Abstract:
Since the beginning of the early 20th century bed rugs have been interesting research objects, but only in passing researchers have paid attention to rugs woven with rags. Noone has taken a closer look upon the reasons why people have woven these rugs. What does the materials of the rag – like recycled garments and interior textiles in the form of clothing rags – have to tell about the times when these rugs were woven? Did the rag rugs have any specific function or were the materials available and therefore used? The main sources of information are 21 rag rugs from Roslagen in Uppland, with a varied amount of rags. They were woven during the latter half of 19th century, and estate inventories from Väddö- and Häverö Ship District have altogether given some answers to the primary question of this paper: in wich way can the examined bed rugs bear witness to the use and value of recycled textile materials and the view of these in the context of the community where they were manufactured and used? This research doesn´t give an answer to whether these rugs have any particular function or not. Instead these rag rugs can be looked upon as representing a general development of the society towards an increasing amount of textiles surplus material. This being due an increasing consumption of factory-made clothing and textiles, manufactured in factories, as well as the paper mills development from producing paper made of cellulose rather than textile waste. To this we can add a principle lingering on from the 19th century, of domestic production and a thrift of resources. This resulted in an obvious recycling of discarded textiles. The home weaving of interior textiles was still strong by the end of the 19th century, and in Rosagen there was also a long tradition of weaving and of using rugs in the beds. At the same time there was, in the coastal regions of Roslagen, a local need for warming covers in boats and boat houses. This demand was related to the shooting of seals and other hunting in the coastal areas, as well as in the fishing- and maritime trades.

Abstract:
The very impetus of this study — to examine the representations of craft in literature — defies the functional binaries so long attributed to art and craft. This study examines the literary formulations of textile crafts and their makers in Canadian works of fiction at the turn of the twenty-first century. Included are three Canadian novels published after 1990: Margaret Atwood's Alias Grace (1996), Austin Clarke's The Polished Hoe (2002) and Rohinton Mistry's A Fine Balance (1995). Through close analysis of these patchwork novels, I suggest ways of reading quilts and other textile crafts as a recontextualization of the forms of the past (through the workings of displacement and parody) in Canadian literature. Chapter One proposes theoretical reconceptualizations of crafts culminating in the 1990s and establishes three paradigms that structure my analysis in each of the chapters: the relations of textile crafts with (a) narrative, (b) trickery, and (c) a dehierarchical and plural aesthetic. In the subsequent chapters, each one dealing with a single novel, I explore the reassembled quality of the narratives and variations of the spider-weaver archetypes they represent, both of which I consider fundamental to the patchwork novel. In Chapter Two, I posit the patchwork quilt in Atwood's Alias Grace as a model for the processes of recollection and fragmentation involved in historiographic metafiction. Chapter Three establishes the crafted object in Clarke's The Polished Hoe as a site of struggle and an embodiment of the collective and composite nature of heritage in the neoslave narrative. Cette étude contribue à remettre en question la célèbre dichotomie entre l'art et l'artisanat en se penchant sur les représentations de l'artisanat dans la littérature. Plus spécifiquement, cette étude vise à explorer les représentations de l'artisanat textile et de la figure de l'artisan dans le roman canadien au tournant du vingt-et-unième siècle, à travers trois romans publiés après 1990 : Alias Grace (1996) de Margaret Atwood, The Polished Hoe (2002) d'Austin Clarke et A Fine Balance (1995) de Rohinton Mistry. Une analyse de ces trois romans-patchwork et du rapiéçage qui en informe leur structure et leur contenu nous révèle une nouvelle façon de conceptualiser l'artisanat tout en remettant en contexte des formes traditionnelles du passé (tels que tissage, tressage, couture) dans la littérature canadienne contemporaine. Abstract:
The Self-Cycling Fermentation (SCF) technique was applied to a culture of Acinetobacter calcoaceticus RAG-1. Both insoluble and soluble carbon substrates were used as the limiting nutrient. The disappearance of the carbon source was found to correspond with the attainment of a minimum in dissolved oxygen concentration. Cellular occurences, averaged out with asynchronous batch cultures, were magnified by the temporal alignment of metabolic events brought about by the synchronization associated with SCF's. The cell population doubled only once per cycle, thus establishing an equality between cycle time and doubling time. Parameters of primary interest were biomass concentration, total bioemulsifier (emulsan) production, cycle time, and residual carbon concentration. Cycle to cycle variation of these parameters was, in most cases, insignificant. The carbon substrate was completely utilized in all cases in which it was measured, giving this technique an advantage over chemostat-type fermentations. The dissolved oxygen profiles followed throughout a cycle were found to be repeatable. A characteristic shape was associated with each carbon source which can be related to the growth of the organism. Finally, the specific emulsan productivity of SCF's was found to be comparable to those achieved by systems presently implemented.

Nach der Verstaatlichung, 1751, wurde im saarländischen Steinkohlerevier durchgehend bis Mitte 2012 Kohle gefördert. Seit Ende des 18. Jahrhunderts wurden auch Riss- und Kartenwerke geführt, die seit dieser Zeit die Lagerstätte und alle bergbaulichen Aktivitäten dokumentierten. Mehrere tausend Rissplatten auf Kartonbasis, komplette Riss-Kartographien auf Leinen, aluminium-kaschierte Papierzeichnungen sowie transparente Zeichenträger als letzte, analoge Innovationsstufe zur Dokumentation bergbaulichen Tuns werden dazu in Schubladen- und Hängeschränken aufbewahrt. Nach dem Ende der Gewinnung wurden alle Archivarien zu einem Standort mit dem Ziel zusammengeführt, hier ein zweckgebundenes Raumangebot zu schaffen, wo für alle Unterlagen eine optimierte Archivierung bereit gestellt wird. Damit soll forthin eine bestmögliche Nutzung der Archivinhalte, z.B. für Recherchen zu Betriebsplanverfahren oder Historien ermöglicht werden. Der Beitrag beschreibt die Planungsphase von der Grundlagenermittlung über die Entwicklung einer Ablagestrategie, die bautechnische Planung, die bauliche Umsetzung sowie die Überführung der Risse und Karten aus der bisherigen Aufbewahrung in die neue Lokation unter Anwendung eines speziellen EDV-gestützten Riss-Finde-Systems
In the year 1751, after the first mining operations were nationalized, mining was continuously practiced in the Saarland coal basin until the year 2012. Since the late 18th century, the miners in the Saar region created many kinds of maps, to document the geology, the mineral deposit and all mining activities. Several thousand mining maps on very difficult drawing papers are keeping in many cupboards with much more drawers. After the extraction all the documents from the individual mines were merged into one site with the aim to create a dedicated room, were the documents can remain with an artful computer-system to locate the accomodated mining maps. Only by this way an optimal use of the archive arias for operating shedules or historical researches is henceforth possible. This article describes the planning phase from the basic evaluation about the development of a storage strategy, the civil engineering planning, the construction implementation as well as the conversion of the mining maps from the previous storage to the new location by using a special computerized mapfinder-system

Abstract:
Human herpesvirus 6A (HHV-6A) has yet to be definitively linked to a specific disease. This is due in part to the ubiquitous nature of the virus. Humanized Rag2-/-γc-/- (Rag-hu) mice were tested to determine if these were a suitable animal model to study the virus. Both cell-free and cell-associated virus was used for infection and both were found to be efficient at infecting the mice. Viral DNA was found in the plasma and cellular blood fractions, bone marrow, lymph node, and thymus, indicating successful infection and propagation of the virus in vivo. The CD3+CD4- population was depleted, while the CD3-CD4+ was increased in infected animals. The CD3-CD4+CD8- and CD3+CD4+CD8- populations were depleted and the CD3+CD4+CD8+ population increased when analysis was gated upon CD4+ cells. The CD3-CD4+CD8+ population expanded and the CD3-CD4+CD8- population was reduced when analysis was gated on the CD3- population. Additional flow cytometry analysis revealed increases in CD4+CD8+ double positive cells in the peripheral blood of cell-free infected mice, which could indicate improper T cell selection and a premature departure of these cells from the thymus, possibly contributing to autoimmunity. Previous research has shown that HIV and HHV-6A may have a synergistic effect on one another and that HHV-6A may act as a cofactor in the progression to AIDS. After determining the Rag-hu mouse model was suitable for studying HHV-6A infection, a coinfection of HHV-6A and HIV-1 was performed. Coinfected mice had fewer thymocytes when compared with the HIV-1 only, mock-infected, and to a lesser extent HHV-6A only groups which could indicate increased cell death in the coinfected group as well as possible disruptions in migration of cells, either causing cells to be sequestered in the bone marrow and unable to migrate to the thymus, or causing premature egress of the cells in the thymus due in part to premature upregulation of CCR7, both of which would explain the smaller cellular populations found in the coinfected mouse thymi. Additional studies were performed to determine if a preferential targeting existed between HHV-6A and HIV-1 as these viruses are found simultaneously coinfecting the same cell. Preferential targeting was not observed by cell-associated migration assay, but increased migration of HHV-6A-infected cells was observed in a CCL21 dependent manner. These studies have provided useful information about HHV-6A and its relevance to HIV/AIDS as well as a possible mechanism of the involvement of HHV-6A in multiple sclerosis (MS) and other autoimmune diseases.

Abstract:
The thesis explores the role of human understanding in R.G. Collingwood's philosophy. I examine four major areas: the role of psychology, the unity of mind (i.e. unity of thoughts and emotions), the role of art and the case of fairy tales as a source of historical knowledge. These themes taken together expound a coherent way to see human understanding: with psychology Collingwood suggests the form that human understanding cannot assume the unity of mind is Collingwood's idea of how we experience an activity, that is, as an undivided whole of emotions and thoughts (and in some respects sensations or feelings too), that exist in every activity as elements that cannot be distinguished or separated. When we come to the forms that an activity can take I argue, using art and fairy tales, that human understanding in Collingwood's system should be seen as a shift from the knowledge of the united spirit (as propounded in Speculum Mentis) to the knowledge of the historical consciousness. The knowledge of the united spirit is achieved through a dialectic scale of the different forms of experience, which individually, in isolation from one another, are not epistemologically valid. In the historical consciousness the forms of experience are epistemologically autonomous and are found within history, all being manifestations of the historical mind.

Abstract:
In spite of the evident centrality of philosophical 'realism' in Collingwood's autobiographical account of his own intellectual development, his critique of 'realism' has hardly been investigated as a central theme in his philosophy. Collingwood's arguments against contemporary 'realism' and his stated move beyond 'idealism' have mostly been treated as a minor question subordinate to other questions. By contrast, I have tried in this thesis to reconstruct Collingwood's philosophy as a critical development of the realism/idealism dispute of his day, focusing on his less known early published and unpublished philosophical writings. This has enabled me to clarify his unique definition of 'realism' in terms of a dualistic framework, and understand his philosophy as an attempt to overcome such dualisms in the realms of philosophy. This approach ultimately highlighted the aim of Collingwood's reform of philosophy as the better understanding of the human mind and action. By employing the 'historical' and 'internal' method of analysis, I firstly illustrated how the idea of 'dualism' became an issue in the realism/idealism dispute as it emerged in early twentieth-century British philosophy. This was followed by a biographical sketch in which I demonstrated that Collingwood's educational background was perfectly equipped to refute the 'realist' philosophy in the dispute. Historically contextualising thus, I chronologically restored the formation of his critique of 'realism' as his attempts to synthesise dualisms in logic (subject/predicate), ontology (abstract/concrete), epistemology (subject/object), and ethics (theory/action) during the period between Religion and Philosophy and An Essay on Philosophical Method. Finally, I argued that Collingwood's critique of 'realism' crystallised in his notion of duty, which embodied his characterisation of philosophy as both 'normative' and 'descriptive'. Throughout, I presented a systematic and sustained picture of Collingwood's early philosophy, identifying and unfolding the fertile implications of his critique of 'realism' for his principal concern with the human mind and action.

This thesis focuses on the experiences of the ‘disorderly’ neighbourhood of Rosemary Lane and Rag Fair in the eastern suburbs of London in c.1690 to 1765. Rosemary Lane and Rag Fair possessed one of the most powerfully articulated reputations for disorder of any London street. In the imagination of both novelists and social investigators it was thought to be squalid, dangerous, dirty - the stereotypical ‘den of iniquity’. Using a wide range of material including parish records, Middlesex session papers, eighteenth century newspapers, and digital sources such as the Old Bailey On-Line and London Lives this thesis will explore the streets and alleyways of Rag Fair. It will go beyond the simple perception of a disorderly neighbourhood, to describe the individual communities and forces which created that disorder. It will show that the poor of Rosemary Lane, generally, did not see themselves as a problem waiting to be solved, they were resourceful and they had their own way of surviving - they were active players in a changing City that was shaking off its medieval roots and embracing the modern. By looking in detail at this community; at its structures and divisions, and at its power relations, its self-identity will be revealed.

Abstract:
Resilience Engineering has evolved during the recent century and could be a good complement to the prevailing ideas concerning safety within the air traffic industry. The concept of Resilience Engineering stresses the fact that in order to keep up the high standard of safety, there must be greater attention directed to the importance of being proactive, and to implement measures before dangerous situations arises. The purpose of our work was to develop the Resilience Analysis Grid (RAG) to help LFV, the leading Air Navigation Service Provider in Sweden, to identify their ability to deal with disturbances and unexpected events. By testing our RAG on seven active air traffic controllers and operational managers, we were able to produce a final set of assertions, with a total number of 22 items, which LFV (or other similar organisations) can use as a foundation for future RAG studies. As a first attempt we also rated the answers which gave us an opportunity to produce a star diagram, showing the relationship between the areas covered by the RAG. During the interviews we discovered that resilience is already today in many aspects a big part of the everyday work and that the RAG method can therefore be applicable in the industry with some modification. However, there are certain areas within LFV that we believe there is room for improvements. We believe that the RAG could serve as a helpful tool in identifying these areas as well as assisting LFV in their striving to remain one of the safest organisations in the world.

This thesis argues that the distinctiveness of contemporary British fashion design can be attributed to the history of education in fashion design in the art schools, while the recent prominence and visibility is the result of the expansion of the fashion media. Fashion design had to struggle to achieve disciplinary status in the art schools. Tarnished by its associations with the gendered and low status practice of the dressmaking tradition, and then in the post war years, with the growth of mass culture and popular culture, fashion educators have emphasised the conceptual basis of fashion design. Young fashion designers graduating from art school and entering the world of work develop an occupational identity closer to that of fine artists. This is a not unrealistic strategy given the limited nature of employment opportunities in the commercial fashion sector. But as small scale cultural entrepreneurs relying on a selfemployed and freelance existence, the designers are thwarted in their ability to maintain a steady income by their lack of knowledge of production, sewing and the dressmaking tradition. The current network of urban `micro-economies' of fashion design are also the outcome of the enterprise culture of the 1980s. Trained to think of themselves primarily as creative individuals the designers are ill-equipped to develop a strategy of collaboration and association through which their activities might become more sustainable. While the fashion media has also played a key role in promoting fashion design since the early 1980s, they are overwhelmingly concerned with circulation figures. They produce fashion images which act as luxurious environments for attracting advertising revenue. Consequently they carry little or no coverage on issues relating to employment or livelihoods in fashion. But their workforce is also creative, casualised and freelance. In each case, these young workers are the product of the shift in the UK to an emergent form of cultural capitalism comprising of low pay and the intensification of labour in exchange for the reward of personal creativity. This current sociological investigation aims to open the debate on the potential for the future socialisation of creative labour.

Abstract:
Bacterial endosymbionts can have profound impacts on their host’s ecology. Notably, endosymbionts can protect their hosts against natural enemies and influence host plant interactions. The endosymbiont Candidatus Arsenophonus infects a wide taxonomic range of arthropod hosts, and is suspected of an uncharacterized mutualistic role in hemipterous insects. In the soybean aphid, Aphis glycines, an introduced pest of soybeans in the United States, Arsenophonus is the sole facultative endosymbiont. The focus of this dissertation is to characterize the role of Arsenophonus in the aphid, with an overall emphasis on its impact on aphid management strategies. I first used diagnostic PCR to determine Arsenophonus infection frequency and strain diversity for native and introduced soybean aphids. I found that Arsenophonus infection is a uniform strain that is highly prevalent in soybean aphid. I then determined if Arsenophonus was a defense symbiont by curing two genotypes of soybean aphid of their natural Arsenophonus infection, resulting in infected and uninfected isolines within the same genetic background. I subjected these isolines to assays with three parasitoid species and a common aphid fungal pathogen, Pandora neoaphidis. I did not find differences in parasitism or fungal infections within the treatments. These results indicate that, although Arsenophonus is widespread, the symbiont should not interfere with biological control efforts. I next examined the influence of Arsenophonus on the ability of soybean aphid “biotypes” to colonize resistant Rag plants. I cured three additional soybean aphid biotypes. All isolines were subjected to growth rate assays on resistant Rag versus susceptible soybean. My results indicate that Arsenophonus infected soybean aphids have an increased population growth compared to uninfected aphids regardless of soybean plant type Finally, I induced soybean plants with jasmonic acid (JA) or salicylic acid (SA) to determine the effective plant defense against soybean aphid feeding. I also used Arsenophonus infected and uninfected aphids to determine any interaction between Arsenophonus and plant defense. I found SA treatment decreased soybean aphid population growth for one experiment, but had no effect when replicated. JA treatment had no effect, and there were no interactions between Arsenophonus infection and plant treatments.

L'industrie du béton en générale, et celle du ciment Portland en particulier, est un acteur important dans la politique environnementale. Le besoin de recourir à l'utilisation des sous-produits industriels afin de réduire la consommation des matières premières semble faire l'unanimité au sein de plusieurs intervenants dans le monde. II existe actuellement sur le marché plusieurs matériaux normalisés qui ont fait leur preuve dans l'amélioration des propriétés des bétons. Toutefois, la disponibilité de ces matériaux n'est pas toujours assurée et il faudra faire appel à des moyens de transport pour acheminer le produit désiré à destination, augmentant ainsi le degré de pollution et le coût. Développer des matériaux locaux et écologiques pour remplacer l'utilisation des matériaux usuels et empêcher leur mise en décharge s'avère une nécessité afín de réduire les contraintes économiques et environnementales. Dans le cadre de la chaire industrielle en valorisation du yerre dans les matériaux avec la Société des Alcools du Québec (SAQ), plusieurs thèmes ont été retenus pour examiner le potentiel d'utilisation du verre dans le béton. Le thème abordé dans cette étude concerne le remplacement partiel et total des granulats naturels par du verre concassé afin de produire des bétons qui présentent une apparence architecturale attirante. Plusieurs taux de remplacements des granulats naturels par des granulats de verre ainsi que différentes combinaisons cimentaires et deux rapports eau/liant ont été examinés. L'étude d'optimisation réalisée a montré que la substitution totale des granulats naturels par le verre de même grosseur offre au béton des critères de performances favorables par rapport aux exigences énoncées dans la norme des éléments de maçonnerie CSA-A165-04, des méthodes d'échantillonnages et des essais des briques CAN3-A82.2-M78 et les méthodes d'essai de la résistance à la compression et du module d'élasticité des prismes de maçonnerie CSA S304.1-04. Les résistances en compression obtenues dans les différents bétons incorporant différents liants composés dépassent les limites de 15 et 20 MPa recommandées par la norme CSA A165.2-04 pour les bétons de qualité I et II. De plus, les bétons contenant des granulats de verre développent de bonnes résistances au gel et dégel, au feu, au choc thermique et à l'écaillage. La présence du verre a bien évidement donnée lieu à une réaction alcali-silice très compromettante à la durabilité des bétons. Toutefois, l'utilisation d'un ciment blanc ayant une faible teneur en alcalis (< 0.15 %) avec d'autres ajouts cimentaires tels que le métakaolin, le laitier ou le verre peut réduire substantiellement les expansions à plus de 50 %. La présence du verte broyé ou concassé façonne la texture des bétons et offre une meilleure esthétique architecturale par la couleur blanchâtre de la poudre et le reflet multicolore des granulats.

Metod För att besvara syftet har statistisk data analyserats med hjälp av ett Pearson Chi2-test. Analysen har skett genom att undersöka födelsemånad, kön och inriktning bland antagna elever vid svenska längdskidgymnasier mellan åren 2010-2015. Resultat Resultatet visar att det finns signifikanta bevis på att relative age effect förkommer på svenska längdskidgymnasier (p= <0,05). Vid uppdelning av respektive utbildningsinriktning kunde det endast konstateras förekomst av relative age effect vid Riksidrottsgymnasierna, vid uppdelning av kön går det inte att säkerställa en signifikant förekomst av relative age effect. Slutsatser Studiens resultat visar att det förekommer relative age effect på svenska längdskidgymnasier, dock varierar den mellan de analyserade undervariablerna. Tydligast förekommer relative age effect vid Riksidrottsgymnasierna, medan det vid Nationell Idottsutbildning, kunde det inte konstateras bevis för en signifikant överrepresentation av elever födda under första kvartalet. När flickor och pojkar analyserades separat kunde ingen relative age effect konstateras. Det behövs mer forskning inom området för att få en överblick av konsekvenserna av relative age effect på längdskidgymnasier men även konsekvenserna för svensk längdskidåkning i stort.
Aim The purpose of this study was to investigate if relative age effect occurs at Swedish cross-country ski schools and if there were a differences in the presence among boys and girls, and between Riksidrottsgymnasier (Country-wide sport academy) and Nationell Idrottsutbildning (National sport academy). Method To answer the aim, statistical data were analyzed using a Pearson Chi2-test. The analysis has been done by examining birth month, sex and education among students at Swedish cross-country ski schools between 2010-2015. Results The result shows that there is a significant evidence of relative age effect at Swedish cross-country ski schools (p= <0,05). When the results were partial between Riksidrottsgymnasier and Nationell Idrottsutbildning, the relative age effect was only seen at Riksidrottsgymnasierna. There were no significant evidence for relative age effect when boys and girls where analyzed separately. Conclusion The results show that relative age effect appears on Swedish cross-country ski schools; however, there were some varies between analyzed variables. Strongest evidence for relative age effect was seen at Riksidrottsgymnaiserna. Further research is needed to get a overview of the consequences of relative age effect at cross-country ski schools, and the consequences it has in Swedish cross-country skiing overall, both for younger and older skiers.

Les Déficits Immunitaires Combinés Sévères (DICS) sont des désordres héréditaires observés dans environ 1 cas sur 75,000 naissances. Les DICS sont caractérisés par une déficience profonde du développement T. Le seul traitement existant à l'heure actuelle pour la plupart de ces patients est la greffe de moelle osseuse. Ce traitement est efficace à 80-90 % lorsqu'un donneur familial HLA-compatible est disponible. Dans les autres cas, le taux de succès de la greffe est d'environ 50 %. Le taux de succès est encore plus faible dans le cas des DICS présentant une absence de T et de B (DICS T-B-NJ+), tels que la déficience en RAG-1 ou RAG-2. L'existence d'un modèle murin de cette pathologie nous a permis d'évaluer l'efficacité d'un traitement alternatif à la greffe de moelle osseuse : la thérapie génique ex-vivo. Tout d'abord, l'effet de la greffe de moelle osseuse sur la reconstitution lymphoi͏̈de des souris déficientes en RAG-1 et RAG-2 a été évaluée sur ce modèle dans plusieurs conditions, et notamment après le traitement systémique des souris traitées avec l'interleukine-7. Le transfert rétroviral du gène RAG-2 humain aux cellules souches hématopoi͏̈étiques de souris déficientes en RAG-2 est effectué à l'aide d'un rétrovirus de Moloney MND-RAG2. Après traitement, 100% des souris traitées ont ainsi reconstitué un compartiment lymphoi͏̈de fonctionnel et diversifié, sans effet toxique observé. Les souris traitées possèdent des taux d'immunoglobulines IgG et IgM proches de la normale. De plus, les splénocytes des souris traitées prolifèrent en présence de mitogène. L'existence d'un avantage sélectif a aussi pu être démontré. Les souris déficientes en RAG-1 traitées avec plusieurs vecteurs rétroviraux (deux vecteurs rétroviraux, MND-RAG1 et MFG-RAG1, et un vecteur lentiviral pWPTS-RAG1) contenant la forme humaine de RAG-1 dans les différents organes, observèrent de faibles taux de reconstitution lymphoi͏̈de et une lymphopénie persistante. Les raisons possibles de cette faible reconstitution ont été recherchées, notamment en quantifiant l'expression du transgène RAG-1 dans les différents organes des souris traitées. Ces résultats suggèrent qu'il existe une valeur seuil d'expression de rAG-1 en dessous de laquelle le transfert de gène est efficace. Nos résultats confirment que la thérapie génique pourrait être une alternative possible à la greffe de moelle osseuse pour les déficits en RAG

The objectives of this research project were to identify and compare factors that influence the purchasing decisions of Multifibres’ customers active in each channel. Multifibres manufactures and distributes industrial cleaning rags to three channels or client categories, these being industrial resellers, industrial end-users and the walk-in customers. An extensive literature review revealed that purchasing decisions are influenced by, amongst other factors, the buyer’s role, the internal cognitive processes of the buyer, as well as factors present in the buyer’s business and external environment. An empirical study was conducted utilising in-depth interviews. The most prevalent, emergent themes that buyers attached the greatest weight to when purchasing cleaning rags were: price; quality; service; relationships; and, convenience. These factors were probed, analysed and compared, based on each buyer category’s unique set of characteristics. When motivating their purchasing preferences, resellers emphasised the importance of the business relationship and trust as being paramount, while end-users viewed price and service as the most important factors. Walk-in customers valued the combination of price and convenience as the most important reasons influencing their purchasing decisions.

Les cancers lymphoïdes présentent fréquemment des aberrations chromosomiques. Dans les lymphocytes, cette instabilité génomique peut découler d'une activité anormale de la recombinase V(D)J (i.e. RAG endonuclease), alors que des facteurs de réponse aux dommages à l'ADN (DDR), tels que les protéines Ataxia-telangiectasia mutated (ATM) et p53, sont connus pour empêcher l'apparition de réarrangements chromosomiques et la lymphomagénèse. Le but de ma thèse fut de tester la contribution relative de ces facteurs dans l'émergence des mutations somatiques dans le génome des lymphomes. Pour ce faire, j'ai réalisé du séquençage génome et transcriptome entier de différents lymphomes, issus de modèles murins génétiquement modifiés pour lesquels les activités RAG et DDR étaient altérées. Lors d'une première étude, j'ai identifié des lésions génomiques spécifiques et récurrentes, causées par l'activité « off-target » de RAG et, de manière plus surprenante, un pattern de réarrangements anormaux survenant en l'absence de RAG. J'ai mis en évidence un mécanisme de « Breakage-Fusion-Bridge », en l'absence de RAG, entrainant l'instabilité et l'amplification d'une région génomique s'étendant sur plusieurs mégabases. Plus encore, j'ai également montré que cette amplification mène à son tour à la surexpression de multiples gènes connus ou candidats dans le cancer et se retrouve dans un sous-groupe de leucémies humaines. Afin de rétablir la différentiation des cellules T, bloquée en l'absence des RAG, j'ai utilisé un autre modèle murin exprimant un récepteur des cellules T (TCR) transgéniques. Ce travail a permis de montrer que le stade du développement et l'activité RAG tous les deux déterminent le paysage génomique des lymphomes T et guident la transformation maligne des cellules à travers différentes voies oncogéniques. De plus, j'ai également réalisé la première étude génome entier de lymphomes T déficients pour ATM et ai identifié un nombre important d'anomalies dans ces tumeurs, présentes aux loci des récepteurs aux antigènes mais aussi à des positions ectopiques. Mes résultats suggèrent, qu'en l'absence d'ATM, les cassures de l'ADN induites par les RAG (non réparées ou résolues de manière anormales) déclenchent une instabilité massive au niveau des loci des récepteurs aux antigènes. Cette instabilité se propage ensuite ailleurs dans le génome et affecte des gènes du cancer. De manière plus générale, ma thèse approfondie la compréhension des mécanismes générant des mutations somatiques dans les cancers lymphoïdes, suite à des défauts de recombinaison V(D)J ou de DDR
Lymphoid cancers frequently harbor chromosomal aberrations. Abnormal V(D)J recombinase (i.e RAG endonuclease) activity is thought to promote genomic instability in lymphocytes, while DNA damage response (DDR) factors such as Ataxia-telangiectasia mutated (ATM) and p53 have been shown to suppress aberrant chromosomal rearrangements and lymphomagenesis. During my thesis, to test the relative contribution of these factors in shaping the pattern of somatic mutations in lymphoma genome, I performed whole genome and transcriptome sequencing of several genetically modified mouse lymphoma models in which the activities of RAG and DDR were perturbed. In a first study, I have identified specific recurrent genomic lesions caused by off-target RAG activity and, more surprisingly, a unique pattern of aberrant rearrangements occurring in the absence of RAG. I provided evidence that, in the absence of RAG, Breakage-Fusion-Bridge triggers instability and amplification of a genomic region of several megabases leading to the overexpression of multiple known and putative cancer genes. Importantly, I also showed that this region is found amplified in a subset of human leukemia. Using additional animal models in which blocked T cell differentiation due to the absence of RAG was rescued by the expression of a transgenic T cell receptor, I could demonstrate that both developmental stage and RAG activity determine T cell lymphoma genome landscapes and mediate malignant transformation through distinct oncogenic paths. In addition, I have established the first genome-wide analysis of ATM-deficient T-cell lymphomas and identified a high number of aberrations localized at antigen receptor loci and ectopic locations in these tumors. My results suggest that, in the absence of ATM, aberrantly resolved RAG-induced DNA breaks at antigen receptor loci trigger massive complex rearrangements spreading to ectopic locations and affecting cancer genes. Overall, my studies provide new insights into the mechanisms of somatic mutations arising in lymphoid cancers in the context of aberrant V(D)J recombination and DDR

They have come to get equipped for the dangers of real-life political action ; to get a politics degree which might make them attractive candidates for civil service positions or for an assistantship at party HQ. They are equipping for the future, they want to know about the future, and they want to be prepared for the currents in which they will soon have to swim: the networks, hierarchies and channels of influence in conjunction with which they will have to operate – present networks, today’s hierarchies and perhaps even tomorrow’s. Yet here they are, engaging not only with today’s political problems, but with yesterday’s, or with those of several centuries past; with Plato’s Republic, Hobbes’s Leviathan, and with a whole cast of authors whose works and words belong, as they soon realise, to the problems of their own time, and seem to offer very little for the solution of today’s. Even on the level of ideology, the challenges of Hobbes, of Rousseau, or of Burke to current thinking are weakened by attendant contexts that are no longer happening, by their ill-suitedness to popular revival, and by their undemocratic obsolescence…

South Africa will be implementing Basel II on 1 January 2008. Basel II provides regulatory capital requirements for credit risk, market risk and operational risk. The purpose of capital requirements is to level the playing field for all internationally active banks and to protect consumers against these risks. Although there is an obvious threat of liquidity risk and it is important to correctly measure and manage liquidity risk, it is almost glaringly omitted from Basel II. The result of not managing liquidity risk properly may have dire consequences for banks because a liquidity crisis may happen without warning. Therefore, the aim of this study was to explore current practices and to propose guidelines for effective liquidity risk regulation in South Africa. A literature study and quantitative analysis on liquidity risk in South Africa were conducted to assess whether it is valid for regulators to require banks to hold capital for liquidity risk. This study provides conclusions and recommendations on the regulatory treatment of liquidity risk in South Africa under Basel II. Although Pillar 2 reviews a variety of other risks and not only liquidity risk, it is proposed that the liquidity risk part of such reviews is conducted on the basis of a questionnaire used to determine possible gaps between banks' practices and prescribed criteria regarding the management and measurement of liquidity risk. It is important to note that such an approach has a constraint in terms of the substantial amount of work that would have to be done on the regulation of liquidity risk by both regulators and banks. Therefore resource constraints and the cost versus the benefit of such an approach would have to be considered carefully. The all-encompassing conclusion to this study is that capital would not be an effective mitigant for liquidity risk for a number of reasons. Liquidity risk differs from bank to bank and a general capital charge for all banks may not be sensible, therefore liquidity risk should be analysed on a bank-by-bank basis. In other words, capital could be charged for liquidity risk under Pillar 2(b) of Basel II. Such a capital charge would not serve the purpose of covering losses resulting from liquidity risk, but would instead impose a penalty on banks that are deemed to manage and measure liquidity risk imprudently. Such a penalty would typically be quite small but would serve as an incentive for banks to improve their management and measurement techniques to the desired level as set out by prescribed criteria. The criteria that should be used for determining whether banks measure and manage liquidity risk prudently should be of such a nature that the Bank Supervision Department (BSD) of the South African Reserve Bank (SARB) complies with Basel Core Principle 14: Liquidity Risk in regulating liquidity risk. In addition, it should align the criteria used to the 14 Principles for the sound management of liquidity as prescribed by the Bank for International Settlements and the Institute of International Finance. Furthermore, it is proposed that the BSD should not prescribe to banks which methods to use to report their liquidity risk, because all banks are not the same in terms of size and sophistication. For this reason, banks should be allowed to follow an internal models approach for liquidity risk whereby banks are, subject to regulatory approval, allowed to use their own internal liquidity risk measures to report liquidity risk to the BSD. This approach is similar to the approach followed by the Bundesbank in Germany. A liquidity risk questionnaire could be drafted according to which banks' liquidity risk management and measurement is assessed in terms of the sound Principles for managing liquidity risk and the Basel Core Principles. One questionnaire could be used for the purposes of assessing the quality of banks' liquidity risk management and measurement in terms of a Supervisory Review and Evaluation Process (SREP) as well as for banks applying for approval of an internal models approach for liquidity risk. The same questionnaire could be used for both purposes, or the questionnaire could be divided into two clear sections whereby all banks are required to answer the SREP (or Pillar 2(b)) section, and only banks applying for the use of an internal models approach for liquidity risk are required to complete this section. A further conclusion to this study is that the BSD should publish a framework in which its approach to regulating liquidity risk is described in detail. Some aspects that should be included in such a document include a widely-accepted definition for liquidity risk and guidelines/minimum standards for measurement and management techniques for liquidity risk and the process that will be followed under Pillar 2 of Basel II. If the BSD is concerned about the level of potential liquidity risk in the South African banking system, it should consider having the additional instruments that are eligible as collateral included as instruments eligible for liquid asset reserve requirements. An additional mitigant for liquidity risk may be that the BSD requires banks to report their liquidity risk more frequently than the current monthly basis.

This is a study of the philosopher Robin George Collingwood. It proceeds along three avenues. First contrary to the received interpretation which sees Collingwood's later work. exemplified by the essay on Metaphysics, as accepting complete historical rclatiyism. I argue that the Metaphysics is better understood within the contcxt ofhcImcneutics. and how careful consideration of his henncneutics of history can havc helpful and Illuminating theological applications. Both 'absolute presuppositions' and the controversial notion of 'unconscious thought' function as a critique of subjectiyism. The second avenue investigates the status of the doctrine of re-enactment. In common with recent research I argue that the doctrine is best understood as a transcendental condition of history. Collingwood is, therefore, clarif)ing the conditions for an understanding of the past, rather than providing the historian \\1th a method. I argue that re-enactmcnt is a 'grammatical' investigation into the nature of the historical object and 1 support this argument by a detailed account of the linguistic nature of Collingwood's philosophy. Following the school of anal~1ical philosophy of history, I argue that re-enactment is a thesis about historical explanation as opposed to explanation by general law s. An argument against the limitation of re-enactment to the bounds of historical explanation fonns the third avenue of research. While the doctrine is certainly an e:xample of historical explanation. there is too much evidence pointing to its henneneutical dimension to leave the issue there. 1 argue that Collingwood anticipates man~ of the themes common to the henneneutics of Hans-Georg Gadarner. and I attempt to systematize these using Gadarner as the yardstick. Abstract:
This thesis argues that Collingwood's philosophy is best understood as a diagnosis of and response to a crisis of Western civilisation. The various and complementary aspects of the crisis of civilisation are explored and Collingwood is demonstrated to be working in the traditions of Romanticism and 'historicism'. In Part One, I examine Collingwood's conception of the crisis of modernity in terms of the Romantic idea of the unity of the forms of experience and his philosophy of art. Contemporary civilisation is seen as corrupted by the suppression of emotion and is to be regenerated by art: the expression of emotion and truthful consciousness. In Part Two, the crisis of civilisation is explained as the failure of contemporary civilisation to significantly move beyond a dependency on a Platonic philosophy of being. The solution, it is proposed, is the development of a philosophy of becoming, which reconciles normative thinking with historical change. The criteria for value and truth are located, not in an ideal transcendent world, but in a 'way of life' in the widest sense. In this respect, Collingwood is compared with Ortega y Gasset and Nietzsche. Part Three demonstrates that Collingwood's dialectical philosophy reveals itself in a historicist phenomenology of mind and a dialectical theory of liberalism and civilisation. This dialectical view leads to a critique of the ill effects of capitalism and of the rationalisation and bureaucratisation of contemporary life. On these subjects, the theories of Collingwood and Ortega y Gasset are contrasted with those of Nietzsche and Weber.

The work deals with physical principles of X-ray generation and development of image during projection and projection reconstruction. A proposal of user’s application in a Matlab – Guide is given, which can be used as a laboratory exercise of the simulation of the projection- and projection image reconstruction. The computer program involves an evaluation of a X-ray quality of CT RTG ZS – quantitative assessment of spatial resolution and as well as the acquisition contrast as a function on an object size. The main aim of the work was the comparison of the acquisition contrast at various acquisition projection and projection-reconstruction parameters. Also, the work is illustrated by some results achieved.

The subject of this thesis is a method proposed for automated evaluation of the parameters of X-ray of cystic disorders in human jawbones. The main problem in medical diagnostic is the low repeatability due to the subjective evaluation of images without using a tool for image processing. In this thesis are described the basic steps of image processing, various methods of image segmentation and chosen segmentation method live-wire. Selected segments were processed in the ImageJ Java environment. In the cystic regions their basic statistical and shape properties were evaluated. The obtained values were used for learning the classification model (decision tree) in the environment RapidMiner. This model was used to create a plug-in for automatic classification of the type of cysts in the program ImageJ.

The aim of the thesis is to design and implement a simulator of image reconstruction during x-ray computed tomography fluoroscopy. Apart from quantitative evaluation of particular imaging process parameters influence, the intended program application will lie in optimization of these parameter values. Introductory part is focused on brief theoretical description of x-ray computed tomography imaging process. Emphasis is placed on fluoroscopy-specific approaches, particularly the division of scanned projections into chosen amount of sectors as well as fluoroscopic imaging process evaluation methods. The subsequent part deals with program implementation and its limitations and sketches the possible working framework. Moreover, one of the chapters is devoted to optimization of imaging process parameters measurement. The final part aims at impact analysis of particular process parameters as well as fluoroscopic imaging process optimization approaches. The thesis consists also of a didactic simulator enabling real-time intervention simulation with manual instrument manipulation. Since both simulators are intended for teaching purposes, the thesis is supplemented with a laboratory exercise draft.

Syftet med vår studie är att undersöka vilken betydelse den fysiska inomhusmiljön har i det pedagogiska arbetet, med de yngsta barnen som utgångspunkt. Med inspiration från fenomenografin har vi genomfört kvalitativa intervjuer med åtta förskollärare för att synliggöra deras uppfattningar och erfarenheter av ovan nämnda fenomen. De frågeställningar som vi i studien söker besvara är: Hur planerar, genomför och utvärderar förskollärare arbetet med den fysiska inomhusmiljön för de yngsta barnen? Vad kännetecknar en god fysisk inomhusmiljö för de yngsta barnen? Vilka hinder och utmaningar finns vid utformningen av den fysiska inomhusmiljön för de yngsta barnen? Resultatet av studien visar att förskollärarna planerar den fysiska inomhusmiljön med barngruppen som utgångspunkt, gör förändringar utifrån barnens uttryckta intressen samt utvärderar kontinuerligt i arbetslaget. En god fysisk inomhusmiljö för de yngsta barnen kännetecknas dels i termer av trygghet, dels av utmaningar. De hinder och utmaningar som förskollärarna uppfattar handlar främst om ramfaktorer, vilket material som går att ha tillgängligt, svårigheter i att tolka de yngsta barnens uttryck samt att hitta lämpliga utmaningar för dessa barn. Av resultatet dras slutsatsen att förskollärarna visar på ett professionellt förhållningssätt när det handlar om barnens inflytande samt att det finns en medvetenhet om den fysiska inomhusmiljön.
