two-step mechanism that requires a) its amino acid-dependent recruitment to the lysosome via the Rag GTPases and b) its growth factor-dependent activation by Rheb. mTORC1 senses and integrates multiple upstream signals to phosphorylate a broad number of substrates and modulate the crucial balance between cell anabolism and catabolism. However, whether mTORC1 can differentially regulate specific proteins to selectively respond to such a variety of intracellular and environmental cues is poorly understood. Here we show that Transcription Factor EB (TFEB), a master modulator of lysosomal biogenesis and autophagy, is modulated by mTORC1 via a specific substrate recruitment mechanism that is mediated by Rag GTPases. Differently from the well-characterized mTORC1 substrates S6K and 4E-BP1, which are recruited by mTORC1 via binding to the regulatory subunit Raptor, TFEB interaction with mTORC1 relies on its physical association with active Rag C/D. Owing to this mechanism, TFEB phosphorylation is insensitive to growth factor-mediated activation of Rheb but highly sensitive to amino acid-mediated activation of Rag GTPases. Strikingly, substituting the region of TFEB responsible for its recruitment to mTORC1 with the one of S6K, inverted TFEB phosphorylation behaviour and made it similar to S6K/4E-BP1. Thus, our findings reveal that diversity in mTORC1 substrate recruitment mechanisms enables mTORC1 to induce selective responses to specific nutritional cues.

Abstract:
V(D)J recombination is a process that generates the diversity of the immune repertoire against foreign antigens. During B and T cell development, genes encoding immunoglobulins (Ig) and T cell receptors (TCR) variable region are somatically assembled by selective V (variable), D (diversity) and J (joining) segments pre-existing in the germline. Recombination-activating genes 1 and 2 (RAG1/RAG2), the lymphocyte-specific factors, initiate V(D)J recombination by nicking at the border the heptamer sequence of RSS (recombination signal sequence) and generate four DNA double stranded breaks (DSB) in the cleavage step. After cleavage, RAG1/RAG2 complex are still bound to the DNA ends. In the joining step, DNA breaks are processed and rejoined by non-homologous end joining apparatus, which includes Ku70/Ku80, DNA-PKcs, Artemis, XRCC4 and DNA ligase IV. However, how the cleavage step is linked to the joining step is not yet known. My results suggest that a physical interaction between RAG1/2 and Ku antigen may help coordinate the cleavage stage of V(D)J with the non homologous DNA end rejoining of the mature sequences. Abstract:
Redundant Arrays of Inexpensive Disks or RAID is a popular method of improving the reliability and performance of disk storage. Of various levels of RAID, mirrored or RAID1 and rotating parity or RAID5 configurations have become moat popular. Mirrored or RAID1 provides best overall performance and is easier to configure, but has 100 percent storage overhead for the redundancy. Rotating parity or RAID5, on the other hand, is quite inexpensive for the redundancy it provides, shorn impressive performance for reads and full-stripe writes in normal mode, but the small write performance is poor due to the read-modify-write cycle involved. The performance drops drastically when one of the disks fails and the system enters degraded mode. Also RAID5 is relatively difficult to configure. Typical non-scientific system disk access patterns exhibit very high locality of reference. This thesis presents the design and implementation of an Adaptive Hierarchical RAID array to exploit this high locality. Frequently accessed data is migrated towards the top of the hierarchy and not so frequently acee88ed data is moved down the hierarchy, thus adaptively rearranging itself to the access patterns. Previous work on Adaptive Hierarchical RAID such as HP AutoRAID has explored one part of the design space, namely design of configurable storage at the SGSI level with no interaction with higher level layers like volume manager. This thesis explores a different design point: namely, one that is centered at the volume manager layer. This is important also for the reason that with fibre channel disks and SCSI-3, Storage Area Networks (SAN) no longer need a conventional controller but a modified version of a controller that is more close to a volume manager.

Abstract:
RAID systems (Redundant Arrays of Inexpensive Disks) have dominated backend storage systems for more than two decades and have grown continuously in size and complexity. Currently they face unprecedented challenges from data intensive applications such as image processing, transaction processing and data warehousing. As the size of RAID systems increases, designers are faced with both performance and reliability challenges. These challenges include limited back-end network bandwidth, physical interconnect failures, correlated disk failures and long disk reconstruction time. This thesis studies the scalability of RAID systems in terms of both performance and reliability through simulation, using a discrete event driven simulator for RAID systems (SIMRAID) developed as part of this project. SIMRAID incorporates two benchmark workload generators, based on the SPC-1 and Iometer benchmark specifications. Each component of SIMRAID is highly parameterised, enabling it to explore a large design space. To improve the simulation speed, SIMRAID develops a set of abstraction techniques to extract the behaviour of the interconnection protocol without losing accuracy. Finally, to meet the technology trend toward heterogeneous storage architectures, SIMRAID develops a framework that allows easy modelling of different types of device and interconnection technique.Simulation experiments were first carried out on performance aspects of scalability. They were designed to answer two questions: (1) given a number of disks, which factors affect back-end network bandwidth requirements; (2) given an interconnection network, how many disks can be connected to the system. The results show that the bandwidth requirement per disk is primarily determined by workload features and stripe unit size (a smaller stripe unit size has better scalability than a larger one), with cache size and RAID algorithm having very little effect on this value. The maximum number of disks is limited, as would be expected, by the back-end network bandwidth. Studies of reliability have led to three proposals to improve the reliability and scalability of RAID systems. Firstly, a novel data layout called PCDSDF is proposed. PCDSDF combines the advantages of orthogonal data layouts and parity declustering data layouts, so that it can not only survivemultiple disk failures caused by physical interconnect failures or correlated disk failures, but also has a good degraded and rebuild performance. The generating process of PCDSDF is deterministic and time-efficient. The number of stripes per rotation (namely the number of stripes to achieve rebuild workload balance) is small. Analysis shows that the PCDSDF data layout can significantly improve the system reliability. Simulations performed on SIMRAID confirm the good performance of PCDSDF, which is comparable to other parity declustering data layouts, such as RELPR. Secondly, a system architecture and rebuilding mechanism have been designed, aimed at fast disk reconstruction. This architecture is based on parity declustering data layouts and a disk-oriented reconstruction algorithm. It uses stripe groups instead of stripes as the basic distribution unit so that it can make use of the sequential nature of the rebuilding workload. The design space of system factors such as parity declustering ratio, chunk size, private buffer size of surviving disks and free buffer size are explored to provide guidelines for storage system design. Thirdly, an efficient distributed hot spare allocation and assignment algorithm for general parity declustering data layouts has been developed. This algorithm avoids conflict problems in the process of assigning distributed spare space for the units on the failed disk. Simulation results show that it effectively solves the write bottleneck problem and, at the same time, there is only a small increase in the average response time to user requests.

Das maligne Melanom zählt zu den aggressivsten und behandlungsresistentesten aller Krebsarten. In den letzten 20 Jahren hat sich die Rate der Melanom-Erkrankungen innerhalb der weißen Bevölkerung verdreifacht. Mittlerweile liegen eine Reihe von Untersuchungen zu den molekularbiologischen Mechanismen der Entwicklung und Progression des malignen Melanoms vor. Aktuelle Forschungsvorhaben beschäftigen sich vor allem mit der Identifizierung Melanom-spezifischer Biomarker, die diagnostische und prognostische Informationen liefern sowie die Entwicklung einer zielgerichteten, kombinierten und individualisierten Therapie des metastasierenden Melanoms ermöglichen. In diesem Kontext soll die vorliegende Arbeit einen weiteren Beitrag zum Verständnis der Metastasierungskaskade und der daran beteiligten Proteine leisten. Aufgrund der Überexpression in einer Reihe von Tumoren und seiner geringen Molmasse von lediglich 11,5 kDa bietet sich das S100A4-Protein als Marker mit hoher prognostischer Signifikanz für verschiedene Tumorentitäten an. Jedoch ist die Beteiligung von S100A4 bei der Ausbildung des invasiven Tumorphänotyps noch nicht vollständig aufgeklärt. S100A4 besitzt zahlreiche intra- und extrazelluläre Bindungspartner, wobei die Metastasierung scheinbar ausschließlich durch das extrazelluläre Protein beeinflusst wird. S100A4 wechselwirkt extrazellulär beispielsweise mit dem Rezeptor für fortgeschrittene Glykierungsendprodukte (RAGE). Ziel dieser Arbeit war es, speziell die Bedeutung von S100A4 und seiner Interaktion mit RAGE für das prometastatische Verhalten von Melanomzellen in vitro und in vivo näher zu charakterisieren. Darüber hinaus sollte die Beteiligung von S100A4 bei der Gehirn-Metastasierung untersucht werden, wobei insbesondere die Regulierung der Endothelzell-Permeabilität und der transendothelialen Migration der Melanomzellen im Vordergrund stand. Im Rahmen dieser Arbeit wurde gezeigt, dass S100A4 und die Interaktion mit RAGE die prometastatischen Eigenschaften der A375-Melanomzellen förderte. Zudem verringerte extrazelluläres S100A4 die Zell-Integrität von Gehirn-Endothelzellen und erleichterte somit die Durchdringung der Blut-Hirn-Schranke. Diese Erkenntnis lässt sich möglicherweise auf andere Blut-Gewebe-Schranken übertragen. Die In-vivo-Orientierungsstudie zeigte, dass S100A4- und RAGE-überexprimierende Zellen zu einer verstärkten disseminierten Metastasierung führten, wobei sich zwei unterschiedliche Verteilungsmuster ergaben. Darüber hinaus führten beide Zelllinien vereinzelt zur Bildung von Gehirnmetastasen, wodurch sich die intrakardiale Injektion durchaus als Modell für weitere Therapiestudien mit dem Augenmerk der S100A4-RAGE-stimulierten Metastasierung eignet. Die genauere Kenntnis regulativer Mechanismen bei der Synthese und Sekretion von S100A4 sowie die pathophysiologische Differenzierung der S100A4-Interaktion mit RAGE eröffnen neue Wege, die S100A4-vermittelten Effekte therapeutisch zu beeinflussen. Daraus lassen sich möglicherweise neue zielgerichtete Radionuklid-basierte Therapieansätze für das metastasierende Melanom ableiten.

Abstract:
Computer data storage is growing at an astonishing rate. With cloud computing and the growth of the Internet enterprise storage has been predicted to grow at rates as high as 300\% per year. To fulfill this need technologies such as Redundant Array of Independent Disks or RAID are being used in industry today. Not only does RAID increase I/O performance but also provides redundancy measures to protect against hardware failure. Even though RAID has existed for some time now and is well understood, proprietary optimizations such as command scheduling and cache strategies that are employed by current RAID controllers are not well known. This thesis presents a model for RAID 5 that incorporates these features and describes the overall function of hardware RAID controllers. Also a python implementation of this model, Accurate Hardware RAID Simulator (AHRS) is presented and validated against a current hardware RAID controller. It is shown that AHRS can reproduce the behavior of a hardware RAID system with an accuracy of 97.92\% on average compared to a LSI hardware RAID controller.

Abstract:
Human herpesvirus 6A (HHV-6A) has yet to be definitively linked to a specific disease. This is due in part to the ubiquitous nature of the virus. Humanized Rag2-/-γc-/- (Rag-hu) mice were tested to determine if these were a suitable animal model to study the virus. Both cell-free and cell-associated virus was used for infection and both were found to be efficient at infecting the mice. Viral DNA was found in the plasma and cellular blood fractions, bone marrow, lymph node, and thymus, indicating successful infection and propagation of the virus in vivo. The CD3+CD4- population was depleted, while the CD3-CD4+ was increased in infected animals. The CD3-CD4+CD8- and CD3+CD4+CD8- populations were depleted and the CD3+CD4+CD8+ population increased when analysis was gated upon CD4+ cells. The CD3-CD4+CD8+ population expanded and the CD3-CD4+CD8- population was reduced when analysis was gated on the CD3- population. Additional flow cytometry analysis revealed increases in CD4+CD8+ double positive cells in the peripheral blood of cell-free infected mice, which could indicate improper T cell selection and a premature departure of these cells from the thymus, possibly contributing to autoimmunity. Previous research has shown that HIV and HHV-6A may have a synergistic effect on one another and that HHV-6A may act as a cofactor in the progression to AIDS. After determining the Rag-hu mouse model was suitable for studying HHV-6A infection, a coinfection of HHV-6A and HIV-1 was performed. Coinfected mice had fewer thymocytes when compared with the HIV-1 only, mock-infected, and to a lesser extent HHV-6A only groups which could indicate increased cell death in the coinfected group as well as possible disruptions in migration of cells, either causing cells to be sequestered in the bone marrow and unable to migrate to the thymus, or causing premature egress of the cells in the thymus due in part to premature upregulation of CCR7, both of which would explain the smaller cellular populations found in the coinfected mouse thymi. Additional studies were performed to determine if a preferential targeting existed between HHV-6A and HIV-1 as these viruses are found simultaneously coinfecting the same cell. Preferential targeting was not observed by cell-associated migration assay, but increased migration of HHV-6A-infected cells was observed in a CCL21 dependent manner. These studies have provided useful information about HHV-6A and its relevance to HIV/AIDS as well as a possible mechanism of the involvement of HHV-6A in multiple sclerosis (MS) and other autoimmune diseases.

O pinhão-manso (Jatropha curcas L.) é uma planta versátil e com diversos usos potenciais em especial para produção de biodiesel. O óleo obtido de suas sementes é seu produto mais rentável e atualmente desperta grande interesse econômico em diversos setores. A caracterização molecular e a avaliação do potencial reprodutivo dos acessos de pinhão-manso são aspectos ainda pouco abordados no Brasil, sendo necessária a caracterização de acessos brasileiros e suas potencialidades. Os objetivos do presente trabalho foram analisar a diversidade genética e a divergência genética em acessos comerciais de pinhão-manso, coletados no estado de São Paulo (Alvinlândia, Lins, Itatinga e Jales), Minas Gerais (Janaúba) e Mato Grosso do Sul (Dourados). Primeiramente, foram comparadas as técnicas de RAPD (Random Amplified Polymorphic DNA), RAPD-RFLP (Restrinction Fragment Length Polymorphism a partir do RAPD) e AFLP (Amplified Fragment Length Polymorphism), quanto à potencialidade de detectar polimorfismos dentro dos acessos e entre os acessos. Posteriormente, os acessos foram caracterizados através de AFLP. Além disso, este trabalho visou detectar se há diferença nas estruturas e no desenvolvimento floral e na taxa de viabilidade polínica dos acessos de São Paulo (Alvinlândia), Minas Gerais e Mato Grosso do Sul. Nessa parte do estudo, foram analisadas e identificadas as estruturas dos órgãos florais e do grão de pólen através das técnicas de microscopia de luz, eletrônica de varredura e transmissão. Na comparação entre as técnicas moleculares, apenas AFLP permitiu a detecção de polimorfismos, com base em um acesso escolhido para os testes (Alvinlândia, SP). As análises de todos acessos através de AFLP mostraram considerável polimorfismo, que foi refletido nas estimativas de diversidade genética de Nei e Índice de Shannon. Na análise de agrupamento, as amostras de cada acesso foram agrupadas coerentemente. Os acessos de Dourados e de Itatinga formaram um grupo separado, apresentando cerca de 76% de similaridade com o segundo grupo (Alvinlândia, Jales, Lins, Janaúba). Dentro deste último, os acessos de São Paulo ficaram agrupados (com cerca de 84% de similaridade), enquanto que Janaúba mostrou-se separado, com 82% de similaridade genética. Estes resultados indicam que os acessos de São Paulo provavelmente tenham origem a partir de Janaúba. No entanto, Itatinga provavelmente tenha origem a partir de Dourados. Quanto à morfologia floral, não foi observada nenhuma diferença a nível microscópico na formação e nas estruturas dos órgãos florais. Quando comparada a viabilidade polínica entre os dois acessos coletados em campo (Minas Gerais e Mato Grosso do Sul), notou-se que há diferença significativa na taxa de viabilidade, mesmo os valores sendo muito próximos (Mato Grosso do Sul - 81,91% e Minas Gerais - 77,20%), mostra que tanto os acessos de Minas Gerais como os de Mato Grosso do Sul podem ser utilizados como parentais masculinos em programas de melhoramento genético. A divergência genética moderada e a baixa variabilidade morfológica sugerem que coletas de materiais em poucos locais poderiam representar a diversidade genética da espécie. Estudos mais aprofundados, empregando técnicas como de microssatélites e mais acessos, são 11 necessários para o conhecimento da diversidade e estrutura genética do pinhão-manso no Brasil
Physic nut (Jatropha curcas L) is a very versatile plant with several potential uses especially for the production of biodiesel. The oil obtained from its seeds is the most profitable product of this specie and it is currently attracting great interest in various economic sectors. Molecular characterization and evaluation of reproductive potential of the accessions of pysic nut have still little attention in Brazil, it still need the characterization of Brazilian accessions and its potenciality. The ains of this study were: to analyze the genetic diversity and genetic divergence in commercial accessions of physic nut, from the States of São Paulo (Alvinlândia, Lins, Jales and Itatinga), Minas Gerais (Janaúba)and Mato Grosso do Sul (Dourados). Firstly we compared the RAPD (Random Amplified Polymorphic DNA) RAPD-RFLP (Fragment Length Polymorphism Restriction from RAPD) and AFLP (Amplified Fragment Length Polymprphism) regarding the potencial to detect polymorphism within the accessions and among accessions. Subsequently, the accessions were characterized by AFLP. Moreover this work aimed to detect whether there are differenced on flower structures and development and the rate of pollen viability on the accessions of São Paulo (Alvinlândia), Minas Gerias and Mato Grosso do Sul. On this part of the study were analysed and identified the structures of the floral organs and pollen grain through the techniques of, light microscopy, scanning electron and transmission microscopy. Comparing the molecular techniques, only AFLP allowed the detection of polymorphisms based on the accession chosen for testing (Alvinlândia, SP). Analyses of all accesses by AFLP showed considerable polymorphism, wich was reflected in estimates of genetic diversity index of Nei and Shannon. In cluster analysis, samples of each accessions were grouped consistently. The accessions of Dourados and Itatinga formed a separated group, with approximately 76% similarity with the second group (Alvinlândia, Jales, Lins, Janaúba). Within the last, accessions of São Paulo werte grouped (about 84% similarity), while Janaúba showed p separately, with 82% of genetic similarity. These results indicates that the accessions of São Paulo probably originates from Janaúba. However Itatinga probably originates from Dourados. As floral morphology, we observed no difference at microscope level on the formation and on the structures of floral organs. When compared the pollen grain viability between the two accessions collected in the field (Minas Gerais and Mato Grosso do Sul), it was noted that the viability rated were significantly different, even though the values being very close (Mato Grosso do Sul Minas Gerais 81,91% - 77,20%), shows that both the accessions of Minas Gerais as the Mato Grosso do Sul can be used as male parental in breeding programs. The moderate and low genetic divergence suggests that morphological variability collections of materials in a few locations could represent the genetis diversity of the species. Further studies employing techniques such as microsatellite and more accessions are required for an understanding of diversity and genetic structure of physic nut in Brazil

Denna B-uppsats tar upp prestanda och återställning mellan två vanligt förekommande mjukvarubaserade RAID-system. Mjukvarubaserad RAID har blivit mer och mer intressant eftersom det är ett billigt och bra sätt att skydda sig mot hårdvarufel på hårddiskar. Tidigare har det inneburit prestandaförluster om man inte har använt en hårdvarubaserad RAID kontroller men med dagens snabba processorer är det inte längre så. Hårddiskar går för eller senare sönder och för ett RAID-system som ska vara igång tjugofyra timmar om dygnet är det viktigt att det även då fungerar med bra prestanda. Denna undersökning visar hur mycket sämre prestandan blir under sådana omständigheter samt hur lång återställningstid man kan förvänta sig på ett datorssystem uppbyggt av delar för hemmabruk. Undersökningen utfördes på två olika typer mjukvarubaserad RAID, ett med mdadm på Ubuntu/Linux och ett med RAID-Z på OpenSolaris. På de systemen mättes läs- och skrivprestanda med programmet bonnie++ som skriver och läser en fil som var dubbelt så stor som primärminnet i datorn. Resultatet visar att prestandan försämras med 36 % när en hårddisk går sönder på ett system med mdadm men med RAID-Z ökar istället prestandan. På ett system med mdadm är återställningstiden för ett degraderat system är ungefär lika lång oavsett hur mycket information som finns lagrad på hårddiskarna. Däremot går det fortare med RAID-Z eftersom den på ett intelligent sätt bara återställer den del av hårddisken som används för lagring av filer.

I detta arbete har målet varit att numeriskt beräkna en lösning till värmeledningsekvationen med rörlig rand med en slumpvandringsmetod. Analytiska lösningar samt lösningar med andra numeriska metoder nns det gott om, det stora problemet ligger i den rörliga randen. Första halvan behandlar en del teori kring värmeledningsekvationen, i synnerhet lösningar med Dirichletvillkor. I den andra halvan behandlas slumpvandringsmetoden för den homogena värmeledningsekvationen. Metoden appliceras först på enkla exempel, för att sedan lösa Stefanproblemet. I slutet av arbetet görs en kort felanalys för den numeriska lösningen av Stefanproblemet. Detta arbete har, som sagt, behandlat en lösning via en slumpprocess till ett exempel på värmeledningsekvationen med rörlig rand. Numeriska metoder genom slumpprocesser för att lösa partiella dierentialekvationer med just rörlig rand är något som det idag forskas på, varpå detta arbete ligger i linje med vad som är aktuellt för ämnet. Exempelvis så behandlar [1] en del kring vad som är aktuellt för s.k. free boundary problems och i [2] diskuteras det aktuella metoder för att utvidga slumpprocesser till ickelinjära problem.

Abstract:
The thesis argues that psychoanalysis is a necessary component of cultural analysis. It is argued that existing syntheses of psychoanalysis and political theories tend to limit the recognition of the relative autonomy of psychic reality by offering accounts of the social determination of subjectivity. The contemporary reappropriation of psychoanalysis by feminist theorists has formulated new explanations of the social position of women as the 'second sex'. The challenge of feminism to traditional theories of culture and society includes questions of how sexual difference informs the transformation of thought into language, how language determines theory, and how theory conceptualises the difference between subjectivity and objectivity. The contradictions within existing syntheses of structuralism, Marxism and feminism are described, and the differences between psychoanalysis and sociology are traced through the the critical reception of Freud's Totem and Taboo by anthropologists. The validity of Freud's concept of the Oedipus complex is explored, and it is suggested that despite the limited acceptance by anthropologists, Totem and Taboo contains a valid theory of the relation of the subject to society. Freud's work is relocated within the paradigm of evolutionary biology to provide a materialist analysis of psychic structure that is not based on linguistics. A study of the origins of language reveals the complexity of the historical factors determining the co-evolution of representation, the maternal function, and the structuration of psychic reality. New discoveries about the pre-Oedipal dyad that underlies the Oedipus complex have shown the effects of infantile dependence and maternal care on adult subjectivity, and it is argued that factors such as the unconscious fear of dependency and of women are of particular significance for feminist thought. It is argued that the theory of pre-Oedipal and prelinguistic subjectivity can make intelligible aspects of ideologies of racism and sexism that are not fully explained by sociological or political theory. The mechanism of projection or projective identification, it is argued, provides a specifically psychoanalytic contribution to existing theories of culture.

Redundant Array of Independent Disks (RAID) provides an inexpensive, fault-tolerant storage solution using commodity hard-drives. RAID storage systems have recently surged in popularity amongst enterprise clients, as they provide economical, scalable, high-performance solutions for their storage requirements. The performance of RAID systems is negatively affected by the overhead required to manage and access multiple drives, and multiple disk failures can result in data loss. As RAID has developed, various improvements have been devised by both academia and business to address these shortcomings. These improvements have suggested improved architectures to increase performance and new coding techniques to protect against data loss in the case of drive failure. Evaluating the effect on performance of these improvements is greatly simplified by the use of discrete-event, software simulations. The RAID Operations Simulator for Testing Implementations (RÖSTI) was developed to support such simulation experiments.

Knowledge of the genetic variability of Brachiaria ruziziensis is required before you start a breeding program. This study investigated the genetic variability of 37 samples of Brachiaria ruziziensis, from the collection held by EMBRAPA Dairy Cattle by molecular markers and long primers. The DNA was extracted from leaves by CTAB method, we used 14 decamer primers and primers 9 long. The data were used for analysis of molecular variance (AMOVA) using the squared distances (Excoffier et al., 1992) using the program Arlequin (Excoffier et al., 2006). We found variation between species of 47.42% and 36.07% for RAPD primers and long, respectively and within the species observed variation of 52.58% and 63.93%. A dendrogram was constructed with the UPGMA clustering algorithm (Unweighted Pair-Group Method using an Arithmetic Average) using this analysis to the program NTSYS 2.0 (ROHLF 1998). Both markers were able to group the samples where genotypes 1 to 5 the most similar to each other. RAPD using primers short and long primers are effective to estimate the variability in Brachiaria ruziziensis.
O conhecimento da variabilidade genética de Brachiaria ruziziensis é necessário antes de iniciar-se um programa de melhoramento. Este trabalho objetivou estudar a variabilidade genética de 37 amostras de Brachiaria ruziziensis, provenientes de coleta realizada pela EMBRAPA Gado de leite por meio de marcadores moleculares RAPD e primers longos. O DNA das folhas foi extraído pelo método CTAB, utilizaram-se 14 primers decâmeros e 9 primers longos. Os dados foram utilizados para análise de variância molecular (AMOVA) utilizando-se as distâncias ao quadrado (EXCOFFIER et al., 1992) com auxílio do programa Arlequin (EXCOFFIER et al., 2006). Encontrou-se variação entre espécies de 47,42% e 36,07% para RAPD e primers longos, respectivamente e dentro da espécies observou-se variação de 52,58% e 63,93%. Foi construído um dendrograma com o algoritmo de agrupamento UPGMA (Unweighted Pair-Group Method Using an Arithmetic Average) utilizando-se para essa análise o programa NTSYS 2.0 (ROHLF, 1998).Ambos marcadores conseguiram agrupar os genótipos onde as amostras 1 a 5 foram as mais semelhantes entre si. RAPD utilizando-se primers curtos e primers longos são eficientes para estimar a variabilidade em Brachiaria ruziziensis.

Les évènements cardiovasculaires sont la première cause de mortalité chez les patients insuffisants rénaux chroniques. Ces complications font suite à des modifications structurelles et fonctionnelles de la paroi vasculaire regroupées sous le terme de vasculopathie urémique. Parallèlement à ces modifications vasculaires, l’urémie s’accompagne d’une accumulation de substances non épurées par le rein appelées toxines urémiques, telles que les produits de la glycation avancée (AGEs) ou les ligands de RAGE. Ces toxines peuvent interagir avec leur récepteur, RAGE, qui est pro- inflammatoire et impliqué dans le remodelage artériel. Cette thèse a consisté en l’étude, chez la souris, du rôle de l’accumulation des ligands de RAGE et de leur interaction avec celui-ci dans le développement de l’athérosclérose, des calcifications vasculaires et de la thrombose artérielle au cours de l’insuffisance rénale chronique (IRC). Dans un premier temps, nous avons montré que l’IRC conduisait à une accumulation des AGEs et des ligands de RAGE sériques et tissulaires, ainsi qu’une augmentation de l’expression de RAGE au sein de la paroi vasculaire participant à la formation des plaques d’athérome. Dans un second temps, nous avons démontré que RAGE participait aux calcifications vasculaires favorisant l’expression de co-transporteur de phosphate inorganique (Pit-1), induisant la différenciation des cellules musculaires lisses en cellules « osteoblast-like ». Enfin, nous avons montré que RAGE participait à la formation d’un thrombus artériel dû à une hyperactivité plaquettaire. En conclusion, cette thèse a permis de renforcer le concept que l’axe ligands de RAGE/RAGE est un acteur important dans la vasculopathie urémique
Cardiovascular events are the primary cause of morbidity and mortality in chronic kidney disease patients. These complications are due to structural and functional changes in the vascular wall named uraemic vasculopathy. Alongside these vascular changes, uremia is accompanied by the retention of various solutes that are normally excreted by the kidneys called uremic toxins, such as the products of advanced glycation (AGEs) or the ligands of RAGE. These toxins may interact with their receptor, RAGE, which is pro-inflammatory and involved in arterial remodeling. The aim of this thesis was to study, in mice, the role of the accumulation of RAGE ligands and their interaction with it in the development of atherosclerosis, vascular calcification and arterial thrombosis in chronic renal failure (CKD). Initially, we showed that CKD leads to an accumulation of serum and tissue AGEs and RAGE ligands, as well as an increase in RAGE expression in the vascular wall involved in atheroma plaque formation. Secondly, we have demonstrated that RAGE is involved in vascular calcification promoting the expression of inorganic phosphate cotransporter (Pit-1), inducing the differentiation of smooth muscle cells "osteoblast-like". Finally, we showed that RAGE participated in the formation of arterial thrombus due to platelet hyperactivity. In conclusion this thesis consolidates that RAGE-RAGE ligands axis is an important actor in uremic vasculopathy

This study was undertaken to describe how the community responded to the incorporation of the Merafong Municipality into the North West Province. The study examined the protracted conflict in Khutsong as well as why the strategies to end the conflict failed. The study was concerned about latent conflict; how differences are raised; dissatisfaction amongst members of the communities and that power asymmetry is the main driver of emotions leading to aggressive behaviour. Service delivery protests are characterised by violence in South Africa and more often communities turn violent in their engagement with their leaders or council officials. This is because it is believed that violence is the only language that authorities understand and they have nothing to lose as government property gets destroyed. Late invitations of third parties who have regulatory skills (negotiations, mediation, conciliation or judicial processes) when conflicts begin, lead to their escalation. A qualitative research method was chosen for this study. A qualitative case study better explained the phenomenon of interest. It allowed me to collect data from the respondents who were active members of the community through interviews. Twelve members of the community (from politicians, ordinary members, educators and the clergy) were part of the respondents. The report was of the respondents own personal experiences as active members of the community. Collected data from the transcripts was coded. Codes with similar meaning were put together in the same categories and the themes were formed from the categories. The qualitative content analysis method was used to analyse the data. From the analysis, it emerged that there was lack of communication and consultation from the side of the government before the implementation of the 16th Amendment Bill which was aimed at removing the cross border municipalities. The announcement was finally made of the incorporation of the Merafong Municipality into North West Province in 2005. The announcement was met with anger and frustration by the Khutsong Demarcation Forum and the community whose expectations were that the incorporation will not go ahead given the reasons they put forth when they met the government officials in the meetings after the demarcation board had proposed it. Violence was the only option for the community to show that their talks with government officials failed and that they are seriously opposed to the incorporation. That led to a protracted violence in Khutsong until 2009 when the Cross-Boundary Municipalities Laws Repeal and Related Matters Amendment Act of 2009 act was passed to reincorporate the Merafong Municipality back into Gauteng.

Exchange rates are one of the most essential determinants of a country's economic performance in terms of level of trade. Since the exchange rate is one of the best indicators of competitiveness, this study sought to examine the behaviour of the rand against other emerging countries in the South African exchange market. The study explored the trends and estimated the forecasting accuracy of six currency markets using ARCH-family and Random walk models over the period 1994 to 2013.The six currency markets examined were the Rand/Dollar, Rand/Pound, Rand/Euro, Rand/Yen and Rand/Pula. The Rand exchange rates did exhibit the characteristics of volatility clustering and asymmetric effects suggesting volatility of the Rand. Exchange rates tend to rise when there is more bad news in the financial market than good news and positive shocks imply a higher next period conditional variance than negative shocks of the same sign.
