Abstract:
A study was designed to screen and analyze the efficient phosphate solubilizing bacteria (PSBs) from wheat rhizosphere. Five biovars of Pseudomonas fluorescens (PSM1, PSM2, PSM3, PSM4 and PSM5) were isolated from wheat rhizosphere and Bacillus megaterium MTCC 8755 procured from microbial type culture collection (MTCC) Chandigarh, India. The P. fluorescens biovar PSM1 was observed to be most efficient phosphate solubilizer. Inoculation of P. fluorescens PSM1 and B. megaterium MTCC 8755, alone and in combination, caused the highest phosphate solubilization at pH 5. At this pH, maximum phosphate solubilization was observed with B. megaterium MTCC 8755 inoculation (8.2 mg mL-1) on sixth day of incubation with P. fluorescens PSM1 (8 mg mL-1) on seventh day of incubation and with dual bacterial treatment (10.5 mg mL-1) on the fourth day of incubation. A correlation coefficient of linear regression equation of phosphate solubilization with pH indicated that pH value of the medium was directly correlated with tricalcium phosphate solubilization. The study will help in choosing soil pH specific PSB inoculant for optimizing plant growth.

Persistent slip markings (PSMs) were experimentally studied in 316L steel fatigued to early stages of the fatigue life. High resolution SEM, combined with focused ion beam (FIB) technique and atomic force microscopy (AFM) were used to assess the true shape of PSMs in their early stage of development. General features of PSMs in fatigued metals are extrusions and intrusions. Their characteristic features were determined. They were discussed in relation with the theories of surface relief formation and fatigue crack initiation based on the formation, migration and annihilation of point defects in the bands of intensive cyclic slip - persistent slip bands (PSBs)

Abstract:
In this work, polystyrene microspheres (PSMS) and PSMS-based polymer nanocomposites have been prepared. PSMS were first synthesized by dispersion polymerization of styrene monomer. The microsphere- coated-multi-walled carbon nanotube (PSMS/MWCNT) nanocomposite was prepared by incorporating MWCNT into PSMS matrix. Aniline monomer was then introduced by means of in situ oxidative polymerization (PANI) route to PSMS, MWCNT, and PSMS/MWCNT. Fourier transform infrared spectroscopy depicted the composite formation of PANI and MWCNT with PSMS matrix. PANI polymer was successfully polymerized in PSMS matrix and on carbon nanotube walls. According to scanning electron microscopy, all nanocomposites exhibited core-shell morphology. Glass transition temperature (Tg) of PSMS/PANI, PANI/MWCNT, and PSMS/PANI/MWCNT were found as 245℃, 287℃, and 289℃, while maximum decomposition temperature (Tmax) was 387℃, 575℃, and 580℃, respectively. Tetrahydrofuran was shown to be a suitable solvent for dispersing the nanocomposite. Toluene can also be used as suitable solvent for these nanocomposites. X-ray diffraction analysis depicted amorphous form of PSMS; however, PANI and MWCNT reduced the amorphous nature of the microspheres. PANI/MWCNT showed the highest crystallinity among all nanocomposites. Moreover, PSMS/PANI/MWCNT nanocomposite revealed electromagnetic interference shielding effectiveness of ∼23.2 dB.

Two fatigued materials with f.c.c. lattice, i.e. pure polycrystalline copper and austenitic Sanicro 25 stainless steel, were subjected to the study of the persistent slip markings (PSMs) developed on the surface of the suitably oriented grains. They were observed using scanning electron microscopy (SEM) and thin surface FIB lamellae were prepared and studied by transmission electron microscopy (TEM). The aim was to correlate the specimen surface profile with the underlying internal dislocation structure. The localization of the intensive cyclic slip into persistent slip bands (PSBs) of the material was observed and associated with the PSMs on the specimen surface. Extrusions, intrusions and the dislocation structure appertaining to them were analysed, documented and discussed in relation to the models of fatigue crack initiation.

Abstract:
Purpose: We assessed clinical and tumor biological features and evaluated their association with positive surgical margins (PSMs) and positive apical margins (PAMs) variability after radical prostatectomy (RP) in men with non-metastasis prostate cancer (nmPCa) in our institute. Patients and methods: During the period from January 2013 to December 2017, clinical and pathological data were collected in 200 patients with nmPCa undergoing RP in the Urological department of Peking University Cancer Hospital & Institute. Surgical and apical margins were stated negative and positive, separately. A dichotomous logistic regression model was used to assess clinical and tumor biological features including age, total prostate volume (TPV), biopsy positive cores (BPC), D’Amico risk grade, tumor clinical stage, International Society of Urologic Pathology (ISUP) grade, tPSA, f/t and pelvic lymph nodes (PLN) invasion, and their association with PSMs and PAMs was evaluated. Results: Overall, men with nmPCa in this study had a high ISUP grade (58.5% grade 3–5), high risk grade (89.4%) and high clinical T stage (56% cT3-4). PSMs were detected in 106 patients; the rate of PSMs was 53%. Among patients with PSMs, 83% were PAMs; the overall rate of PAMs was 44%. Among patients with PSMs, high risk (OR, 1.439; p = 0.023), cT3a (OR, 1.737; p = 0.045), cT3b (OR, 5.286; p < 0.001), cT4 (OR, 6.12; p < 0.001), ISUP Grade 4 (OR, 2; p = 0.034) and Grade 5 (OR, 6.167; p < 0.001) and PLN invasion (OR, 6; p = 0.019) were strongly associated with PSMs using a dichotomous logistic regression univariable model, and high risk (OR, 6; p = 0.019), cT3a (OR, 5.116; p = 0.048), cT3b (OR, 9.194; p = 0.008), cT4 (OR, 4.58; p = 0.01), ISUP Grade 4 (OR, 7.04; p = 0.035), Grade 5 (OR, 16.514; p = 0.002) and PLN invasion (OR, 5.516; p = 0.03) were independently associated with PSMs by using multivariable analysis. Among patients with PAMs, cT3b (OR, 2.667; p = 0.004), cT4 (OR, 3; p = 0.034) and proportion of BPC (OR, 4.594; p = 0.027) were strongly associated with PAMs by using a dichotomous logistic regression univariable model, and cT3b (OR, 3.899; p = 0.02), cT4 (OR, 2.8; p = 0.041) and proportion of BPC (OR, 5.247; p = 0.04) were independently associated with PSMs by using multivariable analysis. Conclusions: Patients with nmPCa in our institute had high risk, high ISUP grade and high clinical stage. Tumor biological factors were strongly associated with PSMs and PAMs, and PLN invasion was independently associated with PSMs. The risk factors influenced the status of surgical margins, and apical margins were different.

Cyclic plastic straining in crystalline materials is localized to persistent slip bands (PSBs) and results in formation of persistent slip markings (PSMs) consisting of extrusions and intrusions. Intensive plastic strain in PSBs results in dislocation interactions and formation of point defects. The extended model based on point defect formation, migration and annihilation is presented describing surface relief formation in the form of extrusion-intrusion pairs. Point defect migration and resulting mass transfer is the principle source of cyclic slip irreversibility leading to crack-like defects - intrusions. Fatigue cracks start in the tip of sharp intrusions.

Abstract:
The study analyses strategies and persuasive appeals in the public service messages aired on Pakistani TV channels during 2016-2018, that measures the effects of PSMs on audience behaviour. The PSMs fails to deliver messages due to dissimilar socio-economic backdrop and scope of the audience understanding. The purposive sampling of six selected PSM is on the bases of health themed PSM’s. Objective is to investigate the characteristics of ads, role of the message, major health issues, sources of the message and appeals. The AIDA model, persuasion theory and social responsibility theory used to filter the textual analysis of the selected PSMs. It was found that the PSMs presented health issues while using celebrity endorsement and persuasive message appeals as persuasive techniques. The PSMs such as breast cancer persuaded women for self-care, calcium deficiency PSM to calcium intakes, heat stroke PSM to precautionary measures during emergency situations, the malnutrition PSM to women healthy diet during pregnancy. The study recommends that advertisers, producers and TV channels should adopt audience perception based important strategies for social welfare, while the state must promote them.

Background:Human immunodeficiency virus-1 (HIV-1) mutates rapidly to escape host immune pressure. This results in the generation of positively selected mutations (PSM) throughout the viral genome. Escape mutations in Nef, one of the accessory proteins of HIV-1, which plays an important role in viral pathogenicity have previously been identified in several large cohort studies, but the evolution of PSMs overtime in various HIV-1 subtypes remains unknown.Methods:161 clade A1, 3093 clade B, 647 clade C and 115 clade D HIV-1 nef sequences were obtained from the HIV Database of Los Alamos National Laboratory and aligned using MEGA 6.0. The sequences from each clade were grouped based on the year of collection. Quasi analysis was used to identify PSMs and the number and locations of PSMs were compared among different subtypes.Results:PSMs for all four subtypes were distributed across the sequence of Nef, and conserved residues F90, W113, PxxPxR (a.a 72-77) remain unaltered overtime. The frequency of PSMs was stable among subtype B sequences but increased overtime for other subtypes. Phylogenetic analysis shows that sequences containing PSMs tend to cluster together at both inter and intra- subtype levels.Conclusion:Identification of PSMs and their changes overtime within various subtypes of HIV-1 is important in defining global viral evolutionary patterns that can provide insights for designing therapeutic strategies.

Abstract:
Legionella pneumophila (Lp) est une bactérie ubiquitaire dans les environnements aqueux et responsable d’une pneumopathie potentiellement sévère : la légionellose. La majorité des souches impliquées appartiennent au sérogroupe 1 (Lp1) et à un sous- groupe spécifique de souches portant un épitope particulier dites mAb3/1+. Cependant, la différence de distribution entre les souches retrouvées dans l’environnement et celles impliquées en clinique n’est pas clairement élucidée. Notre travail a porté sur la détection de deux facteurs de virulence de Lp. Nous avons voulu mettre en évidence l’existence de Phenols Soluble Modulines (PSMs), peptides uniquement décrit chez Staphylocoques et avons ainsi pu démontrer l’activité de peptides prédits par analyse in silico chez Lp capables d’activer la réponse inflammatoire par la voie du NF-?B et sont dotés d’une action cytotoxique. Notre deuxième axe d’étude a porté sur le lipopolysaccharide (LPS) de Lp. Afin de vérifier si la prédominance de certaines souches était liée à un biais diagnostique, nous avons voulu tout d’abord vérifier la sensibilité de 3 tests urinaires diagnostiques envers le LPS extrait de souches de différents sous- groupes de Lp1 et sérogroupes de Lp et avons ainsi pu montrer que ces tests sont capables de détecter tous les LPS de Lp1. La sensibilité envers le LPS des autres sérogroupes est très variable mais reste insuffisante pour permettre leur détection. Nous avons ensuite utilisé ces LPS extraits pour vérifier la réponse immunitaire innée en fonction des souches de Lp1. Ainsi les souches mAb3/1+ activent moins le système immunitaire que les souches mAb3/1-, ce qui pourrait expliquer alors une moins bonne clairance de ces souches permettant leur multiplication à l’origine d’une infection. Au final, notre travail a permis d’étudier deux facteurs de virulence potentiels au sein de Lp, pouvant expliquer partiellement la prédominance de certaines souches en pathologie humaine
Legionella pneumophila (Lp) is a ubiquitous intracellular bacterium found widely in the environment and is the cause of an opportunistic infection named legionellosis. The majority of the strains involved belong to serogroup 1 (Lp1) and to a specific subgroup named mAb3/1+, linked to a specific epitope expressed at the cell membrane. However the distribution difference between the strains found in the environment and the ones involved in pathology is not fully understood. We here studied two virulence factors of Lp. We first demonstrated the existence of Phenols Soluble Modulines (PSMs), smalls peptides that only have been described for Staphylococcus and found that the peptides that were predicted for Lp by in silico analysis were able to activate the innate immune response by NF-?B pathway and were able to have a cytotoxic activity. We also studied the lipopolysaccharide (LPS) of Lp. To found out if the predominance of some strains was linked to a diagnosis biais, we first evaluated the sensitivity of 3 urinary antigens tests against extracted LPS of strains belonging to all the sous-groups of Lp1 and serogroups of Lp. We then demonstrated that those tests are able to detect all LPS of Lp1, independently of mAb3/1 character. The sensitivities of the 3 tests were very variable for the other serogroups of Lp, but were too low to be able to detect those LPS in practice. We then used these extracted LPS to evaluate the innate immune response for different strains of Lp1. We demonstrated that mAb3/1- strains needed lower dose of LPS to activate the innate immune response than mAb3/1+ strains, which could be linked to a better clearance of the bacteria from the host, which doesn’t develop an infection. This work has studied two potentially virulent factors of Lp, which could partially explain the predominance of some strains of Lp in human pathology

Abstract:
Pokročilá vysoce legovaná austenitická nerezová ocel Sanicro 25 s Fe-Ni-Cr matricí byla studována za podmínek nízkocyklové únavy za pokojové a vysoké teploty 700 °C. Široká škála moderních experimentálních technik byla použita ke studiu vzájemně souvisejících efektů chemického složení slitiny, mikrostrukturních změn a deformačních mechanismů, které určují odolnost materiálu vůči poškození. Hlavní úsilí bylo zaměřeno na studium iniciace únavových trhlin a růstu krátkých trhlin, tedy dvě stádia, která hrají zásadní roli ve výsledné celkové délce únavového života materiálu v provozu. • Vnitřní deformační mechanismy byly korelovány s vývojem povrchového reliéfu, který je pozorován ve formě persistentních skluzových stop na povrchu. Bylo zjištěno, že vysoce planární charakter dislokačního skluzu způsobuje vysokou lokalizaci cyklické plastické deformace do persistentních skluzových pásů, což v důsledku vede k nukleaci “Stage I” trhlin, která je spojena s přítomností persistentních skluzových stop na povrchu ve všech studovaných vzorcích. Bylo zjištěno, že praskání dvojčatových hraníc je taktéž spojeno s přítomností persistentních skluzových stop podél povrchové stopy dvojčatové roviny. • Interkrystalická iniciace únavové trhliny byla pozorována pouze zřídka, a to za podmínek zatěžování amplitudami vysoké deformace. Bylo zjištěno že interkrystalická iniciace je spojena s přítomností persistentních skluzových stop na hranicích zrn. Hranice zrn praskají za podmínek externího tahového zatížení zejména z důvodu vysokého počtu nekompatibilit na hranicích zrn, které jsou způsobené tvarem persistentních skluzových stop. • Mechanismy růstu přirozených krátkých trhlin byly studovány na vzorcích vystavených nízkocyklove únava s nízkou i vysokou deformací. Role mikrostruktury byla analyzována pomocí experimentálních technik a diskutována. • Rychlosti šíření nejdelších trhlin byly měřeny na vzorcích s mělkým vrubem. Výsledky byly analyzovány použitím přístupů lomové mechaniky založených na amplitudě KI a J-integrálu stejně jako na amplitudě plastické deformace. Všechny přístupy byly diskutovány v souvislosti s Mansonovými-Coffinovými křivkami únavové životnosti. Jednoduchý mocninový zákon růstu krátkých trhlin založený na amplitudě plastické deformace ukazuje velice dobrou korelaci se zákonem únavové životnosti. • Byla studována role oxidace v podmínkách cyklického zatěžování za vysokých teplot. Bylo zjištěno, že křehké praskání zoxidovaných hranic zrn hraje hlavní roli v počátečních stádiích nukleace trhlin. Později po iniciaci se dráha růstu trhliny mění preferenčně na transkrystalickou. Dráha šíření trhlin je velmi podobná dráze zjištěné při cyklování za pokojové teploty.

Private Information Retrieval is the problem of querying two servers to nd a value in a database, while keeping the index private. We extend this problem to Generalized Wildcard PIR, where we instead query an aggregate of the entries whose indices match a pattern, called a generalized wildcard, which species what values each segment of the indices may take. We give a construction for this variant with similar communication to that of the best PIR protocols known. We study information theoretic models in cryptography, namely Private Information Retrieval, Conditional Disclosure of Secrets, and Private Simultaneous Messages. We give extensions of PIR and CDS in the area of generalized wildcards, and give constructions for those extensions. We discuss directions towards more ecient protocols, and raise open questions.

This report documents an MSc Thesis work of sub-transmission reliability study using PSS/Eperformed at Vattenfall. The network that is examined in this thesis is a sub-transmission network.Vattenfall Distribution Sweden is going to invest and rebuild the network in their 70 and 130 kVmeshed sub transmission network. To invest and rebuild in a network is very expensive. The gridowners want a reliable system that has a high reliability at a low cost.The main focus in this thesis is to examine different substation layouts. Calculating the failureintensities and unavailability for the different substation layouts gives the opportunity to do a life cyclecosts (LCC) on the different substations. With help of LCC calculations it is possible to find thesubstation that gives the lowest cost during a life time. Choosing right equipment in the grid will helpthe Vattenfall network company lower their equipment costs.The thesis work consists of mainly three parts as follows. Network modeling in PSS/EA subset of the existing sub transmission network was used in the project. This subset was taken outfrom the complete network model and modeled in PSS/E. Validation of PSS/E reliability modulesPSS/E reliability modules were new and have not been used earlier by the Vattenfall networkcompany. To be sure that the program calculates correctly a validation was performed. The main focusof the validation has been in looking if the program calculates the interrupted power correctly. Reliability analysis of different substation layoutsFor each of the substation layout the reliability was calculated. On component level (e.g. breaker,disconnector) different types of configuration were studied. To find out the best layout from aneconomical point of view a LCC calculation was also performed.The LCC calculation on the different substation layouts showed that the investment costs affect theresult very much. When the active power through the substation is low it is preferred to use asubstation that has low investment costs. At active power that is 30 MW or higher it is preferred to usethe substation with higher reliability (having higher investment cost). The result shows that using acombined breaker and disconnector instead of circuit breaker could lower the failure intensity andunavailability of substations.

Saliency detection is a long standing problem in computer vision. Tremendous efforts have been focused on exploring a universal saliency model across users despite their differences in gender, race, age, etc. Yet recent psychology studies suggest that saliency is highly specific than universal: individuals exhibit heterogeneous gaze patterns when viewing an identical scene containing multiple salient objects. In this paper, we first show that such heterogeneity is common and critical for reliable saliency prediction. Our study also produces the first database of personalized saliency maps (PSMs). We model PSM based on universal saliency map (USM) shared by different participants and adopt a multi-task CNN framework to estimate the discrepancy between PSM and USM. Comprehensive experiments demonstrate that our new PSM model and prediction scheme are effective and reliable.

As many pipeline operators embark on the journey of developing a Pipeline Safety Management System (PSMS), the first question is typically, “Where do we begin?” Management systems can be intimidating, and the thought of taking on the task of developing one can seem overwhelming. Companies want to know if it is necessary to start from scratch, if they can use existing processes or programs, and which step to take first for a successful PSMS. There are many ways to begin, but one of the most effective ways is to first ask two questions, “What do we already have?” and “What are our biggest problems?” Armed with these answers, a path forward can be developed, and the foundation for the management system can begin to take shape. One effective way to choose where to begin when developing the PSMS is to determine which elements have been related to the root causes of incidents and near misses in the past. Likewise, continuing to determine and monitor the causes of incidents after the implementation of the PSMS will provide guidance for continual improvement of the management system. Using the elements and sub-elements of existing management system standards or practices, such as API RP 1173, Pipeline Safety Management System Requirements [1], as a starting point for determining root causes is a good way to break down, categorize, and trend the causes of each incident. Combining these with a gap analysis of both the undocumented and documented processes and procedures will provide a basis for determining the priorities for development and implementation of each management system element.

Abstract:
Pressure sensitive paint (PSP) technique is based on the interaction of oxygen molecules with luminescent molecules, and it seems suitable for analyses of high Knudsen number flows which require diagnostic tools in the molecular level. However, application of the PSP technique to micro-devices is still very difficult because of the thickness of PSP layers of the order of microns and the aggregation of luminescent molecules caused by the “painting” methods. To resolve the problems of ordinary PSPs mentioned above, we adopt the Langmuir-Blodgett (LB) method to fabricate pressure sensitive molecular films (PSMFs) having nanometer order thickness. The fundamental properties of the PSMFs such as the pressure sensitivity and the surface roughness are examined, to evaluate the feasibility of the PSMF technique for pressure measurement around micro-devices. A PSMF based on palladium (II) mesoporphyrin IX shows high pressure sensitivity in low pressure regime, while a PSMF based on platinum (II) mesoporphyrin IX is optimum for atmospheric pressure. It is also clarified that the PSMFs do not degrade the roughness of solid surfaces. The results indicate the feasibility of the PSMF technique for pressure measurement in high Knudsen number flows such as micro flows.

Inferences from probability-sampling theory (more commonly called “design-based sampling theory”) often rely on the asymptotic normality of nearly unbiased estimators. When constructing a two-sided confidence interval for a mean, the ad hoc practice of determining the degrees of freedom of a probability-sampling variance estimator by subtracting the number of its variance strata from the number of variance primary sampling units (PSUs) can be justified by making usually untenable assumptions about the PSUs. We will investigate the effectiveness of this conventional and an alternative method for determining the effective degrees of freedom of a probability-sampling variance estimator under a stratified cluster sample.
