Abstract:
To establish an efficient screening system for differentially expressed genes of a phytopathogenic fungus Colletotrichum lagenarium, we constructed an equalized (normalized) cDNA library from C. lagenarium and used this library for differential screening. For the isolation of genes involved in infection-related developments of conidia, conidia undergoing appressorium differentiation were selected as the source of materials for construction of the cDNA library. The equalization of cDNA was performed twice using a kinetic method, and the products were cloned into a plasmid vector. Colony hybridization with nine probes of different abundance showed a reduction in abundance variation from at least 276-fold in the original library to 10-fold in the equalized cDNA library, which demonstrated that the cDNA was successfully equalized. By differential hybridization of 1900 cDNA clones in the equalized cDNA library and RNA blot analysis of candidate clones, we identified 11 independent cDNA clones, designated CAD1 through CAD11, that were expressed in appressorium-differentiating conidia, but not in vegetative mycelia. The transcripts of CAD1 and CAD2 hardly accumulated in preincubated conidia, whereas those of CAD3 and CAD4 accumulated highly and slightly, respectively. The amount of the four CAD transcripts increased at the early stage of the appressorium formation process. Sequence analysis of CAD1 revealed that CAD1 would encode for 101 amino acid polypeptides, which showed homology to metallothioneins. Deduced amino acid sequence of CAD2 would encode 278 amino acid polypeptides, and showed high homology to genes in aflatoxin, and sterigmatocystin gene clusters of Aspergillus parasiticus and A. nidulans, respectively. Abstract The pathological process underlying Alzheimer disease (AD) unfolds predominantly in the cerebral cortex with the gradual appearance and regional progression of abnormal tau. Intraneuronal tau pathology progresses from the temporal transentorhinal and entorhinal regions into neocortical fields/areas of the temporal allocortex. Here, based on 95 cases staged for AD-related neurofibrillary changes, we propose an ordered progression of abnormal tau in the temporal allocortex. Initially, abnormal tau was limited to distal dendritic segments followed by tau in cell bodies of projection neurons of the transentorhinal/entorhinal layer pre-α. Next, abnormal distal dendrites accumulated in the prosubiculum and extended into the CA1 stratum oriens and lacunosum. Subsequently, altered dendrites developed in the CA2/CA3 stratum oriens and stratum lacunosum-moleculare, combined with tau-positive thorny excrescences of CA3/CA4 mossy cells. Finally, granule cells of the dentate fascia became involved. Such a progression might recapitulate a sequence of transsynaptic spreading of abnormal tau from 1 projection neuron to the next: From pre-α cells to distal dendrites in the prosubiculum and CA1; then, from CA1 or prosubicular pyramids to CA2 principal cells and CA3/CA4 mossy cells; finally, from CA4 mossy cells to dentate granule cells. The lesions are additive: Those from the previous steps persist.

La ricerca si applica allo studio di decorazioni compositive a matrice geometrica originate dalle mutevoli combinazioni di forme elementari: la divisione regolare del piano con forme chiuse che lo ricoprono completamente, senza sovrapporsi e senza mai lasciare spazi vuoti. Operando su di esso, si inizia a quantificarlo, individuando ogni sua singola parte: la misurazione conduce alla scomposizione in forme elementari di figure complesse. Nello sviluppo di tali configurazioni, il problema della replica è sempre presente, dalla mera idea progettuale fino alle azioni di restauro, in quanto dalla ripetitività delle singole parti – fondamento del suo stesso esistere – prende origine l’intera composizione. Si è cercato, quindi, a partire da un’attenta analisi delle leggi simmetriche che dettano le logiche generative del disegno ornamentale, per una ben precisa canonizzazione di temi e motivi, di identificare gli schemi geometrici che consentano il controllo razionale della decorazione tramite l’individuazione dei procedimenti della iterazione in serie. Non si restituisce semplicemente il dettaglio così come ci appare, ma la scomposizione teorica dell’opera permette di individuare, nella successiva ricomposizione grafica, la natura e la posizione di ogni singolo elemento: uno sguardo critico che guida a una corretta e realistica riproduzione. I modelli formali tendono a divenire così modelli geometrici, simbolo di una struttura più profonda, che è possibile cogliere con l’aiuto di precisi schemi topologici. Attraverso i casi studio esaminati, ed in particolare in funzione del sistema decorativo riscontrato, è stato possibile individuare procedure metodologiche di rilievo ritenute idonee ad esplicitare il processo geometrico di base al fine di facilitare la successiva fase di replica sia in ambiente digitale che fisico e di favorire una funzionale sistematizzazione dell’ornato. In particolare si è ricorso ad un’innovativa metodologia, detta di photo scanning, che permette l’acquisizione simultanea di nuvole di punti e di texture. E così, nota la legge armonica che domina l’intera composizione, definita dal movimento rigido di un modulo predefinito su di un reticolo piano, la decorazione può facilmente riprodursi partendo dalla semplice unità quale entità che può essere, con l’utilizzo di macchine a controllo numerico, reiterata meccanicamente. La procedura è stata applicata a casi studi idonei per analizzare i principi della ripetibilità in serie e delle operazioni di laboratorio; ciò ha consentito, grazie alla ri-progettazione digitale e alla produzione con gli strumenti della tecnologia a controllo numerico, di codificare la riproducibilità tecnica, immateriale e materiale, del decoro architettonico. Le procedure e le strumentazioni descritte possono essere un potente metodo per il controllo, la verifica e il monitoraggio del decoro architettonico, permettendo di realizzare database metrici interattivi in grado di fornire in qualsiasi momento informazioni sull’oggetto rilevato. Impostare un modello di schedatura dei rivestimenti geometrici costituisce uno strumento di catalogazione funzionale sia alla tutela e alla fruizione del bene immobile, sia alla definizione di un metodo di approccio allo studio dei gruppi di simmetria. Si sarà così in grado di stabilire l’origine degli schemi decorativi, i percorsi di distribuzione, lo sviluppo di tendenze locali. [a cura dell'autore]

La condizione digitale che ha gradualmente ibridato le nostre esistenze, trasformando atomi in bit, si è oggi cementificata sulla nostra società, arricchendone la postmodernità e determinando una nuova liquidità acuitasi con l’avvento di internet. Un momento storico segnato da una nuova maturità del digitale, evidente nel nostro diverso rapporto con i dati, e nella diffusione di metodi di machine learning avanzato, che promettono una nuova capacità di comprensione della complessità contemporanea e nel frattempo contribuiscono alla propagazione dell’apparato tecnico sul mondo. Questi cambiamenti, tanto profondi da toccare la nostra cultura, stanno modificando il nostro modo di interpretare e istituire lo spazio, e quindi di abitarlo: condizioni che hanno sicuramente delle ripercussioni sul progetto di architettura nella sua qualità di attività umana rivolta all’uomo. L’incremento di complessità che ha toccato la nostra disciplina con la postmodernità ha nel frattempo trovato nuovo sostegno nella decostruzione Derridiana, in un momento storico segnato da una grande enfasi sulle opportunità degli strumenti digitali, che abbiamo accolto all’interno della nostra disciplina dapprima esclusivamente come mezzi di rappresentazione e che hanno poi determinato l’emergere di nuovi approcci basati sulle potenzialità inclusive di continuità e variazione. Nessuno tra i protagonisti della prima svolta digitale immaginava probabilmente, gli effetti che la cultura digitale produce oggi sul progetto di architettura, forte di quasi trent’anni di sperimentazioni e cambiamenti, tanto metodologici e formali quanto organizzativi e strumentali, a partire dalla ascesa del BIM sino alle nuove possibilità algoritmiche ben rappresentate dai linguaggi di programmazione visuale e dalle simulazioni numeriche. Strumenti su cui si è concentrato lo slancio verso il digitale, che intanto in architettura ha vissuto una seconda svolta, identificata da Carpo negli approcci progettuali oggi possibili grazie a una nuova disponibilità di dati. Una condizione che inevitabilmente tocca tanto la scienza quanto l’architettura, e che non è tuttavia sufficiente a descrivere una contemporaneità in cui la tecnica dispiega le ali sull’architettura, incidendo il significato del nostro ruolo all’interno della società. A partire da queste ramificate considerazioni, consapevole della complessità con cui dobbiamo dialogare nel tentativo di ricostruire una visione il più possibile neutrale, storica e organica della fase che l’architettura sta vivendo, è necessario, a mio avviso, un approccio olistico: inclusivo, capace tanto di estendersi fino ad acquisire una prospettiva filosofica, così come di scendere verso il dettaglio tecnico, operativo, metodologico, strumentale e relazionale. Un proposito che cerco di mantenere vivo all’interno di tutto il mio lavoro di tesi, condensazione di tre anni di ricerca, che nelle sue diverse fasi guarda alle mutazioni che la tecnica digitale sta producendo nella società e quindi nel progetto di architettura. Il mio percorso è arricchito da dieci interviste raccolte con importanti protagonisti dell’architettura contemporanea, che ringrazio sin da ora per la loro grande disponibilità. Queste testimonianze mi hanno permesso di toccare con mano le complessità della progettazione contemporanea e rappresentano un polo di questa tesi, ugualmente volta a fornire un’interpretazione storica delle sfide poste in essere dalla contemporaneità e quindi all’identificazione delle responsabilità che dobbiamo assumerci per mantenere l’uomo al centro del nostro fare.
The digital condition that has gradually hybridized our lives, transforming atoms into bits, has now cemented itself in our society, enriching post-modernity and determining a new form of liquidity that has sharpened with the advent of the internet. It is a historical moment marked by a new digital maturity, evident in our diverse relationship to data and in the spread of advanced machine learning methods, which both promise a new understanding of contemporary complexity as well as contribute to the propagation of the technical apparatus throughout the world. These changes, so profound as to affect our culture, are changing our way of perceiving space, and therefore of inhabiting it: conditions that undoubtedly have repercussions on architectural design in its capacity as a human activity geared towards human beings. The increased complexity that has touched our discipline with Postmodernism has meanwhile found new support in Derridian deconstruction, in a historical moment marked by great emphasis on the opportunities that digital tools offer. These are means we first welcomed into our discipline exclusively as tools for representation, and ones that then themselves determined the emergence of new approaches based on the inclusive potential of continuity and variation. None of the protagonists of the first digital turn could probably have imagined the effects that digital culture would now be having on architectural design. A digital culture that has become increasingly stronger due to almost thirty years of both methodological and formal experimentation, as well as to organizational and instrumental changes, starting with the rise of BIM to new algorithmic possibilities represented by visual programming languages and numerical simulations. These have been the primary tools of concentration in the push towards digital, a digital which today has reached a second turn in the field of architecture, identified by Carpo in new design approaches that are now possible thanks to the larger availability of data. A condition that inevitably affects both science and architectural design, but which, nevertheless, fails to fully share a contemporaneity where technology spreads its wings as far as architecture is concerned, thus affecting the meaning of our role within society. With these multifaceted considerations as a starting point, and fully aware of how complex the dialogue we must engage in in order to reconstruct a neutral, historical, and organic as possible vision of the phase that architecture is experiencing, it is my opinion a holistic approach must be established by us. One that is both inclusive and capable of expanding to the point of acquiring a philosophical perspective, as well as being able to attend to areas that cover technical, operational, methodological, instrumental, and relational details. This objective is one I have striven to keep alive throughout the three years of my doctoral research, which in its various phases looks at the mutations that digital technology is producing in society and therefore in architectural design. My research is enriched by the inclusion of ten interviews with prominent protagonists of contemporary architecture, for whose time and availability I am grateful. These testimonials allowed me to see the complexities of contemporary design up close and personal, and they represent a central part of this thesis, which equally aims to provide a historical interpretation of the challenges posed by contemporaneity and to identify the responsibilities that we must uphold for human beings to remain at the centre of our work.

Abstract:
Este trabalho descreve uma metodologia para a geração de programas CNC para otimizar um sistema produtivo. Quanto às informações geométricas, o sistema caracteriza-se por utilizar os ciclos de usinagem que o comando da máquina disponibiliza, por possibilitar a aplicação de ciclos desenvolvidos com recursos de programação parametrizada e por gerar novos ciclos, através de uma linguagem genérica, proposta neste trabalho, em um editor de programação parametrizada. Quanto aos dados tecnológicos, ou seja, as condições de usinagem definidas no código de programação, o sistema permite a otimização dos parâmetros de corte através de um banco de dados com informações realimentadas com resultados obtidos da produção. A análise da viabilidade da metodologia proposta é realizada através da implementação de um protótipo de um sistema dedicado à programação de tornos CNC. O protótipo é composto de aplicativos computacionais desenvolvidos e orientados à otimização do sistema produtivo. A metodologia para geração de funções CAD/CAM foi implementada e testada mostrando resultados eficientes. O protótipo implementado como um conjunto de ferramentas aplicativas integradas é de fácil operação e utilização unindo todos os módulos projetados.
This study describes a methodology for the generation of CNC programs to optimize a productive system. Regarding geometrical information, the system is characterized by the use of machining cycles that the machine tool control provides because a allows the application of cycles developed with parameterized programming and by generation of new cycles, through a generic language, proposed in this study on a parameterized programming editor. As for technological data, i.e. the machining conditions set out in the programming code, the system allows the optimization of cutting parameters through a data base with information fed back with results of production. The feasibility analysis of the proposed methodology is accomplished through the implementation of a prototype of a system dedicated to programming CNC lathes. The prototype consists of computer applications developed and oriented to the optimization of the production system. The methodology for the generation of CAD/CAM functions has been implemented and tested showing effective results. The prototype implemented as an integrated set of applicative tools is of easy operation and use connecting all designed modules.

Este estudo apresenta a utilização de recursos CAD e CAE como ferramentas de desenvolvimento de novos produtos com proposição ao aumento dos índices de confiabilidade. Para atingir esta meta buscou-se a implementação de um componente do sistema de conformação mecânica por injeção em uma indústria fabricante de chuveiros elétricos. A aplicação de ferramentas qualitativas detectou as falhas críticas do referido sistema e delimitou uma especificação de projeto para concepção de alternativa de solução. A análise por elementos finitos (FEA), permitiu a construção de um novo modelo de agulha para o molde de injeção em consonância às premissas estabelecidas. Decorrido o período da implantação e ajustes pertinentes coletou-se novas amostras da aplicação prática, cujo objetivo foi comparar aos resultados do sistema convencional. Os resultados consolidados corroboram para um aumento significativo dos critérios de desempenho observados, sobretudo no que concerne às funções taxa de falhas, MTTF, e confiabilidade, permitindo-se inferir que os recursos computacionais, constituem-se como alternativa insofismável para resolução dos problemas projetuais desta natureza.

La tesi “Restituzione grafica della cosiddetta Casa Colonica a sud dell’area di Palazzo di Villa Adriana” si pone l'obiettivo di fornire una documentazione digitale della cosiddetta Casa Colonica mediante un modello tridimensionale realizzato con il software 3D Leica Geosystems Cyclone e l'estrazione da questo di elaborati tecnici significativi. La struttura della tesi segue le fasi del processo che a partire dal rilievo porta alla generazione di elaborati bidimensionali di documentazione tecnica di Beni Culturali, articolandosi in tre fasi ben distinte: la prima riguarda la ricerca storica della Villa imperiale, studiando approssimativamente l’intero complesso della villa per poi approfondire il caso di studio; la seconda riguarda la fase di rilievo spiegando la metodologia di rilievo, la strumentazione utilizzata e le varie operazioni svolte per ottenere il rilievo completo sotto forma di nuvola di punti; la terza riguarda la realizzazione degli elaborati: pianta della Casa Colonica; sezione longitudinale e trasversale. iSIGHT-FD manages the computer software required to execute simulation-based design processes, including commercial CAD/CAE software, internally developed programs, and Excel spreadsheets. The software enables the rapid integration of these programs and automates their execution to accelerate the evaluation of many more design alternatives. It provides design exploration and optimization technology to help the user find better designs than those that can be found manually. Inside of this framework, a set of interoperable CAD and CAE components is used to define how changes in the geometry affect the product performance. In the past year Engineous worked with several of its partners to create a seamless and automated process. We will illustrate how to setup and drive geometric changes in CAD models using the translation technologies component in iSIGHT-FD. The user can set loads & boundary conditions on the part/assembly that move with the geometric modifications. The changed geometry with loads & boundary conditions can be automatically meshed and analyzed with FEM solvers and pre/postprocessors. The user is allowed to store and view parametric and non-parametric results for every run. These 3D CAE & CAD results can then be easily embedded into Microsoft office.

Abstract:
Assembly models can be regarded as a kernel for product development processes where they can efficiently contribute to many product simulation behaviors. Assembly models are often containing 3D B-Rep CAD models, possibly with geometric constraints between the components and bill of materials. However, these models are often difficult to process for simulations because algorithms often face a very large diversity of configurations. One origin of such difficulties can be found in companies’ practice where components may be represented differently from one company to another and their interfaces as well. In any case, interfaces between components are not explicit, which leads to tedious model processing tasks. This paper illustrates preparation of assembly models to ease CAE through an analysis of company practices, showing that a concept of conventional representations is an important starting point to efficient treatments of assemblies. In addition, it is described how interfaces and conventional representations can be combined to derive functional and mechanical information from geometric models of components. Illustrations of the proposed approach is given throughout the paper using various standard components.

We study the potential benefits of adding a lottery component to cut the main risks associated with standard negotiated and rule-based auction procurement procedures. We show that adopting a two stage approach in which bureaucrats first negotiate with a small number of bidders to assess their eligibility and, next, rely on a lottery to award the contract reduces corruption risks often observed in negotiated procedures. For rule-based procedures, we show that a “third-price lottery” in which the two highest bidders are selected with equal probability and the project is contracted at a price corresponding to the third highest bid can reduce limited liability, renegotiation, bid rigging and collusion risks.
