Full textMacrophages are functionally polarized into inflammatory and microbicidal (M1) and immunoregulatory (M2) cells. If circulating monocytes may be polarized is not known. We determined the transcriptional signatures of human monocytes stimulated with IFN-γ and IL-4, known to induce the polarization of macrophages into M1 and M2 cells, respectively, using microarrays and real-time RT-PCR. We found that monocytes exhibited an early pattern of activation specific to IFN-γ or IL-4 and a late pattern of activation common to both agonists. The selected biomarkers of early and late responses were tested in patients with Q fever. We showed that the kinetic model of monocyte activation enables a dynamic approach for the evaluation of patients with acute Q fever or Q fever endocarditis. On the other hand, it is known that the prevalence of Q fever is related to sex and is higher in men than in women. Based on previous studies on an experimental model of infection by Coxiella burnetii, the agent of Q fever, we hypothesized that circadian genes are differently modulated in men and women with Q fever. We showed that the expression of the Per2 gene was significantly increased in males with acute Q fever compared with healthy volunteers but did not differ in females with Q fever and healthy females. These results suggest that that the modulation of circadian genes is associated with a human infectious disease. We also found that the expression of LNX1 and LNX2 genes that encode two enzymes involved in protein degradation is increased in Q fever endocarditis but not in acute Q fever

Full textIn pregnant women Q fever presents obstetrical complications. The principal way of human contamination by Coxiella burnetii, the agent of Q fever, is due to aerosols from placentas of infected animals. The nature of placenta cells that are targeted by C. burnetii remains unknown. I showed that C. burnetii infects BeWo and JEG trophoblastic cells and that organisms intensively replicated in BeWo cells and survived in JEG cells. A microarray analysis showed that C. burnetii induced a specific inflammatory response in BeWo cells. These results suggest that trophoblasts may serve as a reservoir for C. burnetii. Placenta macrophages placentaires may also targeted by C. burnetii. I showed that placenta CD14+ macrophages were characterized by phenotypic, transcriptional and functional properties different from those of circulating monocytes and monocyte-derived macrophages. In addition, placenta CD14+ macrophages differentiate into multinucleated giant cells that may regulate the cytolytic activity of macrophages in the placenta context since placenta CD14+ macrophages were not polarized in M1 or M2 macrophages. While M1/M2 polarization of macrophages is well established, that of monocytes remains an important question. We activated monocytes with canonical agonists of M1 and M2 profiles in macrophages using microarrays. The early response, 6 hours, of monocytes corresponded to a type M1/M2 response but the delayed response, 18 hours, did not correspond to the M1/M2 dichotomy, demonstrating a new level of heterogeneity of myeloid cells

Full textIn this thesis, we are interesting in Integer-Valued Polynomials on a subset E of a discrete valuation domain V and we compute generalized factorials as defined by Bhargava. In a the first part, we study what we call Legendre subsets, whose factorial sequence satisfies a similar Legendre formula. We prove, by using v-orderings (introduced by Bhargava) and very well distributed sequences (introduces by Amice), that the precompact regular subsets are the only Legendre subsets. We then extend a Conrad's work about normal bases of continuous functions by replacing his construction for continuous functions on V by continuous functions on a subset E of V. Denoting by qn the number of classes of E modulo Mn (where M is the maximal ideal of V ), we prove that when qn divides qn+1, a necessary and sufficient condition to obtain normal bases by qn-digit expansion is that the subset E is a Legendre subset. In the general case, we characterize all the subsets E of V such that the qn-digit principle works. We give an example of normal bases of integer-valued polynomials, obtained by qn-digit expansion. Finally, we extend the notion of factorial of a subset E of V d. First, we give necessary conditions for this work, based on a Mulay's article. Second, we generalize the notion of v-ordering and construct a basis of the V -module of integer-valued polynomials in several indeterminates, linked with the v-ordering. Finally, we give a new definition for the notion of factorial and we give examples

Full textThe main objective of this doctoral thesis is the analysis of the role of a regional daily newspaper as a local social actor within a particular society: Reunion Island. More precisely, the present work is a monograph on le "Quotidien de la Réunion", in the context of a 21 year period (1976-1997). Within the general theoretical framework which has been adopted in order to make this case study, one is interested in the questions of the + building up; of events and news and of how newspapers deal with social movements and actions. In order to confirm the hypothesis that a local paper does fulfil a role of social actor facing social movements, we have applied the methods of discourse analysis to journalistic forms. This methodology may be regarded as essentially inspired by semiotics. We have analysed a corpus of articles and compared the accounts (by "Le Quotidien") of three movements : (I) the strong desire to save the + young ; "Quotidien" in 1977, (II) the support given to Télé Freedom in 1991 and (III) the demonstration of civil servants and students in 1997. The analysis of the argumentative and rhetorical processes which are specific features of the paper's editorial strategy permits us to ascertain its value system and its positions on the local scene, and also to emphasize certain contradictions and discrepancies between its editorial principles and its journalistic practices

Full textThe concept of the business ecosystem, introduced by James Moore from 1993, and Industry 4.0, which emerged in Germany in 2011, together form a phenomenon that has not yet been fully explored. These ecosystems are multidimensional, as heterogeneous elements connect to form complex sets that are difficult to analyze, and we have not identified any tools for studying them without an a priori model. To overcome this gap, we propose a modeling and analysis method using digital traces of the phenomenon to generate a simplicial complex. This method mainly uses Atkin's Q-analysis, which we have modernized by developing a graph version of the Q-analysis algorithm and pushing back the limits we have identified. The whole process is automated within the Hype calculation application. The main advantage of Q-analysis is its ability to calculate similarity between heterogeneous elements, with respect to dimensions that differ from one another, and without knowing the model of the phenomenon. We use Q-analysis to obtain business ecosystems candidates related to Industry 4.0 in the Grand Est region whose final objective is to help analysts understand how an ecosystem emerges or disappears, and thus facilitate their creation around specific topics. Thanks to a database that we have built up from the digital data, we have obtained encouraging preliminary results despite the poor quality of the dataset. We believe that by improving the quality of the data with richer dimensions, we will obtain even more convincing results

Full textThis thesis is part of a research program on Conformal field theory and representations of Lie algebra of dollar\frak{sl}_2dollar (real, complex, dollarpdollar-adic, dollarqdollar-derfomed). We study a real, dollarpdollar-adic and dollarqdollar-deformed versions of a triple integral appeared in physics in connection with the Liouville model of the Conformal field theory. These integrals turn out to be connected with the Dyson-Macdonald constant term identities. We also give another approach to compute the complex case by using Bernstein-Reznikov's technique. The main idea is to apply invariant functionals on principal series representations of dollarG=SL(2,\mathbb C)dollar. Finally, one defines a dollarqdollar-deformation of Jacquet-Langlands principal series representations of dollarGL_2(\mathbb R)dollar and prove the uniqueness of an invariant triple functional on them by using method of H. Y. Loke. Alongside, we find out some relations to similar differential equations in [NSU]

Full textThe epidemic risk for legionellosis is very serious, indeed 2,000 to 3,000 cases ofthis illness are counted each year in France; JO to 15% ofthem are fatal. This disease is due to the bacteria from the Legionella genus, which includes 47 different species, but Legionella pneumophila sg 1 is responsible for 90% of the infection. To reduce contamination risks, a steady follow-up ofwater network is essential. That is why, the A. F. NOR. Has published in April 2006 a standard project on the « Detection and quantification of Legionella and/or L. Pneumophila by concentration and genic amplification by polymerase chain reaction (PCR) ». It questions the CUITent carrying out of water analysis. According to the cUITently used standard on detection and quantification of Legionellae by filtration and culture, 10 days are needed to have final results. This analysis time can be significantly reduced by coupling the quantitative PCR (q-PCR), developed in the new standard, wih MALDI-ToF mass spectrometry. Analyses will require two steps : 1- A quick screening of bacteria in the sample by MALDI- ToF mass spectrometry. This technique is efficient and fast for the detection and identification ofbacteria. Lndeed, whole bacterial cells are analyzed without Iysis step 2- If Legionella are detected, their quantification is carried out in general and more particularly for L. Pneumophila by q-PCR. Two probes are used, one specific for Legionella spp. And the other for L. Pneumophila. This step allows the quantification of these two kinds of species. The combination of these two techniques is particularly interesting in the cost and time-consuming aspects

Full textThe development of analytical tools for organic geochemistry analysis has increased these past years. This development has allowed answering many questions about organic matter composition. However, many issues remain to be clarified including the characterization of high molecular weight fractions and monitoring the reactivity of organic matter. This thesis has focused on both (i) existing method improvements for fossil organic geochemistry analysis but also on (ii) developing a new type of coupling between the size exclusion chromatography (SEC) and the APPI-QTOF mass spectrometry for high molecular weight weakly polar fractions. Adjustments on APPI-QTOF mass spectrometry have allowed a better understanding of polyaromatic organic contaminant reactivity in presence of mineral matrices. The success of this coupling has allowed a better understanding of the structure of asphaltenes. However despite the "simplification" obtained by the SEC, the large amount of information remains difficult to interpret and time-consuming. A mathematical model has been developed based on numerical and statistical analysis of mass spectra, allowing direct comparison of mass spectra and being able to identify several types of information such as origins of samples, monitoring of physico-chemical processes and also the efficiency of soil recovery treatments as well as the identification of analytical protocols

Full textThe main objective of this work was to develop non-targeted metabolomics analyses of bottled wines in order to decipher chemical informations from the time-related evolution of their composition. This original research was based on the hypothesis that, when analyzed, bottled wines would still hold chemical memories of envionmental parameters (vineyard management, oenological practices, climate, terroir…) at the moment of their elaboration, even after several years of ageing. A second hypothesis was that in order to anticipate the future evolution of the wine quality in terms of chemical composition, it is necessary to know what it was in the past. To that purpose, and for the first time in wine science, Fourier Transform Ion Cyclotron Resonance – Mass Spectrometry (FTICR-MS), Liquid Chromatography coupled with mass spectrometry (UPLC-Q-ToF-MS), Excitation Emission Matrix Fluorescence (EEMF) and multivariate statistics were used in combination. Methodological develoments revealed the advantage of coupling exact mass measurements by FTICR-MS to isomeric discrimination by UPLC-Q-ToF-MS in order to extend the range of detectable metabolites. Such tools were applied to the identification of ageing markers in vertical series of red and white wines from Burgundy, including very old wines (unknown vintages) considered as evolution end points, thus introducing the concept of verticalomics. The characterization of series of white wines from Burgundy (Chardonnay) revealed that chemical spaces specifically related to eonological practices (SO2 addition at pressing, settling level, and permeability of the stopper) could indeed be deciphered although the vintage signatures were confirmed to be the most significant. Similar experiments on Champagne wines (Chardonnay, and blends of Chardonnay, Pinot noir and Pinot Meunier) after the "prise de mousse" and the ageing "sur lattes" further highlighted the hormesis effect associated with the oxygenation of wine. Finally, non-targeted analyses of series of grape extracts and corresponding wines from different appelations – though elaborated by the same winemaker – revealed that terroir-related signatures could be indeed read in wines, in particular after a few years of bottle ageing. Altogether our results provide an unprecedented comprehensive description of the chemical composition of wine and its modification through ageing

Full textThis thesis studies the asymptotic behaviors for Markov processes conditioned not to hit moving boundaries. The first chapter deals with this problem for discrete-time Markov chains defined on finite state space considering periodic boundaries. Even if the notions of quasi-stationary distributions and quasi-limiting distributions are not well-defined considering moving boundaries, the existence of a quasi-ergodic distribution and the Q-process are shown. In the second chapter, the previous results are extended to Markov processes satisfying some global inhomogeneous conditions introduced by N. Champagnat and D. Villemonais. In the periodic case, the existence and uniqueness of a quasi-ergodic distribution are proved. When the boundary stabilizes at infinity, we obtain moreover the existence and uniqueness of a quasi-limiting distribution. The third chapter deals with the quasi-stationarity for the "renormalized" Brownian motion absorbed at {-1,1}. The law of this process depends on a parameter K and a phase transition is observed for its quasi-stationarity, whose the critical parameter is equal to 1/2. Finally, the last chapter extend the results obtained in the second chapter to Markov processes satisfying some criteria weaker than the global Champagnat-Villemonais conditions. In particular, we obtain under these conditions a mixing property, the existence of the Q-process and the existence of a quasi-ergodic distribution for some behaviors of the boundary

Full textHybridisation is a complex process with diverse consequences on species evolution. Hybridisation is difficult to study when species are not clearly morphologically distinguished. Our aim was to study the role of hybridisation in the evolution of the European white oak species complex. We used population genetic tools to quantify contemporary interspecific gene flow and to study the mating system of four oak species. A fast genetic analysis protocol was developed and genetic assignment methods were first tested by simulation and then used to determine the species of each tree and to identify hybrids. These methods revealed that hybrid percentages were between 10 to 30% depending on the natural population studied and that all species pairs were involved. We showed that the census number of species in the stands had an influence on hybridisation dynamics and on introgression direction. We studied the mating system of these species in controlled crosses and in the forest to understand the maintenance of species despite interspecific gene flow. Several reproductive barriers contribute to a partial isolation of species, mostly pre-reproductive and prezygotic. A paternity analysis of maternal progenies sampled in the forest showed that first generation hybridisation was rare but that F1 hybrids were fertile and were mating mostly with one of the two parental species, creating numerous backcrosses that explain the high percentages of hybrids observed in the natural populations studied. Hybridisation and introgression are active processes in oaks and contribute to the evolution of the species complex

Full textThe intermetallic compounds of the RCu2 series (R = rare earths) show a large variety of magnetic structures depending on temperature and external field, mainly caused by the interplay of an indirect exchange interaction and the anisotropy of the crystalline electric field. The RCu2 compounds crystallize in the CeCu2 structure, which can be described as an orthorhombic distortion of the hexagonal AlB2 structure. The aim of the present work is the investigation of RCu2 compounds by using resonant and nonresonant magnetic x-ray scattering, supplemented by neutron scattering. Because of the complementarity of magnetic x-ray and neutron scattering this investigation reveals new details of the magnetic structures of the studied compounds. Magnetic properties and magnetic structures are investigated as well as magnetic phase transitions and magneto-elastic coupling in the pseudohexagonal compounds. The correlation between the crystallographic structure and the magnetic structures is studied for different rare earths (R = Nd, Gd, Tb, Dy) but also for partial substitution of magnetic rare earths by magnetic or nonmagnetic ions on the rare earth site (Tb(1-x)DyxCu2, Tb(1-x)PrxCu2, Dy(1-x)YxCu2). Resonant magnetic x-ray scattering is the only method available to investigate the element specific magnetism in crystals with different magnetic ions. By the study of the Tb resonance and the Dy resonance on Tb0.5Dy0.5Cu2 an unexpected developement of the magnetic structures with temperature is observed. The results of supplementary neutron scattering experiments are presented and analyzed by Rietveld refinement of the crystallographic and magnetic structures, mainly focussed on TbCu2. In summary, new insights into the different magnetic structures of RCu2 compounds are gained. The strong magneto-elastic coupling is studied by different scattering methods applied to this problem for the first time

Full textHair fiber is a sophisticated biological material for which the details of its molecular structure failed to be well understood because of the lack of data regarding protein expression and organization. In this context, improvement on human hair cells knowledge with proteomic approaches has been performed in this PhD. This work has been structured in four parts : establishment of the knowledge on hair biology and its molecular structure obtained by literature mining. Development of a well-suited proteomic strategy for the special case of hair proteins analysis. This strategy is mainly based on the improvement of nanoLC-ESI-Q-TOF coupling for high-throughput peptide sequencing. Cell type isolation to study the proteome of cortical and cuticular hair cells. We have compared the semi quantitative expression of the identified proteins to investigate their location in those proteomes. These analyses led to protein expression evidence for 60% of keratin associated proteins predicted genes (KAP) compared to the only 20% previously evidenced (according to Uniprot). Use of complementary approaches to attempt to refine the quantification of the major proteins of cortical cells, to evidence structural properties for specific protein sequences and to propose hypotheses for the main KAP genes origin
