Abstract:
The purpose of this study was to evaluate the potential of micelle to change the pharmacokinetics of quercetin (QUT), with a primary goal of enhancing its oral bioavailability. QUT-loaded methoxy poly(ethylene glycol)-b-poly(L-lactic acid) micelle (QUT-loaded MPEG-b-PLLA micelle) was prepared by a thin-film hydration method, resulting in a particle size of 88.5 nm. A liquid chromatography tandem-mass spectrometry (LC-MS/MS) method was developed and validated for determination of QUT in rat plasma. The chromatographic separation was performed on an Agilent Eclipse-C18 (4.6 mm × 50 mm, 3.5 μm) with an isocratic mobile phase system consisting of water and methanol (30:70, v/v) at a flow rate of 0.4 mL/min. Calibration curves were linear over the concentration ranges of 2.5–2000 ng/mL for QUT. The micelle was orally administered at a single does in rats, and the pharmacokinetic parameters were evaluated and compared with that administered with the QUT aqueous suspension. The results show that the micelle was able to increase the QUT’s oral bioavailability 9-fold compared to the QUT aqueous suspension. These results suggest that methoxy poly(ethylene glycol)-b-poly(L-lactic acid) is a potential carrier for the oral delivery of QUT.

Abstract:
The three enzymes necessary to catabolize quinate to protocatechuate are inducible by quinic acid, and transcription of their corresponding genes is controlled by the action of a positively acting activator gene and a negatively acting repressor gene. Transformed strains of Aspergillus nidulans containing multiple copies of the activator gene (QUTA) but single copies of the other QUT genes retain normal regulation of the gene cluster and do not show any overexpression of the three quinic acid catabolic enzymes. Transformed strains containing equal multiple copies of the activator gene (QUTA) and QUTE (encoding catabolic 3-dehydroquinase), but single copies of the other QUT genes, retain normal regulation of the QUT gene cluster, but selectively overexpress the QUTE gene upon quinic acid induction. Data are presented that strongly suggested that the gene QUTG, which is physically located within the QUT gene cluster and for which no function has been identified, is not required for expression of the gene cluster and does not encode a chlorogenic acid esterase.

Abstract:
In 2002, a project team was established at the Queensland University of Technology (QUT) to build and implement an e-portfolio for each of the 40,000 students. Its success depended on adopting a systemic approach where access was made available to students as a routine part of their life at QUT. The project team adopted a comprehensive model, so the system could meet a variety of purposes and audience needs and it was customised to integrate with existing QUT systems. Business requirements were commissioned and the project was implemented in manageable steps interspersed with trials and feedback. The underlying philosophy, decisions made, steps taken, feedback and future developments are outlined.

Abstract:
Purpose The purpose of this paper is to measure the prevalence, evaluate the trend and identify the factors associated with the non-performance of qualitative urine test (QUT) among pregnant women living in the extreme south of Brazil between 2007 and 2016. Design/methodology/approach All births occurred in the local maternity wards from January 1 to December 31 of 2007, 2010, 2013 and 2016. Mothers were interviewed within 48h after delivery. The outcome was the non-performance of QUT during pregnancy. χ2 test was used to compare proportions and Poisson regression with robust variance adjustment for the multivariate analysis. The effect measure used was the prevalence ratio. Findings Of the 10,331 new mothers identified, 10,004 (96.8 percent) performed at least one prenatal visit. The prevalence of non-performance of QUT was 3.3 percent (95% CI 2.9–3.7 percent), ranging from 1.5 percent in 2007 to 5.3 percent in 2016 (p<0.001). The analysis showed that not living with a companion, having under four years of schooling, living with seven or more people in the household, having five or more children, having had one to three prenatal visits and not having been supplemented with ferrous sulfate during pregnancy showed a significantly higher prevalence rate to the non-performance of QUT. Originality/value The rate of non-performance of this test among pregnant women has clearly increased. Mothers at higher risk of unfavorable outcomes in pregnancy were the ones with the highest probability of not performing QUT. Increasing the number of prenatal visits is a high-impact measure toward the performance of this test.

Abstract:
QUTR (qutR-encoded transcription-repressing protein) is a multi-domain repressor protein active in the signal-transduction pathway that regulates transcription of the quinic acid utilization (qut) gene cluster in Aspergillus nidulans. In the presence of quinate, production of mRNA from the eight genes of the qut pathway is stimulated by the activator protein QUTA (qutA-encoded transcription-activating protein). Mutations in the qutR gene alter QUTR function such that the transcription of the qut gene cluster is permanently on (constitutive phenotype) or is insensitive to the presence of quinate (super-repressed phenotype). These mutant phenotypes imply that the QUTR protein plays a key role in signal recognition and transduction, and we have used deletion analysis to determine which regions of the QUTR protein are involved in these functions. We show that the QUTR protein recognizes and binds to the QUTA protein in vitro and that the N-terminal 88 amino acids of QUTR are sufficient to inactivate QUTA function in vivo. Deletion analysis and domain-swap experiments imply that the two C-terminal domains of QUTR are mainly involved in signal recognition.

Abstract. Elevated particle number concentration (PNC) observed during nucleation events could make a significant contribution to the total particle load and thus air pollution in urban environments. Therefore, a field measurement study of PNC was conducted to investigate the temporal and spatial variations of PNC within the urban airshed of Brisbane, Australia. PNC was monitored at urban (QUT), roadside (WOO) and semi-urban (ROC) areas around the Brisbane region during 2009. The results showed that morning traffic exhaust emissions were the main contributor to high PNCs at QUT and WOO which contributed 5.5% and 5.1 5 during the week, respectively, with a less significant contribution on weekends. PNC peaks were observed around noon, which correlated with the highest solar radiation levels at all three stations, thus suggesting that high PNC levels were likely to be associated with new particle formation caused by photochemical reactions. Wind rose plots showed relatively higher PNC for the NE direction, which was associated with industrial pollution, accounting for 12%, 9% and 14% of overall PNC at QUT, WOO and ROC, respectively. Although there was no significant correlation between PNC at each station, the variation of PNC was well correlated among three stations during regional nucleation events. In addition, PNC at ROC was significantly influenced by upwind urban pollution during the nucleation burst events, with the average enrichment factor of 15.4. This study provides an insight into the influence of regional nucleation events on PNC in the Brisbane region and is the first study to quantify the effect of urban pollution on semi-urban PNC through the nucleation events.

Abstract:
Oxidative stress caused by generation of free radicals and related reactive oxygen species (ROS) at the sites of deposition has been proposed as a mechanism for many of the adverse health outcomes associated with exposure to particulate matter (PM). Recently, a new profluorescent nitroxide molecular probe (BPEAnit) developed at QUT was applied in an entirely novel, rapid and non-cell based assay for assessing the oxidative potential of particles (i.e. potential of particles to induce oxidative stress). The technique was applied on particles produced by several combustion sources, namely cigarette smoke, diesel exhaust and wood smoke. One of the main findings from the initial studies undertaken at QUT was that the oxidative potential per PM mass significantly varies for different combustion sources as well as the type of fuel used and combustion conditions. However, possibly the most important finding from our studies was that there was a strong correlation between the organic fraction of particles and the oxidative potential measured by the PFN assay, which clearly highlights the importance of organic species in particle-induced toxicity.

Abstract:
Classification of fish image is a complex issue in the field of pattern recognition. Fish classification is a complicated task. Physical shape, size, orientation etc. made it complex to classify. Selection of appropriate feature is also a great issue in image classification. Classification of fish image is very important in fishing service and agricultural field, fish industry, survey applications of fisheries and in other related area. For the assessment and counting of fishes, classification of fish image is also necessary as it can save time. This paper presents a fish image classification method with the robust Gist feature and Gray Level Co-occurrence Matrix (GLCM) feature. Noise removal and resizing of image is applied as pre-processing task. Gist and GLCM feature are combined to make a better feature matrix. Features are also tested separately. But combined feature vector performs better than individual. Classification is made on ten types of raw images of fish from two datasets -QUT and F4K dataset. The feature set is trained with different machine learning models. Among them, XgBoost performs with 90.2% and 98.08% accuracy for QUT and F4K dataset respectively.

The aim of this study was to compare the usability of this new app with a clinical ISy device as measured by effectiveness, efficiency and satisfaction.MethodsIn this mixed-methods randomised usability study, healthy volunteers (aged 39.2±12.2 years, n=24) compared inspirations using the QUT Inspire app and a Triflo II clinical ISy device. A post-test questionnaire and a semi-structured interview explored dimensions of usability regarding the new app.ResultsThe duration of inspirations performed using the QUT Inspire app (7.3±2.0 s) were comparable with use of the Triflo II ISy device (7.5±2.3 s). No artefacts arising from the order of device testing were identified. App users held their phones adjacent but not proximal to their mouths (13.6±6.4 cm), notwithstanding instructions to keep the phone less than 5 cm away for optimal breath sound detection. The use of onscreen text or video instructional materials did not result in a significant reduction in this distance. Participants reported clear preferences for the app (100%, n=24) to motivate persistence with repeated inspirations. App gamification features such as a timer (75%, n=18) and breath counter (83.3%, n=20) were well regarded. Analysis of semi-structured interviews identified four main themes arising from this study: visual reward from responsive app animations, clinical look and feel influencing credibility, perceived effort affecting engagement and selective adoption of gamification features.ConclusionThis study demonstrates that a virtual ISy app can be effective, efficient and have high satisfaction. Improvements informed by this research include use of additional phone sensors to optimise sound detection and minimising the distance that phones are held from the user’s mouth. Further research in randomised controlled trials are needed to evaluate performance of this app in clinical contexts where ISy is currently employed.

Abstract:
A major challenge for Arrow Energy has been balancing rapid growth while continuing to operate as an effective business. To meet this challenge, this extended abstract looks at one of the strategies: a leadership program designed in co-operation with Queensland University of Technology (QUT). The leadership program aims to establish a leadership culture that drives company performance and facilitates change, develops leadership networks to support Arrow's business objectives, and strengthens the talent pool to help in succession planning through commitment to individual development. Two programs were custom designed by QUT and launched in 2012: one for senior leadership and the other for emerging leaders at Arrow. Careful participant selection was vital to the program's success. Both programs were designed to provide participants with a highly experiential learning approach, far removed from traditional training programs they had previously attended. Participants were immersed in exploring real issues and challenges facing the company, as well as looking at how well their leadership approach fitted the needs of a rapidly growing organisation. The course design and use of innovative learning experiences have made the program successful. An important measure of this success has been the leadership development of each individual and the establishment of strong informal networks that are part of a mature organisation. Transfer of learning to the workplace was achieved by immersion in team-based leadership project initiatives, which reinforced the major theme of the program: leadership is a journey that requires sustained focus and development.

"Characterization of the 3-dehydroquinase domain of the pentafunctional AROM protein, and the quinate dehydrogenase from Aspergillus nidulans, and the overproduction of the type II 3-dehydroquinase from neurospora crassa." Full text
Abstract:
The AROM protein of Aspergillus nidulans is a multidomain pentafunctional polypeptide that is active as a dimer and catalyses steps 2-6 in the prechorismate section of the shikimate pathway. The three C-terminal domains (including the type I 3-dehydroquinase) of the AROM protein are homologous with the qutR-encoded QUTR protein that represses transcription of the eight genes comprising the quinic acid utilization (qut) gene cluster, and the two N-terminal domains are homologous with the qutA-encoded QUTA protein that transcribes the qut genes. As part of a larger research programme designed to compare the structures of the three proteins and to probe the domain structure and interaction within each protein, we have overproduced and purified the 3-dehydroquinase domain of the AROM protein. Additionally we have overproduced and purified the qutB-encoded quinate dehydrogenase and overproduced the qa-2 encoded type II 3-dehydroquinase of Neurospora crassa. We report that the AROM 3-dehydroquinase domain has a monomeric native state, with an apparent kcat./Km ratio that is approx. 160-fold lower than the value for the native N. crassa AROM protein. The AROM protein 3-dehydroquinase domain is sensitive to inactivation by borohydride in the presence of the substrate 3-dehydroquinate, confirming that it is a typical type I 3-dehydroquinase. The purified quinate dehydrogenase is bifunctional, being able to metabolize shikimate as a substrate. The apparent Km values for quinate (450 microM), shikimate (1.7 mM) and NAD+ (150 microM) are all similar to values reported for the qa-3-encoded enzyme from N. crassa.

Abstract:
To determine the constancy of uteroplacental O2 uptake (VO2) during uterine quiescence and both spontaneous and oxytocin-induced uterine contractions, we have developed a method to measure VO2 continuously and reproducibly. In seven ewes during uterine quiescence, total uterine blood flow (Qut) averaged 200 ml X min-1 X kg uterine contents-1, with intra-animal SD of 18 min X min-1 X kg-1 and interanimal SD of 86 ml X min-1 X kg-1. Uterine arteriovenous O2 content difference averaged 4.2 +/- 0.5 ml X dl-1. VO2 averaged 8.2 +/- 1.2 ml X min-1 X kg-1, with interanimal SD of 3.6 ml X min-1 X kg-1. During 21 spontaneous prelabor contractions, VO2 decreased 3.8% to 8.7 +/- 0.3 ml X min-1 X kg-1 (NS) during the contraction and subsequently increased 12.3% to 10.1 +/- 0.3 ml X min-1 X kg-1 (P less than 0.05) near the end of the contraction. With oxytocin-induced contractions, VO2 fell 10% to 6.4 +/- 0.2 ml X min-1 X kg-1 at the contraction onset (P less than 0.05) but by the end of the contraction had increased 6.1% to 7.6 +/- 0.2 ml X min-1 X kg-1 (NS). In conclusion, uteroplacental VO2 was measured continuously and reproducibly using relatively straightforward methodology; uteroplacental blood flow, arteriovenous O2 content difference, and uteroplacental O2 uptake were normally distributed with coefficients of variation less than 15%; and during both spontaneous and oxytocin-induced uterine contractions Qut decreased significantly, while VO2 initially decreased moderately and then showed an overshoot, during the contraction.

The feasibility of real-time calculation of parameters for an internal combustion engine via reconfigurable hardware implementation is investigated as an alternative to software computation. A detailed in-hardware field programmable gate array (FPGA)-based design is developed and evaluated using input crank angle and in-cylinder pressure data from fully instrumented diesel engines in the QUT Biofuel Engine Research Facility (BERF). Results indicate the feasibility of employing a hardware-based implementation for real-time processing for speeds comparable to the data sampling rate currently used in the facility, with acceptably low level of discrepancies between hardware and software-based calculation of key engine parameters.

Abstract:
Cameron, Verhoeven and Court have noted that many screen producers do not see their tertiary education as being beneficial to their careers. We hypothesise that universities traditionally have not trained students in producing skills because of the division of labour between arts and business faculties, and because their focus on art rather than entertainment has downplayed the importance of producing. This article presents a Scholarship of Teaching and Learning (SOTL) whole-of-program evaluation of a new cross-faculty Bachelor of Entertainment Industries degree at QUT, devoted to providing students with graduate attributes for producing, including creative skills (understanding story, the aesthetics of entertainment, etc.), business skills (business models, finance, marketing, etc.) and legal skills (contracts, copyright, etc.). Stakeholder evaluations suggest that entertainment producers are highly supportive of this new course.

Abstract:
This special issue is the product of a workshop on innovations in policing and preventing gender violence in the Global South, hosted by Queensland University of Technology (QUT) Centre for Justice 3-4 December 2019. The event was attended by scholars from Brazil, Pacific Island communities, Bangladesh, Argentina, and several Australian jurisdictions. Hence the articles in this special issue reflect the diverse nationalities present at the event. A central aim of the workshop realised in this special issue is the stimulation of innovation in understanding the policing and prevention of gender violence through novel international collaborations and cross-fertilization. It reverses the assumptions that underpin the epistemic injustice of the social sciences, that innovations generally flow only from the Global North to the Global South. This special issue shows that it can be the other way round.

Abstract:
This study investigated the impact of Queensland University of Technology (QUT) student engagement with co-curricular services in 2020. With limited institutional data, it is often not possible to gauge the cumulative impact of all retention and teaching activities, and while any one support activity cannot carry the weight of improving student outcomes, it is important to consider the impact of each as a contributing factor. We asked ourselves: How exactly do we determine how students’ connection to services is impacting learning? How do they connect to wider university expectations such as widening participation initiatives or retention activities? And how do we use evaluative strategies productively to mature our service design so that we can move beyond “satisfaction” to “impact”? Despite methodological constraints, converging lines of evidence indicate that engagement with services in 2020 had a small, but statistically significant, positive impact on grade point average (GPA), progression, and retention.
