Abstract:
Mutant Frigid A (fgdA) of Dictyostelium discoideum is defective in a functional Ga2 subunit of a G protein and is characterized by a complete blockade of the cyclic AMP-mediated sensory transduction steps, including cyclic AMP relay, chemotaxis and the cyclic GMP response. Folic acid-mediated transmembrane signal transduction was investigated in this mutant; the results show that: (1) cell surface folic acid receptors are present in fgdA mutants. (2) Folic acid induces intracellular responses, including activation of guanylate cyclase and chemotaxis. (3) The inhibitory effect of GTP on folic acid binding to membranes is present. (4) GTPγS binding and highaffinity GTPase are stimulated by folic acid. These data strongly suggest that folic acid receptors are coupled to guanylate cyclase and chemotaxis via a Ga protein that is different from Ga2. The results imply that surface receptors for cyclic AMP and folic acid are coupled to different G proteins.

Surface proximal convoluted tubules (PCT) in rats were microinfused in situ with [3H]folic acid to study the role of folate binding protein (FBP) in the kidney brush-border membrane for renal conservation and transport of folate [3H]folic acid absorption was linearly related to tubular length of PCT and occurred largely in this segment of the tubule. Unlabeled folate derivatives inhibited [3H]folic acid absorption, the extent of which was dependent on the type of unlabeled folate used and its concentration. At equivalent concentrations, inhibition was most effective with unlabeled folic acid, slightly lower than with 5-methyltetrahydrofolate and least effective with methotrexate. Comparisons between [3H]folic acid absorption before and after infusion of a saturating dose of unlabeled folic acid or repetitive injections of [3H]folic acid into the same tubular site revealed continuous and rapid regeneration of unsaturated folic acid uptake sites with an apparent half-life of 28.75 +/- 8.75 s. Determination of [3H] retained in the tubule at various periods after microinfusion of [3H]folic acid revealed slow cellular disappearance with an apparent half-life of 47.3 +/- 5.4 min. It is proposed that the brush-border FBP functions as a receptor of infused folic acid and that following the binding of the ligand the folic acid/FBP complex undergoes a rapid change that results in the internalization of folic acid and regeneration of unsaturated binding sites at the membrane surface. Internalized folic acid is slowly released into renal capillaries.

Abstract:
In this study, soy protein isolate was hydrolyzed by compound enzymes to give aqueous soy protein with low molecular weights. Folic acid modified and free soy protein nanoparticles were successfully prepared by a desolvation method as target-specific drug delivery, respectively. Ultraviolet spectrophotometry demonstrated that folic acid was successfully grafted onto soy protein. The shape and size of folic acid modified soy protein nanoparticles were detected by transmission electron microscopy, scanning electron microscope, and dynamic light scattering. In addition, a series of characteristics including kinetic stability, pH stability, and time stability were also performed. Doxorubicin was successfully loaded into folic acid modified soy protein nanoparticles, and the encapsulation and loading efficiencies were 96.7% and 23%, respectively. Doxorubicin-loaded folic acid modified soy protein nanoparticles exhibited faster drug release rate than soy protein nanoparticles in PBS solution (pH = 5). The tumor penetration and antitumor experiments were done using three-dimensional multicellular tumor spheroids as the in vitro model. The results proved that folic acid modified soy protein nanoparticles display higher penetration and accumulation than soy protein nanoparticles, therefore possessing efficient growth inhibitory ability against multicellular tumor spheroids.

Abstract:
Diabetic retinopathy (DR) is a severe complication of diabetes, which seriously affects the life quality of patients. Because of the damage caused by DR, there is an urgent need to develop effective drugs. Folic acid, a water-soluble vitamin, is one of the vitamin B complexes. Folic acid is widely found in the meat and vegetables. In the clinic, low folic acid levels in the body may have a certain correlation with DR. However, there is no relevant basic research proving a relationship between folic acid levels and DR. The purpose of this study was therefore to investigate whether folic acid has a protective effect on the retinal vascular endothelial cells against high glucose levels. Moreover, the molecular mechanism of action of folic acid was further explored. The results showed that folic acid significantly suppressed the cell viability, tube length, migrated cells and the percentage of BrdU+ cells compared with the high glucose group. Moreover, folic acid decreased the mRNA expression of TEAD1 and the protein expression of TEAD1 and YAP1. These findings indicate that folic acid can protect retinal vascular endothelial cells from high glucose-induced injury by regulating the proteins in the Hippo signaling pathway.

AbstractObjectiveTo validate intakes of protein, folic acid, retinol and n–3 fatty acids estimated from a food-frequency questionnaire in week 25 of pregnancy (FFQ-25).DesignValidation was done against a 7-day weighed food diary (FD) and biomarkers of the nutrients in gestation week 32–38.Subjects and settingThe FFQ-25 to be validated was used in the Danish National Birth Cohort comprising 101 042 pregnant Danish women, of whom 88 participated in the present validation study.ResultsEstimated intakes of protein, retinol and folic acid did not differ significantly between the two dietary methods, but intake of n–3 fatty acids was one third larger when estimated from the FFQ-25. The intakes estimated from the two dietary methods were all significantly correlated, ranging from 0.20 for retinol intake to 0.57 for folic acid intake. Sensitivities of being correctly classified into low and high quintiles were between 0.22 and 0.77, and specificities were between 0.62 and 0.89. Urinary protein content did not correlate significantly with protein estimated from the FFQ (r = 0.17, P > 0.05), but did with intake estimated from the FD (r = 0.56, P < 0.0001). Erythrocyte folate correlated significantly with the estimated total intake from the FFQ (r = 0.55, P < 0.0001) and the FD (r = 0.52, P < 0.0001). No correlations with plasma retinol were found. Erythrocyte eicosapentaenoic acid (C20:5n-3) correlated significantly with n–3 fatty acids intake estimated from both the FFQ-25 (r = 0.37, P < 0.001) and the FD (r = 0.62, P < 0.0001).ConclusionThe FFQ-25 gives reasonable valid estimates of protein, retinol and folic acid intakes, but seems to overestimate intake of n–3 fatty acids.

Abstract:
Protein particles have been reported as the potential carriers for the co-encapsulation of bioactive components. In this study, lysozyme, a basic protein, was used to simultaneously encapsulate folic acid and α-tocopherol at pH 4.0. The encapsulation efficiency and loading capacity of folic acid or α-tocopherol increased with its respective concentration. Folic acid had no influence on the encapsulation of α-tocopherol. However, the encapsulation of folic acid was improved by α-tocopherol below 40 μg/mL but reduced by α-tocopherol at higher concentrations. The encapsulation by lysozyme shielded folic acid, α-tocopherol, or both partially from the attack of 2,2′-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical cation. No masking effect of lysozyme encapsulation on α-tocopherol was found in DPPH antioxidant activity assay. Furthermore, the DNA coating was used to improve the dispersion of lysozyme with folic acid and α-tocopherol. The lysozyme/DNA particles with folic acid and α-tocopherol showed a homogenous size distribution of 180–220 nm with ζ-potential values between −33 and −36 mV. The release and bioaccessibility of folic acid in lysozyme/DNA with α-tocopherol were similar to that of folic acid alone, while the release of α-tocopherol was delayed and its bioaccessibility was improved by encapsulation in lysozyme/DNA with folic acid. The data gathered here would provide guidance for the use of lysozyme-based co-encapsulating carriers in the development of functional foods.

Maternal alcohol consumption during pregnancy can induce central nervous system abnormalities in the fetus, and folic acid supplementation can reverse some of the effects. The objective of the present study was to investigate prenatal alcohol exposure-induced fetal brain proteome alteration and the protective effect of folic acid using proteomic techniques. Alcohol (5·0 g/kg) was given intragastrically from gestational day (GD) 6 to15, with or without 60·0 mg folic acid/kg given intragastrically during GD1–16 to pregnant Balb/c mice. The control group received distilled water only. Results of litter evaluation on GD18 showed that supplementation of folic acid reversed the prevalence of microcephaly induced by alcohol. Proteomic analysis indicated that, under the dosage of the present investigation, folic acid mainly reversed the alcohol-altered proteins involved in energy production, signal pathways and protein translation, which are all important for central nervous system development.

Targeted drug delivery system is an effective method for the diagnosis and treatment of cancer, it has received much attention because of its low side effects and therapeutic efficacy. Folic acid receptor is highly expressed on the surface of most cancer cells, but is low or not expressed on the surface of normal cells, and the ligand folate has a high affinity. Folic acid receptor is attached to drug carriers and can be targeted to cancer cells. This paper introduces folic acid and folate receptors, briefly describes the mechanism of action of folic acid receptor-mediated targeted drug delivery, discusses the progress on four types of folic acid-folate receptor-mediated cancer treatment: folate acid-conjugated magnetic nanoparticles, drug binding of small-molecule folic acid, folic acid receptor-bound protein, and folic acid-conjugated polysialic acid. It also analyzes the favorable points and future development trends of each treatment mechanism.

It has been demonstrated that proteins covalently conjugated to folic acid may be taken up by cells via endocytosis after binding to a folate binding protein (FBP) in the cell membrane. The proteins taken up in this manner remain catalytically active and they may modify physiological processes occurring in the cytosol. Confocal fluorescence microscopy of KB cells incubated with FITC-bovine serum albumin-folic acid conjugates showed that after uptake, the conjugates resided in large vesicular structures. The purpose of the present study was to determine the subcellular localization of protein-folic acid conjugates in KB cells using folic acid-bovine serum albumin-colloidal gold (F-BSA-CG) as a tracer. F-BSA-CG conjugates were taken up via uncoated pits or caveolae, and resided primarily in multivesicular bodies (MVBs) and other tubular endosomes at early time points (15-60 min). At later time points (6 hours), conjugates were still contained in MVBs but some were also found in secondary lysosomes or free in the cytoplasm. Coincubation of KB cells with transferrin-colloidal gold (TF-CG) and F-BSA-CG resulted in colocalization of TF-CG and F-BSA-CG within endosomal elements at times later than 15 minutes, indicating that the caveolae-mediated F-BSA-CG endocytic pathway converged with a pathway utilized by clathrin-coated pits.

The water-soluble B vitamins, folate and folic acid, play an important role in reproductive health, but little is known about the effects of folic acid on infertility. The present study tested the hypothesis that folic acid affects oocyte maturation, a possible cause of female infertility. We have studied the in vitro maturation of mouse and Xenopus oocytes. Hypoxanthine (Hx) was used as an inhibitor of mouse oocyte maturation to mimic in vivo conditions by maintaining high levels of cyclic-AMP. The frequency of first polar body (PB1) formation and germinal vesicle breakdown (GVBD) in mouse oocytes was decreased by Hx. This effect was counteracted by folic acid added to the medium. PB1 extrusion and GVBD percentages rose to 27·7 and 40·0 % from 12·8 and 19·9 %, respectively, by exposure to 500 μm-folic acid. Folic acid also restored the spindle configuration, which had been elongated by Hx, as well as normalising the distribution of cortical granules (CG). In folic acid-treated Xenopus eggs, extracellular signal-regulated kinase 1 was phosphorylated, cyclin B2 and Mos were up-regulated and the frequency of GVBD was accelerated. Taken together, the findings suggest that folic acid facilitates oocyte maturation by altering the expression and phosphorylation of proteins involved in M-phase-promoting factor and mitogen-activated protein kinase pathways, as well as causing changes in spindle configuration and CG migration.

Abstract:
MTHFD is a folate-dependent trifunctional protein comprised of three activities: N$ sp5$,N$ sp{10}$-methylenetetrahydrofolate dehydrogenase, N$ sp5$,N$ sp{10}$-methenyltetrahydrofolate cyclohydrolase and N$ sp{10}$-formyltetrahydrofolate synthetase. The enzymes catalyse the sequential interconversion of tetrahydrofolate derivatives required for purine, methionine and thymidylate synthesis. A Chinese hamster ovary cell line, reported to have reduced cyclohydrolase activity, was studied to characterize the nature of its mutation.
Enzymatic assays showed reduced activities of all three enzymes. Immunoblotting and immunoprecipitation of radiolabelled cell extracts indicated that the gene product was greatly reduced or absent in the mutant. Southern analysis showed no differences between normal and mutant cells, indicating that the defect was not due to a major gene rearrangement. RNA analysis, by Northern blotting and by RNA amplification using the polymerase chain reaction, showed that a mRNA for MTHFD of normal size was present in mutant cells. These results suggest that the mutation is post-transcriptional and that it disrupts the synthesis of MTHFD.

Abstract:
The objective of this study was to determine Thai women's nutrition awareness of folic acid and its role in the prevention of neural tube defects. Three hundred and two Thai women between 18-45 years completed a 30 itemized survey. There was a significant lack of nutrition awareness of folic acid. Many women (n= 274; 91.9%) had not heard of neural tube defects (NTD) and only 8.1 % (n=24) had heard of spina bifida/NTD. More than 50% (n=176) were unaware of NTD. Only 11.6% of the women (n=35) thought consuming vitamins during pregnancy would reduce the risk of birth defects. More than 80% of the women reported the best time to take folic acid supplement was during pregnancy. Less than 20% of the women indicated taking folic acid supplement before or after pregnancy. In this population, nutrition awareness of folic acid was minimal.

La regulation de la voie de biosynthese de l'acide folique chez bacillus subtilis a ete analysee tant au niveau genetique que enzymatique. Tout d'abord, le premier enzyme de la voie de biosynthese, gtp cyclohydrolase i, a ete surexprime dans b. Subtilis et purifie. Nous avons determine les constantes cinetiques de cet enzyme et recherche de possibles inhibiteurs de l'activite enzymatique. Ces travaux nous ont permi de montrer que la dephosphorylation de l'intermediaire dihydroneopterine triphosphate en dihydroneopterine monophosphate pouvait se produire spontanement in vitro par une simple reaction chimique en presence de cations divalents. D'autre part, nous avons isole et sequence la partie 3' de l'operon acide folique chez bacillus subtilis. Trois nouveaux genes ont ainsi ete decouverts (orf3, orf4, et lyss), ils font parti integrante de l'unite de transcription acide folique. La presence des deux phases ouvertes de lecture, orf3 et orf4, suggere une fonction regulatrice pour ces deux genes a un niveau qui reste a determiner. Nous avons montre que l'operon acide folique est regule de deux facons differentes. D'une part, l'expression depend fortement de la phase de croissance, et d'autre part, nous avons montre l'existence d'un mecanisme de regulation par attenuation de transcription en aval du gene trpg. Nous avons aussi prepare des anticorps diriges contre tous les enzymes de la voie de biosynthese de l'acide folique. A l'aide de ces anticorps, nous avons demontre l'existence de la regulation de traduction du gene trpg par l'intermediaire de la proteine regulatrice trap (tryptophan rna-binding attenuation protein). Nous presentons aussi certaines caracteristiques pour les autres enzymes de la voie de biosynthese. Certains parametres genetiques et enzymatiques qui regulent la synthese d'acide folique dans bacillus subtilis sont decrits dans cette these

AbstractA woman’s body mass index and nutritional status should be assessed in the antenatal period. The evidence indicates that maternal anemia is a risk factor for preterm birth. Ensuring adequate levels of iron and folic acid is essential for general pregnancy health and outcomes. To reduce the risk of pre-eclampsia, daily calcium supplementation for populations with low dietary calcium intake may be advised, although negative interactions between iron and calcium supplements may occur so these two nutrients should be administered several hours apart. In undernourished populations, balanced energy and protein supplementation should also be recommended for pregnant women (though not specifically linked to a reduction in preterm birth). For populations at risk of vitamin D deficiency, possible benefits for general pregnancy outcomes may be gained from vitamin D supplementation. Where dietary zinc is low, it has been suggested that zinc supplementation may reduce the risk of preterm birth. However, further research is required to clarify the benefits of supplementation. For example, vitamin D in combination with calcium may increase the risk of preterm birth. In the antenatal period, the most important focus should be on promoting a good quality diet in general, rather than a specific supplementation regime.

Abstract:
Demands for plant-based food in the European Union are growing especially nowadays. Pulses are common and regional; they have an excellent nutritive value but its consumption in food industry is still low. In last decades extrusion technology has become extremely popular in food development. The aim of this study was to investigate the latest findings about pea Pisum sativum L. and beans Phaseolus vulgaris L. flour suitability for high-moisture food extrusion. Monographic method was used to analyse pulse seeds chemical content, possible pre-treatment methods, functional properties, covering the latest information about high-moisture extrusion, raw materials and technical parameters used. Pulses are a good source of protein, carbohydrates, minerals and vitamins. Chemical content, functional properties of yellow peas and different varieties of beans, except grey peas are well described in scientific literature. Germination can be promising pre-treatment for pulse seeds, it increases folic acid content, water absorption capacity and reduces the amount of antinutrients. Extrusion technology increases ready product protein digestibility and induces changes in antinutrient activity. High-moisture food extrusion characterises with moisture of raw materials above 40.00% where mostly protein concentrate and isolates from pulses are used.

Mung bean [Vigna radiata] is an important grain legume crop in Asia, particularly in India. Mungbean is consumed in the United States by a niche market in Asian cuisines, mostly as bean sprout, but is on the rise for use as an alternative plant protein as it is very high in micronutrients such as zinc, iron, and folic acid while also being low in fat and high in protein. Currently, more than two-thirds of mungbeans consumed in the United States are imported and nearly all domestically farmed mungbeans are produced in Oklahoma. This project explores the viability for mungbean to be produced by Virginia tobacco farmers as an alternative specialty or rotation crop since the demand for tobacco is on the decline. Two cultivars commercially available in the United States, Berken and OK2000, were planted in two locations in southern piedmont Virginia with an early and late planting date to determine the optimal time to plant mungbean and if it is a feasible crop in Virginia. Location played the most determining factor in overall seed size (g/100 seeds) for both cultivars--which is an indicator of quality--but the overall yield was not affected by location or planting date for either cultivar. Berken yielded an average of 771.03 lbs/ ac and OK2000 yielded an average of 728.81 lbs/ac as compared to 1000 lbs/ acre being considered “good yield.” This study suggests that an early versus late planting date has minimal effect on overall yield of mungbean, and with proper management techniques, there is potential for production in Virginia. This project is important to the AOCS community because it looks into the potential for mungbean to be grown in greater acreage within the USA as an alternative plant protein crop.

Abstract:
Traditional food plays an important role in local identity, consumer behaviour, the transfer of cultural heritage to future generations and the interaction of this heritage with the rest of the world. Local products are, as a rule, natural products, which have a high nutritional and biological value and are perfectly inscribed in the national food traditions. Until the advent of agriculture, the fi rst foods for our ancestors were those picked up from nature. Th ey had a longer life expectancy and a stronger immune system, and this is probably due to their diet rich in herbs and raw products. Nettles play an important role in nutrition and health, especially in spring aft er the relative shortage of fresh sources of vitamins in the winter period, when the nutritional qualities of vegetables and food are low. Th e preliminary study carried out in the fi eld, found that nettles were used in various traditional dishes in various regions of the Republic of Moldova such as: soups, broths, stews, sauces, fasting food, pilafs, salads, pies, canned food for the winter, spices, drinks, tinctures, etc. Th ese plants were also used in traditional medicine, methods of care and health, craft smanship (making textile fi bers, coarse fabrics, fi shing ropes, paper), fodder for animals, destruction of insects and pests in the garden, etc. According to popular beliefs, many customs and superstitions are related to nettles. Th ey are associated with their protective force, with their endowment to heal and maintain people’s health, to remove unclean forces; bundles of nettles were hung at the entrance and corners of the house, animal stables, etc. In the framework of the study of the state program 20.8009.0807.17 analyzing the composition and physico-chemical properties of nettles and of dishes with the use of nettles, a fairly high biological value of these plants was noted. Nettles are rich in substances of protein nature, having a large number of amino acids, carbohydrates, vitamins C, B2, and K pantothenic acid, folic acid, chlorophyll, carotene, calcium salts, magnesium, iron, silicon, phosphates, etc.

Abstract:
The concept of food as medicine is not new. The use of foods to prevent and/or treat certain diseases can be found in ancient drawings and writings. The most famous statement came from Hippocrates, who said “Let food be thy medicine.” It is the position of the Academy of Nutrition and Dietetics to recognize that although all foods provide some level of physiological function, the term, “functional foods” is defined as whole foods along with fortified, enriched, or enhanced foods that have a potentially beneficial effect on health when consumed as part of a varied diet regularly at effective levels based on significant standards of evidence. The most prominent results indicated that high total intake of fruits and vegetables, and some of their specific subgroups including berries, citrus, and green leafy vegetables, may promote higher levels of optimism and self-efficacy, as well as reduce the level of psychological distress, ambiguity, and cancer fatalism, and protect against depressive symptoms. Flavonoids are a class of organic polyphenolic compounds found in varying concentrations in plant-based whole foods such as berries, tea, cocoa, soybeans, and grains. Recent studies suggest that flavonoids can be beneficial to both cognitive and physiological health. As such, long term chronic supplementation with flavonoids has been investigated extensively, particularly concerning cognitive ageing and related neurodegenerative disorders. Less attention has been given to the acute effect of flavonoids on cognitive outcomes, within the immediate 0–6 h post ingestion. Therefore, the general recommendation to consume at least 5 portions of fruit and vegetables a day may be beneficial also for mental health. Immediate cognitive enhancement is often desirable in academic and work environments, such as during an exam or assessment. Besides, support a positive role for the nutrients EPA, DHA, magnesium, alpha-tocopherol, and folic acid, either alone or in combination with drugs, in the preservation of normal brain function and mental well-being. In this study, the effects of consumption of some functions on mood, cognitive function and mental health were investigated. Scientific findings support the combination of micro and macronutrients in a balanced and varied diet along with a healthy lifestyle for the maintenance of normal brain function, improvement of mental abilities, concentration, memory and alertness. Food components actively participate in the generation of nerve impulses by influencing neurotransmitters that activate different parts of the brain, thereby regulating our mental abilities, emotions and mood.
