Abstract:
Purpose The purpose of this paper is to add information on which voices contributed to the scientific management narrative from Frederick Taylor’s 1915 death to the early 1930s with a focus on the role of labor union representatives. The strategy is to analyze the role of labor representatives as participants in Taylor Society meetings and publications. The research contributes to the management history literature by bolstering the picture of the Taylor Society as a liberal, pro-labor organization. The research also shows that the Taylor Society was an early proponent of the idea that assembling diverse groups for dialogue improves organizational problem-solving. Design/methodology/approach The research analyzes historical sources including all issues of the Society’s bulletin from 1914 to 1933 and unpublished material from the Morris Cooke papers and the papers in the Frederick Taylor archive at Stevens Institute of Technology. Findings Taylor Society leaders took a proactive view of encouraging labor voices to join managers and academics in society meetings. At the beginning, few labor leaders spoke at the society, and often, at least some of their comments were critical of scientific management. By 1925, labor participation increased with William Green, American Federation of Labor (AFL) president appearing several times. In addition, labor leaders became positively inclined toward having scientific management experts working in industrial settings. The labor leaders who participated at Taylor Society meetings in the late 1920s and early 1930s considered scientific management insights as useful for labor and wanted to cooperate with the researchers. Originality/value The paper augments a revisionist view of interwar scientific management as progressive and pro-labor, a contested point in the management history literature. The research also shows how the Taylor Society was an early proponent of the importance of diversity, at least in the areas of gender and socioeconomic status, for effective problem-solving.

Abstract:
This article considers imperial Roman and German forms of liberal elite consensus on “proper religious diversity” to set the stage for an examination of the contemporary form of liberal consensus discernible in a recent public talk given by Charles Taylor and Rowan Williams. In each case, attention is drawn to the ways in which “proper religious diversity” is defined to serve ideological and theological agendas. Romanitas, Germanentum, and the Taylor–Williams consensus are cannibalistic theological systems: each uses a public stance of reasonable openmindedness regarding “proper religious diversity” to build and police a theological position that arrogates the perceived value of selected “religious” traditions by re-making them in its own image. Imperial Romanitas and Germanentum served in this way to absorb a diversity of traditions deemed palatable and digestible into overarching theological visions which were convenient to those in power. When Taylor and Williams use the platform of Religious Studies to promote the modern liberal view of “diversity in (true) religion” as a source of pro-social order, their efforts serve a similar crypto-theological and imperialist project.

Abstract:
Purpose This paper aims to provide evidence of pro-worker orientation and acceptance of socialist idealism in scientific management, with particular focus on Walter Polakov. Design/methodology/approach A range of original texts have been examined to identify the ideas expressed or accepted by the early scientific managers. These include Bulletin of the Taylor Society and the early publications of the socialist engineer and scientific manager Walter Polakov. Findings This paper shows how an avowed socialist is outspoken but unremarkable for the members of the Taylor Society in the 1910s and 1920s, contrary to the views expressed in textbooks and other histories which assert a deep antiworker bias in scientific management. Research limitations/implications This is limited to a historical analysis of the role and extent of involvement of the Marxist engineer Walter Polakov in the US scientific management movement in the 1910s and 1920s. Originality/value This paper offers insights into the workings of the Taylor Society using a biographical approach. In so doing, it demonstrates, in a new way, the verity of claims that the original proponents of scientific management were not authoritarian or anti-worker in their views or ideals, but, rather, open to progressive and socialist ideals.

Článek se věnuje nejnovější knize Charlese Taylora Secular Age (2007). Poukazuje na její kořeny v hegeliánství a v novějších kritikách modernity a probírá některé námitky vůči Taylorově koncepci včetně Taylorových odpovědí. Taylor představuje dva typy současných etických projektů. Na jedné straně tzv. výlučný humanismus, na druhé straně imanentní antihumanismus. A smyslem jeho genealogie moderního sekularismu je ukázat vzájemnou podmíněnost mezi antihumanismem, humanismem a religiozitou s tím, že bez nějaké formy transcendence se každý západní hodnotový systém vystavuje nepřekonatelným tenzím, aby následně navrhl koncept originality volající k následování jako spojnici hlavního nároku křesťanství ztělesněného v pojmu lásky k bližnímu s moderním uměním jako náhražkovým náboženstvím. Taylorova práce je nejen v tomto ohledu postavena na jeho celoživotním zájmu o Hegelovu filosofii, v níž se hledá takový způsob, jak spojit etiku autenticity se smyslem pro společenství, který by překonal omezení reflektující racionality. Taylorovo obohacení Hegela můžeme chápat jako snahu nahradit tradiční nárok lásky k bližnímu srozumitelnějším pojmem podpory rozmanitosti v jednotě (resp. pojmem kultury). The essay examines Charles Taylor’s latest book The Secular Age (2007) by unveiling its Hegelian roots together with its debt to other critics of modernity and by considering some objections as well as Taylor’s answers. Taylor sketches two types of contemporary ethical projects. On the one hand he considers exclusive humanism, on the other the immanent antihumanism. The moral of Taylor’s story of modern secularism comes to his claim that the anti-humanism is enabled by humanism and humanism again by religiosity. According to him this comes to indispensability of religion: namely, some notion of transcendence is necessary for coherence of any set of values available to western man. Actually the originality inspiring its following is what he understands as the key element that is shared by the central claim of Christianity embodied in the notion of love (agape) overcoming the rigorism of explicit rules on the one hand and modern art as Ersatzreligion.(Not only) this aspect of Taylor’s recent work is rooted in his lifelong interest in Hegel, who looked for such pairing of the ethics of authenticity with a sense for community that could overcome the limits of reflective rationality. We can interpret Taylor’s amendement to Hegel as a substitution of „support for differentiation in unity“ (or simply „culture“) for „love for neighbour“.

Abstract: In this research, the researcher used the descriptive qualitative method to analyze the data of figurative language and stylistics discovered in Taylor Swift's selected song lyrics on the Folklore album. This research is intended to find out the types and meanings of figurative language used in Folklore album, and to find out the types and functions of stylistics used in Folklore album. The researcher found some types of figurative language which is used in Folklore album there are Metaphor 12, Allegory 4, Metonymy 3, Verbal irony 7, Hyperbole 7, Personification 7, Symbol 14, Paradox 11, Simile 12, Synecdoche totem pro parte 1, and Understatement 1. Moreover, it is possible to conclude that figurative language and style of language significantly contribute to song lyrics. So that's why the writer or songwriter uses figurative language and any style of language in sentences to make the song more engaging and to assist readers to think more about the message and the meaning behind it.

Abstract:
Increasing secularization, pluralization, and individualization have done much to weaken denominational identities and traditional religiosity in most Western countries since the 1960s, with the effect that—to echo Niklas Luhmann—being religious requires purely religious reasons. This also applies to young people, for whom religion is still an option, but precisely one option among others, and according to Charles Taylor quite a challenging one. In our article, we want to focus on young people who actively engage with faith and religion, and who take up a different position with regard to religion than their peers during their adolescence. The data are in-depth interviews with families with three generations present. We will explore the ways in which teenagers (aged 12–19) and young adults (aged 22–25) are confronted with religious issues, as well as how they decide upon these issues and justify their decisions. We will argue that both the societal context and the life phase of adolescence or young adulthood make it likely that a person will base decisions regarding religion upon the criterion of authenticity. Our findings demonstrate that especially positioning towards the question of belief can be a lengthy and conflictual process. We identified two main forms of religiosity that are socially accepted in contemporary society: deriving a sense of social responsibility from faith and transforming and translating belief and religious experience into secular contexts.

Free axial vibrations of non-uniform rods are investigated by a proposed method, which results in a series solution. In a special case, with the proposed method an exact solution with a concise form can be obtained, which imply four types of profiles with variation in geometry or material properties. However, the WKB (Wentzel-Kramers-Brillouin) method leads to a series solution, which is a Taylor expansion of the results of the pro-posed method. For the arbitrary non-uniform rods, the comparison indicates that the WKB method is simpler, but the convergent speed of the series solution resulting from the proposed method is faster than that of the WKB method, which is also validated numerically using an exact solution of a kind of non-uniform rods with Kummer functions.

Abstract:
Purpose This paper aims to estimate a New Keynesian small open economy dynamic stochastic general equilibrium (DSGE) model for Egypt using Bayesian techniques and data for the period FY2004/2005:Q1-FY2015/2016:Q4 to assess monetary and fiscal policy interactions and their impact on economic stabilization. Outcomes of monetary and fiscal authority commitment to policy instruments, interest rate, government spending and taxes, are evaluated using Taylor-type and optimal simple rules. Design/methodology/approach The study extends the stylized micro-founded small open economy New Keynesian DSGE model, proposed by Lubik and Schorfheide (2007), by explicitly introducing fiscal policy behavior into the model (Fragetta and Kirsanova, 2010 and Çebi, 2011). The model is calibrated using quarterly data for Egypt on key macroeconomic variables during FY2004/2005:Q1-FY2015/2016:Q4; and Bayesian methods are used in estimation. Findings The results show that monetary and fiscal policy instruments in Egypt contribute to economic stability through their effects on inflation, output and debt stock. The monetary policy Taylor rule estimates reveal that the Central Bank of Egypt (CBE) attaches significant importance to anti-inflationary policy and (to a lesser extent) to output targeting but responds weakly to nominal exchange rate variations. CBE decisions are significantly influenced by interest rate smoothing. Egyptian fiscal policy has an important role in output and government debt stabilization. Additionally, the fiscal authority chooses pro-cyclical government spending and counter-cyclical tax policies for output stabilization. Again, past values of the fiscal instruments are influential in the evolution of the future fiscal policy-making process. Originality/value A few studies have examined the interaction between monetary and fiscal policy in Egypt within a unified framework. The presented paper integrates the monetary and fiscal policy analysis within a unified dynamic general equilibrium open economy rational expectations framework. Without such a framework, it would not be easy to jointly analyze monetary and fiscal transmission mechanisms for output, inflation and debt. Also, it would be neither possible to contrast the outcome of monetary and fiscal authorities commitment to a simple Taylor instrument rule vis-à-vis optimal policy outcomes nor to assess the behavior of monetary and fiscal agents in macroeconomic stability in context of an active/passive policy decisions framework.

ABSTRACT Family as informal caregivers have an important role in caring for family members with HIV/AIDS. Therefore, the well-being of family caregivers becomes very important because physical and mental health disorders can have a negative impact on the management of family members with chronic diseases. This study aims to analyse the role of social support and coping strategies with the mental health of family caregivers of people with HIV/AIDS (PLWHA). The research method used was systematic review through article searches that were adjusted to the formulation of research questions with the PICO formula. Data were obtained from Pro Quest, Pub Med, Sage Journal and Taylor & Francis from 2014 to 2023. Article titles and abstracts were searched and selected based on inclusion criteria. The search results found 9 articles with studies showing that there was a significant association between family support, coping strategies, and mental health in family caregivers of PLWHA. The presence of social support and adaptive coping strategies support the mental health of families caring for family members with HIV/AIDS. Keywords : Social Support, Coping Strategy, Caregiver of HIV, Mental Health ABSTRAK Keluarga sebagai pengasuh informal memiliki peran penting dalam merawat anggota keluarga dengan HIV/AIDS. Oleh karena itu, kesejahteraan pengasuh keluarga menjadi sangat penting karena gangguan kesehatan fisik dan mental dapat berdampak negatif pada pengelolaan anggota keluarga dengan penyakit kronis. Penelitian ini bertujuan untuk menganalisis peran dukungan sosial dan strategi koping dengan kesehatan mental caregiver keluarga yang merawat orang dengan HIV/AIDS (ODHA). Metode penelitian yang digunakan adalah systematic review melalui penelusuran artikel yang disesuaikan dengan rumusan pertanyaan penelitian dengan rumus PICO. Data diperoleh dari Pro Quest, Pub Med, Sage Journal dan Taylor & Francis dari tahun 2014 hingga 2023. Judul artikel dan abstrak ditelusuri dan dipilih berdasarkan kriteria inklusi. Hasil pencarian menemukan 9 artikel dengan penelitian yang menunjukkan bahwa ada hubungan yang signifikan antara dukungan keluarga, strategi koping, dan kesehatan mental pada keluarga yang merawat ODHA. Adanya dukungan sosial dan strategi koping yang adaptif mendukung kesehatan mental keluarga yang merawat Orang Dengan HIV/AIDS (ODHA). Článek představuje koncept nových sociálních rizik vymezený autory jako Esping-Andersen (1999, Taylor-Gooby (2004, 2005) a Armingeon, Bonoli (2006) a obecné reformní trendy sociálních států v reakci na tato rizika. První část se věnuje rozvoji a reformním trendům v západoevropských zemích. Druhá část se zaměřuje na specifičnost nových sociálních rizik a reformních trendů v zemích střední a východní Evropy. Článek dochází k závěru, že obecné trendy reforem sociálních států v reakci na vynořující se nová sociální rizika v zemích západní Evropy a v zemích střední a východní Evropy jsou podobné. Důraz je v nich kladen na posílení konkurenceschopnosti a zaměstnatelnosti a na rozšiřování příležitostí, například celoživotním vzděláváním, nástroji aktivní politiky zaměstnanosti, ale také socializací pečovatelských služeb pro usnadnění harmonizace práce a rodinného života. Zároveň dochází k posílení principu vlastní zodpovědnosti za sociální blaho, k individualizaci sociálních rizik a zapojení dalších sociálních aktérů (především zaměstnavatelů) do řešení problémů s nimi spojených.

Dalam artikel ini saya ingin membandingkan empati dari sudut pandang fenomenologi dan Immanuel Kant. Artikel ini menawarkan cara pandang secara filosofis-etis. Fenomenologi menekankan pengalaman sebagai dasar untuk berempati. Sementara itu, Kant lebih menitikberatkan pada aspek rasio. Bogdan dan Taylor, sebagaimana dikatakan oleh Armada, mencetuskan gagasan one of them. Konsep one of them ini juga terkait dengan terminologi sorge dari Heidegger yang dijelaskan secara mendalam oleh Schutz sebagai kesadaran di sini-sekarang-kemudian (here-now-then). One of them dan sorge mendapat kepenuhannya dalam pengalaman. Namun di sisi lain Kant menolak pengalaman sebagai dasar etika. Pengalaman seringkali menampakkan partikularitasnya sehingga etika menjadi relatif dan bias. Dari pro-kontra dua pandangan ini timbul pertanyaan, apakah mungkin empati itu bertolak dari pengalaman dan terjadi secara rasional? Pertalian antara pengalaman dan rasio nantinya akan terlihat dalam pemaknaan yang mendalam atas relasi. In this article I will compare the value of empathy from the phenomenology’s and Immanuel Kant’s point of views. Phenomenology emphasizes empirical aspect as empathy basis. Whilst Kant counts heavily on rational aspect. Armada, citing Bogdan an Taylor, triggers a concept ‘one of them’. The concept relates to sorge described by Heidegger and elaborated by Schutz as a conciousness and cognition of here-now-then. The concepts of one of them and sorge have their completeness in empirical experience. In other hand, Kant rejects empirical experience as ethical base. Experiences are often observed in their particularity, hence ethics will be biased and relative. The disagreement between Kant and phenomenology arises a very question, is empirical and rational empathy posible? The interwoven connection of them is in deep import of relation.

Abstract:
This paper investigates the relationship between the quality of institutions and the cyclical properties of macroeconomic policies in the Brazilian economy in the recent period. We extend the monetary and fiscal policy rules proposed by Taylor (2000) to incorporate a proxy for institutional quality. In the empirical analysis, we estimate reaction functions for monetary and fiscal policies by the Markov-Switching method. This methodology allows us to analyze how changes in the quality of institutions might influence the guidance of the fiscal and monetary policies over the sample period. The major results maintain that both monetary and fiscal policies are significantly countercyclical in periods that exhibit higher levels of institutional quality and are pro-cyclical or acyclical in periods which exhibit lower levels of institutional quality. Thus, the quality of institutions plays a key role in the government's ability to implement countercyclical monetary and fiscal policies to stabilize the Brazilian economy over the business cycle.

Research of conscious and unconscious emotional processes at 167 patients with multiple sclerosis in a condition of remission was conducted. The conscious and unconscious alarm (according to tests Taylor and Lusher), conscious and unconscious aggression (according to Bass-darky test and Hand test) in comparison to indicators of personal scales on the Standardized multidimensional personal questionnaire and the clinical and anamnestic of data was studied. At most of patients the high level of conscious and unconscious anxiety, the increased unconscious aggression at standard values of the general conscious aggression is revealed. The correlation analysis between indicators of emotional pro- cesses of the personality and clinical and anamnestic data revealed reliable interrelation of conscious and unconscious alarm, and also a depression (according to the test of Zung) with the phenomena of personal disadaptation and disease severity while conscious and unconscious aggression (on the general indicators) such relationship was no that is important for psychological diagnostics and rehabilitation.

At two localities adjacent to Hatherton Glacier, Darwin Mountains, a thick glacio-lacustrine sequence underlies the main diamictite facies of the Darwin Tillite (Metschel Tillite), and at one of these locations a glacio-lacustrine sequence also conformably overlies the diamictite. These deposits record the initial advance and final retreat of the Gondwana ice cap. Other, pro- and periglacial phases within these sequences record several minor advances and retreats of the ice sheet. Carbonaceous sediments above and below the main diamictite phase show that vegetation was established near the ice margin, suggesting that present-day ice margin temperatures are colder than those experienced during the Permo-Carboniferous (Gondwana) glaciation. It appears that there was little or no erosion of the Devonian Taylor Group which underlies the glacial sequence. The apparent erosional nature of the contact between the Permo-Carboniferous glacial sequence and the overlying Permian coal measures is attributed to surface winnowing of unconsolidated tills and locally the units are conformable.

AbstractThe possibility of referential null subjects in Old English has been the subject of conflicting assertions. Hulk and van Kemenade (1995:245) stated that “the phenomenon of referentialpro-drop does not exist in Old English,” but van Gelderen (2000:137) claimed that “Old English has pro-drop.” This paper presents a systematic quantitative investigation of referential null subjects in Old English, drawing on the York-Toronto-Helsinki Parsed Corpus of Old English Prose (YCOE; Taylor, Warner, Pintzuk, & Beths, 2003) and the York-Helsinki Parsed Corpus of Old English Poetry (YCOEP; Pintzuk & Plug, 2001). The results indicate substantial variation between texts. In those texts that systematically exhibit null subjects, these are much rarer in subordinate clauses, with first- and second-person null subjects also being rare. I argue that the theory of identification of null subjects by rich verbal agreement is not sufficient to explain the Old English phenomenon, and instead I develop an account based on Holmberg's (2010) analysis of partial null subject languages.

Background:Baricitinib (BARI) is a JAK1/JAK2 inhibitor which provides improvements to clinical signs, symptoms, and patient-reported outcomes (PROs) in patients with rheumatoid arthritis [1, 2].Objectives:The effect of BARI on the relationship between disease activity and pain has been explored previously [3]. The purpose of this post hoc analysis was to determine the association between additional PROs (physical function, fatigue, and duration of morning joint stiffness) and disease activity status after 12 weeks of treatment and to evaluate whether patients with an inadequate response to methotrexate treated with BARI 4 mg experienced greater PRO improvement than patients treated with either placebo (PBO) or adalimumab (ADA) across all levels of disease activity.Methods:Data for these analyses were derived from the Phase 3 study RA-BEAM (N=1305; NCT01710358). Pain was evaluated using a 0-100 mm visual analog scale, physical function was assessed using the Health Assessment Questionnaire-Disability Index (HAQ-DI), fatigue was measured using the Functional Assessment of Chronic Illness Therapy-Fatigue (FACIT-F) scale, and duration of morning joint stiffness (MJS, minutes) was reported by the patient. Disease activity was measured using the Clinical Disease Activity Index (CDAI) and categorized as remission (REM, ≤2.8), low disease activity (LDA, >2.8 to ≤10), moderate disease activity (MDA, >10 to ≤22), or high disease activity (HDA, >22). Linear regression was used to model the relationship between change in PROs at Week 12 (response) and CDAI values at Week 12 (primary explanatory variable) to evaluate the extent of improvement in PROs with BARI relative to PBO and ADA across a spectrum of disease activity levels. Last observation carried forward was used to impute missing values.Results:At baseline, 91% of patients were classified as having HDA and 9% as having MDA by CDAI across all treatment groups. After 12 weeks of treatment, 2%, 7%, and 9% of patients achieved REM; 16%, 27%, and 33% of patients achieved LDA; and 33%, 40%, and 38% of patients achieved MDA with PBO, ADA, and BARI, respectively [3].At Week 12, the estimated changes in measures of pain and physical function, as well as duration of MJS, for BARI 4 mg were greater than both PBO and ADA at all disease activity level threshold values of CDAI (Table 1). Objective: This in vitro study aims to evaluate the effect of clinical sandblasting with 50 μm aluminum oxide and 30 μm silica-coated particles on the surface roughness of zirconia cores and the subsequent effect on their fracture resistance after veneering with composite using a specific repair kit. Materials and Methods: Zirconia cores (n=21) were digitally designed and milled from ZirCAD LT B1 (IPS e.max ZirCAD ) blocks using arum 5x-300 Pro (ARUM DENTISTRY). These cores were randomly divided into three groups: Group A: n=8, sandblasted with 50 μm aluminum oxide, and veneered with packable Z350 composite. Group B: n=8, sandblasted with 30 μm silicacoated particles and veneered with packable Z350 composite. Group C: control group (n=5), sandblasted in the laboratory with 110 μm aluminum oxide and veneered with porcelain (Vintage Zr PRO - SHOFU Dental GmbH). All the specimens were tested for surface roughness by the TAYLOR-HOBSON profilometer. After adding veneering material, all the specimens were subjected to a fracture resistance test through a universal testing machine. Results: One-way ANOVA test showed a significantly higher surface roughness in group B compared to group A. Fracture resistance values showed no significant difference between all the groups. Conclusion: Silica-coated particles produced higher surface roughness than aluminum oxide alone. The fracture resistance values of all the groups were above the acceptable clinical limit. Slot antenna elements are quite popular for use in flat-panel arrays, due to their compactness and relatively large bandwidth. According to Babinet’s Principle, slots and dipoles are duals in performance. Therefore, a vertically placed slot is horizontally polarized and a horizontally placed slot is vertically polarized. A rectangular slot array lends itself to a broad range of applications, e.g., tactical and weather radar. Such arrays can be gimbaled to point their beam in any azimuth and elevation angle of interest. In this effort, a slot antenna element is designed in WIPL-D Pro to operate over the 7.5 - 8.5 GHz frequency range. The element is designed to resonate at 8 GHz. The slot element is used to generate a 6x12 slot array normal to the xy-plane. Separate Taylor tapers (nbar = 5) are applied to the 6-row and 12-column elements to provide 25 dB and 30 dB sidelobe level (SLL) suppression, respectively. The input impedance and pattern performance for a single slot, as well as the slot array’s radiation performance show excellent broadband performance.

Ivanchenko et al. “Inventory of stress symptoms”, the test “Probability of Stress Development” by J.H. Taylor adapted by T.A. Nemchin, self-assessment test of stress resistance by S. Couchen and G. Willianson, methods for determining the dominant state of L.V. Kulikova. The results of diagnostics of high school students’ stress resistance, likelihood of stress development in them, inventory of stress symptoms and dominant states, as well as correlation relations of the studied variables are presented. An analysis of the effectiveness of the program developed by the authors to increase the stress resistance of an individual in high school age, implemented in a secondary school, is pro-vided.

Objectives/aimsElevated levels of pro-inflammatory cytokines are associated with a range of mental illnesses, and also with childhood trauma.1–4 Evidence suggests that inflammation occurring as a result of childhood trauma could play a role in the pathogenesis of psychiatric disorders.5 6 However, previous research does not account for confounders, including psychotropic medication and illness duration, that may affect inflammation levels.7 The aim of this study was to investigate the relationship between childhood trauma exposure and levels of inflammation in young people with emerging mental health disorders with confounding factors removed.Methods10 patients, aged 16–25, with a range of psychiatric disorders and no history of psychotropic medication were recruited from Birmingham mental health services, along with 16 healthy controls. Childhood trauma exposure was assessed with the Childhood Trauma Questionnaire (CTQ), and blood samples were analysed for serum concentrations of pro- inflammatory cytokines IL-6 and IL-1β.ResultsThere was no significant difference in serum cytokines between patients and healthy controls, or those with and without childhood trauma (p>0.05). Post-hoc sample size calculations estimated 197 patients with and 197 without childhood trauma would be needed to detect a significant 2 pg/ml difference in cytokine levels (80% power, 5% significance). There was a weak positive, but non-significant, correlation between both cytokines and CTQ score in the patient group (rS=0.1–0.3, p>0.05). Multiple linear regression of CTQ score, sex and BMI found that none of these variables significantly predicted cytokine concentration in our population.ConclusionsThere is preliminary evidence of an association between childhood trauma and inflammation in patients with mental illness, but due to small sample size no definitive conclusions can be drawn from this data. However, recently various studies have shown its adverse effect on glucose control among diabetic patients and the U.S. Food and Drug Administration have revised statin drug labels to include information that increases in fasting serum glucose and glycated hemoglobin levels have been reported. This systematic review objective is to evaluate the risks and benefits of statins in glucose control management of type 2 diabetes patients based on the 44 published journal articles included and obtained through MEDLINE full text, PubMed, Science Direct, Pro Quest, SAGE, Taylor and Francis Online, Google Scholar, High Wire, and Elsevier Clinical Key. Statins were found to affect glucose control through several ways, namely, by affecting insulin production and secretion by β-pancreatic cells, insulin resistance, insulin uptake by the muscles and adipocytes and production of adipokines. Current evidence available shows that most of the statins give unfavorable side effects with regards to glucose control among diabetic patients. A dose-dependent and time-dependent effect was also observed in some statins which may be present among other statins as well.

Abstract Anthracyclines such as daunorubicin and doxorubicin (DOX) are currently used as chemotherapeutic agents in the prevention of various cancers including breast cancer, lung cancer, leukemia, and lymphoma. Although, anthracyclines are successful in controlling various cancers growth, the major drawback is the unwanted cardiotoxicity. Specifically, high doses of DOX used for the therapy of advanced cancers could cause life threating conditions. Therefore, specific interventions are required to decrease the cardiac toxicity associated with the DOX. Benfotiamine, a lipid soluble vitamin B1 derivative has shown be a potent anti-oxidant and anti-inflammatory agent. However, the efficacy of benfotiamine in the prevention of cardiotoxicity associated with the anthracyclines is not known. In this study, we examined the effect of benfotiamine in the prevention of DOX-induced cytotoxicity in the human umbilical vascular endothelial cells (HUVECs). Treatment of HUVECs with DOX caused significant endothelial cells death and benfotiamine in a concentration-dependent manner prevented the dox-induced endothelial cell death. Further, benfotiamine prevents the DOX-induced apoptosis in endothelial cells by preventing the activation of caspase-3. Benfotiamine also prevents DOX-induced reactive oxygen species generation. Our results also indicate that benfotiamine regulates DOX-induced expression of pro-apoptotic mediators such as BAD, phosphor-P53, and pro-caspase-3, and anti-apoptotic mediators such as BCL-2, BCL-x, IAPs, and HSP and others such as FADD, DR5, and SMAC/diablo. We plan to further explore how benfotiamine prevents DOX-induced cardiotoxicity using human cardiac myocytes and mouse models. Thus, based on our cell culture studies, benfotiamine through its potent anti-oxidative property could prevent endothelial cytotoxicity and suggests that it could be further developed as adjuvant therapy in controlling cardiotoxicity associated with the anthracycline chemotherapy. Classic methods of machining cylindrical gears, such as hobbing or circumferential chiseling, require the use of expensive special machine tools and dedicated tools, which makes production unprofitable, especially in small and medium series. Today, special attention is paid to the technology of making gears using universal CNC (computer numerical control) machine tools with standard cheap tools. On the basis of the presented mathematical model, a software was developed to generate a code that controls a machine tool for machining cylindrical gears with straight and modified tooth line using the multipass method. Made of steel 16MnCr5, gear wheels with a straight tooth line and with a longitudinally modified convex-convex tooth line were machined on a five-axis CNC milling machine DMG MORI CMX50U, using solid carbide milling cutters (cylindrical and ball end) for processing. The manufactured gears were inspected on a ZEISS coordinate measuring machine, using the software Gear Pro Involute. The conformity of the outline, the tooth line, and the gear pitch were assessed. The side surfaces of the teeth after machining according to the planned strategy were also assessed; the tests were carried out using the optical microscope Alicona Infinite Focus G5 and the contact profilographometer Taylor Hobson, Talysurf 120. The presented method is able to provide a very good quality of machined gears in relation to competing methods. The great advantage of this method is the use of a tool that is not geometrically related to the shape of the machined gear profile, which allows the production of cylindrical gears with a tooth and profile line other than the standard.

Abstract MDM2 inhibition has gained popularity in recent years as a therapeutic strategy in targeting both solid tumors and hematological malignancies, and several small molecule inhibitors of the p53-MDM2 binding interface have progressed into clinical trials in recent years. Alongside direct stabilization of p53, MDM2 has further been identified to have immune implications in terms of CD8+ T-cell mediated anti-tumor immunity and checkpoint-blockade therapies. However, little work has been done to explore the therapeutic targeting potential of related protein MDMX (MDM4). Pan-cancer analyses conducted by our lab (in part presented at the ASCO 2022 Annual Meeting) identify an important role of MDMX in several tumor types. Particularly, MDMX amplification is associated with a higher risk of brain and liver metastasis, and MDMX amplification itself also presents with significant frequency (10%) in glioblastoma multiforme (GBM). Notably, MDMX amplification also served as a better predictor of reduced overall survival in NSCLC following checkpoint-blockade. We have also previously reported on associations between MDMX amplification and CDKN2A deep deletions in melanoma, a predictor of favorable response to checkpoint blockade. Such findings establish the need for further exploration to be done and the role of MDMX both in cell phenotype and potential immune regulation to be better characterized. Preliminary data acquired from transient overexpression experiments using lipofection of MDMX cDNA and MDMX inhibitor XI-006 (NSC207895) alone and in combination with chemotherapy further supports the viability of MDMX-inhibition as a therapeutic strategy in several cell lines. Overexpression of MDMX in HCT116 p53WT cells demonstrably confers remarkable resistance to MDMX inhibitor XI-006 at a 72-hour timepoint (IC50 of 104.75 uM in overexpressing cells versus 29.03 uM in WT), and treatment with oxaliplatin reveals both reduced basal and post-treatment expression of p53-upregulated modulator of apoptosis (PUMA). Further mechanistic workup of this phenomenon is underway, particularly with an emphasis on immuno-oncological therapeutic translations. Altogether, initial findings ultimately establish the need for further investigation into the mechanistic basis of MDMX-correlated clinical outcomes and the therapeutic potential of MDMX inhibition. Abstract The tumor microenvironment (TME) plays a crucial role in cancer progression. It consists of various elements including stromal cells, proteins, and physical factors that support the growth and spread of cancer. Targeting the TME has become an appealing strategy for cancer treatment. Tumor-associated macrophages (TAMs) are an abundant stromal cell type in the TME and, due to their plasticity, TAMs can be exploited by cancer cells and the broader stromal reaction to promote a pro-tumoral response. TAMs have been implicated in facilitating a variety of pro-tumoral pathways including resistance to anti-cancer therapeutics, immune suppression, angiogenesis, and metastasis. Our group has recently characterized a subpopulation of pro-angiogenic and immune-suppressive TAMs which can be identified by their expression of the lymphatic vessel endothelial hyaluronan receptor-1 (LYVE-1). LYVE-1+ TAMs reside spatially proximal to blood vasculature and adopt a collaborative multi-cellular ‘nest’ arrangement which may support their biological activity. LYVE-1+ TAMs selectively express high levels of the immunomodulatory enzyme heme oxygenase-1 (HO-1), an enzyme which catabolizes heme to generate the biologically active catabolites carbon monoxide (CO), biliverdin and ferrous iron. Genetic inactivation of HO-1 in LYVE-1+ TAMs results in improved anti-tumoral CD8 T-cell responses and enhanced response to chemotherapy, resulting in prolonged tumor control in a spontaneous MMTV-PyMT murine model of breast cancer. Unfortunately, current HO-1 inhibitors are not orally bioavailable, which limits their utility as immunotherapeutics for the treatment of cancer. In response to this hurdle, we have developed a next generation HO-1 inhibitor called KCL-HO-1i. We demonstrate in pharmacokinetic studies that KCL-HO-1i is orally bioavailable in murine models with a serum half-life of around 3hours. Furthermore, orally delivering KCL-HO-1i alongside standard of care chemotherapy is able to deliver durable tumor control in MMTV-PyMT mice. We have characterized the response of the TME to KCL-HO-1i using flow cytometry, immunofluorescence and RNA sequencing approaches and demonstrate that KCL-HO-1i supports a broader switch from an immunological ‘cold’ to ‘hot’ TME which provides a more favourable immune landscape for improving the response to chemotherapeutic drugs. En segundo lugar, considera la respuesta rortyana a Taylor y sus propios argumentos contra el realismo de Taylor. Los descauerdos principales entre Rorty y Taylor son los siguientes: 1) Rorty dice que el pragmatismo supera la epistemología, Taylor, por el contrario, dice que el realismo no-comprometido es el que supera la tradición epistemológica popular; 2) ellos están en desacuerdo sobre cómo entender cierto vocabulario: Taylor afirma que siempre es posible conocer qué vocabulario describe mejor al mundo, pero Rorty parece sostener que los vocabularios son inconmensurables; 3) Taylor piensa a la verdad como una relación de correspondencia con el mundo; Rorty, sin embargo, rechaza esta concepción. Abstract Given an absolutely irreducible Galois representation : GE → GLN (k), E a number field, k a finite field of characteristic l > 2, and a finite set of places Q of E containing all places above l and ∞ and all where ∞ ramifies, there have been defined many functors representing strict equivalence classes of deformations of such a representation, e.g. by Mazur or Wiles in [15] or [26], with various conditions on the behaviour of the deformations at the places in Q and with the condition that the deformations are unramified outside Q. Those functors are known to be representable. For as above, our goal is to present a rather general class of global deformation functors that satisfy local deformation conditions and to investigate for those, under what conditions the global deformation functor is determined by the local deformation functors. We will give precise conditions under which the local functors for all places in Q are sufficient to describe the global functor, first in a coarse form, then in a refined form using auxiliary primes as done by Taylor and Wiles in [24]. This has several consequences. The strongest is that one can derive ring theoretic results for the universal deformation space by Mazur if one uses results of Diamond and Wiles, cf. [11] and [26], and if one has a good understanding of all local situations. Furthermore it is easier to understand what happens under increasing the ramification as done by Boston and Ramakrishna in [6] and [20], [21]. Finally we shall reinterpret the results in the case of a tame representation by directly considering presentations of certain pro-l Galois groups and revisiting the prime-to-adjoint principle of Boston, cf. Abstract Introduction: Children with H3K27-altered diffuse midline gliomas (DMG-H3K27a), have a 5-year survival rate of only 2% following diagnosis. While in-depth genomic characterization has been performed in DMG-H3K27a, very few druggable targets have been identified. A common feature of DMG-H3K27a is infiltration of microglia, macrophages, other myeloid cells, collectively referred to as GAMs. Cellular crosstalk between non-tumor and tumor cells in the tumor microenvironment (TME) can both promote and or inhibit tumor growth, thus representing an opportunity in the pursuit of novel therapeutics. We have recently determined that DMG-H3K27a tumor cells stimulate microglial secretion of pro-tumor growth factors. Hypothesis: Microglial-derived growth factors activate receptor tyrosine kinases (RTKs) via paracrine signaling in DMG-H3K27a. Methods: We used scRNA-seq and RNA-seq datasets and, in parallel, in vitro studies, such as co-culture studies, ELISA assays, western blotting, and growth assays to test our hypothesis. In addition, we are developing a mouse-human chimeric neutralizing antibodies of our top targets for in vivo studies. Preliminary Results: Microglial cells secrete growth factors into the TME, which promotes DMG-H3K27a cell proliferation and growth. Targeting growth factors with neutralizing antibodies attenuates glioma cell proliferation in co-culture studies. Conclusion: This research uncovers an important signaling network between GAMs and DMG-H3K27a that can be therapeutically targeted. Background:The efficacy and safety of baricitinib (BARI), an oral selective Janus kinase (JAK)1/JAK2 inhibitor, were evaluated in the randomized, controlled trial, RA-BEAM (NCT01710358), in patients (pts) with active rheumatoid arthritis (RA) and inadequate responses (IR) to methotrexate (MTX).1,2,3Objectives:To compare the time to onset and magnitude of improvement across different patient-reported outcomes (PROs) of BARI, adalimumab (ADA) and placebo (PBO) during the first 12 weeks of treatment in RA-BEAM.Methods:1,305 patients on stable background MTX were randomized 3:3:2 to PBO, BARI 4 mg, or ADA 40 mg. In this intent-to-treat analysis, least-squares mean changes and percentage changes from baseline were assessed up to Week 12 for pain (0-100 mm visual analog scale [VAS]), SF-36 physical component summary (PCS, 0-100), morning joint stiffness (MJS) severity (0-10), Health Assessment Questionnaire-Disability Index (HAQ-DI, 0-3), Functional Assessment of Chronic Illness Therapy-Fatigue (FACIT-F, 0-52), and Patient Global Assessment of disease activity (PtGA, 0-100 mm VAS) scores. PROs were compared between treatments with ANCOVA; the model included change from baseline as the response variable, baseline of interest, regional baseline, joint erosion status, and treatment as explanatory variables. Last-observation-carried-forward was applied to impute missing data. Speed of onset and magnitude of PRO improvement are presented in spydergrams.Results:Statistically significant improvements (P<0.05) with BARI and ADA vs. PBO were reported as early as Week 1 for pain, MJS severity, HAQ-DI, and PtGA and at Week 4 for FACIT-F and SF-36 PCS scores. Statistically significantly larger improvements (P<0.05) with BARI vs. ADA were observed as early as Week 2 for pain, PtGA, Week 3 for MJS severity, and Week 4 for HAQ-DI and SF-36 PCS scores. Para ello se recurre a una lectura crítica de las fuentes originales. Taylor justifica la necesidad de ofrecer salarios justos, pero propugna también por no compartir las ganancias equitativamente con los trabajadores de la empresa. Asimismo, más allá de una búsqueda aislada de un mero aumento de la eficiencia en la producción de la empresa, encontramos en su obra un Taylor que oscila entre una pretensión ulterior de explotar a los trabajadores y una pretensión ulterior de mejorar las condiciones de la sociedad en su conjunto, incluyendo las condiciones de estos mismos trabajadores, mediante el aumento precisamente de tal eficiencia.

Taylor—Couette flow is ideal for studying drag-reducing polymer additives because, unlike turbulent boundary layers, the instabilities are better understood. Video records of laser-induced fluorescence experiments with and without polymers will be presented. Polyethylene-oxide (PEO) ‘oceans’ were used in concentrations of 20 and 100 p.p.m. In the Taylor number range, 3 × 104 ≤ Ta ≤ 108, Newtonian flow consisted of Taylor vortices which span the gap between cylinders and much smaller Görtler vortices at the inner cylinder wall. Measurements of core-to-core separation between counter-rotating vortices were made to estimate the Görtler instability wavenumber. These measurements show that PEO addition increases the Görtler wavenumber for a given Taylor number. At the lower Taylor numbers, Görtler vortex formation was suppressed by PEO. This implies that polymers directly affect the evolution of centrifugal instabilities.

Abstract:
O estudo da evapotranspiração é essencial para o planejamento agrícola, bem como o manejo da irrigação é essencial em regiões como o semiárido proporcionado um desenvolvimento agrícola. O objetivo do trabalho foi avaliar equações empíricas em comparação ao método padrão de estimativa da evapotranspiração. Foram avaliadas três equações em comparação ao método de Penman-Monteith para estimativa de ET0, os métodos avaliados foram o de Hamon, Hargreaves e Samani e o de Priestley-Taylor. Utilizando dados diários do ano de 2015 para o município de Caruaru – PE as equações foram comparadas a partir de correlação, coeficiente de exatidão (d), coeficiente de desempenho (c) e regressão linear. Os métodos de Hargreaves e Samani e o de Hamon apresentaram desempenho abaixo do aceitável, mesmo apresentando coeficientes de correlação satisfatórios, esses métodos tendem a subestimar a ET0 para a região, já o método de Priestley-Taylor apresentou-se um melhor desempenho na correlação e coeficiente de exatidão. Os métodos de Hargreaves e Samani e o de Hamon não são recomendados para a localidade, sendo o de Priestley-Taylor o método mais preciso e de desempenho ótimo.

La filosofía política de Taylor recupera elementos clave del republicanismo clásico y, al mismo tiempo, no desestima principios esenciales del liberalismo igualitario. En oposición a la teoría de la justicia de Rawls, que critica por afirmar un conjunto unívoco de principios carentes de referente histórico, Taylor fija su atención en las aspiraciones reales de las sociedades modernas. Considera que estas se ven determinadas por visiones divergentes, pero complementarias: una aspiración republicana a la igual libertad y deliberación en común (principio de igual participación) y una aspiración a la prosperidad individual e ingresos diferenciales (principio de contribución). Piensa también que los rasgos privatistas de la cultura actual (movilidad social, consumismo, crecimiento de las megalópolis, erosión del sentido comunitario) alimentan una ideología atomista que favorece el principio de contribución y debilita la dimensión republicana de nuestra sociedad.

Abstract:
Background:Rheumatoid arthritis (RA) is a systemic inflammatory disease which includes increased innate and myeloid immune cell activation. Filgotinib (FIL), an oral JAK1 inhibitor, has shown safety and efficacy in three phase 3 studies (FINCH1-3) in adults with moderately-to-severely active RA. The baseline (BL) neutrophil-to-lymphocyte ratio (NLR) in RA has been associated with a positive response to anti-tumor necrosis factor (TNF) therapy1 and a negative response to DMARD triple therapy2. We previously reported a BL signature of clinical response in the FINCH2 (bDMARD-IR) population which included a neutrophil component3.Objectives:We conducted a post-hoc analysis to explore whether the BL NLR was associated with response to treatment in the FINCH studies.Methods:Clinical data of 3273 RA patients (pts) enrolled in the FINCH clinical trials (FINCH3, methotrexate (MTX)-naïve: NCT02886728; FINCH1, MTX-Inadequate Responder (IR): NCT02889796; FINCH2, bDMARD-IR: NCT02873936) were retrospectively analyzed for a relationship between the BL NLR and composite clinical endpoints (ACR-N, DAS28CRP, or CDAI) or PROs (Pain VAS, FACIT Fatigue, HAQ-DI) through week 24. Pts were classified as High or Low BL NLR using a cutpoint (2.7) identified as an independent predictor of treatment failure in a published RA study2. Adjusted clinical outcomes were estimated based on mixed effects linear regression models including geographic region and demographics covariates.Results:57% of pts enrolled in the FINCH trials were classified as BL NLR-High (Table 1) and FINCH3 NLR-High pts showed higher BL DAS28(CRP). FINCH1 and FINCH3 FIL+MTX-arm NLR-High pts demonstrated significantly better DAS28(CRP) response compared to NLR-Low pts (Figure 1). DAS28(CRP) differences between NLR-High and NLR-Low were detectable as early as Week 2 for FIL200mg + MTX and were sustained through Week 24. FINCH1 and FINCH3 FIL200mg + MTX NLR-High pts also demonstrated sustained clinical and PRO improvements over NLR-Low, including CDAI, ACR-N, Pain VAS, FACIT Fatigue, and HAQ-DI. The strength of these associations was dose-dependent; pts that received FIL100mg + MTX demonstrated weaker but directionally consistent trends in both populations. No significant association between NLR subgroup and clinical efficacy was observed in FINCH2 FIL+MTX-arm pts, FIL-monotherapy (FINCH3) pts, or adalimumab+MTX (FINCH1) pts.Conclusion:In FINCH1 (MTX-IR) and FINCH3 (MTX-naïve), FIL200mg + MTX -arm NLR-High pts demonstrate better sustained clinical response and PRO scores compared to NLR-Low pts. These data are the first to report an association between the BL NLR and therapeutic response in large randomized RA clinical trials. O objetivo deste artigo é traduzir o texto De motu Nervi tensi (Sobre o movimento de uma corda tensa) de 1713 do matemático inglês Brook Taylor (1685-1731), cujo intuito é descrever o movimento de uma corda vibrante com forte ênfase no seu número de vibrações em um determinado tempo. Fazendo uso da estrutura axiomática para cumprir tal finalidade, Taylor lança mão de princípios geométricos e mecânicos, este último com forte fundamento na obra Principia de Isaac Newton. Um aspecto notável do texto de Taylor é o estabelecimento de uma relação com o pêndulo, com intuito de calcular o tempo períodico da corda vibrante. Tendo em vista preservar maior fidelidadade à obra, utiliza-se como critério de tradução respeitar a sintaxe do texto original sempre que possível.

Abstract INTRODUCTION/METHODS: The multistep process of the tumor invasion-metastasis cascade is a major barrier to effective therapy. Mutations of the tumor suppressor p53 are frequently detected in esophageal squamous cell carcinoma (ESCC) cases, which in turn correlate with high metastatic rates. To understand the mutant p53-mediated mechanisms in promoting ESCC metastasis, we conducted RNA-Seq, ChIP-Seq and cytokine array on isogenic primary and metastatic tumor cells harvested from our mouse model of esophageal cancer, L2-Cre; LSL-Trp53R172H; Rosa26LSL-YFP, in which we have identified macrophage colony stimulating factor 1 (CSF1) to be a direct gene target of p53-R172H. We have targeted CSF1/CSF1R signaling to assess its pro-tumorigenic roles. In addition to studying tumor intrinsic mechanisms, we have established multiplex immunofluorescence and flow cytometry approaches to characterize the changes in tumor microenvironment (TME). Overall, this study aims to investigate the role and mediators of CSF1 signaling through its cognate receptor CSF1R by which missense p53 mutations can promote lung metastasis. RESULTS/DISCUSSION: We demonstrate that metastatic ESCC has increased Csf1 expression compared to primary tumors and this is dependent upon p53 mutation status, which is reinforced by TCGA data and patient-derived tissue microarrays (TMAs). The TMA analysis also suggested that CSF1 expression is higher in tumors that are poorly differentiated. Furthermore, through the proximity ligation assay (PLA), we have identified that bromodomain and extra-terminal (BET) protein BRD4, which binds to acetylated histones to regulate transcription interacts with p53-R172H at higher levels compared to wild-type p53 to induce Csf1 expression. We show that the BRD4/CSF1/CSF1R signaling axis fosters tumor invasion, subcutaneous tumor growth and metastatic burden in lungs in a mutant p53 background. In accordance, targeting CSF1/CSF1R signaling reduces phosphorylation of STAT3 in the metastatic tumors, which suggests a mechanism for increased invasiveness and metastasis. In parallel, upon inhibiting this axis, we have identified decreased infiltration of F4/80+CD163+ and F4/80+CD206+ M2-polarized macrophages, as well as CD31+ endothelial cells, and increased number of CD3+CD8+ cytotoxic T-cells at the metastatic tumor sites, indicating that the CSF1/CSF1R pathway plays a critical role in shaping the pro-metastatic and immunosuppressive TME. Finally, analysis of squamous cell carcinoma (SCC) datasets reveals that specific p53 mutations are associated with differential survival rates and CSF1 expression. CONCLUSION: We have demonstrated novel roles and mechanisms of mutant p53-dependent CSF1-CSF1R signaling pathway in fostering ESCC tumor invasion and lung metastasis that may be applicable to other SCCs. Abstract INTRODUCTION: Metastasis is associated with more than 90% of cancer-related mortality, and thus, there is a compelling need for innovative therapeutic breakthroughs. TP53 mutations are present in 60-70% of human cancers, especially in squamous cell cancers. For example, TP53 mutations are detected in up to 80% of esophageal squamous cell carcinomas (ESCCs). Accumulating evidence suggests that certain missense mutant forms of p53 such as R175 (R172 in mouse), R273 and R282 can acquire neomorphic pro-oncogenic activities that are referred to as gain-of-function (GOF). To elucidate novel mutant p53-dependent mechanisms in promoting metastasis, we conducted RNA-Seq, p53 ChIP-Seq and H3K27ac CUT&RUN-Seq on primary and metastatic tumor cells harvested from our mouse model of ESCC harboring Trp53R172H/−, Trp53−/− and Trp53+/+. Herein, we have identified p53-R172H gene targets in metastatic ESCC. One of the dominant effectors of p53-R172H-mediated metastasis was Colony stimulating factor-1 (Csf-1) signaling through its cognate receptor (Csf-1r), which we recently published in Cancer Discovery (PMID: 37676642). Overall, this study aims to investigate the GOF properties and mediators of mutant p53 in promoting ESCC metastasis. RESULTS/DISCUSSION: Using ex vivo cultures and in vivo tail-vein injections, we demonstrated increased colonization and metastatic capabilities of ESCC cells harboring p53-R172H in comparison to the tumor cells with null and wild-type p53. p53-R172H occupies more genes in the metastatic ESCCs than in the primary tumors that is independent of p53 expression levels. We identified 72 unique targets of p53-R172H with altered gene occupancy and expression that occurs during the transition from primary tumor to metastasis. Such targets in metastatic ESCC are exemplified by Csf-1 and Birc5 (encoding anti-apoptosis protein Survivin) that are also enriched with H3K27ac in their promoters. They are upregulated during metastasis compared to the primary tumors dependent upon p53 mutation status, which is reinforced by TCGA data and ESCC patient-derived tissue microarrays (TMAs). Interestingly, other frequently detected “hotspot” p53 mutations including R282W and R273C, which we introduced into the ESCC cells using base editing promote differential pro-tumorigenic activities. Indeed, the analysis of SCC datasets in TCGA reveals that specific p53 mutations are associated with altered overall survival, as well as differences in the enriched pathways. CONCLUSION: We have demonstrated GOF properties and mediators of p53-R172H in promoting ESCC metastasis that may be applicable to other SCCs. We are further expanding our analyses to additional “hotspot” p53 mutations to dissect the mechanisms of distinct p53 mutations in fostering ESCC metastasis, which can open up new avenues for therapeutic applications. Este artigo compara e contrapõe a abordagem naturalista pragmatista para a peculiaridade da linguagem, exemplificada, principalmente, mas, não exclusivamente, por John Dewey, com a extensa abordagem de Charles Taylor em seu O animal linguístico. Taylor, inspirado pelas obras de Hamann, Herder, e Humboldt, conta com recursos filosóficos e conceituais diferentes para o delineamento do que ele denomina de ‘a forma’ da capacidade linguística humana. Porém, Dewey e Taylor chegam a posições que se sobrepõem sem se identificar: a linguagem é a característica definidora constitutiva dos seres humanos. Seres humanos são definidos pelo surgimento da ‘como’ consciência, uma ‘ruptura’ em nossa imersão imediata no mundo, e, como Peirce e Dewey mostraram de maneira tão lúcida, um reflexivo estar consciente do uso de signos e sistemas de signos de todos os tipos. Esses sistemas potencializam e transformam nosso acesso ao mundo e a nós mesmos. Eles não apenas rotulam um mudo já existente. Eles criam âmbitos de significados e valores que não surgiram sem eles. A distinção crucial de Taylor entre os modelos designativo e constitutivo da linguagem é apoiada plenamente pela consideração pragmatista da linguagem, a qual Taylor não declara. Essa distinção mostrará ser de importância especial para Dewey e Taylor na criação de paisagens existencialmente vitais de significado incorporados nas autodescrições e nas práticas delicadas das artes de auto-reflexão. Tanto Dewey quanto Taylor mostram que assim como as texturas abertas da experiência crescem por suas extremidades, assim a própria linguagem possui sua própria “extremidade” e nos aponta para os domínios “liminares” que sustentam o limiar do sentido para além do totalmente dizível. Esses domínios, que eles mostram de maneiras diferentes mas complementares, são acessados como realidades por formas não discursivas que abrangem as obras de arte, o que Taylor denomina de ‘representações,’ e rituais performativos e restaurativos, tanto pessoais, cívicos e religiosos que incorporam os significados. Dewey e Taylor, divergem, entretanto, sobre se e de que maneira estes domínios precisam transcender a natureza.

Currently, the implementation of innovative non-invasive methods for controlling the state of the human organism under stress conditions is highly actual. Long-time intellectual burdens lead to the chronic stress and may result in the exhaustion of antioxidant system. The aim of the research is to study the characteristics of the antioxidant status in saliva under the conditions of intensive intellectual activities. In order to carry out the experiment, we collected the cohort of 123students aged from 20 to 22 years of the second and third years of the V.F.Voino-Yasenetsky Krasnoyarsk State Medical University. The examined materials, namely saliva, had been taken by direct spitting into test-tubes. Saliva was collected twice: firstly background sampling had been taken during the course of regular classes when the students were supposed to be in the state of emotional balance; secondly, the test sampling had been made in the time of examination session characterized by considerable intellectual burdens. We carried out questionnaire survey for the students using Spielberger-Hanin and Nemchin-Taylor tests. With the help of these tests, we determined the situational and personal anxiety as well as the predisposition to stress development. Antioxidant status was evaluated by Н2О2-luminol-dependent chemiluminescence technique. Chemiluminescent tests were performed by tablet luminometer TriStar LB 941, (made by Berthold). We had proved the existence of the association between the antioxidant status of saliva and the state of intellectual tension. We have noted that during a session in a group with the high level of uneasiness indices have grown, in a group with the low level of uneasiness they have decreased. We can suppose that in the time of examination sessions there is an increase in the common level of anxiety under the lowering of antioxidant activity. In intellectual stress, the antioxidant system performs the interception of radicals less intensively, because the speed of active forms of oxygen (AFO) neutralization is decreased. So, under the conditions of intensive intellectual activities, the degradation of antioxidant protection is observed. It is caused, supposedly, by the lowering of the activity of the enzymes of peroxidase protection. The pro-oxidant system also works less efficiently. This is confirmed by the decline of such markers of CL-fluorescence as maximum intensity, amplitude and light sum, which showed AFO number.

Abstract Activated mutants of KRAS comprise the major oncogenic drivers in lung (LAC), colorectal (CRC), and pancreatic ductal (PDAC) adenocarcinoma. Recent success in covalently targeting one KRAS mutant (KRASG12C) led to FDA approval of the first anti-KRAS therapy (G12Ci). However, both primary and treatment-induced acquired resistance to G12Ci have been observed. While analyses of relapsed patients have identified reactivation of the key KRAS effector signaling network as a driver of resistance, the mechanisms in ~50% of patients are not known. To identify potential resistance mechanisms, we applied a CRISPR-Cas9 loss-of-function screen targeting the druggable genome. In addition to genes discovered recently in relapsed patients (e.g., PTEN, NF1), we also identified loss of multiple components of the Hippo tumor suppressor pathway as drivers of G12Ci resistance (NF2, LATS1/2, TAOK1/2 and STK3/4). We therefore determined if activation of the functionally related transcriptional co-activators, YAP1 and TAZ, normally inhibited by Hippo signaling, can drive resistance to G12Ci. We first determined that ectopic expression of constitutively activated YAP1/TAZ was sufficient to impair the anti-proliferative and pro-apoptotic effects of G12Ci treatment in KRASG12C-mutant LAC, CRC, and PDAC cell lines. Conversely, genetic suppression of YAP1/TAZ enhanced G12Ci sensitivity. YAP1/TAZ requires association with TEAD and other transcription factors to regulate transcription. We determined that YAP1/TAZ mutants deficient in TEAD binding failed to drive resistance to G12Ci treatment. Further supporting a role for TEAD, both overexpression of a TEAD dominant negative mutant and treatment with pan-TEAD pharmacological inhibitors phenocopied the effects of YAP1/TAZ genetic suppression and sensitized KRASG12C mutant cancer cells to G12Ci. Finally, transcriptional analyses support a model where YAP1/TAZ-TEAD overcomes KRASG12C addiction by restoring a subset of KRAS-dependent gene transcription. In summary, our observations support YAP1/TAZ-TEAD signaling as a novel driver of resistance to KRAS inhibition and support the use of TEAD inhibitors to enhance the anti-tumor efficacy of KRAS-targeted therapies. Abstract:
Abstract While checkpoint inhibitor treatment has extended the survival of many cancer patients, most initial responders will later develop disease progression through a variety of poorly understood acquired resistance mechanisms (CPI-AR). One such recently described mechanism involves the transcriptional hyperactivation of genes associated with interferon (IFN) signaling, JAK/STAT, and antigen processing/presentation pathways. Aberrant IFN regulation in the setting of AR suggests that tumor cell intrinsic mechanisms have conferred a growth advantage despite the presence of immune-mediated pressure unleashed by PD-1/L1 antibody blockade. Analysis of transcripts progressively upregulated during acquisition of AR in vivo and transcripts upregulated by IFN exposure of isolated AR cell lines in vitro identified the E3 ubiquitin ligase TRIM7 as a candidate mediator of the AR phenotype. A specific pocket on the PRY/SPRY domain of TRIM7 has been shown to bind viral peptides, resulting in ubiquitination and degradation as a mechanism to mitigate viral propagation. Here we describe the evaluation of small molecule inhibitors (SMI) designed to the TRIM7 PRY/SPRY viral-pocket domain, where binding specificity was confirmed using mass spec analysis. These SMI competitively inhibited TRIM7 mediated stabilization of RACO-1, resulting in decreased cancer cell proliferation which was associated with altered c-Jun/AP-1 transcription. TRIM7 SMI also disrupted STING and MAVS ubiquitination and restored IFN responsiveness and sensitivity to PD-1 inhibitor treatment. Finally, given the position of TRIM7 downstream of growth factor receptor and kras signaling, combination of TRIM7 inhibitors with kras, braf, and/or MEK inhibitors was shown to additively suppress proliferation of tumor cells with kras mutations. These results provide evidence that upregulation of TRIM7 mediates both a pro-proliferative and IFN resistant phenotype in the setting of CPI-AR, and that inhibition of TRIM7 with small molecule inhibitors may prevent or reverse AR to PD-1/L1 blockade.
