La Tomographie par Emission de Positrons (TEP) est une technique d’imagerie médicale utilisée largement dans le traitement du cancer et dans la recherche neurobiologique, afin d’imager l’activité biologique des organes. Il s’agit de détecter deux photons de 511 keV produits par l’annihilation d’un positron dans les tissus, ce qui permet d’en reconstruire la carte 3D. En mesurant avec une très bonne précision la différence de temps de détection des deux photons, il sera possible d’améliorer la qualité d’image (technique du temps de vol). Dans ce manuscrit, nous présentons le développement de deux détecteurs innovants, rapides et efficaces, pour la détection de la lumière Cherenkov produite par la conversion des photons de 511 keV. Le premier, destiné à un scanner clinique (cerveau) et pré-clinique à haute précision spatiale, utilise comme milieu de détection du TriMéthylBismuth. Le second, pouvant être utilisé pour construire un scanner corps entier, met en œuvre un cristal de PbF₂ comme radiateur Cherenkov. Dans les deux configurations, un photomultiplicateur à micro-canaux (MCP-PMT) est utilisé pour détecter les photons Cherenkov. Notre électronique de détection montre une résolution temporelle limitée à 5 ps (RMS). La chaîne de détection est limitée par les performances du MCP-PMT. Après étalonnage, nous avons mesuré une efficacité de 25 % (grande pour un détecteur Cherenkov), et de résolution temporelle de 200 ps (FWHM).Nous exposons les facteurs limitant la résolution temporelle des détecteurs et proposons des développements qui permettront d’en améliorer les performances<br>Positron Emission Tomography (PET) is a nuclear imaging technique widely used in oncology and neuroscience to observe biological activity in the body. Detection of two gamma quanta with the energy 511 keV emitted by positron annihilation in tissues allows one to reconstruct the tracer activity distribution in the body of the patient. Additional measurement of the difference in time detection between the two photons lets us to improve significantly the quality of the reconstructed image (time-of-flight method).In this manuscript, we present the development of two innovative detectors, fast and efficient, used to detect Cherenkov light produced by electrons from the photo-ionization conversions of 511 keV gamma quanta. The first one, intended for use in a brain PET scanner of a high spatial resolution, uses TriMethylBismuth for the detection medium. The second one, planned to be used to construct a whole-body PET scanner, enforces a PbF₂ crystal as Cherenkov radiator. In both configurations a micro-channel photo-multiplier (MCP-PMT) is used to detect Cherenkov photons. We commissioned an electronic detection chain with a time resolution limited to 5 ps (RMS). Using precise MCP-PMT calibration, we were able to develop simultaneously detectors with high efficiency, up to 25 %, and time resolution as good as 200 ps (FWHM).We highlight the limitations of detectors time resolution and suggest several developments in order to improve performances of Cherenkov light detectors

[ES] El crecimiento de las plantas se ve afectado por el estrés abiótico, sequía, salinidad o altas temperaturas. La transducción de señales de estrés abiótico es fundamental para generar una respuesta fisiológica adecuada, que implica la participación de diferentes hormonas vegetales, siendo el ácido abscísico (ABA) el regulador hormonal crítico en la regulación de la respuesta de la planta a situaciones de estrés por déficit hídrico. La vía de señalización de ABA y los componentes principales están bien caracterizados molecular y bioquímicamente. Los receptores de ABA "Pyrabactin Resistance 1"(PYR)/"PYR1-LIKE" (PYL)/ "Regulatory Component of ABA Receptor" (RCAR) juegan un papel importante en la regulación cuantitativa de la señalización ABA tanto en semillas como en tejidos vegetativos. Aunque la función bioquímica de los receptores PYR/PYL/RCARs de ABA, está bien caracterizada, se conoce poco sobre otros aspectos con relevancia biológica, como sus modificaciones postraduccionales o la regulación de su vida media. Uno de los avances recientes en este campo ha sido el descubrimiento de una nueva familia de E3 ligasas llamadas RSL1/RFAs ("RING-finger-ABA-related") que consta de al menos 10 miembros, reguladores clave de la estabilidad de los receptores PYR/PYL/RCAR de ABA en tejidos de raíces y hojas, regulando su degradación en diferentes ubicaciones celulares. Un estudio detallado de esta familia génica reveló que RSL1/RFA se caracterizan estructuralmente por la presencia de tres dominios RING putativos en tándem, denominados "RING1-IN BETWEEN RING-RING2" (RBR), y en consecuencia pertenecen a la familia de E3 ligasas de tipo RBR. Cinco miembros de la familia RSL1/RFA, RSL1 y RFA6-RFA9, contienen un dominio TM en el extremo C-terminal, lo que sugiere que RFA6-RFA9 también se localizan en la membrana plasmática. Sin embargo, otros miembros de las E3 ligasas como RFA1-RFA5 carecen del dominio TM C-terminal y su caracterización funcional, así como su ubicación celular, aún no se conocen. Nosotros mostramos que la E3 ligasa RFA1 se localiza en núcleo y citosol, mientras que RFA4 muestra una localización específica en el núcleo promoviendo la degradación nuclear de los receptores ABA. Por lo tanto, los miembros de la familia RSL1/RFA interactúan con los receptores ABA en la membrana plasmática, el citosol y el núcleo, dirigiéndolos a su degradación a través de la vía endosomal/vacuolar (en el caso de RSL1) o el proteosoma 26S (para RFA1 y RFA4). Realizando tanto mutagénesis como ensayos bioquímicos para identificar la cisteína 361 (Cys361) en RFA4 como la Cys del sitio activo, que es una característica distintiva de las E3 ligasas de tipo RBR. Demostramos mediante análisis de inmunotransferencia del mutante con pérdida de función de rfa1rfa4 que los niveles endógenos de los receptores de ABA PYR1 y PYL4 aumentan en comparación con las plantas de tipo silvestre. Hemos identificado una enzima E2, "Ubiquitin Conjugating Enzyme 26" (UBC26), como la enzima nuclear canónica E2 que interactúa con la E3 ligasa RFA4 y forma complejos UBC26-RFA4-Receptor, formando agregados nucleares. Generamos alelos ubc26 con pérdida de función que mostraban una mayor sensibilidad a ABA y acumulación de receptores ABA en comparación con el tipo silvestre. En definitiva, hemos revelado un sofisticado sistema de ubiquitinación de receptores ABA en diferentes ubicaciones subcelulares llevado a cabo a través de la familia de E3 ligasas RSL1/RFA de tipo RBR. Por otro lado, hemos iniciado pruebas bioquímicas para identificar la S-acilación en el dominio TM de RSL1. Generando RSL1C334S, RSL1 C5S y RSL1C6S mediante mutagénesis y RSL1ΔTM que presenta una delección del dominio TM. Signal transduction of abiotic stress is crucial to generate an appropriated physiological response, which involves the participation of different plant hormones, being abscisic acid (ABA) the critical hormonal regulator in regulating the plant's response to situations of stress due to water deficit. The ABA signaling pathway and the major components of the pathway are well characterized molecularly and biochemically. Pyrabactin Resistance 1 (PYR)/PYR1-LIKE (PYL)/Regulatory Component of ABA Receptor (RCAR) ABA receptors play an important role in quantitative regulation of ABA signaling both in seeds and vegetative tissues. Although the biochemical function of the PYR/PYL/RCAR ABA receptors has been well established in recent years, little is known about other aspects with biological relevance, such as their post-translational modifications or the regulation of their half-life. One of the recent advances in this field has been the discovery of a new family of E3 ligases called RSL1/RFAs (RING-finger-ABA-related) that consists of at least 10 members, which are key regulators of the stability of PYR/PYL/RCARs in root and leaf tissues, and regulate the degradation of ABA receptors at different cellular locations. Further inspection of the gene family revealed that RSL1/RFAs are structurally characterized by the presence of three putative RING domains in tandem, named as RING1-IN BETWEEN RING (IBR)-RING2, and accordingly they belong to the RBR-type E3 ligase family. Five members of the RSL1/RFA family, that is, RSL1 and RFA6-RFA9, contain a TM domain at the C-terminal end of the proteins, which suggests that RFA6-RFA9 are also localized in plasma membrane. However, other members of this family of E3 ligases such as RFA1-RFA5 lack the C-terminal TM domain and their functional characterization, as well as their cellular location, has not been investigated yet. In this study we show that the E3 ligase RFA1 is localized both in the nucleus and in the cytosol, while RFA4 shows a specific localization in the nucleus promoting the nuclear degradation of ABA receptors. Therefore, we members of the RSL1/RFA family interact with ABA receptors at the plasma membrane, cytosol and nucleus, targeting them for degradation via the endosomal/vacuolar pathway (in the case of RSL1) or the 26S- proteasome (for RFA1 and RFA4). We provide information on the physiological function of these RBR-type E3 ligases, which are hardly explored in plants. Additionally, we performed mutagenesis and biochemical assays to identify Cys361 in RFA4 as the active site cysteine, which is a distinctive feature of RBR-type E3 ligases. We have shown by immunoblot analysis of the rfa1rfa4 loss-of-function mutant that endogenous levels of ABA receptors PYR1 and PYL4 are increased compared to wild-type plants. On the other hand, we have identified an E2 enzyme, Ubiquitin Conjugating Enzyme 26 (UBC26), as the canonical nuclear enzyme E2 that interacts with the E3 ligase RFA4 and forms UBC26-RFA4-Receptor complexes, forming nuclear aggregates. We also generated loss-of function ubc26 alleles that exhibited higher sensitivity to ABA and accumulation of ABA receptors compared to wild type. We have revealed a sophisticated ubiquitination system of ABA receptors in different subcellular locations carried out through the RBR-type RSL1/RFA family of E3 ligases. We have proceeded with the biochemical and genetic study of the different members of the family. We have started biochemical tests to identify the S-acylation in the TM domain of RSL1. To this end, we have generated RSL1C334S, RSL1 C5S and RSL1C6S by mutagenesis as well as RSL1ΔTM, a deletion of the TM domain. Initial studies have shown that Cys residues close to the TM domain are S-acylated. Finally, we have also generated new combined mutants: rsl1rfa1, rsl1rfa5, rfa1rfa5 and rsl1rfa1rfa5.<br>Fernández López, MA. (2021). La sequía y la salinidad representan estreses ambientales que afectan de forma crítica el crecimiento de las plantas y limitan enormemente su potencial agrícola. La fitohormona ácido abcísico (ABA) juega un papel fundamental en la coordinación de la respuesta y adaptación de las plantas a este tipo de estreses, así como en la regulación del crecimiento y desarrollo vegetal. Elementos intermediarios de la ruta de señalización ya habían sido caracterizados, pero aún se desconocía el mecanismo de percepción y transducción de señal de la hormona. Este trabajo de tesis ha contribuido a la caracterización de una nueva familia de receptores intracelulares de la hormona ABA, formada por 14 miembros y denominada PYR/PYL (de pyrabactin resistance / PYR1-like) /RCAR (de Regulatory Component of Abscisic acid Receptor), y a su caracterización estructural y bioquímica. Estas proteínas son capaces de unir de forma específica la hormona ABA. La unión de la hormona induce en estos receptores un cambio conformacional, que les permite regular la actividad de los reguladores negativos de la ruta: fosfatasas del grupo A como ABI1, ABI2 o HAB1 ( Leung et al., 1994; Meyer et al.,1994; Saez et al., 2004). To establish accurate and early stage tumour diagnosis, quantitative molecular imaging modalities such as positron emission tomography (PET) are being employed. Molecular probes containing a positron emitting isotope (e.g. ¹⁸F, ⁶⁴Cu) are designed to target biomarkers or metabolic processes within the tumour. A therapeutic molecule could then be conjugated to this diagnostic probe to create a multifunctional construct (MFC). MFCs are a new concept in drug design offering the potential for both diagnosis and therapy. This work exploits the chelating properties of saturated tetraazamacrocycles known herein as macrocycles as well as their known affinity for the chemokine receptor CXCR4, a cell surface receptor involved in embryonic development and cell migration known to be overexpressed in at least twenty three different tumour types, along with the photodynamic activity of porphyrin molecules to create novel MFCs with diagnostic and therapeutic properties. A series of configurationally restricted copper(II), zinc(II) and nickel(II) mono-ring macrocycles bearing a methylamine pendant arm are presented. Monomacrocycles possess the potential for improved pharmacological properties. Biological studies assessing the affinity and potency of these macrocycles as CXCR4 antagonists demonstrated that zinc(II) monomacrocycles were the most potent in their class. A side bridged (SB) zinc(II) monocyclen compound; [Zn9]²⁺ synthesised in 64% yield, demonstrated the highest affinity for CXCR4 in its class showing a 2.5-fold higher affinity in comparison to the clinically licensed antagonist AMD3100 (IC₅₀ = 12 nM vs. 31 nM). In response to the high affinity observed for a meta-substituted biscyclen ligand (L48) a novel cross bridged (CB) meta-substituted biscyclen compound bearing a methylamine pendant arm 20 was synthesised along with its copper(II), nickel(II) and zinc(II) complexes in 77%, 88% and 90% yields, respectively. These metal complexes displayed high affinity for CXCR4 when competed with a CXCR4 specific antibody (% inhibition >85%) and required sub-micromolar concentrations to reduce the ‘normal’ signalling of CXCR4 by 50%. [Cu₂20]⁴⁺ has the potential to be a targeted PET probe through the use of ⁶⁴Cu and so a detailed biological evaluation into its binding mode was undertaken. These assays concluded that [Cu₂20]⁴⁺ exerts its antagonistic effect by locking the receptor on the surface of the cell, stopping the internalisation and recycling processes, which would result in formation of a new receptor. An analogous meta-substituted biscyclam ligand (28) and its copper(II), zinc(II) and nickel(II) complexes were synthesised in 74%, 58% and 46% yields, respectively. These metal complexes also demonstrated high affinity for CXCR4 (IC₅₀ < 0.1 μM). In fact [Cu₂28]⁴+ showed unexpectedly high affinity for CXCR4 demonstrating almost a 3-fold higher affinity than AMD3100 (IC₅₀ = 11nM vs. 31 nM), the potential of [Cu₂28]⁴⁺ as a targeted PET probe should also be considered. Various organic spacer arms were attached to bismacrocycle 20, to produce a CXCR4 targeting probe with a functional arm for further conjugation which would also act to prevent steric hindrance. An acidic terminating chain was successfully attached to bismacrocycle 20 in quantitative yield, via ring-opening of diglycolic anhydride as well as an amine terminating variant (93% yield). A biotin tag was also attached to bimacrocycle 20 and its copper(II) complex was successfully isolated in a 94% yield. Addition of a spacer arm was found to have no significantly detrimental effect on binding affinity to CXCR4. A series of known water and organic soluble metalloporphyrins were synthesised. Subsequent conjugation to macrocyclic compounds proved challenging but analytical data for the conjugation of bismacrocycle 20 and metalloporphyrin [Zn59] indicated a successful reaction had occurred. This reaction represents important steps towards the synthesis of MFCs with diagnostic and therapeutic properties.

Abstract The general goal of this thesis is the development of new generation photoactivable smart nanomaterials that can be modulated into tremendous way to achieve multimodal anticancer and antimicrobial therapeutic actions. Here the cyclodextrins act as the building block for the development of the nanocarrier in order to convey the therapeutic payload to the desired site. With this goal in mind, we have developed several kinds of nanosystems and are discussed in each chapter. The entire dissertation is divided into four sections based on the types of materials and the sections are further divided into twelve chapters based on the functional behavior of each nanosystems.

Atualmente a bioadsorção vem se apresentando como uma boa alternativa às tecnologias existentes. Este trabalho teve como objetivo a investigação da macrófita Azolla pinnata como bioadsorvente de cinco íons metálicos: Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+ em soluções sintéticas. A caracterização do biomaterial foi realizada utilizando-se técnicas como Microscopia Eletrônica de Varredura (MEV) para análise da superfície do material; análise de Espectroscopia de Energia Dispersiva de Raios-X (EDX), que mostrou que qualitativamente a diminuição de íons de competição (Ca, Na e K) após o processo de adsorção, evidenciando que a troca iônica pode ser um dos mecanismos do processo; e análise de Infravermelho por Transformada de Fourier (FTIR), em que se pôde constatar que alguns grupos funcionais como carboxilas e grupos sulfonados são fundamentais no processo. A cinética de adsorção foi ajustada pelos modelos de pseudoprimeira e pseudossegunda ordem, sendo que o melhor ajuste variou de acordo com o íon estudado. O modelo de difusão intrapartícula mostrou que muitos mecanismos podem ser limitantes na taxa com a qual ocorre o processo. Os dados de equilíbrio foi ajustado pelos modelos de isoterma de Langmuir e de Freundlich em duas temperaturas, 25 e 45 ºC. O processo foi favorecido pelo acréscimo de temperatura para todos os íons. O modelo de Langmuir se ajustou melhor aos dados experimentais de todos os íons e o valor de capacidade máxima de adsorção (qmax) a 25 ºC foi de 0,359; 0,448; 0,254; 0,472 e 0,357 mmol/g para Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+, respectivamente. Os eluentes HCl, EDTA e CaCl2 foram testados no processo de dessorção em batelada e o cloreto de cálcio se mostrou eficiente além de não causar danos ao material nem diminuir a capacidade de adsorção da macrófita. Ensaios dinâmicos em coluna de leito fixo foram realiados para os íons Cu2+ e Pb2+ com soluções mono e bicomponentes. Nos ensaios em sistema binários, pode-se constatar uma grande preferência pela adsorção de íons chumbo e, portanto, uma grande queda na adsorção de cobre. Foram testados ciclos de adsorção/dessorção em coluna utilizando o CaCl2 como eluente e, em três ciclos, a macrófita manteve suas propriedades adsortivas. Com estes resultados, pode-se constatar que a realização deste trabalho contribuiu para a avaliação deste material como bioadsorvente alternativo e que, especificamente, macrófita A. pinnata mostrou-se atrativa para utilização nos processos de tratamento e/ou estabilização de metais pesados presentes em baixas concentrações em efluentes industriais<br>Resumo: No processo Kraft para produção de papel e celulose, os cavacos de madeira são cozidos em um digestor com uma solução aquosa denominada licor branco, constituída de hidróxido de sódio (NaOH) e sulfeto de sódio (Na2S). Durante o cozimento uma pasta fibrosa é produzida. Esta pasta é filtrada e segue para o branqueamento. O produto filtrado chama-se licor negro, devido à mudança da sua cor nesta etapa. O licor negro possui aproximadamente 15 % de sólidos em massa. Ele é bombeado para a unidade de recuperação, na qual ele será concentrado em evaporadores de múltiplosefeitos até 75 % de sólidos em massa. Em seguida, ele é bombeado para uma caldeira onde é queimado, para recuperar os sais inorgânicos e, principalmente, fornecer calor para geração de calor. Durante a evaporação do licor negro, quando a porcentagem em sólidos ultrapassa 50 %, ocorre a precipitação de sais inorgânicos. Estes sais se depositam nas superfícies de troca térmica dos evaporadores diminuindo sua eficiência e causando prejuízos à indústria. Os principais sais que causam este problema são o carbonato de sódio (Na2CO3) e o sulfato de sódio (Na2SO4). O objetivo do presente trabalho foi testar e validar um método experimental para a determinação da quantidade de Na2CO3 presente em amostras industriais de licor negro. Além disso, também foram analisadas algumas propriedades físico-químicas do licor negro, como massa específica, teor de sólidos e viscosidade. Com os resultados obtidos, tentou-se obter uma correlação entre o teor de carbonato de sódio e as demais propriedades do licor que foram determinadas. Os resultados das análises das características físico-químicas demonstraram coerência e boa reprodutibilidade, porém, os resultados das análises de teor de carbonato de sódio não foram satisfatórios e mostraram que a técnica deve ser aperfeiçoada<br>Abstract: Research in the technology field has been looking for evaluating efficient and economic techniques and/or process for effluents that contains heavy metal. The biodsorption process has been seemed as a good alternative to the existing technology. This work had the aim to investigate the use of Azolla pinnata macrophyte as biosorbent of five metallic ions: Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+ in synthetic solutions. The biomaterial characterization was carried out by techniques as scanning electron microscopy (SEM) for the analyze of material superficies; the energy dispersive X-ray spectroscopy (EDX), which showed, qualitatively, a decreasing in the competitions ions (Ca, Na e K) after the sorption process, indicating that the ion exchange can be one o the process mechanism; and Fourier Transform Infrared Spectroscopy (FTIR), in which it was possible implied functional groups as carboxyl and sulfonate play a important role in the adsorption. The kinetics was adjusted by primer and second order models and the model which fit better depends on each ion. The intra-particle diffusion model showed that several mechanisms may be limiting to the process rate. The equilibrium data were adjusted by Langmuir and Freundlich models in two temperatures: 25 and 45 ºC. The process was favored by the increasing in the temperature for all five ions. The Langmuir model fit better the experimental data and the maximum adsorption capacity (qmax) were 0,359; 0,448; 0,254; 0,472 and 0,357 mmol/g for Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+, respectively. The HCl, EDTA and CaCl2 eluents were tested in the desorption process and the calcium chloride was efficient and did not cause any damage in the biomaterial or decreasing the biosorption capacity. Dynamics runs were carried out in fixed-bed in mono and bi-components solutions. In the binary were found a great preference for lead instead of copper and as a result, a significant decrease in the removal of copper. Adsorption/desorption cycles were tested in fixed-bed system using CaCl2 as eluent. After three cycles no damage, weight lost or significant decrease in the biosorption capacity was noticed. These results indicating that the A.pinnata macrophyte is an attractive potential new biosorbent to be used in heavy metal removal especially in low concentrations effluents<br>Abstract: In the Kraft process for pulp and paper production, wood chips are cooked in a digester with an aqueous solution named white liquor, consisted by sodium hydroxide (NaOH) and sodium sulfide (Na2S). During this cooking process, wood chips are transformed generating a fibrous paste and a liquid solution. This paste is filtered and separated from the liquid solution, named black liquor due to its color. The paste goes to the bleaching stage to produce cellulose. Black liquor contains inorganic and organic (mainly lignin) compounds from wood. Initially this black liquor contains approximately 15 mass/% of solids and is sent to the recovery unit to have this concentration raised to approximately 75 mass/% to be burned in a recovery boiler in order to generate power and recover inorganic reactants, recycling them to the process. The concentration process of black liquor occurs in a multiple effect evaporators unit. During this process, when the concentration of solids exceeds 50%, the precipitation of inorganic salts (mainly Na2CO3 and Na2SO4) begins. These salts are deposited in the heat transfer surfaces of the evaporator tubes, reducing their efficiency, leading to a reduction in the production capacity. The scope of this work was testing and validate an experimental method to estimate the content of Na2CO3 present in industrial black liquor samples. Moreover, some black liquor's physical-chemical properties as density, solids content and viscosity were also analyzed. An attempt to develop a predictive model to evaluate sodium carbonate content from the other properties was made. The results obtained for physical-chemical properties have shown a good consistency. Este trabajo de investigación nace por el amor a nuestras mascotas, el querer darles una mejor calidad de vida y la necesidad que tenemos los dueños de mascotas de tener una atención inmediata y de calidad para nuestros amigos peludos cuando pase alguna emergencia. De esa necesidad es que nace el seguro para mascotas SAFE PET, que ofrece un paquete estándar que incluyan varios beneficios y coberturas para las mascotas afiliadas. La principal fuente de ingreso de SAFE PET será la venta de nuestras pólizas, nuestra inversión inicial será de s/ 124,890 que la financiaremos el 60% los 4 socios de la empresa y 40% con un préstamo a una entidad financiera. Por las investigaciones que realizamos mediante encuestas a dueños de mascotas, estos en su mayoría nos manifestaba el querer tener un seguro para su mascota, y luego gracias nuestra landing page pudimos observar que del 100% de visitas a nuestra landing el 54.6% dejó sus datos porque le interesa nuestro seguro, este porcentaje es mucho mayor al que esperábamos y eso nos alienta mucho más a ser uno de los primero en el país en dedicarnos exclusivamente en seguro integral de mascotas. Finalmente, luego del estudio financiero realizado a nuestra empresa hemos considerado en el análisis de sensibilidad COK (89.71%) y un costo promedio ponderado de capital de WACC (74.53%), para terminar después de todo nuestro plan financiero podemos concluir que SAFE PET, es una empresa viable económicamente.<br>This research was born for our love to pets and we want to give them a better life quality and the need that pet owners to have immediate and quality care for our furry friends when an emergency happens. From that need is born SAFE PET, that is a pet insurance, which offers a standard package that includes many benefits and coverages for affiliated pets. The main source of income for SAFE PET will be the sale of our policies, our initial investment will be S/ 124,890 which will be financed by 60% the 4 partners of the company and 40% with a loan to a financial institution. El presente trabajo plantea la puesta en marcha de un negocio innovador, para ello, se ha considerado los factores externos e internos que podrían repercutir en el desarrollo del mismo. Asimismo, se ha evaluado el contexto actual y se ha elaborado el respectivo estudio que sustenta la viabilidad del proyecto. Hoy en día las personas se preocupan más por la seguridad de sus mascotas, existe una tendencia petfriendly que ha llevado a la sociedad a incrementar el consumo de alimentos balanceados, gastos en veterinarias y spas caninos. Considerando que no existe en la actualidad un negocio similar al planteado, los alumnos autores del presente trabajo han considerado que la creación de un seguro para mascotas en el Perú sería altamente rentable. Para sustentar la idea expuesta, se ha tomado en consideración todos los aspectos concernientes a la creación de la persona jurídica, los objetivos estratégicos, políticas internas y externas, análisis financieros y la rentabilidad del proyecto. Después de analizar los aspectos antes mencionados, y de considerar todos los aspectos que puedan influir en su desarrollo, se llega a la conclusión de que el negocio es viable.<br>This work proposes the start-up of an innovative business, for this, the external and internal factors that could have an impact on its development have been considered. Likewise, the current context has been evaluated and the respective study that supports the viability of the project has been prepared. Nowadays people care more about the safety of their pets, there is a petfriendly trend that has led society to increase consumption of feed, veterinary expenses and canine spas. Considering that there is currently no business similar to that proposed, the student authors of the present work have considered that the creation of pet insurance in Peru would be highly profitable. To support the idea presented, all aspects concerning the creation of the legal entity, strategic objectives, internal and external policies, financial analysis and the profitability of the project have been taken into consideration. Asimismo, contribuye a fortalecer la tenencia responsable de mascotas en los hogares de familia. TESIS PARA OPTAR AL GRADO DE MAGÍSTER EN ADMINISTRACIÓN<br>PTY Garden supone la creación de una empresa dedicada a ofrecer el servicio de jardinería en el distrito de La Chorrera, provincia de Panamá Oeste, distante a 39.1 Kms del centro de la ciudad de Panamá. Su actividad principal será el mantenimiento, diseño y ejecución de zonas verdes y jardines en áreas residenciales. El proyecto cuenta con varios elementos diferenciadores, como lo son la apuesta por las nuevas tecnologías e innovación a la hora de diseñar paisajes vinculados a la jardinería Feng-shui y al uso de técnicas para la conservación y mantenimiento de jardines. El servicio está orientado a hombres y mujeres mayores de 25 años, que estén interesados en el mantenimiento de los jardines de sus residencias, por lo que se estima un tamaño de mercado de USD$15,114,390.00 dólares al año. La elección de la provincia de Panamá Oeste responde a 3 motivos principales: por un lado, en los últimos años la provincia ha presentado un crecimiento demográfico significativo, siendo el punto de mayor producción residencial del país. Fenómeno que se ha dado debido a que el 70% de los proyectos residenciales son de interés preferencial, ya que la Ciudad de Panamá no es capaz de cubrir la gran demanda de viviendas que exige la población, por lo que recurren a áreas aledañas a la ciudad donde residir. Otro motivo es que Panamá Oeste es considerada la “Ciudad Dormitorio”, ya que la mayoría de los ciudadanos que residen en esta zona, trabajan en la ciudad en jornadas extendidas, por lo que no disponen de tiempo para realizar actividades como la jardinería. El último y uno de los más relevantes motivos es que en La Chorrera no existe una empresa que brinde el servicio de jardinería, más bien empresas de jardinera en la ciudad deben viajar hasta la provincia para prestar sus servicios. Su equipo gestor está representado por Milagros Macias, Lic. En Ingeniera Marítima, con conocimiento y experiencia en servicio al cliente y manejo de personal. Además cuenta con habilidades en el diseño de jardines utilizando los módulos de AutoCAD. El análisis financiero realizado muestra que el proyecto es factible y que requiere de una inversión inicial de USD$45,000 dólares. El VAN que arroja es de USD$106,227.36 dólares, la TIR de 76% garantiza la capacidad adquisitiva si la tasa de descuento subiera y el análisis de sensibilidad muestra que existe un 69% de probabilidades de éxito en su ejecución, convirtiéndolo en un proyecto que espera una buena rentabilidad.

Cette thèse a pour objectif l’élaboration de nanoparticules hybrides à base de silice par microémulsion inverse. Les nanoparticules de silice constituent une matrice de base permettant de confiner et de protéger des molécules organiques et/ou des nanoparticules métalliques. L’incorporation combinée de différentes entités dans la silice ouvre ainsi de larges perspectives de par l'introduction de nouvelles propriétés liées à la structure hybride. Afin d’élaborer de tels objets, nous avons utilisé des micelles inverses à base d'eau, de Triton X-100, d'hexanol et de cyclohexane comme milieu réactionnel. L’influence des conditions opératoires sur le contrôle de la taille des micelles inverses a d'abord été étudiée. Ces micelles inverses ont ensuite été mises à profit comme nanoréacteurs pour la synthèse de nanoparticules de silice par procédé sol-gel en utilisant les précurseurs alkoxysilanes adéquats. Nous avons regardé dans quelle mesure il était possible de contrôler la taille des nanoparticules de silice en fonction du pourcentage d’eau par rapport au tensioactif. Il a ainsi été possible d’accéder de façon reproductible à des nanoparticules avec de tailles variables, de 30 nm à 200 nm. Nous avons ensuite regardé qu'il était possible d'encapsuler au sein de cette matrice nanométrique des fluorophores et des nanoparticules d’or et d’argent de façon contrôlée. En vue d’assurer une bonne stabilisation colloïdale en solution, ces nanoparticules hybrides ont été fonctionnalisées d'une part par ajout d'un silane fonctionnel et d'autre part par click chemistry. Nous avons ainsi pu montrer qu’il est possible d’effectuer dans un même milieu micellaire l’ensemble des processus de fabrication de la nanoparticule hybride, de la matrice de silice à sa fonctionnalisation en passant par l’incorporation d’entités fonctionnelles. Cette méthode de synthèse séquentielle nous a ainsi permis de supprimer les étapes de purification et de redispersion qui peuvent s’avérer problématiques dans les procédés classiques. L’ensemble de ce travail a été mis à profit pour la conception d’un agent antibactérien à base de nanoparticules argent/silice capables d’empêcher la prolifération bactérienne grâce au relargage progressif des ions argent. Les tests effectués en solution comme sur le coton et le polyéthylène téréphtalate imprégnés montrent effectivement un caractère antibactérien certain de ces systèmes<br>This thesis aims at developing hybrid nanoparticles based on silica by reverse microemulsion. The silica nanoparticles are the basic matrix containing and protecting organic molecules and/or metallic nanoparticles. The combined incorporation of different entities within the silica opens wide prospects for the introduction of new properties related to the hybrid structure. To develop such objects, we used reverse micelles based on water, Triton X-100, hexanol and cyclohexan as reaction medium. The influence of operating conditions on the control of the size of reverse micelles was first studied. These micelles were then set to be used as nanoreactors for the synthesis of silica nanoparticles by sol-gel using suitable alkoxysilanes precursors. We monitored how it was possible to control the size of silica nanoparticles based on the water to surfactant ratio. It was thus possible to prepare in a reproducible way nanoparticles with sizes varying from 30 nm to 200 nm. We then investigated the possibility to encapsulate, in this nanoscaled matrix, fluorophores and nanoparticles of gold and silver in a controlled manner. To ensure a good colloidal stability in solution, these hybrid nanoparticles were, on the one hand, modified by adding a functional silane and, on the other hand, by click chemistry. We have thus shown that it is possible to perform, in a same micellar media, all of manufacturing process of the hybrid nanoparticle, from the silica matrix to its functionalization passing by the incorporation of functional entities. This method of sequential synthesis allowed us to bypass the purification and redispersion steps that can be problematic in the conventional methods. All this work has been extended to the design of an antibacterial agent based of silver/silica nanoparticles, capable of preventing bacterial growth through the gradual release of silver ions. Tests conducted in solution on the impregnated cotton and polyethylene terephtalate indeed show an interesting antibacterial character of these systems

Recently, systems that integrate positron emission tomography and magnetic resonance imaging (PET/MR) have become available for clinical use. This new technology, which combines the high spatial resolution and superior soft-tissue contrast of MR with the picomolar sensitivity, quantitative capabilities, and wide array of tracers of PET, has the potential to benefit patients and provide insights that were previously unattainable in standalone systems. Simultaneous measurement of PET and MR parameters provides complementary information, allowing for a more complete assessment of disease, as well as cross validation and calibration of MR and PET measurements and techniques. To take full advantage of such a multi-modal system, accurate quantification of the PET data is necessary. Due to the low spatial resolution of PET – which can be further reduced by external factors like patient motion – and the inherent lack of anatomic detail, accurate quantification can be challenging. The simultaneously acquired MR information provides an opportunity to optimize PET quantification and analysis. In order to fully realize the benefits provided by the simultaneously acquired MR data, the MR data cannot be treated as discrete sequences, but as the continuous flow of information. This is due to differences in the time required for data collection to generate PET and MR images. This work describes the development and optimization of a pipeline for the reconstruction and analysis of PET data in a brain-dedicated prototype PET/MR system, the BrainPET (Siemens Healthcare). First, the performance of the BrainPET system was optimized for neurological imaging. MR-hardware interference and characteristics of the PET camera were quantified and a method for multimodal alignment was developed. To simplify and streamline the reconstruction and quantification process, a platform was designed which utilizes the functionality of a number of specialized brain imaging analysis software packages in an automated fashion. Second, MR-based methods addressing specific challenges to PET quantification were addressed. Simultaneously acquired structural MR data was used to correct the PET data for attenuation and partial volume effects. The use of MR data for motion correction was addressed and a unified algorithm which derives motion estimates from the PET data when MR data is unavailable was presented. Finally, the value of the optimized PET processing for neurological studies was evaluated in three instances: first an upper limit on the physiologic noise introduced by MR imaging on cerebral metabolism was estimated using PET and found to be minimal; next the benefit of MR-based motion correction and partial volume effect correction were estimated in a patient study; and lastly, a method to derive the PET radiotracer input function from the PET data using multiple MR sequences was presented.<br>Biophysics

[EN] ABSTRACT Abscisic acid (ABA) signaling plays a critical role in regulating root growth and root system architecture. ABA-mediated growth promotion and root tropic response under water stress are key responses for plant survival under limiting water conditions. In this work, we have explored the role of Arabidopsis (Arabidopsis thaliana) PYR/PYL/RCAR receptors (PYRABACTIN RESISTANCE1 (PYR1)/PYR1 LIKE (PYL)/REGULATORY COMPONENTS OF ABA RECEPTORS) for root ABA signaling. As a result, we discovered that PYL8 plays a nonredundant role for the regulation of root ABA sensitivity. Unexpectedly, given the multigenic nature and partial functional redundancy observed in the PYR/PYL family, the single pyl8 mutant showed reduced sensitivity to ABA-mediated root growth inhibition. This effect was due to the lack of PYL8-mediated inhibition of several clade A phosphatases type 2C (PP2Cs), since PYL8 interacted in vivo with at least five PP2Cs, namely HYPERSENSITIVE TO ABA1 (HAB1), HAB2, ABAINSENSITIVE1 (ABI1), ABI2, and PP2CA/ABA-HYPERSENSITIVE GERMINATION3 as revealed by tandem affinity purification and mass spectrometry proteomic approaches. Membrane-delimited abscisic acid (ABA) signal transduction plays a critical role in early ABA signaling, but the molecular mechanisms linking core signaling components to the plasma membrane are unclear. We show that transient calciumdependent interactions of PYR/PYL/RCAR ABA receptors with membranes are mediated through a 10-member family of C2-domain ABA-related (CAR) proteins in Arabidopsis thaliana. Specifically, we found that PYL4 interacted in an ABA-independent manner with CAR1 in both the plasma membrane and nucleus of plant cells. CAR1 belongs to a plant-specific gene family encoding CAR1 to CAR10 proteins, and bimolecular fluorescence complementation and coimmunoprecipitation assays showed that PYL4-CAR1 as well as other PYR/PYL-CAR pairs interacted in plant cells. The crystal structure of CAR4 was solved, which revealed that, in addition to a classical calcium-dependent lipid binding C2 domain, a specific CAR signature is likely responsible for the interaction with PYR/PYL/RCAR receptors and their recruitment to phospholipid vesicles. This interaction is relevant for PYR/PYL/RCAR function and ABA signaling, since different car triple mutants affected in CAR1, CAR4, CAR5, and CAR9 genes showed reduced sensitivity to ABA in seedling establishment and root growth assays. In summary, we identified PYR/PYL/RCAR-interacting partners that mediate a transient Ca2+-dependent interaction with phospholipid vesicles, which affects PYR/PYL/RCAR subcellular localization and positively regulates ABA signaling.<br>[ES] RESUMEN La señalización por la hormona vegetal ácido abscísico (ABA) desempeña un papel crítico en la regulación del crecimiento de la raíz y en la arquitectura del sistema radical. La promoción de crecimiento de la raíz en condiciones de estrés hídrico mediada por ABA es clave para la supervivencia de las plantas bajo condiciones limitantes de agua. En este trabajo, hemos explorado el papel de los receptores PYR/PYL/RCAR (PYRABACTIN RESISTANCE1 (PYR1)/PYR1 LIKE (PYL)/ REGULATORY COMPONENTS OF ABA RECEPTORS) de Arabidopsis (Arabidopsis thaliana) en la ruta de señalización de ABA en raíz. Así, hemos descubierto que el receptor de ABA PYL8 juega un papel no redundante en la regulación de la percepción de ABA en raíz. Inesperadamente, dada la naturaleza multigénica y la redundancia funcional parcial observada en la familia PYR/PYL/RCAR, el mutante pyl8 fue el único mutante sencillo de pérdida de función de los receptores PYR/PYL/RCAR que mostraba una sensibilidad reducida a la inhibición del crecimiento mediada por ABA en raíz. Este efecto se debe a la falta de inhibición mediada por PYL8 de varias fosfatasas del grupo A tipo 2C (PP2Cs), ya que PYL8 es capaz de interactuar in vivo con al menos cinco PP2Cs, denominadas HYPERSENSITIVE TO ABA1 (HAB1), HAB2, ABAINSENSITIVE1 (ABI1), ABI2, and PP2CA/ABA-HYPERSENSITIVE GERMINATION3 según lo han revelado la purificación por afinidad en tándem (TAP por sus siglas en inglés) y estudios proteómicos de espectrometría de masas. La transducción de la señal del ABA localizada en la membrana plasmática celular juega un papel crucial en los pasos iniciales de la señalización de la fitohormona, pero los mecanismos moleculares que unen los componentes básicos de la señalización y la membrana plasmática no están claros. Estudiando las interacciones de los receptores del ABA PYR/PYL/RCAR con la membrana plasmática hemos encontrado que éstos pueden interaccionar transitoriamente con ella de forma dependiente de calcio gracias a una familia de proteínas con dominios C2 relacionadas con la ruta de señalización de ABA (denominadas C2-domain ABA-related (CAR) proteins). Específicamente, se encontró que PYL4 interacciona de manera independiente de ABA con CAR1 tanto en la membrana plasmática como en el núcleo de las células vegetales. La proteína CAR1 pertenece a una familia multigénica constituida por 10 miembros en Arabidopsis thaliana, desde CAR1 hasta CAR10, y que solo se encuentra en plantas. Los ensayos de complementación bi-molecular de fluorescencia y de co-immunoprecipitación confirmaron la interacción en células vegetales tanto de PYL4-CAR1 como de otras parejas de PYR/PYL-CAR. La cristalización de la proteína CAR4 reveló que, además de un dominio C2 clásico de unión a lípidos dependiente de calcio, las proteínas de la familia CAR presentan un dominio específico que probablemente es responsable de la interacción con los receptores PYR/PYL/RCAR y de su posterior reclutamiento a las vesículas de fosfolípidos. Esta interacción es relevante para la función de los receptores PYR/PYL/RCAR en la señalización del ABA, ya que diferentes mutantes triples car de pérdida de función, que tienen afectados los genes CAR1, CAR4, CAR5, y CAR9, demostraron una reducción de la sensibilidad al ABA en ensayos de establecimiento de plántula y crecimiento de la raíz. En resumen, hemos identificado nueva familia de proteínas que son capaces mediar las interacciones transitorias dependientes de Ca2+ con vesículas de fosfolípidos, lo que a su vez afecta localización de PYR/PYL/RCAR y regula positivamente la señalización de ABA.<br>[CAT] RESUM La senyalització per l'hormona vegetal àcid abcíssic (ABA) exerceix un paper crític en la regulació del creixement de l'arrel i també en l'arquitectura del sistema radical. La promoció del creixement de l'arrel en condicions d'estrés hídric, regulada per ABA és clau per la supervivència de les plantes sota condicions limitants d'aigua. Amb aquest treball, hem investigat el paper dels receptors PYR/PYL/RCAR (PYRABACTIN RESISTANCE1 (PYR1)/PYR1 LIKE (PYL)/ REGULATORY COMPONENTS OF ABA RECEPTORS) d'Arabidopsis (Arabidopsis thaliana) en el camí de senyalització d'ABA en arrel. Així, hem descobert que el receptor d'ABA PYL8 exerceix un paper no redundant en la regulació de la percepció d'ABA en arrel. Inesperadament, donada la naturalesa multigènica i la redundància funcional parcial que s'observa en la família PYR/PYL/RCAR, el mutant pyl8 va ser l'únic mutant senzill de pèrdua de funció dels receptors PYR/PYL/RCAR que mostrava una sensibilitat reduïda a la inhibició del creixement mitjançada per l'ABA en l'arrel. Doncs aquest efecte es deu a la falta d'inhibició regulada per PYL8 de diverses fosfatases del grup A tipus 2C (PP2Cs), ja que PYL8 té la capacitat d'interactuar in vivo almenys amb cinc PP2Cs, anomenades HYPERSENSITIVE TO ABA1 (HAB1), HAB2, ABAINSENSITIVE1 (ABI1), ABI2, and PP2CA/ABAHYPERSENSITIVE GERMINATION3 segons ho han revelat per una banda la purificació per afinitat en tàndem (TAP són les seues sigles en anglés) i per altra banda, estudis proteòmics d'espectrometria de masses. Pel que fa a la transducció del senyal del l'ABA, la qual es localitza en la membrana plasmàtica cel¿lular, juga un paper molt important en els primers instants de la senyalització de la fitohormona, no obstant això els mecanismes moleculars que uneixen els components bàsics d'aquesta senyalització amb la membrana plasmàtica, no es troben del tot clars. Per tant, s'han estudiat les interaccions que tenen els receptors del ABA PYR/PYL/RCAR amb la membrana plasmàtica, i hem trobat que aquests tenen la capacitat d'interaccionar transitòriament amb la membrana de forma dependent al calci, gràcies a una família de proteïnes amb domini C2, les quals es troben relacionades amb la ruta de senyalització d'ABA(anomenades C2domain ABArelated (CAR) proteins).Específicament, es va trobar que PYL4 interacciona d'una manera independent al ABA amb CAR1, tant en la membrana plasmàtica, com en el nucli de les cèl¿lules vegetals. La proteïna CAR1 pertany a la família multigènica constituïda per 10 components en Arabidopsis thaliana, des de CAR1 fins CAR10, que tan sols es troba en plantes. Els assajos de complementació bimolecular de fluorescència i de co-immunoprecipitació, van confirmar la interacció en cèl¿lules vegetals, tant de PYL4CAR1 com d'altres parelles de PYR/PYL-CAR. La cristal¿lització de la proteïna CAR4 va revelar que, a més d'un domini C2 clàssic de unió a lípids dependent del calci, les proteïnes de la família CAR presenten un domini PYR/PYL/RCAR, i del seu posterior reclutament a les vesícules fosfolipídiques. Doncs, aquesta interacció és rellevant en la funció dels receptors PYR/PYL/RCAR, ja que participa en la senyalització del l'ABA. Aquesta interacció es clau per a la funció dels receptors, ja que diferents mutants triples car de pèrdua de funció, els quals posseïxen afectats els gens CAR1, CAR4, CAR5 i CAR9, van mostrar una reducció de la sensibilitat a l'ABA en assajos d'establiment de plàntula i creixement de l'arrel. This thesis looks at the study of nanocomposites of Poly(ethylene terephthalate) and organoclays. Two methods of materials blending are investigated for the production of the nanocomposites: solvent blending and melt blending. The main objectives were the investigation of the influence of organoclays and processing conditions on morphological, rheological, mechanical properties, crystal structure and isothermal crystallization kinetics of the nanocomposite and a comparison with unfilled PET. In solvent blending, the use of long sonication time and epoxy led to the formation of a two-dimensional network structure of long, thin particles in a solvent blended PET nanocomposite at low clay loading. The clay network structure seemed not to affect the tensile properties. The long, thin particles were able to be separated and dispersed further by high shear in a twin screw extruder, resulting in a high level of separation and dispersion. The crystallization of the solvent blended nanocomposite was not only influenced by the nanoclay but also by the residual solvent. The extent of clay dispersion did not affect the crystallization of the solvent blended sample. Both solvent blended and melt blended nanocomposites showed that increasing the amount of surfactant improved the degree of nanoclay dispersion in the PET that led to an enhancement in the tensile properties of the nanocomposite compared to the unfilled polymer. The degradation of the organoclay during melt blending did not limit the nanoclay dispersion in the PET. The low thermal stability of the organoclay reduced the strength of the crystalline nanocomposite but it did not affect the strength of the amorphous nanocomposite. In contrast to the solvent blended sample, the extent of clay dispersion influenced the crystallization of the melt blended sample. The poorly dispersed particles were more efficient in nucleating PET crystallization than the well dispersed particles. The crystallization rate of PET increased as the surfactant concentration decreased.

Los jugos naturales a base frutas y verduras, mejor conocidos como jugos verdes , está entrando en un período de rápida evolución, debido al cambio producido en los patrones de consumo en segmentos de la población panameña, cada vez más interesada en el consumo de alimentos naturales para mejorar sus hábitos alimenticios y salud. Además, este mismo segmento de personas optan por productos más prácticos, al no disponer de tiempo suficiente para su preparación. En Panamá existen algunas tiendas dedicadas a ofrecer jugos naturales de frutas y verduras, de la cual destaca GREEN BAR, la cual es percibida como la más especializada hasta el momento, adicional se encuentra ECO-FRUIT el cual adicional vende comida orgánica ligera, y FRESCHO, quien hace entregas a domicilio. La propuesta de valor radica en brindar una experiencia de consumo basada principalmente en servicio personalizado y la oferta de realizar las mezclas de frutas y vegetales al gusto del consumidor. El mercado objetivo son individuos comprendidos entre los 25 y 45 años, que tienen en común hábitos alimenticios y rutina de vida. Hemos identificado los siguientes segmentos: los pragmáticos y los racionales. Debido a que Barra Verde PTY es una marca totalmente nueva, el objetivo de este plan de mercadeo es: Generar conocimiento de marca y lograr que los consumidores asocien a Barra Verde PTY como la mejor experiencia de servicio en la categoría. Por lo tanto el posicionamiento a futuro, deseado es que Barra Verde PTY, se perciba como la mejor experiencia de compra en el mercado de Panamá en su categoría. La estrategia para conseguir los objetivos ya mencionados estará fundamentada en un Plan Comunicacional, teniendo como iniciativas y tácticas, conjuntas de mercadeo digital, promocional y relacional. La tienda está localizada en la ciudad de Panamá, en la exclusiva zona de Costa del Este, caracterizada por la alta presencia de corporaciones nacionales y multinacionales. El objetivo del presente trabajo de investigación es desarrollar la propuesta de negocios, a través de una plataforma virtual, para la atención de mascotas domésticas, dirigido a los dueños de estas mascotas y centro veterinarios que en los últimos años han tenido un crecimiento del 76% a Nivel Nacional y un 91% solo en Lima Metropolitana. Se ha identificado un mercado de dueños de mascotas (usuarios), en amplio crecimiento que busca cada día, centros veterinarios especializados que brinden un servicio de alta calidad, conectividad en línea, con información actualizada y al mismo tiempo a costos mínimos necesarios. Así mismos centros veterinarios (clientes) que buscan los conecten con su público objetivo a través de las redes sociales y todo medio informático que incremente su cartera de clientes y que permitan desarrollar la publicidad de sus servicios a bajos costos. En la primera parte del trabajo se ha desarrollado tanto el planeamiento estratégico con el fin de recopilar información valiosa que ha permitido corroborar supuestos y determinar los puntos más valorados por los usuarios y clientes con respecto a la propuesta de valor y así poder utilizarlas en la fase posterior, es decir en nuestro plan de marketing. En efecto, en dicho plan, se identificó claramente el público objetivo y se establecieron los medios mediante los cuales se realizará la publicidad y difusión de la propuesta además de todos los componentes del marketing mix. En las secciones posteriores se detalla en plan de operaciones, la estructura organizacional y de recursos humanos, y finalmente se ejecuta un análisis financiero, el cual ha permitido confirmar la factibilidad del proyecto.<br>The objective of this research work is to develop the business proposal, through a virtual platform, for the care of domestic pets, aimed at the owners of these pets and veterinary centers that in recent years have had a growth of 76% at the National Level and 91% only in Metropolitan Lima. A growing market of pet owners (users) has been identified that is looking every day for specialized veterinary centers that provide a high quality service, online connectivity, with updated information and at the same time at the minimum necessary costs. Likewise, veterinary centers (clients) that seek to connect them with their target audience through social networks and all computer media that increase their client portfolio and that allow them to develop the advertising of their services at low costs. In the first part of the work, the strategic planning has been developed in order to collect valuable information that has allowed to corroborate assumptions and determine the points most valued by users and customers with respect to the value proposition and thus be able to use them in the phase later, that is, in our marketing plan. Indeed, in said plan, the target audience was clearly identified and the means by which the proposal will be publicized and disseminated, as well as all the components of the marketing mix, were established. In the subsequent sections, the operations plan, the organizational and human resources structure are detailed, and finally a financial analysis is carried out, which has confirmed the feasibility of the project.<br>Trabajo de investigación

El presente proyecto consiste en un modelo de negocio para la creación de una empresa llamada Service Pet 360 que unifica los servicios principales para atender a perros y gatos, los cuales abarcan servicios de salud, alimentación, entretenimiento, etc. que llegará al cliente mediante una página web donde se podrán encontrar diferentes empresas reconocidas que trabajan con calidad y compromiso para la total seguridad de nuestros clientes (perros y gatos), de esta forma le otorgamos a nuestros usuarios (dueños de mascotas) la posibilidad de escoger entre una cartera de opciones de atención, productos de la más alta calidad y que se encuentren cerca de su ubicación. The Walking Pet, que conecta a través de una aplicación para dispositivos móviles y/o de una página responsive web (que se adapta a varios dispositivos y sistemas operativos), una base de datos de paseadores con una base de datos de clientes registrados para que se acuerden servicios de paseos de perros. El modelo de negocio es uno del tipo de economías colaborativas: los paseadores no son empleados del modelo, son los proveedores directos de la solución del problema. Los ingresos del modelo de negocio son las comisiones que The Walking Pet cobre a los paseadores, de acuerdo a la cantidad de perros que conformen el grupo de salida del paseo. La escala de comisiones es la siguiente por paseos de 30 minutos a una tarifa de S/15 por paseo por perro. A mayor número de perros por grupo de salida, mayores las comisiones porcentuales para el modelo, y a su vez a valores absolutos, mayores los ingresos percibidos por los paseadores (por grupo de salida). La propuesta de valor de nuestro modelo de negocio se fundamenta en cuatro características: Confianza, Seguridad, Oportunidad y paseos en pequeños grupos. Para la característica de seguridad, nos apoyamos en las tecnologías digitales de geo localización y de transmisión de imágenes y videos por internet. La oportunidad es aquella característica que permite al cliente seleccionar el horario o la agenda que mejor se acomode a su necesidad y para ello se ha de contar con la suficiente cantidad de paseadores registrados. Inicialmente foi avaliada a miscibilidade da blenda de poli(tereftalatode etileno) [PET] com poliestireno [PS]. Sendo esta blenda imiscível em toda a faixa de composição, foi estudada a influência da adição de SMA nas propriedades térmicas e na morfologia da blenda PET/PS/SMA. O parâmetro de interação de Flory-Huggings (c12), cujo valor está relacionado com as interações específicas entre os componentes, foi calculado a partir das temperaturas de fusão de equilíbrio do componente semicristalino puro e na blenda, como igual a +0,024. O valor positivo sugere interações desfavoráveis entre os componentes, em concordância com os outros métodos. A viabilidade de reutilização do PET em mistura com outro polímero, foi analisada através de sucessivos processos de extusão, de maneira que as propriedades microscópicas dos componentes puros e das blendas foram estudadas. A pequena redução da massa molar média do PET, não deve prejudicar o reaproveitamento do PET pós-consumo. Para as blendas PET/PS/SMA, o aumento da adesão interfacial, de certo modo, minimiza o efeito causado pela redução de massa molar durante o processo de reciclagem.

Atualmente, o alerta prévio de desastres naturais e ambientais iminentes, a previsão acurada do clima e compreensão detalhada do status dos recursos hídricos globais são assuntos cotidianos extremamente importantes para a comunidade global. Os Serviços Nacionais de Meteorologia e Hidrologia em todo o mundo são responsáveis por fornecer essas informações, que são necessárias para a proteção do meio ambiente, desenvolvimento econômico (transporte, energia, agricultura, etc.) e a segurança da vida e da propriedade. Neste contexto, um dos serviços amplamente utilizado mundialmente é o Sistema Internacional de Coleta de Dados (International Data Collection System) que é composto por redes de satélites geoestacionários e de órbita baixa (não-geoestacionário). O INPE desenvolve e opera o Sistema Brasileiro de Coleta de Dados Ambientais (SBCDA) que é composto basicamente pelos satélites de Coleta de Dados (SCDs) 1 e 2, e Satélite Sino Brasileiro de Recursos Terrestres (CBERS) 4, e pelas cerca de 1100 Plataformas de Coleta de Dados (PCDs) distribuídas no território brasileiro, e pelas Estações Terrenas de Recepção (ETRs) em Cuiabá-MT, e Alcântara-MA, e pelo Centro de Missão (CM) no Centro Regional do Nordeste do INPE (INPE/CRN) em Natal, RN. O SBCDA opera desde 1993 utilizando uma tecnologia analógica que vem se tornando obsoleta, a qual naturalmente deverá ser substituída por sistemas digitais com decodificação e armazenamento de dados a bordo. Neste cenário, a sincronização de frequência e fase da portadora e a estimação de tempo de símbolo é de fundamental importância para a recepção correta do sinal que chega ao receptor. Portanto, foi proposto o desenvolvimento de uma solução para estas funções utilizando processamento digital de sinais através de códigos em MatLab. Para a implementação da sincronização de frequência foi utilizado um PLL Digital de segunda ordem, levando em consideração requisitos como o efeito Doppler. Para o estimador de tempo de símbolo foi utilizado o algoritmo com alimentação direta x (feedforward), o qual utiliza uma estrutura de simples implementação, uma simplificação do proposto em [16]. Também foi adotada uma solução simples de um interpolador linear para determinação do tempo de atraso. O resultado apresentado pelo sincronizador de frequência / fase foi satisfatório para operação em parte da faixa. O estimador de tempo de símbolos também apresentou resultado satisfatório para Eb/N0 na faixa de 0 a 11 dB, com uma perda menor que 0,5 dB em relação ao valor teórico.<br>Nowadays previous warning of impending natural and environmental disasters, accurate climate prediction, and detailed understanding of the status of global water resources are everyday issues that are extremely important to the global community. The National Meteorological and Hydrological Services throughout the world are responsible for providing this information, which is necessary for the protection of the environment, economic development (transportation, energy, agriculture, etc.) and the safety of life and property. In this context, one of the services widely used worldwide is the International Data Collection System which is composed of geostationary and low-orbit (nongeostationary) satellite networks. INPE develops and operates the Brazilian Data Collection System (BDCS), which is basically composed by the Data Collection Satellite (DCS) 1 and 2, the China-Brazilian Earth Resources Satellite (CBERS) 4, 1100+ Data Collection Platforms (DCPs) distributed in the Brazilian territory, two Receiving Ground Stations (RGS) at Cuiabá-MT, and Alcântara-MA, and one MISSION Center (MC) at the INPE Northeast Regional Center (INPE/NRC) at Natal, RN The BDCS has been operating since 1993 using an analog technology that has become obsolete and must be replaced by digital systems with on-board data decoding and storage. In this scenario, carrier frequency and phase synchronization and symbol time estimation have a fundamental importance for the correct reception of the signal arriving at the receiver. Therefore, it was proposed the development of a solution for these functions using digital signal processing using MatLab codes. For the implementation of frequency synchronization, the chosen solution uses xii a second-order Digital PLL, taking into account requirements such as the Doppler Effect. For the symbol time estimator we used the feedforward algorithm, which uses a simple implementation structure, a simplification of the one proposed in [16]. Also a simple solution of a linear interpolator was used to determine the delay time. The result presented by the frequency / phase synchronizer was satisfactory for operation in part of the range. The symbol time estimator also presented satisfactory results for Eb / N0 in the 0 to 11 dB range, with a loss of less than 0.5 dB over the theoretical value.

This thesis deals with the recycling of polyethylene terephthalate and its subsequent processing into the form of a regranulate meeting the parameters required for food use. A brief description of recycling technology, PET flake quality rating and input material contamination is provided. Additionally, legislation regarding recycling for food purposes is discussed and regranulation technologies are described. The suitability of these technologies for processing specific flake samples has been confirmed. In the experimental part, differences in the regranulate samples are compared using DSC and FT-IR. It has been found that there are no major differences between food quality samples. Tests were carried out on 200 PET bottles (bottle, label), on which the input material was divided into 16 groups. Groups that are unsuitable for recycling with this technology was defined. In addition, contaminants of the final product were examined. Their approximate composition was determined and the relationship between the labels of the bottles at the inlet and the impurities at the outlet was found. Appropriate solutions have been proposed to reduce this contamination

We studied the H- 'H POT. +' e HE- 'H POT. +' molecules introducing the screening effects via modification of the nuclear repulsion energy between atoms allowing thus to understanding the effects in the bond length and energy. The relevance of this work is of prime importance in material science due to many technological applications as molecules confined to surfaces, in liquid helium, neutral plasma and catalysis. All this applications has a common background of knowledge: the influence of the material physical and chemical properties with the distance and energy between the atoms in the chemical composition of the material.

A vida útil das hortaliças é beneficiada pela conservação em temperatura em torno de 10oC. Em climas quentes e secos, como no semi-árido nordestino, esta condição é possível mediante equipamentos de refrigeração que demandam custos e energia elétrica, condição esta incompatível com as das populações de baixa renda. O sistema Pot in Pot desenvolvido na Nigéria, consiste na superposição de dois potes de cerâmica de diferentes tamanhos dispostos um dentro do outro e de uma camada de areia molhada inserida entre eles que proporciona o resfriamento do pote interno pela evaporação da água, conseguindo-se aumentar o tempo de vida útil de vegetais. Com o objetivo de avaliar o efeito deste sistema de conservação sobre hortaliças produzidas no Nordeste brasileiro foi desenvolvida esta pesquisa comparando o sistema Pot in Pot (27±0,40C e 83±0,6%UR) com o ambiente de uma câmara climatizada para a condição semelhante a do semi-árido nordestino (37±0,7ºC e 40±0,5%UR) e com outro sistema convencional de conservação: refrigeração em geladeira doméstica (7±0,30C e 53±1%UR), utilizando-se amostras sem embalagem e amostras embaladas em polietileno de alta densidade. Dois tipos de hortaliças foram utilizados: pimentão e tomate, adquiridos no comércio atacadista. Os experimentos foram realizados em 3 repetições e em cada utilizando-se 72 unidades de tomate e 80 de pimentão. Todas as amostras foram selecionadas, limpas, identificadas, pesadas e fotografadas. Em seguida foi retirada uma unidade de cada amostra para ensaios laboratoriais de pH, sólidos solúveis totais (SST), acidez total titulável (ATT) e textura, correspondendo ao tempo zero do experimento. As demais amostras foram divididas em 2 porções iguais onde uma delas foi submetida a sanitizacão. Em seguida, todas as amostras sanitizadas ou não, foram armazenadas nos 3 diferentes ambientes. A cada 7 dias, as amostras foram analisadas quanto às características visuais (cor, brilho, frescor, turgência e enrugamento), perdas de peso, pH, acidez total titulável, sólidos solúveis totais e textura até apresentarem sinais de alteração que as tornaram impróprias para o consumo. Nas análises físico-químicas utilisou-se métodos preconizados por AOAC. Os resultados demonstraram diferenças significativas (p<0,05) entre os ambientes e os períodos de armazenamento. Concluiu-se que: o uso do sistema Pot in Pot proporcionou aumento da vida útil das hortaliças estudadas e foi o mais eficiente para a conservação do tomate; a refrigeração com embalagem foi o melhor ambiente para o pimentão; as características visuais e o aparecimento de fungos foram limitantes na vida útil de pimentão e a turgescência para a conservação de tomate; a sanitização com cloro não contribuiu para a conservação das hortaliças; o aparecimento de fungos em todos os sistemas utilizados aponta para a necessidade da associação de fungicida específico e que a eficiência do sistema Pot in Pot é dependente das características intrínsecas das hortaliças

Denna rapport har för avsikt att att beskriva arbetet kring utvecklingen av verktyget PET – Plate Evaluation Tool, som är en applikation avsedd att skanna, presentera, spara samt utvärdera målade plåtar i målningslinjer på SSAB Special Steels i Oxelösund. Till hjälp används det öppna biblioteket OpenCV som är ett bibliotek för datorseende. Med hjälp av detta vävs en bild samman när plåtar passerar en eller två kameror. Släpljus från en ljusramp som genererar en homogen ljusbild används för att tydligare framhäva brister. Verktyget identifierar sedan bristerna på plåtens undersida i syfte att hjälpa operatören att se dessa. Resultaten presenteras sedan bl.a. i ett webb-verktyg som använder sig av Java EE-standarden samt PrimeFaces. Webb-verktyget skapar spårbarhet som förenklar mycket arbete kring eventuella reklamationer, eller andra kvalitetsförbättringar och vilket genomslag dessa får. För operatörernas del presenteras resultaten i en skräddarsydd bild skriven i Java där operatören kan analysera bilden i mer detalj. Vidare undersöks och jämförs sedan vilka filter och parametrar som är lämpligast, och hittar utvalda brister i plåtar på bästa sätt. Projektet undersöker också möjligheten att genom denna teknik kunna utläsa stämplar som är stansade i plåten. Applikationen har fått mycket bra bemötande från de som använder den mest, nämligen operatörerna på berörda målningslinjer. De två filter som skrevs har visat sig kunna identifiera de brister de skall på ett bra sätt och kvalitén på de bilder som vävs samman har visat sig vara så pass bra att stämplar kan utläsas.

This thesis presents three classes of mechanism that may explain reported associations between pet ownership and health benefits. The first suggests any association is noncausal. Studies 1-3 examine candidate factors to explain both health advantages and likelihood of pet ownership. Type A behaviour was hypothesised to be associated with higher risk of illness and lower likelihood of pet ownership. Hardiness was hypothesised to be associated with better health and increased likelihood of pet ownership. Neither hypothesis was supported. The second class of explanation suggests that pets indirectly effect health by acting as social facilitators of human-human interactions. Enhanced social contact may lead to health advantages. Studies 4-6 examine the robustness of the catalysis effect of Pets and its impact on owners' social networks. Whilst the catalysis effect was found to be robust in generating social contacts, these were superficial and not regarded as providing relationship functions likely to enhance health. The third class of explanation suggests pets have direct effects on health through the nature of the relationship with the owner, or through physiological effects such as reduced cardiovascular arousal to stress. Study 7 indicates that pets serve valuable supportive functions for normal children. Study 8 found that young people with autism demonstrate positive behaviours within their relationships with pets which they do not with people. Study 9 found the relationships between people with physical disabilities and their service dogs serve many supportive, as well as instrumental, functions and that this is associated with better self-perceived health. Studies 10 and 11 found no evidence that pet presence moderates cardiovascular reactivity to a laboratory stress task. Little evidence was found of an association between pet ownership and health advantages, although it is clear that pets can be significant and valued relationships for their owners.

En el presente trabajo de investigación, se desarrolla la validación de la idea de negocio de la creación del aplicativo Pet & Vet Club. Se trata de una plataforma de servicios integrados para mascotas: teleconsultas, entrenamiento y tienda online de productos. Este negocio está dirigido a los dueños de mascotas, tanto hombres como mujeres entre los 18 y 50 años, residentes de Lima Metropolitana y pertenecientes a los niveles socioeconómicos A y B. Se estableció la tendencia de crecimiento de servicios relacionados a las mascotas como marco de referencia para determinar el potencial del modelo de negocio. Tras el análisis de fuentes primarias y secundarias, se pudo observar que tanto el mercado de teleconsultas como el de compras online para mascotas presentan un importante desarrollo que puede ser aprovechado por la presente aplicación, debido a que no existen competidores con la oferta exacta. La propuesta de valor del modelo de negocio demostró su viabilidad en el mercado limeño con el objetivo de resolver las necesidades de los nuevos consumidores. Por último, se realizó el plan financiero del proyecto con la finalidad de evaluar la viabilidad del proyecto. Se identificó la inversión inicial necesaria, los costos operativos, los gastos administrativos y de ventas, así como también se realizó una estimación de las ventas en base al concierge y a fuentes secundarias. Finalmente, se concluyó que el proyecto es viable, dado que, se obtuvo un VAN positivo, una TIR mayor a la tasa de descuento y el PRD es de 2,72 años.<br>In this research, the validation of the business idea for the creation of the Pet & Vet Club application is developed. It is a platform of integrated services for pets: teleconsultations, training and online products store. This business is aimed at pet owners, both men and women between 18 and 50 years old, who reside in Metropolitan Lima and belong to the A and B socioeconomic levels. The growth trend of pet-related services was established as a frame of reference to determine the potential of the business model. After the analysis of primary and secondary sources, it was observed that both the market for teleconsultations and the online shopping for pets present an important development that can be taken advantage of by this application, since there are no competitors with the exact offer. The value proposition demonstrated its viability in the Lima market in order to meet the needs of new consumers. Finally, the financial plan of the project was carried out in order to evaluate the feasibility of the project. The initial investment, operating costs, administrative and sales expenses were identified, as well as an estimate of sales based on the concierge and secondary sources. Finally, it was concluded that the project is viable given a positive NPV, an IRR greater than the discount rate and the DPP of 2.72 years.<br>Trabajo de investigación

The financial situation of limited company PFT is evaluated in the period 2008 to 2013 through techniques of financial analysis. Then the resulting values are compared with the recommended values, sector averages and values of selected competing companies with limited liability. Multivariate methods of intercompany comparison are applied for a more comprehensive assessment of financial management. The conclusion is focused on summarizing the results, identifying strengths and weaknesses and proposing appropriate measures.
